A model system is described where Azospirillum and germinated wheat seeds were grown in association for a week and then assayed for nitrogen fixation (C2H2-reduction) and denitrification (N2O-formation) activities. The association performed C2H2-reduction and N2O-formation under microaerobic conditions. Both activities were measurable after already 3–5 h of incubation with substantial rates and were strictly dependent on the presence of both plants and bacteria. During the week of the growth of the association, the bacteria had lived exclusively from the carbon compounds supplied by the roots of the plants. C2H2-reduction activity by the association was more or less the same with all the Azospirillum brasilense strains, but lower with A. lipoferum and with the A. amazonense strains tested. Two nitrogenase negative mutants of Azospirillum brasilense showed virtually no activity in the association. C2H2-reduction activity was strongly dependent on the growth temperature of the association. Denitrification (N2O-formation) was high also at higher temperatures and at pH-values in the medium around 7.8 but not at neutrality and was strictly dependent on nitrate. The Azospirillum strain used strongly determined the rate of the N2O-formation in the association. Strains of the obligately aerobic nitrogen fixing organismAzotobacter chroococcum were constructed which contained defined chromosomal deletions in which the nitrogenase structural genenifHDK cluster (nifH for the polypeptide of the Fe-protein component of nitrogenase andnifD andnifK for the alpha and beta subunits respectively of the MoFe-protein component of the enzyme) was replaced by a kanamycin resistance gene. N2 fixation was nevertheless observed in deletion strains though only in a molybdenum-deficient medium or in spontaneously arising tungstate-resistant derivatives. In comparison with the parent strain growing in molybdenum-sufficient medium, diazotrophic growth was slow and the nitrogenase activity in vivo was characterised by disproportionately low rates of C2H2-reduction compared to H2-evolution and relative insensitivity of H2-evolution to inhibition by C2H2. The findings show reiteration of functional structural genes for nitrogenase inA. chroococcum consistent with our previous observation of twonifH genes in this organism and detection in this work of a secondnifK-like sequence in the genomes of both parent and deletion strains whenA. chroococcum nifK DNA was used as a probe.  相似文献

Azospirillum isolates were obtained from rhizosphere soil and roots of three cactaceae species growing under arid conditions. All Azospirillum isolates from rhizosphere and roots ofStenocereus pruinosus andStenocereus stellatus were identified asA. brasilense; isolates of surface-sterilized roots fromOpuntia ficus-indica were bothA. brasilense andA. lipoferum. Azospirilla per g of fresh root in the three species ranged from 70×103 to 11×103. The most active strains in terms of C2H2 reduction (25–49.6 nmol/h·ml) and indoleacetic acid (IAA) production (36.5–77 μg/ml) were those identified asA. brasilense and isolated from Stenocereus roots.A. lipoferum isolated from Opuntia roots produced low amounts of IAA (6.5–17.5 μg/ml) and low C2H2-reduction activity (17.8–21.2 nmol/h·ml).  相似文献

Summary Model experiments were performed to investigate the nitrogen fixation (C2H2 reduction) and denitrification (N2O formation) capabilities ofAzospirillum spp. in association with wheat. Plants and bacteria were grown together for a week and then assayed for activities. This association performed C2H2 reduction or N2O formation, depending on the concentrations of nitrate and oxygen in the vessels. Both activities depended on theAzospirillum strains used. The newly isolatedAzospirillum amazonense strains Y1 and Y6 showed significant C2H2 reduction and low N2O formation in association with wheat under the conditions employed and are possibly useful in practice. The potential for N2 fixation by heterocystous cyanobacteria isolated from soils of different geographical areas was determined as nitrogenase activity (NA) using the acetylene reduction assay. Morphology of cyanobacteria had the largest influence on NA determined under light conditions. NA was generally higher in species lacking thick slime sheaths. The highest value (1446 nmol/h C2H4 per g fresh biomass) was found in the strain of branched cyanobacterium Hassalia (A Has1) from the polar region. A quadratic relationship between NA and biomass was detected in the Tolypothrix group under light conditions. The decline of NA in dark relative to light conditions ranged from 37 to 100 % and differed among strains from distinct geographical areas. Unlike the NA of temperate and tropical strains, whose decline in dark relative to light was 24 and 17 %, respectively, the NA of polar strains declined to 1 % in the dark. This difference was explained by adaptation to different light conditions in temperate, tropical, and polar habitats. NA was not related to the frequency of heterocysts in strains of the colony-forming cyanobacterium Nostoc. Colony morphology and life cycle are therefore more important for NA then heterocyst frequency. NA values probably reflect the environmental conditions where the cyanobacterium was isolated and the physiological and morphological state of the strain.  相似文献

Summary A two-year field study was undertaken using15N isotope techniques to differentiate between stimulation of N uptake and N2 fixation in Western Canadian cultivars of spring wheat (Triticum aestivum L. emend Thell) and durum (T. turgidum L. emend Bowden) in response to inoculation with N2-fixing bacteria. Bacterial inoculation either had no effect or lowered the % N derived from the fertilizer and the fertilizer use efficiency. Despite the depression of fertilizer uptake, inoculants did not alter the relative uptake from soil and fertilizer-N pools indicating that bacterial inoculation did not alter rooting patterns. Nitrogen-15 isotope dilution indicated that N2 fixation did occur. In 1984, % plant N derived from the atmosphere (% Ndfa) due to inoculation with Bacillus C-11-25 averaged 23.9% while that withAzospirillum brasilense ATCC 29729 (Cd) averaged 15.5%. In 1985, higher soil N levels reduced these values by approximately one-half. Cultivar x inoculant interactions, while significant, were not consistent across years. However, these interactions did not affect cultivars ‘Cadet’ and ‘Rescue’. In agreement with previous results, ‘Cadet’ performed well with all inoculants in both years while ‘Rescue’ performed poorly. Among 1984 treatments, the N increament in inoculated plants was positively correlated with % Ndfa but no such correlation existed in 1985. N2 fixation averaged over all cultivars and strains was 17.9 and 6.7 kg N fixed ha−1 in 1984 and 1985, respectively. Highest rates of N2 fixation were estimated at 52.4 kg N ha−1 for ‘Cadet’ in 1984 and 31.3 kg N ha−1 for ‘Owens’ in 1985, both inoculated with Bacillus C-11-25, an isolate from southern Alberta soils. Inoculation with either ofAzospirillum brasilense strain Cd (ATCC29729) or 245 did not result in as consistent or as high N2 fixation, suggesting that these wheats had not evolved genetic compatability with this exogenous microorganism. These agronomically significant amounts of N2 fixation occurred under optimally controlled experimental conditions in the field. It is yet to be determined if N2 fixation would occur in response to bacterial inoculation under dryland conditions commonly occurring in Western Canada. Summary Two different strains, An 1 and An 2, were obtained from root nodules ofAlnus nitida Endl., collected from one locality in the area of its natural habitat near Bahrin, District Swat, Pakistan. The light and electron microscopy of the isolates revealed the occurrence of septate and branched hyphae bearing sporangia and vesicles. The strains differed in their growth requirements, nitrogen-fixing ability and production of extracellular pigments, thus indicating the existence of more than oneFrankia strain in the same locality. In the absence of combined nitrogen in the medium strain An 1 formed vesicles and fixed N2 (up to 200 nmol C2H4. mg protein–1.h–1), while strain An 2 under the experimental conditions formed only few vesicles and fixed N2 at a very low rate (ca 10 nmol C2H4. mg protein–1 .h–1). The nitrogenase activity of strain An 1 was strongly affected by the O2 concentration.Frankia An 1 and An 2 were infective and effective onA. nitida andA. glutinosa but not onDatisca cannabina andElaeagnus umbellata. An investigation was conducted to study the effect of biofertilisers (Glomus fasciculatum, Azospirillum brasilense, Azotobacter chroococcum and Micro Phos) and pesticides (carbofuran and bavistin) on the management of root-rot and root-knot disease complex of balsam. The individual application of biofertilisers (G. fasciculatum, A. brasilense and A. chroococcum) significantly improved the plant growth parameters viz., length, dry weight and number of flowers compared to untreated-uninoculated plants. The simultaneous inoculation of plant with Meloidogyne javanica and Macrophomina phaseolina in the pots treated with either of the biofertilisers (G. fasciculatum, A. brasilense and A. chroococcum) and pesticides (carbofuran/bavistin) significantly improved the plant growth parameters and reduced the reproduction factor, number of galls and intensity of root-rot compared to untreated-inoculated plants. On the other hand, the plants treated with Micro Phos neither significantly improved the plant growth parameters nor reduced the reproduction factor, number of galls and intensity of root-rot. Among the biofertilisers and pesticides, carbofuran was found to be the most effective in the management of disease complex of balsam followed by bavistin, G. fasciculatum, A. brasilense and A. chroococcum.  相似文献

Summary Under controlled conditions in pots filled with sand, vermiculite and field soil, inoculation withA. brasilense-Cd ATCC 29729 or withAzotobacter chroococcum caused increases above controls in the weight and N content of panicles ofSetaria italica. In no case, however, did N increases in test plants exceed the initial total N content. High acetylene reduction activities (1,000–2,000 nmole/h/pot) could be found only in plants inoculated withAzospirillum. Inoculation withAzospirillum (strain-Cd) in the field caused a significant increase above noninoculated controls of 18.5% in shoot dry weight, ofSetaria italica. Azotobacter caused a non significant increase of 8%. No significant differences were found between yields ofSetaria italica grown in soil inoculated withA. chroococcum, and those of plants grown in the presence ofA. vinelandii. A. brasilense-Cd was more effective in the field thanA. brasilense Sp-7 ATCC 29145. Out of ten isolates ofAzospirillum spp. isolated from the root and rhizosphere of wheat plant, seven belonged toA. brasilense and three belonged toA. lipoferum. All eight isolates ofAzotobacter spp. belonged toAzotobacter chroococcum. Two strains, one fromA. lipoferum and another fromA. chroococcum having high nitrogen fixing capacity with negative test for denitrification were used as inoculant to supplement the nitrogen need of wheat crop. Significant increases in the yield of wheat grain and uptake of nitrogen by the crop over the control were found in pot tests when the seeds were inoculated either withAzospirillum spp. orAzotobacter spp. or the combination of both the inoculants.  相似文献

Summary Two experiments were performed to examine the effects of inoculation of field grown wheat with various Azospirillum strains. In the first experiment the soil was sterilized with methyl bromide to reduce the Azospirillum population and15N labelled fertilizer was added to all treatments. Two strains ofAzospirillum brasilense isolated from surface sterilized wheat roots and theA. brasilense type strain Sp7 all produced similar increases in grain yield and N content. From the15N and acetylene reduction data it was apparent that these increases were not due to N2 fixation. In the second experiment performed in the same (unsterilized) soil, twoA. brasilense strains (Sp245, Sp246) and oneA. amazonense strain (Am YTr), all isolated from wheat roots, produced responses of dry matter and N content while the response to the strain Sp7 was much smaller. These data confirm earlier results which indicate that if natural Azospirillum populations in the soil are high (the normal situation under Brazilian conditions), strains which are isolated from wheat roots are better able to produce inoculation responses than strains isolated from other sources. The inoculation of a nitrate reductase negative mutant of the strain Sp245 produced only a very small inoculation response in wheat. This suggests that the much greater inoculation response of the original strain was not due to N2 fixation but to an increased nitrate assimilation due to the nitrate reductase activity of the bacteria in the roots. Two phosphate-solubilizing analogue-resistant mutants ofAzotobacter chroococcum showed a positive interactive effect on sunflower (Helianthus annuus). Plant height, leaf area, seed mass, seed yield, protein and oil content increased significantly by inoculation withA. chroococcum mutants. Both levels of fertilizers applied (80N+30P2O5 and 80N+60P2O5 kg/hm2 showed increased seed yield. Likewise, inoculation resulted in a significantly higher yield. Application ofP-solubilizingA. chroococcum strains with the lower fertilizer level showed no significant difference on plant growth parameters when compared with the higher fertilizer level, indicating that inoculation of P-solubilizingAzotobacter strains can reduce the necessary levels of fertilization from 60P2O5 to 30P2O5 kg/hm2.  相似文献

Root colonization studies, employing immunofluorescence and using locally isolated strains, showed thatEnterbacter sp. QH7 andEnterobacter agglomerans AX12 attached more readily to the roots of most plants compared withAzospirillum brasilense JM82. Heat treatment of either root or inoculum significantly decreased the adsorption of bacteria to the root surface. Kallar grass and rice root exudates sustained the growth ofA. brasilense JM82,Enterobacter sp. QH7 andE. agglomerans AX12 in Hoagland and Fahraeus medium. All the strains colonized kallar grass and rice roots in an axenic culture system. However, in studies involving mixed cultures,A. brasilense JM82 was inhibited byEnterobacter sp. QH7 in kallar grass rhizosphere and the simultaneous presence ofEnterobacter sp. QH7 andE. agglomerans AX12 suppressed the growth ofA. brasilense JM82 in rice rhizosphere. The bacterial colonization pattern changed from dispersed to aggregated within 3 days of inoculation. The colonization sites corresponded mainly to the areas where root mucigel was present. The mutant strain Azospirillum brasilenseSp7.2.3 with impaired lectin activity exhibited poorer cell aggregation than its parent strain A. brasilenseSp7(S) both in the exponential and stationary growth phases. The pretreatment of bacterial cells with the specific haptens (L-fucose and D-galactose) of a lectin located at the cell surface of the mutant strain was found to inhibit the aggregation of azospirilla. The specific binding of the A. brasilenseSp7(S) lectin to the extracellular polysaccharide-containing complexes of this strain was revealed by dot immunoblotting on nitrocellulose membrane filters. The interaction of the lectins of A. brasilense75, A. brasilenseSp7, and A. lipoferum59b with the polysaccharide-containing complexes that were isolated from these strains was not specific. No interstrain cross-interaction between the exopolysaccharides and lectins of azospirilla was found. A coflocculation of A. brasilenseSp7 cells with Bacillus polymyxa1460 cells was shown. The involvement of autogenous lectins in the aggregation of bacterial cells is discussed.  相似文献

The nitrogen-fixing grey alder,Alnus incana (L.) Moench, has a potential use in forest soil restoration and as part of energy forestry plantations. As a first step to estimate nitrogen fixation byA. incana under field conditions we performed studies on nitrogenase activity and its possible relation to abiotic factors and growth of the alders. Nitrogenase activity was measured as acetylene reduction activity (ARA) on eleven 1-year-old seedlings ofA. incana inoculated with a local source ofFrankia and planted in an experimental plot located in Umeå, northern Sweden. Each alder was planted into an open-ended cylinder which was closed with a gas tight lid around the stem base to serve as cuvette during ARA measurements. Propane served as tracer gas. ARA was measured in the middle of the day at 15 occasions during 26 June to 29 September 1987. Growth was recorded as leaf area and top shoot length at each ARA measurement until the end of August. Weather conditions were recorded for the whole growing season.Maximal ARA was recorded in late July or early August and ranged from 1.86 to 106mol C2H4plant–1h–1. Final leaf area ranged from 0.022 to 0.124 m2. A relationship between ARA and the number of hours of sunshine during the same day was observed. ARA in relation to soil temperature increased during the study period, except for the last measurements. ARA in relation to leaf area was initially high but decreased later on. It is suggested that as leaves got older their contribution to photosynthesis per unit leaf area decreased and their potential to deliver nitrogen for retranslocation within plant increased. Both of these events would cause reduced ARA per unit leaf area. The data on ARA, growth, and abiotic factors taken together supported the view that sunshine and weather conditions affected photosynthesis and thereby delivery of assimilates to the nodules.  相似文献

Summary Cultivars ofPhaseolus vulgaris (L.) from contrasting geographic locations were cultivated under fields conditions for measurements of nitrogenase and nitrate reductase activities. A first trial with two cultivars indicated that a tropical cultivar B-789 has a higher nitrogenase activity than a temperate one Elsa. And the converse was true for the nitrate reductase activity. While where a post flowering application was made, a renewal of nitrate reductase activity occurred. Further similar comparisons of both enzymatic activities upon eight tropical and temperate cultivars of equivalent vegetative cycles indicated, on the average, that tropical cultivars have a higher level of (C2H2) reduction and a lower nitrate reductase activity than temperature cultivars. These observations suggest that there exists an inverse relationship between the two enzymatic activities in common beans, and there probably exists genetic variability for a possible improvement of N-fixation ability. An early application of N-fertiliser upon the Elsa and B-789 plots promotes later nitrogenase activity while a post flowering application shows obvious a renewal of nitrate reductase activity. Thus, analyses of nitrate reductase and nitrogenase activities of a common bean crop at different physiological stages may give us an indication of the best time to apply supplementary nitrogen fertilisation to common beans to increase seed yield.  相似文献

Indigenous strains isolated from rhizosphere may contain highly competent genotypes to enhance the plant growth and often perform better than the introduced isolates. The present study deals with the characterisation of plant growth-promoting (PGP) attributes and antagonistic activity of Azotobacter chroococcum AZO2 against Macrophomina phaseolina causing charcoal rot disease and their effect on the growth of sesame (Sesamum indicum L.). Eight strains of Azotobacter were isolated from sesame rhizosphere on nitrogen-free medium, which exhibited significant PGP parameters such as phosphate solubilisation, indole acetic acid and siderophore production. The strain A. chroococcum AZO2 (EU274299) was characterised by 16S rDNA gene sequencing. Amplification of 781 bp nif H gene confirms nitrogenase activity of all the strains. A. chroococcum AZO2 exhibited strong antagonistic activities against M. phaseolina causing 81% colony growth inhibition and resulted in hyphal perforations, empty cell (halo) formation, hyphal twisting, shrinking and lysis of fungal mycelia along with degeneration of sclerotia. A. chroococcum AZO2 produced chitinase that caused degradation and digestion of the cell wall component of M. phaseolina. Different vegetative and reproductive parameters of sesame were found to be enhanced significantly upon application of A. chroococcum AZO2 + half doses of chemical fertilisers. A. chroococcum AZO2 was also found to be an effective root coloniser, plant growth promoter and potential antagonistic bacterium. It can be concluded that A. chroococcum AZO2 strain bears the characteristics of technological applications for inoculant preparation and growth enhancement of sesame besides being utilised as a better PGP bacterium as well as an effective agent for biocontrol of M. phaseolina.  相似文献

Summary The growth of threeAzotobacter chroococcum strains was investigated in a number of media. The cultures were incubated at 29°C. and studied by means of a Wild phase contrast microscope. The experiments have clearly demonstrated that the morphology and reproduction ofA. chroococcum are influenced greatly on the one hand by conditions of nutrition and on the other hand also by the properties of the strains. Although all strains used in this work were able to produce gonidia and regenerative rods, these were not found in all media. The formation of filamentous cells, and vegetative cells inside such cells, depended also on the medium and, in some cases, on the strain. Cysts could sometimes resist a temperature of 50°C. for 5.5 minutes, but not a single strain with cysts survived temperatures of 60°C. or higher. Spores could not be detected by the usual staining methods.  相似文献

Aquaspirillum magnetotacticum strain MS-1 and two nonmagnetic mutants derived from it reduced C2H2 microaerobically but not anaerobically even with NO3 ?. This organism apparently is not capable of NO3 ?-dependent nitrogen fixation. Cells ofA. magnetotacticum reduced C2H2 at rates comparable to those ofAzospirillum lipoferum grown under similar conditions, but much lower than that ofAzotobacter vinelandii grown aerobically. Cells ofA. magnetotacticum in anaerobic cultures lacking NO3 ? did not reduce C2H2 until O2 was introduced. Optimum rates of C2H2 reduction byA. magnetotacticum were obtained at 200 Pa O2. C2H2 reduction was inhibited by more than 1 kPa O2 or 0.2 mM NO3 ? or NH4 +. These results suggest thatA. magnetotacticum fixes N2 only under microaerobic, N-limited conditions.  相似文献

An alternative protocol was developed for in vitro propagation of photinia (Photinia × fraseri Dress), an ornamental shrub, using the plant growth-promoting rhizobacteria (PGPR) Azospirillum brasilense and Azotobacter chroococcum during rhizogenesis. Shoot tips from four-year-old mature plants, cut in spring and summer, were used as initial explants. They were cultured on Murashige–Skoog (MS) medium with Gamborg’s vitamins, N6-benzyladenine (BA: 11.1 μM) and gibberellic acid (GA3: 1.3 μM), obtaining 63% of established explants. The highest shoot length (22.9 mm) and multiplication rate (4.3) was achieved by cultivating for four weeks in the same basal medium supplemented with 4.4 μM BA. Both auxin induction and bacterial inoculation were used for rooting. Elongated shoots were treated with two concentrations of indole-3-butyric acid (IBA: 4.9 or 49.2 μM) during 6 days for auxin induction. Then, the shoots were transferred to an auxin-free medium and inoculated with A. brasilense Cd, Sp7 or A. chroococcum (local strain). Bacterial inoculation induced earlier rooting of photinia shoots. A. brasilense Cd with 49.2 μM IBA pulse showed a significant increase (P ≤ 0.05) in root fresh and dry weight (105%, 137%), root surface area (65%) and shoot fresh and dry weight (32%, 62%). A. brasilense Sp7 enhanced the root fresh weight (34%) and root surface area (41%) while no significant differences with A. chroococcum inoculation were detected. The PGPR inoculated micro-cuttings in combination with auxin induction pulses may play a useful role in root organogenesis of micropropagated plants.  相似文献
