为了进一步阐明SPD对大鼠纹状体突触后D1受体的激动作用特性,本文应用反磷酸化在体内测定及放射配体结合方法,分别观察SPD对6-OHDA损毁大鼠纹状体DARPP-32体内磷酸化作用及突触后D1受体密度的影响.结果表明:皮下给予SPD(20,40 mg/kg,21 d),损毁侧纹状体DARPP-32体外[32P]的掺入量较健侧下降50%(P<0.01).换言之,损毁侧纹状体内DARPP-32的磷酸化程度增加了.然而,SPD使损毁导致D1受体上调的作用减弱(Bmax 从385.0±26.1 fmol/mg 降至319.7±20.1 fmol/mg水平).因此,SPD激动D1受体,使6-OHDA损毁大鼠纹状体内DARPP-32磷酸化作用加强,而受体密度减少.这是SPD调节脑内D1受体信号转导功能的重要机制.  相似文献

本研究通过GDP体外处理大鼠脑组织块,观察GDP对脑线粒体脱耦联蛋白(uncoupling proteins,UCPs)活性、UCP4和UCP5表达的影响,以探讨嘌呤核苷酸对大鼠脑UCPs的调节作用.取Sprague-Dawley大鼠双侧大脑半球,将脑组织切成约8-10 mm3的脑组织块,与含1 mmol/L GDP的孵育介质共孵育30 min后,匀浆并差速离心分离提取大鼠脑组织线粒体,采用[3H]-GTP结合法测定UCPs活性,并以Scatehard作图法计算两者结合的解离常数(Kd)和最大结合量(Bmax);RT-PCR和Western blot分别检测UCP4和UCP5的mRNA和蛋白表达.结果显示,1 mmol/L GDP可降低体外大鼠脑组织线粒体中UCPs与[3H]-GTP结合的Bmax,提高Kd,但对脑纰织中UCP4和UCP5 mRNA和蛋白表达量的改变无统计学意义.上述结果提示,GDP可直接抑制体外大鼠脑组织中UCPs的活性,但并不影响UCP4和UCP5的表达.  相似文献

用放射配基结合分析法,研究了不同温度条件下蟾蜍心脏各部位肾上腺素能受体的类型和含量。结果表明,常温蟾蜍心肌膜在37℃同β-受体的特异性标记配基~3H-双氢心得舒(~3H-DHA)的最大结合(B_(max))及Kd值分别为:全心,55.11±6.22fmol/mg蛋白及2.15±0.42nmol/L;窦房,55.80±7.03fmol/mg蛋白及2.65±0.37 nmol/L;心室,54.27±3.06 fmol/mg蛋白及1.84±0.14 nmol/L,同样温度条件下,窦房及心室心肌膜同α-受体的特异性标记配基~3H-双氢麦角隐亭(~3H-DHE)没有特异性结合,经过5—8℃,10d以上低温服习的蟾蜍,在10℃测试,其α-,β-受体的结合量和亲和力无异于常温蟾蜍。上述结果提示,蟾蜍心肌只有β-受体,温度对受体的类型和含量无明显影响,这种受体类型和含量在相当大的温度范围内保持不变。  相似文献

在测定蚕豆(Vicia faba L.)保卫细胞原生质体质膜ABA结合蛋白的解离常数(Kd)和最大结合容量的基础上,进一步研究了几种因子(pH、光)对Kd和最大结合容量的影响。结果显示:pH并不改变结合蛋白的Kd值,而仅影响每个原生质体结合的分子数;光、暗处理的结果表明,光可使结合蛋白的Kd值增大,每个原生质体结合的ABA分子数减少,而暗处理可使结合蛋白的Kd值减小及每个原生质体结合的分子数增多,说明黑暗可提高气孔保卫细胞对ABA的敏感性,而光可以降低气孔保卫细胞对ABA的敏感性。因此,光、暗可以通过调节ABA结合蛋白的Kd值来调节气孔对ABA的敏感性。  相似文献

本实验使用荧光指示剂Fura-2与Tb~(3+),检测了不同年龄组大鼠的不同脑区(海马、皮层、间脑、小脑)突触体内游离钙与膜结合钙水平。结果显示,与青年对照组相比,老年大鼠大部分脑区(海马、皮层、间脑)突触体内游离钙水平显著增高,尤其是海马突触体内游离钙增高极为显著;其突触体膜结合钙水平表现为:海马、小脑两脑区明显升高,而皮层、间脑两脑区明显下降,呈现一种全脑范围内的钙水平失衡。提示动物的衰老与其脑内钙自体平衡失调有关。  相似文献

本文采用腺苷亲和层析法从大鼠脂肪细胞膜上分离出了一种亚基分子量为38kD的腺苷结合蛋白质。此蛋白在SDS-聚丙烯酰胺凝胶电泳上显示单一带,糖蛋白染色阳性;能与[8-~3H]腺苷特异结合(Kd=0.269nmol/L,Bmax=6.05pmol/mg.Pr);结合抑制实验表明它与腺苷A_1受体激动剂R-PIA、A_2受体激动剂NECA和腺苷的亲和力大小顺序为:R-PIA>腺苷>NECA。这表明所分离出的38kD蛋白是大鼠脂肪细胞膜上的腺苷A_1受体。  相似文献

为探讨烫伤引起病理性应激时大鼠肾胞液醛固酮结合活性的变化及可能的调节机制,以[~3H]-醛固酮为配体,用放射性配基-受体结合分析法测定了正常对照组、轻度烫伤组和重度烫伤组大鼠肾胞液醛固酮结合活性的结合容量(Rt)和表观解离常数(Kd);采用体内注射TNF-α、IL-1β中和抗体和α-促黑色素细胞刺激激素(α-melanocyte-stimulatinghormone,α-MSH)和合成肽KPV(Ac-D-Lys-L-Pro-D-Val)等措施调节其改变.结果发现,肾胞液存在两种不同结合容量、不同亲和力的醛固酮结合活性受体.与正常对照组的Rt(Rt141.6±7.2fmol/mgpro;Rt2317.6±70.0fmol/mgpro)相比,轻烫组的Rt(Rt141.4±5.0fmol/mgpro;Rt2314.8±45.7fmol/mgpro)无显著差异(P>0.05;P>0.05);重烫组的Rt(Rt122.4±5.4fmol/mgpro;Rt2196.3±32.5fmol/mgpro)则显著下降(P<0.01;P<0.01).体内注射TNF-α与IL-1β中和抗体、α-MSH及KPV均能明显提高重烫组Rt值.结果提示,重度烫伤大鼠肾胞浆醛固酮结合活性降低,TNF-α、IL-1β中和抗体、α-MSH及KPV均可防止重度烫伤引起病理性应激时醛固酮结合活性降低.  相似文献

胰岛素能影响卵巢功能,例如,离体颗粒细胞持续生长需要胰岛素,胰岛素也促进LH诱导的黄体细胞生成孕酮等。除猪颗粒细胞的资料外,人类卵巢组织也有胰岛素受体。Jarrett等在剖腹手术时取得人类卵巢组织样品,作质膜受体测定。质膜与猪[~(125)Ⅰ]胰岛素共同保温,Scatchard分析表明存在双相曲线。高亲和力结合位平均浓度为57.4±7.9(±SEM)fmol/mg蛋白质,解离常数为3.5±0.9nM。在月经周期中结合亲和力和结合位点数目皆无明显变化。上皮生长因子或高血糖素都不影响胰岛素与卵巢质膜的结合。因此,人类卵巢质膜存在高亲和力、特异性和可饱和的胰岛素结合位。胰岛素可能直接参与卵巢功能。  相似文献

目的 :观察大鼠心肌浆网 (sarcoplasmicreticulum ,SR)和核被膜 (nuclearenvelope ,NE)ryanodine受体 (RyR)与配体结合特点及其蛋白质磷酸化调节。方法 :采用差速和等密度梯度离心分离心肌SR和NE ,用放射受体分析法研究RyR的特征。结果 :NE上存在高亲和力RyR ,其最大结合 (Bmax)为SRRyR的 1.7% ,解离常数 (Kd)为SR的6 0 %。分别用PKA和PKC磷酸化后 ,SR上该受体的Bmax各增加 3.7和 1.2倍 ,而NE上的该受体Bmax各增加 2 .2和 3.1倍 ,Kd均无显著改变。结论 :NE上存在比SR密度低但亲和力高的RyR ,能被PKA和PKC激活 ,而且对PKC较PKA更敏感  相似文献

We present, herein, the evidence for lactoferrin (Lf) binding sites in brain endothelial capillary cells (BCECs) and mouse brain. The results from confocal microscopy showed the presence of Lf receptors on the surface of BCECs and the receptor-mediated endocytosis for Lf to enter the cells. Saturation binding analyses revealed that Lf receptors exhibited two classes of binding sites in BCECs (high affinity: dissociation constant (K (d)) = 6.77 nM, binding site density (B (max)) = 10.3 fmol bound/mug protein; low affinity: K (d) = 4815 nM, B (max) = 1190 fmol bound/mug protein) and membrane preparations of mouse brain (high affinity: K (d) = 10.61 nM, B (max) = 410 fmol bound/mug protein; low affinity: K (d) = 2228 nM, B (max) = 51641 fmol bound/mug protein). The distribution study indicated the effective uptake of (125)I-Lf in brain after intravenous administration. The present study provides experimental evidence for the application of Lf as a novel ligand for brain targeting.  相似文献

We localized melatonin binding sites in different brain regions (optic tectum, telencephalon, cerebellum, hypothalamus, olfactory bulbs, and medulla oblongata) of Senegal sole, a species of aquaculture interest, and checked day/night changes in density (B(max)) at mid-light (ZT06) and mid-dark (ZT18). Plasma melatonin was measured using a radioimmunoassay, while binding assays were performed using 2-[(125)I]iodomelatonin as a radioligand. Plasma melatonin concentrations were significantly lower at mid-light (189.5+/-46 pg/ml) than mid-dark (455.5+/-163 pg/ml). Values of B(max) were statistically significantly higher in the optic tectum (5.6+/-0.6 and 12.3+/-1 fmol/mg prot, at mid-light and mid-dark, respectively) and in the cerebellum (7.7+/-1.1 and 10.6+/-1.3 fmol/mg prot, at mid-light and mid-dark, respectively). Significant day/night differences were only observed in these two tissues. These results show for the first time the distribution of melatonin binding sites within the brain of a flatfish species and their lack of down-regulation.  相似文献

The distribution of high-affinity binding sites for gamma-[3H]hydroxybutyrate in coronal sections of rat brain was studied by quantitative autoradiographic techniques. Binding sites for this naturally occurring substance, which may possibly have a neurotransmitter role, are concentrated in some restricted areas of the brain, particularly in the limbic system. The hippocampus (especially field CA1 of Ammon's horn, at 292 fmol/mg of tissue), septum (72 fmol/mg of tissue), and cortex (frontal, 113 fmol/mg of tissue; parietal, 103 fmol/mg of tissue; cingulate, 114 fmol/mg of tissue; and entorhinal, 134 fmol/mg of tissue) show pronounced labeling with gamma-[3H]hydroxybutyrate. Binding is much lower in caudatus-putamen (50 fmol/mg of tissue), thalamus, and hypothalamus. Caudal parts of the brain (cerebellum, pons, and medulla) are practically devoid of binding sites. These results strongly support a functional role of endogenous gamma-hydroxybutyrate in particularly restricted areas of the rat brain.  相似文献

The in vivo effect of trazodone on the density of [(3)H]5-HT binding sites and 5-HT(1A) receptors and adenylyl cyclase (AC) response was studied in regions of rat brain. The chronic administration of trazodone (10 mg/Kg body wt, 40 days) resulted in a significant downregulation of [(3)H]5-HT binding sites and 5-HT(1A) receptors in cortex and hippocampus. Trazodone significantly (p < 0.0001) decreased the density of [(3)H]5-HT binding sites in cortex (42.6 +/- 3.6 fmol/mg protein, 65%) and hippocampus (12.6 +/- 1.6 fmol/mg protein, 87%) when compared to control values of 121.9 +/- 5.4 and 99.3 +/- 7.5 fmol/mg protein in these regions, respectively. Similarly there was a significant (p < 0.0001) decrease in the density of 5-HT(1A) receptors in both cortex (7.2 +/- 0.5 fmol/mg protein, 70%) and hippocampus (6.3 +/- 1.2 fmol/mg protein, 79%) when compared to control values of 24.2 +/- 2.1 and 30.6 +/- 3.7 fmol/mg protein, in these regions respectively. However, the affinity of [(3)H]5-HT to 5-HT binding sites (1.83 +/- 0.26 nM, p < 0.0001) and [(3)H]8-OH-DPAT to 5-HT(1A) receptors (0.60 +/- 0.06 nM, p < 0.05) was significantly decreased only in cortex when compared to the control K(d) values of 0.88 +/- 0.04 nM and 0.47 +/- 0.02 nM in these regions, respectively.The basal AC activity did not alter in treated rats, where as, the inhibition of forskolin-stimulated AC activity by 5-HT (10 microM) was significantly (p < 0.0001) decreased both in cortex (43%) and hippocampus (40%) when compared to control levels. In conclusion, chronic treatment with trazodone results in downregulation of 5-HT(1A) receptors in cortex and hippocampus along with concomitant increased AC response, suggesting the involvement of 5-HT(1A) receptor-mediated AC response in the mechanism of action of trazodone.  相似文献

[3H]Fluphenazine was used to label both D-1 and D-2 dopamine receptors in mouse striatal membranes. The D-1 and D-2 specific binding of [3H]fluphenazine was discriminated by the dopamine antagonists SCH-23390 (D-1 selective) and spiperone (D-2 selective). Saturation analyses of these two sites yielded a D-1 receptor density in mouse striatum of 1,400 fmol/mg of protein and a D-2 receptor density of 700 fmol/mg of protein. The affinity of [3H]fluphenazine for the D-2 site was slightly greater than for the D-1 site; the equilibrium dissociation constant (KD) was 0.7 versus 3.2 nM, respectively. Assay conditions are described that reduce nonspecific binding of [3H]fluphenazine to acceptable levels (35% of total binding at 1 nM [3H]fluphenazine). By comparison of displacement curves from a series of dopaminergic and nondopaminergic ligands, the pharmacological specificity of [3H]fluphenazine binding in mouse striatum was demonstrated to be dopaminergic. Only small amounts of dopamine-specific (apomorphine-sensitive) [3H]fluphenazine binding were found in other brain regions. However, chlorpromazine displaced considerable [3H]fluphenazine from all brain regions, including cerebellum, suggesting the presence of a [3H]fluphenazine binding site with a phenothiazine specificity.  相似文献

Abstract— [125I]Diiodo α-bungarotoxin ([125I]2BuTx) and [3H]quinuclidinylbenzilate ([3H]QNB) binding sites were measured in post-nuclear membrane fractions prepared from whole brains or brain regions of several species. Species studied included Drosophila melanogaster (fruit fly), Torpedo californiea (electric ray), Carassius auratus (goldfish), Ram pipiens (grass frog), Kana cutesheiana (bullfrog), Rattus norvegicus (rat, Sprague-Dawley), Mus muscalus (mouse, Swiss random, C58/J, LG/J), Oryctolagus cuniculus (rabbit, New Zealand Whitc), and Bos (cow). Acetyl-CoA: choline O -acetyltransferase (EC 2.3.1.6) levels were also determined in the post nuclear supernatants and correlated with the number of binding sites.
All species and regions except Drosophila had 16–150 fold more [3H]QNB binding sites than [125I]2BuTx binding sites. Brain regions with the highest levels of [125I]2BuTx binding were Drosophila heads (300 fmol/mg), goldfish optic tectum (80fmol/mg), and rat and mouse hippocampus (3040 fmol/mg). The highest levels of [3H]QNB binding were seen in rat and mouse caudate (1.3–1.6 pmol/mg). Lowest levels of [3H]QNB and [125I]2BuTx binding were seen in cerebellum. The utility of [125I]2BuTx and [3H]QNB binding as quantitative measures of nicotinic and muscarinic acetylcholine receptors in CNS is discussed.  相似文献

Based on non-competitive binding interactions we suggested that mu and delta receptors associate as a mu/delta receptor complex in rat brain. We hypothesized that the same non-competitive binding interactions observed in rat brain will be seen in CHO cells that co-express mu and delta receptors, but not in cells that express just mu or delta receptors. We used CHO cells expressing the cloned human mu receptor, cloned human delta receptor, or cloned mouse delta/human mu ("dimer cell"). Cell membranes were prepared from intact cells pretreated with 100nM SUPERFIT. [(3)H][d-Ala(2),d-Leu(5)]enkephalin binding assays followed published procedures. SUPERFIT, a delta-selective irreversible ligand, decreased [(3)H][d-Ala(2),d-Leu(5)]enkephalin binding to delta receptors by approximately 75% and to mu receptors by approximately 50% in dimer cells. SUPERFIT treatment did not decrease [(3)H][d-Ala(2),d-Leu(5)]enkephalin binding to mu cells. The IC(50) values observed in SUPERFIT-treated dimer cells were: [d-Pen(2),d-Pen(5)]enkephalin (1820nM) and morphine (171nM). Saturation binding experiments with SUPERFIT-treated dimer cells showed that [d-Pen(2),d-Pen(5)]enkephalin (5000nM) was a competitive inhibitor. In contrast, morphine (1000nM) lowered the B(max) from 1944fmol/mg to 1276fmol/mg protein (35% decrease). Both [d-Pen(2),d-Pen(5)]enkephalin and morphine competitively inhibited [(3)H][d-Ala(2),d-Leu(5)]enkephalin binding to SUPERFIT-treated mu cells. The results indicate that the mu-delta opioid receptor complex defined on the basis of non-competitive binding interactions in rat brain over 20 years ago likely occurs as a consequence of the formation of mu-delta heterodimers. SUPERFIT-treated dimer cells may provide a useful model to study the properties of mu-delta heterodimers.  相似文献

Competition between cold phenytoin and [3H]phenytoin binding was observed in normal human brain. Binding was observed in all areas examined. The highest number of sites was in the amygdala (a total of 717.71 fmol/mg protein) and the lowest in the Brodman area (BA) 4 of the motor cortex (153.91 fmol/mg protein) and cerebellar cortex (154.4 fmol/mg protein). In three areas, amygdala, cortex area BA 38 (inferior parietal lobe), and cortex area BA 8 (premotor cortex), two sets of binding sites were observed. In these areas the Kd for the higher affinity sites ranged from 35 to 116 nM, and for the lower affinity site, from 328 to 866 nM. In the four areas where only one binding site was observed the KdS ranged from 164 to 311 nM and the Scatchard plot was linear.  相似文献

[3H]5-hydroxytryptamine ([3H]serotonin) binds with high affinity (KD 2-12 nM) to a finite number of sites on brain astroglial cells. The number of binding sites in the C6 glioma line is decreased significantly (Bmax = 315 versus 30 fmol/mg) by homogenization. In intact primary cultures, derived from newborn rat brain, the number of binding sites is far greater in cultures of immature astrocytes than in cultures treated with dibutyryl cyclic AMP (Bmax = 1,520 versus 580 fmol/mg). The external plexiform layer of the olfactory bulb is among the brain regions where insulin receptors are most abundant. In vitro binding of porcine 125I-insulin to membranes of dendrodendritic synaptosomes isolated from adult rat olfactory bulbs was studied to test the hypothesis that dendrodendritic synapses are major insulin-receptive sites in the external plexiform layer of olfactory bulbs. Of the specific insulin binding sites present in a total particulate fraction from the olfactory bulbs, approximately half were recovered in the dendrodendritic synaptosome fraction. The only other subcellular fraction to which substantial insulin binding was observed was the conventional (axodendritic/axosomatic) synaptosome fraction. Analysis of equilibrium binding of insulin to dendrodendritic synaptosomal membranes, at total insulin concentrations of 0.5-1,000 nM, revealed binding site heterogeneity consistent with a two-site model for insulin binding to a high-affinity (KD = 6 nM), low-capacity (Bmax = 110 fmol/mg of protein) site and a low-affinity (KD = 190 nM), high-capacity (Bmax = 570 fmol/mg of protein) site. The results indicate that the intense labeling of the external plexiform layer of the olfactory bulb in autoradiographic studies of insulin binding can be attributed to insulin receptors on dendrodendritic synaptic membranes in this region.  相似文献
