将带1-2个芽眼的唐菖蒲球茎切块接种到附加1.0mg/LBA的MS基本培养基上可诱导休眠芽萌动、无菌芽转移至附加3.0mg/LBA的培养基上可分化产生丛生芽。丛生芽的继代增殖宜采用附加1.5mg/LBA的培养基生根培养,以MS+NAA 0.1-0.5mg/L或MS+IBA 1.5mg/L效果最佳。试管苗移栽存活率可达50%左右。  相似文献

本研究以‘正午’牡丹腋芽为外植体,建立了高效的牡丹微繁殖体系。腋芽的初始培养基为WPM+0.5mg/L BA+0.2mg/L GA3,培养50d后,一个丛生芽平均可切分为13个繁殖体用于增殖培养;增殖培养基为WPM[1668mg/L Ca(NO3)2·4H2O]+0.5mg/L BA+0.2mg/L GA3,以35d为一个继代培养周期,增殖率为3.0,共继代培养7次;生根培养时,先将无根苗在复壮培养基[1/2MS(296mg/L CaCl2)+0.5g/L活性炭]上培养20d,再转入根诱导培养基[1/2 MS(296 mg/L CaCl2)+1.0 mg/L腐胺+1.0 mg/L IBA]培养30d,最后转入根形成培养基[1/2 MS(296mg/L CaCl2)+4.0g/L活性炭]培养20d,其生根率达77.2%;驯化与移栽基质为珍珠岩∶蛭石∶草炭土=1∶1∶1,组培苗移栽成活率高达92.1%。这表明以‘正午’牡丹腋芽建立的微繁殖体系具备规模化商业生产的价值。  相似文献

以绒毛皂荚(Gleditsia vestita Chun et How ex B.G.Li)种子为外植体,通过愈伤组织诱导、不定芽分化增殖、生根和驯化移栽,建立了绒毛皂荚组织培养的快速繁殖体系。结果表明:1)绒毛皂荚子叶和胚轴在MS+6-BA3.0 mg/L+KT 0.4 mg/L+IAA 0.3 mg/L培养基上同步诱导出愈伤和芽,愈伤组织诱导率达到90.5%,同步分化率高达77.9%;2)不定芽增殖的最佳培养基为1/2MS+6-BA 1.0 mg/L+IBA 0.3 mg/L+NAA 0.2 mg/L,培养20 d后,增殖倍数为4.0;3)MS+NAA 0.5 mg/L+IAA 0.25 mg/L条件下,生根率高达100%,且移栽后植株长势好,成活率达90%以上。  相似文献

为保护野生资源、实现人工栽培,本研究以葶苈(Draba nemorosa)嫩茎为材料,采用组织培养方法进行愈伤组织诱导与分化、不定芽生根与试管苗生根继代增殖培养,以及移栽和定植研究。结果表明,MS+6-BA 0.4 mg/L+2,4-D 2.5 mg/L是愈伤组织诱导培养和继代增殖培养的理想培养基;MS+6-BA 0.6 mg/L+NAA 0.1 mg/L 是愈伤组织分化培养和不定芽继代增殖培养的理想培养基;1/3MS+IAA 0.6 mg/L是不定芽生根培养和生根继代增殖培养的理想培养基;试管苗移栽成活率为86.8%,定植成活率为96.4%;定植苗保持了野生葶苈的植物学性状。  相似文献

采用黄山药的带芽茎段为外植体,试验了不同激素处理对芽诱导的影响,通过继代培养诱导丛生芽使大量增殖,经诱导生根和炼苗移栽而完成植株的快速繁殖再生。结果表明,MS 6-BA2.0mg/L NAA0.5-1.0mg/L为芽诱导的最适培养基,MS 6-BA1.0mg/L NAA0.5-1.0mg/L为继代增殖的最佳培养基,月增殖系数约为3倍,1/2MS 1BA0.5mg/L培养基诱导生根相对较好,诱导率为50%,组培苗经炼苗后,移栽成活率可达80%。  相似文献

以北海道黄杨的茎段为试材、研究其再生植株在不同外源激素配比下的适宜培养基。试验结果表明,北海道黄杨茎段初代培养的培养基以MS培养基附加BA1.5mg/L,NAA0.5mg/L较好。第一次继代培养的培养基以MS培养基(铁盐加倍)附加BA1.5mg/L,NAA0.5mg/L较为合话,第二次及往后的继代培养的培养基以MS培养基(铁盐加倍)附加BA1.0mg/L、IBA0.2mg/L较适宜,并且,随着继代次数的增加,继代苗的分生率提高,生根培养基为1/2MS(铁盐加倍) NAA0.2mg/L及1/2MS(铁盐加倍)移栽时,以蛭石;腐叶土(1:1)为移栽基质,成活率达100%。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
