Oxygen uptake of growth hormone transgenic coho salmon Oncorhynchus kisutch was measured in individual fish with a closed-system respirometer and was compared with that of similar-sized non-transgenic control coho salmon during starvation and when fed a fixed ration or to satiation. Transgenic and control fish did not differ in their standard oxygen uptake after 4 days of starvation, although control fish had a higher routine oxygen uptake, scope for spontaneous activity and initial acclimation oxygen uptake. During feeding, transgenic fish ate significantly more than control fish, and had an overall oxygen uptake that was 1·7 times greater than control fish. When fish that had eaten the same per cent body mass were compared, transgenic fish had an oxygen uptake that was 1·4 times greater than control fish. Differences in oxygen uptake in growth hormone transgenic coho salmon and non-transgenic fish appear to be due to the effects of feeding, acclimation and activity level, and not to a difference in basal metabolism.  相似文献

Uptake of [35S]sulfate by segments of rat costal cartilage during culture was greatly stimulated when freshly prepared phenylmethylsulfonyl fluoride or diisopropylfluorophosphate was included in the incubation medium. By contrast, hydrolysed diisopropylfluorophosphate, sodium fluoride or soybean trypsin inhibitor did not stimulate [35S]sulfate uptake. Incorporation of four other radioactive precursors of cartilage synthesis was almost completely suppressed during cartilage incubation in the presence of phenylmethylsulfonyl fluoride. However, stimulation of [35S]sulfate binding by the latter was shown to occur at sites other than on glycosaminoglycan molecules and to a similar degree with both active and inactivated cartilage. These and other data indicate that the stimulatory effect of phenylmethylsulfonyl fluoride on [35S]sulfate uptake is independent of normal metabolic processes, and may involve the binding of phenylmethylsulfonyl fluoride to cartilage proteins.  相似文献

Uptake of sulfate by yeast requires the presence of a metabolic substrate and is dependent on the time during which the cells have been metabolizing in the absence of sulfate. At low concentrations of sulfate, uptake can be described by simple saturation kinetics. Uptake of sulfate is accompanied by a net proton influx of 3 H+ and an efflux of 1 K+ for each sulfate ion taken up. Divalent cations stimulate sulfate uptake at low concentrations of sulfate; the maximal rate of uptake is not significantly affected but Km is lowered. Stimulation by divalent cations shows an optimum at a cation concentration of about 4 mM. Monovalent cations are less effective, trivalent cations are more effective in stimulating sulfate uptake. The results are qualitatively in accordance with the notion, that the effect of cations is due to an effect via the surface potential.  相似文献

Summary

Among the many functions of follicle cells in the insect ovary is the regulation of the entrance into the follicle of the vitellogenin circulating in the hemolymph. The vitellogenin enters the follicle via large spaces which appear between the follicle cells. The appearance of these spaces (patency) is a result of a reduction in volume of the follicle cells caused by the action of juvenile hormone which activates a juvenile-hormone-sensitive Na+K+ ATPase via a pathway involving protein kinase C. A putative juvenile hormone receptor protein has been identified in membranes from follicle cells. An antigonadotropin, a small neuropeptide, antagonizes the action of juvenile hormone on the follicle cells.  相似文献

Sexual maturation profoundly affects population dynamics, but the degrees to which genetic, top-down, and bottom-up controls affect age at maturity are unclear. Salmonid fishes have plastic age at maturity, and we consider genetic and environmental effects on this trait by developing fitness functions for coho salmon (Oncorhynchus kisutch). The functions are based on size-specific survival and reproductive success, where reproductive success is the product of fecundity and ability to defend nests (females) or the product of sperm volume and ability to mate (males). We model genetic and bottom-up controls (e.g., food availability) with an environmentally explicit growth function and top-down control (predation mortality) with survival functions that consider both size-dependent and size-independent mortality. For females, we predict that early maturation rarely maximizes fitness, but males can maximize fitness by maturing early if they grow well in freshwater. We predict that early maturation is most affected by the bottom-up effects of resource distribution at sea, followed by bottom-up and genotypic effects in freshwater. Top-down processes are predicted to have strong effects on the likelihood of delayed maturation.  相似文献

The epizootiology and histopathology of the myxosporean Parvicapsula sp. was studied during monthly health surveys of 4 groups of coho salmon Oncorhynchus kisutch at a commercial farm in Puget Sound, Washington, USA, from 1984 to 1986. No Parvicapsula sp. was detected in histological samples taken from juvenile fish in fresh water, but the parasite was detected in fish from all groups 2 to 8 mo after transfer to seawater net pens. Groups placed in seawater net pens in November and December had a higher prevalence of infection, and a longer period of continuous detected infection, than those introduced into net pens in May. For the groups transferred to seawater in November and December, the highest infection prevalence (45 to 90%) was detected during the following March and April. Among 13 tissues examined histologically, only the pseudobranch and kidney were positive for Parvicapsula sp., with 26 (62%) of 42 positive fish showing infections only in the pseudobranch, 5 (12%) showing infections only in the kidney, and 11 (26%) showing infections in both organs. Both the pseudobranch and kidney were apparent primary infection sites, but pseudobranch infections appeared to persist longer in a population. Pseudobranch infections were frequently heavy and associated with extensive inflammation and necrosis of filament and lamellar tissues. The kidney had been the only infection site reported for Parvicapsula sp. in previous studies of coho salmon.  相似文献

Juvenile coho salmon Oncorhynchus kisutch from the Quinsam, Big Qualicum and Puntledge Rivers, British Columbia, Canada, discriminated between the chemical emanations of similarly aged salmon from their own and another population. However, common rearing during the embryonic, larval and early free-swimming stages rendered families more attractive to each other than they were otherwise. Preference among populations was also influenced by quantity of odour produced by the fish. Some fish, notably the Quinsam River population, appeared to produce greater quantities of attractants than other fish. Finally, the salmon also preferred water conditioned by faeces of members of their own population over blank water and, in one of two experiments, over water conditioned by faeces of non-population members, suggesting that at least some of the odourants mediating intraspecific discriminations are present in faeces. These results confirm earlier studies demonstrating that salmonid populations differ in odours distinguishable by conspecifics but indicate that experiments investigating this phenomenon must be designed carefully to avoid bias owing to differences other than population-specific traits. The role of population recognition in the lives of salmonids is unclear; it might be a manifestation of more general imprinting for homing, of sibling recognition, or it might play a role in mate choice.  相似文献

The importance of environmental ions to olfactory sensitivity in fish has not been clarified by electrophysiological experiments. We tested the ability of juvenile coho salmon, Oncorhynchus kisutch (Walbaum), to detect L-serine after acclimation to either fresh water or sea water. The olfactory threshold was identical in the two waters, indicating that the sensitivity of olfaction was not affected by ionic content of the solutions bathing the sensory tissue.  相似文献

The cranial skeleton of the lamprey, a primitive vertebrate, consists of cartilaginous structures that differ from vertebrate cartilages in having a noncollagenous extracellular matrix. Novel matrix proteins found in these cartilages include lamprin in the annular cartilage and an unidentified protein in the branchial cartilages. Both show biochemical similarities to elastin. The inextractability of these proteins, even to chemical cleavage by cyanogen bromide, indicates a polymer with extensive covalent cross-linking. Here we report on the type of cross-linking. Lysyl pyridinoline was found in high concentration in the elastin-like protein of lamprey branchial cartilage at a ratio of 7:1 to hydroxylysyl pyridinoline, the form that dominates in vertebrate collagens. Both forms of pyridinoline cross-link were absent from annular cartilage and desmosine cross-links, which are characteristic of vertebrate elastin, were not detected in either form of lamprey cartilage. Pyridinoline cross-links are considered to be characteristic of collagen, so their presence in an elastin-like protein in a primitive cartilage poses evolutionary questions about the tissue, the protein, and the cross-linking mechanism.  相似文献

1. Yearling coho salmon were fed diets containing the flesh of sexually mature adult coho salmon from Lakes Ontario, Erie and Michigan; Pacific Ocean coho salmon and a commercial trout chow were used to prepare control diets. The fish were fed the experimental diets for 27 weeks. Samples were taken of fed fish and fish fed for 27 weeks and then food deprived for 4 weeks. 2. Total accumulated organochlorine in the recipient was directly proportional to dietary total organochlorine levels, although there were considerable interlake differences in the degree of clearance of specific organochlorine compounds. 3. There was evidence of changes in thyroid physiology, hematocrit, liver structure and inter-renal gland activity in recipients fed the Great Lakes' coho salmon, apparently correlated with organochlorine bioaccumulation.  相似文献

This work was designed to find the cause of the delay in hydrogen sulfide dissimilation in Desulfotomaculum acetoxidans DSM 771, which is dependent on the sulfate uptake. This bacterium grown without addition of any aromatic compound was shown by spectrum analysis with the methylene method to contain hydroxy-benzoate derivatives. The presence of these compounds was confirmed by HPLC in fractions obtained from cell walls after 15 days of culture. The test with 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt seemed to indicate the presence of peroxidase, which probably oxidized benzoate to its hydroxy derivatives. The test with 5-sulfo-salicylic acid proved the ability of the investigated strain to utilize arylsulfates and to reduce sulfate group to hydrogen sulfide. On the basis of the above data, we propose the following sequence of reactions: 1, benzoate secretion; 2, benzoate hydroxylation; 3, sulfonation of hydroxy-benzoate derivatives.  相似文献

The ability of bacteria to cause rapid uptake of choline sulfate in plants, i.e., effectiveness, was studied using Pseudomonas tolaasii and excised roots of barley (Hordeum vulgare L.). Once effective, bacteria remained so after being killed by treatments which cause little damage to their outer structure. However, effectiveness was destroyed by disruption of the cell wall, protein reagents, a mild heat treatment or removal of Mg2+. Effective bacteria adsorbed choline sulfate. This adsorption had characteristics similar to those of bacterial effectiveness (magnesium requirement, high substrate specificity). These results indicate that a proteinaceous structure on the bacterial surface binds and, somehow, transfers choline sulfate to the plant.  相似文献

Synopsis Gastric evacuation of multiple meals of recently emerged sockeye salmon, Oncorhynchus nerka, consumed by juvenile coho salmon, O. kisutch, were compared with that expected from a single meal evacuation rate model developed for the estimation of food consumption by coho in the field. Significant interaction occurred between meals consumed two hours apart (p<0.001). Evacuation of the first of two meals (29.9% of initial weight remaining) was significantly faster (p<0.05) than that of a single meal (36.1% of initial weight remaining) after four hours, whereas evacuation of the second meal (72.2% of initial weight remaining) was significantly slower (p<0.001) than that of single meals (50.6% of initial weight remaining) after two hours. The total weight of the multiple meal remaining after four (0.191 g), six (0.138 g) and eight (0.070 g) hours averaged within 6% of that predicted by the single meal evacuation model (0.186 g, 0.115 g and 0.073 g, respectively). These data suggest that the stomach evacuation model based on single meals is adequate for estimating the evacuation of prey consumed by continuously feeding coho salmon.  相似文献

After chondroitinase digestion of bovine nasal and tracheal cartilage proteoglycans, subsequent treatment with trypsin or trypsin followed by chymotrypsin yielded two major types of polypeptide-glycosaminoglycan fragments which could be separated by Sepharose 6B chromatography. One fragment, located close to the hyaluronic acid-binding region of the protein core, had a high relative keratan sulfate content. This fragment contained about 60% of the total keratan sulfate, but less than 10% of the total chondroitin sulfate present in the original proteoglycan preparation. The weight average molecular weight of the keratan sulfate-enriched fragment was 122,000, as determined by sedimentation equilibrium centrifugation. The chemical and physical data indicate that this fragment contains an average of 10 to 15 keratan sulfate chains, if the average molecular weight of individual chains is assumed to be about 8,000, and about 5 chondroitin sulfate chains attached to a peptide of about 20,000 daltons. The other population of fragments was derived from the other end of the proteoglycan molecule, the chondroitin sulfate-enriched region, and contained mainly chondroitin sulfate chains. About 90% of the total chondroitin sulfate, but only 20 to 30% of the total keratan sulfate was recovered in these fragments. On the average, approximately 5 chondroitin sulfate chains and 1 keratan sulfate chain could be linked to the same peptide. Another 10 to 20% of the total keratan sulfate, originally found in or near the hyaluronic acid-binding region, was not separated from the chondroitin sulfate-enriched fragments. Hydroxylamine could be used to liberate a large molecular size, chondroitin sulfate-enriched fragment (Kav 0.54 on Sepharose 2B) from the proteoglycan aggregates. The remainder of the protein core, containing the keratan sulfate-enriched region, was bound to hyaluronic acid with the link proteins and recovered in the void volume on the Sepharose 2B column.  相似文献

Sulfate complexation of Cd in nutrient solution has been shown to have little impact on Cd uptake by plants. This study examined the effect of sulfate added to soil on Cd concentrations in soil solution and Cd uptake by Swiss chard (Beta vulgaris L. cv. Fordhook Giant). Swiss chard was grown in soil which was wetted with complete nutrient solution containing equivalent salt concentrations of NaNO3 or Na2SO4. Plant growth was reduced by increasing both NO3 and SO4 concentrations in soil solution, with growth reductions similar for both salts. The Cd concentration in soil solution increased P< 0.05) more consistently with increasing concentrations of SO4 compared to NO3 in soil solution. Solution speciation, calculated with GEOCHEM-PC, showed significant increases of Cd2+ activities with increasing salt rates. Shoot Cd content in 19-day-old Swiss chard plants was marginally but significantly P <0.05) increased with increasing SO4 concentration but no effect was observed with increasing NO3 concentration. These results are compared with earlier work on the marked effect of Cl- salinity on Cd availability in Swiss chard. Possible mechanisms explaining the smaller effect of SO4 compared to Cl on Cd availability are proposed.  相似文献
