A pH‐sensitive and double functional nanoprobe was designed and synthesized in a water‐soluble system using thioglycolic acid (TGA) and mercapto‐acetohydrazide (TGH) as the stabilizers. TGA is biocompatible because the carboxyl group is easily linked to biological macromolecules. At the same time, the hydrazide on TGH reacts with the aldehyde on poly(ethylene glycol) (PEG) and forms a hydrazone bond. The hydrazone bond ruptured at specific pH values and exhibited pH‐stimuli‐responsive characteristics. As an optical imaging probe, the PEG–TGA/TGH‐capped CdTe quantum dots (QDs) had high quality, with a fluorescence efficiency of 25–30%, and remained stable for at least five months. This pH‐responsive factor can be used for the effective release of CdTe QDs under the acidic interstitial extracellular environment of tumor cells. This allows the prepared pH‐stimuli‐responsive nanoprobes to show fluorescence signals for use in cancer cell imaging. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

CdTe nanocrystals (NCs) were fabricated through an organic synthesis. The growth and properties of CdTe NCs depended strongly on the preparation conditions. In a reaction system of octadecene and tetradecylphosphonic acid (TDPA), the growth was slow. CdTe NCs with cubic‐like morphology were created in trioctylamine (TOA) using octadecylphosphonic acid (ODPA)‐CdO or TDPA‐CdO as precursors. The TOA and ODPA system gives rise to NCs with high photoluminescence (PL) efficiencies (12%). A CdxZn1‐xS shell coating on the CdTe core, gave rise to tunable dark red PL (630–670 nm). The morphology and PL properties of the CdTe cores were drastically affected by shell coating and this determined the properties of CdTe/CdxZn1‐xS NCs. Small CdTe NCs were easily coated with CdxZn1‐xS shells. The resulting core/shell NCs revealed a spherical morphology. However, shell growth became slow when large CdTe cores were used. This is ascribed to the cores with a cubic‐like morphology. CdS interlayer plays an important role for the formation of the CdTe/CdxZn1‐xS NCs because the experimental result indicated it is difficult to coat CdTe NCs with a ZnS shell. The core/shell NCs benefited from a CdxZn1‐xS composite shell because CdTe/CdS NCs created via a similar procedure revealed a low PL efficiency. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Water‐soluble thioglycolic acid (TGA)‐capped core/shell CdTe/CdS quantum dots (QDs) were synthesized. The interactions of rhein and emodin with TGA‐CdTe/CdS QDs were evaluated by fluorescence and ultraviolet‐visible absorption spectroscopy. Experimental results showed that the high fluorescence intensity of TGA‐CdTe/CdS QDs could be effectively quenched in the presence of rhein (or emodin) at 570 nm, which may have resulted from an electron transfer process from excited TGA‐CdTe/CdS QDs to rhein (or emodin). The quenching intensity was in proportion to the concentration of both rhein and emodin in a certain range. Under optimized conditions, the linear ranges of TGA‐CdTe/CdS QDs fluorescence intensity versus the concentration of rhein and emodin were 0.09650–60 µg/mL and 0.1175–70 µg/mL with a correlation coefficient of 0.9984 and 0.9965, respectively. The corresponding detection limits (3σ/S) of rhein and emodin were 28.9 and 35.2 ng/mL, respectively. This proposed method was applied to determine rhein and emodin in human urine samples successfully with remarkable advantages such as high sensitivity, short analysis time, low cost and easy operation. Novel, water‐soluble CdTe quantum dots (QDs) capped with β‐cyclodextrin (β‐CD) and ~ 4.0 nm in diameter were synthesized in aqueous solution, and characterized using transmission electron microscopy (TEM). A fluorescence‐sensing system based on the photoinduced electron transfer (PET) of (mono‐6‐thio‐β‐CD)–CdTe QDs was then designed to measure the interaction of phenothiazine dyes [methylene blue (MB) and methylene green (MG)] with herring sperm DNA (hsDNA). This fluorescence‐sensing system was based on a fluorescence “OFF–ON” mode. First, MB/MG adsorbed on the surface of (mono‐6‐thio‐β‐CD)–CdTe QDs effectively quenches the fluorescence of (mono‐6‐thio‐β‐CD)–CdTe QDs through PET. Then, addition of hsDNA restores the fluorescence intensity of (mono‐6‐thio‐β‐CD)–CdTe QDs, because hsDNA can bind with MB/MG and remove it from the as‐prepared (mono‐6‐thio‐β‐CD)–CdTe QDs. In addition, detailed reaction mechanisms of the (mono‐6‐thio‐β‐CD)–CdTe QDs–MB/MG–hsDNA solution system were studied using optical methods, by comparison with the TGA–CdTe QDs–MB/MG–hsDNA solution system. l ‐glutathione capped highly fluorescent CdTe quantum dots (QDs) were prepared by an aqueous approach and used as fluorescent labels to link albumin bovine serum (BSA) and rat anti‐mouse CD4, which was expressed on mouse T‐lymphocyte and mouse spleen tissue. The sharp and narrow emission peaks showed that the as‐prepared QDs have desirable dispersibility, uniformity and good fluorescence properties. Both CdTe–BSA and CdTe–CD4 conjugates showed an enhancement of fluorescence intensity over that of bare CdTe QDs. The experimental result of gel electrophoresis confirmed the successful conjugation of CdTe–BSA and CdTe–CD4. The fluorescent microscopic images of CdTe–CD4 labeled mouse T‐lymphocyte cells and mouse spleen tissue were compared with that obtained from fluorescein isothiocyanate labeling. It was demonstrated that the CdTe QDs‐based probe exhibited much better photostability and fluorescence intensity than fluorescein isothiocyanate, showing a good application potential in the immuno‐labeling of cells and tissues. The interactions between thioglycolic acid‐capped‐CuInS2/ZnS quantum dots (CuInS2/ZnS/TGA QDs) and tyrosine kinase inhibitor (TKI) were investigated using fluorescence, ultraviolet–visible spectrometry and Fourier transform infrared spectrometry. The results indicated that the fluorescence intensity of CuInS2/ZnS/TGA could be quenched by imatinib, dasatinib, nilotinib, gefitinib and erlotinib, which hinted that CuInS2/ZnS/TGA QDs could be used in the detection of TKI in active pharmaceutical ingredients (API). Calibration curves showed good linear correlation and low detection limits. The average recovery was between 98 and 102%. Moreover, the nature of the fluorescence quenching mechanism of CuInS2/ZnS/TGA QDs by TKI was discussed. A ground state complex was formed by hydrogen bonding between the carboxyl group of CuInS2/ZnS/TGA QDs and the amino group of TKI. Water‐soluble glutathione (GSH)‐capped core/shell CdTe/CdS quantum dots (QDs) were synthesized. In pH 5.4 sodium phosphate buffer medium, the interaction between GSH‐CdTe/CdS QDs and sanguinarine (SA) was investigated by spectroscopic methods, including fluorescence spectroscopy and ultraviolet‐visible absorption spectroscopy. Addition of SA to GSH‐CdTe/CdS QDs results in fluorescence quenching of GSH‐CdTe/CdS QDs. Quenching intensity was in proportion to the concentration of SA in a certain range. Investigation of the quenching mechanism, proved that the fluorescence quenching of GSH‐CdTe/CdS QDs by SA is a result of electron transfer. Based on the quenching of the fluorescence of GSH‐CdTe/CdS QDs by SA, a novel, simple, rapid and specific method for SA determination was proposed. The detection limit for SA was 3.4 ng/mL and the quantitative determination range was 0.2–40.0 µg/mL with a correlation coefficient of 0.9988. CdTe quantum dots (QDs) capped with different stabilizers, i.e. thioglycolic acid (TGA), 3‐mercaptopropionic acid (MPA) and glutathione (GSH) were investigated as fluorescent probes for the determination of Cu2+. The stabilizer was shown to play an important role in both the sensitivity and selectivity for the determination of Cu2+. TGA‐capped CdTe QDs showed the highest sensitivity, followed by the MPA and GSH‐capped CdTe QDs, respectively. The TGA‐ and MPA‐capped CdTe QDs were not selective for Cu2+ that was affected by Ag+. The GSH‐capped CdTe QDs were insensitive to Ag+ and were used to determine Cu2+ in water samples. Under optimal conditions, quenching of the fluorescence intensity (F0/F) increased linearly with the concentration of Cu2+ over a range of 0.10–4.0 µg/mL and the detection limit was 0.06 µg/mL. The developed method was successfully applied to the determination of Cu2+ in water samples. Good recoveries of 93–104%, with a relative standard deviation of < 6% demonstrated that the developed simple method was accurate and reliable. The quenching mechanisms were also described. Here we report a facile one‐pot method for the preparation of high‐quality CdSe nanocrystals (NCs) in aqueous solution under an air atmosphere. Compared with the traditional use of NaHSe or H2Se, the more stable sodium selenite is utilized as the Se source for preparing highly luminescent CdSe nanocrystals. By using mercaptosuccinic acid (MSA) as the capping agent and borate–citrate acid as the buffering solution, CdSe nanocrystals with high quantum yield (up to 70%) have been synthesized conveniently. The influence of different experimental parameters, such as the pH of the precursor solution, the molar ratio of Cd2+ to Na2SeO3 and Cd2+ to MSA on the CdSe nanocrystals, has been systematically investigated. The chemiluminescence (CL) behaviour of the luminol–potassium periodate system enhanced by CdTe quantum dots capped with thioglycolic acid (TGA–CdTe QDs) was studied using kinetic experiments, CL spectra, UV–vis absorption spectra and fluorescence spectra. The production of oxygen‐containing reactant intermediates (O2?? and OH?) in the present CL system was verified by CL. The possible CL mechanism was discussed in detail. Furthermore, theophylline (THP) was determined based on its enhancement of the CL intensity of the CdTe QDs–luminol–potassium periodate system coupled with a flow‐injection technique. Under these optimized conditions, the linear range was found to be from 1.0 × 10?8 to 1.0 × 10?5 g/mL with a detection limit of 2.8 × 10?9 g/mL (3σ). The recoveries for the determination of THP in tablets were from 98.2 to 99.6%.  相似文献

A novel fluorescence assay system for glucose was developed with thioglycollic acid (TGA)‐capped CdTe quantum dots (QDs) as probes. The luminescence quantum yield of the TGA‐capped CdTe QDs was highly sensitive to H2O2 and pH. In the presence of glucose oxidase, glucose is oxidized to yield, gluconic acid and H2O2. H2O2 and H+ (dissociated from gluconic acid) intensively quenched the fluorescence of QDs. The experimental results showed that the quenched fluorescence was proportional to the glucose concentration within the range of 0.01–5.0 mm under optimized experimental conditions. Compared with most of the existing methods, this newly developed system possesses many advantages, including simplicity, low cost, high flexibility, and good sensitivity. Water-soluble gold nanoparticles with an average diameter of 5 nm were prepared with carboxylic acid terminated thiol ligands. These ligands contain zero to eight methylene moieties. CdTe nanocrystals with an average diameter of 5 nm were synthesized with aminoethanethiol capping. These nanocrystals displayed characteristic absorption and emission spectra of quantum dots. The amine terminated CdTe nanocrystals and carboxylic-acid-terminated gold nanoparticles were conjugated in aqueous solution at pH 5.0 by electrostatic interaction, and the conjugation was monitored with fluorescence spectroscopy. The CdTe nanocrystals were significantly quenched upon binding with gold nanoparticles. The quenching efficiency was affected by both the concentration of gold nanoparticles in the complex and the length of spacer between the CdTe nanocrystal and Au nanoparticle. The observed quenching was explained using Förster resonance energy transfer (FRET) mechanism, and the Förster distance was estimated to be 3.8 nm between the donor–acceptor pair.  相似文献

A rapid, simple and sensitive label‐free fluorescence method was developed for the determination of trace amounts of an important drug, heparin. This new method was based on water‐soluble glutathione‐capped CdTe quantum dots (CdTe QDs) as the luminescent probe. CdTe QDs were prepared according to the published protocol and the sizes of these nanoparticles were verified through transmission electron microscopy (TEM), X‐ray diffraction (XRD) and dynamic light scattering (DLS) with an average particle size of about 7 nm. The fluorescence intensity of glutathione‐capped CdTe QDs increased with increasing heparin concentration. These changes were followed as the analytical signal. Effective variables such as pH, QD concentration and incubation time were optimized. At the optimum conditions, with this optical method, heparin could be measured within the range 10.0–200.0 ng mL?1 with a low limit of detection, 2.0 ng mL?1. This work reports for the first time a potential‐based nano‐electrochemiluminescent (ECL) pH sensor, using anatase TiO2 nanocrystals (NCs) as the ECL probe. The first ECL peak potential of the TiO2 NCs shifted negatively with increasing pH, showing a linear range from −0.47 V (vs Ag/AgCl) at pH 3 to −1.06 V at pH 10. This phenomenon was attributed to the absorption of ‘potential‐determining ions’ of OH on the surface of TiO2 NCs, leading to larger impedance of the electron injection. Other common ‘potential‐determining ions’, such as phosphate, induced a slight potential shift of 0.03 V at a concentration of 0.1 M. Using urease as an enzyme model, a urea biosensor was developed by the simultaneous modification of urease and TiO2 NCs on indium–tin oxide (ITO) electrodes. The biosensor, measured on the basis of the pH increase caused by the enzyme catalysis reaction, had a linear range of 0.01–2.0 mM, with a potential shift of 0.175 V. The as‐prepared pH sensor, which has simple construction procedures and acceptable sensitivity and selectivity, may provide new avenues for the construction of ECL bioanalytical methodologies. In this paper, we synthesized a papain‐stabilized fluorescent Au nanocluster (NC) probe and studied its interaction with dopamine. As fluorescence of papain–Au NCs is quenched in the presence of dopamine under alkaline conditions, we were able to establish a simple, selective analytical method for dopamine determination. By studying the fluorescence lifetime and dynamic light scattering of the NCs before and after interaction with dopamine, we found that this fluorescence quenching mechanism was possibly due to dopamine self‐polymerization that produced polydopamine that cross‐linked papain–Au NCs. Based on this new phenomenon, we proposed a highly selective analytical method for dopamine detection. Other small organic molecules, such as amino acids, ascorbic acid and uric acid did not interfere with dopamine detection. Dopamine in the range 20–100 μM can be linearly detected by the fluorescence quenching ratio of gold nanoclusters. Dopamine detection could be visually realized by watching color changes of papain–Au NCs under UV light or daylight, as both fluorescence and absorption of the papain–Au NCs changed during dopamine polymerization.  相似文献

The present study reports a one‐step synthesis method for the preparation of cationic gold nanoclusters (Au NCs). Polyethyleneimine (PEI), a positively charged hyperbranched polyamine, was selected as the capping reagent. Glutathione showed a synergistic effect on the formation of the small size of cationic Au NCs. The prepared cationic Au NCs have a size less than 2 nm and carry a positive charge in solution with pH less than 11. The cationic PEI–Au NCs‐triggered luminol chemiluminescence (CL) reactions showed slow and intense CL profiles. The maximum CL intensity can be obtained within 10 min and the CL signal maintained almost the same within 30 min. A linear increase of CL intensity was observed in the presence of an increasing concentration of cationic Au NCs ranging from 0.030 μM to 15 μM. The linear response of the cationic Au NCs in the CL reaction and the glow‐type CL profile make the proposed CL reaction have broad application prospects in the field of biological analysis and CL imaging.  相似文献

Here we present a facile one-pot method to prepare high-quality CdTe nanocrystals in aqueous phase. In contrast to the use of oxygen-sensitive NaHTe or H(2)Te as Te source in the current synthetic methods, we employ more stable sodium tellurite as the Te source for preparing highly luminescent CdTe nanocrystals in aqueous solution. By selecting mercaptosuccinic acid (MSA) as capping agent and providing the borate-citrate acid buffering solution, CdTe nanocrystals with high quantum yield (QY >70% at pH range 5.0-8.0) can be conveniently prepared by this method. The influence of parameters such as the pH value of the precursor solution and the molar ratio of Cd(2+) to Na(2)TeO(3) on the QY of CdTe nanocrystals was systematically investigated in our experiments. Under optimal conditions, the QY of CdTe nanocrystals is even high up to 83%. The biological application of luminescent MSA-CdTe to HEK 293 cell imaging was also illustrated.  相似文献

Glibenclamide (GB), as a sulfonylurea‐based medication is commonly prescribed for the treatment of type 2 diabetes. Due to its increasing consumption, there is a need to develop a simple, fast, and reliable detection method to follow its concentration in pharmaceutical and biological samples. Herein, a novel fluorometric method is developed for the sensitive measurement of GB. The method is based on the enhancing effect of GB on the fluorescence emission of mercaptopropionic acid (MPA) capped cadmium telluride quantum dots (CdTe QDs). QDs were synthesized in aqueous solution and were characterized by fluorescence spectroscopy, transmission electron microscopy (TEM), X‐ray diffraction (XRD) and Fourier transform infrared spectroscopy (FT‐IR). Fluorescence intensity of QDs was enhanced by adding GB in a very low concentration. The effect of operative factors such as pH, buffer, contact time and concentration of CdTe QDs were investigated and in the optimized condition, a linear increase was achieved for the emission intensity of QDs by increasing GB concentration in the range 49–345 ng mL?1, with a detection limit of 17.84 ng mL?1. The offered method has an acceptable precision (relative standard deviations were < 2.8%) and was satisfactorily applied for the determination of GB in pharmaceutical products and human urine samples.  相似文献

A fluorescence biosensor for organophosphorus pesticides was developed. A pH indicator, CdTe quantum dots, were used as an optical transducer of the inhibition of enzyme by analyte. Through the intervening agency of chitosan, the recognition elements (acetylcholinesterase and CdTe) were immobilized onto the surface of quartz by electrostatic attraction to form a self‐assembled multilayer film. In the absence of pesticide, acetylcholine was biocatalytically hydrolysed to yield acetic acid and choline. The released acid resulted in pH decrease, which was sensed by the immobilized pH indicator (CdTe). In the presence of pesticide, the action of acetylcholine was reduced; the fluorescence intensity of the film changed and was related to the concentration of pesticide. This multilayer film could be used as the biosensor for monocrotophos, with a detection limit of 3.20 × 10?8 mol/L; the sensitivity was 100 times higher than that of CdTe in aqueous solution. Recently great progress has been achieved in highly effective hybrid solar cells fabricated using aqueous materials. The state‐of‐the‐art energy conversion efficiency has been close to 5% with high photocurrent. However, charge separation and transport mechanism in the aqueous‐processed hybrid solar cells are rarely reported and are usually assumed to be similar to oil‐phase processed systems; that is, self‐assembly polymers are mainly responsible for charge separation and carrier transport. To date, this assumption has prohibited further improvement of the conversion efficiency in aqueous‐processed hybrid systems by adopting any appropriate technique routes. Here, ultrafast carrier dynamics in these hybrid solar cells consisting of poly(p‐phenylenevinylene) (PPV)‐based aqueous polymers and water‐solution CdTe nanocrystals (NCs) are investigated in detail. Self‐charge separation in grown CdTe NC partly capped CdS shell layers after anneal treatment is unambiguously identified. Different from their oil‐soluble counterparts, these core/shell nanocrystals do not have the restrictions of quantum confinement and surface ligands, form effective charge transport networks, and play a dominant role in the charge separation and carrier transport processes. These findings provide a greater understanding on the fundamental photophysics in aqueous‐processed hybrid systems.  相似文献
