The genetic origin of kiwifruit (Actinidia deliciosa var.deliciosa) was studied using phylogenetic analysis of DNA sequences derived from the polygalacturonase gene. Results indicate that hexaploid kiwifruit had an allopolyploid origin with the diploidA. chinensis contributing one genome (genome A) and another (as yet unidentified) diploid species contributing a second genome (genome B). The results leave open the question of whether a third, distinct species contributed to the hexaploid kiwifruit genome. A tetraploid race ofA. chinensis is also suggested to be allopolyploid containing genomes A and B.  相似文献

? Premise of the study: Microsatellite makers were developed for Clematoclethra scandens to investigate its population genetics and speciation. ? Methods and Results: A total of 36 microsatellite markers were isolated using the Fast Isolation by AFLP of Sequences COntaining repeats (FIASCO) method. Their polymorphisms were assessed in two natural populations. The results showed that 30 markers displayed prominent polymorphisms and six markers were monomorphic. ? Conclusions: These microsatellite loci will facilitate further studies on population genetics and speciation of C. scandens.  相似文献

首次报道了刚毛藤山柳(Clematoclethra scandens subsp.scandens)的花药和早期胚珠发育的胚胎学特征:花药4室;发育完整的花药壁6层,为基本型花药壁,由外至内分别为表皮、药室内壁、两层中层和两层绒毡层;腺质型绒毡层为2核或3核,纤维性加厚发生于药室内壁;小孢子母细胞减数分裂为同时型胞质分裂,小孢子四分体多为四面体型;成熟花粉粒为2-细胞型,3孔沟或稀4孔沟。早期胚珠为近倒生,单珠被,薄珠心,珠孔由单珠被构成。比较胚胎学研究认为藤山柳属和猕猴桃科的其它两个属都具有单珠被胚珠及其他相似的花药发育特征,这些特征和形态分类系统范畴的第伦桃科和山茶科有明显区别。因此,本文支持花粉形态学、细胞学和分子系统学的观点,认为藤山柳属与水东哥属和猕猴桃属构成一个较好的单系,一并放在猕猴桃科。  相似文献

Saurauia oroquensis, a new species from Colombia remarkably allied toS. bullosa Wawra, is described. It is distinguished from the latter by the absence of stellate trichomes on the lower leaf surface, by the homotrichous nature of sepal pubescence (limited only to the exposed parts in bud, with the imbricated parts and inner surface completely glabrous), and by the much lower number of stamens per flower. This new taxon is endemic to the Colombia-Venezuelan border region (Táchira-Norte de Santander).  相似文献

The pollen-pistil interaction has been examined in kiwifruit (Actinidia deliciosa). In this species a large number of seeds are produced in each fruit and a great many pollen grains germinate and grow to reach the ovules. This growth is assisted by an abundant secretion that is present all along the pistilar tract. At anthesis, the stigma is covered by a secretion where the pollen grains germinate and grow. The stylar transmitting tissue is initially rich in starch reserves, but the starch gradually disappears and, concomitantly, an abundant secretion that stains for carbohydrates appears in all of the intercellular spaces. Pollen tube growth relies on this secretion since it is depleted after pollen tube passage, while in unpollinated flowers it remains unaltered throughout the flower life-span. In the ovary a similar situation occurs. The placental surface, where the pollen tubes grow before reaching the ovules, is covered by a number of obturators. At anthesis, these obturators are rich in starch reserves and have an abundant secretion on their outer surface. As time passes, starch disappears while the secretion increases. It is in this secretion that the pollen tubes grow on their way toward the ovules. These observations are discussed in terms of the support given by the pistil to pollen tube growth to achieve the highly successful reproductive performance of this species.  相似文献

Microsatellite marker transfer across species in the dioecious genus Actinidia (kiwifruit) could offer an efficient and time-effective technique for use during trait transfer for vine and fruit improvement in breeding programmes. We evaluated the cross-species amplification of 20 EST-derived microsatellite markers that were fully informative in an Actinidia chinensis mapping family. We tested all 20 markers on 120 genotypes belonging to 21 species, 5 with varieties and/or chromosome races. These 26 taxa included 16 diploids, 7 tetraploids, 2 hexaploids and 1 octaploid, and represented all four taxonomic sections in the genus. All 20 markers showed some level of cross-species amplification. The most successful marker amplified in all genotypes from all species from all sections of the genus, the least successful amplified fragments only in A. chinensis and A. deliciosa. One species, A. glaucophylla, failed to amplify with all but 2 markers. PIC (Polymorphism information content) values were high, with 14 of 17 markers recording values of 0.90 and above. Sequence data demonstrated the presence of the microsatellite in all the amplified products. Sequence homology was less 5′ of the microsatellite and increased toward the start codon of the translated region of the EST from which the marker was derived. The data confirm that EST-derived microsatellite markers from Actinidia species show cross-species amplification with high levels of polymorphism which could make them useful markers in breeding programmes.  相似文献

Evolutionary relationships within Actinidia, a genus known for the contrasting mode of inheritance of its plastids and mitochondria, were studied. The phylogenetic analysis is based on chloroplast (cp) and mitochondrial (mt) restriction site and sequence data (matK, psbC-trnS, rbcL, and trnL-trnF for cpDNA; nad1-2/3 and nad4-1/2 for mtDNA). The analysis of cp sequence data confirms the hypothesis that the four currently recognized sections are not monophyletic. The detection of incongruences among phylogenies (mtDNA vs. cpDNA tree) coupled with the detection of intraspecific polymorphisms confirms some of the reticulations previously emphasized, diagnoses new hybridization/introgression events, and provides evidence for multiple origin of at least two polyploid taxa. A number of hybridization/introgression events at the diploid, tetraploid, and possibly hexaploid levels are documented. The extensive reticulate evolution undergone by Actinidia could account for the lack of clear morphological discontinuities at the species level.  相似文献

There are various arguments on classification of the genus Actinidia Lindl., a genus with approximately 63 species, 59 of which have been found in China. The paper investigated the characteristics of foliar trichomes of 35 taxa from China under optical microscope, including size, celluar structure, distribution and density. According to their micromorphological characteristics, foliar trichomes can be classified into the following six categories: 1) single-cell hairs; 2) uniseriate hairs, including linear, bulbous, twisted, straight-walled, and bent-walled hairs; 3) multiseriate hairs, including twisted, straight-walled and gradually sharpening, straight-walled and suddenly sharpening, bent-walled and gradually sharpening, and suddenly sharpening hairs; 4) multiseriate thick hairs, including pillar hairs, gradually sharpening thick hairs, and suddenly sharpening thick hairs; 5) stellate hairs, including parenchyma-stellate and sclerenchyma-stellate (normal state and special states such as rosulate, peltate-stellate, and overlopping-stellate) hairs; and 6) dichotomous hairs. On the basis of the micromorphological characteristics of foliar trichomes in Actinidia, with Clematoclethra lasioclada as an outgroup, both the quantitative cladistic analysis and phenetic analysis were performed using Wagner method and UPGMA clustering method respectively to reconstruct the phylogeny of Actinidia in China. The phylogenetic tree generated by cladistic analysis suggests that the sect. Leiocarpae be a monophyletic group, but other three sections, i.e., sect. Maculatae, sect. Strigosae and sect. Stellatae, be non-monophyletic groups. The results obtained from the phenetic analysis reflect relationships among the taxa of Actinidia in China, especially a close relationship between A. chinensis and A. deliciosa, and a relatively remote relationship between A. callosa var. henryi and A. callosa var. discolor. In conclusion, the micromorphological characters of foliar trichomes and the methods of quantitative taxonomic analysis are of key importance tostudies on phylogenetic and phenetic relationships of Actinidia.  相似文献

Chromaphyosemion killifishes are a karyotypically highly diverse group of small, sexually dimorphic fishes living in rainforest rivulets in tropical West and Central Africa. In the present study, we used various chromosome banding and staining techniques to analyse the karyotypes of 13 populations representing seven described species ( Chromaphyosemion loennbergii , Chromaphyosemion punctulatum , Chromaphyosemion splendopleure , Chromaphyosemion volcanum , Chromaphyosemion malumbresi , Chromaphyosemion melanogaster , Chromaphyosemion bitaeniatum ) and two undescribed forms ( Chromaphyosemion cf. lugens , Chromaphyosemion sp. Rio Muni GEMHS00/41). Diploid chromosome numbers (2 n ) and the number of chromosome arms (NF) ranged from 2 n  = 24 in C. malumbresi to 2 n  = 40 in C. bitaeniatum and from NF = 40 in C. volcanum and C. cf. lugens to NF = 54 in one population of C. loennbergii . A tentative XX/XY sex chromosome system was revealed in C. loennbergii , C. melanogaster , C. malumbresi , and Chromaphyosemion sp. Rio Muni GEMHS00/41. Mapping cytogenetic data for all described Chromaphyosemion species onto a recently published mitochondrial DNA phylogeny revealed a complex pattern of chromosomal evolution with several independent reductions of 2 n and independent modifications of NF and nucleolus organizer region phenotypes. Together with the results of preliminary crossing and mate choice experiments, the cytogenetic and molecular phylogenetic data suggest that, contrary to previous hypotheses, chromosomal rearrangements are probably not the most important and certainly not the only factor driving speciation in Chromaphyosemion killifishes.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society , 2008, 94 , 143–153.  相似文献

Interspecific hybridization is widespread among plants. Understanding the phylogenetic relationships among species is necessary for revealing the potential hybridization events. Actinidia, best known as kiwifruit genus found throughout a wide range in eastern Asian from Indonesia to Siberia. In this study, phylogenetic relationships of Actinidia species with sympatric distributions were investigated using three chloroplast introns (trnL-F, atpB-rbcL and rpl32-trnL) and three Exon primed intron-crossing (EPIC) markers. Chloroplast phylogeny supports non-monophyly of the five species studied excluding Actinidia fulvicoma var. lanata. The non-monophyly was also revealed by EPIC markers. Our results showed EPIC markers are more variable and informative for phylogenetic inference than that of chloroplast markers. The incongruences between loci from the plastid and nuclear DNA phylogenic trees may stem from incomplete lineage sorting or historical introgression hybridization. Incomplete lineage sorting may explain the non-monophyly between Actinidia chrysantha (section Maculatae) and other four species (section Stellatae), and introgression hybridization and high level of interspecific gene flow may explain the non-monophyly among the species of sect. Stellatae. Thus, natural hybridization and introgression may be common in Actinidia with sympatric distribution.  相似文献

A series of chloroplast and nuclear probes were used to examine restriction fragment length polymorphisms (RFLPs) in kiwifruit (Actinidia deliciosa) and three of its closest relatives. The four species fell into two pairs, withA. chinensis andA. deliciosa closely related but some distance away from the other two species,A. latifolia andA. eriantha. 猕猴桃自然居群SSR遗传变异的空间自相关分析   总被引:10,自引:2,他引:10  
本研究采用空间自相关分析方法对同域分布的中华猕猴桃(Actinidiachinensis)和美味猕猴桃(A.deliciosa)自然居群SSR遗传变异的空间结构进行了研究,以探讨猕猴桃自然居群遗传变异的分布特征。选用的9对SSR引物在两物种中共扩增出104个等位基因。选择频率在20–80%的SSR等位基因,运用等样本对频率方法分别对同域分布的中华猕猴桃和美味猕猴桃各1个居群及其中华/美味猕猴桃复合居群进行了空间自相关系数Moran’sI值计算。结果表明:中华猕猴桃和美味猕猴桃的遗传变异在居群内均存在着一定程度的空间结构,尽管近半数或半数以上的等位基因在居群内表现为随机分布的空间模式,但也有相当比例(29.6–48.0%)的等位基因在种内居群中和复合居群中(河南西峡51.0%,陕西商南44.7%)呈现渐变、衰退、双向衰退或侵扰模式。而且其居群内遗传变异的空间分布规律,不论是在种内还是复合居群中都基本一致:相距在100m以内,特别是30m范围内的个体间的等位基因表现出显著性的正相关,但随着地理距离的增大逐渐显示出负相关,说明猕猴桃属植物的有效传粉距离可能在100m左右,种子散播主要集中在30m的近距离内。猕猴桃自然居群遗传变异的空间结构是其传粉和种子散播等生物学特性与生境共同作用的结果,其中种子近距离的散播、花粉传播的有限距离及人为干扰是最主要的因素。本研究结果揭示了这两个近缘物种居群遗传变异的空间分布特征及相互关系,有助于进一步探讨猕猴桃属植物的遗传变异、居群扩散及其地理系统发育进化等方面的规律,并为制定相应的保育策略和措施提供基础数据和科学依据。  相似文献

Karyology and reproductive biology of a marine population of the species complex Gyratrix hermaphroditus, from Roscoff (Brittany, France), have been investigated. A diploid complement of six chromosomes was determined from spermatogonial mitotic figures. One chromosome pair is metacentric, the second is intermediate between meta- and submetacentric, and the third is subtelocentric.In this population, regular meiosis occurs in both female and male germ lines, and the animals reproduce only by means of amphimictic eggs. Certain specimens of the population showed the elimination of one of the three bivalents during the first meiotic division in spermatogenesis. It seems that such animals produce normal and aneuploid sperm simultaneously; the aneuploid sperm are not capable of fertilization.The Roscoff population differs in its karyotype (2n = 6) from freshwater populations, which are either diploid (2n = 4) or polyploid (3n = 6, 4n = 8). These results suggest that aneuploidy played a role in the differentiation of freshwater populations from an originally marine species complex.  相似文献
