In Experiment 1, 24 suckled beef cows were assigned to 4 treatment groups (6 cows/group). Group I cows calved spontaneously. Parturition was induced in Groups 2, 3 and 4 with 20 mg dexamethasone (DEX) 8 to 12 days prior to expected calving date. Additionally, cows in Groups 3 and 4 received 8 mg triamcinalone acetonide (TA) 6 days prior to DEX treatment. Animals in Group 4 also received 10 mg estradiol benzoate (EB) with TA, and on alternate days until DEX, when 20 mg EB was given. Gonadotropin releasing hormone (GnRH, 100 mug) was given intramuscular (IM) to all cows on days 2 or 3 postpartum. Plasma LH increased (P< .05) following GnRH treatment in Groups 2, 3 and 4, but not in Group 1. LH release (area under the curve) following GnRH was greater (P< .05) for cows in Group 4 compared to cows in Groups 1, 2 or 3, and differences in LH release between Groups 1, 2 or 3 were not significant. In Experiment II, 36 mature Hereford cows were assigned to a 2 x 3 factorial experiment (6 cows/group). Groups 1 and 2, 3 and 5, and 4 and 6 received 0, 100, or 200 mug GnRH (IM) at 78 hr postpartum, respectively. In addition, cows in Groups 2, 5 and 6 received 5 mg EB at 36 hr postpartum. Plasma LH concentrations were not different (P <.05) among groups from 36 to 78 hr postpartum. A surge of LH in response to EB treatment was not detected at 54 to 62 hr (18 to 26 hr post EB), indicating a lack of response by the positive feedback mechanism at this early time postpartum. Mean plasma LH concentrations were elevated 78 to 82 hr postpartum for Groups 3 through 6. Treatment with EB at 36 hr caused a significantly greater (P< .05) response to GnRH with 200 mug of GnRH releasing more LH than 100 mug of GnRH.  相似文献

Prolonged reduction in energy intake in beef heifers has been reported to suppress ovulation but the mechanisms involved are poorly understood. The objective of this study was to examine whether changes in the pattern of LH secretion following each of three different tests predicted the functional state of the hypothalamo-pituitary-ovarian (H-P-O) axis. Test 1 examined the ratio of LH secretion during the 1h before and 2h after naloxone (NAL) administration. The other two tests assessed the LH surge following an exogenous oestradiol positive feedback signal (Test 2) or exogenous progesterone priming (Test 3). In phases 1 and 3, each of 8 weeks duration, the heifers were fed 100% of their maintenance energy requirements. In phase 2, of 9 weeks duration, they were fed 50% of their maintenance energy requirements. Oestrus was induced in all heifers by PG administration at the start of the experiment.Heifers were administered a naloxone challenge of 50, 100, 200 or 400mg naloxone hydrochloride i.v. (one dose per heifer) during the mid-luteal period of phase 1 and all four naloxone treated heifers received 400mg naloxone hydrochloride at the end of phases 2 and 3. Doses of 10, 20 or 40 mg oestradiol benzoate (EB) i.m. were each administered to two of the remaining heifers during the mid-luteal period of phase 1. One heifer on each dose of oestradiol benzoate in phase 1 had the same dose administered at the end of phases 2 and 3. The progesterone challenge was administered to three heifers by insertion of a PRID for 12 days starting in the middle of phase 2.In Test 1, the ratio of LH secretion before and after naloxone administration in phase 1 was 1:1 (50mg), 1:4 (100mg), 1:4 (200mg) and 1:9 (400mg) (50mg versus 100mg and 100mg versus 200mg doses, P<0.05); 50mg versus 400mg doses, P<0.001). In phase 2, this ratio was 1:1 and there was no response to 400mg dose of naloxone in any of the four heifers. In phase 3, the ratio depended on the ovarian activity in the heifer and ranged from 1:1 to 1:4 (P<0.05). In Test 3, a positive oestradiol feedback signal was detected in cyclic heifers in phases 1-3 but not in the acyclic heifer in phase 2. Heifers challenge with exogenous progesterone did not have oestradiol or LH values above threshold levels.We conclude that all three tests successfully predicted the functional state of the hypothalamo-pituitary-ovarian axis. In nutritionally undernourished beef heifers onset of ovarian acyclicity is either preceded or accompanied by the loss of a positive feedback signal (Test 2) and progesterone priming ability (Test 3), and that a plasma LH ratio of > or =1:2 following naloxone challenge (Test 1) is a sign of recovery of the functional state of the hypothalamo-pituitary-ovarian axis.  相似文献

A study was conducted to determine reproductive response of primiparous beef heifers to an ovulation induction regimen during restricted dietary energy intake. Thirty-seven Barzona x Hereford heifers, maintained under drylot conditions, were utilized. Heifers were restricted in TDN on a pen basis to approximately 50% of N.R.C. recommendations for the first 90 days postpartum, then received 120% for 80 days thereafter. All animals received control injections (C) or 30 mg progesterone on day 15 postpartum followed in 48 hours by 2 mg estradiol-17beta (PE). Treated heifers not ovulating at first treatment and/or not cycling, were re-treated at 60 day postpartum, with non-cycling C heifers receiving control injections. Intact fertile bulls were maintained with the heifers from day 1 to 170 days postpartum, with visual observations for signs of estrus and breeding activity conducted twice daily during this period. At first treatment, seven of 18 heifers ovulated, one conceived and four continued to cycle. At second treatment, three of 13 conceived and seven returned to a synchronized estrus 15 to 21 days later. Although intervals to first estrous behavior and estrus favored PE heifers (P< .05), by 90 and 170 days postpartum no advantage in interval to conception or number conceiving was observed.  相似文献

The hypothesis that luteinizing hormone (LH) secretion in prepubertal females is responsive to estradiol negative feedback and that decreased feedback occurs as puberty approaches was tested in heifers. In the first experiment, seven heifers were maintained prepubertal by dietary energy restriction until 508 days of age (Day 0). All heifers were placed on a high-energy diet on Day 0 at which time they received no additional treatment (CONT), were ovariectomized (OVX) or were ovariectomized and subcutaneously implanted with estradiol-17 beta (OVX-E2). This feeding regimen was used to synchronize reproductive state in all heifers. A second experiment was performed with 16 prepubertal heifers using the same treatments at 266 days (Day 0) of age (CONT, OVX and OVX-E2) but no dietary intake manipulation. In both experiments, LH secretion increased rapidly following ovariectomy in OVX heifers. In the initial experiment, LH secretion was maintained at a low level in OVX-E2 heifers until a synchronous rapid increase was noted coincidental with puberty in the CONT heifer. In the second experiment, LH secretion increased gradually in OVX-E2 heifers and attained castrate levels coincidental with puberty in CONT heifers. A gradual increase in LH secretion occurred as puberty approached in CONT heifers. These results indicate that: a) LH secretion in prepubertal heifers is responsive to estradiol negative feedback; and b) estradiol negative feedback decreases during the prepubertal period in beef heifers.  相似文献

The working hypothesis in the present study was that changes in concentrations and secretory patterns of luteinizing hormone (LH), 17 beta estradiol (E2), and progesterone in sexually mature beef heifers fed diets deficient in energy are related to changes in body weight of the animals. Another important component of the study was to determine if concentrations and secretion patterns of the reproductive hormones changed over time as feeding of the experimental diets continued. Twelve Red Angus X Hereford heifers (20 mo of age; 355 +/- 7 kg) were assigned randomly to receive a low- (L, n = 7) or high- (H, n = 5) energy diet for 100 days (Day 0 = day of initiation of dietary treatment). All heifers were exhibiting estrous cycles at regular intervals when the experiment was initiated and continued to exhibit estrous cycles at regular intervals throughout the study. Stage of the estrous cycle was synchronized in all 12 heifers by administration of prostaglandin F2 alpha (PGF2 alpha) on two occasions (Days 45 and 75) during the experiment. Serial blood samples (taken at 12-min intervals for 4 h) were collected at 0, 12, 24, 36, 48, and 60 h after the PGF2 alpha injections (Days 45-47 and 75-77) to determine patterns of LH secretion during the follicular phase of the estrous cycle. In addition, serial blood samples (taken at 20-min intervals for 18 h) to monitor LH secretion during the luteal phase of the estrous cycle, in which the stage of the cycle was standardized between heifers, were obtained (Days 59 and 89).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Five studies were conducted in yearling heifers which were either puberal or prepuberal utilizing the Syncro-Mate-B (SMB) treatment (G. D. Searle & Co., Chicago, Ill.). In trial 1 the pregnancy rate in Brahman crossbred heifers after 21 days of breeding was 56% and 28% greater (P<.005) in the SMB treated groups than in the nontreated groups. Pregnancy rate in 3 4 Simmental heifers (trial 2) was increased 39% (P<.05) after 21 days of breeding. Hereford and crossbred heifers (trial 3) weighing less than 250 kg failed to respond to SMB treatment whereas heifers weighing greater then 250 kg had increased reproductive performance. Conversely the results obtained in trial 4 using Brahman crossbred heifers was not influenced by weight or SMB treatment. Crossbred Santa Gertrudis heifers (trial 5) weighing greater then 250 kg and treated with SMB had improved reproductive performance compared to the control groups. While in treated heifers weighing less than 250 kg reproductive performance was far from optimum.  相似文献

In the first experiment, the effect of the stress of blood collection (via tail vessel puncture) on serum luteinizing hormone (LH) was evaluated in six nonsuckled first calf Brangus heifers. The animals were bled on days 22 and 31 postpartum at 15 minute intervals for a period of two hours. Blood was processed to yield serum and analyzed for LH via radioimmunoassay (RIA). There were no significant differences or fluctuations in serum LH levels between bleeding periods or between cows. Serum LH concentrations in nonsuckled cows were not affected by the stress of blood collection. In the second experiment, 24 first calf Brangus heifers were randomly assigned to one of four treatment groups. Treatment 1 cows were suckled once daily for approximately 30 min starting day 21 postpartum. Treatment 2 cows were suckled twice daily for approximately 30 min each time, starting 21 days postpartum. Treatment 3 cows were suckled once daily for approximately 30 min starting 30 days postpartum. Treatment 4 cows were suckled twice daily for approximately 30 min each time starting 30 days postpartum. Each cow was bled via tail vessel puncture on days one and nine following the start of each treatment. The blood sampling regime was similar to that used in Experiment 1 and consisted of four presuckling samples taken at 15 min intervals, one midsuckling sample (the calf was allowed to suckle for 15 min) and four postsuckling samples taken at 15 min intervals. Blood was collected, processed to yield serum and assayed for LH via RIA. Suckling intensity (SI) was found to have a significant effect on serum LH levels. The once daily suckled cows had higher (P<.01) mean serum LH levels than did the twice daily suckled cows (1.70 +/- .03 and 1.53 +/- .03 ng/ml, respectively). The LH concentrations decreased (P<.01) from the first to last bleeding time (BT). The mean serum LH levels for the presuckling, midsuckling and the first postsuckling samples were higher (P<.05) than the last postsuckling sample. The mean serum LH level for the first time period prior to suckling was higher (P<.05) than the last postsuckling sample. The mean serum LH level for the first time period prior to suckling was higher (P<.05) than the last two periods after suckling (1.73 +/- .08 ng/ml vs 1.51 +/- .06 and 1.41 +/- .06 ng/ml). Bleeding day (BD) and weaning day (WD) did not alter serum LH levels. The interactions found to be significant (P<.01) were SIxBD, SIxWD, BDxWD and BTxSIxBDxWD.  相似文献

The effects of dietary energy and suckling on adrenal function and luteinizing hormone (LH) concentrations were investigated in primiparous postpartum cows. Ten heifers were assigned at calving to either high (22.8 Mcal/day) or low (15.2 Mcal/day) energy diets. Blood samples were collected every 15 minutes for 8 hours on 28, 42, and 56 days post partum. Calves were allowed to suckle ad libitum during sampling periods. Serum samples were analyzed by radioimmunoassay for LH and cortisol. Concentrations of catecholamines were quantified by reverse-phase HPLC. Body weights were decreased (P<0.01) by low energy intake. In addition, low energy diet cows had lower mean LH concentrations (0.97 +/- 0.09 vs 1.57 +/- 0.07 ng/ml), P<0.05) than high energy diet cows. Luteinizing hormone concentrations in high energy diet cows increased with days post partum, resulting in a treatment-by-time interaction (P<0.005). Treatment did not affect mean cortisol concentrations. However, within 15 minutes of suckling cortisol release was significantly above baseline in 77% of the observed suckling events. Dihydroxyphenylalanine (DOPA) increased in high energy diet cows compared with that of low energy diet cows (2,833 +/- 243 vs 1,294 +/- 243 pg/ml, P<0.01). Norepinephrine (NE) and 3,4-dihydroxyphenylacetic acid (DOPAC) were not influenced by treatment. Plasma NE decreased during the postpartum interval (P<0.005). These data suggest that reduced energy intake may prevent the increase in LH associated with increasing days post partum and alter adrenal function. In addition, spontaneous suckling events elicit a release of cortisol.  相似文献

Twenty-seven dairy cows were evenly assigned to one of three groups and given an intramuscular injection of 2 mg estradiol benzoate. Cows in group 1 were greater than 30 days postpartum at treatment and had been diagnosed via rectal palpation to have ovarian cysts. Cows in groups 2 and 3 were 12 to 14 and 30 to 40 days postpartum, respectively. Blood plasma was collected from all cows before treatment and then every three hours for 36 hours post-treatment. Concentrations of LH, estradiol-17 beta and progesterone in plasma were determined by radioimmunoassay. Four, zero and five cows in groups 1, 2 and 3, respectively, had concentrations of progesterone greater than 1.0 ng/ml before estradiol benzoate treatment. None of these cows had a peak LH release greater than 5 ng/ml following estradiol benzoate treatment. The numbers of cows with progesterone concentrations less than 1 ng/ml that released LH (>5 ng/ml) in response to estradiol benzoate were 3 of 5, 3 of 9, and 4 of 4 for groups 1, 2, and 3, respectively; the proportion for group 3 was higher (P<.05) than for group 2. Of the cows that released LH, mean peak LH concentrations were 33.3+/-5.4, 14.8+/-7.2 and 24.6+/-9.8 ng/ml for groups 1, 2 and 3, respectively, and the duration of the LH increase was 8.0+/-1.0, 8.0+/-2.0 and 13.0+/-4.0 hours. The time from estradiol benzoate treatment to peak LH release for cows with ovarian cysts (25+/-2 hours) was delayed (P<.05) compared with that for cows 30 to 40 days postpartum without ovarian cysts (16+/-1 hour). In summary, responsiveness to estradiol benzoate is regained between 2 to 4 weeks postpartum in most cows. In addition, some cows with ovarian cysts can release LH in response to estradiol benzoate, but peak LH release is delayed compared to cows at a comparable stage postpartum without ovarian cysts.  相似文献

Brahman cows (n = 49) and primiparous heifers (n = 11), Brahman x Hereford primiparous F1 heifers (n = 86) and Simmental x Brahman primiparous F1 heifers (n = 13) were randomly allotted by breed, age and date of calving to one of eight treatment groups: 1) control; 2) once-daily suckling; 3) lasalocid (200 mg/hd/d); 4) alfaprostol (5 mg intermuscular injections on Days 21 and 32 post partum); 5) lasalocid + once-daily suckling; 6) alfaprostol + once daily suckling; 7) alfaprostol + lasalocid; 8) alfaprostol + lasalocid + once daily suckling. All animals received 2.3 kg/hd/d of a concentrate (6 corn : 1 cottonseed meal) and lasalocid was mixed and fed in the concentrate. Body weights and condition scores were taken on Day 1 post partum and every 28 d thereafter. All animals were maintained with sterile marker bulls with Brahman and Simmental x Brahman cattle artificially inseminated at first estrus. Blood samples were collected at weekly intervals starting on Day 21 post partum until estrus and at nine to twelve days post estrus when the ovaries were palpated for corpora lutea. After the first postpartum estrus with a corpora lutea, cows were placed with fertile bulls. Mean serum progesterone concentrations were below 0.5 ng/ml prior to treatment. Calf weight gains to 90 d were not affected by age (P > 0.10) but were lower in the once-daily suckling group (P < 0.05). Treatment did not affect cow weight or condition score (P > 0.10). Cows had a shorter postpartum interval (P < 0.0001) than heifers. Once-daily suckling shortened postpartum interval (P < 0.0001) and positively influenced the cumulative frequency of return to estrus by 40 d post partum (P < 0.02). Alfaprostol did not affect postpartum interval (P > 0.10) but did increase the cumulative frequency of return to estrus by 90 d post partum (P < 0.03). Lasalocid did not affect postpartum interval or cumulative frequency of return to estrus (P > 0.10). Both once-daily suckling and alfaprostol were effective in increasing the numbers of animals inseminated by 90 d post partum. The once-daily suckling + alfaprostol treatment resulted in the shortest postpartum interval.  相似文献

The objectives were to examine the effects of dietary energy and protein density on age and body composition at puberty, and on ovarian follicular dynamics during the pre- and peripubertal periods in Holstein heifers. In Phase 1, heifers were randomly allotted (n=10 per diet) at 100 kg body weight (BW) to diets with either low (P1L), medium (P1M) or high (P1H) energy and protein formulated for an average daily gain (ADG) of 0.5, 0.8 or 1.1 kg per day, respectively. During Phase 2 (P2), all heifers were fed ad libitum a common diet formulated for an ADG of 0.8 kg per day. Half the animals within the high (n=5) and low groups (n=5) entered P2 either at 12 months of age (P2H-12; P2L-12) or at 330 kg BW (P2H-330; P2L-330). Heifers fed P1H, P1M, P1L, and P2L-12 diets attained puberty at approximately 9, 11, 16, and 14 months of age, respectively (P<0.01). Urea space estimates of body fat and protein percent, and back-fat thickness, were lower in P1L heifers compared to P1H or P1M heifers at similar chronological ages (P<0.05) but did not differ at puberty (P>0.10). Compared to P1L heifers, P1H heifers had high amplitude LH pulses at 8 months, and high frequency low amplitude LH pulses at 10 months of age (P<0.05). The mean diameter (mm) of the dominant follicle was smaller (P<0.05) in P1L heifers (10.6) compared to P1H (12.8) or P1M (12.2) heifers at 8 months. Maximum size and growth rate of the nonovulatory dominant follicle increased with age (P<0.05) but did not differ between P1H and P1M heifers at puberty. The diameter (mm) of the nonovulatory dominant follicle, and the first and second ovulatory follicles were larger in P2L-12 heifers (14.0, 14.7, and 14.9) compared to P1M heifers (13.1, 12.5, and 11.9), while the peak progesterone levels and CL growth were lower (P<0.05) in the first cycle. In conclusion, dairy heifers attained puberty at a constant body weight and body composition independent of dietary manipulation, the size of dominant follicles increased with age in association with increased LH support, and heifers realimented from a low energy diet developed larger first ovulatory follicles and smaller CL with lower peak progesterone concentrations in the first cycle.  相似文献

Angus and Hereford yearling heifers (469) were bred to either Hereford or Angus bulls and fed either a high level (13.3 Mcal of ME) or a low level of nutrition (7.3 Mcal of ME) for the last 116 days of pregnancy. The level of calving difficulty did not differ between heifers receiving the high (H) and low (L) level of nutrition the first year (36% and 34%) but was significantly higher (P < .01) in heifers receiving H the second year (33% vs. 16%). Calves born to heifers receiving H weighed at birth 2.7 kg more (P < .01) both years than calves born to heifers receiving L. In the first year of the study the size of the pelvic opening was 10 cm larger 1 week before calving (P < .01) in heifers receiving H than in heifers receiving L. Calf survival was not related to level of nutrition, breed of dam or breed of sire either year of the study.  相似文献

This study was conducted to determine the efficacy of feeding melengestrol acetate (MGA) for 14 days and administering prostaglandin F(2)alpha (PGF) 17 days after MGA to synchronize or induce estrus in yearling beef heifers. The study involved 56 Angus (n = 19), Hereford (n = 15) and Simmental (n = 22) heifers that were assigned by breed and pubertal status to either MGA+PGF or to control groups. Heifers in the synchronized group were fed 0.5 mg MGA per head per day for 14 days from a grain carrier and were injected with 25 mg, i.m. PGF 17 days after the last daily feeding of MGA. Control heifers were fed from a grain carrier without MGA and were not treated with PGF. Heifers were classified as pubertal when concentrations of progesterono in the serum exceeded 1 ng/ml in 1 of 2 samples collected prior to the initiation of treatments. Blood samples were collected 7 days before and on the day that treatment with MGA or carrier began and 7 days before and on the day that PGF was administered. Progesterone concentrations in the serum were elevated ( > 1 ng/ml) in 61% (17 28 ) of the MGA+PGF-treated heifers and in 61% (17 28 ) of the control heifers prior to feeding MGA. However, concentrations of progesterone in the serum at the time PGF was administered differed (P<0.05) between MGA+PGF and control groups. Concentrations of progesterone in the serum exceeded 1 ng/ml in 100% (28 28 ) of the MGA+PGF-treated heifers and in 71% (20 28 ) of control heifers at the time PGF was administered (P<0.05). All heifers were inseminated 12 hours after the first detected estrus. Twenty-two of 28 (79%) of the MGA+PGF-treated heifers exhibited estrus within 6 days after PGF compared with 9 of 28 (32%) of control heifers (P<0.05). The conception rate at first service did not differ between MGA+PGF and control groups (64% and 67%, respectively). Synchronized pregnancy rates were higher (P<0.05) for MGA+PGF-treated heifers than for control heifers (14 28 , 50% vs 6 28 , 21%). Increased concentrations of progesterone in serum at the time PGF was administered and higher pregnancy rates during the synchronized period among MGA+PGF-treated heifers demonstrate the efficacy of this treatment for use in estrus synchronization. Moreover, this treatment may have a potential effect on inducing puberty in breeding age heifers.  相似文献

A total of eight Simmental heifers (114 ± 3.2 days old and weighing 118 ± 3.8 kg BW) were used to study the effects of feeding method on intake and animal behaviour in a crossover design experiment. Treatments consisted of feeding concentrate and chopped barley straw as (1) choice (CH; concentrate and straw in separate feedbunks) or (2) total mixed ration (TMR; concentrate and straw in one feedbunk). Feeds were offered on an ad libitum basis, but always maintaining a concentrate to straw ratio of 90 to 10. The experiment was performed in two 21-day periods, and sampling was carried out in the last week of each period. At the end of each period, treatment was changed for heifers; hence, the final number of animals per treatment was eight. Intake was recorded over 7 consecutive days. BW was recorded at the beginning and the end of the experiment and on day 21 of each experimental period. Barley straw was coarsely chopped with a chopping machine. Once chopped, all the straw was handled for particle size separation using the 2-screen Penn State Particle Separator and only material of more than 8 mm was used to feed the heifers. Animal behaviour was video-recorded for 24 h on day 2 and day 6 of each experimental period. Concentrate intake and total dry matter intake of heifers fed with the CH feeding method were higher (P < 0.01 and P < 0.05) than when fed with TMR (5.1 and 5.3 v. 4.7 and 5.0 kg dry matter (DM)/day, respectively). Conversely, barley straw was consumed in higher amounts in heifers fed with the TMR feeding method (0.3 v. 0.2 kg DM/day, respectively; P = 0.001). The total NDF intake was similar in both treatments. In contrast, NDF intake from barley straw and physically effective NDF intake were higher in heifers fed with the TMR feeding method than when fed with CH. Feeding method used to feed heifers did not affect the consumption of the different kinds of barley straw particles and eating and drinking behaviours but affected ruminating behaviour. Heifers fed TMR spent more time ruminating than heifers fed concentrate and barley straw separately (376 v. 287 min/day, respectively; P < 0.01). TMR as the feeding method in intensive beef production systems could be a good approach to promote roughage intake.  相似文献

The role of endogenous opioids and nutrition on the inhibition of luteinizing hormone (LH) secretion during the postpartum period was investigated in a Spanish breed of sheep lambing in the mid-late breeding season. Two groups of adult Rasa Aragonesa ewes housed in individual pens and lambing on 30 December were fed during the suckling period to provide maintenance requirements and the production of 1.1 (M; n=8) or 0.55 (L; n=8) kg of milk per day. On days 10, 20 and 30 after lambing, the effect of a treatment with the opiate receptor antagonist naloxone (1 mg/kg at four hourly intervals) on LH secretion was assessed in half of the ewes of each group, the remaining females receiving four saline injections. After weaning, animals were fed to provide requirements for maintenance of liveweight. Blood samples were collected twice a week from day 20 postpartum until the end of March, and assayed for progesterone and prolactin. Although underfed ewes showed significantly lower mean plasma concentrations during the control period on day 20 postpartum, nutrition did not seem to modify LH secretion before naloxone or saline injections. Moreover, no differences between nutritional groups in the response to naloxone injections on pattern of LH secretion were found. In fact, naloxone treatment induced an increase of mean LH concentrations on days 10, 20 and 30 postpartum (at least, P<0.05), of LH pulse frequency on days 20 and 30 (P<0.05), and of LH pulse amplitude on days 10 and 20 (P<0.05). Underfed ewes during the postpartum period showed a slower decline in plasma prolactin levels, with significant differences on days 29, 36 and 39 after lambing (P<0.05). Only 3 M ewes ovulated before the onset of the seasonal anoestrus period. It is concluded that endogenous opioids are involved in the inhibition of LH secretion during the early suckling period of a reduced seasonality breed of sheep without any influence of nutrition on the response to naloxone treatment; however, ewes underfed before weaning failed to reactivate their cyclicity prior to the onset of the seasonal anoestrus.  相似文献

Fall-lambing western range ewes were fed either a high-or low-energy ration the last two months of gestation and were fed protein and energy in excess postpartum. GnRH was infused for 10 hours on day 5 or 26 postpartum. Blood samples were collected at 20-minute intervals during infusion and radioimmunoassayed for LH. Net weight change from day 60 prepartum to day 1 postpartum was 0.67 +/- 1 kg vs. -6.9 +/- 1.5 kg (mean +/- SE) for the high-and low-energy groups, respectively (P < 0.05). Nutritional treatment had no effect on LH response at either day 5 or day 26 postpartum, althoughthe day 26 LH response to GnRH was greater (P < 0.05) than the day 5 LH response. LH response was greater (P < 0.05) in ewes that gained weight prepartum vs. ewes that lost weight prepartum, but only in ewes infused on day 5 postpartum.  相似文献

The effect of dietary energy and weight class on the fertility of yearling beef heifers (Angus, Hereford, and Angus x Hereford) was investigated over 2 years. In year 1, heifers (n=58) were classed as heavy (HW; > or =340 kg) or light weight (LW; <340kg) and then assigned to receive either a low (LE; 0.23 kg/day) or high energy (HE; 0.68 kg/day) diet. In year 2, heifers (n=60) were also classified as heavy (> or =335kg) or light weight (<335 kg), but the energy content of the diet was raised so that heifers on the LE and HE were targeted to gain 0.46 and 0.79 kg/day, respectively. Heifers in the four groups, LELW (n=14 and 12), LEHW (n=16 and 17), HELW (n=13 and 15), and HEHW (n=15 and 16) received restricted amounts of concentrate (HE > LE) and free choice hay over 47 or 42 days (year 1 and year 2, respectively). To synchronize estrus, heifers were fed capsules containing MGA (0.5 mg/animal each day) beginning 11 days before the end of the feeding trial (day 0), PGF(2alpha) (25mg i.m.) and estradiol benzoate (Ebeta; 400 microg i.m.) was given on days 8 and 10, respectively. Estrous behavior was observed (days 10 and 11) and all heifers were inseminated on day 11. Following AI, heifers were re-grouped and a bull was introduced (days 27-39) for the second service in both years. Pregnancy diagnosis for the first (days 41-42) and second services (days 69-97) was performed by transrectal ultrasonography. Transrectal ultrasonic observations of ovarian follicle number and size were completed for a subset of heifers (n=5-8) from each experimental group at the end of the feeding trial. The effect of year was not significant for any of the reproductive performance variables measured. The mean ADG was (0.72 +/- 0.04 kg/day) and was greater in LW than HW heifers and in heifers in the HE than LE treatment groups (P <0.05). In heifers receiving the LE diet, ADG was lower in HW than LW heifers (weight x diet; P=0.02; 0.54 +/- 0.04 and 0.62 +/- 0.03 kg/day for HW and LW heifers, respectively). The diameter of the largest follicle was greater in heifers receiving the HE diet (P < 0.05; 11.3 +/- 0.4 mm) than those on the LE diet (10.3 +/- 0.3), and in LW (P <0.05) compared to HW heifers. The HE diet increased the size of the largest follicle in LW but not HW heifers (diet x weight, P <0.05). The percentage of pubertal heifers at the end of the feeding period (59.3%), estrous response (56.4%), conception rate (47.7%), ovulation rate (88.9%), and first service pregnancy rate (36.2%) were not significantly affected by initial weight or diet. There was a tendency for first service pregnancy rates to be greater in LW than HW heifers consuming the LE diet (diet x weight, P <0.1; 54.2 +/- 15 and 30.3 +/- 10% for LELW and LEHW heifers, respectively). Pregnancy rate after two services was greater (P=0.01) in LW (82 +/- 10%) than in HW (64.5 +/- 10%). The LE diet achieved moderate rates of gain and allowed high level of reproductive performance in LW but not HW heifers.  相似文献

Suckling may prolong the anovulatory period postpartum by 1) a neural-mediated inhibition of luteinizing hormone-releasing hormone (LHRH)-induced gonadotropin secretion, or 2) an inhibitory effect of hormones released by suckling on gonadotropin secretion and/or action at the ovary. In the present investigation we considered whether a suckling event caused 1) acute inhibition of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion, and 2) release of glucocorticoids and/or prolactin (PRL). Six Hereford cows remained intact and six were ovariectomized (ovx) on day 7 postpartum. Calves remained with their dams continuously. Cows were bled at 10-min intervals during 6 consecutive hr on days 14, 28 and 42 postpartum. Both LH and FSH were released episodically by day 14 in intact and ovx cows, but suckling did not acutely affect LH and FSH secretion. A PRL release accompanied suckling 67, 96 and 95% of the time. However, among all instances where PRL was released on days 14, 28 and 42 postpartum, 67, 29 and 37% occurred independent of a suckling event. Glucocorticoids were not released by suckling in intact cows but were released in ovx cows. We conclude that suckling does not acutely affect LH or FSH concentrations in serum of cows postpartum, that PRL concentrations usually increase in serum coincident with suckling but can be released at other times, and suckling-induced glucocorticoid release depends upon the presence of the ovary.  相似文献

Twenty suckled CharloixxHereford beef cows (5 cows/group) were assigned at random to receive 100 microg GnRH (IM) at either 2 to 3, 7 to 8, 15 to 16, or 31 to 32 days postpartum, Groups 1 through 4, respectively. Blood samples for hormone determinations were collected at time 0 (pre-GnRH), every half hr for 3 hr, and at 4.0 hr and 6.0 hr post-GnRH. Mean plasma LH, estradiol-17beta, or progesterone concentrations were not different among groups prior to GnRH. Plasma LH increased (P<.05) following GnRH in Groups 2, 3 and 4, but not in Group 1. Peak GnRH induced LH release was greater (P<.05) in Groups 3 and 4 than in Groups 1 or 2. Correlation coefficients between days postpartum and peak LH release (r=.72), and estradiol-17beta concentrations and time of LH peak (r=-.42) were significant (P<.05). These data indicate that LH release in response to GnRH, in suckled beef cows is not fully restored until 15 to 16 days postpartum.  相似文献

Beef heifers, 13 months old, were fed to achieve high (7.6 +/- 0.2 units) or low (3.9 +/- 0.1 units) body condition by feeding them one of two diets for 20 weeks. During week 17 of the growth phase, all heifers were ovariectomized. From week 20 to week 27 (restriction phase), all heifers were fed a daily diet containing 0.071 MJ metabolizable energy kg-1 body weight. At weekly intervals throughout the restriction phase, blood samples were collected at 10-min intervals for 11 h to determine the pattern of secretion of luteinizing hormone (LH), the amount of LH released in response to 750 ng (pituitary responsiveness) and 50 micrograms LH-releasing hormone (LHRH, releasable stores) and mean concentrations of follicle-stimulating hormone (FSH) in the circulation. Body weight declined during the restriction phase in a similar fashion in heifers with high and low body condition and changes in body weight were unrelated to mean concentrations of LH and FSH and frequency of LH pulses. Amplitude of LH pulses and responsiveness to 750 ng LHRH increased in a linear fashion with weight loss in heifers with low but not in those with high body condition. The amount of LH released in response to 50 micrograms LHRH decreased with increasing weight loss in heifers with high but not with low body condition, indicating that releasable pools of LH declined with increased weight loss in heifers with high body condition.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
