The Andes of South America hosts perhaps the highest amphibian species diversity in the world, and a sizable component of that diversity is comprised of direct-developing frogs of the genus Pristimantis (Anura: Craugastoridae). In order to better understand the initial stages of species formation in these frogs, this study quantified local-scale spatial genetic structuring in three species of Pristimantis. DNA sequences of two mitochondrial gene fragments (16S and COI) were obtained from P. brevifrons, P. palmeri and P. jubatus at different locations in the Cordillera Occidental. We found high levels of genetic diversity in the three species, with highly structured populations (as measured by F ST) in P. brevifrons and P. palmeri while P. jubatus showed panmixia. Large effective population sizes, inferred from the high levels of genetic diversity, were found in the three species and two highly divergent lineages were detected within P. jubatus and P. palmeri. Estimated divergence times among populations within P. brevifrons and P. palmeri coincide with the Pleistocene, perhaps due to similar responses to climatic cycling or recent geological history. Such insights have important implications for linking alpha and beta diversity, suggesting regional scale patterns may be associated with local scale processes in promoting differentiation among populations in the Andes.  相似文献

Weed populations can have high genetic plasticity and rapid responses to environmental selection pressures. For example, 100-fold amplification of the 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) gene evolved in the weed species Amaranthus palmeri to confer resistance to glyphosate, the world’s most important herbicide. However, the gene amplification mechanism is unknown. We sequenced the EPSPS gene and genomic regions flanking EPSPS loci in A. palmeri, and searched for mobile genetic elements or repetitive sequences. The EPSPS gene was 10,229 bp, containing 8 exons and 7 introns. The gene amplification likely proceeded through a DNA-mediated mechanism, as introns exist in the amplified gene copies and the entire amplified sequence is at least 30 kb in length. Our data support the presence of two EPSPS loci in susceptible (S) A. palmeri, and that only one of these was amplified in glyphosate-resistant (R) A. palmeri. The EPSPS gene amplification event likely occurred recently, as no sequence polymorphisms were found within introns of amplified EPSPS copies from R individuals. Sequences with homology to miniature inverted-repeat transposable elements (MITEs) were identified next to EPSPS gene copies only in R individuals. Additionally, a putative Activator (Ac) transposase and a repetitive sequence region were associated with amplified EPSPS genes. The mechanism controlling this DNA-mediated amplification remains unknown. Further investigation is necessary to determine if the gene amplification may have proceeded via DNA transposon-mediated replication, and/or unequal recombination between different genomic regions resulting in replication of the EPSPS gene.  相似文献

Field studies were conducted to evaluate the capacity of Amaranthus palmeri to grow and reproduce following incomplete physical control in Gossypium hirsutum fields. A. palmeri plants that emerged simultaneously with a G. hirsutum crop were selected for use. Treatments included severing the main stem of flowering plants at heights of 0, 3 and 15 cm above the soil level. A non‐cut/intact control, in which the apical meristem was not removed, was also included. Six weeks after treatment, intact A. palmeri plants had grown to a mean height of 210 cm (SE = 38) and produced 477 408 (SE = 81 250) seeds per plant. Thirty‐five percent of the A. palmeri plants cut back to a height of 15 cm above the soil level did not recover from the treatment; survivors regrew to a mean height of 102 cm (51% reduction, compared to intact plants) and produced 116 000 seeds per plant (73% reduction). A. palmeri plants cut to 3 cm above the soil level had a mortality rate of 64%, an 82% reduction in final plant height, and produced 28 000 seeds per plant. When stems were severed at the soil surface, plant mortality was 95%; final plant height and seed production of survivors were reduced by 95 and 99%, respectively, relative to the control. G. hirsutum seeded yields exceeded 3 t ha?1 when A. palmeri plants were cut back to at least 15 cm, whereas yield was reduced 50% in the control treatment, where A. palmeri growth was not interrupted by cutting. In conclusion, while there is immediate benefit of removing A. palmeri plants in terms of G. hirsutum yield, incomplete stem removal can have multi‐season implications. Results demonstrate that severely pruned A. palmeri plants can resume growth, reach reproductive maturity and produce viable seed, which have the potential to repopulate soil seedbanks.  相似文献

Background and Aims

The analysis of herbarium specimens has previously been used to prospect for ‘new’ hyperaccumulators, while the use of foliar manganese (Mn) concentrations as a taxonomic tool has been suggested. On the basis of their geographic and taxonomic affiliations to known Mn hyperaccumulators, six eastern Australian genera from the Queensland Herbarium collection were sampled for leaf tissue analyses.

Methods

ICP-OES was used to measure Mn and other elemental concentrations in 47 species within the genera Austromyrtus, Lenwebbia, Gossia (Myrtaceae), Macadamia (Proteaceae), Maytenus and Denhamia (Celastraceae).

Key Results

The resulting data demonstrated (a) up to seven ‘new’ Mn hyperaccumulators, mostly tropical rainforest species; (b) that one of these ‘new’ Mn hyperaccumulators also had notably elevated foliar Ni concentrations; (c) evidence of an interrelationship between foliar Mn and Al uptake among the Macadamias; (d) considerable variability of Mn hyperaccumulation within Gossia; and (e) the possibility that Maytenus cunninghamii may include subspecies.

Conclusions

Gossia bamagensis, G. fragrantissima, G. sankowsiorum, G. gonoclada and Maytenus cunninghamii were identified as ‘new’ Mn hyperaccumulators, while Gossia lucida and G. shepherdii are possible ‘new’ Mn hyperaccumulators. Of the three Myrtaceae genera examined, Mn hyperaccumulation appears restricted to Gossia, supporting its recent taxonomic revision. In the context of this present investigation and existing information, a reassesment of the general definition of Mn hyperaccumulation may be warranted. Morphological variation of Maytenus cunninghamii at two extremities was consistent with variation in Mn accumulation, indicating two possible ‘new’ subspecies. Although caution should be exercised in interpreting the data, surveying herbarium specimens by chemical analysis has provided an effective means of assessing foliar Mn accumulation. The inheritance of glyphosate resistance in two Amaranthus palmeri populations (R1 and R2) was examined in reciprocal crosses (RC) and second reciprocal crosses (2RC) between glyphosate-resistant (R) and -susceptible (S) parents of this dioecious species. R populations and Female-R × Male-S crosses contain higher 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) gene copy numbers than the S population. EPSPS expression, EPSPS enzyme activity, EPSPS protein quantity, and level of resistance to glyphosate correlated positively with genomic EPSPS relative copy number. Transfer of resistance was more influenced by the female than the male parent in spite of the fact that the multiple copies of EPSPS are amplified in the nuclear genome. This led us to hypothesize that this perplexing pattern of inheritance may result from apomictic seed production in A. palmeri. We confirmed that reproductively isolated R and S female plants produced seeds, indicating that A. palmeri can produce seeds both sexually and apomictically (facultative apomixis). This apomictic trait accounts for the low copy number inheritance in the Female-S × Male-R offsprings. Apomixis may also enhance the stability of the glyphosate resistance trait in the R populations in the absence of reproductive partners.  相似文献

Nonnative Lehmann lovegrass (Eragrostis lehmanniana) has invaded large areas of the Southwestern United States, and its impact on native plants is not fully understood. Palmer’s agave (Agave palmeri), an important resource for many pollinators, is a key native plant potentially threatened by E. lehmanniana. Understanding potential impacts of E. lehmanniana on A. palmeri is critical for anticipating the future of the desert community where they coexist and for addressing management concerns about associated threatened and endangered species. Our study provides strong indications that E. lehmanniana negatively impacts A. palmeri in several ways. Areas of high E. lehmanniana abundance were associated with significantly lower densities and greater relative frequencies of small A. palmeri, suggesting that E. lehmanniana may exclude A. palmeri. There were no significant differences in species richness, abundance, or community composition when comparing flower associates associated with A. palmeri in areas of high and low E. lehmanniana abundance. However, we did find significantly lower connectedness within the pollination network associated with A. palmeri in areas with high E. lehmanniana abundance. Although E. lehmanniana forms thick stands that would presumably increase fire frequency, there was no significant association between E. lehmanniana and fire frequency. Interestingly, medium to high densities of A. palmeri were associated with areas of greater fire frequency. The complex ramifications of E. lehmanniana invasion for the long-lived A. palmeri and interlinked desert community warrant continued study, as these species are likely to continue to be found in close association due to their similar soil preferences.  相似文献

Background and Aims

A deeper understanding of mutualism can be reached by studying systems with measurable costs and benefits. Most studies of this type focus on an unusual class of obligate, species-specific pollination mutualisms. The interaction between Datura wrightii (Solanaceae) and the hawkmoth Manduca sexta offers similar advantages but greater generality. Adult moths both nectar at and deposit eggs on the same plant; larvae are herbivorous. The antagonistic component of this interaction has been well studied. Here the role of M. sexta as a pollinator of D. wrightii, particularly in the context of this moth''s frequent nectaring visits to the bat-pollinated plant Agave palmeri, is documented.

Methods

Hand-pollinations were used to determine breeding system and the reproductive consequences of mixed loads of A. palmeri and D. wrightii pollen. Plants and moths were caged overnight to assess whether nectaring visits led to fruit and seed set. Finally, pollen deposited on field-collected stigmas was identified, with a particular focus on documenting the presence of D. wrightii and A. palmeri grains.

Key Results

Datura wrightii is highly self-compatible, and a visit that deposits either outcross or self pollen almost doubles fruit and seed set compared with unvisited flowers. Manduca sexta transferred enough pollen to produce fruit and seed sets comparable to hand-pollination treatments. Agave palmeri did not interfere with D. wrightii success: in the field, stigmas received almost pure D. wrightii pollen, and hand-addition of large quantities of A. palmeri pollen had no measurable effect on fruit and seed set.

Conclusions

The floral visitation component of the D. wrightii–M. sexta interaction is indeed mutualistic. This finding is essential background to future development of this interaction as a model system for studying mutualism''s costs and benefits. It is already proving valuable for dissecting third-species effects on the outcome of mutualism. Results indicate that M. sexta''s heavy visitation to A. palmeri has no negative effect on the benefits conferred to D. wrightii. However, it can be predicted to augment M. sexta populations to the point where the costs of the interaction begin to exceed its benefits.Key words: Datura wrightii, Agave palmeri, pollination, herbivory, mutualism  相似文献

There is increasing recognition that both competition and facilitation are important drivers of plant community dynamics in arid and semi-arid environments. Decades of research have provided a litany of examples of the potential for shrubs as nurse plants for establishment of desirable species, especially in water-limited environments. However, interactions with the existing understory community may alter the outcome of interactions between shrubs and understory plants. A manipulative experiment was conducted to disentangle interactions between a native forb species (Penstemon palmeri A. Gray), a native shrub (Artemisia tridentata Nutt.), and a diverse understory of exotic and native forbs and grasses in a semi-arid shrubland of Northern Utah, USA. Seedlings of P. palmeri were transplanted in a factorial design: (1) beneath shrub canopies or into their interspaces and (2) with understory interactions retained or removed. Transplant survival was tracked for roughly 1 year. Shrubs appeared to facilitate P. palmeri survival while interactions with the existing understory community were equivalently negative, leading to overall neutral interactions. Further, positive shrub interactions and negative understory interactions appeared to operate independently and simultaneously. While the debate over the importance of facilitation and competition in driving plant community dynamics continues, our observations strongly suggest that both have considerable effects on plant establishment in A. tridentata communities. Furthermore, our results inform the conservation and restoration of P. palmeri populations, and suggest the utility of nurse shrubs and/or understory thinning as strategies for increasing the diversity of desirable species in the arid and semi-arid western United States shrublands.  相似文献

Longitudinal sections of seeds of representatives of all the genera were examined with the light microscope. Two broad groups based on shape of the seeds, ornamentation of the external surface of their testa, and the position of the embryo relative to the endosperm were recognised. A well-preserved isolated graptolite Prolinograptus packhami Rickards and Wright, 1997, well known from Australia, is described for the first time in Europe. The genus has now been recognized in three continents (Australia, Europe and North America). The Australian P. packhami occurs in the Neocucullograptus inexpectatus/Neocucullograptus kozlowskii biozonal interval, while the form from the Mielnik IG-1 core, Poland, appears earlier, in the Bohemograptus praecornutus Biozone. Prolinograptus, of unclear origin, is one of the least-known genera of the subfamily Linograptinae. P. packhami, along with Neodiversograptus palmeri and Prolinograptus orangensis, form a group of very diminutive specimens, with slender stipes and thecal apertures showing varying degrees of eversion. Scanning electron microscope examination revealed the presence of some micro-ornamentation on the rhabdosome surface of the Polish Prolinograptus, similar to that known in retiolitids (Retiolitidae).  相似文献

Four conidial germination types namely, polygoni (syn. Pseudoidium), cichoracearum (syn. Reticuloidium), pannosa (syn. Fibroidium) and fuliginea (syn. Magnicellulatae) are commonly used as an aid in the identification of the Oidium anamorphs of powdery mildews. However, results of germination tests and a survey of the literature showed that these types did not adequately distinguish all taxa and did not reflect the range of species covered. Hence two new main types, Striatoidium and Blumeria, are proposed for the newly created genus Neoerysiphe and for the unique pattern of B. graminis. Two new names, orthotubus and brevitubus subtypes of Fibroidium, are proposed for the pannosa and fuliginea types respectively. Also proposed is a special longitubus pattern for the long, undifferentiated, negatively hydrotropic germ tubes prevalent in Erysiphe trifolii and species in Golovinomyces sect. Depressi. The recognition of the Striatoidium type of N. galeopsidis as distinct from the Pseudoidium type of E. elevata facilitated the detection of a simultaneous infection of Catalpa by these two powdery mildews. A key is provided for the identification of Oidium genera based on germination types. A review of germination patterns in the tribe Phyllactinieae found no consistent differences amongst the genera. Golovinomyces sect. Depressi is re-described to accommodate Golovinomyces spp. often having a longitubus pattern of germination. It includes G. cichoracearum var. latisporus, now considered a separate species based on its germination type, other anamorphic morphology and previous molecular sequence analyses. A new combination, Golovinomyces ambrosiae, is proposed for this species. Other anomalies within G. cichoracearum s. lat. were addressed by proposing another new combination, G. fischeri for the former G. cichoracearum var. fischeri that differs from G. cichoracearum s. str. in having larger chasmothecia and a well distinguished anamorph, and by proposing a new species, G. sonchicola, that is biologically, phylogenetically and morphologically distinct from G. cichoracearum s. str.  相似文献

The new genus Groveromyia gen. n. was described from the Late Eocene Rovno amber. The genus includes 4 species: G. occlusa sp. n., G. digna sp. n., G. astrosa sp. n., and G. concinna (Fedotova, 2005) comb. n. (Bryocrypta). Some changes in the biometric parameters of the genera and species were revealed relative to those in the recent and the Late Eocene Stomatosematidi. Keys to the recent and fossil genera of the tribe Stomatosematidi and the species of the genus Groveromyia are given.  相似文献

Amplification of the EPSPS gene has been previously identified as the glyphosate resistance mechanism in many populations of Amaranthus palmeri, a major weed pest in US agriculture. Here, we evaluate the effects of EPSPS gene amplification on both the level of glyphosate resistance and fitness cost of resistance. A. palmeri individuals resistant to glyphosate by expressing a wide range of EPSPS gene copy numbers were evaluated under competitive conditions in the presence or absence of glyphosate. Survival rates to glyphosate and fitness traits of plants under intra-specific competition were assessed. Plants with higher amplification of the EPSPS gene (53-fold) showed high levels of glyphosate resistance, whereas less amplification of the EPSPS gene (21-fold) endowed a lower level of glyphosate resistance. Without glyphosate but under competitive conditions, plants exhibiting up to 76-fold EPSPS gene amplification exhibited similar height, and biomass allocation to vegetative and reproductive organs, compared to glyphosate susceptible A. palmeri plants with no amplification of the EPSPS gene. Both the additive effects of EPSPS gene amplification on the level of glyphosate resistance and the lack of associated fitness costs are key factors contributing to EPSPS gene amplification as a widespread and important glyphosate resistance mechanism likely to become much more evident in weed plant species.  相似文献

The taxonomic composition ofPhrygilanthus has long been a source of confusion. Until recently the genus was considered to be distributed from Mexico, Central and South America, to Australia, New Guinea, and the Philippines. Analyses of floral morphology, inflorescence structure, life form, fruit type, and especially chromosome numbers indicate thatPhrygilanthus, as classically circumscribed, is a highly heterogeneous and artificial assemblage containing elements now referable to the following ten genera:Cecarria, Desmaria, Gaiadendron, Ligaria, Muellerina, Notanthera, Psittacanthus, Struthanthus, Tripodanthus, andTristerix. Phrygilanthus itself is nomenclaturally illegitimate and must be replaced by Notanthera. A synopsis of the genera segregated from thePhrygilanthus group is included. A key to the New World genera of Loranthaceae is also appended as well as a nomenclator indicating the present generic status of taxa at one time or another assigned toPhrygilanthus. One genus,Cecarria, is described as new; five new combinations are made:Cecarria obtusifolia (Merr.) Barlow,Psittacanthus palmeri (Wats.) In a survey of the higher plants for families with rosmarinic acid-accumulating species we could show for the first time, that some species of the family Marantaceae of the order Zingiberales accumulate rosmarinic acid. Other compounds detected in Marantaceae are chlorogenic acid and rutin (quercetin 3-O-rutinoside). Out of 35 species coming from 9 different genera extracted and analysed, two species of Maranta (Maranta leuconeura, Maranta depressa) and one Thalia species (Thalia geniculata) showed the presence of rosmarinic acid. The two Maranta species additionally contained chlorogenic acid, which was also present in Stromanthe amabilis. Rutin was detected in the genera Calathea, Ctenanthe, Maranta, Pleiostachya and Thalia. For a comparison, species from six other families of the Zingiberales were analysed as well.  相似文献

Delftia sp. I24 and a moderately halophilic Ochrobactrum sp. G21 are able to hydrolyse dihydropyrimidines and hydantoins D-specific. The genes being with the utmost probability involved in dihydropyrimidine and hydantoin degradation of these two microorganisms were cloned into an appropriate vector and transformed into E. coli. The putative gene cluster of Delftia sp. I24 included four genes: an incomplete NADPH-dependent glutamate synthase (gltB), dihydropyrimidine dehydrogenase (pydA), permease (hyuP) and an incomplete d-hydantoinase (hyuH). The hydantoinase gene sequence was completed by PCR amplification. The putative gene cluster of Ochrobactrum sp. G21 comprised nine ORFs, six being potentially involved in hydantoin-hydrolysation: carbamoylase (hyuC), d-hydantoinase (hyuH), two transporters (OrfS1 and OrfS2) and two permeases (hyuP1 and hyuP2). Expression of the d-hydantoinases from Delftia sp. I24 and from Ochrobactrum sp. G21 in E. coli, followed by biotransformation assays confirmed hydantoinase activity. This is the first report of the genetical organization of hydantoin-degradation within the genera Delftia and Ochrobactrum. Phylogenetic analysis of the two “novel” hydantoinases and known hydantoinases and dihydropyrimidinases, including putative protein sequences, revealed that they can be classed with some exceptions in the following groups: l-hydantoinases (l-Hyd), Rhizobiales family (Rhizo-Fam), Comamonadacae family (Com-Fam), Pseudomonas family (Pseud-Fam), Bacilli family (Bac-Fam) and Agrobacterium family (Agro-Fam). The highly conserved “histidine motif” for the superfamily of amidohydrolases could be found for all hydantoinases of this study but differences were found in the substrate recognition sites, whereas some of the above mentioned groups showed to posses the same recognition sites as known hydantoinases.  相似文献

Background

Rutaceae subfamily Rutoideae (46 genera, c. 660 species) is diverse in both rainforests and sclerophyll vegetation of Australasia. Australia and New Caledonia are centres of endemism with a number of genera and species distributed disjunctly between the two regions. Our aim was to generate a high-level molecular phylogeny for the Australasian Rutoideae and identify major clades as a framework for assessing morphological and biogeographic patterns and taxonomy.

Methodology/Principal Findings

Phylogenetic analyses were based on chloroplast genes, rbcL and atpB, for 108 samples (78 new here), including 38 of 46 Australasian genera. Results were integrated with those from other molecular studies to produce a supertree for Rutaceae worldwide, including 115 of 154 genera. Australasian clades are poorly matched with existing tribal classifications, and genera Philotheca and Boronia are not monophyletic. Major sclerophyll lineages in Australia belong to two separate clades, each with an early divergence between rainforest and sclerophyll taxa. Dehiscent fruits with seeds ejected at maturity (often associated with myrmecochory) are inferred as ancestral; derived states include woody capsules with winged seeds, samaras, fleshy drupes, and retention and display of seeds in dehisced fruits (the last two states adaptations to bird dispersal, with multiple origins among rainforest genera). Patterns of relationship and levels of sequence divergence in some taxa, mostly species, with bird-dispersed (Acronychia, Sarcomelicope, Halfordia and Melicope) or winged (Flindersia) seeds are consistent with recent long-distance dispersal between Australia and New Caledonia. Other deeper Australian/New Caledonian divergences, some involving ant-dispersed taxa (e.g., Neoschmidia), suggest older vicariance.

Conclusions/Significance

This comprehensive molecular phylogeny of the Australasian Rutoideae gives a broad overview of the group’s evolutionary and biogeographic history. Deficiencies of infrafamilial classifications of Rutoideae have long been recognised, and our results provide a basis for taxonomic revision and a necessary framework for more focused studies of genera and species.  相似文献

The Late Miocene Italian brackish Loxoconchidae are herein discussed and illustrated. Three genera and two subgenera have been recognized in the brackish Italian basins: Loxoconcha, Loxocorniculina, Loxoconchissa (Loxoconchissa) and Loxoconchissa (Loxocaspia). Taking into account the diagnostic characters of several Loxoconchidae genera, in this paper Loxocorniculina is raised at a generic rank, Loxocaspia is confirmed as a subgenus within genus Loxoconchissa and several new species are established: Loxoconchissa (Loxoconchissa) kinoi nov. sp., Loxoconchissa (Loxocaspia) cosentinoi nov. sp., Loxoconchissa (Loxocaspia) nuda nov. sp., Loxoconchissa (Loxocaspia) punctata nov. sp., Loxoconchissa (Loxocaspia) reticulata nov. sp., Loxoconchissa (Loxocaspia) tuberosa nov. sp. and Loxoconchissa (Loxocaspia) velonae nov. sp. Loxocorniculina is a typical Paratethyan genus which widespread into the Palaeo-Mediterranean during the late Messinian Lago-Mare event. Loxoconchissa was known to be widespread only in the Paratethyan realm and in this paper it is signalled for the first time in the Late Tortonian-early Messinian of Italy. The palaeobiogeography of these genera is discussed and the observed continuous distribution of Loxocorniculina against the disjunct distribution of Loxoconchissa leads to suggest that this latter genus underwent a passive dispersal via aquatic birds.  相似文献
