We examined the effects of two enkephalin metabolites, des-tyr-[leu]enkephalin and tyr-gly-gly, on one-way active avoidance conditioning in mice. These metabolites are products of the two major enkephalin hydrolyzing enzymes in plasma, aminopeptidase and angiotensin converting enzyme. Like [leu]enkephalin from which it may be formed, tyr-gly-gly impaired avoidance acquisition, and its dose-response function for this effect was U-shaped. Also like [leu]enkephalin, tyr-gly-gly did not alter locomotor activity. On the other hand, des-tyr-[leu]enkephalin, at the doses tested, was without effect on avoidance conditioning but produced decreased locomotion. These data suggest that the tyrosine end of the enkephalin molecule may be important for its effects on conditioning. Because of their low opioid potencies, it is unlikely that the behavioral actions of tyr-gly-gly and des-tyr-[leu]enkephalin are mediated through opioid receptors.  相似文献

Brain-derived neurotrophic factor (BDNF) was studied initially for its role in sensory neuron development. Ablation of this gene in mice leads to death shortly after birth, and abnormalities have been found in both the peripheral and central nervous systems. BDNF and its tyrosine kinase receptor, TrkB, are expressed in hypothalamic nuclei associated with satiety and locomotor activity. In heterozygous mice, BDNF gene expression is reduced and we find that all heterozygous mice exhibit abnormalities in eating behavior or locomotor activity. We also observe this phenotype in independently derived inbred and hybrid BDNF mutant strains. Infusion with BDNF or NT4/5 can transiently reverse the eating behavior and obesity. Thus, we identify a novel non-neurotrophic function for neurotrophins and indicate a role in behavior that is remarkably sensitive to alterations in BDNF activity.  相似文献

DGAVP facilitates consolidation and retrieval of active and passive avoidance behavior. In this study it was tested whether the long-term behavioral effects of DGAVP are the consequence of an initial increase in behavioral arousal during the learning phase. Animals that were preestimated in an open field test to be low active showed a lower number of conditioned avoidance responses (CAR's) during acquisition and extinction of a shuttle-box task than high active rats. DGAVP was administered 40 min prior to the 1st acquisition session. The immediate effect of DGAVP was a shift in the bell-shaped curve of the relation between arousal and performance (13); an increase in acquisition performance was observed with a low dose of DGAVP (0.1 microgram), while a decrease was found with a high dose of DGAVP (1 microgram). A dose-dependent inhibition of extinction was found in both low and high active animals. These results suggest an immediate effect of DGAVP on the rate of acquisition behavior, which may be a direct consequence of its arousing properties, and a long-term effect on extinction, indicating the formation of memory traces specific for vasopressin-related neuropeptides.  相似文献

Patients with schizophrenia frequently exhibit behavioral abnormalities associated with its pathological symptoms. Therefore, a quantitative evaluation of behavioral dynamics could contribute to objective diagnoses of schizophrenia. However, such an approach has not been fully established because of the absence of quantitative biobehavioral measures. Recently, we studied the dynamical properties of locomotor activity, specifically how resting and active periods are interwoven in daily life. We discovered universal statistical laws ("behavioral organization") and their alterations in patients with major depressive disorder. In this study, we evaluated behavioral organization of schizophrenic patients (n?=?19) and healthy subjects (n?=?11) using locomotor activity data, acquired by actigraphy, to investigate whether the laws could provide objective and quantitative measures for a possible diagnosis and assessment of symptoms. Specifically, we evaluated the cumulative distributions of resting and active periods, defined as the periods with physical activity counts successively below and above a predefined threshold, respectively. Here we report alterations in the laws governing resting and active periods; resting periods obeyed a power-law cumulative distribution with significantly lower parameter values (power-law scaling exponents), whereas active periods followed a stretched exponential distribution with significantly lower parameter values (stretching exponents), in patients. Our findings indicate enhanced persistency of both lower and higher locomotor activity periods in patients with schizophrenia, probably reflecting schizophrenic pathophysiology.  相似文献

Spontaneous locomotor activity (SLA) of the male rats possesses significant individual differences. The daily volume of this activity is sufficiently stable for each separate individual. Animals with high and low SLA also differ in daily dynamics of this parameter. The behaviour of the rats with low SLA was characterized by lesser orienting activity and greater emotional reactivity in comparison to the rats with high SLA. By means of factor analysis it was established that, along with searching activity, emotional reactivity and alimentary motivation, the need in motor activity is one of the main inner factors, determining various manifestations of adaptive behaviour.  相似文献

Exploratory behavior and locomotor activity is enhanced in male rat pups (aged 10 to 20 days) whose dams received a chronic dose (1.25 ml/l) of Tween 80 via their drinking water. This enhancement manifests itself during the diurnal period of the day. These data suggest that Tween 80 has an effect on the CNS which could lead to misinterpretation of results in toxicology studies that use this compound as a dosage vehicle.  相似文献

In an effort to discover if various classes of drugs could be differentiated on the basis of their ability to inhibit spontaneous and methamphetamine-induced locomotor activity, a series of drugs with a variety of pharmacological effects was tested for activity as inhibitors of spontaneous and methamphetamine-induced locomotor activity in mice. When ED50 values for the inhibition of spontaneous locomotor activity are compared to those for inhibition of locomotor activity in the presence of methamphetamine, it is found that approximately the same doses produce 50% inhibition of locomotor activity in both cases. It appears that effects on locomotor activity in the presence and absence of methamphetamine, as described in this paper, cannot be used to differentiate among the various classes of drugs tested. This finding implies that interpretation of anti-methamphetamine activity in drugs must be done with caution.  相似文献

Summary Perch-hopping activity patterns of pinealectomized and normal house sparrows,Passer domesticus, were examined by power spectral analysis and by analysis of daily totals. Power spectral analysis strengthened our previous conclusion that circadian periodicity is absent from the activity of pinealectomized birds kept in constant conditions. The spectral analyses also revealed that obvious ultradian rhythms are not present in sparrow perching activity. Pinealectomy had no effect on the daily activity totals.  相似文献

Scale invariance is a feature of complex biological systems, and abnormality of multi-scale behaviour may serve as an indicator of pathology. The hypothalamic suprachiasmatic nucleus (SCN) is a major node in central neural networks responsible for regulating multi-scale behaviour in measures of human locomotor activity. SCN also is implicated in the pathophysiology of bipolar disorder (BD) or manic-depressive illness, a severe, episodic disorder of mood, cognition and behaviour. Here, we investigated scaling behaviour in actigraphically recorded human motility data for potential indicators of BD, particularly its manic phase. A proposed index of scaling behaviour (Vulnerability Index [VI]) derived from such data distinguished between: [i] healthy subjects at high versus low risk of mood disorders; [ii] currently clinically stable BD patients versus matched controls; and [iii] among clinical states in BD patients.  相似文献

The five most 5' HoxD genes, which are related to the Drosophila Abd-B gene, play an important role in patterning axial and appendicular skeletal elements and the nervous system of developing vertebrate embryos. Three of these genes, Hoxd11, Hoxd12, and Hoxd13, act synergistically to pattern the hindlimb autopod. In this study, we examine the combined effects of two additional 5' HoxD genes, Hoxd9 and Hoxd10. Both of these genes are expressed posteriorly in overlapping domains in the developing neural tube and axial mesoderm as well as in developing limbs. Locomotor behavior in animals carrying a double mutation in these two genes was altered; these alterations included changes in gait, mobility, and adduction. Morphological analysis showed alterations in axial and appendicular skeletal structure, hindlimb peripheral nerve organization and projection, and distal hindlimb musculature. These morphological alterations are likely to provide the substrate for the observed alterations in locomotor behavior. The alterations observed in double-mutant mice are distinct from the phenotypes observed in mice carrying single mutations in either gene, but exhibit most of the features of both individual phenotypes. This suggests that the combined activity of two adjacent Hox genes provides more patterning information than activity of each gene alone. These observations support the idea that adjacent Hox genes with overlapping expression patterns may interact functionally to provide patterning information to the same regions of developing mouse embryos.  相似文献

In repeated short-term tests (four sessions, each of 45-minute duration), and one longer test (a 23.5-hour session), behavior of rats was evaluated in a long, narrow shuttlebox. One side of the box was exposed to an electric field at various strengths, while a visually identical opposite side was shielded from exposure. In the short-term tests, rats generally remained shielded from electric fields of 90 kV/m and greater during the first session, and maintained this response in subsequent sessions. In the longer test, this same preference response was demonstrated at field strengths of 75 kV/m and greater; however, at 25 and 50 kV/m, rats exhibited a statistically significant preference for the exposed region of the shuttlebox, but only during the light portion of a 12-hour light: 12-hour dark cycle. Exposed animals made more traverses than sham-exposed controls between the two ends of the shuttlebox during the first hour of the test. The experimental data support the hypothesis that the observed behavioral effects are the result of direct interaction of the electric field with the animal, and not the result of secondary factors such as electric shock, corona discharge, audible noise, ozone, or vibration of the experimental apparatus.  相似文献

To determine whether luteinizing hormone-releasing hormone (LHRH) contributes to the sexual behavior and locomotor activity of sexually inexperienced male rats, subcutaneous injections of LHRH (500 ng) were given to males. The males showed significant facilitation of a few aspects of sexual behavior 2h after LHRH injection, compared with saline-injected controls. However, the locomotor activity of males injected with LHRH showed no significant change. These results may indicate that LHRH mechanisms play a direct role in the normal regulation of sexual arousal of male behavior in rats.  相似文献

本文研究了根田鼠母体捕食应激对其子代运动及探究行为的作用。母体自妊娠中期至分娩期间,每天被短暂且重复地暴露给艾鼬。分娩后,对母体及其子代正常饲养。在子代性成熟后,分别测定其运动和探究行为。母体捕食应激显著降低子代的运动时间和频率。在探究行为中,母体捕食应激子代的探究潜伏期和探究时间均显著大于对照子代;其次,母体捕食应激雌性子代的探究次数显著小于对照。另外,个体的运动水平以及探究潜伏期和探究次数均存在性别差异。本研究结果表明,田鼠类母体捕食应激对其子代运动和探究行为有显著的抑制作用,由此可能对子代适合度,乃至种群波动产生负效应。  相似文献

The effects of caffeine and its interaction with ethanol were examined in a test of social behavior and a holeboard test of exploration and locomotion. Male mice were injected i.p. with 15, 30 or 60 mg/kg caffeine alone or in combination with 2 g/kg ethanol. The animals were then put in pairs into a familiar arena, or examined individually in the holeboard. Only the highest dose of caffeine (60 mg/kg) had a significant effect on the time spent in social interaction and motor activity in the social behavior test: both measures were reduced. The duration and frequency of avoidance-irritability behavior was dose-dependently increased by caffeine. In the holeboard, caffeine caused a dose-dependent increase in locomotor activity. 30 mg/kg caffeine reversed the ethanol-induced reduction of time spent in social interaction, and 60 mg/kg caffeine antagonized the ethanol-induced increase in locomotor activity in both the social behavior and holeboard tests. Caffeine's effects on ethanol-induced behavioral changes are compared with those of other drugs.  相似文献

Interleukin-6 (IL-6) is a major cytokine in the central nervous system, secreted by different brain cells and with roles in a number of physiological functions. We herewith confirm and expand the importance of astrocytic production of and response to IL-6 by using transgenic mice deficient in astrocytic IL-6 (Ast-IL-6 KO) or in its receptor (Ast-IL-6R KO) in full C57Bl/6 genetic background. A major prosurvival effect of astrocytic IL-6 at early ages was clearly demonstrated. Robust effects were also evident in the control of activity and anxiety in the hole-board and elevated plus-maze, and in spatial learning in the Morris water-maze. The results also suggest an inhibitory role of IL-6 in the mechanism controlling the consolidation of hippocampus-dependent spatial learning. Less robust effects of astrocytic IL-6 system were also observed in despair behavior in the tail suspension test, and social behavior in the dominance and resident–intruder tests. The behavioral phenotype was highly dependent on age and/or sex in some cases. The phenotype of Ast-IL-6R KO mice mimicked only partially that of Ast-IL-6KO mice, which indicates both a role of astrocytes in behavior and the participation of other cells besides astrocytes. No evidences of altered function of the hypothalamic–pituitary–adrenal axis were observed. These results demonstrate that astrocytic IL-6 (acting at least partially in astrocytes) regulates normal behavior in mice.  相似文献
