摘要:【目的】继杂交瘤技术后,重组抗体技术是新一代的抗体制备技术。然而如何用原核系统中较多地表达具有生物活性的单链抗体,避免包涵体形成仍是一个需要探讨的问题。【方法】将目的基因scFv-H4克隆到载体pET22b上,分别转入大肠杆菌BL21(DE3)和Origami(DE3)中,通过改变诱导温度和IPTG浓度,比较具有生物活性的蛋白量以及包涵体的量。【结果】在BL21(DE3)中,pET22b能产生大量表达scFv-H4,而BL21(DE3) 的含有trxA和gor双突变的衍生菌Origami(DE3)表达的scFv-H4的总量较少,但是具有生物活性的蛋白量较多(35 mg/L培养物),具有生物活性的蛋白比例也较BL21(DE3)高。另外IPTG的浓度对scFv-H4表达没有显著影响,而较高的诱导温度会促使表达的蛋白形成包涵体。【结论】在较低的温度下,pET22b能在Origami(DE3)能较好地表达具有生物活性的scFv-H4,减少包涵体的比例,为后续的抗体性质研究和改造奠定了基础。  相似文献

Antibodies currently constitute the most rapidly growing class of human therapeutics; however, the high-yield production of recombinant antibodies and antibody fragments is a real challenge. High expression of active single-chain antibody fragment (scFv) in Escherichia coli has not been successful, as the protein contains three intramolecular disulfide bonds that are difficult to form correctly in the bacterial intracellular environment. To solve this problem, we fused the scFv gene against VEGF165 with a small ubiquitin-related modifier gene (SUMO) by synthesizing an artificial SUMO–scFv fusion gene that was highly expressed in the BL21(DE3) strain. The optimal expression level of the soluble fusion protein, SUMO–scFv, was up to 28.5% of the total cellular protein. The fusion protein was purified by Ni nitrilotriacetic acid (NTA) affinity chromatography and cleaved by a SUMO-specific protease to obtain the native scFv, which was further purified by Ni-NTA affinity chromatography. The result of the high-performance liquid chromatography showed that the purity of the recombinant cleaved scFv was greater than 98%. The primary structure of the purified scFv was confirmed by N-terminal amino acid sequencing and matrix-assisted laser desorption/ionization time-of-flight mass spectroscopy analysis. In vitro activity assay demonstrated that the recombinant scFv could dose-dependently inhibit VEGF165-induced human umbilical vein-derived endothelial cell proliferation. The expression strategy presented in this study allows convenient high yield and easy purification of recombinant scFv with native sequences.  相似文献

为了实现特异性识别斑马鱼卵黄蛋白原的噬菌体展示单链抗体的可溶性表达,将不能以可溶性蛋白形式表达的、只能以噬菌体展示形式特异性识别斑马鱼卵黄蛋白原的单链抗体F5的基因,克隆到pET 32a载体并转化入大肠杆菌ori DE3中。结果表明,通过诱导表达,可获得可溶性的并且仍特异性识别斑马鱼卵黄蛋白原的单链抗体32a-F5。噬菌体展示单链抗体不能可溶性表达是噬菌体展示技术应用中常见的问题,该方法提供了一种表达可溶性单链抗体的可行性方案。  相似文献

人工合成了芋螺毒素SO3的基因片段,通过链延伸方法获得了SO3的全长基因。在此基础上,将SO3基因插入到含抗大鼠转铁蛋白受体的单链抗体基因的pTIG-Trx表达载体中,构建含抗大鼠转铁蛋白受体单链抗体和SO3融合基因的重组表达载体,转化大肠杆菌BL21(DE3)和Origmia(DE3)/DsbC并得到了表达,该融合蛋白主要以包涵体形式存在。为进一步研究芋螺毒素SO3跨血脑屏障转运和治疗作用奠定了基础。  相似文献

Recombinant expression and EGFR-binding activity assay of single chain nimotuzumab (nimotuzumab scFv) is reported in this study. The scFv was produced in VH-linker-VL format in Origami? 2(DE3)pLysS bacterial cells and purified using Ni-TNA resin column. 3-D structure prediction using I-TASSER (Iterative Threading Assembly Refinement) server and analyzing the predicted models using YASARA (Yet Another Scientific Artificial Reality Application) viewer revealed that VH and VL domains assemble into a correctly-folded single chain antibody that is able to bind EGFR. The scFv was evaluated in ELISA and western blot tests and proven to be able to bind EGFR-overexpressing cancer cells (A-431 cells in an efficient manner but unable to recognize cancer cells expressing low levels of EGFR (MCF-7 breast cancer cells).  相似文献

Human epidermal growth factor (hEGF) can stimulate the division of various cell types and has potential clinical applications. However, the high expression of active hEGF in Escherichia coli has not been successful, as the protein contains three intra-molecular disulfide bonds that are difficult to form correctly in the bacterial intracellular environment. To solve this problem, we fused the hEGF gene with a small ubiquitin-related modifier gene (SUMO) by synthesizing an artificial SUMO-hEGF fusion gene that was highly expressed in Origami (DE3) strain. The optimal expression level of the soluble fusion protein, SUMO-hEGF, was up to 38.9% of the total cellular protein. The fusion protein was purified by Ni-NTA affinity chromatography and cleaved by a SUMO-specific protease to obtain the native hEGF, which was further purified by Ni-NTA affinity chromatography. The result of the reverse-phase HPLC showed that the purity of the recombinant cleaved hEGF was greater than 98%. The primary structure of the purified hEGF was confirmed by N-terminal amino acid sequencing and MALDI-TOF mass spectroscopy analysis. Using the method of methylthiazoletetrazolium, the mitogenic activity on Balb/c 3T3 cells of the purified hEGF was comparable to that of commercial hEGF.  相似文献

目的:应用噬菌体展示技术筛选针对表皮生长因子受体突变体Ш (epidermal growth factor receptor variant type Ⅲ, EGFRvIII)的单链抗体 (single chain Fv, scFv)。方法:利用原核表达纯化的人EGFRvIIIex蛋白和高表达EGFRvIIIex的小鼠成纤维细胞系NIH3T3免疫小鼠,扩增VH和VL片段并拼装成scFv 基因,连接至噬菌粒pCANTAB 5E,电击转化Hpd3cells,构建噬菌体单链抗体库,并进行3轮富集筛选。在第4轮筛选时,采用了降低抗原浓度的方法。然后将筛选得到的阳性克隆测序分析,转化E.coli HB2151,IPTG 诱导可溶性scFv 的表达。结果:构建了库容为7.9×107 的噬菌体单链抗体库。经过第4轮低浓度抗原筛选,得到了较高亲和力的克隆。取单个阳性克隆测序分析结果表明,该抗EGFRvIII scFv 基因序列长807 bp,编码268个氨基酸。IPTG诱导后表达的可溶性scFv 可分别与纯化的EGFRvIIIex抗原以及细胞表面的EGFRvIIIex结合。结论:利用噬菌体抗体库筛选得到了高亲和力的抗EGFRvIII scFv,为开发针对EGFRvIII的抗体药物提供了靶向载体分子。  相似文献

We previously isolated phage antibodies from a phage library displaying human single chain antibodies (scFvs) by screening with a mannotriose (Man3)-bearing lipid. Of four independent scFv genes originally characterized, 5A3 gene products were purified as fusion proteins such as a scFv-human IgG1 Fc form, but stable clones secreting 1A4 and 1G4 scFv-Fc proteins had never been established. Thus, bacterial expression systems were used to purify 1A4 and 1G4 scFv gene products as soluble forms. Purification of 1A4 and 1G4 scFv proteins from inclusion bodies was also carried out together with purification of 5A3 scFv protein in order to compare their Man3-binding abilities. The present studies demonstrated that 1A4 and 1G4 scFv proteins have a higher affinity for Man3 than 5A3 scFv protein, which may determine whether scFv-Fc proteins expressed in mammalian cells are retained in the ER or secreted. Furthermore, the inhibitory effects of anti-Man3 1G4 scFv and anti-Tn antigen scFv proteins on MCF-7 cell growth were evaluated. Despite the fact that no obvious difference was detected in cell growth, microscopic observations revealed inhibition of foci formation in cells grown in the presence of the anti-carbohydrate scFv proteins. 前期研究结果发现,SCYL1-BP1具有细胞周期调控功能,同时具有肿瘤抑制因子的特性。目的:采用基因工程技术,构建SCYL1-BP1的大肠杆菌重组表达菌株,以获得足够量的高纯度目的蛋白,为后面进行一系列药理学检测及新药安全性测试奠定基础。方法:利用从人胎脑cDNA文库中克隆得到SCYL1-BP1基因克隆为模板,经PCR扩增,通过酶切位点克隆到新型原核表达载体pET-28b-SUMO上,转化大肠杆菌表达菌BL21(DE3)。经IPTG诱导表达,摸索优化表达条件,表达产物经Ni柱进行亲和层析纯化,后再进行SDS-PAGE和Western blot等分析鉴定。结果:成功构建了SCYL1-BP1的原核表达工程菌BL21(DE3)/pET-28b-SUMO-SCYL1BP1。SDS-PAGE和Western blot检验结果表明,诱导表达的融合蛋白His6-SUMO-SCYL1BP1的分子量约为65 kDa,主要以可溶的形式存在,且能被His标签抗体和SCYL1-BP1单克隆抗体特异性识别。结论:原核表达并纯化了人SCYL1-BP1融合蛋白,为其后续功能研究及性质实验奠定基础。  相似文献

Overproduction of recombinant proteins in Escherichia coli is often hampered by their failure to fold correctly, leading to their accumulation within inclusion bodies. To overcome the problem, a variety of techniques aimed at soluble expression have been developed including low temperature expression and/or fusion of soluble tags and chaperones. However, a general protocol for bacterial expression of disulfide bond-containing proteins has hitherto not been established. Single chain Fv fragments (scFvs) are disulfide bond-containing proteins often difficult to express in soluble forms in E. coli. We here examine in detail the E. coli expression of a scFv originating from an anti-carbohydrate MLS128 antibody as a model system. We combine three techniques: (1) tagging scFv with thioredoxin, DsbC and protein disulfide isomerase (PDI), (2) expressing the proteins at low temperature using the pCold vector system, and (3) using Origami E. coli strains with mutations in the thioredoxin reductase and glutathione reductase genes. We observed a high expression level of soluble MLS128-scFv in the Origami strain only when PDI is used as a tag. The recombinant protein retains full binding activity towards synthetic carbohydrate antigens. The developed "pCold-PDI" vector has potential for overproduction of other scFvs and disulfide-containing proteins in the Origami strains.  相似文献

采用重组PCR技术获得抗多药耐药相关蛋白3(multidrug resistance protein 3, MRP3)的单链抗体(scFv)与人源可溶性肿瘤坏死因子相关凋亡诱导配体(soluble TNF-related apoptosis inducing ligand, sTRAIL)的融合蛋白质的基因编码序列, 利用原核表达载体pMAL-c2,构建含麦芽糖结合蛋白(maltose binding protein, MBP)标签肽的antiMRP3(scFv)-sTRAIL融合蛋白, 经亲和层析柱纯化. 获得纯化的antiMRP3(scFv)-sTRAIL融合蛋白,用MRP3阳性U251多形性胶质母细胞瘤做增殖抑制实验、细胞凋亡诱导实验,结果均显示具有明显的活性, 而MBP无明显作用. 上述结果表明,成功表达了antiMRP3(scFv)-sTRAIL融合蛋白, 该融合蛋白具有诱导U251多形性胶质母细胞瘤细胞凋亡的活性, 为开发靶向性抗肿瘤药物奠定了基础.  相似文献

为了建立一种核酸酶P1(Nuclease P1,NP1)的原核表达纯化系统,首先采用重叠延伸PCR将22段寡核苷酸拼接,获得人工合成的NP1基因。将其克隆至分泌型表达载体pMAL-p4X获得重组质粒pMAL-p4X-NP1,然后将重组载体转化T7 Express和Origami B(DE3)菌株诱导表达,利用Amylose亲和层析柱纯化获得重组蛋白,并对其活性、热稳定性和金属离子依赖性进行系统分析。SDS-PAGE结果显示,重组蛋白MBP-NP1(Maltose binding protein-NP1)在T7 Express和Origami B(DE3)菌株中均可表达,且以可溶性形式存在。活性检测表明Origami B(DE3)菌株中获得的重组蛋白活性高于T7 Express菌株(75.48 U/mg:51.50 U/mg);利用蛋白酶Factor Xa切除MBP标签后,两种重组蛋白的比活力均有提高,分别为258.13 U/mg和139.20 U/mg。重组NP1表现出良好的热稳定性,80℃温浴30 min后重组酶仍具有90%以上的活力。2.0 mmol/L Zn2+对NP1有比较明显的激活作用,相同浓度的Cu2+则对该酶有强烈的抑制作用。该研究实现了NP1在大肠杆菌系统中的功能性表达,为NP1纯酶的制备提供一个替代途径。  相似文献

Divercin V41, a class IIa bacteriocin with strong antilisterial activity, is produced by Carnobacterium divergens V41. To express a recombinant version of divercin V41, we constructed a synthetic gene that encodes the mature divercin V41 peptide and then overexpressed the gene in pET-32b by using the T7 RNA polymerase promoter in the Escherichia coli Origami (DE3)(pLysS) strain. The DvnRV41 peptide was expressed as a translational fusion protein with thioredoxin and accumulated in the cell cytoplasm in a soluble anti-Listeria active form. The fusion protein was then purified and cleaved to obtain pure, soluble, folded DvnRV41 (462 microg per 20 ml of culture). This paper describes the first design of a synthetic bacteriocin gene and the first bacteriocin expressed in the E. coli cytoplasm.  相似文献

Many eukaryotic proteins have been produced successfully in Escherichia coli. However, not every gene can be expressed efficiently in this organism. Most proteins, especially those with multiple disulfide bonds, have been shown to form insoluble protein or inclusion body in E. coli. An inactive form of protein would require an in vitro refolding step to regain biological functions. In this study, we described the system for soluble expression of a single-chain variable fragment (scFv) against hepatocellular carcinoma (Hep27scFv) by coexpressing Dsb protein and enhancing with medium additives. The results revealed that overexpression of DsbABCD protein showed marked effect on the soluble production of Hep27scFv, presumably facilitating correct folding. The optimal condition for soluble scFv expression could be obtained by adding 0.5M sorbitol to the culture medium. The competitive enzyme-linked immunosorbent assay (ELISA) indicated that soluble scFv expressed by our method retains binding activity toward the same epitope on a hepatocellular carcinoma cell line (HCC-S102) recognized by intact antibody (Ab) (Hep27 Mab). Here, we report an effective method for soluble expression of scFv in E. coli by the Dsb coexpression system with the addition of sorbitol medium additive. This method might be applicable for high-yield soluble expression of proteins with multiple disulfide bonds.  相似文献

DNA encoding a His-tagged 11S globulin from Amaranthus hypochondriacus (amarantin) was successfully expressed in Escherichia coli strains BL21 (DE3) and Origami (DE3). The two strains produced different accumulation patterns. Whereas most of the proamarantin expressed in BL21 (DE3) was localized in inclusion bodies, that produced in Origami (DE3) was soluble (76 mg/L). Sucrose density gradient ultracentrifugation analysis of the expressed soluble proamarantin revealed that the protein was assembled into trimers. Treatment of proamarantin trimers in vitro using purified asparaginyl endopeptidase resulted in the appearance of peptides of the sizes expected for acidic and basic chains. Because the proamarantin assembles into trimers with the expected sedimentation characteristics and is cleaved into acidic and basic chains rather than being degraded, the results suggest that the protein folding occurring in E. coli is similar to that taking place in seeds. The His-tagged proamarantin was purified in a single step by immobilized metal affinity chromatography with a final yield of 48 mg/L. The overexpression of proamarantin in E. coli, together with the one-step purification will facilitate further investigation of this storage protein through site-directed mutagenesis.  相似文献

The effects of subcellular localization on single-chain antibody (scFv) expression levels in transgenic tobacco was evaluated using an scFv construct of a model antibody possessing different targeting signals. For translocation into the secretory pathway a secretory signal sequence preceded the scFv gene (scFv-S). For cytosolic expression the scFv antibody gene lacked such a signal sequence (scFv-C). Also, both constructs were provided with the endoplasmic reticulum (ER) retention signal KDEL (scFv-SK and scFv-CK, respectively). The expression of the different scFv constructs in transgenic tobacco plants was controlled by a CaMV 35S promoter with double enhancer. The scFv-S and scFv-SK antibody genes reached expression levels of 0.01% and 1% of the total soluble protein, respectively. Surprisingly, scFv-CK transformants showed considerable expression of up to 0.2% whereas scFv-C transformants did not show any accumulation of the scFv antibody. The differences in protein expression levels could not be explained by the steady-state levels of the mRNAs. Transient expression assays with leaf protoplasts confirmed these expression levels observed in transgenic plants, although the expression level of the scFv-S construct was higher. Furthermore, these assays showed that both the secretory signal and the ER retention signal were recognized in the plant cells. The scFv-CK protein was located intracellularly, presumably in the cytosol. The increase in scFv protein stability in the presence of the KDEL retention signal is discussed.  相似文献

探讨大肠杆菌细胞质氧化还原环境对重组蛋白溶解性的影响。选择含有1对二硫键的牛碱性成纤维细胞生长因子(BbFGF)作为简单蛋白的模式分子,选择含有2对二硫键的人抗HBsAg单链抗体(HBscFv)作为复杂蛋白的模式分子,分别构建表达质粒并转化普通宿主菌和还原酶缺陷型宿主菌E. coli Origami(DE3),比较表达产物的溶解性和纯化产物的活性。结果发现,BbFGF在普通宿主菌中大部分形成包涵体,在Origami(DE3)中为可溶性表达,但表达量降低。两种工程菌的表达产物经离子交换和肝素亲和层析两步纯化后,MTT法测定活性,发现来自还原酶缺陷型宿主菌的BbFGF活性高于普通宿主菌表达产物,二者的ED50分别是1.6 ng/mL和2.2 ng/mL;HBscFv在两种宿主菌中均形成包涵体,包涵体以6 mol/L盐酸胍缓冲液溶解后,镍离子螯合亲和层析纯化并透析复性,间接ELISA测定抗原结合活性,发现二者活性无明显差异,但在Origami(DE3)菌体破碎后的的上清中可检测到HBscFv活性,纯化后产量为1~2 mg/L,而在普通宿主菌破碎后的上清中检测不到HBscFv活性。上述结果说明,改变宿主菌细胞质氧化还原环境对于含有1~2对二硫键的重组蛋白的可溶性表达具有明显促进作用。  相似文献

Amarantin acidic subunit has the potential to be employed as a functional and a nutraceutical protein. To evaluate both possibilities this protein was produced in recombinant Escherichia coli Origami (DE3) harboring the expression plasmid pET-AC6His. Three different expression factors were assayed: inductor concentration, temperature and time of the amarantin acidic subunit accumulation. The results indicated that a 0.3 mmol/L concentration of isopropyl-beta-D-thiogalactoside, at 37 degrees C and 6 h after induction were favorable for high expression of amarantin acidic subunit, mostly in the form of inclusion bodies. The protein was purified from soluble fraction by immobilized metal affinity chromatography, up to 30 mg amarantin acidic subunit/L Terrific broth culture were obtained. Sucrose density gradient ultracentrifugation analysis of the expressed soluble amarantin acidic subunit revealed that it was assembled in monomers. The expression of the amarantin acidic subunit, together with the one-step purification will facilitate further investigation of this storage protein through site-directed mutagenesis.  相似文献

用人肝癌细胞系SMMC7721免疫小鼠,提取脾细胞总RNA,构建单链抗体库,从中筛选到1个与人肝癌细胞系HepG2特异性结合的噬菌体-单链抗体。此单链抗体与HepG2细胞结合滴度比正常肝细胞低100倍以上。构建表达质粒pTrx-scFv5-56,单链抗体scFv5-56在大肠杆菌BL21(DE3)中成功地进行了可溶性表达。  相似文献

目的:构建PET-28a-SPA原核表达载体,在大肠杆菌BL21(DE3)中实现其高效可溶性表达,测定对肿瘤细胞的凋亡效果。方法:本实验在获得凋亡蛋白融合基因的基础上,成功地构建了重组表达质粒PET-28a-SPA,将阳性重组质粒转化表达受体菌BL21(DE3)感受态细胞中,经IPTG诱导表达,表达产物经聚丙烯酰胺凝胶电泳检测和Western blot检测,并采用MTT法检测其对肿瘤细胞的增殖抑制。结果:表达产物经聚丙烯酰胺凝胶电泳检测,凋亡蛋白融合基因获得高效表达,软件分析表明表达蛋白占菌体蛋白20%左右。上清表达量约为10%。上清蛋白经纯化后,Western blot结果显示,利用凋亡蛋白单克隆抗体可以很好地和所表达的蛋白带特异性结合,并且对A549肺癌细胞及Hela细胞具有一定的凋亡作用。结论:所获凋亡蛋白以高效胞质可溶形式表达,为其研制有效的肿瘤免疫治疗靶向药物提供一定的基础。  相似文献
