The diversity of freshwater bacterioplankton communities has not been extensively studied despite their key role in foodwebs and the cycling of carbon and associated major elements. In order to explore and characterize the composition of bacterioplankton associated with cyanobacterial blooms, large 16S rRNA clone libraries from four lakes experiencing such blooms were analysed. The four libraries contained 1461 clones, of which 559 were prokaryotic sequences of non-cyanobacterial origin. These clones were classified into 158 operational taxonomic units affiliated mainly with bacterial divisions commonly found in freshwater systems, e.g. Proteobacteria, Bacteriodetes, Actinobacteria, Verrucomicrobia and Planctomycetes. Richness and evenness of non-cyanobacterial clones were similar to other clone libraries obtained for freshwater bacterioplankton, suggesting that bacterial communities accompanying cyanobacterial blooms are as diverse as non-bloom communities. Many of the identified operational taxonomic units grouped with known freshwater clusters but the libraries also contained novel clusters of bacterial sequences that may be characteristic for cyanobacterial blooms. About 25% of the operational taxonomic units were detected in more than one lake. Even so, 16S rRNA heterogeneity analysis demonstrated large differences in community composition between lakes regardless of their similar characteristics and close proximity. Hence even the similar environmental conditions created by different cyanobacterial blooms may foster very dissimilar bacterial communities, which could indicate that the genetic diversity in lake bacteria have been underestimated in the past.  相似文献

We characterized the bacterioplankton community and its seasonal dynamics in two neighbouring hypertrophic lakes by denaturing gradient gel electrophoresis (DGGE) analysis of short (193 bp) 16S ribosomal DNA polymerase chain reaction (PCR) products obtained with primers specific for the domain Bacteria. Lake Blankaart is turbid and has a high phytoplankton biomass and episodic cyanobacterial blooms, whereas biomanipulated Lake Visvijver is characterized by clearwater conditions and the establishment of a dense charophyte vegetation. Both lakes were dominated by bacterial groups commonly found in freshwater habitats (e.g. ACK4 cluster of Actinomycetes; ACK stands for clones isolated from the Adirondack mountain lakes) . Yet, cluster analysis and principal components analysis (PCA) revealed that taxon composition of the bacterioplankton community of the two lakes differs substantially and consistently throughout the season. During the study year (1998), the bacterioplankton community of both lakes showed a distinct seasonal pattern. Lake Blankaart showed a clear differentiation between winter, spring, summer and autumn. In Lake Visvijver, summer samples differed greatly from spring, autumn and winter samples. We hypothesize that the contrasting bacterioplankton in the two neighbouring shallow lakes is determined largely by the presence or absence of macrophytes.  相似文献

The contribution of bacterioplankton to total plankton respirationwas measured in two eutrophic Danish lakes and in experimentalenclosures treated with planktivorous fish and nutrients. Bacterialrespiration was calculated from measured oxygen uptake ratesin particles passing a 1.0-µm pore size filter, the rateswere then corrected for the size distribution of glucose uptake.During summer the respiration of the planktonic bacteria contributed{small tilde}50% of the community respiration in the two lakes.Prolific phytoplankton growth induced by biomanipulation andnutrient addition created situations where the contributionof the bacteria decreased to 20%. High bacterial contributionsto community respiration were found when the phytoplankton biomassdecreased. Simultaneous measurements of bacterial respirationand production (by means of [3H]thymidine incorporation) allowedan estimation of bacterial growth yield, which ranged from 9to 66%. In the two lakes the growth yield was constant witha mean of 29 ± 5% (±SD, RQ = 1). The variabilityof the growth yield was larger in the enclosures. The wide range(9–66%) was mainly caused by changes in bacterial netproduction without concomitant changes in respiration. The discussionincludes an evaluation of the oxygen uptake method in size fractionatedsamples and the availability of labile organic substrates asa factor controlling bacterial growth yield. Genetic diversity of cyanobacteria in four eutrophic lakes   总被引:1,自引:0,他引:1  
Recent studies indicate genetic diversity of cyanobacteria in eutrophic lakes is not represented well by culture collections or morphology. Yet, few studies have investigated genetic richness and evenness of cyanobacteria using culture-independent methods. We compared the genetic structure of cyanobacteria supported by four neighboring eutrophic lakes during the ice-free season. The partial phycobilincpcB/A genes plus intergenic spacer (PC-IGS) was used as a genetic marker.Sequences were phylogeneticallygrouped by maximum likelihood into genotypes representing sub-genera of the major taxa. Genotypes fell into genera commonly observed by microscopy in these lakes including Microcystis, Aphanizomenon, Chroococcus, Anabaena, and Cylindrospermopsis. Only three genotypes were shared among all four lakes, despite significant water flowage between lakes.A Parsimony P-test indicated lakes were significantly (p=0.01) clustered on the maximum likelihood tree. Pairwise differences using Unifrac distance were moderately or not significant. Analysis of molecular variance (AMOVA) indicated genetic variation among all genotypes (φ=0.06, p<0.001) and 94% of variability occurred within lakes rather than between lakes (6%), explaining the lack of pairwise differences between lakes. Lorenze curves of genotype abundance in each lake showed genetic structure was only moderately uneven (Gini coefficients of 0.37-0.5) indicating lakes did not support dominant genotypes. Overall, results from this study suggest diversity of cyanobacteria is shaped by heterogeneity within lakes (temporally or spatially) and relatively even population structures.  相似文献

Microcystin concentrations in two Dutch lakes with an important Planktothrix component were related to the dynamics of cyanobacterial genotypes and biovolumes. Genotype composition was analysed by using denaturing gradient gel electrophoresis (DGGE) profiling of the intergenic transcribed spacer region of the rrn operon (rRNA-ITS), and biovolumes were measured by using microscopy. In Lake Tjeukemeer, microcystins were present throughout summer (maximum concentration 30 microg l(-1)) while cyanobacterial diversity was low and very constant. The dominant phototroph was Planktothrix agardhii. In contrast, Lake Klinckenberg showed a high microcystin peak (up to 140 microg l(-1)) of short duration. In this lake, cyanobacterial diversity was higher and very dynamic with apparent genotype successions. Several genotypes derived from DGGE field profiles matched with genotypes from cultures isolated from field samples. The microcystin peak measured in Lake Klinckenberg could be confidently linked to a bloom of Planktothrix rubescens, as microscopic and genotypic analysis showed identity of bloom samples and a toxin-producing P. rubescens culture. Toxin-producing genotypes were detected in the microbial community before they reached densities at which they were detected by using microscopy. Cyanobacterial biovolumes provided additional insights in bloom dynamics. In both lakes, the microcystin content per cell was highest at the onset of the blooms. Our results suggest that while genotypic characterization of a lake can be valuable for detection of toxic organisms, for some lakes a monitoring of algal biomass has sufficient predictive value for an assessment of toxin production.  相似文献

With an unprecedented decade-long time series from a temperate eutrophic lake, we analyzed bacterial and environmental co-occurrence networks to gain insight into seasonal dynamics at the community level. We found that (1) bacterial co-occurrence networks were non-random, (2) season explained the network complexity and (3) co-occurrence network complexity was negatively correlated with the underlying community diversity across different seasons. Network complexity was not related to the variance of associated environmental factors. Temperature and productivity may drive changes in diversity across seasons in temperate aquatic systems, much as they control diversity across latitude. While the implications of bacterioplankton network structure on ecosystem function are still largely unknown, network analysis, in conjunction with traditional multivariate techniques, continues to increase our understanding of bacterioplankton temporal dynamics.  相似文献

Summary Food resources in the environment and in the diets of small fish inhabiting two water bodies in a tropical savanna were studied during both wet and dry seasons. During the wet season (high water, abundant food) most fish species in both habitats fed predominantly on vegetation-dwelling invertebrates. Most fish species switched to alternative foods (algae and detritus) following the drastic decline in invertebrate food available towards the end of the dry season. In one habitat, this change in diet was accompanied by an increase in the volume of food intake. In the second habitat, only two larger species foraged intensively, while smaller species showed low food intake or almost ceased feeding. These differences may be explained by the high risk of predation for small fish in the second habitat. Dietary overlaps among fish species were high at the end of the dry season and moderate in the wet season. However, critical analysis of such factors as food abundance, the size and number of shared prey, and diet breadth showed that all significant overlaps were ecologically unimportant i.e. there was only weak competition for food.  相似文献

The population dynamics of Daphnia species (D.cucullata, D.ambigua,D.galeata) in two small, productive lakes were studied and experimentswith field enclosures were carried out to elucidate the ecologicalniches of the introduced, alien species D.ambigua. Daphnia ambiguocoexisted temporally with the similarly sized D.cucullata, butlived in deeper water. Although both species had the same clutchsizes, similar proportions of ovigerous females, and death rates.D.cucullata was nurnerically dominant over D.ambigua. This suggeststhat the observed difference in abundance is simply a resultof the slower population growth rates of D.ambigua at lowertemperatures in greater depths. The experiments with field enclosuresshowed that when D.culcullata and D.ambigua were forced to livein the same layer of water, D.ambigua was numerically dominantover D.culullata both in the enclosures with and without fish.As the difference between the abundances of the two speciesdecreased in the enclosure with fish, D.cucullata seems to bebetter protected against fish predation than D.ambigua. In theenclosures without fish, D.ambigua and D.cucullata were numericallysurpassed by the larger species D.galeara. SUMMARY. Changes in bacterial populations and certain physical and chemical variables in Esthwaite Water between June and September 1975 were studied and compared with results obtained from 1972 to 1974 in the hypolimnia of Blelham Tarn and the Lund tubes. The counts of total bacteria ranged between 1 and 7 × 106ml−1 and were highest in the anoxic hypolimnion. The bacterial genera examined in more detail constituted only a small percentage of this count and included Ochrobium (104ml−1), Naumanniella (103ml−1), Leptothrix (102ml−1), Planctomyces (103ml−1), and Metallogenium (102ml−1). The iron bacteria appear to grow best in the oxycline where there was not only sufficient oxygen for aerobic growth but also a plentiful supply of reduced iron. Planctomyces numbers increased as the thermocline became depressed in September. The results from Blelham Tarn might be interpreted as further evidence of growth by iron bacteria in the absence of dissolved oxygen, but other explanations are possible. Examination of the results by multiple regression analysis showed that it was possible to explain a significant proportion of the bacterial variation (with the notable exception of the Planctomyces counts) in spite of considerable intercorrelation of the regressor variables.  相似文献

The seasonal variations of limnological (water temperature, light availability, turbidity, and chlorophyll a concentration) parameters were recorded continuously from January 2004 to February 2005 at two freshwater lakes: Oyako-ike and Hotoke-ike, Sôya Coast, East Antarctica. Water was in a liquid phase throughout the year, with temperatures ranging from 0 to 10°C. The maximum photosynthetically active radiation in Lake Oyako-ike was 23.16 mol m?2 day?1 (at 3.8 m) and Hotoke-ike was 53.01 mol m?2 day?1 (at 2.2 m) in summer, and chlorophyll a concentration ranged from ca. 0.5 to 2.5 μg L?1 (Oyako-ike) and from ca. 0.1 to 0.8 μg L?1 (Hotoke-ike) during the study period. Increase in chlorophyll a fluorescence occurred under dim-light conditions when the lakes were covered with ice in spring and autumn, but the signals were minimum in ice-free summer in both the lakes. During spring and summer, as a result of decreasing snow cover, the chlorophyll a concentration similarly decreased when PAR was relatively high, following periods of heavy winds. The autumnal and spring increase occurred under different PAR levels (ca. 20-fold and 90-fold stronger, respectively, in autumn in both the lakes). Differences in the autumn and spring increases suggest that the spring algal community is more shade-adapted than the autumn algal community. Antarctic phytoplankton appears especially adapted to low-light levels and inhibited by strong light regimes.  相似文献

Despite research demonstrating that water and nutrient availability exert strong effects on multiple ecosystem processes in tropical forests, little is known about the effect of these factors on the demography and population dynamics of tropical trees. Over the course of 5 years, we monitored two common Amazonian secondary forest species—Lacistema pubescens and Myrcia sylvatica—in dry-season irrigation, litter-removal and control plots. We then evaluated the effects of altered water and nutrient availability on population demography and dynamics using matrix models and life table response experiments. Our results show that despite prolonged experimental manipulation of water and nutrient availability, there were nearly no consistent and unidirectional treatment effects on the demography of either species. The patterns and significance of observed treatment effects were largely dependent on cross-year variability not related to rainfall patterns, and disappeared once we pooled data across years. Furthermore, most of these transient treatment effects had little effect on population growth rates. Our results suggest that despite major experimental manipulations of water and nutrient availability—factors considered critical to the ecology of tropical pioneer tree species—autogenic light limitation appears to be the primary regulator of tree demography at early/mid successional stages. Indeed, the effects of light availability may completely override those of other factors thought to influence the successional development of Amazonian secondary forests.  相似文献

In the context of this study two concepts were applied for the development of rule-based agents of algal populations: (1) rule discovery by means of a hybrid evolutionary algorithms (HEA) and rigorous k-fold cross-validation, and (2) rule generalisation by means of merged time-series data of lakes belonging to the same lake category. The rule-based agents developed during this study proved to be both explanatory and predictive. It has been demonstrated that the interpretation of the rules can be brought into the context of empirical and causal knowledge on chlorophyll-a dynamics as well as population dynamics of Microcystis and Oscillatoria under specific water quality conditions. The k-fold cross-validation of the agents based on measured data of each year of similar lakes revealed good forecasting accuracy resulting in r2 values ranging between 0.39 and 0.63.  相似文献

Tropical macrophytes sold in the live garden trade are perceived as unlikely to invade temperate regions owing to climate mismatches. Here we study two tropical macrophytes (Pistia stratiotes and Eichhornia crassipes) not previously considered an invasion risk but which were recently discovered in the Great Lakes, and determine mechanisms that may be responsible for their continued presence including human introduction, reproduction through viable seeds and tolerance of winter conditions. Surveys conducted in 2011 and 2012 revealed recurrent presence of one or both species at some sites. Macrophytes in in situ enclosures failed to survive winter conditions, with plant health declining progressively prior to mortality. Water hyacinth seeds were field-collected, identified using Sanger sequencing, and germinated at 28°C with or without scarification. Germination was highest for scarified versus non-scarified seeds. Human introduction was observed at two sites, one involving both species, the other only water hyacinth. These species likely persist through a combination of annual reintroduction (both species) and possibly by production of viable seed (water hyacinth). Macrophytes, particularly water hyacinth, that were not previously viewed as a threat to the Great Lakes owing to environmental incompatibility may need to be reassessed.  相似文献

The bacterioplankton assemblages of eight maritime Antarctic lakes with a wide range of trophic status and geographic span (six lakes from Hope Bay, Antarctic Peninsula and two from Potter Peninsula, King George Island) were described using denaturing gradient gel electrophoresis and band sequencing during two consecutive austral summers (2003–2004). Analyses of the gels identified a total of 230 bands spread across 57 different positions. Among those bands, 14 were shared between lakes from Hope Bay and Potter Peninsula, 17 were observed only in particular lakes, and 17 were registered both years in the same lake. We successfully reamplified and sequenced 43 bands located in 36 different positions belonging to Bacteroidetes, Actinobacteria, Betaproteobacteria and Cyanobacteria. The closest matches for 63% of the sequenced bands were from Antarctic or from other cold environment clones and sequences already in the databases, suggesting the widespread dominance of microbial communities adapted to cold habitats. The results of the multivariate analyses (Cluster Analysis and CCA) indicated that the nutrient status of the lake influences the bacterioplankton assemblages. This article explores factors that shape population structure in novel environments that have received scant theoretical attention: cities. Urban bird populations exhibit higher densities and lower diversity. Some work suggests this may result from lower predation pressure and more predictable and abundant resources. These factors may lead to populations with few winners and many losers regarding access to food, body condition, and reproductive success. We explore these hypotheses with an individual-energy-based competition model with two phenotypes of differing foraging ability. We show that low frequency resource fluctuations favor strong competitors and vice versa. We show that low predation skews equilibrium populations in favor of weak competitors and vice versa. Increasing the time between resource pulses can thus shift population structure from weak to strong competitor dominance. Given recent evidence for more constant resource input and lower predation in urban areas, the model helps understand observed urban bird population structure.  相似文献

1. The significance of microbial diversity in processing dissolved organic matter (DOM) is largely unknown. We investigated the range of functional diversity in the bacterioplankton from a eutrophic river by profiling extracellular enzyme activities (EEA) and substrate-induced respiration (SIR) patterns.
2. The EEA profiles consisted of assays for 21 hydrolases, measured using fluorogenic substrates arrayed on 96-well microplates. Commercially available BiOLOG® GN and ECO plates, which contained 120 different substrates, were used for the SIR profiles.
3. The EEA data were more dynamic than the SIR. Five enzymes, leucine aminopeptidase, alkaline phosphatase, alanine aminopeptidase, arginine aminopeptidase and β-glucosidase, showed consistently high activity; ten others were ubiquitous at lower activity levels; the remainder were detected intermittently. The SIR data showed less temporal variability. With one exception (citrate), the 20 substrates that generated the largest responses were all saccharides or their derivatives.
4. The EEA and SIR data did not generally correlate. Both methods were effective for ordinating bacterioplankton although, unlike the SIR, the EEA ordination followed a clear temporal trajectory.
5. Because the SIR profiles are based on a culture response, whilst the EEA profiles measure activity of the extant community, the latter appear to be more directly linked to the mechanics of DOM processing.  相似文献

Zeekoevlei is the largest freshwater lake in South Africa and has been suffering from hyper-eutrophic conditions since last few decades. We have used total P (TP), dissolved phosphate (PO4 3−), organic P (OP), calcium (Ca) and iron (Fe) bound P fractions to investigate the relevant physical, chemical and biological processes responsible for sedimentation and retention of P and to study phosphorus (P) dynamics in this shallow lake. In addition, redox proxies (V/Cr and Th/U ratios) are used to study the prevailing redox conditions in sediments. Adsorption by CaCO3 and planktonic assimilation of P are found to control P sedimentation in Zeekoevlei. Low concentration of the labile OP fraction in surface sediments restricts the release of P by bacterial remineralisation. Low molar Ca/P and Fe/P ratios indicate low P retention capacity of sediments, and P is most likely released by desorption from wind-induced resuspended sediments and mixing of pore water with the overlying water column. Ciliate community composition was investigated in four subtropical eutrophic lakes with cyanobacterial blooms in three seasons during 2009–2010 using the quantitative protargol staining approach. Among the four lakes, Lake Taihu and Chaohu are situated in the mid-lower Yangtze Plain near sea level, and Lake Dianchi and Xingyunhu are located on the Yunnan–Kweichow Plateau. A total of 102 ciliate species, representing 15 orders and 66 genera, were studied. Ciliate communities in Lake Taihu and Chaohu were characterized by abundant algivorous and mixotrophic oligotrichs and prostomatids, while Lake Dianchi and Xingyunhu ciliate communities were primarily characterized by voracious bacterivorous scuticociliates and peritrichids. Cluster analysis based on the transformed square root of ciliate abundance showed that samples from Lake Taihu and Chaohu composed one group whereas samples from Lake Dianchi and Xingyunhu composed another group in each season. Our results indicated that all dominant ciliate species are ubiquitous and that species dominance is regulated by different factors in different subtropical eutrophic lakes.  相似文献

The aim of this work was to test the efficacy of molecular techniques for detecting toxigenic cyanobacteria in environmental water samples collected from freshwater lakes, rivers and reservoirs in Portugal. Of 26 environmental samples tested, 21 were found to contain Microcystis using a genus-specific polymerase chain reaction (PCR). Another primer pair was applied to the same DNA template to test for the presence of microcystin synthetase genes. This primer pair resulted in the formation of a PCR product in 15 of the samples containing Microcystis and one sample that did not give a positive result in the Microcystis genus-specific PCR. A restriction assay using the enzyme EcoRV was then applied to show that in most cases, the gene fragment was from toxigenic strains of Microcystis and, in one above-mentioned case, from a microcystin-producing strain of Planktothrix. All environmental samples were examined microscopically to confirm the presence of cyanobacteria species. Samples were also tested for the presence of microcystins using the ELISA plate assay. There was good agreement between the results obtained with molecular techniques and those obtained from microscopy and chemical methods. The PCR techniques applied in this paper were found to be useful, particularly when the concentration of the target organism was very low compared with other organisms. This technique can be used to detect inocula for cyanobacterial populations and therefore provide a useful tool for assessing under which conditions particular species can grow into bloom populations.  相似文献
