A crosslinked dinucleoside, 1,2-(diguanosin-7-yl) ethane, has been isolated from the reaction of guanosine with the antitumor agent, BCNU. The formation of this product suggests that DNA crosslinking, which may be responsible for the cytotoxicity of BCNU, could occur through such dinucleosides.  相似文献

BCNU has been reacted with polycytidylic acid and two derivatives of CMP, 3-hydroxyethyl-CMP and 3,N4-ethano-CMP, have been identified in the acid hydrolysate of the polymer. Their formation accounts for some of the reaction of BCNU with nucleic acids, and may be related to the mechanism of action of this compound.  相似文献

BCNU, labelled with 14C in the chloroethyl groups, decomposes in neutral aqueous solution to release half of its radioactivity as volatile products. These have been identified by a combination of gas chromatography and mass spectrometry as vinyl chloride, acetaldehyde, dichloroethane, and chloroethanol. This set of products is consistent with the existence of chloroethylcarbonium ions as reactive intermediates which would produce the previously described substituted nucleotides.  相似文献

The heterocyclic amino-compounds 11a, 13a, 13b, and 17 reacted with formaldehyde and p-thiocresol (14) in alcoholic solution to give the corresponding N-methylphenylthiomethyl derivatives (16, 15a, 15b, and 18a, respectively) in satisfactory to good yields. The reactions were catalyzed by acetic acid. 2-N-Methylguanosine (6a) was obtained in good yield by treatment of 15b with sodium borohydride followed by acidic hydrolysis, or alternatively by Raney nickel desulfurization of 15a followed by ammonolysis of the product. Sodium borohydride reduction of 18a gave 21 in good yield. 2-N,2-N-Dimethylguanosine (6b) was obtained from 19a in three steps.  相似文献

When 32Pi-labeled rabbit platelets were incubated with 5 × 10?10m 1-O-alkyl-2-acetyl-sn-glyceryl-3-phosphorylcholine (AGEPC), either in the presence or absence (0.1 mm EGTA) of added Ca2+, there was a three- to five-fold increase in the [32P]phosphatidic acid (PA) pool within 15 to 20 s. This event was followed by a gradual decrease in the [32P]PA level to near basal level in 5 min. AGEPC effected this change in [32P]PA in a characteristic dose- and time-dependent manner. Polar head group analogs of AGEPC, such as AGEDME and AGEMME, also effected an increase in PA labeling at levels. comparable to those previously reported for their activity toward rabbit platelets [K. Satouchi, R. N. Pinckard, L. M., McManus, and D. J. Hanahan (1981) J. Biol. Chem.256, 4425–4432]. Other analogs, i.e., lysoGEPC and the enantiomer, sn-1-AGEPC, which are inactive toward rabbit platelets, also showed no effect on the level of [32P]PA. The finding that the PA level in rabbit platelets could be manipulated by the addition of AGEPC, without any added Ca2+, provided an excellent model system for establishinig a correlation between the uptake of Ca2+, serotonin release, and PA level. Thus, PA must be regarded as a sensitive indicator of a reaction mechanism important to the platelet response to AGEPC, and could be the focal point in promoting calcium uptake during the stimulation process.  相似文献

Influences of dithiothreitol (DTT), p-chloromercuriphenyl sulfonate (PCMPS) and ascorbate on CuCl2-induced elevation of [3H]cimetidine binding were investigated in brain membranes of rats. CuCl2 (10–500 μM) elevated specific [3H]cimetidine binding in a concentration-dependent manner. There were two types of [3H]cimetidine binding in the presence of 50 μM CuCl2: high affinity binding with Kd = 1.97 nM and low affinity with Kd = 21.6 nM. PCMPS (10 and 100 μM) reduced the binding in both media with and without CuCl2. DTT (1–30 μM) or ascorbate (0.1 and 1.0 mM) markedly elevated the binding in the presence of CuCl2 but showed no effect and ascorbate rather inhibited the binding in the absence of CuCl2. DTT (0.1 mM) diminished the binding in the presence and absence of CuCl2. CuCl2 (50 μM) significantly (P < 0.01) increased the IC50 of histamine for [3H]cimetidine binding and the effect was greater than that from 100 μM GTP. It is suggested that sulfhydryl groups sensitive to PCMPS could interact with Cu2+ and thus be involved in an elevation of cimetidine binding. Cu2+ seems to regulate affinity of agonist binding for cimetidine binding sites presumably by acting on cimetidine binding sites and/or GTP binding regulatory proteins.  相似文献

The release of [3H]GABA which is newly synthesized from [3H]l-glutamic acid (GLU) has been examined using striatal slices obtained from the rat brain. It was found that 8–10% of [3H]GLU transported was converted to [3H]GABA during the incubation of striatal slices in the presence of nipecotic acid (5 × 10?5 M). Nipecotic acid was added to the medium in order to prevent possible reuptake of [3H]GABA released during its synthesis, and it was found to have no significant effect on the formation of [3H]GABA from [3H]GLU as well as on the uptake of [3H]GLU. The application of high potassium (60 mM) stimulation exhibited a significant enhancement of the release of this newly synthesized [3H]GABA in a Ca2+ dependent manner. Kinetic analysis revealed that the evoked release of newly synthesized [3H]GABA was approximately two times greater than that of previously-loaded [3H]GABA, whereas no significant difference was observed in the spontaneous release. An immobilization stress in water failed to affect the release of newly synthesized [3H]GABA from striatal slices despite the occurrence of a significant enhancement of GABA formation in this structure.These results suggest that newly synthesized GABA may be preferentially released from its nerve terminals in response to the excitation of neurons at least in the striatum as compared with previously accumulated GABA.  相似文献

The crystal structure of hexaaquamanganese(II) bis{bis(N-salicylideneglycinato)manganate(III)} dihydrate has been determined by X-ray analysis. The complex crystallizes in the monoclinic space group I2|a, with unit-cell dimensions a = 37.431(5), b = 12.100(1), c = 9.448(1) Å, β = 92.31(1)°. The structure was deduced by the direct method and refined by the block-diagonal least-squares technique to a final R value of 0.062 for 3904 observed reflections. The Mn(II) is octahedrally ligated by six water molecules, while Mn(III) is octahedrally chelated by two salicylideneglycinate ligands, of which one is nearly planar and the other considerably bent.It was discovered that the crystal is, as a whole, of a ‘sandwich’ structure made of one central sheet containing hexaaquamanganese(II)'s and the water molecules of crystallization, and two outside sheets containing bis(N-salicylideneglycinato)manganate(III)'s.  相似文献

A new automated system for the analysis of nitrate via reduction with a high-pressure cadmium column is described. Samples of urine, saliva, deproteinized plasma, gastric juice, and milk can be analyzed for nitrate, nitrite, or both with a lower limit of detection of 1.0 nmol NO3? or NO2?/ml. The system allows quantitative reduction of nitrate and automatically eliminates interference from other compounds normally present in urine and other biological fluids. Analysis rate is 30 samples per hour, with preparation for most samples limited to simple dilution with distilled water. The application of gas chromatography/mass spectrometry for the analysis of 15NO3? in urine after derivatization to 15NO2-benzene is also described.  相似文献

Starting from 2-acetamido-4,6-di-O-acetyl-2-deoxy-3-O-(methyl 2,3,4-tri-O-acetyl-β-D-glucopyranosyluronate)-α-D-glucopyranose (20), a crystalline intermediate prepared by a conventional sequence of reactions, the total synthesis of N-acetyl-hyalobiosyluronic dolichyl diphosphate was achieved. One of the key steps involved the transformation of the disaccharide 20 into the methyloxazoline 26, which was then converted into the stable, crystalline disaccharide phosphate derivative in ~30% yield. The methyloxazoline 26 was directly prepared from the corresponding methyl α-glycoside by acetolysis. Similarly, the allyl α-glycoside was transformed into 26.  相似文献

Ca2+ inhibited the Mg2+-dependent and K+-stimulated p-nitrophenylphosphatase activity of a highly purified preparation of dog kidney (Na+ + K+)-ATPase. In the absence of K+, however, a Mg2+-dependent and Ca2+-stimulated phosphatase was observed, the maximal velocity of which, at pH 7.2, was about 20% of that of the K+-stimulated phosphatase. The Ca2+-stimulated phosphatase, like the K+-stimulated activity, was inhibited by either ouabain or Na+ or ATP. Ouabain sensitivity was decreased with increase in Ca2+, but the K0.5 values of the inhibitory effects of Na+ and ATP were independent of Ca2+ concentration. Optimal pH was 7.0 for Ca2+-stimulated activity, and 7.8–8.2 for the K+-stimulated activity. The ratio of the two activities was the same in several enzyme preparations in different states of purity. The data indicate that (a) Ca2+-stimulated phosphatase is catalyzed by (Na+ + K+)-ATPase; (b) there is a site of Ca2+ action different from the site at which Ca2+ inhibits in competition with Mg2+; and (c) Ca2+ stimulation can not be explained easily by the action of Ca2+ at either the Na+ site or the K+ site.  相似文献

The susceptibility of induced corpora lutea (CL) of prepuberal gilts and spontaneously formed CL of mature gilts to prostaglandin F (PGF) luteolysis was studied. Prepuberal gilts (120 to 130 days of age) were induced to ovulate with Pregnant Mare Serum Gonadotropin and Human Chorionic Gonadotropin (HCG). The day following HCG was designated as Day 0. Mature gilts which had displayed two or more estrous cycles of 18 to 22 days were used (onset of estrus = Day 0). Gilts were laparotomized on Day 6 to 9, their CL marked with sterile charcoal and totally hysterectomized. On Day 20, gilts were injected IM with either distilled water (DW), 2.5 mg PGF or 5.0 mg PGF. An additional group of prepuberal gilts was injected with 1.25 mg PGF, a dose of PGF equivalent, on a per kilogram body weight basis, to the 2.5 mg PGF dose given to the mature gilts. The percentages of luteal regression on Day 27 to 30 for mature and prepuberal gilts given DW, 2.5 mg PGF and 5.0 mg PGF were 0.0 vs 4.4, 43.5 vs 96.8 and 47.7 vs 91.6, respectively; the percentage of luteal regression for the prepuberal gilts given 1.25 mg PGF was 75.1. These results indicate that induced CL of the prepuberal gilt were more susceptible to PGF luteolysis than spontaneously formed CL of the mature gilt and that pregnancy failure in the prepuberal gilt could be due to increased susceptibility of induced CL to the natural luteolysin.  相似文献

The discrete [UO2F4(H20)]2? anion has site symmetry m2m, its virtually linear uranyl moiety being surrounded by fluoro and aquo ligands occupying the vertices of a pentagon in the equatorial plane. Watet molecules serve to link the complex anions by hydrogen bonds into layers, between which the organic cations are accommodated.  相似文献

A gas chromatographic-mass spectrometric method for analysis of L-[2-15N]- and L-[5-15N]glutamine is described. The method is based on direct acylation of glutamine with trifluoroacetic anhydride and the formation of the N,N-bis-trifluoroacetyl-L-glutamine derivative. This simple and sensitive method is capable of detecting approximately 0.5 atom% excess 15N in as little as 10 microliter of plasma with a mean coefficient of variance of 11.6%. The method was applied to determine the appearance of 15N enrichment in plasma amino-N and amide-N of glutamine in a healthy adult volunteer during a constant infusion of 15NH4Cl. A plateau level of 3.7 and 2.6 atom% excess was observed in amide-N and amino-N, respectively, at 1 and 2 h after 15NH4Cl infusion was started.  相似文献

Homoursodeoxycholic acid and [11,12-3H]homoursodeoxycholic acid were synthesized from ursodeoxycholic acid and homocholic acid, respectively. Ursodeoxycholic acid (Ia) was converted to 3α,7β-diformoxy-5β-cholan-24-oic acid (Ib) using formic acid. Reaction of the diformoxy derivative (Ib) with thionyl chloride yielded the acid chloride (II) which was treated with diazomethane to produce 3α,7β-diformoxy-25-diazo-25-homo-5β-cholan-24-one (III). Homoursodeoxycholic acid (IV) was formed from the diazoketone (III) by means of the Wolff rearrangement of the Arndt-Eistert synthesis.N-Bromosuccinimide oxidation of homocholic acid (V), which was prepared from cholic acid by the same procedure described above, afforded 3α,12α-dihydroxy-7-oxo-25-homo-5β-cholan-25-oic acid (VI). Reduction of the 7-ketohomodeoxycholic acid (VI) with sodium in 1-propanol gave 3α,7β,12α-trihydroxy-25-homo-5β-cholan-25-oic acid (VII). The methyl ester of 7-epihomocholic acid (VII) was partially acetylated to give methyl 3α,7β-diacetoxy-12α-hydroxy-25-homo-5β-cholan-25-oate (VIII) using a mixture of acetic anhydride, pyridine and benzene. Dehydration of the diacetoxy derivative (VIII) with phosphorus oxychloride yielded methyl 3α,7β-diacetoxy-25-homo-5β-chol-11-en-25-oate (IX). Reduction of the unsaturated ester (IX) with tritium gas in the presence of platinum oxide catalyst followed by alkaline hydrolysis gave [11,12-3H]homoursodeoxycholic acid.  相似文献

Reaction of phenol with an alkylthiolate-ligated double cubane complex effects phenolate substitution at the terminal positions; the product can be isolated as its benzyltriethylammonium salt. The phenolate cluster possesses unaltered magnetic properties and blue shifted optical spectra, and undergoes ligand exchange reactions with electrophiles as expected for terminal phenolate substitution. Increased isotropic proton NMR shifts and large negative shifts in corresponding first and second reduction potentials are consistent with increased donation of electron density to the [MoFe3S4]3+ cores for phenolate versus thiophenolate terminal ligands to iron. Similar behavior has been observed for Fe4S4, Fe2S2 and MoS2Fe systems.  相似文献

A large dose of dimethylnitrosamine was administered to rats by two different dosing regimens, one being eleven intraperitoneal injections of 5 mg/kg body wt. at 12-h intervals (a dosing regimen strongly carcinogenic for the kidney but not the liver), and the other being a continuous dosing over several weeks by adding 8.5 mg of dimethylnitrosamine to each litre of drinking water giving an approximate daily dose of 0.7 mg/kg body wt. This treatment is known to be strongly carcinogenic for the liver but not the kidney. The accumulation in DNA of liver and kidney of the methylated purines 7-methylguanine and O6-methylguanine under each regimen were measured and compared. With the eleven-injection regimen there was a build up of O6-methylguanine in the DNA of the susceptible organ, the kidney, whilst in the liver virtually no accumulation was detected. Under the prolonged, low concentration regimen the liver, in spite of its susceptibility to the carcinogen did not accumulate O6-methylguanine. The results are discussed in terms of the hypothesis that production of O6-methylguanine and its persistence in the DNA of the target organ are responsible for the carcinogenic action of dimethylnitrosamine.  相似文献

(Des-Histidine1) (N?-phenylthiocarbamoyllysine12)-glucagon, synthesized by the one-step Edman degradation procedure is a competitive inhibitor of glucagon action in the rat liver plasma membrane adenylate cyclase system. However, in the perfused rat liver, the compound did not inhibit glucagon stimulated glycogenolysis even when used at a concentration 100-fold in excess of native glucagon. Instead, it showed a weak potency, but full agonist activity, stimulating liver glycogenolysis to 100% of the level obtained by glucagon. These results are discussed in terms of the possible mechanism(s) of glucagon action.  相似文献

The crystal structure of the compound [(Ph2PCH2CH2PPh2)Pt(3,5-Me2pzH)2][BF4]2·CH2Cl2 has shown that the ligands around the Pt atom are approximately in a square planar coordination, whereas the pyrazole rings point roughly in the same direction, away from the coordination plane. This unusual conformation is probably due to the hydrogen bonds with fluorine atoms of the BF4? anion. Bond distances and angles are compared with those in other pyrazole complexes.  相似文献
