1. Glycogen concentrations in liver and skeletal muscle were compared in rainbow trout and in chicks of two genetic sources. 2. Tissue glycogen concentrations were determined during feeding and after feed withdrawal in response to diets high in carbohydrate and oil, respectively. 3. Livers of trout and chicks were heavier and glycogen concentrations were higher in both liver and muscle of trout and chicks fed high-carbohydrate diets. 4. Feed withdrawal resulted in gradual but steady declines in trout glycogen over a 16-day period but caused sharp declines in liver glycogen in chicks followed by a rebound and a more gradual decline within a 5-day period. 5. Feed withdrawal from trout caused declines in muscle glycogen followed by rebounds which occurred more rapidly when the high-carbohydrate diet had been fed. 6. Feed withdrawal had little effect on muscle glycogen in broiler-type chicks. In White Leghorn chicks there was a general decline in muscle glycogen which showed marked fluctuations when the high-fat diet had been fed.  相似文献

The effects of different protein, lipid and carbohydrate diets on growth and energy storage in tench, Tinca tinca L., were studied. Over a 2-month period fish were fed four different diets: control, protein-enriched, carbohydrate-enriched and lipid-enriched. The best growth rates were obtained with the control and protein-enriched diets; the carbohydrate diet produced the worst results (lowest specific growth rate, weight gain, nutritional index and hepatosomatic index). These results suggest that it is not advisable to reduce dietary fish protein below 35%, and that it is not possible to obtain a protein-sparing effect of either lipids or carbohydrates, at least in our experimental conditions. The high-protein diet resulted in the storage of energy excess as muscle proteins and hepatic glycogen. Tench fed the high-carbohydrate diet stored carbohydrates as muscle glycogen and reduced plasma triglycerides. Finally, both liver and muscle lipid content were in positive correlation to dietary lipid.  相似文献

1. The turnover rate of L-[1-14C]leucine was increased by 35% in lactating rats compared with virgin rats. Starvation or removal of pups (24 h) returned the value to that of the virgin rat. 2. Incorporation of L-[U-14C]leucine into lipid and protein of mammary glands of lactating rats in vivo increased 7-fold and 6-fold respectively compared with glands of virgin rats. Lactation caused no change in the incorporation of L-[U-14C]leucine into hepatic lipid and protein. 3. The production of 14CO2 from L[l-14C]leucine (in the presence of glucose) was similar in isolated acini from glands of fed (chow) and starved lactating rats. Feeding with a 'cafeteria' diet caused a slight decrease, and removal of pups a large decrease, in the oxidative decarboxylation of leucine. 4. Oxidation of L-[2-14C]leucine to 14CO2 was increased about 3-fold in acini from starved lactating rats or lactating rats fed on a 'cafeteria' diet compared with rats fed on a chow diet. Insulin decreased the formation of 14CO2 in all three situations. 5. Incorporation of L-[U-14C]- and [2-14C]-leucine into lipid was decreased in acini from starved lactating rats and lactating rats fed on a 'cafeteria' diet. Insulin tended to increase the conversion of [2-14C]leucine into lipid, but this was significant only in the case of the acini from 'cafeteria'-fed rats. 6. Experiments with (-)-hydroxycitrate indicate that the major route for conversion of leucine carbon into lipid in acini is via citrate translocation from the mitochondria. 7. Intestinal preparations from rainbow trout fed a diet rich in wax esters incorporated [1(-14)C]hexadecanoic acid and [1(-14)C]hexadecanol into triacylglycerols at the same rate. The ratio of the number of H atoms from C1 of hexadecanol to the number of molecules of hexadecanol incorporated into triacylglycerols was 1.6 : 3.0. [U-14C]Glucose was incorporated much faster into the glycerol moiety of triacylglycerols than was [U-14C]aspartic acid. We conclude that the oxidation of absorbed fatty alcohol to fatty acid and its subsequent incorporation into triacylglycerols is closely linked with the reductive formation of triacylglycerol-glycerol from glucose. The ability of trout intestines to metabolise fatty alcohol to triacylglycerols was the same in fish fed wax esters as in those fed triacylglycerols.  相似文献

We hypothesize that variations in dietary carbohydrate levels produce changes in glucosensor parameters in previously characterized glucosensing areas (hypothalamus and hindbrain) related with the regulation of food intake of a carnivorous fish species like rainbow trout. Therefore, we fed trout with standard, carbohydrate-free (CF) or high-carbohydrate (HC) diets for 10 days to assess changes in glucosensing system and food intake. Fish fed CF diet displayed hypoglycemia and increased food intake. Fish fed a HC diet displayed hyperglycemia and decreased food intake. Changes in food intake due to dietary carbohydrates were accompanied in hypothalamus and hindbrain of fish fed with HC diet by changes in parameters involved in glucosensing, such as increased glucose, glucose 6-phosphate, and glycogen levels and increased glucokinase (GK), glycogen synthase, and pyruvate kinase activities as well as increased GK and GLUT2 expression. All those results address for the first time in fish, despite the relative intolerance to glucose of carnivorous species, that dietary carbohydrates are important regulators of the glucosensing system in carnivorous fish, suggesting that the information generated by this system can be associated with the changes observed in food intake.  相似文献

Fed animals have a higher resting metabolic rate in the thermoneutral zone than fasting ones. The metabolic increase is due to the specific dynamic action of food. With a decline of environmental temperature this increase in metabolism either declines or remains unchanged; decisive is whether the heat is used for thermoregulation or not (Mejsnar and Jansky 1971). The objective of our work was to find out to what extent a single intake of a diet with a different ratio of nutrients can influence resting metabolism in the golden hamster and whether this heat can be used for thermoregulation in the cold. Female golden hamsters aged 6-8 weeks kept at a constant temperature of 22 +/-1 degrees C with twelve-hour alternation of light (6 a.m. - 6 p.m.) and darkness ( 6 p.m. - 6 a.m.) were used for the experiments. The oxygen consumption was assessed after a single intake of a standard, high-carbohydrate (76 cal.% starch), high-fat (80 cal.% margarine) and high-protein (82 cal.% casein) diet-for detailed composition see Fábry (1959). The food was given at 6.m. after previous 20 hours of fasting. Animals were then transferred into the respiration chamber and kept there for three hours, including one hour when they were left to settle down; during this period the oxygen consumption was not measured. Oxygen consumption measurement started at 9 a.m. and lasted till 11 a.m. The metabolism of the animals at rest was assessed at temperatures of 10, 20 and 30 degrees C by measuring the oxygen consumption by the interferometric method (Wollschitt et al. 1935). The results are expressed in ml of oxygen per g of body weight per hour. The relationship between the metabolism at rest and environmental temperatures in hamsters given a single dose of standard, high-proetin, high-fat or high-carbohydrate diet is apparent from Table 1. The maximum increase of oxygen consumption after administration of the experimental diets was found at a temperature of 30 degrees C. At an environmental temperature of 20 degrees C the administration of the high-protein and high-fat diet causes roughly the same increase of metabolism. The high-carbohydrate diet increase is only one third of thevalues found, in the remaining two diets at the same temperature and is non-significant. At the environmental temperature of 10 degrees C all the diets used increased the oxygen consumption insignificantly. The changes in metabolism at different environmental temperatures after administration of various diets expressed as percentage of metabolism at 30 degrees C in animals fed the standard diet indicate that the specific dynamic action of the high-protein and high-fats diets is lower at lowered temperatures. We may thus assume that the heat produced as a result of specific dynamic action of the high-protein and high-fat diets is perhaps used for thermoregulation. The role of specific dynamic action of high-carbohydrate diet for thermoregulation is not clear from our experiments. Gastrointestinal (GI) integrity and function are regulated by nutrition and growth factors. The discovery of ghrelin, a natural growth hormone (GH) secretagogue produced by the gastrointestinal (GI) tract, is a potential link between diet and growth signals. The aim of this study was to evaluate macronutrient effect on ghrelin expression and secretion in addition to some possible function in intestinal trophic status. Wistar rats were fed a high-carbohydrate, high-protein (HP), high-fat or standard (St) diet. Animals received the same daily food volume and caloric intake. After 7 days, animals were fasted for 24 h and blood and tissue samples were obtained just before feeding or at 2 or 6 h after feeding. Fasting high-protein-fed rats had higher ghrelin plasma levels than with rats fed the high-carbohydrate, high-fat or standard diets. Two-hours after refeeding, ghrelin plasma levels had decreased in all groups with a slight recovery at 6 h after refeeding, except in the high-protein group. Ghrelin plasma levels in rats fed with the high-protein diet correlated negatively with their GH and insulin-like growth factor 1 (IGF-1) plasma concentrations which were also the lowest among the study groups. In conclusion, ghrelin secretion was nutritionally manipulated because a protein-enriched diet increased its levels.  相似文献

The effect of feeding an isocaloric and isonitrogenous trout diet that contained different levels of digestible carbohydrate (cerelose) to rainbow trout at either 10 or 15° C on liver glycogen and liver weight was determined in two fasting studies of 12 and 41 days duration. Trout fed diets with increased levels of digestible carbohydrate (HC) had significantly higher liver-body weight ratios (LW) and liver glycogen (LG) than trout reared on low digestible carbohydrate diets (HF). Both LW and LG declined in fasting trout previously fed HC diets but declined little in fasting trout previously fed HF diets. Trout reared at 10° C had higher LW and LG than trout reared at 15° C on either the HC or HF diets. During fasting, the trout reared on HC diets at 10° C required a longer period of time for the LG and LW to decline to the levels of trout reared on the low carbohydrate diets, than did trout reared on the HC diets at 15° C. The results indicate that both pre-fasting diet and water temperature can affect liver glycogen utilization and liver weight in fasting trout. Prolonged elevation of LW and LG in fasting trout could jeopardize the survival rate of stocked trout, particularly at low water temperatures.  相似文献

Blood glucose was significantly decreased by insulin (4 I.U./kg). Glucagon (1 mg/kg) and Cortisol (5 mg/kg) administration produced a significant hyperglycaemia. Insulin administration did not modify liver glycogen levels. Glucagon showed a marked liver glycogen mobilization. Cortisol stimulated liver glycogen deposition. Insulin and Glucagon showed a significant inverse effect on gluconeogenesis from (U-14C)glutamate, decreasing and increasing 14C-glucose formation respectively. Hormonal treatments did not influence the very low levels of incorporation of (U-14C)glutamate into liver and muscle glycogen.  相似文献

1. Inter-organ relationships between glucose, lactate and amino acids were studied by determination of plasma concentrations in different blood vessels of anaesthetized rats fed on either a high-carbohydrate diet [13% (w/w) casein, 79% (w/w) starch] or a high-protein diet [50% (w/w) casein, 42% (w/w) starch]. The period of food intake was limited (09:00-17:00h), and blood was collected 4h after the start of this period (13:00h). 2. Glucose absorption was considerable only in rats fed on a high-carbohydrate diet. Portal-vein-artery differences in plasma lactate concentration were higher in rats fed on this diet, but not proportional to glucose absorption. Aspartate, glutamate and glutamine were apparently converted into alanine, but when dietary protein intake was high, a net absorption of glutamine occurred. 3. The liver removed glucose from the blood in rats fed on a high-carbohydrate diet, but glucose was released into the blood in rats fed on the high-protein diet, probably as a result of gluconeogenesis. Lactate uptake was very low when amino acid availability was high. 4. In rats on a high-protein diet, increased uptake of amino acids, except for ornithine, was associated with a rise in portal-vein plasma concentrations, and in many cases with a decrease in hepatic concentrations. 5. Hepatic concentrations of pyruvate and 2-oxo-glutarate decreased without a concomitant change in the concentrations of lactate and malate in rats fed on the high-protein diet, in spite of an increased supply of pyruvate precursors (e.g. alanine, serine, glycine), suggesting increased pyruvate transport into mitochondria. 6. High postprandial concentrations of plasma glucose and lactate resulted in high uptakes of these metabolites in peripheral tissues of rats on both diets. Glutamine was released peripherally in both cases, whereas alanine was taken up in rats fed on a high-carbohydrate diet, but released when the amino acid supply increased. 7. It is concluded that: the small intestine is the main site of lactate production, and the peripheral tissues are the main site for lactate utilization; during increased ureogenesis in fed rats, lactate is poorly utilized by the liver; the gut is the main site of alanine production in rats fed on a high-carbohydrate diet and the liver utilizes most of the alanine introduced into the portal-vein plasma in both cases.  相似文献

Improper eating habits such as high-fat or high-carbohydrate diets are responsible for metabolic changes resulting in impaired glucose tolerance, hyperinsulinemia, insulin resistance, and ultimately diabetes. Although the essentiality of trivalent chromium for humans has been recently questioned by researchers, pharmacological dosages of this element can improve insulin sensitivity in experimental animals and diabetic subjects. The aim of the study was to assess the preventive potential of the supplementary chromium(III) propionate complex (CrProp) in rats fed a high-fat diet. The experiment was conducted on 32 male Wistar rats divided into four groups and fed the following diets: the control (C, AIN-93G), high-fat diets (HF, 40 % energy from fat), and a high-fat diet supplemented with CrProp at dosages of 10 and 50 mg Cr/kg diet (HF?+?Cr10 and HF?+?Cr50, respectively). After 8 weeks, high-fat feeding led to an increased body mass, hyperinsulinemia, insulin resistance, a decreased serum urea concentration, accumulation of lipid droplets in hepatocytes, and increased renal Fe and splenic Cu contents. Supplementary CrProp in both dosages did not alleviate these changes but increased renal Cr content and normalized splenic Cu content in high-fat-fed rats. Supplementary CrProp does not prevent the development of insulin resistance in rats fed a high-fat diet.  相似文献

The effect of increased available dietary carbohydrate on the tolerance of waterborne copper by rainbow trout was investigated. Trout were reared for six to eight weeks on one of four isocaloric and isonitrogenous diets which varied in available carbohydrate content due to either processing (extrusion v. steam-pelleting) or supplementation with cerelose (0 or 21%). The incipient lethal level (ILL) of copper for trout reared on the low-available-carbohydrate steam-pelleted diet was 350 μg I?1, significantly higher than the 276 μg I?1 shown for trout reared on the high-available-carbohydrate extruded diet. The ILL of copper for the low-carbohydrate group (0% cerelose) was 408 μg I?1, significantly higher than the ILL of 246 μg I?1 of trout reared on the high-carbohydrate diet (21% cerelose). Increased available dietary carbohydrate resulted in increased liver glycogen content and liver-body weight ratios and decreased liver protein content. Significant correlations were evident between waterborne copper tolerance and liver glycogen, liver-body weight ratios and liver protein. Relative to those on high carbohydrate diets, livers of fish on low-carbohydrate diets contained higher concentrations of copper after seven days of exposure to 0 or 196 μg I?1 of copper. The elevated liver glycogen levels, resulting from increased dietary carbohydrate, are thought to have impaired liver function, thus reducing copper tolerance.  相似文献

Three coenzyme A (CoA) molecular species, i.e., acetyl-CoA, malonyl-CoA, and nonesterified CoA (CoASH), in 13 types of fasted rat tissue were analyzed. A relatively larger pool size of total CoA, consisting of acetyl-CoA, malonyl-CoA, and CoASH, was observed in the medulla oblongata, liver, heart, and brown adipose tissue. Focusing on changes in the CoA pool size in response to the nutrient composition of the diet given, total CoA pools in rats continuously fed a high-fat diet for 4 weeks were significantly higher in the hypothalamus, cerebellum, and kidney, and significantly lower in the liver and skeletal muscle than those of rats fed a high-carbohydrate or high-protein diet. In particular, reductions in the liver were remarkable and were caused by decreased CoASH levels. Consequently, the total CoA pool size was reduced by approximately one-fifth of the hepatic contents of rats fed the other diets. In the hypothalamus, which monitors energy balance, all three CoA molecular species measured were at higher levels when rats were fed the high-fat diet. Thus, it was of interest that feeding rats a high-fat diet affected the behaviors of CoA pools in the hypothalamus, liver, and skeletal muscle, suggesting a significant relationship between CoA pools, especially malonyl-CoA and/or CoASH pools, and lipid metabolism in vivo.  相似文献

The effects of proline on lipogenesis in isolated rat hepatocytes were determined and compared with those of lactate, an established lipogenic precursor. Proline or lactate plus pyruvate increased lipogenesis (measured with 3H2O) in hepatocytes from fed rats depleted of glycogen in vitro and in hepatocytes from starved rats. Lactate plus pyruvate but not proline increased lipogenesis in hepatocytes from starved rats. ( - )-Hydroxycitrate, an inhibitor of ATP-citrate lyase, partially inhibited incorporation into saponifiable fatty acid of 3H from 3H2O and 14C from [U-14C]lactate with hepatocytes from fed rats. Incorporation of 14C from [U-14C]proline was completely inhibited. Similar complete inhibition of incorporation of 14C from [U-14C]proline by ( - )-hydroxycitrate was observed with glycogen-depleted hepatocytes or hepatocytes from starved rats. Inhibition of phosphoenolpyruvate carboxykinase by 3-mercaptopicolinate did not inhibit the incorporation into saponifiable fatty acid of 3H from 3H2O or 14C from [U-14C]proline or [U-14C]lactate. Both 3-mercaptopicolinate and ( - )-hydroxycitrate increased lipogenesis (measured with 3H2O) in the absence or presence of lactate or proline with hepatocytes from starved rats. The results are discussed with reference to the roles of phosphoenolpyruvate carboxykinase, mitochondrial citrate efflux, ATP-citrate lyase and acetyl-CoA carboxylase in proline- or lactate-stimulated lipogenesis.  相似文献

In parallel with increased prevalence of overweight people in affluent societies are individuals trying to lose weight, often using low-carbohydrate diets. Nevertheless, long-term metabolic consequences of those diets, usually high in (saturated) fat, remain unclear. Therefore, we investigated long-term effects of high-fat diets with different carbohydrate/protein ratios on energy balance and fuel homeostasis in obese (fa/fa) Zucker and lean Wistar rats. Animals were fed high-carbohydrate (HC), high-fat (HsF), or low-carbohydrate, high-fat, high-protein (LC-HsF-HP) diets for 60 days. Both lines fed the LC-HsF-HP diet displayed reduced energy intake compared with those fed the HsF diet (Zucker, -3.7%) or the HC diet (Wistar rats, -12.4%). This was not associated with lower weight gain relative to HC fed rats, because of increased food efficiencies in each line fed HsF and particularly LC-HsF-HP food. Zucker rats were less glucose tolerant than Wistar rats. Lowest glucose tolerances were found in HsF and particularly in LC-HsF-HP-fed animals irrespective of line, but this paralleled reduced plasma adiponectin levels, elevated plasma resistin levels, higher retroperitoneal fat masses, and reduced insulin sensitivity (indexed by insulin-induced hypoglycemia) only in Wistar rats. In Zucker rats, however, improved insulin responses during glucose tolerance testing and tendency toward increased insulin sensitivities were observed with HsF or LC-HsF-HP feeding relative to HC feeding. Thus, despite adverse consequences of LC-HsF diets on blood glucose homeostasis, principal differences exist in the underlying hormonal regulatory mechanisms, which could have benefits for B-cell functioning and insulin action in the obese state but not in the lean state.  相似文献

Previous studies have shown that rainbow trout fed on diets containing whole protein have superior growth rates compared to fish fed on diets of similar amino acid composition but containing a high proportion of free amino acids. The influence of several nutritional factors on the uptake of radioactivity from food pellets containing either [U-I4C] protein or [U-14C] amino acids into the systemic blood of trout has been investigated. The time taken for radioactivity in the free amino acid fraction of blood to reach a peak after a meal containing [U-14C] protein had been given was much shorter, and the level of radioactivity in the blood higher, in trout with almost empty stomachs than in fish with almost full stomachs; uptake of radioactivity into blood amino acids was also more rapid and reached much higher concentrations when pellets containing [U-14C] amino acids were fed than when [U-14C] protein was fed. Incorporation of radioactivity into blood protein continued for a much longer period and reached higher levels when a pellet containing [U-14C] protein was fed than when a pellet containing [U-14C] amino acids was fed. Previous dietary history (low or high protein intake) did not appear to affect the rate of absorption of amino acids from either protein or free amino acid pellets. The uptake rates from pellets containing free amino acids could be slowed by mixing the dietary amino acids with albumin. The distribution, postabsorption, of radioactivity in the different fractions of blood and liver suggested that incorporation of carbon residues into glycogen and lipid from an amino acid diet was greater than from a protein diet. The converse was true of incorporation of radioactivity into tissue protein.  相似文献

Yearling coho salmon were fed either a commercial trout (TC) diet or a diet consisting of homogenized adult Pacific Ocean coho salmon (PS), supplemented with vitamins and minerals. The hepatosomatic index of the TC-fed fish killed 12 h after the last feed or after 4 or 8 weeks of fasting were significantly larger ( P 0.05) than comparable PS-fed groups.
'Light' and'dark' hepatocyte cell types were evident in all groups although the dark cells were less numerous in the fasted fish. Hepatocytes of fasted fish contained markedly more endoplasmic reticulum than fed fish. Hepatocytic glycogen content of the TC-fed fish was higher than in PS-fed fish. Whereas in the TC-fed group hepatic glycogen was still evident in fish fasted for 8 weeks, the hepatocytes of the fasted PS-fed group contained little or no glycogen. The suitability of commercial fish diets for salmonids is discussed.  相似文献

Glucose and lipid metabolism in pancreatic islet organs is poorly characterized. In the present study, using as a model the carnivorous rainbow trout, a glucose-intolerant fish, we assessed mRNA expression levels of several genes involved in glucose and lipid metabolism (including ATP-citrate lyase; carnitine palmitoyltransferase-1 isoforms, CPT; the mitochondrial isoform of the phosphoenolpyrutave carboxykinase, mPEPCK and pyruvate kinase, PK) and glucosensing (glucose transporter type 2, Glut2; glucokinase, GK and the potassium channel, KATP) in Brockmann bodies. We evaluated the response of these parameters to changes in feeding status (food deprived vs. fed fish) as well as to changes in the amount of carbohydrate (dextrin) in the diet. A general inhibition of the glycolytic (including the glucosensing marker GK) and β-oxidation pathways was found when comparing fed versus food-deprived fish. When comparing fish feeding on either low- or high-carbohydrate diets, we found that some genes related to lipid metabolism were more controlled by the feeding status than by the carbohydrate content (fatty acid synthase, CPTs). Findings are discussed in the context of pancreatic regulation of glucose and lipid metabolism in fish, and show that while trout pancreatic metabolism can partially adapt to a high-carbohydrate diet, some of the molecular actors studied seem to be poorly regulated (KATP) and may contribute to the glucose intolerance observed in this species when fed high-carbohydrate diets.  相似文献

Maslinic acid is a triterpene present in a considerable proportion in solid residues from olive-oil production. In the present work the effects of maslinic acid on growth, protein-turnover rates and nucleic-acid concentration on liver were investigated in the rainbow trout. Five groups of 120 fish of a mean body mass of 20 g were fed for 225 days with diets containing 0, 1, 5, 25 and 250 mg of maslinic acid per kg diet. At the end of the experiment, whole-body and liver weight and growth rate of trout fed with maslinic acid were higher than controls. The highest weight increase was registered for the group fed 250 mg kg(-1), representing a 29% increase over controls. The total hepatic DNA or liver cell hyperplasia levels in trout fed with 25 and 250 mg of maslinic acid kg(-1) were 37% and 68% higher than controls. Also in these same groups of trout, fractional and absolute hepatic protein-synthesis rates were significantly higher than in control, and significant increments in hepatic protein-synthesis efficiency and protein-synthesis capacity were reported. In close agreement with these results, microscopy studies showed that trout fed on 25 and 250 mg kg(-1) hepatocytes appeared to be more compact, with a larger rough-endoplasmic reticulum and larger glycogen stores than controls. These results suggest that maslinic acid can act as a growth factor when added to trout diet.  相似文献

Carbamoyl phosphate synthetase I (CPS-I) is the most abundant protein of rat liver mitochondria. Biochemical measurements in liver homogenates have shown that the liver from rats fed a high-protein diet contains more CPS-I per gram tissue protein than controls. However, there is no information on changes in the intact tissue at the cellular and mitochondrial level. Therefore, monoclonal antibodies to beef liver CPS-I were produced by the hybridoma technique. Four clones, C-241/1A, B, C, and D secreted immunogammaglobulin (IgG) IgG1. Using C-241/C, we measured by electron microscopy immunogold procedures the labeling of CPS-I in mitochondria from liver of rats fed high protein (casein, 50 and 80% of total food intake) diets. CPS-I (expressed as gold particles/micron2 of mitochondrial cross-sectional area) was greater than in mitochondria from control rats (20% casein diet), whether the rats were fed for 1, 6, or 14 months on the high-protein diets. The immunocytochemical measurements shown here demonstrate that the increase in the level of CPS-I in high-protein diets is a reflection of both the larger number of CPS-I molecules per mitochondrial area and the larger proportion of the total hepatocyte volume occupied by mitochondria. Similar measurements were carried out with glutamate dehydrogenase (GDH) using previously characterized monoclonal antibodies. No differences in GDH labeling were found with high-protein diets. Interestingly, when mitochondria from hepatocytes of rats fed a high-protein diet were divided into two subpopulations on the basis of mitochondrial cross-sectional size (i.e., greater or less than 0.7 micron2), the large mitochondria had 1.2 times more CPS-I and 0.8 times less GDH than the small mitochondria nearby.  相似文献
