采用0℃~7.2℃模型的累积低温计算方法,以加权法为萌芽计算标准,通过对29份观赏桃Prunus persica Batsch.(L.)品种资源的需冷量研究,结果表明:我国重瓣桃花品种的需冷量分布范围为400h~1250h,并以900h以上的品种为主.红花垂枝桃、红花重瓣寿星桃、鸳鸯垂枝、红叶桃、洒红桃和菊花桃等名优品种的需冷量分别为926h、930h、1188h、1112h、1214h 和1250h,郑州地区需冷量的满足时间基本在1月底之后.  相似文献

‘龙山红’是从四川省龙门山南段的野生彩色猕猴桃中筛选出的一个两性花株系,也是国内首次发现雌雄同体且充分发育的两性花彩色猕猴桃株系。本文首次报道温度(低温层积,变温)和赤霉素处理对两性花猕猴桃新株系‘龙山红’种子萌发的影响。通过低温层积、变温、GA3及其组合处理后的试验表明,低温层积、变温、GA3都能有效打破休眠,促进萌发,但物理温度处理法对种子萌发的效果更好。  相似文献

以酿酒葡萄‘赤霞珠’(红)和‘霞多丽’(白)为试材,通过考察其自然休眠过程和打破休眠(单氰胺处理)过程中冬芽组织活性氧及抗氧化系统的变化,探讨葡萄休眠机制。结果表明:(1)在陕西杨陵地区,早熟品种‘霞多丽’在12月20日左右进入深度休眠,第2年1月29日进入休眠解除阶段;晚熟品种‘赤霞珠’在1月9日左右达到深度休眠,2月18日左右进入休眠解除阶段;单氰胺处理可促进葡萄休眠的解除。(2)葡萄休眠过程中,活性氧O2.-和H2O2含量在休眠初期逐渐增加,深度休眠时含量最高,休眠解除过程中含量降低,从而促进休眠的解除;超氧化物歧化酶(SOD)活性在休眠中一直降低,休眠解除后活性上升;过氧化物酶(POD)活性在休眠前期较低,随后活性上升,休眠解除过程中活性又降低;过氧化氢酶(CAT)活性在休眠过程中持续上升,随后维持在较高水平;抗坏血酸-还原态的谷胱甘肽(ASA-GSH)循环在休眠前期较弱,休眠解除过程中逐渐增强。研究发现,葡萄进入休眠后,其冬芽抗氧化能力逐渐降低,至深度休眠时抗氧化能力降至最低,随着低温积累需冷量得到满足,活化了抗氧化系统进而抗氧化能力迅速提高,休眠被解除。  相似文献

自然条件下,4月至7月下旬期间,中国水仙鳞茎的呼吸速率逐渐下降,7月底至8月底开始上升之后又下降。30℃高温下的鳞茎发芽延缓,发芽率下降;15℃低温有利于打破休眠,7月下旬前的鳞茎,其休眠解除所需的低温处理时间长,GA3不能破除,还延缓其休眠时间,萌发率也下降;而7月下旬后的鳞茎,短时间低温或施用GA3均能破除其休眠,提高鳞茎的发芽率和发芽整齐度;乙烯在任何阶段都有助于水仙鳞茎休眠的解除。自然条件下7月下旬前水仙鳞茎可能是内生休眠,其休眠较难打破,需要长时间的低温或施用适当浓度的乙烯,7月下旬后可能是环境休眠,短时间的低温、GA3或乙烯都能破除其休眠。  相似文献

采用高效液相色谱法(HPLC)分析了剥鳞与激素处理对大樱桃花芽休眠解除及内源生长素(IAA)、赤霉素(GAD、玉米素(ZT)和脱落酸(ABA)变化的影响。结果表明,花芽中的ABA主要分布于鳞片内,鳞片中的GA3和ZT含量远低于去鳞芽,也低于完整芽。剥鳞能明显增加休眠花芽中内源GA2和ZT的含量,降低ABA的含量,对IAA含量的影响不大。剥鳞降低了ABA/GA3、ABA/ZT的比值,使花芽向促进生长、抑制休眠的方向转化。同时,休眠前、后期剥鳞均能明显提高萌芽率,中期剥鳞效果不明显。剥鳞后施用外源激素随休眠时期不同而有不同的破眠效果,早期剥鳞GA3的效果最好,6-BA次之,IAA最差;中期破眠效果不如早期,GA。和6-BA没有明显差别;后期以6-BA效果最好,其次是GA3和IAA;3次处理中ABA均明显抑制花芽萌发。  相似文献

以3年生盆栽‘曙光’油桃为材料,研究油桃自然休眠过程中50℃高温和单氰胺对花芽呼吸代谢的影响.结果表明:高温和单氰胺均可以打破油桃的自然休眠,导致休眠花芽呼吸代谢显著下降,其呼吸代谢的衰减可持续数小时.主要呼吸途径三羧酸循环(TCA)和磷酸戊糖途径(PPP)的运行均受到影响.未经破眠处理的花芽TCA和PPP均呈衰减趋势,而高温和单氰胺诱导了早期呼吸衰减结束后PPP的迅速活化.高温还表现出对TCA恢复的诱导,而单氰胺在96 h内未表现出这种作用.在高温和单氰胺打破自然休眠的机制中,呼吸衰减和随后出现的PPP活化可能是重要的组成部分.  相似文献

以采自江西吉安官山林场5年生东京野茉莉当年自然带壳种子为材料,通过对其种子吸水率、不同层积时期种子内生理生化指标的测定、种子萌发抑制物分析,并利用各类不同药剂处理进行发芽试验,以探寻东京野茉莉种子的休眠机理及破眠方法。结果表明:(1)休眠的原因主要包括种皮障碍、缺少萌发所需激素以及种皮、胚中存在萌发抑制物,其中种皮障碍和抑制物的存在是限制种子萌发的首要因素。(2)GA3处理结合自然低温层积30d即可解除东京野茉莉种子胚的休眠,但种皮障碍始终是其种子萌发的限制因素。(3)GA3、NAA、6-BA等药剂处理均可促进种子的萌发,并以刻伤种子后用500mg/L GA3处理24h为破除该种子休眠的最有效方法。  相似文献

以2年生低需冷量设施栽培适宜品种“春捷毛桃”为试材,研究桃植株进入休眠和发展抗冻性的自然诱导因子及Ca2+在此过程中的作用.结果表明:短日照和自然低温单个因子或其共同作用均能诱导桃植株停止生长,然后进入休眠状态和发展抗冻性,但短日照和自然低温的作用机制不同.短日照首先诱导桃植株进入休眠状态,然后诱导抗冻性发展;而自然低温则是首先诱导桃植株抗冻性发展,而后诱导其进入休眠状态;当短日照和自然低温共同作用即自然条件下时,短日照起主导作用,自然低温起辅助促进作用.短日照处理研究表明,短日照诱导桃植株停止生长、休眠诱导和抗冻性发展离不开Ca2+的作用,在此过程中Ca2+起着传递短日照信号的作用;在人工补光长日照条件下,随着温度的降低,Ca2+逐渐由细胞间隙、细胞壁和液泡流入细胞质和核内,同时植株生长速度变缓并最终完全停止,随后进入休眠状态并发展抗冻性.表明Ca2+作为传递自然低温信号的信使在自然低温诱导桃植株停止生长、抗冻性发展和休眠过程中起着重要作用.  相似文献

以12个葡萄品种1年生枝条为试材,通过0、-15、-18、-21、-24、-27、-30、-133℃8个低温处理24h后,测定不同低温胁迫下葡萄枝条的,6-含水量、束缚水含量、自由水含量和束缚水与自由水的比值。结果表明:随着处理温度的降低,同一品种枝条的总含水量基本恒定,自由水含量呈先下降再升高的趋势,束缚水含量和束缚水与自由水的比值均呈现为先升高再下降的趋势;束缚水与自由水的比值随低温胁迫的变化呈Cubic方程,不同品种束缚水与自由水的比值达到最高点的温度不同,‘双红’、‘北冰红’、‘左优红’的拐点温度在-25℃以下,‘775’、‘巨峰’的拐点温度在-20℃左右,‘雷司令’、‘黑比诺’、‘霞多丽’的拐点温度在-18℃左右,‘赤霞珠’、‘梅鹿特’、‘白比诺’、‘红地球’拐点温度在-16~17℃。对葡萄休眠枝条进行低温处理后,通过测定束缚水与自由水含水量变化鉴定葡萄品种的抗寒性是可行的。  相似文献

Background

Recently, many studies utilizing next generation sequencing have investigated plant evolution and domestication in annual crops. Peach, Prunus persica, is a typical perennial fruit crop that has ornamental and edible varieties. Unlike other fruit crops, cultivated peach includes a large number of phenotypes but few polymorphisms. In this study, we explore the genetic basis of domestication in peach and the influence of humans on its evolution.

Results

We perform large-scale resequencing of 10 wild and 74 cultivated peach varieties, including 9 ornamental, 23 breeding, and 42 landrace lines. We identify 4.6 million SNPs, a large number of which could explain the phenotypic variation in cultivated peach. Population analysis shows a single domestication event, the speciation of P. persica from wild peach. Ornamental and edible peach both belong to P. persica, along with another geographically separated subgroup, Prunus ferganensis.We identify 147 and 262 genes under edible and ornamental selection, respectively. Some of these genes are associated with important biological features. We perform a population heterozygosity analysis in different plants that indicates that free recombination effects could affect domestication history. By applying artificial selection during the domestication of the peach and facilitating its asexual propagation, humans have caused a sharp decline of the heterozygote ratio of SNPs.

Conclusions

Our analyses enhance our knowledge of the domestication history of perennial fruit crops, and the dataset we generated could be useful for future research on comparative population genomics.

Dormancy is a condition that delays or inhibits growth in seed, vegetative buds, and floral buds. In peach, seed germination occurs when seed accumulate sufficient stratification and growing degree hours to break dormancy and begin growing. Correlations have been reported between mean seed stratification requirements and mean bud chilling requirements among Prunus families, but an individual seed’s germination date and subsequent vegetative and floral bud break date are not correlated. Prior to this study, the genetic factors involved in regulating seed dormancy and their location on the peach genomic map were unknown. Segregating F2 seed were collected from a high?×?low chill F1 peach hybrid in 2005, 2006, and 2008. Germination date and growth habit was measured after the stratification requirement of the 2005 seed was fully met. The seed collected in 2006 and 2008 received varying amounts of stratification, which enabled data on stratification requirement, heat requirement, and growth habit to be collected. Genomic DNA was extracted from seedling leaf tissue and screened with SSR markers selected from the Prunus reference map at an average resolution of 20 cM. Seed dormancy quantitative trait loci (QTLs) were detected on G1, G4, G6/8, and G7. The QTLs detected on G6/8 and G7 were discovered in the same region as QTLs associated with floral bud chilling requirement and bloom time in peach.  相似文献

近年来,我国观赏桃新品种日渐繁多、名称混乱、市场难以监管,同时用以区分品种的SSR指纹图谱的构建方法在研究界无统一的科学标准,尤其是构成最终引物组合的核心引物的确定,具体操作流程层出不穷、五花八门。为探索筛选SSR指纹图谱核心引物的科学方法,同时构建观赏桃SSR指纹图谱,该研究选用35对已报道的SSR引物对22份观赏桃种质进行试验。结果表明:通过PCR扩增与分析,多态性较高的8对引物——候选引物总共扩增出31个多态性条带,变幅为3~5个,PIC值变幅为0.458~0.668,MI值变幅为1.374~3.340。采用"随机组合"法对8对引物进行C_8~1、C_8~2、C_8~3…依次分析,得到区分能力最强的3种不同的最少引物组合方式——候选组合,并能区分出18份种质,从中发现区分能力最强的3种引物组合方式并不都是由引物PIC值、alleles数量或MI值等多态性指标最高的引物组成,而是由互补性最强的引物组成。选用组合内各引物多态性条带总数最多的组合方式"4-3"(BPPCT001+BPPCT015a+BPPCT017+BPPCT025)为22份观赏桃种质构建了指纹图谱。基于此,通过常规多态性指标筛选候选引物可以确定出单对引物鉴别能力最强的少量引物;通过"随机组合"筛选候选组合可以进一步确定出引物之间互补性最强的几种组合方式;根据组合内各引物的多态性条带总数确定最终核心引物可以确定出可扩容性最大的引物组合。该研究最终建立了候选引物——候选组合——核心引物组合"三步法"确定SSR指纹图谱核心引物组合的科学方法,不仅为22份供试观赏桃种质构建了SSR指纹图谱,也为其它作物SSR指纹图谱的构建提供了新的思路。  相似文献

Background and Aims

The aim was to assess the amounts of macro- (N, P, K, Ca and Mg) and micro-elements (Fe, Mn, Cu and Zn) lost by peach trees (Prunus persica L. Batsch) in all the nutrient removal events (pruning, flower abscission, fruit thinning, fruit harvest and leaf fall), as well as those stored in the permanent structures of the tree (roots, trunk and main branches).

Methods

Three peach cultivars were used. The biomass and nutrient composition of materials lost by trees at the different events were measured during 3 years. The biomass and nutrient composition of permanent tree structures were also measured after full tree excavation.

Results

Winter pruning and leaf fall were the events where most nutrients were removed. Nutrient losses and total requirements are given as amounts of nutrients needed per tree and also as amounts necessary to produce a t of fresh fruit.

Conclusions

The allocation of all nutrients analyzed in the different plant parts was similar in different types of peach trees, with each element having a typical “fingerprint” allocation pattern. Peach tree materials removed at tree pruning and leaf fall include substantial amounts of nutrients that could be recycled to improve soil fertility and tree nutrition. Poorly known tree materials such as flowers and fruit stones contain measurable amounts of nutrients.  相似文献

A 2-season trial was conducted to verify the effects of foliar applications of some dormancy-breaking substances (DBS) on dormancy release in buds of ‘Anna’ apple (Malus sylvestris, Mill) trees, as well as on metabolic changes in the contents of phytohormones, proline and arginine in buds during their release from dormancy. The efficiency of early bud break induced by Dormex?, potassium nitrate, mineral oil, calcium nitrate and thiourea was noticed in varying degrees. Although Dormex? was distinguished, all DBS hastened bud break, shortened flowering duration, improved bud break% and fruit-set%, increased the contents of gibberellic acid, indole-3-acetic acid, proline and arginine, but reduced abscisic acid content in buds as compared to the control. These results were positively reflected in the final tree yield. Accordingly, it is concluded that the use of Dormex?, at a rate of 4 %, could be recommended for reaching bud break as early as possible and improving ‘Anna’ apple tree yield under the short winters in Egypt and similar regions by regulating the contents of proline, arginine and phytohormones in buds.  相似文献

Background

Winter chill is one of the defining characteristics of a location''s suitability for the production of many tree crops. We mapped and investigated observed historic and projected future changes in winter chill in California, quantified with two different chilling models (Chilling Hours, Dynamic Model).

Methodology/Principal Findings

Based on hourly and daily temperature records, winter chill was modeled for two past temperature scenarios (1950 and 2000), and 18 future scenarios (average conditions during 2041–2060 and 2080–2099 under each of the B1, A1B and A2 IPCC greenhouse gas emissions scenarios, for the CSIRO-MK3, HadCM3 and MIROC climate models). For each scenario, 100 replications of the yearly temperature record were produced, using a stochastic weather generator. We then introduced and mapped a novel climatic statistic, “safe winter chill”, the 10% quantile of the resulting chilling distributions. This metric can be interpreted as the amount of chilling that growers can safely expect under each scenario. Winter chill declined substantially for all emissions scenarios, with the area of safe winter chill for many tree species or cultivars decreasing 50–75% by mid-21st century, and 90–100% by late century.

Conclusions/Significance

Both chilling models consistently projected climatic conditions by the middle to end of the 21st century that will no longer support some of the main tree crops currently grown in California, with the Chilling Hours Model projecting greater changes than the Dynamic Model. The tree crop industry in California will likely need to develop agricultural adaptation measures (e.g. low-chill varieties and dormancy-breaking chemicals) to cope with these projected changes. Most trees from temperate climates require the accumulation of winter chill and subsequent heat during their dormant phase to resume growth and initiate flowering in the following spring. Global warming could reduce chill and hence hamper the cultivation of high-chill species such as cherries. Yet determining chilling and heat requirements requires large-scale controlled-forcing experiments, and estimates are thus often unavailable. Where long-term phenology datasets exist, partial least squares (PLS) regression can be used as an alternative, to determine climatic requirements statistically. Bloom dates of cherry cv. ‘Schneiders späte Knorpelkirsche’ trees in Klein-Altendorf, Germany, from 24 growing seasons were correlated with 11-day running means of daily mean temperature. Based on the output of the PLS regression, five candidate chilling periods ranging in length from 17 to 102 days, and one forcing phase of 66 days were delineated. Among three common chill models used to quantify chill, the Dynamic Model showed the lowest variation in chill, indicating that it may be more accurate than the Utah and Chilling Hours Models. Based on the longest candidate chilling phase with the earliest starting date, cv. ‘Schneiders späte Knorpelkirsche’ cherries at Bonn exhibited a chilling requirement of 68.6?±?5.7 chill portions (or 1,375?±?178 chilling hours or 1,410?±?238 Utah chill units) and a heat requirement of 3,473?±?1,236 growing degree hours. Closer investigation of the distinct chilling phases detected by PLS regression could contribute to our understanding of dormancy processes and thus help fruit and nut growers identify suitable tree cultivars for a future in which static climatic conditions can no longer be assumed. Sweet cherry cultivars have different chilling and heat requirements for breaking rest and flowering. The knowledge of these requirements may be valuable in the selection of the appropriate cultivars for producers and to avoid losses caused by an inadequate cultivar selection in a particular area. Determination of chilling and heat requirements is also important within a breeding programme, when choosing parents to obtain early flowering cultivars. Chilling requirements of seven cherry cultivars growing in south-eastern Spain were calculated using different methods (hours below 7 °C, Utah and Dynamic model), which were compared. Recording hourly average temperatures at several locations during 2 years, the Utah and Dynamic models performed better than hours below 7 °C. Different chilling requirements and slight differences in heat requirements were observed in the studied cultivars. ‘Cristobalina’ and ‘Brooks’, the earlier-flowering cultivars, were those with the lowest chilling requirements. ‘Burlat’, ‘New Star’ and ‘Somerset’ had medium chilling and heat requirements for flowering, and ‘Marvin’ showed the highest values and also the latest blooming date. All the studied cultivars may have their chilling requirements satisfied in the region of Murcia, if grown at least 650 m above sea level. Some cultivars, such as ‘Cristobalina’ and ‘Brooks’, could successfully break dormancy already when grown at an altitude above 325 m.  相似文献

Sequence-specific amplified polymorphism (SSAP) technology is a novel, anchored PCR approach derived from AFLP, which amplifies the region between a transposon insertion and an adjacent restriction site and have higher levels of polymorphism. In the current study, we developed 16 SSAP markers based on the long terminal repeat (LTR) sequences of Ty1-copia retrotransposons in the peach and used them for DNA profiling of 52 individual peaches: 44 peach cultivars and 8 ornamental peaches. These primer combinations produced a total of 1,553 fragments and 1,517 polymorphic bands with a polymorphism percentage of 97.7%. Furthermore, the Shannon's information index of each primer combination ranged from 0.1593 to 0.4456. Neighbor-joining analyses revealed two main genetic clusters, corresponding to the fruit flesh types: (A-1) MF (melting flesh) with clingstone and ornamental peaches; (A-2) MF with freestone and NMF (non-melting flesh) with clingstone. Finally, cluster analyses revealed that all ornamental peaches are closely related to the MF with clingstone peach cultivars. The application of these primer combinations identified using SSAP will facilitate future cultivar identification and germplasm management in peaches.  相似文献
