Previous studies have successfully shown evidence for parasitic infections in human remains from various archaeological sites. However, in the case of Korea, since there have been very few paleoparasitological reports published, pre-20th century parasitic infection patterns remain obscure. Therefore, in order to partly fill this gap, we are reporting on a case of paleoparasitic infection from the feces of a 15th century child mummy from Yangju, Korea. In the course of the present study, we found the eggs of Clonorchis sinensis, Ascaris lumbricoides, and Trichuris trichiura in the feces of the mummy. Trichuris trichiura eggs were found in far greater numbers than other parasite eggs; in fact, intact bipolar plugs were clearly observed and even the larvae were still visible in some eggs. The eggs of C. sinensis and A. lumbricoides were also well preserved, though not in as great a number. Since we could find a number of well-preserved larvae-containing eggs, we are encouraged that successful extraction, amplification, and sequence determination of ancient DNA from the paleoparasite eggs might be possible in future studies. With additional paleoparasitological investigation using feces from Korean mummies, we hope that a history of parasite infection in Korea will be reconstructed.  相似文献

Paleoparasitological research has made important contributions to our understanding of parasite evolution and ecology since the 1960s. Since then, most studies have focused on paleoparasitological evidence from single sites or samples. With the development of high throughput sequencing techniques, new avenues of investigation for paleoparasitological material are opening up. Here, I provide an overview of recent developments and highlight how these results will broaden the scope of the field, placing paleoparasitology at the interface of a wide array of studies, including parasitology, climate change, human evolution, and evolutionary processes.  相似文献

Three techniques to extract parasite remains from archaeological sediments were tested. The aim was to improve the sensibility of recommended paleoparasitological techniques applied in archaeological remains. Sediment collected from the pelvic girdle of a human body found in Cabo Vírgenes, Santa Cruz, Argentina, associated to a Spanish settlement founded in 1584 known as Nombre de Jesús, was used to search for parasites. Sediment close to the skull was used as control. The techniques recommended by Jones, Reinhard, and Dittmar and Teejen were used and compared with the modified technique presented here, developed to improve the sensibility to detect parasite remains. Positive results were obtained only with the modified technique, resulting in the finding of Trichuris trichiura eggs in the sediment.  相似文献

Organic remains attributable to one regurgitated pellet were examined. The pellet, belonging to a bird of prey and collected from a cave of Southern Patagonia, was dated at 6,540 +/- 110 yr. With standard paleoparasitological procedures, eggs of Capillaria sp. and a mite, Demodex sp., were found. The parasites found in the pellet belong to a rodent ingested by the bird. The present report constitutes the first paleoparasitological study of a regurgitated pellet.  相似文献

The present paper reports the first paleoparasitological results obtained from coprolites of fossil rodent middens and demonstrates the potential of rodent middens as a source of paleoparasitological evidences in South America. Ten fossil rodent middens from northwestern Patagonia, Argentina, were studied. Five coprolites of each midden were fully processed, rehydrated, homogenized, subjected to spontaneous sedimentation, and examined through light microscopy. Eight of the 10 examined rodent middens contained parasite eggs. The eggs of parasites were assigned to Heteroxynema (Cavioxyura) viscaciae Sutton & Hugot, 1989 and Helminthoxys sp. (Nematoda: Oxyuridae), Trichuris sp. (Nematoda: Trichuridae) and one unidentified nematode. Fossil rodent middens were assigned to Lagidium viscacia (Caviomorph: Chinchillidae). The excellent preservation of parasite remains in coprolites from fossil rodent middens provided an opportunity to perform paleoparasitological inferences. The results of this papers demonstrates that fossil rodent middens offer an excellent opportunity for the recovery of parasite remains for future paleoparasitological studies in the southwest of South America.  相似文献

Paleoparasitology is the application of conventional or molecular investigative techniques to archeological samples in order to reveal parasitic infection patterns among past populations. Although pioneering studies already have reported key paleoparasitological findings around the world, the same sorts of studies had not, until very recently, been conducted in sufficient numbers in Korea. Mummified remains of individuals dating to the Korean Joseon Dynasty actually have proved very meaningful to concerned researchers, owing particularly to their superb preservation status, which makes them ideal subjects for paleoparasitological studies. Over the past several years, our study series on Korean mummies has yielded very pertinent data on parasitic infection patterns prevailing among certain Joseon Dynasty populations. In this short review, we summarized the findings and achievements of our recent paleoparasitological examinations of Joseon mummies and discussed about the prospects for future research in this vein.  相似文献

Methyl halide-degrading bacteria are a diverse group of organisms that are found in both terrestrial and marine environments. They potentially play an important role in mitigating ozone depletion resulting from methyl chloride and methyl bromide emissions. The first step in the pathway(s) of methyl halide degradation involves a methyltransferase and, recently, the presence of this pathway has been studied in a number of bacteria. This paper reviews the biochemistry and genetics of methyl halide utilization in the aerobic bacteria Methylobacterium chloromethanicum CM4T, Hyphomicrobium chloromethanicum CM2T, Aminobacter strain IMB-1 and Aminobacter strain CC495. These bacteria are able to use methyl halides as a sole source of carbon and energy, are all members of the alpha-Proteobacteria and were isolated from a variety of polluted and pristine terrestrial environments. An understanding of the genetics of these bacteria identified a unique gene (cmuA) involved in the degradation of methyl halides, which codes for a protein (CmuA) with unique methyltransferase and corrinoid functions. This unique functional gene, cmuA, is being used to develop molecular ecology techniques to examine the diversity and distribution of methyl halide-utilizing bacteria in the environment and hopefully to understand their role in methyl halide degradation in different environments. These techniques will also enable the detection of potentially novel methyl halide-degrading bacteria.  相似文献

We present the results of a paleoparasitologic, paleogenetic and paleobotanic analysis of coprolites recovered during the excavation of the church La Concepción in Santa Cruz de Tenerife. Coprolites (n = 4) were rehydrated and a multidisciplinary analysis was conducted. The paleobotanic analysis showed numerous silicates, seeds and fruits of the family Moraceae. In the paleoparasitologic study, Ascaris sp. eggs (n = 344) were identified. The paleogenetic results confirmed the Ascaris sp. infection as well as the European origin of human remains. These findings contribute to our knowledge of ancient helminthes infections and are the first paleoparasitological record of Ascaris sp. infection in Spain.  相似文献

Excavations on Buldir Island, Aleutian Islands, Alaska, have revealed the remains of several features dated between the 13th and 17th centuries A.D. Soil from an open-air workshop, represented by 2 excavated pits, and a structure built of whale bones were sampled for evidence of parasites and microfloristic remains. Two groups of helminth eggs (Diphyllobothrium and Ankylostomidae) were identified in samples from the open-air pits; microfloristical remains were found in all samples. This is the first paleoparasitological analysis conducted on an Aleutian midden.  相似文献

In this study, we have used a technique designed to target short fragments containing informative mitochondrial substitutions to extend the temporal limits of DNA recovery and study the molecular phylogeny of Ursus deningeri. We present a cladistic analysis using DNA recovered from 400 kyr old U. deningeri remains, which demonstrates U. deningeri's relation to Ursus spelaeus. This study extends the limits of recovery from skeletal remains by almost 300 kyr. Plant material from permafrost environments has yielded DNA of this age in earlier studies, and our data suggest that DNA in teeth from cave environments may be equally well preserved.  相似文献

The inositol phosphates are a group of organic phosphorus compounds found widely in the natural environment, but that represent the greatest gap in our understanding of the global phosphorus cycle. They exist as inositols in various states of phosphorylation (bound to between one and six phosphate groups) and isomeric forms (e.g. myo, D-chiro, scyllo, neo), although myo-inositol hexakisphosphate is by far the most prevalent form in nature. In terrestrial environments, inositol phosphates are principally derived from plants and accumulate in soils to become the dominant class of organic phosphorus compounds. Inositol phosphates are also present in large amounts in aquatic environments, where they may contribute to eutrophication. Despite the prevalence of inositol phosphates in the environment, their cycling, mobility and bioavailability are poorly understood. This is largely related to analytical difficulties associated with the extraction, separation and detection of inositol phosphates in environmental samples. This review summarizes the current knowledge of inositol phosphates in the environment and the analytical techniques currently available for their detection in environmental samples. Recent advances in technology, such as the development of suitable chromatographic and capillary electrophoresis separation techniques, should help to elucidate some of the more pertinent questions regarding inositol phosphates in the natural environment.  相似文献

This study examines fossil microorganisms found in iron-rich deposits in an extreme acidic environment, the Tinto River in SW Spain. Both electron microscopy (SEM and TEM) and non-destructive in situ microanalytical techniques (EDS, EMP and XPS) were used to determine the role of permineralization and encrustation in preserving microorganisms forming biofilms in the sediments. Unicellular algae were preserved by silica permineralization of their cell walls. Bacterial biofilms were preserved as molds by epicellular deposition of schwertmannite around them. In the case of fungi and filamentous algae, we observed permineralization of cell structures by schwertmannite in the sediments. The extracellular polymeric matrix around the cells was also preserved through permineralization of the fibrillar component. The process of permineralization and deposition of iron-rich precipitates present in the acidic waters of Rio Tinto served to preserve many microfossils in an oxidizing environment, in which organic compounds would not normally be expected to persist. Studies of microbial fossil formation mechanisms in modern extreme environments should focus on defining criteria to identify inorganic traces of microbial life in past environments on Earth or other planets.  相似文献

The knowledge of the mature sperm proteome is undoubtedly the basis for understanding sperm function, the mechanisms responsible for fertilization, the reasons for infertility and possible treatments. The methods of sperm protein extraction depend mainly on the proteins of interest and the protein separation techniques that will be employed. The isolation of the membrane proteins appears to be most problematic step. Nevertheless, two-dimensional electrophoresis and mass spectrometry have become the main techniques used in human sperm protein analysis. We outline the present techniques used to examine the sperm proteome and data generated from studies on the human sperm and different types of male infertility. We present the most characteristic proteins that are involved in sperm function. Their value as biomarkers for diagnosis and treatment of infertility would require further validation. We focus on selected and critical studies of the human sperm proteome to present our subjective view of this fast-moving field.  相似文献

Paleoparasitology is the science that uses parasitological techniques for diagnosing parasitic diseases in the past. Advances in molecular biology brought new insights into this field allowing the study of archaeological material. However, due to technical limitations a proper diagnosis and confirmation of the presence of parasites is not always possible, especially in scarce and degraded archaeological remains. In this study, we developed a Molecular Paleoparasitological Hybridization (MPH) approach using ancient DNA (aDNA) hybridization to confirm and complement paleoparasitological diagnosis. Eight molecular targets from four helminth parasites were included: Ascaris sp., Trichuris trichiura, Enterobius vermicularis, and Strongyloides stercoralis. The MPH analysis using 18th century human remains from Praça XV cemetery (CPXV), Rio de Janeiro, Brazil, revealed for the first time the presence E. vermicularis aDNA (50%) in archaeological sites of Brazil. Besides, the results confirmed T. trichiura and Ascaris sp. infections. The prevalence of infection by Ascaris sp. and E. vermicularis increased considerably when MPH was applied. However, a lower aDNA detection of T. trichiura (40%) was observed when compared to the diagnosis by paleoparasitological analysis (70%). Therefore, based on these data, we suggest a combination of Paleoparasitological and MPH approaches to verify the real panorama of intestinal parasite infection in human archeological samples.  相似文献

Diatoms are abundant and diverse in many Antarctic freshwaters, with a general trend of decreasing diversity moving southwards. They form an important component of many benthic algal communities in streams and standing waters but are generally less common in the phytoplankton. Diatoms are excellent ecological indicator species and, because their remains are preserved in many sedimentary environments, there appears to be a great potential for palaeoenvironmental reconstruction and the examination of past diversity. A lack of taxonomic precision and consistency, coupled with the insufficient collection from some geographical areas, makes the estimation of the number of Antarctic diatom species problematic.  相似文献

A molecular paleoparasitological diagnostic approach was developed for Enterobius vermicularis. Ancient DNA was extracted from 27 coprolites from archaeological sites in Chile and USA. Enzymatic amplification of human mtDNA sequences confirmed the human origin. We designed primers specific to the E. vermicularis 5S ribosomal RNA spacer region and they allowed reproducible polymerase chain reaction identification of ancient material. We suggested that the paleoparasitological microscopic identification could accompany molecular diagnosis, which also opens the possibility of sequence analysis to understand parasite-host evolution.  相似文献

During a paleoparasitological survey of several animal mummies (Cavia aperea f. porcellus and Canis familiaris) from Chiribaya Baja, an archaeological site in Southern Peru, an unexpected find was made. In the well preserved fur, large numbers of mummified fleas (Pulex simulans/irritans) that parasitized the animals during life were encountered. Due to the relative recent event of the host mummification and the outstanding preservation of the fleas, an attempt for the retrieval of DNA was made. A DNA extraction and sequencing protocol for archaeological ectoparasitic remains has been established, taking additional studies for tissue and protein preservation into account. Tissue preservation was assessed with transmission electron microscopy and the protein preservation was tested through the racemisation ratios of aspartic acid. Regions of the 28S rDNA gene were successfully amplified and sequenced. Further research perspectives are outlined.  相似文献

The African giant rat Cricetomys gambianus Waterhouse is found in a wide variety of habitats in Ibadan district. These include an abandoned farm in a degraded deciduous forest, a zoological garden complex, Agodi; and a forest plantation of teak (Tectona grandis L.F.), human habitation and derived Guinea savanna. They located their burrows in the most heavily shaded parts of the habitats. Burrows were found mostly inside deserted termite mounds and amongst roots of trees probably to provide insulation from heat. They are nocturnal, omnivorous rodents, feeding on a wide variety of root crops, fruits, molluscs and insects. Fruits of the oil palm (Elaeis guineensis Jacq.) or their remains were found in all burrows. Information on habitat preferences, food habits, as well as that obtained from the dimensions and design of burrows which are reported in this paper later provided a basis for the design of cages and husbandry techniques for a programme of domestication of this rodent, which is currently being carried out at University of Ibadan, Nigeria.  相似文献

Ion channels are found in most plant membranes. They catalyse the rapid passive uniport of particular ions with varying selectivity. Planar lipid-bilayer (PLB) techniques have been developed to study the electrical activities of single ion channels in well-defined lipid and aqueous environments. They greatly facilitate both the biophysical and biochemical characterisation of ion channels and complement both conventional impaling electrode and membrane-patch voltage-clamping (patch-clamping) electrophysiological techniques applied in vivo. Bilayers can be formed across the end of patch-clamp pipettes or across apertures in specifically designed chambers. Ion channels in native membranes and purified, genetically altered or synthetic ion channels, proteins and peptides can all be studied in PLBs. The main applications of PLBs are (1) to study ion channels in membranes inaccessible to patch-clamp electrodes, (2) to provide a functional assay system during channel-protein purification and (3) to investigate the relationship between the molecular structure of ion channels and their conductance properties. In the present article we describe the techniques available for reconstitution and analysis of ion channels in PLBs and discuss how the PLB technique has been, and may be, useful to the study of plant ion channels.  相似文献

Ancient DNA has been discovered in many types of preserved biological material, including bones, mummies, museum skins, insects in amber and plant fossils, and has become an important research tool in disciplines as diverse as archaeology, conservation biology and forensic science. In archaeology, ancient DNA can contribute both to the interpretation of individual sites and to the development of hypotheses about past populations. Site interpretation is aided by DNA-based sex typing of fragmentary human bones, and by the use of genetic techniques to assess the degree of kinship between the remains of different individuals. On a broader scale, population migrations can be traced by studying genetic markers in ancient DNA, as in recent studies of the colonisation of the Pacific islands, while ancient DNA in preserved plant remains can provide information on the development of agriculture.  相似文献
