The concentration of copper which caused 50% mortality of bluegill in 96 h (96-h LC50) was between 4 and 16 ppm of the metal, copper. During the 6–8 day treatment period, sublethal concentrations of copper elicited a hypoactive locomotor response which was dependent on both concentration and time of exposure. Fish treated in 0.04, 0.08 and 0.4 ppm of copper were only 67, 61 and 44% as active, respectively, as they were prior to treatment. Locomotor activity decreased dramatically during the first 4 days of exposure to copper and remained low during further exposure.  相似文献

The concentration of hydrogen ions which caused 50% mortality of bluegill in 96 h (96 h LC50) was between pH 3.5 and 3.0 for four acids; sulphuric, nitric, phosphoric and hydrochloric. Therefore, it appears that any contribution by the anions of these acids to fish toxicity is similar. This suggests that the quantity rather than the quality of acids is the primary factor in fish toxicity brought about by acid precipitation.
At sublethal concentrations of acids, bluegill became hypoactive with respect to their swimming behaviour.  相似文献

The present study investigated lead effects on gill morphology, hematocrit, blood sodium, glucose, lipids, protein, and cholesterol of Prochilodus lineatus exposed to two sublethal lead concentrations for 96 h. Preliminary series of short-term static toxicity tests were run to determine LC50 (96 h) of lead in P. lineatus, which was 95 mg Pb.L-1. Therefore, lead concentrations tested in the sublethal experiments were 24 and 71 mg Pb.L-1, which correspond to 25% and 75% of the LC50 (96 h), respectively. Gills of P. lineatus exposed to both lead concentrations during 96 h presented a higher occurrence of histopathological lesions such as epithelial lifting, hyperplasia, and lamellar aneurism. P. lineatus did not show significant alterations in hematocrit during exposure to both lead concentrations. Fish exposed to the highest lead concentration showed a significant decrease in Na+ plasma concentration after 48 h, possibly reflecting a sodium influx rate decrease. P. lineatus exposed to both lead concentrations presented a "classical general adaptation syndrome to stress", as hyperglycemia associated with lowered lipids and proteins was reported. Stress-response magnitude was dose-dependent. While the response to the lowest lead concentration might represent adaptation, the highest concentration seems to characterize exhaustion.  相似文献

The effect of exposure to sub-lethal concentrations of cypermethrin, a synthetic pyrethroid pesticide, on biochemical parameters of muscle, blood and enzyme activities in brain, liver and kidney of the Indian major carp, Labeo rohita was studied. The sub-lethal exposure studies were done for up to 45 days at 1/10 and 1/50 of 96 h LC(50) of cypermethrin. The 96 h LC(50) was found to be 0.139 ppm. RNA levels decreased while DNA levels were elevated. Acid phosphatase was unchanged while alkaline phosphatase was depleted. Brain acetylcholinesterase activity was decreased significantly (P<0.05) over a period of 45 days at both cypermethrin concentrations. Lactate dehydrogenase activity in brain and liver was elevated, but inhibited in kidney. Succinate dehydrogenase and ATPase activities were depleted in brain, kidney and liver. There was a decrease in serum protein level over control at both concentrations of the pyrethroid. Blood glucose level and total leucocytes were elevated compared with controls at either concentration from day 15 to day 45. Haemoglobin percentage and total erythrocytes decreased in both sub-lethal concentrations. Extracts of the herb Datura stramonium were effective in countering the toxicity of this pesticide. Our data suggest that sub-lethal exposure of cypermethrin alters the biochemical, haematological parameters and enzymes of organs tissue and exert stress on the fish. Plant extracts may be useful in counteracting some of these effects.  相似文献

Marine planktonic algae are frequently exposed to metallic contaminants. Because heavy metals can be assimilated and accumulated by algal cells, they can then be transferred to higher trophic levels of food chains. We studied the effects of cadmium on protein production and the growth of the marine prasinophyte Tetraselmis gracilis (Kylin) Butcher. By means of toxicological assays, we estimated the LC50 of cadmium as 3.2 ppm and 1.8 ppm after 48 h and 96 h of exposure to this heavy metal, respectively. The growth curves and survival percentages of cell cultures in the presence of cadmium were determined, and a proportional reduction of both parameters with increasing metal concentrations was found. When chronically exposed to sublethal concentrations of cadmium, T. gracilis contained high levels of superoxide dismutase (SOD) activity, one of the main enzymes of the cell's antioxidant defense mechanism. Under these growth conditions, total SOD activity in crude extracts was increased by 41% (at 1.5 ppm) and 107% (at 3.0 ppm). Assays of SOD activity in nondenaturing polyacrylamide gels also showed a similar induction by cadmium. These results show that cadmium has potentially toxic properties since it significantly inhibited the growth of T. gracilis at low concentrations and promoted the induction of SOD activity, suggestive of an oxidative stress state. Besides being the first report of SOD in T. gracilis, this work describes experimental evidence of SOD induction by cadmium in this species.  相似文献

The effects of selenium (Se) on ruminant microbial fermentation were investigated in vitro using rumen microflora collected from a rumen-fistulated dairy cow. First, the effects ofl-selenomethionine (SeMet; at 0.2 or 2 ppm Se) in the presence or absence of wheat bran (WB, 500 mg per incubation flask) were evaluated. Second, the effects of several forms of Se (elemental Se: 50 ppm Se; sodium selenite: 2 ppm Se; SeMet: 2 ppm Se) were compared. Results showed that the amounts of short-chain fatty acids (SCFAs) tended to be increased by SeMet treatment, whereas SeMet in the presence of WB transiently suppressed fermentation. The addition of SeMet tended to increase the production of acetate while reducing the production of butyrate with and without WB supplementation. Among the different Se compounds tested, the amounts of SCFAs were greater with SeMet treatment, which yielded a higher proportion of acetate compared to other treatments. Selenite did not influence the total SCFAs concentrations; however, it increased the relative proportion of butyrate at the expense of acetate. Elemental Se did not significantly affect fermentation. Higher bacterial Se concentrations were observed for selenite than for SeMet. It was concluded that Se supplementation can influence rumen microbial fermentation and that Se compounds differ in this regard.  相似文献

The diterpenoids eunicin, eupalmerin acetate, and crassin acetate derived from octocorals were found to be toxic to the rotifer Brachionus plicatilis Müller and the amphipod Parhyale hawaiensis (Dana) at estimated concentrations ranging from 1 ppm to 100 ppm. Exposure of animals to a concentration ? 1 ppm resulted in either loss of swimming activity or mortality at 48 h. No ciliary loss was evident in the rotifer as had been reported in ciliated larvae of the nudibranch Phestilla sibogae Bergh.The cembranolides may serve as chemical defenses against predation and in the competition for space amongst the sessile organisms on coral reefs.  相似文献

The basal activity of Ca2+-ATPase in two isolates (NL56, UNC) and two clones (D6, W2) of P.falciparum was assessed. The effects of various concentrations of chloroquine phosphate and toxic concentrations of lead acetate were also evaluated in the clones and strains of P.falciparum. The Ca2+-ATPase activity was measured by monitoring the rate of release of inorganic phosphate from the gamma-position of ATP on spectrophotometer at 820nm wavelength. The various concentrations of chloroquine (3, 6, 9, 12, 18μg/ml) and lead acetate (5, 10, 20, 30, 40μg/ml) on Ca2+-ATPase activity were measured respectively. Chloroquine phosphate inhibited Ca2+-ATPase activity in both the isolates and the cloned strains of P.falciparum in concentration dependent manner. Median Inhibitory concentration of chloroquine (MIC50) estimated from the plot of activity against chloroquine concentration was found to be 2.6mg/ml at pH 7.4 for both the isolates and cloned strains examined. Lead acetate at concentrations 5-20μg/ml inhibited Ca2+-ATPase activity in concentration dependent manner in clone W2 (Chloroquine resistant strain) while the same range of concentrations of lead acetate stimulated the activity of the enzyme in clone D6 (Chloroquine sensitive strain).The inhibitory effect of lead acetate on the enzyme in clone D6 was observed at concentrations above 20μg/ml. The result also suggests that lead ions could modulate and moderate calcium ion homeostasis in P. falciparum via its effect on Ca2+-ATPase activity. Also sufficient influx of lead ions into P. falciparum may transform the biochemical or bioenergetics nature of chloroquine sensitive strain of P. falciparum (D6) to that similar to chloroquine resistant strain (W2). In conclusion, inhibition of Ca2+-ATPase activity of P.falciparum may be part of the mechanism of action of chloroquine in its use as chemotherapy for malaria. The study implies that populations simultaneously exposed to lead pollution and malaria infection may experience failure in chloroquine therapy.  相似文献

Phenylalanine ammonia lyase (PAL) activity was measured using HPLC in pea leaves following exogenous application of L-phenylalanine and ferulic acid. Treatment with different concentrations (50, 100, 150 ppm) of L-phenylalanine caused increased activity of PAL activity in comparison to control. In pea leaves treated with 50 ppm L-phenylalanine, maximum PAL activity was observed after 72 h of treatment. Application of ferulic acid first reduced PAL activity at lower concentration (50 ppm) but it further increased at higher concentrations of the compound (100 and 150 ppm) in pea leaves compared to control. Minimum PAL activity was 0.19 nM cinnamic acid/min/g fresh wt after 24 h at 50 ppm and then increased with time. Treatment with both compounds significantly increased the accumulation of phenolic acids and salicylic acid and reduced conidial germination of Erysiphe pisi on pea leaves. They were equally effective at 100 and 150 ppm in reducing conidial germination. Conidial germination on L-phenylalanine-treated leaves was 26% after 24 h and that on ferulic acid treated leaves 34% compared to control (46%). Foliar application of different concentrations of L-phenylalanine increased the level of ferulic acid in the leaves of pea plants. Maximum enzyme activity in terms of the accumulation of cinnamic acid (79.3 and 83.5 μg/g fresh wt) was observed following the application of L-phenylalanine after 24 and 48 h respectively. At 50 ppm, cinnamic acid accumulation in pea leaves was 35.6 and 39.4 μg/g fresh wt and 74.3 and 86.5 μg/g fresh wt at 100 ppm.  相似文献

The plasma membrane/mitochondrial fractions of Penaeus indicus postlarvae contain Mg2+-dependent ATPase, Na+,K+-stimulated ATPase, Na+-stimulated ATPase and K+-stimulated ATPase. The Na+,K+-activated, Mg2+-dependent ATPase was investigated further in relation to different pH and temperature conditions, and at various concentrations of protein, ouabain, ATP and ions in the incubation medium. In vitro and in vivo effects of lead were studied on the enzyme activity. In vitro lead inhibited the enzyme activity in a concentration-dependent manner with an IC50 value of 204.4 microM. In correlation with in vitro studies, in vivo investigations (both concentration and time dependent) of lead also indicated a gradual inhibition in enzyme activity. A maximum decrease of 85.3% was observed at LC50 (7.2 ppm) of lead for concentration-dependent experiments. In time-dependent studies, the decrease was maximal (81.7%) at 30 days of sublethal exposure (1.44 ppm). In addition, the substrate- and ion-dependent kinetics of Na+,K+-ATPase was studied in relation to in vitro exposure of lead; these studies suggest a non-competitive type of inhibition.  相似文献

The effects of sublethal doses of lead (as acetate) on blood parameters of adult male Bufo arenarum were studied. Toads received one single injection with 10, 25, 50 or 100 mg/kg of body weight, equivalent to approximately 1/90-1/10 of the 120 h-LD50; seven days after the injections, the hematocrit and the blood delta-aminolevulinic acid dehydratase (ALAD) activity were measured. Hematocrit of lead-injected animals did not exhibit significant changes respective to controls that received sodium acetate (range 29.8-38.8%). Blood lead concentrations were positively and significantly correlated with the injected metal doses. Blood ALAD activity declined proportionately to the doses of the metal as well as to its whole blood concentration. Because of its sensitivity and specificity, it was concluded that the activity of delta-ALAD may be adopted as a reliable biomarker of Bufo arenarum experimental lead intoxication.  相似文献

Selenium added to the diet significantly lowers the incidence of spontaneous mammary adenocarcinoma in female inbred C3H/St mice infected with the Bittner Milk Factor. Lead, 5 ppm, added to the drinking water in the form of the acetate, diminishes the uptake of selenium and reduces its anticarcinogenic effects, causing mammary tumors to appear with the same high incidence as in Se-unsupplemented controls. At higher lead concentrations in the drinking water (25 ppm), the overall tumor incidence is lowered, but tumor growth is significantly accelerated and the survival of tumor-bearing mice is shortened. Under the conditions of administration chosen, lead acts as a selenium antagonist and lowers the concentrations of selenium in liver, kidney, and spleen. The deposition of selenium, copper, and arsenic in bone is increased as compared to lead-unexposed controls.  相似文献

A comparative analysis of the influence of different nitric oxide forms on the character of the dehydration structuring of human serum samples was carried out. The effects of an NO-containing gas flow that was generated by a Plazon device (800 and 80 ppm), an experimental NO generator (20, 50, 75, and 100 ppm), as well as by glutathione-containing dinitrosyl iron complexes (3 mM/L) were investigated using 15 healthy donors. The influence of endogenous sodium on serum crystallization of intact and NO treated blood samples was evaluated. It was found that the effect of NO on the crystallogenic properties of blood serum is directly determined by its concentration and form (free or bound), as well as by the presence of reactive oxygen species in a gas flow. The most pronounced stimulatory effect was observed for the bound NO form, dinitrosyl iron complexes with glutathione ligands. Low NO concentrations modulated the crystallogenic properties of blood serum, while the best stimulatory effect was demonstrated by the gas flow that contained 20 ppm nitric oxide. In contrast, high NO concentrations (800 ppm) inhibited the crystallogenic activity of the biological medium due to an increase in the destruction of structural elements by many times, which lead to the formation of an additional band in the marginal zone of the microscope specimen.  相似文献

Modifications in the enzyme activity of lysozyme, a protein implied in the defence barrier of the organism, can be a good biomarker of alterations in the immune system as a result of exposure to toxic metal, such as lead. The aim of this work was to evaluate the effect of a 200?ppm dose of lead on lysozyme activity in blood, kidney, and lung, and also on tissue structure. Previously, the effect of lead acetate on lysozyme activity in vitro was determined; the in vitro results indicated that lead produced a decrease in enzyme activity. The activity loss was 16?% at 200?ppm of lead. Lead acetate was administered to Wistar rats by oral and intraperitoneal injections. An increase in lysozyme activity was observed in blood when lead was administered by introperitoneal route and in kidney by the oral route. The possible immunostimulation in kidney was discarded because of the structural alterations observed in the tissue. In lung, the decrease in specific lysozyme activity, for both routes of lead exposure, seems to indicate immunosupression, which was in accordance with the structural alterations observed in this tissue.  相似文献

Treatment with both the compounds significantly reduced conidial germination of Erysiphe pisi on pea leaves. They were equally effective at 100 and 150 ppm in reducing conidial germination. The conidial germination on l-phenylalanine-treated leaves was 26% after 24 h and that on ferulic acid-treated leaves was 34% as compared to the control (46%). Foliar application of different concentrations of l-phenylalanine increased the level of ferulic acid in the leaves of pea plants. Maximum accumulation of ferulic acid (79.3 and 83.5 μg/g fresh wt.) was observed following the application of l-phenylalanine after 24 h and 48 h, respectively. At 50 ppm, ferulic acid accumulation in pea leaves was 35.6 and 39.4 μg/g fresh wt. and 74.3 and 86.5 μg/g fresh wt. at 100 ppm.  相似文献

Efficacy of two tropical plant extracts for the control of mosquitoes   总被引:10,自引:0,他引:10  
The larvicidal activity of seed and leaf extracts of Calophyllum inophyllum and leaf extract of Rhinacanthus nasutus on the juveniles of Culex quinquefasciatus , Anopheles stephensi and Aedes aegypti was determined. Ethyl acetate (EA) soluble fractions of C. inophyllum and petroleum ether (PE) fraction of R. nasutus extracts showed very high larvicidal activity. Irrespective of the species of the mosquitoes tested, at concentrations ranging from 3.91 to 9.39 ppm, 9.04 to 35.49 ppm and 13.21 to 28.92 ppm of the active fractions of seed and leaf extracts of C. inophyllum and the leaf extract of R. nasutus , respectively, killed 50% of the treated larvae. The fractions also interfered with adult emergence. Exposure of the mosquitoes at any stage of their larval development to less than 0.86, 5.49 and 6.81 ppm of the active fractions of seed and leaf extracts of C. inophyllum and the leaf extract of R. nasutus , respectively, inhibited 50% of the treated larvae from emerging as adults. Experiments under laboratory and semi-field conditions showed that the activity of the extracts persisted for up to 10 days.  相似文献

The insecticidial and biological activity of the cyano-substituted neonicotinoid acetamiprid was determined against the western subterranean termite, Reticulitermes hesperus Banks (Isoptera: Rhinotermitidae). Acetamiprid was very active against termites in topical applications, with an LD50 = 0.02 ng per termite. Even though acetamiprid was extremely toxic in topical applications, deposits > or =50 ppm on sand were required to consistently provide >90% kill of termites within 7 d after a 1-h exposure. Termites were quickly affected by brief exposures to sand treated with 1 ppm acetamiprid and within 1 h, their locomotion was dramatically impaired. Acetamiprid was transferred from donors to recipients only when donors were held on deposits > or =50 ppm for 1 h. Deposits even as low as 1 ppm were repellent and termites failed to tunnel into treated sand, and there was no significant mortality. Exposure to acetamiprid impaired locomotion of termites as did other slow-acting neonicotinoids, such as imidacloprid. Acetamiprid was repellent at all concentrations tested, acting like type I pyrethroid treatments in soil. A new subcategory of type III soil termiticides is proposed that incorporates the sublethal and delayed effects observed in neonicotinoid insecticides, and repellency at certain concentrations.  相似文献

目的:探讨铅锌联合染毒对乳鼠颅骨成骨细胞增殖分化的影响。方法:分离并培养原代成骨细胞,加入不同浓度铅、锌培养48h,检测其对成骨细胞增殖的作用;用碱性磷酸酶试剂盒检测ALP活力。结果:在染铅48h后,当铅浓度≥10μmol/L时,细胞增殖功能下降(P<0.05);加锌干预48h后,铅+锌组细胞增殖功能均高于各自单独染铅组,其中铅(1μmol/L、10μmol/L)+锌(50μmol/L)组、铅(10)+锌(100)组与对照组间的差异具有统计学意义(P<0.05)。铅干预48h后,100μmol/L铅组的ALP活力显著下(P<0.05),给予锌干预的铅锌联合染毒组,各组ALP活力均有增加,其中铅(1μmol/L、10μmol/L)+锌(50μmol/L)组ALP活力均高于对照组,而铅(100μmol/L)+锌(50μmol/L)组ALP活力低于对照组,差异均有统计学意义(P<0.05)。结论:铅对成骨细胞有毒性作用,影响其增殖和分化功能;50μmol/L锌在一定程度上可以拮抗铅对成骨细胞增殖和分化功能的损伤,且对ALP活力的作用更显著,为铅中毒骨病的防治提供一定的科学依据。  相似文献

This study was designed to evaluate the effects of different amounts of dietary vitamin E on platelet tocopherol levels and 12-lipoxygenase activity when exogenous arachidonic acid was used as substrate. Weanling male Sprague-Dawley rats were fed diets containing 0, 50, and 5000 ppm of D-alpha-tocopherol acetate for 4 months. Platelet tocopherol was increased with increasing concentrations of dietary vitamin E; however, the conversion of exogenously added arachidonate by platelet to 12-HETE (12-hydroxyeicosatetraenoic acid) and thromboxane B2 from these three dietary groups was essentially the same. This study provides direct evidence that platelet 12-lipoxygenase activity is independent of its vitamin E content when exogenously added arachidonate was used as substrate.  相似文献
