The bloom‐forming cyanobacterium Microcystis aeruginosa Kütz 854 was cultured with 1.05 W·m?2 UV‐B for 3 h every day, and its growth, pigments, and photosynthesis were investigated. The specific growth rates represented by chl a concentration and OD750 were inhibited 8% and 9% by UV‐B exposure, respectively. Six days of UV‐B treatment significantly reduced cellular contents of phycocyanin and allophycocyanin by 32% and 62%, respectively, and markedly increased the carotenoid content by 27%, but had little effect on the chl a content. The initial values of optimal photosynthetic efficiency for UV‐B treated samples were, respectively, 52%, 87%, and 93% of controls on days 4, 7, and 10 of growth. The light‐saturated photosynthetic rates at day 6 were significantly lower than controls grown without UV‐B. The probability of electron transfer beyond QA decreased during UV‐B exposure, and this indicated that the acceptor side of PSII was one of main damage sites. The adaptation of M. aeruginosa 854 to UV‐B radiation could be observed from light‐saturated photosynthetic rates on day 13 and diurnal changes of chl fluorescence during the late growth phase. When both exposed to higher UV‐B, samples cultured under 1.05 W·m?2 UV‐B for 9 days recovered faster than controls. It is suggested that M. aeruginosa 854 had at least three adaptive strategies to cope with the enhanced UV‐B: increasing the synthesis of carotenoids to counteract reactive oxidants caused by UV‐B exposure, degrading phycocyanin and allophycocyanin to avoid further damage to DNA and reaction centers, and enhancing the repair of UV‐B induced damage to the photosynthetic apparatus.  相似文献

This study assessed the implication of oxidative stress in the mortality of cells of Microcystis aeruginosa Kütz. Cultures grown at 25°C were exposed to 32°C, darkness, and hydrogen peroxide (0.5 mM) for 96 h. The cellular abundance, chl a concentration and content, maximum photochemical efficiency of PSII (Fv/Fm ratio), intracellular oxidative stress (determined with dihydrorhodamine 123 [DHR]), cell mortality (revealed by SYTOX‐labeling of DNA), and activation of caspase 3–like proteins were assessed every 24 h. The presence of DNA degradation in cells of M. aeruginosa was also assessed using a terminal deoxynucletidyl transferase‐mediated dUTP nick end labeling (TUNEL) assay at 96 h. Transferring cultures from 25°C to 32°C was generally beneficial to the cells. The cellular abundance and chl a concentration increased, and the mortality remained low (except for a transient burst at 72 h) as did the oxidative stress. In darkness, cells did not divide, and the Fv/Fm continuously decreased with time. The slow increase in intracellular oxidative stress coincided with the activation of caspase 3–like proteins and a 15% and 17% increase in mortality and TUNEL‐positive cells, respectively. Exposure to hydrogen peroxide had the most detrimental effect on cells as growth ceased and the Fv/Fm declined to near zero in less than 24 h. The 2‐fold increase in oxidative stress matched the activation of caspase 3–like proteins and a 40% and 37% increase in mortality and TUNEL‐positive cells, respectively. These results demonstrate the implication of oxidative stress in the stress response and mortality of M. aeruginosa.  相似文献

The processes of CO2 acquisition were characterized for the acid‐tolerant, free‐living chlorophyte alga, CPCC 508. rDNA data indicate an affiliation to the genus Coccomyxa, but distinct from other known members of the genus. The alga grows over a wide range of pH from 3.0 to 9.0. External carbonic anhydrase (CA) was detected in cells grown above pH 5, with the activity increasing marginally from pH 7 to 9, but most of the CA activity was internal. The capacity for HCO3? uptake of cells treated with the CA inhibitor acetazolamide (AZA), was investigated by comparing the calculated rate of uncatalyzed CO2 formation with the rate of photosynthesis. Active bicarbonate transport occurred in cells grown in media above pH 7.0. Monitoring CO2 uptake and O2 evolution by membrane‐inlet mass spectrometry demonstrated that air‐grown cells reduced the CO2 concentration in the medium to an equilibrium concentration of 15 μM, but AZA‐treated cells caused a drop in extracellular CO2 concentration to a compensation concentration of 27 μM at pH 8.0. CO2‐pulsing experiments with cells in the light indicated that the cells do not actively take up CO2. An internal pool of unfixed inorganic carbon was not detected at the CO2 compensation concentration, probably because of the lack of active CO2 uptake, but was detectable at times before compensation point was reached. These results indicate that this free‐living Coccomyxa possesses a CO2‐concentrating mechanism (CCM) due to an active bicarbonate‐uptake system, unlike the Coccomyxa sp. occurring in symbiotic association with lichens.  相似文献

Growth and photosynthesis of an edible cyanobacterium, Ge‐Xian‐Mi (Nostoc), were investigated with differently sized colonies. Both photosynthesis and growth were dependent on the colony size. Compared with larger ones, smaller colonies grew faster regardless of the levels of light and temperature for culture and showed higher values of maximal net photosynthetic rate, apparent quantum yield, light‐saturating and compensating points, and dark respiration. The ratios of chl a content and mass to surface area of a colony increased and that of chl a to mass or mass to volume of a colony decreased with increased colonial sizes. A Ge‐Xian‐Mi colony appeared to increase its chl a content per surface area, enhancing the light‐shading effect; however, at the same time it decreased its mass density on a volume basis, minimizing the enhanced effects of shading and diffusion barrier caused by the thickening outer layer with increasing colony size during growth.  相似文献

The influence of temperature and inorganic carbon (Ci) concentration on photosynthesis was examined in whole corals and samples of cultured symbiotic dinoflagellates (Symbiodinium sp.) using combined measurements from a membrane inlet mass spectrometer and chl a fluorometer. In whole corals, O2 production at 26°C was significantly limited at Ci concentrations below ambient seawater (~2.2 mM). Further additions of Ci up to ~10 mM caused no further stimulation of oxygenic photosynthesis. Following exposure to 30°C (2 d), net oxygen production decreased significantly in whole corals, as a result of reduced production of photosynthetically derived oxygen rather than increased host consumption. Whole corals maintained a rate of oxygen evolution around eight times lower than cultured Symbiodinium sp. at inorganic carbon concentrations <2 mM, but cultures displayed greater levels of photoinhibition following heat treatment (30°C, 2 d). Whole corals and cultured zooxanthellae differed considerably in their responses to Ci concentration and moderate heat stress, demonstrating that cultured Symbiodinium make an incongruous model for those in hospite. Reduced net oxygen evolution, in whole corals, under conditions of low Ci (<2 mM) has been interpreted in terms of possible sink limitation leading to increased nonphotochemical energy dissipation. The advantages of combined measurement of net gas exchange and fluorometry offered by this method are discussed.  相似文献

The mechanism of inorganic carbon (Ci) acquisition by the economic brown macroalga, Hizikia fusiforme (Harv.) Okamura (Sargassaceae), was investigated to characterize its photosynthetic physiology. Both intracellular and extracellular carbonic anhydrase (CA) were detected, with the external CA activity accounting for about 5% of the total. Hizikia fusiforme showed higher rates of photosynthetic oxygen evolution at alkaline pH than those theoretically derived from the rates of uncatalyzed CO2 production from bicarbonate and exhibited a high pH compensation point (pH 9.66). The external CA inhibitor, acetazolamide, significantly depressed the photosynthetic oxygen evolution, whereas the anion‐exchanger inhibitor 4,4′‐diisothiocyano‐stilbene‐2,2′‐disulfonate had no inhibitory effect on it, implying the alga was capable of using HCO3? as a source of Ci for its photosynthesis via the mediation of the external CA. CO2 concentrations in the culture media affected its photosynthetic properties. A high level of CO2 (10,000 ppmv) resulted in a decrease in the external CA activity; however, a low CO2 level (20 ppmv) led to no changes in the external CA activity but raised the intracellular CA activity. Parallel to the reduction in the external CA activity at the high CO2 was a reduction in the photosynthetic CO2 affinity. Decreased activity of the external CA in the high CO2 grown samples led to reduced sensitiveness of photosynthesis to the addition of acetazolamide at alkaline pH. It was clearly indicated that H. fusiforme, which showed CO2‐limited photosynthesis with the half‐saturating concentration of Ci exceeding that of seawater, did not operate active HCO3? uptake but used it via the extracellular CA for its photosynthetic carbon fixation.  相似文献

The combined effects of different light and aqueous CO2 conditions were assessed for the Southern Ocean diatom Proboscia alata (Brightwell) Sundström in laboratory experiments. Selected culture conditions (light and CO2(aq)) were representative for the natural ranges in the modern Southern Ocean. Light conditions were 40 (low) and 240 (high) μmol photons · m?2 · s?1. The three CO2(aq) conditions ranged from 8 to 34 μmol · kg?1 CO2(aq) (equivalent to a pCO2 from 137 to 598 μatm, respectively). Clear morphological changes were induced by these different CO2(aq) conditions. Cells in low [CO2(aq)] formed spirals, while many cells in high [CO2(aq)] disintegrated. Cell size and volume were significantly affected by the different CO2(aq) concentrations. Increasing CO2(aq) concentrations led to an increase in particulate organic carbon concentrations per cell in the high light cultures, with exactly the opposite happening in the low light cultures. However, other parameters measured were not influenced by the range of CO2(aq) treatments. This included growth rates, chlorophyll a concentration and photosynthetic yield (FV/FM). Different light treatments had a large effect on nutrient uptake. High light conditions caused an increased nutrient uptake rate compared to cells grown in low light conditions. Light and CO2 conditions co‐determined in various ways the response of P. alata to changing environmental conditions. Overall P. alata appeared to be well adapted to the natural variability in light availability and CO2(aq) concentration of the modern Southern Ocean. Nevertheless, our results showed that P. alata is susceptible to future changes in inorganic carbon concentrations in the Southern Ocean.  相似文献

Transfer of N2 and CO2 fixation products from the bloom forming blue-green alga, Anabaena oscillarioides Bory, to attached and free swimming bacteria is common during active growth of the former. Incubation with 15N2 and 14CO2 followed by size fractionation filtration reveals that: i) magnitudes of fixed N and C excretion, relative to N2 and CO2 fixation, are dictated by dissolved inorganic carbon (DIC) availability for A. oscillarioides photosynthetic production, ii) associated bacteria exhibit preferences for recently fixed excreted N compounds, iii) bacterial utilization of excreted N is independent of ambient light conditions, and iv) lag times between N2 fixation and detectable bacterial assimilation of excreted fixed N compounds are ca. 1–2 h. Both 14NH4Cl dilution and Hg(NH3)2 Cl2 precipitation techniques indicate that NH3 is a major excretion product from A. oscillarioides, particularly during DIC limited growth. Active N and C excretion and transfer to associated bacteria are features of viable A. oscillarioides filaments. Hence, transfer of these metabolites reflects complex mutualistic, and possibly symbiotic associations rather than solely signaling senescence.  相似文献

The role of carbonic anhydrase (CA) in inorganic carbon acquisition (dissolved inorganic carbon, DIC) was examined in Alboran Sea phytoplankton assemblages. The study area was characterized by a relatively high variability in nutrient concentration and in abundance and taxonomic composition of phytoplankton. Therefore, the relationship between environmental variability and capacity for using HCO3? via external CA (eCA) was examined. Acetazolamide (AZ, an inhibitor of eCA) inhibited the primary productivity (PP) in 50% of the samples, with inhibition percentages ranging from 13% to 60%. The AZ effect was more prominent in the samples that exhibited PP >1 mg C · m?3 · h?1, indicating that the contribution of eCA to the DIC photosynthetic flux was irrelevant at low PP. The inhibition of primary productivity by AZ was significantly correlated to the abundance of diatoms. However, there was no a relationship between AZ effect and CO2 partial pressure (pCO2) or nutrient concentration, indicating that the variability in the PP percentage supported by eCA was mainly due to differences in taxonomic composition of the phytoplankton assemblages. Ethoxyzolamide (EZ, an inhibitor of both external and internal CA) affected 13 of 14 analyzed samples, with PP inhibition percentages varying from 50% to 95%. The effects of AZ and EZ were partially reversed by doubling DIC concentration. These results imply that CA activity (external and/or internal) was involved in inorganic carbon acquisition in most the samples. However, EZ effect was not correlated with pCO2 or taxonomic composition of the phytoplankton.  相似文献

Inorganic phosphorus (Pi) and carbon (here, CO2) potentially limit the photosynthesis of phytoplankton simultaneously (colimitation). A single Pi limitation generally reduces photosynthesis, but the effect of a colimitation is not known. Therefore, photosynthesis was measured under Pi‐limited conditions and high and low CO2, and osmo‐mixotrophic (i.e., growth in the presence of glucose) conditions that result in colimiting conditions in some cases. The green alga Chlamydomonas acidophila Negoro was used as a model organism because low Pi and CO2 concentrations likely influence its photosynthetic rates in its natural environment. Results showed a decreasing maximum photosynthetic rate (Pmax) and maximum quantum yield (ΦII) with increasing Pi limitation. In addition, a Pi limitation enhanced the relative contribution of dark respiration to Pmax (Rd:Pmax) but did not influence the compensation light intensity. Pmax positively correlated with the cellular RUBISCO content. Osmo‐mixotrophic conditions resulted in similar Pmax, ΦII, and RUBISCO content as in high‐CO2 cultures. The low‐CO2 cultures were colimited by Pi and CO2 and had the highest Pmax, ΦII, and RUBISCO content. Colimiting conditions for Pi and CO2 in C. acidophila resulted in an enhanced mismatch between photosynthesis and growth rates compared to the effect of a single Pi limitation. Primary productivity of colimited phytoplankton could thus be misinterpreted.  相似文献

Chlorella emersonii Shihira et Krauss var. emersonii exhibits ‘C4-like’ gas exchange characteristics when grown at air levels of CO2, but is ‘C3-like’ when grown with extra CO2. The total inorganic carbon concentration, and the free CO2 concentration, averaged over the cell interior are higher in air-adapted cells than can be accounted for by passive CO2 equilibration from the medium and the mean intracellular pH value. The ‘extra’ inorganic C in the air-grown cells probably cannot all be accounted for in terms of binding to proteins and requires an active transport process to account for it. The electrical potential of the cell interior becomes more negative when the ‘CO2 concentrating mechanism’ is operative; this is most readily explained if the active step in inorganic C accumulation is primary active uniport of HCO3?. Since the ‘CO2 concentrating mechanism’ can operate when CO2 is the species crossing the outer permeation barrier, it is suggested that the site of active HCO3? transport in Chlorella (and other eukaryotes) is the chloroplast envelope, and the plasmalemma in cyanobacteria. This scheme explains the obligatory role of the de-repressed carbonic anhydrase in C4-like photosynthesis in algae, but some other data support an explanation of C4-like photosynthesis in terms of special properties of carbonic anhydrase as a carbon donor to RuBP carboxylase-oxygenase.  相似文献

The short‐term and long‐term effects of elevated CO2 on photosynthesis and respiration were examined in cultures of the marine brown macroalga Hizikia fusiformis (Harv.) Okamura grown under ambient (375 μL · L?1) and elevated (700 μL · L?1) CO2 concentrations and at low and high N availability. Short‐term exposure to CO2 enrichment stimulated photosynthesis, and this stimulation was maintained with prolonged growth at elevated CO2, regardless of the N levels in culture, indicating no down‐regulation of photosynthesis with prolonged growth at elevated CO2. However, the photosynthetic rate of low‐N‐grown H. fusiformis was more responsive to CO2 enrichment than that of high‐N‐grown algae. Elevation of CO2 concentration increased the value of K1/2(Ci) (the half‐saturation constant) for photosynthesis, whereas high N supply lowered it. Neither short‐term nor long‐term CO2 enrichment had inhibitory effects on respiration rate, irrespective of the N supply, under which the algae were grown. Under high‐N growth, the Q10 value of respiration was higher in the elevated‐CO2‐grown algae than the ambient‐CO2‐grown algae. Either short‐ or long‐term exposure to CO2 enrichment decreased respiration as a proportion of gross photosynthesis (Pg) in low‐N‐grown H. fusiformis. It was proposed that in a future world of higher atmospheric CO2 concentration and simultaneous coastal eutrophication, the respiratory carbon flux would be more sensitive to changing temperature.  相似文献

The low CO2 concentration in seawater poses severe restrictions on photosynthesis, especially on those species with form II RUBISCO. We found that the potentially toxic dinoflagellate Protoceratium reticulatum Clap. et J. Lachm. possesses a form II RUBISCO. To cast some light on the mechanisms this organism undergoes to cope with low CO2 availability, we compared cells grown at atmospheric (370 ppm) and high (5000 ppm) CO2 concentrations, with respect to a number of physiological parameters related to dissolved inorganic carbon (DIC) acquisition and assimilation. The photosynthetic affinity for DIC was about one order of magnitude lower in cells cultivated at high [CO2]. End‐point pH‐drift experiments suggest that P. reticulatum was not able to efficiently use HCO3? under our growth conditions. Only internal carbonic anhydrase (CA) activity was detected, and its activity decreased by about 60% in cells cultured at high [CO2]. Antibodies raised against a variety of algal CAs were used for Western blot analysis: P. reticulatum extracts only cross‐reacted with anti‐ß‐CA sera, and the amount of immunoreactive protein decreased in cells grown at high [CO2]. No pyrenoids were observed under all growth conditions. Our data indicate that P. reticulatum has an inducible carbon‐concentrating mechanism (CCM) that operates in the absence of pyrenoids and with little intracellular CO2 accumulation. Calculations on the impact of the CA activity to photosynthetic growth [CO2] suggest that it is an essential component of the CCM of P. reticulatum and is necessary to sustain the photosynthetic rates observed at ambient CO2.  相似文献

The morpho‐functional patterns of photosynthesis, measured as 14C‐fixation and chl fluorescence of PSII, also as affected by different doses of UV radiation in the laboratory were examined in the South Pacific kelp Lessonia nigrescens Bory of the coast of Valdivia, Chile (40°S). The results indicated the existence of longitudinal thallus profiles in physiological performance. In general, blades exhibited higher rates of carbon fixation and pigmentation as compared with stipes and holdfasts. Light‐independent 14C fixation (LICF) was high in meristematic zones of the blades (3.5 μmol 14C·g?1 fresh weight [FW]·h?1), representing 2%–16% (percentage ratio) of the photosynthetic 14C fixation (20 μmol 14C·g?1 FW·h?1). Exposures to UV radiation indicated that biologically effective UV‐B doses (BEDphotoinhibition300) of 200–400 kJ·m?2 (corresponding to current daily doses measured in Valdivia on cloudless summer days) inhibit photosynthetic 14C fixation of blades by 90%, while LICF was reduced by 70%. The percentage ratio of LICF to photosynthetic 14C fixation increased under UV exposure to 45%. Primary light reactions measured as maximum quantum yield (Fv/Fm) and electron transport rate (ETR) indicated a higher UV susceptibility of blades as compared with stipes and holdfasts: after a 48 h exposure to UV‐B, the decrease in the blades was close to 30%, while in the stipes and holdfasts it was <20%. The existence of translocation of labeled carbon along the blades suggests that growth at the meristem may be powered by nonphotosynthetic processes. A possible functional role of LIFC, such as during reduction of photosynthetic carbon fixation due to enhanced UV radiation, is discussed. These results in general support the idea that the UV‐related responses in Lessonia are integrated in the suite of morpho‐functional adaptations of the alga.  相似文献

Trichodesmium N2 fixation has been studied for decades in situ and, recently, in controlled laboratory conditions; yet N2‐fixation rate estimates still vary widely. This variance has made it difficult to accurately estimate the input of new nitrogen (N) by Trichodesmium to the oligotrophic gyres of the world ocean. Field and culture studies demonstrate that trace metal limitation, phosphate availability, the preferential uptake of combined N, light intensity, and temperature may all affect N2 fixation, but the interactions between growth rate and N2 fixation have not been well characterized in this marine diazotroph. To determine the effects of growth rate on N2 fixation, we established phosphorus (P)–limited continuous cultures of Trichodesmium, which we maintained at nine steady‐state growth rates ranging from 0.27 to 0.67 d?1. As growth rate increased, biomass (measured as particulate N) decreased, and N2‐fixation rate increased linearly. The carbon to nitrogen ratio (C:N) varied from 5.5 to 6.2, with a mean of 5.8 ± 0.2 (mean ± SD, N = 9), and decreased significantly with growth rate. The N:P ratio varied from 23.4 to 45.9, with a mean of 30.5 ± 6.6 (mean ± SD, N = 9), and remained relatively constant over the range of growth rates studied. Relative constancy of C:N:P ratios suggests a tight coupling between the uptake of these three macronutrients and steady‐state growth across the range of growth rates. Our work demonstrates that growth rate must be considered when planning studies of the effects of environmental factors on N2 fixation and when modeling the impact of Trichodesmium as a source of new N to oligotrophic regions of the ocean.  相似文献
