This paper presents a study of the metabolic response (dark respiration intensity, photosystem II efficiency, metabolic activity) and the yield of barley treated with 24-epibrassinolide and subjected to high-temperature stress. Transport of exogenously applied 24-epibrassinolide in barley and changes in the profile of brassinosteroids that may occur in tissues after 24-epibrassinolide application were also studied. The water solution of 24-epibrassinolide (0.005 and 0.25 mg dm−3) was applied via infiltration of the first and second leaves of 12-day-old seedlings. Control plants were treated with water solution of hormone solvent (ethanol). Fifteen-day-old plants were subjected to high-temperature stress (42°C for 3 h). The influence of hormone treatment and stress conditions was investigated in the first and second leaves based on measurements of PSII efficiency. The aftereffect of plant treatment was investigated in the seventh leaf (measurements of PS II efficiency, dark respiration intensity, metabolic activity). The transport efficiency of 24-epibrassinolide exogenously applied to the first and second leaves, as well as the profile of other brassinosteroids, was also measured on the seventh leaf. Finally, yield formation was estimated. 24-epibrassinolide showed protective action, which manifested itself in the improved functioning of PSII, but this was observed in case of higher hormone concentration and only for the first, older leaf. The PSII efficiency of the seventh leaf was similar in plants treated with brassinosteroid and in the control plants, whereas the respiration intensity and metabolic activity decreased in plants previously treated with higher concentration of 24-epibrassinolide. The use of a higher hormone concentration at the seedling phase ultimately resulted also in lower crop yield. Brassinosteroids—brassinolide and castasterone—were detected in barley leaves. 24-epibrassinolide was found only in trace amounts in control plants. Its exogenous application directly to the apoplast of the first and second leaves resulted in an increase in the 24-epibrassinolide content in the seventh leaf, but did not depend on whether a high or low concentration had been applied to the plants.  相似文献

Brassinosteroids which show high structural similarity to animal steroid hormones elicit a variety of growth responses when exogeneously applied to plant tissues. Thus far however, the function of endogeneous brassinosteroids in higher plants has been unclear. This paper describes three extremely dwarfed Arabidopsis thaliana mutants, cbb1 (dwf1-6), cbb2 and cbb3, which are impared in cell elongation controlled by brassinosteroids. While cbb1 (dwf1-6) and cbb3 can be phenotypically normalized to wild-type by feeding with brassinosteroids indicating deficiencies of brassinosteroid biosynthesis, cbb2 is brassinosteroid-insensitive and defines a function required for further metabolic conversion necessary for biological activity or for perception/signal transduction of these growth-regulating plant steroid hormones. Expression of the meri5 and TCH4 genes is low in all three cbb mutants and can be restored to wild-type levels by brassinosteroid treatment in the cbb1 (dwf1-6) and cbb3 mutants but are unaffected in the cbb2 mutant. These data indicate that brassinosteroids are essential for proper plant development and play an important role in the control of cell elongation.  相似文献

The isoprenoid biosynthetic pathway provides intermediates for the synthesis of a multitude of natural products which serve numerous biochemical functions in plants: sterols (isoprenoids with a C30 backbone) are essential components of membranes; carotenoids (C40) and chlorophylls (which contain a C20 isoprenoid side-chain) act as photosynthetic pigments; plastoquinone, phylloquinone and ubiquinone (all of which contain long isoprenoid side-chains) participate in electron transport chains; gibberellins (C20), brassinosteroids (C30) and abscisic acid (C15) are phytohormones derived from isoprenoid intermediates; prenylation of proteins (with C15 or C20 isoprenoid moieties) may mediate subcellular targeting and regulation of activity; and several monoterpenes (C10), sesquiterpenes (C15) and diterpenes (C20) have been demonstrated to be involved in plant defense. Here we present a comprehensive analysis of genes coding for enzymes involved in the metabolism of isoprenoid-derived compounds in Arabidopsis thaliana. By combining homology and sequence motif searches with knowledge regarding the phylogenetic distribution of pathways of isoprenoid metabolism across species, candidate genes for these pathways in A. thaliana were obtained. A detailed analysis of the vicinity of chromosome loci for genes of isoprenoid metabolism in A. thaliana provided evidence for the clustering of genes involved in common pathways. Multiple sequence alignments were used to estimate the number of genes in gene families and sequence relationship trees were utilized to classify their individual members. The integration of all these datasets allows the generation of a knowledge-based metabolic map of isoprenoid metabolic pathways in A. thaliana and provides a substantial improvement of the currently available gene annotation.  相似文献

Brassinosteroids are of ubiquitous occurrence in plants and elicit a wide spectrum of physiological responses. In our study, brassinosteroids were isolated and identified in topmost dormant leaves of tea plants. Six brassinosteriods, i.e. 6-deoxocastasterone, 24-epibrassinolide,3-dehydroteasterone, typhasterol, 3-deoxotyphasterol and 28-homodolicholide, were isolated and identified by GC–MS. All the brassinosteroids identified belong to important components of early and late C6 oxidation pathways proposed for brassinosteroids biosynthesis in plants. It suggests that both pathways are operating in tea to produce brassinolide, the most active brassinosteroid biologically.  相似文献

Brassinosteroids are a class of steroidal phytohormones with high growth-promoting properties. The preferred side-chain conformations of 10 brassinosteroids were determined by means of detailed NMR investigations and molecular-modeling studies. Vacuum conformations obtained by simulated annealing calculations and Boltzmann statistical analysis were compared with solution conformations derived from NOE experiments and molecular dynamic simulations, and with X-ray structures. In general, results from simulated annealing calculations and NMR-supported molecular dynamics simulations are in good agreement. For some of the compounds investigated the conformation was less well-defined at the end of the side-chain. It could be shown that the energetically most favorable and most probable conformations also include the conformations obtained by NMR supported molecular-dynamics calculations and by X-ray analysis. For the most bioactive compound brassinolide (1) the majority of conformations show a side-chain bent towards the -face of the steroid skeleton, whereas for the less bioactive brassinosteroids, conformations with straight side-chains or side-chains bent towards the -face are more frequent.Electronic Supplementary Material available.  相似文献

Effect of pre-treatments of 1 and 5 M epibrassinolide or homobrassinolide prior to water stress induction on changes in root nodulation and contents of endogenous abscisic acid (ABA) and cytokinin trans-zeatin riboside (ZR), and nitrogenase activity was investigated in the nodulated roots of Phaseolus vulgaris L. cv. Arka Suvidha. Brassinosteroids in the unstressed plants increased root nodulation, ZR content and nitrogenase activity, and also ameliorated their stress-induced decline in the nodulated roots. The ABA contents in the nodules of control or stressed plants were not altered by brassinosteroids treatment. There was an increase in pod yield by brassinosteroids treatment (5 M) in the irrigated control as well as stressed plants without influencing pod number or pod length. Among the brassinosteroids, epibrassinolide was relatively more effective.  相似文献

Brassinosteroids are found in a wide range of organisms from lower to higher plants. They are steroidal plant hormones implicated in the promotion of plant growth and development. Brassinosteroid metabolism has long been known to be altered in plants responding to abiotic stresses and to undergo profound changes in plants interacting with bacterial, fungal and viral pathogens. Evaluation of biological activity of new synthetic brassinolide analogs   总被引:1,自引:0,他引:1  
The responses of plants to exogenous treatment with new synthetic brassinosteroids (BRs) were assessed and compared with the activity of natural 24-epibrassinolide (24-EPI). Morphological experiments on plants of pea and flax showed that the boundary between stimulatory and inhibitory concentrations of individual BRs and 24-EPI used is very narrow and differs also with the plant species. Moreover brassinosteroids can exhibit effects similar to various other plant hormones. This was proven also in our experiments, where auxin, anti-auxin and cytokinin like effects were achieved by BRs application. One of the explanations of the different morphological effects could be the influence of brassinosteroid application on the level of endogenous hormones. There are changes in the levels of indole-3-acetic acid, 6-benzylaminopurine, trans-zeatin and dihydrozeatin in rape and wheat plants caused by BR 4 and 24-EPI application, but there is no general trend explaining unequivocally their influence. The fact that all tested BRs significantly increased the dry weight accumulation in comparison with non-treated reference rape plants can be accounted for the known BRs characteristics to avoid biotic stresses.  相似文献

Brassinosteroids play an important role in growth and development of plants. They have been reported universally in all the plants. The present study deals with the presence of these compounds in immature tea seeds. Five brassinosteroids, i.e. 6-deoxo-28-norcathasterone, 6-deoxo-28-norteasterone, 3-dehydro-6-deoxo-28-norteasterone, 6-deoxo-28-nortyphasterol and 6-deoxo-28-norcastasterone have been isolated and identified by GC–MS. The identified brassinosteroids and their derivatives are active constituents of late C-6 oxidation pathway, thereby suggesting the biosynthesis of brassinosteroids in tea seeds by late C-6 oxidation pathway.  相似文献

Brassinosteroids are now considered as the sixth group of hormones in plants. As brassinosteroids influence varied growth and development processes such as growth, germination of seeds, rhizogenesis, flowering, senescence and abscission, they are considered as plant hormones with pleiotropic effects. The effect of 28-homobrassinolide and 24-epibrassinolide on ripening of tomato pericarp discs was studied. Application of brassinosteroids to pericarp discs resulted in elevated levels of lycopene and lowered chlorophyll levels. In addition brassinosteroid-treated pericarp discs exhibited decreased ascorbic acid and increased carbohydrate contents. Fruit ripening as induced by brassinosteroids was associated with increase in ethylene production. The study revealed the ability of brassinosteroids in accelerating fruit-senescence.  相似文献

Ligand binding may involve a wide range of structural changes in the receptor protein, from hinge movement of entire domains to small side-chain rearrangements in the binding pocket residues. The analysis of side chain flexibility gives insights valuable to improve docking algorithms and can provide an index of amino-acid side-chain flexibility potentially useful in molecular biology and protein engineering studies. In this study we analyzed side-chain rearrangements upon ligand binding. We constructed two non-redundant databases (980 and 353 entries) of "paired" protein structures in complexed (holo-protein) and uncomplexed (apo-protein) forms from the PDB macromolecular structural database. The number and identity of binding pocket residues that undergo side-chain conformational changes were determined. We show that, in general, only a small number of residues in the pocket undergo such changes (e.g., approximately 85% of cases show changes in three residues or less). The flexibility scale has the following order: Lys > Arg, Gln, Met > Glu, Ile, Leu > Asn, Thr, Val, Tyr, Ser, His, Asp > Cys, Trp, Phe; thus, Lys side chains in binding pockets flex 25 times more often then do the Phe side chains. Normalizing for the number of flexible dihedral bonds in each amino acid attenuates the scale somewhat, however, the clear trend of large, polar amino acids being more flexible in the pocket than aromatic ones remains. We found no correlation between backbone movement of a residue upon ligand binding and the flexibility of its side chain. These results are relevant to 1. Reduction of search space in docking algorithms by inclusion of side-chain flexibility for a limited number of binding pocket residues; and 2. Utilization of the amino acid flexibility scale in protein engineering studies to alter the flexibility of binding pockets.  相似文献

Brassinosteroids are naturally occurring plant growth regulators, which exhibit structural similarities to animal steroid hormones. Recent studies have indicated that besides an essential role in plant growth and development, brassinosteroids also exert anti-stress effects on plants. We show here that tomato plants treated with 24-epibrassinolide (EBR) are more tolerant to high temperature than untreated plants. An analysis of mitochondrial small heat shock proteins (MT-sHSP) in tomato leaves by western blotting revealed that the MT-sHSP did not preferentially accumulate in EBR treated plants at 25 °C. However, treatment of plants at 38 °C induced much more accumulation of MT-sHSP in EBR treated than in untreated plants. Results of this study provide the first direct evidence for EBR induced expression of MT-sHSP, which possibly induced thermotolerance in tomato plants. EBR treated tomato plants had better photosynthetic efficiency. We also observed significantly higher in vitro pollen germination, enhanced pollen tube growth and low pollen bursting in the presence of EBR at 35 °C, a temperature high enough to induce heat-stress symptoms in tomato, indicating a possible role of EBR during plant reproduction.  相似文献

Salt stress is among the major abiotic stresses that adversely affect the global crop production and its adverse impacts are getting more serious in the regions where saline water is used for irrigation. It induces reactive oxygen species, alters the activity of antioxidant system and adversely affects the process of photosynthesis. Various strategies have been employed to mitigate the deleterious effects of salt stress. Presently, the recommended strategies to overcome the adverse effects of salt stress include the use of tolerant cultivars, ameliorative water management and diverse cultural practices. However, none of these approaches have been found to be fully effective under salt stress conditions. An alternative and technically simpler approach to induce salt stress tolerance is the exogenous application of plant growth regulators (PGRs). This technique has gained significant importance during the past decade. PGRs have been implicated to regulate a wide range of metabolic and physiological activities in plants, ranging from cell division and organogenesis to protection against biotic and abiotic stresses. One of the important factors for enhanced plant productivity by PGRs is their efficiency to overcome the salt-induced stress conditions. Recent findings on the effects of brassinosteroids and polyamines on the salt stress tolerance of crops open new avenues to address the salinity problems. This review enlightens the role of brassinosteroids and polyamines in different plant processes like their role in regulation of photosynthesis, antioxidant systems and other related aspects, thereby improving overall performance of plants.  相似文献

Being sessile, plants are subjected to a diverse array of environmental stresses during their life span. Exposure of plants to environmental stresses results in the generation of reactive oxygen species (ROS). These activated oxygen species tend to oxidize various cellular biomolecules like proteins, nucleic acids, and lipids, a process that challenges the core existence of the cell. To prevent the accumulation of these ROS and to sustain their own survival, plants have developed an intricate antioxidative defence system. The antioxidative defence system comprises various enzymatic and nonenzymatic molecules, produced to counter the adverse effect of environmental stresses. A sizable number of these molecules belong to the category of compounds called secondary metabolites. Secondary metabolites are organic compounds that are not directly involved in the growth and development of plants but perform specialized functions under a given set of conditions. Absence of secondary metabolites results in long-term impairment of the plant’s survivability. Such compounds generally include pigments, phenolics, and so on. Plant phenolic compounds such as flavonoids and lignin precursors have been reported to accumulate in response to various biotic and abiotic stresses and are regarded as crucial defence compounds that can scavenge harmful ROS. Another important category of plant metabolites, called brassinosteroids, exhibit stress regulatory and growth-promoting activity and are classified as phytohormones. Elucidation of the physiological and molecular effects of secondary metabolites and brassinosteroids have catapulted them as highly promising and environment-friendly natural substances, suitable for wider application in plant protection and crop yield promotion. The present review focuses on our current understanding of how plants respond to the generation of excessive ROS and the role of secondary metabolites and brassinosteroids in countering the adverse effects of environmental stresses.  相似文献

拟南芥的CPD基因编码一种与植物油菜素内酯(brassinosteroids,BRs)生物合成有关的细胞色素P450类固醇单加氧酶(CYP90 )。为探讨油菜素内酯这类新型植物激素在多年生木本植物中生物合成及作用的分子机理,以拟南芥CPD基因的一个cDNA片段为探针,从一种杂交杨 (Populustremula×tremudelois)的cDNA文库中分离出一个长 1 442bp的cDNA片段,然后再以这个cDNA的 5′区为探针,从这种杂交杨的基因组文库中分离出一个长 1 900bp的基因组DNA(gDNA)片段。测序结果表明,这段cDNA的 5′区与这段gDNA的 3′区重合; 由这段cDNA和gDNA组成的读框编码一个由 476个氨基酸组成的分子量为 63kD的蛋白质。该蛋白与拟南芥CYP90的同源性为 78 32%,比后者仅长 4个氨基酸,在所有已知的功能结构域,其中包括与BR生物合成密切相关的类固醇结合位点,也具有较高的同源性,表明CPD基因在一年生的草本和多年生的木本植物之间具有很高的保守性。系统树分析还表明,CYP90蛋白与番茄和玉米的矮化基因产物、鱼的all trans retinoicacid4 hydroxy lase及微澡青菌(Synechocystissp. )的细胞色素P450在进化上有较密切的联系。  相似文献

Brassinosteroids are plant growth-promoting compounds that exhibit structural similarities to animal steroid hormones. Recent studies have indicated that brassinosteroids are essential for proper plant development. In addition to a role in development, several lines of evidence suggest that brassinosteroids exert anti-stress effects on plants. However, the mechanism by which they modulate plant stress responses is not understood. We show here that Brassica napus and tomato seedlings grown in the presence of 24-epibrassinolide (EBR) are significantly more tolerant to a lethal heat treatment than are control seedlings grown in the absence of the compound. Since a preconditioning treatment of seedlings was not required to observe this effect, we conclude that EBR treatment increases the basic thermotolerance of seedlings. An analysis of heat shock proteins (HSPs) in B. napus seedlings by western blot analysis indicated that the HSPs did not preferentially accumulate in EBR-treated seedlings at the control temperature. However, after heat stress, HSP accumulation was higher in EBR-treated than in untreated seedlings. The results of the present study provide the first direct evidence for EBR-induced expression of HSPs. The higher accumulation of HSPs in EBR-treated seedlings raises the possibility that HSPs contribute, at least in part, to thermotolerance in EBR-treated seedlings. A search for factors other than HSPs, which may directly or indirectly contribute to brassinosteroid-mediated increase in thermotolerance, is underway.  相似文献
