目的:探讨腹腔内注射LiCl引起的内脏不适刺激对大鼠不同味觉溶液摄食的影响。方法:测量给予大鼠腹腔内注射LiCl或生理盐水前后.大鼠5种不同味觉溶液摄食量的改变。结果:腹腔内注射LiCl后,大鼠5种溶液的摄食量均有显著性下降,而对照组则改变不大。结论:内脏不适刺激可以减少大鼠不同味觉溶液的摄食。  相似文献

为探讨家蚕Bombyx mori人工饲料饲养发育不齐的生理原因,从同一蚕品种中挑选出对人工饲料摄食性不同的个体,用电生理学方法测定了下颚瘤状体味觉感器对4种代表性物质(蔗糖、肌醇、大豆粉提取物和柠檬酸)的电生理反应。结果表明,栓锥感器Ss-Ⅰ对蔗糖等促食物质的反应以及栓锥感器Ss-Ⅱ对大豆粉提取物等阻食物质的反应,均存在明显的个体差异。在临界浓度下,低摄食性个体的放电脉冲频率显著高于高摄食性个体。说明低摄食性蚕的味觉反应比高摄食性蚕敏感。  相似文献

本研究以轻度麻醉的大鼠为对象,应用细胞外微电极记录技术,观察并分析了脑桥臂旁核抑制性味觉神经元的自发活动及其对NaCl、HCl、盐酸奎宁(quinine HCl,QHCl))和蔗糖等四种基本味觉刺激的反应。共分析了18个具有自发活动的抑制性味觉神经元,自发放电频率分布在0.2~5.5Hz之间,平均放电频率(2.15±0.31)Hz。18个神经元中,1个神经元对单一味觉刺激呈抑制性反应,其余17个神经元对两种或两种以上的基本味觉刺激发生抑制性反应,且抑制具有潜伏期短、持续时间较长等特征。抑制持续时间5~80S,部分神经元表现为后抑制效应。根据神经元对四种基本味觉刺激呈抑制性反应的程度,将其分为NaCl优势神经元(n=8),HCl优势神经元(n=3),QHCl优势神经元n=3)和蔗糖优势神经元n=4)。其中NaCl优势神经元的反应谐宽最高(0.945)。这些神经元对欣快或厌恶刺激的区别能力较低。结果提示,在脑桥臂旁核存在对味觉刺激起抑制性反应的神经元,这些味觉神经元可能在味觉的调制及对欣快和厌恶刺激的编码中发挥重要的作用。  相似文献

目的:分析慢性吗啡依赖大鼠纳洛酮催瘾戒断条件性位置厌恶(CPA)建立前后、消退后及重建后,与成瘾密切相关的脑区杏仁中央核(CeA)内蛋白激酶A(PKA)蛋白表达的适应性变化,探讨阿片依赖戒断后厌恶动机形成的生物学基础。方法:①将雄性SD大鼠分为实验组(慢性吗啡注射+纳洛酮催瘾组,MN),对照组(慢性吗啡注射+生理盐水催瘾组,MS),慢性生理盐水注射+纳洛酮催瘾组,SN),每组24只。采用慢性吗啡注射(10 mg/kg,BID,ip)后给予一次纳洛酮(0.3 mg/kg)催瘾注射(同时与条件性位置训练箱搭配)建立大鼠CPA模型。②在CPA建立前后、消退后及重建后,采用免疫组织化学方法检测CeA内PKA蛋白表达情况。结果:MN组在CPA建立前后、消退后及重建后CeA内PKA的蛋白表达出现适应性变化(P<0.05),建立后(Day7,134.43±4.481,P<0.05),消退后(Day13,141.01±3.360,P<0.01)及重建后(Day14,137.18±40.330,P<0.05)PKA蛋白表达水平均低于建立前(Day5,124.48±6.722)。而MS组(P>0.05)和SN组(P>0.05)在CPA的各个时间点PKA的蛋白表达变化差异均无显著性。结论:①CeA内PKA蛋白的低表达导致的厌恶的中枢状态,可能是CPA建立的关键的神经机制。②CeA内PKA的适应性变化可能是物质依赖戒断后CPA相关神经可塑性变化的重要分子基础。  相似文献

应用细胞外记录的电生理学方法,在乌拉坦麻醉的大鼠观察了电损毁双侧杏仁中央核前后脑桥臂旁核味觉神经元对四种基本味觉刺激(即氯化钠、盐酸、奎宁和蔗糖)反应的变化。根据对味觉刺激的优势反应,29个记录的味觉神经元中,有14个NaCl优势、9个HCl优势、3个QH2SO4优势和3个蔗糖优势反应神经元。损毁杏仁中央核明显增强臂旁核味觉神经元对盐酸和硫酸奎宁的反应(P<0.01)。氯化钠优势、盐酸优势和奎宁优势反应神经元对盐酸和硫酸奎宁的反应在电损毁杏仁中央核后也明显增强。在破坏杏仁中央核后,臂旁核味觉神经元对氯化钠和硫酸奎宁苦味的分辨能力降低。以上结果提示,杏仁中央核在大鼠脑桥水平的味觉编码中发挥重要作用,它可能是通过参与对味觉的影响来调节机体的摄食行为。  相似文献

为观察Aβ类初级传入纤维是否参与相邻脊髓节段外周末梢之间的信息传递及其相关机制 ,实验自近中端切断一侧T8~T12 脊髓节段背侧皮神经 ,将一支被切断的皮神经的外周端分离成数支细束 ,以单个Aβ纤维放电为指征 ,检测单位的传导速度、适应特性、机械感受阈值、感受野的形状和面积 ;在相邻脊髓节段、也与中枢断离的皮神经干上施加逆向电刺激 ( 0 45mA ,0 1ms,2 0Hz,10s) ,以观察该刺激对Aβ纤维的上述机械感受特性的影响。在 42只大鼠上共记录了 5 0个Aβ类单位。逆向电刺激相邻节段皮神经后 ,60 6% (n =3 3 )的单位感受野增大 ,全部单位的感受野平均面积从 8 94± 6 5 1mm2 显著增加到 2 0 3 4± 16 17mm2 (P <0 0 1)。 81 8% (n =2 0 )的单位感受野形状从点状、圆或与身体长轴垂直的椭圆变成与身体长轴斜行或平行的椭圆。 68 0 % (n =5 0 )的单位机械感受阈值下降 ,全部单位的平均阈值从 2 3 7± 1 2 4mN降至 2 2 9± 1 2 4mN (P <0 0 5 )。上述机械感受特性的改变可持续 5 2 2 3± 9 2 7至 5 6 93± 15 76min。跨节段电刺激后 ,有 5 0 0 % (n =5 0 )的单位同时出现放电的增加 ,但该增加仅持续 1 5 2± 0 46min ,显著短于机械感受特性改变的时程 (P <0 0 1)。有机械感受特性改变的单位也  相似文献

有关内脏痛的问题,已有不少学者进行了研究。但由于内脏痛本身的复杂性,至今其中枢传导途径还不太清楚。本实验刺激大鼠内脏大神经后,利用Fos的免疫组化技术在下列结构中可以发现Fos免疫组化阳性反应的神经元:隔外侧核、斜角带核、丘脑的菱形核、连接核、下丘脑室旁核、中缝背核、兰斑、中缝大核、网状外侧核、脊髓RexedⅠⅡⅢⅩ层等结构。而在对照组的上述结构中没有表达,只在耳蜗背核有较明显的表达。通过上述研究我们发现围绕脊髓中央管和各脑室的周围灰质系统与内脏伤害性刺激有关。这引起了我们极大的兴趣。关于这部分的工作我们正在进行中。  相似文献

探讨电刺激致海马(hippocampus,HPC)癫痫网络的神经信息特征和M型胆碱能受体阻断剂东莨菪碱(scopolamine)对该信息特征的调制作用。实验用雄性SD大鼠45只,体重150 ̄250g。急性强直电(60Hz,2s,0.4 ̄0.6mA)刺激右侧后背HPC(acutetetanizationoftherightposteriordorsalhippocampus,ATPDH),双电极同步记录同侧HPC网络和单个神经元电活动。分析癫痫发作样高频电振荡(ripple)功率谱(powerspec-trum)、尖波连续发放峰间间隔(interpeakinterval,IPI)和单位时间内平均频率(Hz),并同步分析单个神经元放电脉冲间隔(interspikeinterval,ISI)的变化特征。发现:(1)ATPDH诱导的HPC癫痫放电模式主要包括rip-ple和具有稳定频率特征的尖波样连续发放;(2)东莨菪碱(i.p.)可以提前ripple第1组分最大功率(μV2)与单个神经元原发性单位后放电最大ISI出现的时间,对最大ISI的作用更明显;(3)东莨菪碱可以部分再现重复施加ATPDH诱导出现巨大尖波连续发放IPI和神经元放电ISI平行发展特征。结果提示:M胆碱能受体阻断剂东莨菪碱可以同时调制HPC癫痫网络成员电场和细胞的瞬时编码信息;而成员电场ripple功率谱/连续尖波IPI和神经元放电ISI点分布的对比研究,可以用于分析癫痫网络瞬时编码信息和药物生物学效应。  相似文献

目的:探讨激活大鼠ACC脑区的阿片受体降低伤害刺激引起厌恶情绪的作用。方法:将实验大鼠随机分为7组,完全弗氏佐剂(CFA)+生理盐水(NS)组,生理盐水(NS)+生理盐水(NS)组,生理盐水(NS)+μ-阿片受体激动剂([DAla2, NMe-Phe4, Gly-ol5]enkephinlin, DAMGO)组,完全弗氏佐剂(CFA)+ 0.01/0.04/0.2/1 μg/μl DAMGO组(n=6)。实验周期为3 d,第1日测量基础值,第2日预先通过ACC区域给药1 μl,然后将0.08 ml完全弗氏佐剂(CFA)注射到大鼠左后脚掌,第3日观察大鼠的CPA反应、缩足反射潜伏期(PWL)和ACC脑区的电活动。结果:①皮下注射CFA的大鼠,注射前与注射后相比,PWL明显减少(P<0.05);②在笼具痛侧,CFA组大鼠停留的时间明显少于非痛侧(P<0.05);③在ACC脑区预先注射0.04/0.2/1 μg/μl DAMGO可明显减弱C-CPA反应(P<0.05);④在ACC脑区预先注射0.04/0.2/1 μg/μl DAMGO可以降低CFA诱发ACC脑区放电频率的增加(P<0.05)。结论:激活了大鼠ACC脑区上的μ-阿片受体可以降低伤害性刺激诱发的厌恶情绪的发生。  相似文献

To clarify developmental changes in the gustatory system of the rat, integrated taste responses from the chorda tympani (CT) nerve were recorded and analyzed at different postnatal ages. The response magnitude was calculated relative to the response to the standard, 0.1 M NH4Cl. Even at 1 week of age, the CT responded well to all tested 0.1 M chloride salts (NH4Cl, NaCl, LiCl, KCl, RbCl and CsCl). The responses to 0.1 M NaCl and LiCl increased with increasing age of the rat while response magnitudes to KCl, RbCl and CsCl did not change up to 8 weeks. At 1 week, the integrated response pattern was quite similar to that in adult rats for NaCl, HCl and quinine hydrochloride (QHCl). The concentration-response functions for NaCl, HCl, QHCl and sucrose at 2 weeks were essentially the same as those at 8 weeks. These results suggest that taste buds in the 2-week-old rat are functionally mature for the detection of the four basic taste stimuli. The relative magnitude of the responses to the various sugars was smaller at 1 week compared to the adult rat and reached a maximum at weeks 3-4, then decreased gradually with age. Among the six sugars, sucrose was the most effective followed by lactose. From weeks 1-4, the magnitude of the integrated taste response to fructose was smaller than that to lactose except at 3 weeks of age. Maltose, galactose and glucose were less potent stimuli than the other sugars tested. The response magnitude to lactose at 4 weeks had decreased compared to that for the other sugars. Taste responses to the sugars in preweanling and adult rats were not cross-adapted by the individual sugars. These results suggest that after 1 week of age during postnatal development in the rat, taste information from the CT rapidly increases in its importance for feeding behavior.  相似文献

Even though monosodium glutamate (MSG) is a prototypical umami substance, previous studies reported that a conditioned taste aversion (CTA) to MSG, mixed with amiloride to block the taste of sodium, generalizes to sucrose. These findings suggest that the taste of glutamate mimics the taste of sucrose and raise the question of whether glutamate has a broadly tuned sweet taste component. To test this hypothesis, CTA experiments were conducted to test for generalization between MSG and several sweet stimuli: sucrose, glucose, maltose, saccharin and SC-45647. Strong bidirectional generalization was seen between MSG mixed with amiloride and sucrose, glucose, saccharin and SC-45647. Weak generalization was seen between MSG and maltose, and sucrose and maltose. None of the CTAs generalized to NMDA. These findings support the hypothesis that the taste of MSG has broadly tuned, sweet-like characteristics, possibly due to the convergence of afferent signals for MSG, natural sugars and artificial sweeteners.  相似文献

It has been known that umami substances such as monosodium L-glutamate (MSG) and 5'-inosine monophosphate (IMP) elicit a unique taste called 'umami' in humans. One of the characteristics of the umami taste is synergism: the synergistic enhancement of the magnitude of response produced by the addition of 5'-ribonucleotides to MSG. In addition to this well-documented synergism, we report here for the first time on another type of synergism between a glutamate receptor agonist, L-AP4, and sweet substances, by analyzing the chorda tympani responses in rats. The results are as follows: (i) when L-AP4 was mixed with one of the sweet substances, such as sucrose, glucose, fructose and maltose, large synergistic responses were observed. (ii) These synergistic responses, except to L-AP4 + sucrose, were not suppressed by sweet taste suppressants, gurmarin and pronase E. (iii) These synergistic responses were not suppressed by either metabotropic or ionotropic glutamate receptor antagonists. (iv) Fibers that responded well to the binary mixtures of L-AP4 and sweet substances also responded well to NaCl and HCl, but very weakly to sucrose. These findings are different from the characteristics of synergism between glutamate and IMP. The multiple transduction mechanisms for the umami taste in rat taste cells are discussed.  相似文献

Amiloride, a sodium channel blocker, is known to suppress NaCl responses of the chorda tympani (CT) nerve in various mammalian species. In mice, the NaCl suppressing effect of amiloride is reported to differ among strains. In C57BL mice, amiloride inhibits NaCl responses to about 50% of control, whereas no such clear suppression was evident in prior studies with 129 mice. However, evidence from behavioral studies is not entirely consistent with this. Recently, it has been found that genetic backgrounds of 129 mice differ within substrains. 129X1/SvJ (formerly 129/SvJ) mice differ from the 129P3/J (formerly 129/J) strain by 25% of sequence length polymorphisms. Therefore, we examined possible substrain difference between 129P3/J and 129X1/SvJ mice in the amiloride sensitivity of electrophysiologically recorded NaCl responses. Amiloride significantly suppressed CT responses to NaCl without affecting responses to KCl both in 129P3/J and 129X1/SvJ mice. However, the magnitude of the amiloride inhibition was significantly larger (approximately 50% of control in response to 0.01-1.0 M NaCl by 100 microM amiloride) in 129X1/SvJ than in 129P3/J mice (approximately 20% of control in response to 0.03-0.3 M NaCl by 100 microM amiloride). Threshold amiloride concentration for suppression of responses to 0.3 M NaCl was 30 microM in 129P3/J mice, which was higher than that in 129X1/SvJ mice (10 microM). In 129X1/SvJ mice, the threshold amiloride concentration eliciting inhibition of NaCl responses and the magnitude of the inhibition were comparable with those in C57BL/6 mice. These results suggest that amiloride sensitivity of NaCl responses differs even among the 129 substrains, 129P3/J and 129 X1/SvJ, and the substrain difference of 129 mice in amiloride sensitivity is as large as that between two inbred strains (129P3/J and C57BL/6).  相似文献

The topographical relationship of the chorda tympani nerve (chorda tympani) to the tensor tympani muscle in the middle ear of carnivores provides new phylogenetic information. The examination of histological serial sections of 16 carnivore species representing most families revealed two distinct character states concerning the course of the chorda tympani: a hypotensoric state with the nerve running below the insertion tendon of the tensor tympani muscle, and an epitensoric state with the nerve running above the tendon. The shift from the plesiomorphic hypotensoric chorda tympani to the apomorphic epitensoric condition occurred once in carnivore phylogeny: Only in the herpestid species under study does the chorda tympani cross above the tensor tympani muscle. Therefore, we introduce the epitensoric pattern as a new synapomorphy for herpestids. Within the herpestids we find the following structural distinctions: Herpestes javanicus and Galerella sanguinea have a chorda tympani running in a sulcus directly above the insertion of the tensor tympani muscle, whereas in the eusocial herpestid species Suricata suricatta and Mungos mungo the chorda tympani lies far above the insertion of the muscle. The epithelial sodium channel (ENaC) blocker amiloride has been shown to increase the behaviorally measured NaCl detection threshold in mice. In this study, a conditioned taste aversion (CTA) paradigm was used to examine whether 100 microM amiloride has a perceptible taste that could contribute to this observed decrease in behavioral responsiveness. Eighty-four C57BL/6J (B6) and 64 DBA/2J (D2) mice were divided into eight groups (n=8-12 per group), in which half received an injection of 0.15 M LiCl (2 mEq/kg) and the other half an equivalent saline injection, in three conditioning trials. The four conditioned stimuli were 100 microM amiloride hydrochloride, water, 0.1 and 0.3 M NaCl. Neither strain demonstrated acquisition of a CTA to amiloride in a brief-access (BA) taste test (5 s trials in the gustometer). Although 0.3 M NaCl is inherently aversive, its pairing with LiCl led to significantly further decreases in licking during the BA test on salt trials in both strains. The D2 strain clearly avoided 0.1 M NaCl, whereas avoidance of this stimulus was more equivocal in B6 mice. The inefficacy of amiloride to serve as a conditioned stimulus in taste aversion learning involving three LiCl pairings suggests that the effects of this ENaC blocker on taste-related behavioral responses to NaCl are likely due to its pharmacological interference with sodium taste transduction.  相似文献

Adolescent individuals display altered behavioral sensitivity to ethanol, which may contribute to the increased ethanol consumption seen in this age‐group. However, genetics also exert considerable influence on both ethanol intake and sensitivity. Currently there is little research assessing the combined influence of developmental and genetic alcohol sensitivities. Sensitivity to the aversive effects of ethanol using a conditioned taste aversion (CTA) procedure was measured during both adolescence (P30) and adulthood (P75) in eight inbred mouse strains (C57BL/6J, DBA/2J, 129S1/SvImJ, A/J, BALB/cByJ, BTBR T+tf/J, C3H/HeJ and FVB/NJ). Adolescent and adult mice were water deprived, and subsequently provided with access to 0.9% (v/v) NaCl solution for 1 h. Immediately following access mice were administered ethanol (0, 1.5, 2.25 and 3 g/kg, ip). This procedure was repeated in 72 h intervals for a total of five CTA trials. Sensitivity to the aversive effects of ethanol was highly dependent upon both strain and age. Within an inbred strain, adolescent animals were consistently less sensitive to the aversive effects of ethanol than their adult counterparts. However, the dose of ethanol required to produce an aversion response differed as a function of both age and strain.  相似文献

Voluntary running in an activity wheel endowed rats with aversion to a taste solution consumed before the running. This running-based taste aversion was attenuated by extra running opportunities interspersed among the taste–running pairings, but the attenuating effect was reduced by signaling the extra running by another taste cue. These results correspond to the so-called degraded contingency effect and cover-cue effect in the traditional preparations of Pavlovian conditioning.  相似文献

Responses of three groups of neural fibers from the chorda tympani of the hamster to binary mixtures of taste stimuli applied to the tongue were analyzed. The groups displayed different sensitivities to six chemicals at concentrations that had approximately equal effects on the whole nerve. Sucrose-best fibers responded strongly only to sucrose and D-phenylalanine. NaCl-best and HCl-best fibers, responded to four electrolytes: equally to CaCl2 and nearly equally to HCl, but the former responded more to NaCl, and the latter responded more to NH4Cl. The groups of fibers dealt differently with binary mixtures. Sucrose- best fibers responded to a mixture of sucrose and D-phenylalanine as if one of the chemicals had been appropriately increased in concentration, but they responded to a mixture of either one and an electrolyte as if the concentration of sucrose or D-phenylalanine had been reduced. NaCl- best fibers responded to a mixture as if it were a "mixture" of two appropriate concentrations of one chemical, or somewhat less. But, responses of HCl-best fibers to mixtures were greater than that, approaching a sum of responses to components. These results explain effects on the whole nerve, suggest that the sensitivity of a mammalian taste receptor to one chemical can be affected by a second, which may or may not be a stimulus for that receptor, and suggest that some effects of taste mixtures in humans may be the result of peripheral processes.  相似文献

Conditioned taste aversion (CTA) to saccharin induced by orallithium chloride and normal saline were investigated in 270Wistar rats in a CTA paradigm. They were trained to drink for20-min sessions per day in a two-bottle choice, providing saccharinand water simultaneously. Saccharinn neophobia was replacedby a preference with two more exposures. Saccharin was withheldfor 2 days following force-feeding of lithium or saline in orderto assess lithium-illness. Lithium-fed rats showed signs ofillness followed by a strong CTA to saccharin persisting for3–4 days. Saline-fed rats did not suffer from any illnessbut showed a weak and inconsistent CTA lasting for a day ortwo. Scopolamine injections prior to force-feeding did not affectthe pattern of aversion in either group, but significantly suppressedthe revocation of CTA in the saline-fed rats by a second oraldose of saline.  相似文献
