目的探讨口炎清颗粒对肠道菌群失衡的调节作用。方法小鼠随机分为6组:正常对照组、重度失调组、自然恢复组、口炎清低剂量组、口炎清高剂量组和大豆低聚糖对照组。正常对照组小鼠灌服蒸馏水,其他组以头孢曲松钠终浓度为8 g/(kg.d)的剂量,连续灌胃8 d,建立小鼠菌群失衡模型,自然恢复组在失衡模型建立后不使用任何药物,自然恢复7 d;口炎清低剂量组、高剂量组、大豆低聚糖对照组分别灌服相应剂量的药物7 d,观察各组小鼠盲肠指数和肠道内双歧杆菌、类杆菌、韦荣球菌、乳酸杆菌、消化球菌、优杆菌、葡萄球菌、肠球菌、大肠埃希菌、链球菌和酵母菌11种菌群变化。结果与菌群失衡组比较,口炎清组与大豆低聚糖组盲肠指数均显著下降(P〈0.05),呈恢复趋势。自然恢复组肠道菌群种类和数量变化不明显,口炎清组厌氧菌重新出现,尤其是低剂量组双歧杆菌、优杆菌和类杆菌的出现预示着肠道菌群正在逐渐恢复正常。结论一定浓度的口炎清可以明显促进菌群失衡小鼠盲肠大小的恢复,对小鼠肠道菌群有调节作用,具有类似大豆低聚糖的益生元功效。  相似文献

目的 :对山松牌大豆低聚糖对肠道菌群调节的效果进行研究。方法 :通过动物实验和人体试验。结果 :山松牌大豆低聚糖能增殖体内乳杆菌和双歧杆菌数量 ;在较高剂量时 ,能使体内的肠球菌和肠杆菌增殖。采用B/E值 (即双歧杆菌和肠杆菌的比值 )作为指标进行分析发现 ,该制品在低剂量时能更好地优化肠道菌群结构。结论 :大豆低聚糖对肠道菌有较好的调节效果  相似文献

本试验比较研究了不同蛋白源(鱼粉,FM;大豆分离蛋白,SPI)基础饲料中添加大豆低聚糖(SBOS)对牙鲆(Paralichthys olivaceus)消化道(胃、幽门盲囊、前肠、中肠和后肠)和肝脏消化酶活性、饲料表观消化率和消化道(胃、小肠)组织结构的影响。分别以FM、SPI作为主要蛋白源,配制了4种等氮等能饲料。其中,饲料FM、SPI分别以FM、SPI作为主要蛋白源;饲料FMO、SPIO分别在饲料FM、SPI基础上添加10%SBOS(水苏糖:2.61%;棉籽糖:0.61%)。试验表明:①饲料中添加SBOS普遍降低了牙鲆消化道和肝脏蛋白酶活性,但差异均不显著(p > 0.05)。FM基础饲料中添加SBOS显著降低了肝脏脂肪酶活性(p < 0.05),而对消化道脂肪酶活性无显著影响;SPI基础饲料中添加SBOS显著提高了前肠脂肪酶活性,降低了胃和中肠脂肪酶的活性,而对幽门盲囊、后肠和肝脏脂肪酶活性均无显著影响。FM基础饲料中添加SBOS显著降低了中肠淀粉酶活性,提高了胃淀粉酶活性(p < 0.05),而对幽门盲囊、前肠、后肠和肝脏淀粉酶活性无显著影响;SPI基础饲料中添加SBOS显著提高了胃和前肠淀粉酶活性,降低了中肠脂肪酶活性(p < 0.05),而对幽门盲囊、后肠和肝脏淀粉酶活性无显著影响;②FM基础饲料中添加SBOS显著降低了饲料干物质表观消化率(p < 0.05),而对粗蛋白、粗脂肪和能量表观消化率均无显著影响(p > 0.05)。SPI基础饲料中添加SBOS对干物质、粗蛋白、粗脂肪和能量表观消化率均无显著影响;③饲料中添加SBOS对牙鲆胃和小肠组织结构均无明显负面影响。试验结果表明,饲料中添加SBOS对牙鲆消化道和肝脏消化酶活性、饲料表观消化率和消化道组织结构均没有表现出明显的负面效应;大豆蛋白源中含有的SBOS不是影响牙鲆对其利用率差的主要因素。  相似文献

目的研究大豆异黄酮(SI)对小鼠淋巴细胞的辐射防护作用。方法24只雄性昆明小鼠,随机分为正常对照组、辐射对照组和辐射补充0.5%SI组,喂养2周后,4.0Gy照射。照射后24 h处死小鼠,取血、胸腺和脾脏分离淋巴细胞,进行血淋巴细胞计数、观察DNA损伤情况;培养胸腺和脾脏淋巴细胞,检测3H-dT掺入量,观察淋巴细胞的增殖能力,计算淋巴细胞的增殖指数。结果辐射使小鼠胸腺和脾脏淋巴细胞数明显减少、胸腺淋巴细胞增殖能力和脾脏淋巴细胞转化指数降低、血淋巴细胞DNA损伤增加,这些变化均具有统计学意义;补充SI可降低胸腺和脾脏淋巴细胞数的减少幅度,降低胸腺淋巴细胞增殖能力和脾脏淋巴细胞转化指数下降幅度,减少辐射对血淋巴细胞DNA损伤程度,其中SI对胸腺淋巴细胞增殖能力和对血淋巴细胞DNA损伤程度的防护作用与辐射对照组相比有统计学意义。结论大豆异黄酮可对小鼠的血、胸腺和脾脏淋巴细胞有一定的辐射防护作用。  相似文献

We examined the dietary effects of cyclic nigerosylnigerose (CNN), a dietary indigestible oligosaccharide with four D-glucopyranosyl residues linked by alternating alpha-(1-->3)- and alpha-(1-->6) glucosidic linkages, on the intestinal immune function of mice, and the effects were compared with those of alpha-(1-->3)-linked oligosaccharide (nigerooligosaccharides, NOS) or alpha-(1-->6)-linked oligosaccharide (isomaltooligosaccharides, IMO). BALB/c mice were fed with 1-5% CNN, 5% IMO, or 12.5% NOS for 4 weeks, and the intestinal mucosal immune responses were determined. In the 1-5% CNN fed groups, the amounts of IgA in feces increased significantly. In addition, IgA, transforming growth factor-beta1 (TGF-beta1), and interleukin-6 (IL-6) secretion by Peyer's patch (PP) cells were enhanced in CNN fed mice. In the 5% CNN group, pH in the cecum decreased, and the amounts of lactic acid and butyric acid increased. These findings were not observed in the NOS- or IMO-fed group of mice. They suggest that CNN supplementation changes the intestinal environment of microflora and indirectly enhances the immune function in the gut.  相似文献

通过腹腔注射5-FU建立小鼠肠黏膜炎模型,探讨富硒长双歧杆菌(Selenium-enriched Bifidobacterium longum,Se-B.longum)能否改善5-FU所致的小鼠肠黏膜炎。将健康BALB/c小鼠随机分为对照组、5-FU组和Se-B.longum/5-FU组,分别灌胃生理盐水、生理盐水和Se-B.longum(Se0.3 mg/kg BW,1×106bacteria/只)6 d,然后5-FU组和Se-B.longum/5-FU组小鼠均腹腔注射5-FU(250 mg/kg),观察小鼠腹泻及死亡情况,5 d后处死小鼠,计算体重变化、脏器指数及考察肠道组织变化;将健康BALB/c小鼠随机分为对照组、5-FU组和Se-B.longum/5-FU组,分别灌胃生理盐水、生理盐水和Se-B.longum(Se 0.3 mg/kg BW,1×106bacteria/只)6 d,然后5-FU组和Se-B.longum/5-FU组小鼠均腹腔注射5-FU(300 mg/kg),观察小鼠死亡情况,绘制生存曲线。Se-B.longum能缓解5-FU导致的正常小鼠的肠粘膜炎、降低小鼠死亡率。  相似文献

目的:探讨大肠杆菌表达的人源可溶性B淋巴细胞激活因子hsBAFF对小鼠脾脏B淋巴细胞免疫反应及其对胞内游离Ca^2+信号变化的影响。方法:选择20只健康ICR小鼠,雌雄各半,随机分成两组(n=10):①对照组;②hsBAFF实验组。实验组小鼠腹腔注射含hsBAFF(0.1mg/kgbw)的PBS溶液,对照组注射同等剂量的PBS溶液.连续8d。用MTT法检测小鼠脾脏B淋巴细胞的增殖及其对LPS刺激的免疫反应,并用激光共聚焦显微镜分析脾脏B淋巴细胞胞内钙离子水平([Ca^2+]i)变化。结果:hsBAFF注射小鼠的脾脏B淋巴细胞增殖和对LPS刺激的免疫反应均明显高于对照组(P〈0.05);hsBAFF注射组[Ca^2+]i荧光强度维持在相对稳定的高水平上波动,平均荧光强度显著高于对照组(P〈0.01),且荧光强度变化率小于对照组。结论:大肠杆菌表达的hsBAFF能促进B淋巴细胞的增殖和免疫应答,从而增强机体免疫功能。hsBAFF激活小鼠脾脏B淋巴细胞可能与[Ca^2+]i升高有关。  相似文献

The objective of this study was to investigate the effect of purified soybean agglutinin on growth and immune function in rats. Thirty male Sprague-Dawley rats (77.8 +/- 2.6 g) were individually fed casein-cornstarch based diets containing 0, 0.05, 0.10, 0.15 or 0.20% soybean agglutinin (w/w) during a 20-day experiment. Growth declined linearly with increasing the concentration of soybean agglutinin (p < 0.05). The proliferation of lymphocytes in spleen, lymph nodes and blood decreased with an increase in dietary soybean agglutinin (p < 0.05). The concentrations of interleukin-2, interferon-gamma and tumor necrosis factor-alpha in plasma, spleen, and mesenteric lymph nodes as well as plasma concentrations of IgA, IgG and IgM also declined with increasing dose of soybean agglutinin (p < 0.05). The results show that dietary soybean agglutinin has negative effects on growth as well as both cell-mediated and humoral immune function of rats and appears to function in a dose-dependent manner.  相似文献

The present study was designed to evaluate the effects of Prunella Vulgaris (P. vulgaris) on the immune function in mice. The mice were randomly divided into one control group and three treatment groups of 10 mice each. The control group received pure water and the treatment groups received P. vulgaris extract at concentrations of 0.15, 0.30 and 0.90 g/kg BW orally for 30 days, respectively. Changes in cell immune function, non-specific immunity and humoral immunity function were evaluated. Active lymphocytes and T lymphocyte subsets were determined by fluorescence-activated cell sorting (FACS). Certain Serum concentrations of cytokines were measured by enzyme-linked immunosorbent assay (ELISA). The results showed that, for cell immune function, compared with the control group, foot pad thickness in high dose group increased significantly (p<0.01), whereas no significant difference in the proliferative ability of splenic lymphocytes was observed among all groups (p>0.05). For non-specific immunity, NK cell activity increased significantly in a dose-dependent manner in P. vulgaris treated mice (p<0.01), mononuclear-macrophage function in medium and high dose P. vulgaris treated mice were significantly higher than that of the control group (p<0.05). For humoral immunity, no significant differences were observed in terms of the half value of hemolysis (HC50), number of hemolytic plaques and serum IgG level (p>0.05). The percentage of active T and Th lymphocytes of mice peripheral blood in high dose group were significantly higher than that of the control group (p<0.01). There was no significant difference in serum levels of IL-1β, IL-4, IL-10 and IFN-γ among all of the four groups (p>0.05). The data indicated that 0.90 g/kg BW P. vulgaris extract (equivalent to 7.5 g/kg BW crude drug) had some effect on cellular immune function and non-specific immune function in mice.  相似文献

【目的】通过小鼠的急性毒性、30 d喂养及细菌移位试验,对马奶及其制品(酸马奶)中分离出来的7株肠球菌属乳酸菌进行初步安全性评价。【方法】分别对小鼠连续7 d及30 d灌胃不同剂量的肠球菌JHZ9、JHZ15、JHZ17、JHZ22、JHZ25、JHZ28、JNN1,对其一般体征进行观察,记录体重和食物摄入量,最后进行大体解剖,观察肝、肾、脾、心、肺并称重,同时进行细菌移位试验。【结果】除30 d喂养中,试验菌株JHZ17的高剂量组与生理盐水组差异显著(P0.05),7 d急性毒性试验及30 d喂养试验中各试验组小鼠体重、食物摄入量与对照组相比无显著性差异(P0.05),30 d喂养后小鼠各脏器(心、肝、脾、肺、肾)指标与对照组无显著性差异(P0.05),小鼠的经口急性毒性最大耐受剂量(MTD)大于1010 CFU/(mL bw)20 mL/(kg·d),属无毒级别,未发现细菌移位现象。【结论】肠球菌JHZ9、JHZ15、JHZ17、JHZ22、JHZ25、JHZ28、JNN1安全,无毒副作用。  相似文献

Abstract

The objective of this study was to investigate the effect of purified soybean agglutinin on growth and immune function in rats. Thirty male Sprague-Dawley rats (77.8 ± 2.6 g) were individually fed casein-cornstarch based diets containing 0, 0.05, 0.10, 0.15 or 0.20% soybean agglutinin (w/w) during a 20-day experiment. Growth declined linearly with increasing the concentration of soybean agglutinin (p < 0.05). The proliferation of lymphocytes in spleen, lymph nodes and blood decreased with an increase in dietary soybean agglutinin (p < 0.05). The concentrations of interleukin-2, interferon-γ and tumor necrosis factor- in plasma, spleen, and mesenteric lymph nodes as well as plasma concentrations of IgA, IgG and IgM also declined with increasing dose of soybean agglutinin (p < 0.05). 目的 探讨口炎清组分玄参对于小鼠肠道菌群的调节作用.方法 应用抗生素头孢曲松钠灌胃的方法建立小鼠肠道菌群失衡模型.利用观察称量等方法观察小鼠临床表征的变化,以及盲肠指数和脾指数的变化.利用活菌计数法分析肠道优势菌群的变化趋势.结果 (1)菌群失衡组盲肠指数增高(P<0.05);自然恢复组盲肠指数变化不明显;玄参流膏与大豆低聚糖回灌组盲肠指数均显著下降(P<0.05),呈恢复趋势.(2)菌群失衡组脾脏指数有下降趋势,自然恢复组脾脏指数无明显变化;玄参流膏组与大豆低聚糖回灌组脾脏指数均有所上升,呈恢复趋势.(3)菌群失衡组厌氧菌被抑制,需氧球菌大量繁殖.自然恢复组菌群种类和数量变化不明显,玄参组厌氧菌重新出现,尤其是双歧杆菌、乳杆菌、韦荣球菌和类杆菌的出现预示着菌群正在逐渐恢复正常.结论 玄参流膏可以促进菌群失衡小鼠盲肠和脾脏大小的恢复,可以调节小鼠肠道菌群失衡,具有类似大豆低聚糖的益生元功效.  相似文献

目的探讨异麦芽低聚糖对D-半乳糖致衰老大鼠肠黏膜功能的影响。方法Wistar大鼠随机分为3组:(1)青年对照组,(2)衰老对照组,(3)衰老观察组(衰老+异麦芽低聚糖)。采用D-半乳糖造成衰老模型后,应用异麦芽低聚糖灌胃,检测各组肠道菌群、血清IgG和肠黏膜sIgA。结果D-半乳糖致衰老大鼠肠道菌群失调;灌胃异麦芽低聚糖后,衰老大鼠肠道双歧杆菌增加(P〈0.05),肠杆菌和肠球菌数量减少(P〈0.05);血清IgG、肠黏膜sIgA含量增加(P〈0.05)。结论异麦芽低聚糖可改善衰老机体肠黏膜功能。  相似文献

通过研究广叶绣球菌多糖对免疫低下小鼠肠道菌群、细胞因子表达量及短链脂肪酸的影响,探究广叶绣球菌多糖的免疫作用机制。腹腔注射环磷酰胺构建免疫低下小鼠模型,将小鼠分为6组:正常对照组、模型组、广叶绣球菌多糖低、中、高剂量组以及阳性对照组,连续饲养30d后处死取样,HE染色观察小肠组织结构,酶联免疫吸附法测定小肠白细胞介素-6(IL-6)、IL-10、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)的表达水平,高通量测序技术分析小鼠肠道菌群的变化,气-质联用(GC-MS)技术分析盲肠内容物短链脂肪酸(SCFAs)的含量。结果表明,广叶绣球菌多糖高剂量组可显著改善绒毛肿胀和变短现象,提高绒毛长度/隐窝深度的比值(V/C值)和小肠IL-6、IL-10、TNF-α、IFN-γ细胞因子含量(P<0.05或P<0.01),提高拟杆菌属Bacteroides、拟普雷沃菌属Alloprevotella、丁酸弧菌属ButyrivibrioIntestinimonas、链球菌属Streptococcus的相对丰度(P<0.05或P<0.01);各剂量组均可提高盲肠内6种主要短链脂肪酸含量,差异达显著或极显著水平(P<0.05或P<0.01)。试验组与阳性对照组趋势一致。广叶绣球菌多糖可通过改善免疫低下小鼠的肠道粘膜形态,提高小肠细胞因子水平,调节肠道菌群结构,增加SCFAs产生菌的相对丰度,提高短链脂肪酸含量,调节免疫低下小鼠的肠道免疫功能。  相似文献

Numerous studies have demonstrated that dietary sources of nucleosides and nucleotides are important for the maintenance of cellular and humoral immune responses. To determine the immunological effects of feeding a nucleoside-nucleotide mixture to dexamethasone-immunosuppressed C57BL/6 adult mice infected with Cryptosporidium parvum, we examined fecal oocyst shedding, lymphoproliferative responses to concanavalin (Con) A, and C. parvum antigen, interleukin (IL-2), and gamma-interferon (IFN-gamma) production by cultured spleen cells. Mice were fed a nucleotide-free 20% casein diet (control group) or this diet supplemented with a 0. 5% nucleoside-nucleotide mixture before and after inoculation with C. parvum. Spleens from mice receiving the supplemented diet had higher (P < 0.05) Con A and antigen-specific induced cell proliferation than those from control mice. In addition to the increased cell proliferation, the spleen cells from the supplemented mice produced significantly more IL-2 (P < 0.002) and significantly more IFN-gamma (P <; 0.004) than cells from the control mice. Mice fed the supplemented diet excreted fewer (P < 0.05) C. parvum oocysts in the feces than control mice. The cumulative survival rate in the nucleoside-nucleotide mixture-fed group was higher compared with the control group (P < 0.05). We conclude that nucleosides and nucleotides may partially counteract the immunosuppressive effects of dexamethasone in C. parvum-challenged mice.  相似文献

探究桑黄饮片水提物对环磷酰胺所致免疫低下小鼠的免疫功能的影响。本研究以桑黄饮片为原料,采用超声和沸水提取方法得到桑黄饮片水提物,注射环磷酰胺造成小鼠免疫功能低下,通过测定脾脏和胸腺指数、血清免疫球蛋白和细胞因子、脾淋巴细胞增殖、碳粒廓清等实验,对免疫低下小鼠的免疫功能进行研究评价。结果表明,灌胃桑黄饮片水提物的小鼠与模型组相比,免疫器官指数、血清中免疫球蛋白含量、细胞因子水平、脾淋巴细胞增殖程度、巨噬细胞的吞噬速率和吞噬指数、小鼠耳肿胀度均有提高,脾脏苏木精-伊红(HE)染色也表明桑黄饮片水提物可以改善环磷酰胺导致的脾脏组织病理状态。本研究表明桑黄饮片水提物对环磷酰胺所致的免疫抑制小鼠具有良好的免疫保护作用,为桑黄饮片扩大临床应用提供实验依据。  相似文献

根据保健食品功能评价规范?功能学评价程序研究一种双歧杆菌胞外多糖(Bifidobacterium spp. exopolysaccharide, EPS)的免疫调节功能。通过脾淋巴细胞增殖反应、绵羊红细胞诱导小鼠迟发型变态反应(DTH)、血清溶血素测定以及巨噬细胞吞噬实验探讨该EPS的免疫调节活性。EPS分别以高、中、低剂量组连续口服给药10天, 结果发现低剂量的EPS[100 mg/(kg·d)]可以促进脾淋巴细胞增殖; 高、中、低剂量对绵羊红细胞诱导的小鼠迟发型变态反应均无促进作用; 但是, EPS 高、中、低剂量组均能明显提高小鼠血清半数溶血值HC50以及增强小鼠腹腔巨噬细胞吞噬鸡红细胞的吞噬能力。根据规范可知, 该双歧杆菌EPS 具有一定的免疫调节活性。  相似文献

目的:观察大豆低聚糖和所含糖对肠道菌群中双歧杆菌、肠杆菌体外促生长作用。方法:按1%大豆低聚糖、0.36%水苏糖、0.52%蔗糖、0.11%棉子糖含量配制培养液,分别接种各试验菌株,在0、12、24小时测定各培养液吸光度A值(分光光度计.550nm)和pH值。结果:经24小时培养,加大豆低聚糖的双歧杆菌标准株和临床分离株培养液A值分别为>1.5和1.307,大于肠杆菌标准株和临床分离株的1.1和1.173,而其pH值小于肠杆菌;加水苏糖的双歧杆菌培养液A值为1.47,大于蔗糖的1.4和棉子糖的0.53。结论:大豆低聚糖促双歧杆菌生长作用大于促肠杆菌生长,水苏糖是促双歧杆菌增殖生长的主要成分。  相似文献
