American foulbrood (AFB), a severe bacterial disease of honeybee brood, has recently been found in Uruguayan apiaries. Detection of the causative agent, Paenibacillus larvae subspecies larvae, is a very important concern in order to prevent disease dissemination and decrease of honey production. Since spores are the infective forms of this pathogen, in the present work we report the use of polymerase chain reaction (PCR) to detect P. l. subsp. larvae spores from in vitro cultures, larvae with clinical symptoms and experimentally contaminated honey. The set of primers was designed based on the published P. l. subsp. larvae 16S rRNA gene. Using this approach we could amplify the pathogen DNA and obtain a great sensitivity and a notable specificity. Detection limit for spore suspension was a 10–2 dilution of template DNA obtained from 32 spores, as determined by plate count. For artificially contaminated honey, we could detect the PCR product at a 10–3 dilution of template DNA obtained from 170 spores. In addition, when PCR conditions were set to improve specificity, we were able to amplify P. l. subsp. larvae DNA selectively and no cross-reactions were observed with a variety of related bacterial species, including P. l. subsp. pulvifaciens. Since spore detection is very important to confirm the presence of the disease, this method provides a reliable diagnosis of AFB from infected larvae and contaminated honey in a few hours.  相似文献

Biological and immunological factors may influence changes in sex ratios at different points of the schistosome life cycle, resulting in the fact that female schistosomes are significantly outnumbered by males in chronic infections of snails and mammalian hosts. Analysis of this phenomenon has long been hampered by shortcomings in the methods used to determine sex ratios. Here, Robin Gasser describes recently developed molecular methods for sexing schistosome larvae. These have opened the way towards understanding why sex ratios become male biased and allow a proper assessment of its consequences in the epidemiology, diagnosis and pathology of infection.  相似文献

Chemical composition of newborn larvae, muscle larvae and adult Trichinella spiralis. International Journal for Parasitology16: 455–460. The chemical composition of newborn larvae (NBL), muscle larvae (ML) and 4-day-old and 7-day-old adult Trichinella spiralis were compared. Total protein constituted 44.1% of the dry wt of ML, 36.9% of NBL and 30.3% of 7-day-old adult worms. Glycogen content was 16.1% of worm dry wt in ML and 7.8% in NBL and 7.2% of worm dry wt in adults. Trehalose content was 5.2% in NBL, 8.3% in ML and 8.7% in 7-day-old adult worms. Significantly lower levels of trehalose and glycogen were found in 4-day-old worms than were present in 7-day-old worms. Free glucose amounted to 0.1% of dry wt in ML, 0.37% in NBL and 0.87% in 7-day-old adults. RNA accounted for 3.5% of the dry wt of ML, 3.2% of the dry wt of 7-day-old adults and 1.1% of the dry wt of NBL. The DNA content of NBL was 0.23% of worm dry wt, in ML 0.48% of worm dry wt and 0.51% of worm dry wt in 7-day-old adults. The three parasite stages examined agreed closely with regard to types of amino acids in the free pool. Exceptions were as follows: NBL lacked taurine which both adults and ML contained; adults lacked methionine which both ML and NBL contained; and, trace amounts of cysteine were present in ML but absent from the other two stages.  相似文献

AIMS: The aim of the study is to examine the disinfection of wood contaminated with Paenibacillus larvae subsp. larvae spores, in order to find a practical decontamination method for hive materials. METHODS AND RESULTS: The number of viable spores recovered after the treatment, on the surface by swabbing, and in the deeper parts of the wood by scraping, was used to test the efficiency of the disinfection. Our results indicate that chemical disinfection is only complete when high concentrations (> 50%) of the disinfectant are used. Heat treatment in general was found to be very effective. The scorching of wood was not satisfactory as it only killed spores at the surface. CONCLUSION: Complete disinfection is only possible with some heat treatments or by using high concentrations of chemical disinfectants. SIGNIFICANCE AND IMPACT OF THE STUDY: This study puts forward some methods that can provide complete decontamination, which is necessary for an effective control of American foulbrood disease.  相似文献

Pollen, water, or nothing was added to the food of worker honey-bee, Apis mellifera, larvae aged 6–18 hr. Six hr later larvae from each of these three groups received, in their food, either Bacillus larvae spores in water, or water only. Pooled data from seven replicates of this procedure show that spore feeding preceded by pollen feeding resulted in a 71.17% mortality; spore feeding preceded by water feeding resulted in 94.84% mortality; and spore feeding preceded by nonexperimental feeding resulted in 92.75% mortality. Mortality in groups not receiving spores was below 11.00% in all cases. Chi-square analyses showed a highly significant reduction in mortality associated with pollen feeding.  相似文献

The larval drift behaviour of 23 species representing Ephemeroptera, Plecoptera and Trichoptera was investigated in the laboratory using different current regimes. Mayfly nymphs often performed swimming, while caddis larvae were reluctant to do so. Stonefly nymphs were intermediate. In mayflies swimming seemed to be used to reach the substrate as soon as possible. In contrast most stonefly nymphs by swimming prolonged the time spent in the water column. Modes of swimming and sinking posture differed markedly between the orders. Living passively sinking animals often reached bottom faster than dead control specimens, so consequently behaviour did not always express itself in activity. Some caddis larvae spun adherent anchor lines. Differences among taxa seemed more important in explaining swimming activity compared to preferred habitats (as stream, river and lake) in each species. However, observed differences among closely related species indicated subtle differences related to microhabitat to be of profound importance in explaining the alternative behavioural strategies used.  相似文献

Many insects, including Drosophila melanogaster, have a rich repertoire of olfactory behavior. Combination of robust behavioral assays, physiological and molecular tools render D. melanogaster as highly suitable system for olfactory studies. The small number of neurons in the olfactory system of fruit flies, especially the number of sensory neurons in the larval stage, makes the exploration of sensory coding at all stages of its nervous system a potentially tractable goal, which is not possible in the foreseeable future in any mammalian preparation. Advances in physiological recordings, olfactory signaling and detailed analysis of behavior, can place larvae in a position to ask previously unanswerable questions.  相似文献

The larval antennal sensilla of two Zygoptera species, Calopteryx haemorroidalis (Calopterygidae) and Ischnura elegans (Coenagrionidae) are investigated with SEM and TEM. These two species have different antennae (geniculate, setaceous) and live in different environments (lotic, lentic waters). Notwithstanding this, similarities in the kind and distribution of sensilla are outlined: in both species the majority of sensilla types is located on the apical portion of the antenna, namely a composed coeloconic sensillum (possible chemoreceptor), two other coeloconic sensilla (possible thermo-hygroreceptors) and an apical seta (direct contact mechanoreceptor). Other mechanoreceptors, such as filiform hairs sensitive to movements of the surrounding medium or bristles positioned to sense the movements of the flagellar segments, are present on the antenna. Similarities in the antennal sensilla types and distribution are observed also with other dragonfly species, such as Onychogomphus forcipatus and Libellula depressa. A peculiar structure with an internal organization similar to that of a gland is observed in the apical antenna of C. haemorroidalis and I. elegans and it is present also in O. forcipatus and L. depressa. The possible function of this structure is at the moment unknown but deserves further investigations owing to its widespread presence in Odonata larvae.  相似文献

Rates of feeding of larvae of Culex pipiens in distilled water and larval water (water in which larvae were held overnight at a density of 1–2 larvae/ml) were compared by two methods. One method depended on assessing the relative rate of displacement of kaolin from the gut when kaolin-glutted larvae were provided with charcoal powder in the test liquids. The other method involved observation of changes in the time spent filtering by individual larvae transferred between distilled and larval water. Compared with plain water, larval water stimulates increased rates of feeding by the same or other conspecific larvae.
Résumé L'eau dans laquelle on a maintenu des larves pendant une nuit, à la densité de 1–2 larves ml, stimule l'activité de filtration et la prise de nourriture de larves introduites isolément dans ce milieu, par comparaison avec des larves témoins placées dans de l'eau distillée. La comparaison de l'activité alimentaire a été faite selon deux méthodes. La première méthode utilise des larves gavées de poudre de kaolin ajoutée à l'eau du milieu d'élevage; ces larves sont transférées ensuite dans de l'eau distillée ou de l'eau ayant hébergé de nombreuses larves (eau de larves) eau à laquelle on ajoute de la poudre de charbon. La quantité de charbon filtrée et absorbée peut alors être appréciée et comparee pour l'eau distillée et pour l'eau de larves. La seconde méthode est basée sur la mesure du temps passé en activité de filtration, pour des larves isolées introduites dans de l'eau distillée ou de l'eau, ayant hébergé, de larves.Ces expériences démontrent que l'eau d'êlevage des larves groupées renferme un principe phagostimulant apporté par les larves elles-mêmes.

Silkworms (Bombyx mori L.) accumulate 1-deoxynojirimycin (DNJ), a mulberry iminosugar, by feeding on mulberry leaves. DNJ is a potent α-glucosidase inhibitor that helps prevent diabetes. However, iminosugars are toxic to many insects. In this study, we analyzed the concentrations of three major mulberry iminosugars—DNJ, 2-O-α-d-galactopyranosyl-DNJ (GAL-DNJ), and fagomine—in the larvae of mulberry-feeding insect species (two mulberry specialists and six non-specialists), to clarify the differences in accumulation, metabolism, and excretion of iminosugars between specialists and non-specialists. DNJ and fagomine concentrations in the two Bombyx larvae were much higher than those in the other larvae. GAL-DNJ concentrations were low in all species. DNJ and fagomine concentrations in the excrements of Agrotis segetum (Denis and Schiffermüller) and Sarcopolia illoba (Butler) larvae were lower than in those of the other larvae. Further, iminosugar concentrations in the hemolymph of B. mori, B. mandarina Moore, and S. illoba larvae were analyzed. DNJ and fagomine concentrations in the hemolymph of the two Bombyx larvae were much higher than in that of S. illoba. DNJ concentrations in the whole bodies of the two Bombyx larvae decreased as they developed. Similarly, DNJ, and fagomine concentrations in the hemolymph of B. mori larvae decreased with growth.  相似文献

American foulbrood is a severe bacterial disease affecting larvae of the honeybee Apis mellifera and it is caused by Paenibacillus larvae larvae. The disease is present worldwide and cases have been reported in almost all the beekeeping regions of the five continents. During 2001 and 2002 we carried out a nationwide study to assess the presence and amount of P. l. larvae spores in honey samples from Uruguay, combining classic bacteriological, and molecular approaches. The distribution of P. l. larvae spores in honey of the whole country showed a clear pattern and may provide useful data for a control and prevention strategy of American foulbrood.  相似文献
