高等生物的一个重要代谢调控机制是通过对酶的磷酸化和去磷酸化来进行的,哺乳动物的丙酮酸脱氢酶复合体(pyruvate dehydrogenase complex,PDHc)也是如此。PDHc的活性的调节主要是通过对其E1(pyru-vate dehydrogenase,PDH)的磷酸化和去磷酸化来实现的。当机体主要靠储存的脂肪生存而所存的葡萄糖仅供大脑和神经组织等只能依靠葡萄糖来提供能量的器官使用的时候,即葡萄糖缺乏时,就需要抑制PDHc的活性。主要探讨了哺乳动物在特定器官中和特定的一些生理条件下,PDHc活性改变的一些规律。  相似文献

革兰氏阴性菌在生长繁殖过程中需要从外界摄取营养物质。一些小分子营养物质可以自由地通过革兰氏阴性菌的细胞膜,而一些大分子营养物质的转运需要特异性的TonB复合物依赖性的外膜受体进行转运。TonB复合物由TonB、ExbB、ExbD构成,是革兰氏阴性菌对外界营养物质主动转运过程的能量提供单位,在革兰氏阴性菌分布广泛。近年来,对TonB-ExbB-ExbD复合物的功能、结构及作用机制取得了重大研究进展,然而此复合物在不同的细菌也存在功能及作用机制上的差异。基于此背景,本文综述了TonB复合物的功能和结构研究进展,并分析了TonB复合物在革兰氏阴性菌中的分布、进化,比较了不同革兰氏阴性菌此复合物的差异,有助于进一步发现和揭示TonB复合物的新功能与作用机制。  相似文献

Abstract The Saussurea nipponica complex was examined with enzyme electrophoresis. Earlier morphological evidence suggested that the complex comprises S. sugimurai and seven subspecies of S. nipponica . Electrophoretic evidence supports the separation of the two species and the subspecies classification of S. nipponica . Nei's genetic distance ( D = 0.19) between S. sugimurai and S. nipponica exceeds the estimated infraspecific level. Saussurea nipponica is newly defined as a species composed of seven infraspecific taxa which have their own distribution ranges and habitats. They are in a very early stage of differentiation. The extensive morphological diversity in S. nipponica is presumed to be a result of geographical isolation under strong environmental selection.  相似文献

上皮细胞和组织发生癌变时,通常会伴随细胞极性丧失和组织结构紊乱的现象。然而,极性丧失对肿瘤发生的贡献却一直存在争论。随着控制上皮细胞顶-底极性分子机制的逐步揭示,人们发现,这一极性机制与肿瘤发生联系紧密。上皮细胞顶-底极性的确立主要依赖于顶膜区Par复合物与底侧膜区Scrib复合物之间的拮抗。当Scrib复合物活性下调或Par复合物活性上调时,两复合物间的这种相互制约的平衡就会被打破,从而导致肿瘤发生。本文主要综述Scrib复合物和Par复合物如何参与上皮细胞顶-底极性的建立,以及两者间的互作与肿瘤发生的关系。  相似文献

Isoprenoid ether lipids are common to the ‘urkingdom’ archaebacteria, within which members of the Caldariella group of extreme thermoacidophiles and some methanogenic species have lipids based on macrocyclic tetraethers containing two 16,16′-biphytanyl chains. 肝萎缩增生复合征(atrophy-hypertrophy complex,AHC)是指肝组织萎缩和代偿性增生的一种临床病理特征,常见的病因有门静脉流入受阻、肝静脉流出受阻、胆道梗阻等。AHC常伴有区域性肝组织的解剖、病理、代谢功能的改变,包括肝脏沿肝门轴的旋转、萎缩肝叶的纤维化和门管区小门静脉的狭窄或血栓形成等。肝萎缩-增生复合征萎缩肝组织仍具有部分代谢功能和生物转化功能,并可能对维持病变肝脏的正常肝功能具有重要的作用,萎缩肝组织仍具有部分代谢功能和生物转化功能,对萎缩肝组织的代谢分区和代谢功能的进一步研究具有重要意义。本文通过分析近年来国内外有关AHC的文献,探讨AHC的病因、病理变化及萎缩肝组织的代谢功能分化特征的研究进展。  相似文献

Pyruvate dehydrogenase complex (PDHC) was purified from bovine kidney with a specific activity of 12-16 mumol of NADH or acetyl-CoA formed/min/mg protein. The four peptides comprising its three catalytic components were separated by sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Rabbit antibodies against this highly purified PDHC (anti-PDHC) exhibited similar binding affinity to the phospho-PDHC as it did to the PDHC antigen. To test whether there exist brain isozymes of PDHC differing from kidney enzyme, which has been extensively characterized, the PDHCs in bovine brain and kidney were compared using this anti-PDHC. The PDHC activities in the brain and kidney mitochondrial extracts were inhibited to the same degree by varying amounts of anti-PDHC. Brain PDHC was precipitated with the anti-PDHC and resolved by SDS-PAGE. The four brain PDHC peptides isolated immunochemically with anti-PDHC had the same sizes as the kidney PDHC peptides. These PDHC peptides from kidney and brain were further compared by their peptide fragment patterns, which were generated by partial proteolysis with Staphylococcus aureus V8 protease or by CNBr and resolved by SDS-PAGE. The peptide patterns generated with the former method indicated that the alpha and beta peptides of the pyruvate dehydrogenase (E1) component and the peptide of dihydrolipoyl transacetylase (E2) component of kidney PDHC were very similar to the corresponding peptides immunologically isolated from brain. The peptide patterns generated with CNBr further confirmed that the beta E1 and E2 peptides of kidney PDHC were similar to the corresponding peptides from brain.  相似文献

Numerous components of signaling pathways involved in key cellular processes reside on the Golgi complex. Here, we will focus on the roles of signaling proteins that regulate cargo trafficking along the anterograde and retrograde pathways. Emphasis will also be put on the effects of these regulatory proteins on the maintenance of the structure and function of the Golgi, and in particular on the phosphorylation of key components of the transport machinery. These pathways position the Golgi complex as a central hub in the regulation of cell signaling. To date, however, the activation and coordination of these signaling molecules remain a mystery. Being able to describe the interplay between several of these signaling pathways and secretion, and the flow of information through these pathways, will help us to understand how the secretory machinery works and how it interacts with other cellular functions. This will also advance our understanding of how the secretory pathway functions under physiological circumstances, and how its dysregulation can initiate pathological conditions.  相似文献

In this work we studied permeability transition by incubating mitochondria in the presence of 50 M Ca2+ and malate/glutamate as substrates. This condition, besides inducing the release of pyridine nucleotides, promotes the generation of reactive oxygen-derived species by the complex I of the respiratory chain. The latter leads to the opening of the mitochondrial permeability transition pore. Ca2+ release, mitochondrial swelling and collapse of the transmembrane electric potential, were analyzed to assess this process. We propose that the mechanism for pore opening, in addition to the oxidative stress, involves the uncoupling effect of fatty acids providing activation of phospholipase A2, lipid peroxidation, and the oxidation of membrane thiols. This proposal emerges from the data indicating the protective effect of bovine serum albumin and N-ethylmaleimide. The key role of reactive oxygen species was implied based on the fact that the scavenger -phenyl-tert-butyl nitrone inhibited pore opening.  相似文献

The spermatozoon of Hemirhamphodon pogonognalhus shows modifications that are frequent though not obligate in internally fertilizing sperm, notably elongation of the nucleus and extension of the mitochondria of the midpiece as an elongate sheath around the proximal region of the axoneme. These similarities to poecilid and jenynsid sperm are considered homoplasic. As in the mature sperm of all but one investigated teleost, an acrosome is absent. The elongate, blade-shaped, electron-dense nucleus has a mean length of 3.2 μm; its basal implantation fossa, less than one-tenth of the length of the nucleus, houses the anterior half of the distal and only centriole (of triplet construction with satellite rays), a centriolar plug, and a mass connecting the centriole to the wall of the fossa. A unilateral putative centriole adjunct is present. The anterior region of the axoneme is surrounded by a mitochondrial sleeve, and internal to this, separated by a cisterna, by a submitochondrial sleeve. The mitochondrial sleeve unites posteriorly with the submitochondrial sleeve. Between the submitochondrial sleeve and the axoneme is a space, the cytoplasmic canal, that is open to the exterior posteriorly. The discrete, cristate mitochondria, in their sleeve, are unique in investigated atherinomorph sperm in being bilateral, grouped on only two opposing sides of the axoneme, with an arc-shaped ‘intermitochondrial link’ between. The 9 + 2 flagellum is unique for the Animalia in having 23 radial subplasmalemmal rods, repeated longitudinally (periodicity 0.025 pm) in a quasicrystalline array. Internal fertilization is deduced to have arisen in the Exocoetoidei independently of that in the Cyprinidcntiformes.  相似文献

The effects of genetically determined changes in leaf morphology on the characteristics of growth, pigment complex, and productivity were studied in pea plants (Pisum sativum L.). The homeotic afila(af) mutation, which transformed leaflets into tendrils, decreased the leaf area and the chlorophyll (Chl) content per plant (CCP) in the af/af plants 1.5-fold as compared to the wild type (Af/Af). The loss of leaflets in the af/af plants was partly recompensed by expansion of the tendrils and stipules and by extra accumulation of Chl (a + b). The mutation did not affect Chl (a + b) that fell to the share of light-harvesting complexes (LHC) and the ratio of Chl a/b (representing the relative distribution of chlorophylls between LHC and the reaction centers); neither it affected the quantum efficiency of the primary charge separation (F v/F m). The diminished assimilating area (AA) in the af/af plants at the preflowering period did not reduce the final biomass and grain yield. The measurement of the area shaded by plants in the glasshouse experiments and the direct assessment of the vertical profile of solar radiation in the field stand canopies demonstrated that this phenomenon was in particular related to the fact that, in the af/af plants, the solar radiation was available to the apical and subapical leaves (as in the wild-type plants) and also to the lower metamers. As a result, the actively functioning AA expanded, and the photoassimilating potential of the af/af plants was enhanced. Our data presume the direct relationship between plant production and CCP.  相似文献

The nuclear pore complex (NPC) is a giant protein assembly that penetrates the double layers of the nuclear membrane. The overall structure of the NPC has approximately eightfold symmetry and is formed by approximately 30 nucleoporins. The great size and complexity of the NPC have hindered the study of its structure for many years until recent breakthroughs were achieved by integrating the latest high-resolution cryo-electron microscopy (cryo-EM), the emerging artificial intelligence-based modeling and all other available structural information from crystallography and mass spectrometry. Here, we review our latest knowledge of the NPC architecture and the history of its structural study from in vitro to in situ with progressively improved resolutions by cryo-EM, with a particular focus on the latest subnanometer-resolution structural studies. The future directions for structural studies of NPCs are also discussed.  相似文献

在自然生态系统中,鼠类并非完全有害,作为生态系统中的一个组成部分和物流与能流的一环,有其存在的必然性。只有在与人类的生活和生产发生冲突时,鼠类才表现出其危害,生态系统的失衡是鼠类暴发成灾最根本的原因。目前,害鼠对工农业生产造成严重的危害,并威胁着人们的身体健康,害鼠防治的异化则对生态环境造成严重的影响。对鼠害的治理是一项社会性的工程,当地政府的政策法令、组织管理、人们的科学化素质以及科技普及程度,对害鼠的控制有着重要的作用。从长远发展来讲,随着社会的进步,经济的发展,人民生活水平的提高,害鼠种群数量将会受到控制而趋于下降。但中短期而言,社会经济建设的某些活动可能引发局部或大面积害鼠种群的暴发。因此在实施那些措施时,若能注意害鼠种群的发生发展,或对有关项目可能对鼠类生存环境的影响进行预测,并采取有效措施控制害鼠种群,将会减少害鼠带来的各种损失,更有利于我们的经济建设。由此提出,可适时地采用“社会—经济—自然复合生态系统”的思维,并关注目前社会经济活动中重大举措对鼠类群落的影响,科学地制定鼠害控制对策。  相似文献
