Sediment and tailings samples were collected from sites with a contrasting physicochemical gradient to investigate microbial squalene-hopene cyclase (sqhC) composition and distribution in terrestrial environments. Acidobacteria (66%), Alphaproteobacteria (96%) and Gammaproteobacteria (55%) were found to dominate sqhC communities, respectively, at the acidic Dajiuhu Peatland, the alkaline Heshang Cave and strongly acidic tailings of Tongling copper mine in China. Statistical analysis confirmed that pH was the important factor impacting the geographical distribution of sqhC at phylum level. sqhC gene abundance is comparable at the three sites. However, the total amount of hopanoids in per gram total organic matter content (TOC) is 1.75 times higher in the acidic peatland than that in the alkaline Cave and it is below the detecting limit in tailings of Tongling copper mine, inferring the potential impact of pH in regulating the hopanoid production.  相似文献

通过在小麦收获后复种绿肥毛叶苕子研究绿肥的培肥效应。在复种绿肥毛叶苕子的情况下,研究后茬作物小麦生育期土壤微生物生物量碳、氮的时空变化。结果表明,小麦生育期0~20 cm土层土壤微生物生物量碳、氮均表现为有毛叶苕子处理高于无毛叶苕子处理,其中施肥70%化肥+毛苕子翻压还田处理的土壤微生物生物量碳、氮最高。小麦苗期土壤微生物生物量碳、氮含量最低,至抽穗期土壤微生物生物量碳、氮含量达到最高,成熟期又有所降低。微生物熵在各时期的变化与微生物生物量碳一致,绿肥处理表现出明显较高的微生物熵。  相似文献

多环芳烃(Polycyclic aromatic hydrocarbons,PAHs)是土壤中广泛存在的、美国环保总署(USEPA)优先控制的一类有毒(致癌、致突变)的持久性污染物,主要来源于人类活动。土壤微生物多样性是表征土壤质量变化的敏感指标之一。磷脂脂肪酸(PLFAs)分析方法是基于活性微生物细胞膜的PLFAs组分的生化检测技术,克服了传统培养方法只能分离出少量微生物(1%)的缺点。采用PLFAs方法,解析了土壤活性微生物对PAHs污染胁迫的反应。结果表明,土壤微生物分布情况可分为4种类型:Ⅰ型,微生物PLFAs种类最多,占该区土壤微生物PLFAs种类总数的57.7%,PAHs对变量的解释量最小;Ⅱ型,微生物PLFAs占PLFAs总数的30.8%,PAHs对变量的解释量较小;Ⅲ型,微生物PLFAs种类占总数的7.68%,PAHs对变量的解释量较大;Ⅳ型,微生物PLFAs的种类仅占总数的3.85%,PAHs对变量的解释量最大。相关性分析表明:土壤微生物PLFAs的种类、生物量和生态多样性指数与土壤中萘(Nap)、芴(Flu)、蒽(Ant)、苯并[K]荧蒽(Bkf)、苯并[a]芘(Bap)、茚并[1,2,3-cd]芘(Ind)的相对含量呈负相关关系;与苊(Ace)、菲(Phe)、荧蒽(Fla)、芘(Pyr)、苯并[a]蒽(Baa)的相对含量呈正相关关系;与PAHs的种类和浓度呈负相关关系。结果将为开展PAHs污染土壤的生态风险评价和微生物生物修复技术研究提供理论依据。  相似文献

对微生物谷氨酰胺转胺酶(MTG)超滤浓缩的工艺条件进行了探讨及优化。实验采用截留分子量为30 kDa的聚醚砜(PES)膜,当发酵液初始pH为7,超滤浓缩倍数为4倍时,可以得到理想的MTG回收率。同时对超滤液中蛋白酶的变化进行了分析,发现随着超滤倍数的提高蛋白酶也逐渐提高,但在浓缩4倍以后达到较稳定的水平。聚醚砜(PES)超滤膜使用后用稀释的NaOH溶液浸泡清洗处理50 min后,膜通量可以恢复98.12%。  相似文献

微生物燃料电池(Microbial fuel cell,MFC)是一种近几年快速发展的废物处理与能源化技术,可以与污水处理、污染物降解、脱盐等环境技术结合。微生物燃料电池与堆肥技术结合可以在处理日益增长的固体废弃物的同时回收能量,具有很好的发展前景。文中分析了堆肥微生物燃料电池系统的微生物特征,探讨了堆肥过程中影响微生物燃料电池产电性能的因素,包括电极,隔膜,供氧和构型。最后归纳说明了堆肥微生物电池作为一种新的废弃物处理技术的特点:较高的微生物量并可产生较高的电流密度;对不同环境的适应性强;可以自身调节温度,能源利用效率高;质子从阳极向阴极的移动会受到不同堆肥原料的影响。  相似文献

Aims: As a biosafety laboratory, we survey the handling of adenovirus type 5 (Ad5) and HIV1‐derived lentivirus in contained‐use facilities in Switzerland to identify insufficiencies of the safety precautions taken by the laboratories. Methods and Results: In the past 9 years, we took 430 swab samples from various types of surfaces in research laboratories. Samples were examined for Ad5 contaminations by real‐time PCR and infectivity assay or for the presence of lentivirus (HIV1) nucleic acids by real‐time (RT) PCR. Samples collected from centrifuges did not only contain Ad5 DNA more frequently but also exhibited higher numbers of Ad5 and lentiviral (HIV1) DNA copies than swabs from any other area of sampling. Five of ten samples containing infectious Ad5 particles or lentivirus (HIV1) RNA were found in samples taken from centrifuges. Ad5 contamination rates were higher in the tube holder and lower on the inner wall of the rotor chamber in centrifuges that were fitted with aerosol tight covers compared to centrifuges without covers. Conclusions: Our results allowed the comparison of hygiene standards of different laboratories and lead to the identification of centrifuges as hotspots for contaminations. Significance and Impact of the Study: Based on our results, we propose to use the collected data as a tool for rating future swab results. Furthermore, the amount of Ad5 and HIV1‐derived lentivirus DNA could serve as an indicator of the level of good laboratory practice in contained‐use laboratories handling these viral vectors.  相似文献

土壤中蕴藏着高度的微生物多样性,在陆地生态系统中发挥着非常重要的功能,加强对土壤微生物资源的综合管理与开发应用是提升生态系统稳定性与生产力及农产品质量的重要途径。首先,土壤微生物多样性具有全球性的重大意义,有待完善对土壤微生物的检测与监测技术研究,进而实现土壤微生物多样性与土壤功能的耦合以及对土壤质量的评定;其次,土壤微生物作为一种宝贵的生产资料和可持续资源,要加强其在土壤肥力强化与保育、土壤障碍消减与调节、土壤污染控制与修复等3个领域的应用研究。最后,未来土壤微生物学发展将会形成土壤微生物系统学、土壤微生物过程学与土壤微生物功能学3个子学科,要建立土壤微生物种质资源库与遗传信息库,推进土壤微生物生理代谢过程、生物化学过程及生态行为过程的研究,联结土壤微生物与土壤功能的关系,并从土壤中的功能微生物出发对环境变化作出积极响应和主动调控。此外,原创性方法的建立与应用是限制土壤微生物学发展的技术瓶颈,联合生物地理学与生物信息学破译重要基因的特定生态功能,并将其应用到生态模型以及生态系统未知领域的研究中去,是土壤微生物学面临的挑战。  相似文献

本研究旨在开发基于微生物燃料电池(microbial fuel cell,MFC)的微生物电化学传感器对水环境重金属污染进行实时在线监测。构建微生物电化学传感器,采用错流式布水方式、优化外阻值及外循环速率等参数,在最优参数下分析重金属模拟废水浓度对电压的抑制情况。结果表明,当外阻值为130Ω、外循环速率为1.0 mL/min时,MFC电化学传感器的性能最优。在此条件下对1–10 mg/L的Cu2+、0.25–1.25 mg/L的Cd2+、0.25–1.25 mg/L的Cr6+和0.25–1.00 mg/L的Hg2+均有响应,60 min内输出电压最大抑制率分别可达92.95%、73.11%、82.76%和75.80%,线性相关系数均大于0.95,且有较好的生物学重复性。该MFC电化学传感器具有良好的重金属检测性能,可为实现水环境重金属污染实时在线监测技术的实际应用提供理论依据。  相似文献

We analyzed PCR-amplified 16S rRNA genes from native and Fe(III)-enriched surface sediments of a major tidal channel in the Tijuana River Estuary, California, USA. Clones from native sediments were most closely affiliated with photosynthetic taxa (Cyanobacteria, Chloroflexi, and Halochromatium) and microorganisms known to reduce (Desulfatibacillus, Desulfobacterium, and Desulfuromusa) or oxidize (Microcoleus, Phormidium, and Halochromatium) various sulfur species, reflective of the fluctuating redox conditions in the tidal zone. Fe(III) was rapidly reduced in anaerobic microcosms amended with 2-line ferrihydrite, with or without the sulfate reduction inhibitor sodium molybdate. The addition of ferrihydrite without molybdate caused a major shift in community structure to a dominance of the Fe(III)-reducing genus Shewanella, while at the same time the sulfate-reducing and sulfide-oxidizing populations were replaced by taxa known to cycle elemental sulfur. Sediments amended with both ferrihydrite and molybdate were again populated by Shewanella clones, but also numerically important were clones most similar to Marinobacterium, Pseudomonas, and Bacillus, suggesting a role for these taxa in Fe(III) reduction in marine habitats.  相似文献

Production of microbial biomass through fermentation of pretreated rice straw using Penicillium janthinellum (St-F-3B) is reported, with emphasis on the use of non-effluent generating pretreatment procedures. The fungus readily metabolized a rice straw substrate that had been subjected to alkali pretreatment by steaming at atmospheric pressure followed by the neutralization of the alkali. The crude protein content of the microbial biomass averaged 15–20%. The fermentation could be carried out in aerated-agitated fermenters using fertilizer grade nutrient salts to produce a biomass with 17.5% crude protein. When operated on a semicontinuous basis using 20% of the previous batch as inoculum, successive batches produced a biomass product with 12–19% crude protein content in 48 h. The problems of developing a technology for protein from agricultural residues with particular reference to developing countries are discussed.  相似文献

采用传统平扳分离培养方法和PCR—DGGE技术研究了水稻秸秆腐解复合菌系RSS-4在腐解稻秆过。程中菌种区系变化情况。结果表明:平板分离培养方法显示,在稻秆腐解过程中,微生物的数量呈现出先升后降的变化趋势,在整个腐解过程中细菌的数量占优势;DGGE图谱显示,至少有12种细菌和18种真菌的近缘种参与到稻秆的腐解过程。在其腐解过程中,不同腐解阶段真菌的组成呈现出多样性,数量变化差异也较大:细菌DGGE图谱中的条带1、9、10等以及真菌DGGE图谱中的条带8、9、13等为优势菌株,它们贯穿于稻秤腐解的整个过程;细菌中的条带12以及真菌中的条带4在腐解的前期起作用,而后迅速消失;细菌中的条带3、11等以及真菌中的条带3、10等在腐解的后期才出现而起作用;而细菌中的条带2以夏真菌中的条带1、5等仅出现在腐解的莱一时期。  相似文献

The remediation of uranium from soils and groundwater at Department of Energy (DOE) sites across the United States represents a major environmental issue, and bioremediation has exhibited great potential as a strategy to immobilize U in the subsurface. The bioreduction of U(VI) to insoluble U(IV) uraninite has been proposed to be an effective bioremediation process in anaerobic conditions. However, high concentrations of nitrate and low pH found in some contaminated areas have been shown to limit the efficiency of microbial reduction of uranium. In the present study, nonreductive uranium biomineralization promoted by microbial phosphatase activity was investigated in anaerobic conditions in the presence of high nitrate and low pH as an alternative approach to the bioreduction of U(VI). A facultative anaerobe, Rahnella sp. Y9602, isolated from soils at DOE's Oak Ridge Field Research Center (ORFRC), was able to respire anaerobically on nitrate as a terminal electron acceptor in the presence of glycerol-3-phosphate (G3P) as the sole carbon and phosphorus source and hydrolyzed sufficient phosphate to precipitate 95% total uranium after 120 hours in synthetic groundwater at pH 5.5. Synchrotron X-ray diffraction and X-ray absorption spectroscopy identified the mineral formed as chernikovite, a U(VI) autunite-type mineral. The results of this study suggest that in contaminated subsurfaces, such as at the ORFRC, where high concentrations of nitrate and low pH may limit uranium bioreduction, the biomineralization of U(VI) phosphate minerals may be a more attractive approach for in situ remediation providing that a source of organophosphate is supplied for bioremediation.  相似文献

多环芳烃(PAHs)是环境中普遍存在的一类有机污染物,微生物的降解是PAHs去除的主要途径。近年来,有关PAHs微生物降解途径和代谢产物的研究已有很多报道。小分子PAHs一般可以直接被微生物降解,而大分子PAHs则需要微生物以共代谢的方式降解。在过去20年中,微生物降解PAHs的基因相继被发现,各种基因在调控PAHs降解过程中的功能也越来越清晰。本文概述了PAHs微生物降解基因方面的研究进展,详细介绍了微生物对萘、菲的降解基因,最后对PAHs微生物降解基因的应用前景进行了展望。  相似文献

In this study, the use of biodegradable polymers for microencapsulation of naltrexone using solvent evaporation technique is investigated. The use of naltrexone microspheres for the preparation of matrix devices is also studied. For this purpose, poly(L-lactide) (PLA) microspheres containing naltrexone prepared by solvent evaporation technique were compressed at temperatures above the Tg of the polymer. The effect of different process parameters, such as drug/polymer ratio and stirring rate during preparation of microspheres, on the morphology, size distribution, and in vitro drug release of microspheres was studied. As expected, stirring rate influenced particle size distribution of microspheres and hence drug release profiles. By increasing the stirring speed from 400 to 1200 rpm, the mean diameter of microspheres decreased from 251 μm to 104 μm. The drug release rate from smaller microspheres was faster than from larger microspheres. However, drug release from microspheres with low drug content (20% wt/wt) was not affected by the particle size of microspheres. Increasing the drug content of microspheres from 20% to 50% wt/wt led to significantly faster drug release from microspheres. It was also shown that drug release from matrix devices prepared by compression of naltrexone microspheres is much slower than that of microspheres. No burst release was observed with matrix devices. Applying higher compression force, when compressing microspheres to produce tablets, resulted in lower drug release from matrix devices. The results suggest that by regulating different variables, desired release profiles of naltrexone can be achieved using a PLA microparticulate system or matrix devices.  相似文献

Freestanding layered membrane–based devices have broad applications in highly efficient energy‐storage/conversion systems. The liquid–solid interface is considered as a unique yet versatile interface for constructing such layered membrane–based devices. In this review, the authors outline recent developments in the fabrication of soft materials to functionalize layered devices from the aspect of liquid–solid interfacial assembly and engineering arts. Seven liquid–solid interfacial assembly strategies, including flow‐directed, superlattice, solvent‐casting, evaporation‐induced, dip‐coating, spinning, and electrospinning assemblies, are comprehensively highlighted with a focus on their synthetic pathways, formation mechanisms, and interface engineering strategies. Meanwhile, recent representative works on layered membrane–based devices for electrochemical energy applications are presented. Finally, challenges and opportunities of this research area are highlighted in order to stimulate future developments. This review not only offers comprehensive and practical approaches to assemble liquid–solid interfaces with soft materials for various important layered electrochemical energy devices but also sheds lights on fundamental insights by thoughtful discussions on performance enhancement mechanisms of these electrochemical energy systems.  相似文献
