By means of the electron cytochemical method ribonucleoprotein (RNP) particles, condensed chromatin (CCh) and ribosomes of cytoplasm are describe in normo- and hyperchromic neurons (HChN) of the V and VI layers of the sensomotor cortex. The normochromic neurons are characterized by nearly a complete absence of CCh, a great number of fibrillar RNP particles. The ribosomes of cytoplasma are organized as polysomes. This demonstrates a high metabolic activity of the DNA-RNA-protein system in these cells. In nuclei of one HChN group numerous small CCh clumps are revealed, amount of RNP particles does not change noticeably, comparing the nuclei of the normochromic cells. In cytoplasm a partial dissociation of polysomes takes place. All this demonstrates a decreased RNA synthesis in the nucleus and protein in cytoplasm of the given cells. In another HChN group the nucleus is filled with large CCh clumps. The number of fibrillar RNP particles decreases noticeably, and the number of granular ones increases. A complete dissociation of polysomes occurs. This demonstrates that in the cells mentioned not only RNA and protein synthesis is decreased, but the processing and nucleo-cytoplasmic transport of RNA is disturbed. The presence of transitional forms between the neuronal forms described makes it possible to suppose certain cyclicity in the work of their plastic apparatus, the normo- and hyperchromic neurons being morphologic equivalents of certain phases of the cycle.  相似文献

The effects of zinc on the ribonucleoprotein (RNP) constituents of HEp-2 cells have been analyzed. Pulse-chase autoradiographic experiments show a preferential inhibition of nucleolar RNA synthesis and a block in the transport of nucleolar and extranucleolar RNA in zinc-treated cells. Concomitantly with the disturbance in RNA metabolism and in protein synthesis, nucleolar condensation, accumulation of perichromatin granules and fibrils, condensation of interchromatin fibrils, and appearance of dense granular bodies occur. Accumulation of perichromatin fibrils and condensation of interchromatin fibrils appear to be related to the block in the transport of heterogeneous nuclear RNA. Depletion of certain proteins required for the assembly of RNP particles could share in the abnormal behavior of RNA and lead to the accumulation of perichromatin granules and the appearance of dense granular bodies.  相似文献

Summary— The in situ distribution of phosphorus in perichromatin granules (PCGs), and in the surrounding nucleoplasm was investigated in rat liver cells by means of electron spectroscopic imaging of unstained preparations. A 2–3 nm fibril containing high concentration of phosphorus was found to be the main substructural feature of the PCGs revealed in the maps of phosphorus. This fibril is folded within the PCG with no apparent order. Fibrils of similar diameter and phosphorus content were also found in both the halo surrounding the PCG and dispersed in the nucleoplasm. Some of such fibrils are in continuity with those occurring within PCGs. Sometimes these fibrils are grouped forming a stalk connecting the PCG to chromatin. Some stalked PCGs are U-shaped or kidneyshaped, resembling Balbiani ring granules in the process of formation as observed in Chironomus salivary gland cell nuclei. The external fibrils are interpreted as perichromatin fibrils considered to be precursors of PCGs.  相似文献

Ultrastructural changes in the nuclear DNP and RNP components of human NB cells induced by synchronous infection with H-1 parvovirus were studied using Bernhard's EDTA method of staining. Early events (12 h after infection) occurred in the nucleolus. Chromatin within the nucleolar fibrous centers condensed thereby converting the centers to vacuoles. DNP associated with the granular nucleolonema also contracted markedly, causing a disruption of this skein-like structure; it then migrated peripherally forming a heterochromatic cortex surrounding the granular nucleolar vestige. Subsequently (24–36 h after inoculation), condensation of extranucleolar chromatin took place concurrently with the accumulation of extensive amounts of interchromatin granules in the nucleus and cytoplasm. Conglomerates of perichromatin fibrils and interchromatin granules were frequently juxtapposed to the condensing chromatin. Large clumps of interchromatin granules were also closely associated with fragmenting nucleoli, and the apparent transformation of nucleolar granules into interchromatin granules was observed. Accumulation of H-1 protein on chromatin evidently fostered its condensation resulting in the pathology described.  相似文献

Nuclear ribonucleoprotein particles were studied in the central nervous system of the rat after fixation by perfusion with hypotonic-detergent formaldehyde. Beads-on-a-string structures (polyparticles or chains) formed of 14-nm granules related by a 7-nm thick filament were found. These polyparticles are positively contrasted by a preferential method for ribonucleoproteins and they are sensitive to RNase. Perichromatin granules are loosened by this fixation and their internal structure is clearly depicted. They are composed of a tangled mass of 3-nm thick filaments, but lack 14-nm granules. The internal filaments of the perichromatin granules display frequent continuities with polyparticles. These results show that polyparticles correspond to perichromatin fibrils visualized in nuclei by standard fixation procedures.  相似文献

An immunoelectron study of nuclear distribution of pre-mRNA splicing and pre-rRNA processing factors was carried out for oocytes of two turbellarian species: the Baikal endemic Geocentrophora wagini and a cosmopolitan G. baltica. Using monoclonal antibodies against Sm-epitope of small nuclear RNPs (snRNPs) and SR-protein SC35, it has been shown that on different stages of oocyte growth splicing factors (snRNPs and SC35) are distributed within the whole nucleus. A fibrogranular material located near heterochromatin clumps is labeled with these antibodies. A fibrillar part of this material seems to represent perichromatin fibrils. The features of intranuclear distribution of splicing factors in Geocentrophora oocyte nuclei and their ultrastructural features suggest that pre-mRNA synthesis and splicing may occur up to the end of diplotene. In Geocentrophora oocyte nuclei a few nuclear bodies (NBs) were found. Splicing factors (snRNPs and SC35) and fibrillarin were revealed in these NBs. Homology of Geocentrophora oocyte NBs to coiled bodies of oocyte and somatic cell nuclei of other animals is discussed. During diplotene, Geocentrophora oocyte nucleoli were found to lose their granular component and to change to large fibrillar structures named "postnucleoli". The postnucleoli contain both fibrillarin and non-nucleolar spliceosomal components (snRNPs and SC35). Geocentrophora oocyte postnucleoi are compared with similar structures of mammalian oocyte nuclei, taken as an example of morphological convergence of nuclear structure organization in phylogenetically distant animal species.  相似文献

The fate of different nuclear ribonucleoprotein (RNP) components was investigated during spontaneous apoptosis of thymocytes using specific monoclonal antibodies against snRNPs, hnRNPs, and ribosomal proteins at light and electron microscopy levels and by flow cytometry. It was found that, during apoptosis, nuclear RNP-containing structures (perichromatin granules, interchromatin granules, and perichromatin fibrils) segregate in the interchromatin space and cluster into heterogeneous aggregates of granules in which some of the structures may still be recognized morphologically. Along with the progress of apoptosis, the clusters are extruded from the nucleus into the cytoplasm, from which they are finally released via cytoplasmic extrusions. At all these stages, RNPs inside the clusters are always recognized by specific antibodies, even when they bleb out of the cell surface, thus suggesting that degradation of RNPs might be only partial during apoptosis. This could be potentially harmful in genetically susceptible subjects, as the appropriate MHC class II molecules may capture and present normally cryptic self-peptides. It is tempting to speculate that this event might have implications in the etiology of autoimmune diseases.  相似文献

The ultrastructural distribution of nuclear ribonucleoproteins (RNP) has been investigated by incubation of thin sections of mouse or rat liver, embedded in Lowicryl K4M or prepared by cryoultramicrotomy, with antibodies specific for RNP. The antibodies were localized by means of a protein A-colloidal gold complex. Anti-small nuclear (sn)RNP antibodies, specific for determinants of the nucleoplasmic snRNP species containing U1, U2, U4, U5, and U6 RNAs, were found associated preferentially with perichromatin fibrils, interchromatin granules, and coiled bodies. This indicates an early association of snRNP with structural constituents containing newly synthesized heterogeneous nuclear RNA. It also suggests a possible structural role of some snRNPs in nuclear architecture. Antibodies against the core proteins of heterogeneous nuclear RNP particles associate preferentially with the border regions of condensed chromatin, and in particular with perichromatin fibrils and some perichromatin granules. These results are discussed in view of recent knowledge about the possible role of nucleoplasmic RNP-containing components in the functions of the cell nucleus.  相似文献

Small nuclear ribonucleoproteins (snRNPs) containing U1, U2, U4, U5, and U6 small nuclear RNAs were detected by ultrastructural immunocytochemistry in the nuclei of isolated rat hepatocytes using Fab fragments of anti-Sm and anti-RNP autoantibodies. Their localization was carried out in normal cells and in cells treated with two drugs, the adenosine analog DRB and CdCl2, which alter the number and distribution of nuclear RNP components. It was found that more precise determination of the distribution of these small RNAs could be obtained by using two complementary procedures in parallel rather than either one alone. They consisted of an indirect immunoperoxidase labeling carried out before embedment and an indirect immunogold labeling applied to thin sections of cells embedded in Lowicryl K4M. The results indicate that snRNPs are associated with all extranucleolar perichromatin fibrils and granules and interchromatin fibrils, which confirms that they occur in structures involved in the synthesis and processing of hnRNA. The snRNPs are not associated with nucleolar perichromatin granules induced by DRB, which confirms that there may be two kinds of perichromatin granules. The snRNPs are also associated with the still enigmatic interchromatin granules which apparently do not contain hnRNA but at least in DRB-treated cells, also contain ribosomal RNA.  相似文献

Cryoelectron microscopy of vitrified sections has become a powerful tool for investigating the fine structural features of cellular compartments. In the present study, this approach has been applied in order to explore the ultrastructural morphology of the interphase nucleus in different mammalian cultured cells. Rat hepatoma, Chinese hamster ovary and Potorus kidney cells were cryofixed by high-pressure freezing and the cryosections were examined at low temperature by transmission electron microscopy. Our results show that while the contrast of nuclear structural domains is remarkably homogeneous in hydrated sections, some of them can be recognised due to their characteristic texture. Thus, condensed chromatin appears finely granular and the perichromatin region contains rather abundant fibro-granular elements suggesting the presence of dispersed chromatin fibres and of perichromatin fibrils and granules. The interchromatin space looks homogeneous and interchromatin granules have not been identified under these preparative conditions. In the nucleolus, the most striking feature is the granular component, while the other parts of the nucleolar body, which appear less contrasted, are difficult to resolve. The nuclear envelope is easily recognisable with its regular perinuclear space and nuclear pore complexes. Our observations are discussed in the context of results obtained by other, more conventional electron microscopic methods.  相似文献

Using the system of morphometric analysis described in this paper, human peripheral blood T and B lymphocytes, labeled with specific surface markers, can be compared on different analytical levels. They show differences in their surface and the eccentricity of cells, in the relative surfaces occupied by peripheral and central condensed chromatin, in the average surface of the central chromatin clumps and in the number of perichromatin granules per nuclear surface. The morphometric analysis reveals the importance of examining the nuclear and the surface parameters in the characterization of lymphocytes, confirming that a detailed analysis of the nuclear characteristics can contribute to the identification of T and B lymphocytes by transmission electron microscopy.  相似文献

The changes in the number of perichromatin granules (PCG) and the alterations in the RNA content of the interchromatin and perichromatin regions caused by ovariectomy and estradiol injection were studied in rat endometrial fibroblast and myometrial muscle cells. Twelve rats were divided in four groups. A group of rats was fixed without any treatment, the other three groups were ovariectomized and processed 21 days after the operation. One of them was studied without further treatment, and two groups were injected intraperitoneally with 20 micrograms of 17 beta-estradiol hemisuccinate and fixed 0.5 and 2 h after the injection. The frequency of PCG was evaluated in preparations stained with EDTA procedure preferential for RNP. The alterations of RNA content were estimated by post-embedding high resolution in situ hybridization using a total DNA probe labeled with biotinilated nucleotides revealed by streptavidin coupled with 10 nm gold grains. Most of the non-nucleolar labeling is associated to RNP containing fibrils. Perichromatin and interchromatin granules are labeled to a lesser extent. Castration brings about a reduction of the number of PCG and of the numerical density of labeling in endometrial fibroblasts. The injection of estradiol causes a rapid increase in both parameters. On the contrary, the frequency of PCG and intensity of labeling of epithelial endometrial cells and in muscle cells increase after ovariectomy and are reduced by estradiol administration. These results suggest that estradiol may affect differentially various types of target cells in the same organ, and also that PCG are not the only nuclear compartment of pre-mRNA or mRNA altered by the changes in estradiol, the RNP containing fibrils located in the perichromatin and in the interchromatin regions are also involved.  相似文献
