RAPD polymorphisms detected among the flax genotrophs   总被引:6,自引:0,他引:6  
The occurrence of environmentally induced heritable changes in certain flax varieties has been shown to be accompanied by changes in the genomic DNA. A large difference in nuclear DNA contents has been characterized between the extreme types, termed genotrophs. The genomic variation between a series of genotrophs has been studied by the polymerase chain reaction using random arbitrary oligonucleotide primers. A total of 320 primers were used in the reactions and 253 polymorphic bands observed. The polymorphic bands were derived from all parts of the genome, namely the highly repetitive, middle-repetitive and low-copy-number sequences. They were also shown to be distributed thoughout the genome. In one group of genotrophs, all of which were induced by temperature treatment, there was a clustering of the polymorphisms with a high degree of shared polymorphisms. These results are in agreement with earlier studies showing that a dispersed fraction of the genome is susceptible to variation when environmentally induced heritable changes occur.  相似文献

Peroxidase, esterase, and acid phosphatase isozymes of environmentally induced L and S genotrophs, nuclear DNA reversion types, and the orginal plastic (Pl) type of the flax variety Stormont Cirrus have been compared by polyacrylamide gel electrophoresis. Differences were observed in particular line was not correlated with the nuclear DNA amount. The relationship between the isozyme pattern and the phenotypes of the lines in which they are expressed is discussed.  相似文献

Ferguson plots demonstrated that corresponding malate dehydrogenase (MDH) isozymes of Durrant's L and S flax genotrophs differ in apparent molecular weight (MW) and also in net negative charge. The MW differences explain heritable differences in electrophoretic relative mobility (R m) between corresponding L and S isozymes. The MW for each MDH isozyme was higher for L than for S and resulted in a slowerR m for L. The net negative charge for each isozyme was higher for L than for S. MDH isozymes also differ in MW within L and S. MW was lower for isozymes in leaves from the bottom of the stem than in leaves from the top of the stem, particularly in L. Anionic peroxidase isoenzymes from seedling root, hypocotyl and cotyledon regions of the large (L) and small (S) flax genotrophs were separated on acrylamide gels. Tissue cultures were initiated from each of these regions of the seedlings, and maintained for a 200-day period with six transfers. The differences in electrophoretic mobility of the peroxidase isoenzymes between L and S noted in seedlings, and also in main stems of adult plants, were still present in the tissue cultures.  相似文献

Anionic peroxidase isoenzymes, separated on acrylamide gels, were examined in two flax genotrophs and in their reciprocal F2 hybrids. Isoenzyme 1 exhibited a significant difference in Rm between stem base and apex and there was a gradient of decreasing Rm and activity between base and apex. Isoenzyme 2 displayed only the activity gradient. The parents differed significantly in the Rm's and activities of isoenzymes 1 and 2, and the F2's showed complete dominance of the L parent for Rm, with activities being approximately intermediate.  相似文献

The IAA-oxidase and peroxidase capabilities along the length of the main stem tissues of two flax genotrophs L and S and two flax genotypes R and M were examined in vitro. Stem gradients for peroxidase activity increased basipetally in all plant types, as did IAA-oxidase activity gradients at non-rate-limiting concentrations of Mn2+. Correlations between peroxidase activity and non-rate-limited IAA-oxidase activity supported the contention of dual activities on the same molecule. At rate-limiting concentrations of Mn2+, IAA-oxidase activity did not correlate with peroxidase activity. Plant type differences were detected in rate-limited IAA-oxidase activity. This activity was higher in the stem region immediately above the cotyledons (axillary buds) of the more branched types, L and R, than in the sparsely branched types, S and M.  相似文献

The chloroplast DNA of L. usitatissimum var. Stormont Cirrus has been mapped with respect to the recognition sites for the enzymes SalP1, Sst1 and SalG1. It is a circular molecule of about 160 kilobasepairs, with an inverted repeat containing the rDNA. Comparisons between chloroplast DNA of uninduced and induced flax genotrophs show there to be no major structural differences.  相似文献

Flax (Linum usitatissimum) has a genome in which changes have been associated with environmental factors. The inbred flax variety, Stormont Cirrus (Pl), served as the parent, and several lines (termed genotrophs) were derived from this parent. The phenotypes of the genotrophs were stable in a number of different growth environments, unlike the original Pl line in which changes associated with environmental factors continued to occur. These genotrophs differed from the original line in a number of characteristics, but the only known phenotypic characteristic that is shared by all the genotrophs and different from the parental, Pl, line is the lack of changes associated with the original environmental factors. However, some of these genotrophs have changed in both phenotype and nuclear DNA subsequent to their original growth and differentiation from Pl. Representational difference analysis (RDA) has been used to identify differences between Pl and all the genotrophs in an attempt to identify the loci controlling these aspects of plasticity. Subtractions between Pl DNA as a tester (target) and one of the genotrophs (individual RDA) or a mixture of different types of genotroph (L6, S6, C2, and LH) DNAs as a driver were done (combined sample RDA; csRDA). In addition, contrary RDA, where of the genotroph DNA was used as a tester and Pl DNA as a driver, was also executed. Three difference clones (163-4-2, 123-5-2, and 163-13), from 74 primary clones obtained after three rounds of subtractions with Pl DNA as tester were further characterized. In addition, 2 difference products (213-r1 and 213-r9) were characterized from contrary RDA. The clones 163-4-2 and 163-13 from the csRDA showed polymorphisms between Pl and all the genotrophs when PCR was done with primers derived from sequences of the clones, but only the clone 163-13 polymorphism was confirmed by Southern blot analysis. Four of 5 clones (163-4-2, 123-5-2, 163-13 and 213-r9) that have been characterized appear to be associated with structural changes in the DNA. From the contrary csRDA, it was observed that no clones could be recovered from subtractions between a mixture of genotrophs as a tester and Pl as a driver, and several possible explanations have been proposed.  相似文献

Environmentally induced flax genotrophs L and S show heritable shifts in the relative mobilities of peroxidase, esterase, and acid phosphatase isozymes, plus a number of nonspecific glycoproteins. All L isozymes migrated faster than corresponding S isozymes in 10% acrylamide gels. Various aspects of these shifts are reviewed here; it is proposed that posttranslational modification, probably of the carbohydrate moieties of these glycoproteins, underlies the shifts. In the F1 hybrids between Durrant's L and Durrant's S flax genotrophs, the relative mobilities of the anionic peroxidase isozymes were essentially the same as those in the L parent. The isozymes in both parents and their F1's were compared over a range of acrylamide gel concentrations, with plots of log relative mobility against gel concentration. Plots of comparative mobility, relative to the internal standard hemoglobin, against concentration were also examined. Both approaches provided evidence that apparent molecular weight modifications underlay the shift in mobility between the parents and the resemblance of the F1's to L, the parent which was homozygous for the dominant alleles controlling the mobility shift for at least two of the isozymes.  相似文献

Malate dehydrogenase (MDH) band relative mobility (R m) and activity were examined in leaf extracts of Durrant's flax genotrophs, L and S, and flax genotypes, R and M. MDH activity in leaves from just below the inflorescence was higher in the two smaller, sparsely branched plant types, S and M, than in the larger, more branched plant types, L and R. The MDH electrophoretic banding pattern in flax leaf extracts consisted of three major anionic bands, MDH-1, MDH-2, and MDH-3. NoR m differences were detected between corresponding isozymes of genotypes R and M. For the genotrophs, however, all three bands of S migrated faster than the corresponding bands of L. Codominance was absent in F1 hybrids; SR m was dominant for MDH-2 and MDH-3 and LR m was dominant for MDH-1. The observations suggest that MDHR m in L and S may be controlled by a modifier locus (or loci). Previous studies indicate that a modifier locus may also control heritable genotrophic differences in peroxidase (PER) and acid phosphates (AP)R m. Abstract Flax plants were transferred from 30°C to 15°C temperatures, and then back to 30°C during their vegetative growth phase. Reciprocal transfers were also made from 15°C to 30°C, and then back to 15°C for an identical period at the same time. Plants were also retained in a constant 30°C or 15°C in growth chambers with identical lighting conditions. The reciprocal transfer period lasted 6 d. The relative mobility (Rm) of an anionic leaf peroxidase isozyme, reflecting its molecular weight/conformation and/or net charge, was monitored over a period starting before the reciprocal transfers and finishing after the return to original temperatures. Two of the four flax types which were studied showed isozyme relative mobility responses related either to the different constant temperatures, or to the transfer from 15°C to 30°C. The responses were not reversible on return to the original lower temperature. Thus, exposure to 30°C, even for a transient 6 d period, produced a one way Rm shift. This suggests a developmental switch-over in the particular form of peroxidase glycoprotein being synthesized, rather than an adaptive response triggered by external environmental changes.  相似文献

Summary The association of the H-h (hairy-hairless septa) character with plant weight was studied in the coupling and repulsion phases in F2 of reciprocal crosses between large (L) and small (S) genotrophs of flax variety Stormont Cirrus. F2 plants of reciprocal crosses in coupling (LH x Sh) and in repulsion (Lh x SH) giving H-h segregations were grown with their parents at two sowing times. Significant positive and negative associations between h and plant weight were obtained. A model is proposed based on the hypothesis that the H phenotype had changed to the h phenotype at the time of induction by a heterochromatic region extending over this locus. In the heterozygote, stable equilibria of the homozygotes are destroyed and transfer of heterochromatin, or number of reiterated sequences, or a decrease in one homologue and an increase in the other, occur in this region between homologous chromosomes. The amount and direction of the association is dependent upon the frequency of transfer: 0% transfer gives complete positive association; 50% transfer, no association; 100% transfer, complete negative association. This mechanism or heterochromatic transfer preserves the Mendelian ratio of 31 of Hh in the F2. It is also supposed that there must be other controlling elements present as well.  相似文献

Cultivated flax (Linum usitatissimum L.) is the earliest oil and fiber crop and its early domestication history may involve multiple events of domestication for oil, fiber, capsular indehiscence, and winter hardiness. Genetic studies have demonstrated that winter cultivated flax is closely related to oil and fiber cultivated flax and shows little relatedness to its progenitor, pale flax (L. bienne Mill.), but winter hardiness is one major characteristic of pale flax. Here, we assessed the genetic relationships of 48 Linum samples representing pale flax and four trait-specific groups of cultivated flax (dehiscent, fiber, oil, and winter) through population-based resequencing at 24 genomic regions, and revealed a winter group of cultivated flax that displayed close relatedness to the pale flax samples. Overall, the cultivated flax showed a 27% reduction of nucleotide diversity when compared with the pale flax. Recombination frequently occurred at these sampled genomic regions, but the signal of selection and bottleneck was relatively weak. These findings provide some insight into the impact and processes of flax domestication and are significant for expanding our knowledge about early flax domestication, particularly for winter hardiness.  相似文献

In extracts of flax seedlings 4 days after imbibition, isocitrate lyase activity is unstable in comparison to that in extracts from 2.5-day seedlings or to malate syntheses analysed at several stages of development. This instability in extracts of 4-day seedlings is especially pronounced when a large number of seedlings is homogenized per unit volume of Tris-Mg2+-EDTA-dithioerythritol buffer. However, isocitrate lyase can be stabilized when the resultant homogenate is diluted soon after seedling breakage. The pronounced instability in more concentrated extracts is not due to inadequate buffering by the homogenization medium, nor can it be due to polyphenols because added polyvinylpyrrolidone has no effect. Mixing of a heated supernatant from concentrated extract with dilute unheated extract yields the units of stable isocitrate lyase expected in the dilute extract, ruling out stoichiometric inactivation by a heat-stable component. The pronounced instability is attributed to the action of proteinases. A theoretical model assuming a decay process that is first order in isocitrate lyase and first-order in one or more proteinases is in reasonable agreement with the results. Malate synthase and NADP+-isocitrate dehydrogenase are much more stable in concentrated extracts prepared from 4-day flax seedlings. Isocitrate lyase is stable in concentrated extracts of 5-day watermelon seedlings, which is a developmental stage analogous to that for 4-day flax seedlings.  相似文献

Examination of a method of determining total phenolic content showed that it could be standardized and used on plant extracts from two genotypes of flax. The majority of the phenolic content in these extracts could be removed by adding an anion exchange resin during extraction and dialysing the resulting extracts. Examination of phenolic content and the activities and relative mobilities of peroxidase isozymes in both genotypes revealed effects of phenolic removal on these isozyme characteristics; such effects differed between the genotypes.  相似文献
