[背景] 大量制备高纯度的重组腺相关病毒(Recombinant Adeno-Associated Virus,rAAV),是以腺相关病毒(AAV)为投递载体的基因靶向治疗或基因工程药物研发过程中需要解决的重要问题。[目的] 利用层析柱法大量纯化质粒和rAAV细胞培养液,以获得高纯度的rAAV颗粒。[方法] 对组装rAAV的3种质粒用HiPrepTM10 Sepharose 6FF和HiTrap PlasmidSelect Xtra凝胶层析柱纯化,目的是得到组装AAV的超螺旋质粒DNA,然后对组装rAAV过程中的AAV-293细胞培养方法进行优化,组装成功病毒后,再对收集到的细胞提取液用HiLoadTM10Q和HiLoadTM10SP阴阳离子交换层析柱纯化,最后用纯化浓缩后的rAAV感染HT1080细胞,通过流式细胞仪检测荧光表达细胞数,计算浓缩后活病毒感染滴度。[结果] HiPrep和HiTrap层析柱纯化后得到大量高纯度的超螺旋质粒DNA,组装病毒后,用HiLoad柱纯化获得大量高纯度的rAAV颗粒,通过测定,rAAV基因组滴度达到(2.46±0.37)×1012 TCID50/mL (MOI=1.0×104)。[结论] 通过一系列精细化组装纯化制备出高纯度、高滴度rAAV病毒颗粒。  相似文献

PurposeAn analytical model has been developed for the light propagation in columnar phosphors, based on the optical photon propagation physical and geometrical principles.MethodsThis model accounts for the multiple reflections on the sides of the crystal column, as well as for the infinite forward and backward reflections of the propagated optical photon beams created in the crystal bulk. Additionally it considers the lateral propagated optical photon beams after multiple refractions from the neighbor columns and the optical photon attenuation inside the scintillator.The model was used to predict the Detector Optical Gain (DOG), and the angular distribution, of the columnar CsI:Tl scintillators, used in medical imaging.ResultsThe model was validated against CsI:Tl optical photon transmission published results and good agreement was observed. It was, also, found that the DOG is affected by the length of the columns, as well as the incident X-ray energy spectrum. The results of the angular distribution are in accordance with the theory that the longer crystal columns have more directional light distribution.ConclusionsThe results of DOG are in accordance with the use of short crystal columns for lower energies (mammography) and the use of long crystal columns for higher energies (general radiology). Angular distribution was found more directive for long crystal columns.  相似文献

【背景】草莓连年栽培导致土传病害问题突出,施用熏蒸剂进行土壤消毒因效果显著得以广泛应用。但不同熏蒸剂对土壤病原微生物的影响存在较大差异,同时对非靶标微生物和土壤理化性质也会有不同程度的影响。【目的】明确不同熏蒸剂对草莓连作土壤养分和土壤细菌、真菌群落结构的影响,为合理选择熏蒸剂提供科学依据。【方法】以连作土壤为材料设置5个处理:未熏蒸、石灰氮熏蒸、石灰熏蒸、棉隆熏蒸、威百亩熏蒸,测定熏蒸处理后土壤养分含量;采用PacBio测序平台分析土壤微生物多样性的变化。【结果】石灰氮和威百亩处理均增加了碱解氮含量,降低了有机质、有效磷和速效钾含量;棉隆处理土壤中各养分含量均增加;石灰处理除有机质含量增加外,碱解氮、有效磷和速效钾含量均降低;棉隆、石灰和威百亩处理显著降低pH值。5个处理草莓连作土壤样本中获得了1 164个细菌OTU和444个真菌OTU。细菌多样性和丰富度分析发现,4种熏蒸剂处理均增加了土壤细菌群落的丰富度,石灰氮、石灰和威百亩处理增加了土壤细菌菌落的多样性。4种熏蒸剂处理真菌菌落的丰富度低于对照;石灰、棉隆处理真菌菌落的多样性高于对照和石灰氮、威百亩处理低于对照,但差异不显著。在物种组成分析中,从门水平看,变形菌门(Proteobacteria)和芽单胞菌门(Gemmatimonadetes)为优势细菌门;与对照相比,石灰氮、石灰、棉隆处理变形菌门相对丰度增高,威百亩处理相对丰度降低。4种处理均降低了芽单胞菌门的相对丰度。其他门类中,4种处理均增加了浮霉菌门(Planctomycetota)、疣微菌门(Verrucomicrobia)的相对丰度。优势细菌群落分析表明土壤熏蒸减少了芽单胞菌属(Gemmatimonas)、藤黄单胞菌属(Luteimonas)、中慢生根瘤菌属(Mesorhizobium)等细菌的相对丰度,增加了噬几丁质菌属(Chitinophaga)、苍白杆菌属(Ochrobactrum)的相对丰度。子囊菌门(Ascomycota)为优势真菌,石灰氮、石灰、棉隆、威百亩4种处理均增加了子囊菌门的相对丰度。另外还检测到引起草莓根部土传病害的枝孢属(Cladosporium)和镰刀菌属(Fusarium)病菌,熏蒸处理后均降低了枝孢属和镰刀菌属的相对丰度,其中枝孢属在石灰氮、石灰、棉隆、威百亩处理中分别降低了1.35%、1.11%、0.90%和1.31%,镰刀菌属分别降低了0.71%、0.85%、0.19%和0.65%,但差异不显著。4种土壤熏蒸剂均增加了有益真菌毛壳菌属(Chaetomium)的相对丰度。【结论】采用熏蒸剂处理连作土壤改变了微生物群落构成,减少或灭杀土壤中的大部分致病菌属,起到有效防治草莓土传病害的作用,但不能灭杀所有病菌,而且对有益菌和土壤理化性质有不同程度的影响,因此处理后补充有益微生物非常关键。根据对病原菌的灭杀效果,石灰氮、威百亩处理的效果优于其他处理,可作为轮换施用的熏蒸剂,本试验条件下,棉隆是一种弱的处理剂。  相似文献

Disturbed soil columns, 5.8 cm in diameter and 25 cm in length, were used as a basic model to simulate the movement of carbofuran in rice field soil under saturated conditions. Bioaugmentation using a specific carbofuran degrader, Burkholderia sp. PCL3, in free and immobilized cell forms and biostimulation using rice straw as organic amendment were applied with the aim of enhancing the degradation of carbofuran in soil and to prevent the movement of carbofuran along with the flow through. In the abiotic control and the treatment with only indigenous microorganisms, the mass recovery percentage of carbofuran in the effluent was 52.1 and 22.5%, respectively. The application of bioaugmentation or biostimulation significantly enhanced carbofuran degradation in soil and reduced the movement of carbofuran as indicated by a low mass recovery percentage of carbofuran in the effluent of 14.6–15.5%. A low efficiency of carbofuran removal was obtained from the soil column with bioaugmentation together with biostimulation treatments in which the mass recovery percentage of carbofuran in the effluent was in the range of 22.1–22.6%. Sorption of carbofuran to soil, rice straw and corncob, formation of carbofuran metabolite and colony forming unit (CFU) and pH variation with the time were also investigated during column operation.  相似文献

Abstract

Tribenuron methyl (TBM) is widely used in weed control. Due to its phytotoxicity, concerns on TBM pollution to soil have been raised. In this research, TBM concentration in the soil profile and vetiver grass were measured and simulated using HYDRUS-1D and modified PRZM-3 models. The treatments were two herbicide concentrations to soil with vetiver (C1V and C2V) and without vetiver (C1S and C2S). In control treatment (Co) no herbicide was applied to the soil. In general, according to the measured data, TBM soil residues in C1V and C2V treatments were 39.8% and 30.1% lower than that obtained in C1S and C2S treatments, respectively. The TBM was leached to 90?cm soil depth and it was limited to about 50?cm in the treatments with vetiver grass. The simulated herbicide residue in the soil profile in modified PRZM-3 model was more accurate than the HYDRUS-1D model. The dissipation processes of herbicides in soil and solving method of water movement in soil, considered in the modified PRZM-3 model, are more precise than that obtained in the HYDRUS-1D model. However, the prediction of TBM uptake by vetiver in the HYDRUS-1D model was closer to the measured values than that obtained in the modified PRZM-3 model.  相似文献

PurposeTo evaluate the respiratory motion influence on the tridimensional (3D) dose delivery to breast-shaped phantoms using conformal radiotherapy (3D-RT), Field-in Field (FiF), and IMRT planning techniques.MethodsThis study used breast-shaped phantoms filled with MAGIC-f gel dosimeter to simulate the breast, and an oscillation platform to simulate the respiratory motion. The platform allowed motion in the anterior-posterior direction with oscillation amplitudes of 0.34 cm, 0.88 cm, and 1.22 cm. CT images of the static phantom were used for the 3D-RT, FiF, and IMRT treatment planning. Five phantoms were prepared and irradiated for each planning technique evaluated. Phantom 1 was irradiated static, phantoms 2–4 were irradiated moving with the three different motion amplitudes, and phantom 5 was used as a reference. The 3D dose distributions were obtained by relaxometry of magnetic resonance imaging, and the respiratory motion influence in the doses distribution was accessed by gamma evaluations (3%/3mm/15% threshold) comparing the measurements of the phantoms irradiated under movement with the static ones.ResultsThe mean gamma approvals for three oscillatory amplitudes were 96.44%, 93.23%, and 91.65%; 98.42%, 95.66%, and 94.31%; and 94.49%, 93.51%, and 86.62% respectively for 3D-RT, FiF and IMRT treatments. A gamma results profile per slice along the phantom showed that for FiF and IMRT irradiations, most of the failures occurred in the central region of the phantom.ConclusionsBy increasing the respiratory motion movement, the dose distribution variations for the three planning techniques were more pronounced, being the FiF technique variations the smallest one.  相似文献

PurposeThis paper analyzes Tomotherapy-based intracranial stereotactic radiosurgery (HTSRS) of brain metastasis targeting two end-points: 1) evaluation of dose homogeneity, conformity and gradient scores for single and multiple lesions and 2) assay of dosimetric criticality of completion of HTSRS procedures.Methods42 treatment plans of 33 patients (53 brain lesions) treated with HTSRS were analyzed. Dose to healthy brain, homogeneity, conformity and gradient indexes were evaluated for each lesion. Influence of Field Length and multiple lesions cross-talk effect were assessed. Treatment interruption and completion was investigated using radiochromic films in order to examine the delivered dose and its robustness to patient intrafraction movement.ResultsThe average dose homogeneity index was 1.04 ± 0.02 (SD). Average dose conformity and gradient score indexes were 1.4 ± 0.2 and 50 ± 14 respectively. We found a strong correlation of the dose to healthy brain and conformity and gradient indexes with target(s) volume for which analytical functions were obtained. Field Length and cross-talk effect were significantly correlated with poor gradient scores, but were found not to affect dose conformity.ConclusionsHomogeneity and conformity of HTSRS plans achieved excellent scores, while dose falloff and dose to healthy brain were slightly larger when compared with non-coplanar SRS techniques. Care should be given if treating large (>3 cc) or multiple near in-plane lesions in order to reduce dose to healthy brain. Analysis of interrupted treatments suggests splitting HTSRS treatments in two consecutive fractions in order to prevent target miss and overdosage due to patient intrafraction movement.  相似文献

Background: The extent to which nutrient availability influences plant community composition and dynamics has been a focus of ecological enquiry for decades.

Aims: Results from a long-term nitrogen (N) and phosphorus (P) addition experiment in alpine tundra were used to evaluate the importance of the two nutrients in structuring plant communities in three communities that differed in their snow cover amounts and duration and soil moisture characteristics.

Methods: A factorial N and P experiment was established in three meadows differing in initial vegetation composition and soil moisture. Plant and soil characteristics were measured after 20 years, and the dissimilarity among meadows and treatments were measured using permutational analysis of variance.

Results: Plant species richness declined uniformly across the three meadow types and in response to N and N + P additions, while both evenness and the Shannon diversity index finding indicated that nutrient additions had the highest impact on moister habitats. Overall, N impacts overshadowed changes attributed to P additions, and the N and N + P plots in wet meadow sites were the least diverse and scored the lowest dissimilarity averages among treatments. Dissimilarity estimates indicated that the control and P plots in the dry meadow community were more distinct in composition than all other plots, and especially those in the moist or wet meadows. Above-ground biomass of grasses and sedges (graminoids) increased with N additions while forbs appeared to show responses dictated in part by the graminoid responses. The most abundant grass species of moist and wet meadow, Deschampsia cespitosa, dominated N and N + P plots of the wet sites, but did not show a N response in moist areas in spite of its general abundance in moist meadow. Competition from other plant species in the moist areas likely diminished the D. cespitosa response and contributed to the resilience of the community to nutrient enrichment.

Conclusions: Initial community composition, as influenced by the specific moisture regime, appears to control the extent to which changes in nutrient resources can alter plant community structure. Long-term fertilization tends to support most but not all findings obtained from shorter-termed efforts, and wet meadows exhibit the largest changes in plant species numbers and composition when chronically enriched with N.  相似文献

Sugar cane bagasse was water- or alkali-treated at three liquid/solid (L/S) ratios and its digestibility was measured as microbial protein production of Chaetomium cellulolyticum grown on solid-state fermentation columns. The treatments significantly enhanced fungus growth compared to non-treated bagasse, which was used as a control, although the composition of bagasse did not change greatly. Alkali-treated bagasse reached an average protein content of about 7.6% and the lower the L/S ratio, the higher the protein content. L/S ratio did not have an effect in water-treated bagasse. Protein content of water-treated bagasse was also high, approximately 80% of that one of alkali-treated bagasse. BackgroundMonolithic chromatography using convective interaction media (CIM) disks or columns can be used in the separation step of speciation analysis. When different monolithic disks are placed in one housing, forming conjoint liquid chromatography (CLC) monolithic column, two-dimensional separation is achieved in a single chromatographic run.MethodsHere, we assembled low-pressure (maximum 50 bar) CLC monolithic column, which consists of two 0.34 mL shallow CIM monolithic disks and high-pressure CLC column (maximum 150 bar) from 0.1 mL analytical high performance short bed CIMac monolithic disks. Both the CLC columns constructed from affinity Protein G and weak anion exchange diethylamine (DEAE) disks, were applied for the speciation of cisplatin, oxaliplatin and carboplatin in spiked standard serum proteins, spiked human serum and serum of cancer patients. The analytical performances of the CLC columns used were evaluated by comparing their robustness, selectivity, repeatability and reproducibility. The separated serum proteins were detected on-line by ultraviolet (UV) and eluted Pt species by inductively coupled plasma mass spectrometry (ICP-MS). For accurate quantification of the separated Pt species (unbound Pt-based chemotherapeutic from species associated to transferrin (Tf), human serum albumin (HSA) and Immunoglobulin G (IgG)), post column isotope dilution (ID)-ICP-MS was used.ResultsThe data from analyses showed that both tested CLC monolithic columns gave statistically comparable results, with the low-pressure CLC column exhibiting better resolving power and robustness. It also enables more effective cleaning of monolithic disks and to analyse larger series of serum samples than the high-pressure CLC column. Analyses of serum samples of cancer patients treated with cisplatin or carboplatin showed that Pt-chemotherapeutics were bound preferentially to HSA (around 80%). The portion of unbound Pt in general did not exceed 2%, up to 5% of Pt was associated with Tf and approximately 20% with IgG. Column recoveries, calculated as a ratio between the sum of concentrations of Pt species eluted and concentration of total Pt in serum samples, were close to 100%.ConclusionsLow-pressure CLC column exhibited greater potential than high-pressure CLC column, and can be thus recommended for its intended use in speciation analysis of metal-based biomolecules.  相似文献

Chromatographic separation of glucose and fructose using columns of cation exchange resins in the Ca2+ form was analyzed with a dispersion model neglecting the adsorption process. Pulse response experiments were carried out with analysis by the moment method. Elution profiles of the sugars were successfully predicted by numerical inversion of the concentration calculated in the Laplace domain. The predicted elution profiles could be used for estimation of the column efficiency defined as the recovery of the separated sugar per unit time and per unit length of the column.  相似文献

Thylakoid membranes isolated from peas have been subjected to ionic analyses using the technique of neutron activation. This has allowed the analyses of K+, Na+, Mg2+, Ca2+ and Cl? to be measured simultaneously on the same sample. By varying the ionic composition of the suspending medium it has been shown that these chloroplast membranes have no obvious chemical specificity for the inorganic cations studied and that the major controlling factor is the electrostatic neutralization of the surface negative charges. In agreement with the Gouy-Chapman theory and for the conditions used, divalent cations were preferentially attracted to the membrane surface. This finding, together with the ionic analysis of the unwashed thylakoids and of isolated intact chloroplasts, indicated that the major physiological surface cation is Mg2+ and that K+ is probably the main inorganic cation of the stroma. This conclusion is discussed in terms of counterion movement in response to light induced proton pumping at the thylakoid membrane.  相似文献

Remediation of soil and groundwater contaminated by leaking fuel storage tanks may be assisted by plants, although plant effects on abiotic and biotic removal processes remain unclear. The objectives of this study were to investigate abiotic and biotic removal of JP-8, a kerosene-based jet fuel, in soils with plants, and to determine the effects of plant-induced water movement. Loss of JP-8 in a dry-soil, control column was 25% after 5 months, primarily due to volatilization and gas-phase diffusion. By comparison, managed treatments with simulated surface spills averaged 86% mass reduction at 5 months, indicating an important contribution of biodegradation. Overall JP-8 mass reduction was similar in surface and subsurface-irrigated systems, indicating water content, not mode of water application, influences bioremediation in near-surface systems. The JP-8 concentration reductions in soil columns contaminated above a simulated watertable were 36% after 3 months and 50% after 12 months for vegetated columns compared to 26% and 34% in unplanted columns. Downward movement of JP-8 in unplanted columns was double that in planted columns. Near the groundwater table, JP-8 persists longer than near the soil surface. Plants promote upward movement of water and help draw spilled JP-8 to aerobic near-surface soil.  相似文献

Fifty ejaculates, ten from each of 5 mature stallions, were utilized to study the effects of calcium and fatty acids on equine spermatozoa which were isolated in 3% Bovine Serum Albumin (BSA). The ejaculates were evaluated for percent motile spermatozoa, rate of forward movement, debris, primary abnormalities and secondary abnormalities. The isolation procedure consisted of layering 2 ml of diluted semen (100 x 10(6) spermatozoa/ml) over 6 ml of 3% BSA in 13 x 125 mm columns in a water bath (37 degrees C). After 30 min., the top semen layer and upper half of the BSA layer were withdrawn from all columns and the lower half of the BSA was re-evaluated. A 2 x 2 factorial arrangement of treatments was utilized with either the inclusion or omission of calcium or fatty acids in the BSA isolation media. The percent motile spermatozoa and rate of forward movement were increased (P<.01) when fatty acids were included in the isolation media but decreased (P<.01) when they were omitted. The highest percent motile spermatozoa and rate of forward movement were observed with BSA in the presence of fatty acids and omission of calcium. The calcium by fatty acid interaction, stallion effect and stallion by treatment interaction were significant for percent motile spermatozoa. Less debris was observed in all samples of isolated spermatozoa when compared with the initial estimate. Isolation resulted in a reduction of (P<.01) the primary abnormalities. Also, fewer (P<.01) secondary abnormalities were observed after isolation in all treatments except 4 (-FA+Ca) than were found in the ejaculate sample.  相似文献

Aims

A century of atmospheric deposition of sulfur and nitrogen has acidified soils and undermined the health and recruitment of foundational tree species in the northeastern US. However, effects of acidic deposition on the forest understory plant communities of this region are poorly documented. We investigated how forest understory plant species composition and richness varied across gradients of acidic deposition and soil acidity in the Adirondack Mountains of New York State.

Methods

We surveyed understory vegetation and soils in hardwood forests on 20 small watersheds and built models of community composition and richness as functions of soil chemistry, nitrogen and sulfur deposition, and other environmental variables.

Results

Community composition varied significantly with gradients of acidic deposition, soil acidity, and base cation availability (63% variance explained). Several species increased with soil acidity while others decreased. Understory plant richness decreased significantly with increasing soil acidity (r?=?0.60). The best multivariate regression model to predict richness (p?<?0.001, adjusted-R2?=?0.60) reflected positive effects of pH and carbon-to-nitrogen ratio (C:N).

Conclusions

The relationship we found between understory plant communities and a soil-chemical gradient, suggests that soil acidification can reduce diversity and alter the composition of these communities in northern hardwood forests exposed to acidic deposition.

AimThis study retrospectively analysed the results of using optically stimulated radiation dosimeters (OSLDs) for in vivo dose measurements during total skin electron therapy (TSET, also known as TSEI, TSEB, TSEBT, TSI or TBE) treatments of patients with mycosis fungoides.BackgroundTSET treatments are generally delivered to standing patients, using treatment plans that are devised using manual dose calculations that require verification via in vivo dosimetry. Despite the increasing use of OSLDs for radiation dosimetry, there is minimal published guidance on the use of OSLDs for TSET verification.Materials and methodsThis study retrospectively reviewed in vivo dose measurements made during treatments of nine consecutive TSET patients, treated between 2013 and 2018. Landauer nanoDot OSLDs were used to measure the skin dose at reference locations on each patient, as well as at locations of clinical interest such as the head, hands, feet, axilla and groin.Results1301 OSLD measurements were aggregated and analysed, producing results that were in broad agreement with previous TLD studies, while providing additional information about the variation of dose across concave surfaces and potentially guiding future refinement of treatment setup. In many cases these in vivo measurements were used to identify deviations from the planned dose in reference locations and to identify anatomical regions where additional shielding or boost treatments were required.ConclusionsOSLDs can be used to obtain measurements of TSET dose that can inform monitor unit adjustments and identify regions of under and over dosage, while potentially informing continuous quality improvement in TSET treatment delivery.  相似文献

The geometry and electronic structure of exo-tricyclo[3.2.1.02,4]oct-6-ene (exo-TCO) was investigated using DFT methods. The two faces of the endo-pyramidalised double bond of the molecule are not equivalent. The exo face of the double bond has regions with high electron density (q i,HOMO) and greater negative potential. Molecular complexes of exo-TCO with bromine were investigated using the B3LYP/6-311++G(d,p) method; the exo-TCO . . . Br2(exo) molecular complex was found to be relatively more stable than the exo-TCO . . . Br2(endo) complex. The cationic intermediates of the reaction were studied by DFT and MP2 methods. The solvent effect was evaluated using the self-consistent isodensity polarised continuum model (SCI-PCM). The exo-bromonium cation was found to be more stable than the endo-bromonium cation. Exo-facial selectivity due to electronic and steric factors was observed upon addition of bromine to exo-TCO. The multicentre nonclassical delocalised bromocarbonium cation IV and the exo-bridged-bromonium cation I are more stable than the rearrangement cation V. The reaction products are formed via exo-bridged-bromonium I and nonclassical IV cations, which are the most stable intermediates and whose stabilities barely differ. The mechanism of the addition reaction is also discussed.  相似文献

Abstract

This study evaluated remediation of the herbicide sulfentrazone in soils with three different mineralogies (kaolinite, hematite, and gibbsite) and three remediation sulfentrazone treatments (Canavalia ensiformis L., Crotalaria juncea L., and natural attenuation). This study was conducted in a factorial scheme, in triplicate with randomized block design. Sulfentrazone was applied at 0 and 400?g ha?1. We analyzed sulfentrazone residue in the soils by high-performance liquid chromatography and confirmed the results with bioassays of Pennisetum glaucum. Herbicide movement was greater in the kaolinitic soil without plant species. The retention of herbicide in the kaolinitic soil occurred in larger quantities in the 0–12?cm layer, with higher levels found in the treatments with plants. In the hematitic soil with C. juncea, all applied herbicides were concentrated in the 0–12?cm layer. In the other hematitic soil treatments, sulfentrazone was not detected by chemical analysis at any soil depth, although in many treatments, it was detected in the bioassay. Phytoremediation was more efficient with C. ensiformis grown in gibbsitic soil, reducing the sulfentrazone load by approximately 27%. The effect of various electrolytes on the adsorption of poliovirus was measured in 250-cm-long soil columns with ceramic samplers at different depths. Viruses suspended in deionized water moved much farther through the soil than those suspended in tap water, whereas movement in sewage water was intermediate. The salt content of the tap water and sewage water promoted virus adsorption, but evidently the organic compounds in sewage retarded adsorption. When viruses were suspended in chloride solutions of K+, Na+, Ca+, and Mg2+, virus adsorption increased as the cation concentration and valence increased. The depth of virus penetration was related to the ionic strength of the solutions. Virus penetration data for NO3, SO42−, and H2PO4 salts of K+, Na+, and Ca2+ indicated that other anions were more effective than Cl in promoting virus adsorption. Also, NH4+ was more effective than other cations in limiting the penetration depth of viruses. It seems that ions composed of radicals are more effective than ions composed of single atoms in promoting virus adsorption. Al3+ was the most effective ion in limiting virus penetration, probably owing to flocculation of the viruses. Adding AlCl3 concentrations to secondary sewage effluent to provide an Al3+ concentration of 0.1 mM reduced the virus penetration depth to 40 cm. These studies show that the ionic composition of the suspending solutions must be considered in predicting virus penetration depths, and it may be practical to add low concentrations of a flocculating agent such as AlCl3 to sewage water to limit virus movement through very porous soils.  相似文献

ABSTRACT

Background: Functional trait-based approaches have been used to identify patterns of plant species diversity and composition related to environmental variability/changes. Bryophytes are rapidly affected by perturbations and thus their traits and distribution are expected to show well-defined relationships with environmental variability.

Aim: To quantify the impacts of fragmentation on the diversity and structure of epiphytic bryophytes to vertical and horizontal environmental gradients in an Atlantic Forest remnant.

Methods: Six functional traits related to water relations and light tolerance were recorded and one of them, the leaf lobule area in liverworts, was selected for morphometric measurements along the gradients analysed. Functional diversity and composition metrics of bryophytes along gradients were compared.

Results: Functional diversity changed little along the vertical and horizontal gradients. Conversely, the functional composition of traits changed markedly. Traits related to water storage, such as the presence and area of lobules, and to protection against excess light incidence, such as dark pigments, were more numerous in the canopy and at the fragment edge.

Conclusions: Functional composition is more correlated with the vertical and horizontal gradients than functional diversity. The lobule of liverworts stood out as the most relevant trait to explain the adaptive strategies of bryophytes.  相似文献
