首次报道了中国4种蝙蝠的G-带和C-带核型。大长舌果蝠(Eonycteris spelaea)二倍染色体数目(2n)为36,常染色体臂数(FN)为56;马来假吸血蝠(Megaderma spasma)2n=38,FN=70;黑髯墓蝠(Taphozous melanopogon)2n=42,FN=64;皱唇蝠(Chaerephon plicata)2n=48,FN=54。通过C-带显示,除着丝粒异染色质外,在皱唇蝠的许多染色体臂内和马来假吸血蝠染色体的端粒处也有较多的插入异染色质,大长舌果蝠的基因组中既有臂内异染色质也有端粒异染色质。  相似文献

The fruit bat, Eonycteris spelaea , occurs from India through the Philippines to the southeast limit of its distribution in the Lesser Sunda islands of Indonesia. Mitochondrial DNA (mtDNA) variation was examined in Indonesian E. spelaea island populations by amplification of the D-loop and digestion with restriction endonucleases. In addition, microgeographic variation was assessed by investigation of three cave populations within one island. A total of 24 genotypes, comprising two broad clades, was detected. The pattern of mtDNA variation reflects the colonization history of E. spelaea with estimates of haplotype and sequence diversity highest in the older western populations and lowest at the eastern periphery of the species' distribution. These findings may also be associated with an environmental cline from west to east. There is also evidence that genetic distance between populations reflects geographic relationships, especially historical connectedness, as measured by Pleistocene sea-crossing distances. At the microgeographic level, cave populations were heterogeneous and composed of diverse lineages suggesting restricted local interchange.  相似文献

2019年7月于海南岛儋州市石花水洞网捕到1只蝙蝠,形态特征、头骨特征及线粒体Cyt b基因序列鉴定证实其为长舌果蝠(Eonycteris spelaea),该物种为海南岛蝙蝠分布新记录种。其主要鉴别特征为:中等体型,前臂长69.6 mm;第二指无爪;有一短尾,尾长12.9 mm,明显突出于股膜之后;肛腺发达;头骨较大,头骨和吻部相对较长,颅全长35.2 mm,吻长11.4 mm。本文报道了海南岛长舌果蝠的外部形态、头骨特征以及基于Cyt b基因序列的系统发育树结果,并与该物种已报道的数据进行了比较。  相似文献

Chromosomes of palpigrades (Arachnida: Palpigradi), a rare arachnid order with numerous primitive characters, were studied for the first time. We analysed two species of the genus Eukoenenia, namely E. spelaea and E. mirabilis. Their karyotypes are uniform, consisting of a low number of tiny chromosomes that decrease gradually in size. Study of the palpigrade karyotype did not reveal morphologically differentiated sex chromosomes. Analysis of E. spelaea showed that constitutive heterochromatin is scarce, GC-rich, and restricted mostly to presumed centromeric regions. Meiosis is remarkable for the presence of a short diffuse stage and prominent nucleolar activity. During prophase I, nuclei contain a large nucleolus. Prominent knob at the end of one bivalent formed by constitutive heterochromatin is associated to the nucleolus by an adjacent NOR. Presence of a nucleolus-like body at male prophase II suggests activity of NOR also during beginning of the second meiotic division. The data suggest acrocentric morphology of palpigrade chromosomes. Palpigrades do not display holocentric chromosomes which appear to be apomorphic features of a number of arachnid groups. These are: acariform mites, buthid scorpions, and spiders of the superfamily Dysderoidea. Therefore, cytogenetic data do not support a close relationship of palpigrades and acariform mites as suggested previously.  相似文献

During the course of research on the bacterial feeding behavior and resistance of amoebae to virulent pathogens, we isolated a new strain of amoeba from organic rich soil at the margin of freshwater swamp in the northeastern United States. Light microscopic morphology is characteristically heterolobosean, resembling vahlkampfiids, including a broadened, limax shape, and eruptive locomotion, but occasionally becoming more contracted and less elongated with lateral or anterior bulges and somewhat branching sparse, uroidal filaments. Electron microscopic evidence, including mitochondria with flattened cristae surrounded by rough endoplasmic reticulum, further indicates a heterolobosean affinity. The solitary nucleus contains a centrally located nucleolus. Cysts are rounded with occasionally an eccentrically located nucleus. The cyst walls are relatively thin, becoming crenated, and loosely enclosing the cyst when mature. Molecular genetic evidence places this isolate among the Heterolobosea, branching most closely in a clade including Allovahlkampfia spelaea and previously isolated, un-named strains of soil amoebae. Based on differentiated features, including morphology of the uroid, cyst wall structure, and molecular genetic evidence that distinguish it from A. spelaea, a new genus and species, Solumitrus palustris, is proposed for this new heterolobosean.  相似文献

During a two year population ecology study in a cave environment, 15 Eurycea (= Typhlotriton) spelaea were observed ingesting bat guano. Furthermore, E. spelaea capture numbers increased significantly during the time that grey bats (Myotis grisescens) deposited fresh guano. We investigated the hypothesis that this behaviour was not incidental to the capture of invertebrate prey, but a diet switch to an energy-rich detritus in an oligotrophic environment. Stable isotope assays determined that guano may be assimilated into salamander muscle tissue, and nutritional analyses revealed that guano is a comparable food source to potential invertebrate prey items. This is the first report of coprophagy in a salamander and in any amphibian for reasons other than intestinal inoculation. Because many temperate subterranean environments are often energy poor and this limitation is thought to select for increased diet breadth, we predict that coprophagy may be common in subterranean vertebrates where it is not currently recognized.  相似文献

On the archaeological site of Menez-Dregan in Brittany, France, dated 300,000-500,000 years-old, paleoparasitological analysis of cave deposits led to the detection of well-preserved helminth eggs, which morphology and morphometry pointed to the diagnosis of Toxocara canis eggs, a parasite of carnivore mammals. Paleolithic remains suggested a parasitism of the hyena Crocuta spelaea or other canids that inhabited the region.  相似文献

舌头通常具有味觉、消化和免疫等重要功能。由于不同的食物含有不同的营养成分,我们推测舌头中和食性相关的基因在不同食性的动物中可能具有表达差异。为了检验此假说,本研究选取三种蝙蝠(大长舌果蝠(Eonycteris spelaea)、中华菊头蝠(Rhinolophus sinicus)和大足鼠耳蝠(Myotis ricketii))为研究对象,利用RNA高通量测序技术获得了三种蝙蝠舌头的转录组数据。比较转录组分析显示,在三种蝙蝠舌头中,表达量普遍最高的基因主要具有免疫功能,说明这些基因对于维持舌头正常的功能起到不可或缺的作用;另一方面,相对于食虫的中华菊头蝠和大足鼠耳蝠,与糖类代谢相关的基因在食蜜的大长舌果蝠中显著高表达,提示这些基因在这三种蝙蝠的舌头中具有表达差异。我们的研究不仅丰富了蝙蝠的转录组资源,还证实了舌头中参与糖类代谢的基因表达差异可能和蝙蝠食性分化紧密相关。  相似文献

We developed 11 novel microsatellite loci for Sprague's pipit (Anthus spragueii), a songbird endemic to the Northern Great Plains of central North America. These loci were screened in 21 individuals from southern Saskatchewan, Canada. All 11 loci were polymorphic, with the number of alleles per locus ranging from five to 23, polymorphic information content from 0.312 to 0.947, and observed heterozygosity from 0.476 to 0.900. These loci will be suitable for a wide variety of studies on pipit natural history, ecology, and conservation.  相似文献

Six new microsatellite DNA loci are isolated from a genomic library of the sphenodontid reptile tuatara (Sphenodon) and presented here as a tool for identifying individuals for future paternity and kinship studies. These loci, in combination with four previously published loci, are sufficient to discriminate between clutch-mate siblings from Stephens Island and Brothers Island populations. These populations represent high and low levels of genetic diversity in tuatara populations respectively. An estimate of minimum number of fathers of each clutch found no evidence for multiple paternity in any clutch. These newly isolated loci complete the development of an array of genetic tools for use in tuatara to enhance ongoing conservation and management of wild, translocated and captive populations.  相似文献

We identified microsatellite sequences of potential utility in the house sparrow (Passer domesticus) and assigned their predicted genome locations. These sequences included newly isolated house sparrow loci, which we fully characterized. Many of the newly isolated loci were polymorphic in two other species of Passeridae: Berthelot's pipit Anthus berthelotii and zebra finch Taeniopygia guttata. In total, we identified 179 microsatellite markers that were either isolated directly from, or are of known utility in, the house sparrow. Sixty-seven of these markers were designed from unique sequences that we isolated from a house sparrow genomic library. These new markers were combined with 36 house sparrow markers isolated by other studies and 76 markers isolated from other passerine species but known to be polymorphic in the house sparrow. We utilized sequence homology to assign chromosomal locations for these loci in the assembled zebra finch genome. One hundred and thirty-four loci were assigned to 25 different autosomes and eight loci to the Z chromosome. Examination of the genotypes of known-sex house sparrows for 37 of the new loci revealed a W-linked locus and an additional Z-linked locus. Locus Pdoμ2, previously reported as autosomal, was found to be Z-linked. These loci enable the creation of powerful and cost-effective house sparrow multiplex primer sets for population and parentage studies. They can be used to create a house sparrow linkage map and will aid the identification of quantitative trait loci in passerine species.  相似文献

Primers have been developed for nine polymorphic tetranucleotide microsatellite loci isolated from the swift fox (Vulpes velox). These can be amplified in three multiplex reactions and all loci can be scored easily and without ambiguity. The number of alleles per locus ranged from three to 14 and the overall heterozygosities ranged from 0.54 to 1.00. All loci were in Hardy‐Weinberg equilibrium, but two loci were in linkage disequilibrium. These loci will be used to monitor endangered swift fox populations on the Canadian prairies.  相似文献

This study reports the isolation and characterization of seven highly polymorphic microsatellite loci in Silene vulgaris (Caryophyllaceae). The loci were isolated from two libraries constructed from genomic DNA enriched for CA and GA repeats. These markers yielded nine to 40 alleles per locus (mean 22.1) in a survey of 45 individuals from a single population located in the western Swiss Alps. Average observed heterozygosity ranged from 16.2 to 77.4%. These microsatellite loci should be valuable tools for studying fine‐scale genetic structure.  相似文献

We report the isolation of microsatellite loci from three species in the genus Conradina (Lamiaceae). To ensure their utility for multiple species, loci were screened for amplification and variability in all six Conradina species; 11 loci demonstrated high levels of amplification and polymorphism in most species. These 11 loci were characterized in 20 individuals from one population of Conradina brevifolia; alleles per locus ranged from five to 15, and observed heterozygosity ranged from 0.30 to 0.90. These microsatellites will be used to clarify species limits, detect interspecific hybridization, and understand the partitioning of genetic variation in each species of Conradina.  相似文献

Twenty-two nuclear microsatellite loci were isolated from a genomic DNA library derived from Madagascar’s Rousettus madagascariensis. Marker characteristics were determined from a single population (37 individuals) from Fort Dauphin (southeastern Madagascar). Sixteen of the 22 loci were within Hardy–Weinberg expectations. These loci are highly informative with polymorphic information content values ranging between 0.757 and 0.916. These loci will provide valuable information for the study of population genetics and gene flow within this species of bats. Due to the dramatic reduction and alteration of their habitat, data generated utilizing this marker suite will potentially provide additional information for the effective long-term management of this near-threatened bat species.  相似文献

Morphological and genetic analyses of Eonycteris spelaea from 15 islands along the Banda Arc, from Sumatra to Timor and including Kalimantan and Sulawesi, revealed considerable divergence between islands and geographical patterning. On the basis of both morphology and genetics, the populations on the large islands of Greater Sunda (Sumatra, Java, Kalimantan and Sulawesi) are generally distinct from one another and from those on the islands in Nusa Tenggara (Lombok to Timor), which form a more cohesive cluster. These differences may be the result of the Nusa Tenggara populations having been colonized more recently than those on the Greater Sunda, and probably from a single source. All biological measures of the relationships between island populations are positively associated with the extent of the sea-crossing between them, indicating the sea is an important barrier to movement. Multivariate analyses show the presence of a marked trend for body size to increase from west to east. However, individuals from Kalimantan are not consistent with this trend, being smaller than predicted, and on the two outer Banda Arc islands of Sumba and Timor animals are a little larger than predicted from the longitudinal trend. These differences could be due to the relative isolation of these populations or differing environmental conditions. There is also a negative relationship between body size and island area, but this is confounded by the longitudinal trend. We describe the cloning and characterization of nine microsatellite loci from the northern leopard frog. Seven loci consist of tetranucleotide repeats, one locus consists of a dinucleotide repeat and one locus consists of a GT repeat juxtaposed with a GATA repeat. In a sample of 36 frogs from a natural population, polymorphism at these loci ranged from two to 13 alleles per locus with expected heterozygosities ranging from 0.5 to 0.91. These loci will be useful to researchers since this species is used for a broad range of studies.  相似文献

Sixteen dinucleotide microsatellite loci were isolated from the genome of Propithecus verreauxi verreauxi. All loci were polymorphic when genotyped on a minimum of 16 animals. The number of alleles across these loci ranges from two to 11. Additionally, seven of these loci were genotyped across a minimum of 200 animals in order to estimate heterozygosity and their potential for parentage assignment in this population. Using these seven loci, the mean heterozygosity in this population is 0.705, and the combined probability of these seven loci to exclude a random individual from parentage, when one parent is known, is 0.996. These data suggest that these loci will be useful for estimating a variety of population genetic and genealogical parameters in P. v. verreauxi populations.  相似文献

Primers for 16 microsatellite loci were developed for the trumpeter swan (Cygnus buccinator), a species recovering from a recent population bottleneck. In a screen of 158 individuals, the 16 loci were found to have levels of variability ranging from two to seven alleles. No loci were found to be linked, although two loci repeatedly revealed significant departures from Hardy–Weinberg equilibrium. Amplification in the closely related tundra swan (Cygnus columbianus) was successful for all except one locus. These microsatellite loci will be applicable for population genetic analyses and ultimately aid in management efforts.  相似文献
