Elaeocarpus grandis (Blue Quandong) is a common rainforest tree with a widespread distribution across northeastern Australia, while the related species E. sp. Rocky Creek and E. williamsianus are more restricted in their distribution. Highly polymorphic markers are required to provide genetic information on these species, which will aid in developing effective conservation and management strategies. Eight microsatellite loci were isolated and characterized in E. grandis, and tested in the rare species and other species of the Elaeocarpaceae. Cross‐species transferability was most successful within Elaeocarpus, with six and eight loci transferring to E. sp. Rocky Creek and E. williamsianus, respectively.  相似文献

We identified 16 polymorphic nuclear microsatellite loci from an AG-enriched library of Ceratopetalum apetalum. These loci were tested across two populations of C. apetalum, a single population of C. gummiferum and representatives of four other congeneric taxa. Fourteen loci were identified as polymorphic in at least one other species. These microsatellites will be used to look at population dynamics of species with varying distributions and presumed low dispersal capabilities within the genus Ceratopetalum.  相似文献

Wilsonia backhousei is a clonal saltmarsh plant restricted to the southern latitudes of Australasia and threatened in New South Wales. We have identified eight informative microsatellite loci in the species from (AG)n‐ and (AC)n‐enriched libraries. In 48 samples from two populations we detected an average of five alleles per locus (range 2–8, average HE = 0.45), of which 72% were unique to one population or the other. Six of the eight loci were also amplifiable in Wilsonia rotundifolia under the same reaction conditions. The markers will be excellent tools for use in the management and conservation of both species.  相似文献

Ten novel polymorphic microsatellites were isolated from the dace complex (Leuciscus leuciscus), which is a European cyprinid species. Four multiplex polymerase chain reaction sets were optimized in order to genotype 26 polymorphic loci in all, including 16 previously published cyprinid-specific loci. The level of genetic diversity was assessed in 142 dace individuals. We also successfully applied 26 of the microsatellites to 10 related species. These primers thus will be useful to assess population structure of the dace and other cyprinid species, with application for conservation issues and phylogeographical approaches.  相似文献

Wuchang bream (Megalobrama amblycephala) is an economically important fish in China. From a (GT)13‐enriched genomic library, 20 microsatellites were developed. Nine of these 20 loci were polymorphic in a test population with allele numbers ranging from two to four, and the observed and expected heterozygosities ranging from 0.2609 to 0.7826 and from 0.3739 to 0.7546, respectively. In the cross‐species amplifications, six of these nine loci were also polymorphic in white amur bream (Parabramis pekinensis). These polymorphic microsatellite loci are potentially useful for population genetics of Wuchang bream and its closely related species.  相似文献

Ten novel polymorphic microsatellites (seven with perfect motifs) were isolated from vairone species (Telestes souffia and Telestes muticellus), which are endangered European cyprinid species. Together with 11 previously published cyprinid-specific loci, five multiplex sets were optimized, allowing the genotyping of 21 polymorphic loci. The level of genetic diversity was assessed in 97 individuals from the two species T. souffia and T. muticellus. We also successfully applied the 21 microsatellites to nine related species. These primers will thus be useful in assessing population structure of the vairone and other cyprinid species, with application for conservation issues and phylogeographical approaches.  相似文献

We developed eight polymorphic nuclear microsatellite markers for the Swiss stone pine (Pinus cembra L.), of which seven may be amplified in a multiplex polymerase chain reaction. Allelic polymorphism across all loci and 40 individuals representing two populations in the Swiss Alps was high (mean = 7.6 alleles). No significant linkage disequlibrium was displayed between pairs of loci. Significant deviation from Hardy–Weinberg equilibrium was revealed at three loci in one population. Cross–amplification was achieved in two related species within the genus (P. sibirica and P. pumila). Thus, the markers may be useful for population genetic studies in these three pine species. They will be applied in ongoing projects on genetic diversity and patterns of gene flow in P. cembra.  相似文献

We describe the isolation and characterization of 12 highly polymorphic microsatellite loci for the muskrat, Ondatra zibethicus. Microsatellite markers from three other rodent species were cross-amplified in muskrat and one of them was polymorphic. We observed moderate to high levels of genetic variability in these 13 polymorphic loci (five to 22 alleles per locus) with observed heterozygosity ranging from 0.48 to 0.96. These markers will be useful for further studies on population genetic structure in muskrat and potentially in other rodent species.  相似文献

In this study, we developed five microsatellite loci from an enriched genomic library constructed for the pygmy treeshrew (Tupaia minor), and adapted another two from a previous study on the common treeshrew (Tupaia glis), for use in studying mating and dispersal patterns in Bornean treeshrews. We screened 32 plain treeshrew (Tupaia longipes) and 54 large treeshrew (Tupaia tana) individuals at these loci. Polymorphism ranged from two to 13 alleles, and heterozygosity ranged from 0.29 to 0.88. These results indicate the general utility of these microsatellites for genetic analyses in other Tupaia spp.  相似文献

Ten microsatellite loci were developed and described for Pimelodus microstoma, identified in eggs collected from the lower Das Cinzas river. Nine loci were polymorphic, with 02–19 alleles per locus. The observed heterozygosity values were up to 0.898. Five species of the Pimelodidae family were also tested for amplification of loci, among which Pimelodus maculatus was the most successful in the transferability of primers. These loci have proven to be useful for studying the variability and structuring of populations of Pimelodus microstoma.  相似文献

One-hundred and two unique loci were isolated from a microsatellite-enriched black-billed magpie Pica pica genomic library. Sixteen of these new loci, along with 148 existing microsatellite passerine loci, were tested for polymorphism in four unrelated black-billed magpies. We identified a total of 43 unique polymorphic loci (10 and 33 respectively) that we characterized in 24 unrelated individuals from a population in Guadix, Spain. The putative chromosomal locations of loci polymorphic in black-billed magpie were assigned based on the location of their orthologues in the assembled zebra finch and chicken genomes.  相似文献

Crossosoma californicum (Crossosomataceae) is a rare shrub species endemic to the California Channel Islands. Previous studies based on allozymes revealed little genetic variability in this plant species. We have isolated 13 polymorphic microsatellite loci from C. californicum. These loci show intermediate levels of variability, averaging 4.2 alleles per locus and expected heterozygosity of 0.376. Two loci did not fit Hardy–Weinberg expectations with significant deficits of heterozygous genotypes consistent with the presence of null alleles or population subdivision.  相似文献

Nine yellowcheek (Elopichthys bambusa) microsatellite loci were isolated using the fast isolation by AFLP of sequences containing repeats (FIASCO) protocol. Three to eight alleles per locus were detected in 29 samples collected from five populations of E. bambusa. The mean number of alleles was 5.6 ± 1.9 and the level of observed heterozygosities ranged from 0.415 to 0.843. These are the first microsatellite loci characterized from E. bambusa that can be used for estimating genetic diversity, population structure and parentage analysis.  相似文献

Here we describe 32 di‐, tri‐ and tetranucleotide microsatellite loci isolated by PIMA, a polymerase chain reaction (PCR)‐based procedure, for the common snook (Centropomus undecimalis). Five loci were monomorphic, and the remaining loci averaged 6.7 alleles per locus in a sample of 45 common snook. For polymorphic loci, expected heterozygosities ranged from 0.02 to 0.91 (mean = 0.538). Significant departures from Hardy–Weinberg equilibrium expectations occurred in two loci. Exact tests for genotype disequilibrium gave evidence for linkage at one pair of loci. Many cross‐species primer assays yielded PCR fragments of the expected size for 11 species of Centropomus and two species of the confamilial genus Lates.  相似文献

Eleven polymorphic microsatellite loci have been isolated and characterized from random amplified polymorphic DNA product in half-smooth tongue sole, Cynoglossus semilaevis. Twenty-one microsatellites were selected for designing microsatellite primers, of which 11 gave working primer pairs. They had between three and 12 alleles. Observed and expected heterozygosities varied from 0.53 to 0.93, and from 0.52 to 0.80, respectively. Five additional fish species assessed for cross-species amplification revealed between one and three positive amplifications and between zero and three polymorphic loci per species.  相似文献

In the present study, we report 12 polymorphic microsatellite loci developed from a cDNA library from the turbot, Scophthalmus maximus. Observed and expected heterozygosities varied from 0.20 to 1.00 and from 0.18 to 0.78, respectively. No significant linkage disequilibrium between pairs of loci was found, but two loci significantly deviated from Hardy–Weinberg equilibrium after Bonferroni correction. Cross‐species amplifications of these microsatellites in five additional fish species revealed between five and 11 positive amplifications and between zero and four polymorphic loci per species.  相似文献

We have developed six novel, polymorphic microsatellite markers for the scrub hare, Lepus saxatilis. These markers have been tested for 150 scrub hares and 67 Cape hares from different localities across southern Africa. All six loci amplified consistently well and most were highly polymorphic. Four loci amplified consistently in five different leporid genera. The microsatellites presented here will be useful for phylogeographical studies in the scrub hare and other leporidae.  相似文献

Here, we report the isolation and characterization of 11 polymorphic microsatellites in Limodorum abortivum. Allele variability has been characterized in three populations from Southern Italy and France. The number of alleles ranged from one to six per locus with an average of 3.8 alleles per locus. Observed and expected heterozygosity values ranged from 0.000 to 1.000 and from 0.492 to 0.806, respectively, with striking differences among populations. These microsatellites should be valuable tools for studying fine-scale genetic structure of scattered Limodorum abortivum populations, patterns of relationship with closely related taxa and the evolutionary ecology of its mycorrhizal interactions.  相似文献

Nineteen microsatellite markers were developed from Tetratheca paynterae ssp. paynterae, a rare and endangered, leafless, perennial shrub. Twelve loci were polymorphic in T. paynterae ssp. paynterae with two to 14 alleles per locus and mean expected heterozygosity of 0.62. Primer pairs were tested on four other Tetratheca species from ironstone ranges in southern Western Australia. Ten loci were polymorphic in T. paynterae ssp. cremnobata and T. aphylla ssp. aphylla, three in T. harperi and four in T. erubescens. The level of polymorphism was adequate for studies of genetic structure and mating systems in three of the five taxa.  相似文献
