The longer the seeds of dwarf French bean (Phaseolus vulgaris)were soaked in water before sowing, the smaller were the seedlingsthey produced. Soaking for 24 hours removed only little drymatter, but detectable amounts of a gibberellin and a betaine.The amounts of these two removed increased with increased timeof soaking. Long soaking of seeds decreased the amounts of gibberelinand auxin in the cotyledons, and of gibberellin, auxin, andbetaine in the primary leaves of seedlings. Aerating, cooling,or increasing the volume of water in which the seeds were soakedalso retarded the growth of the seedlings, but did not affectthe amount of growth substance removed from the seeds or containedin the cotyledons. The deleterious effect of soaking seeds wasnot alleviated by treating the seeds or seedlings with gibberellicacid or glycine betaine. Although larger seedlings were producedby heavy than by light seeds, their cotyledons and primary leavescontained similar concentrations of growth substances.  相似文献

Soybean seedlings (Glycine max L.) were germinated and dark-grown in water-saturated vermiculite (water potential = −0.01 megapascal) for 48 hours, then transferred either to water-saturated vermiculite or to low water potential vermiculite (water potential = −0.30 megapascal). A decrease in growth rate was detectable within 0.8 hour post-transfer to low water potential vermiculite. A fourfold increase in the abscisic acid content of the elongating region was observed within 0.5 hour. At 24 hours post-transfer, hypocotyl elongation was severely arrested and abscisic acid reached its highest measured level: 3.7 nanograms per milligram dry weight (74-fold increase). A comparison of the polyA+ RNA populations isolated at 24 hours post-transfer from the elongating region of water-saturated and low water potential vermiculite-grown seedlings was made by two-dimensional (isoelectric focusing-sodium dodecyl sulfate) polyacrylamide gel analysis of in vitro translation products. It revealed both increases and decreases in the relative amounts of a number of translation products. Rewatering seedlings grown in low water potential vermiculite at 24 hours post-transfer led to a total recovery in growth rate within 0.5 hour, while abscisic acid in the elongating hypocotyl region required 1 to 2 hours to return to uninduced levels. Application of 1.0 millimolar (±) abscisic acid to well-watered seedlings resulted in a 48% reduction in hypocotyl growth rate during the first 2 hours after treatment. Plants treated with abscisic acid for 24 hours had a lower polysome content than control plants. However, hypocotyl growth inhibition in abscisic acid-treated seedlings preceded the decline in polysome content.  相似文献

Plant water deficits reduced the basipetal transport of auxin in cotyledonary petiole sections taken from cotton (Gossypium hirsutum L.) seedings. A pulse-labeling technique was employed to eliminate complications of uptake or exit of 14C-indoleacetic acid from the tissue. The transport capacity or the relative amount of radioactivity in a 30-minute pulse which was basipetally translocated was approximately 30% per hour in petioles excised from well watered seedlings (plant water potentials of approximately -4 to -8 bars). No cotyledonary leaf abscission took place in well watered seedlings. Plant water potentials from -8 to -12 bars reduced the transport capacity from 30 to 15% per hour, and although the leaves were wilted, cotyledonary abscission did not increase appreciably at these levels of stress. The threshold water potential sufficient to induce leaf abscission was approximately -13 bars and abscission increased with increasing stress while the auxin transport capacity of the petioles remained relatively constant (15% per hour). The basipetal transport capacity of well watered petioles tested under anaerobic conditions and acropetal transport tested under all conditions were typically less than basipetal transport under the most severe stress conditions. Cotyledonary abscission took place during and 24 hours after relief of stress with little or no abscission taking place 48 hours after relief of stress. Although the water potential returned to -4 bars within hours after rewatering the stressed plants, partial recovery of the basipetal transport capacity of the petioles was not apparent until 48 hours after rewatering, and at least 72 hours was required to return the transport capacity to near normal values. These data support the view that decreased levels of auxin reaching the abscission zone from the leaf blade influence the abscission process and further suggest that the length of time that the auxin supply is maximally reduced is more critical than the degree of reduction.  相似文献

Restoration requires techniques similar to those used in agriculture to improve germination and seedling vigor. We treated 6‐year‐old (collected in 2003, S‐2003) and 1‐month‐old (S‐2009) seeds of Dodonaea viscosa with hydropriming (HP). Seeds were made permeable with hot water prior to hydration for 24 or 48 hours (HP‐24 and HP‐48, respectively) followed by dehydration. The resulting seedlings exposed to both HP treatments were sown in a lava field in soil mixed with hydrogel (HG) under the shade projected by five vegetation patches. The effects of these treatments on germination, seedling field survival, and growth were assessed. HP‐24 in S‐2009 and HP‐48 in S‐2003 increased the germination percentage from 22.5 and 31.7% in control seeds (permeable seeds) to 63.3 and 98.3%, respectively. The seedlings‐2009 (from S‐2009) with HG maintained high survival in all vegetation patches. Seedlings‐2003, however, had low survival. The lack of HG was negatively related to the photon flux in each patch. Survival of seedlings‐2009 increased with HG of up to 398.41 µmol m?2 s?1; after which survival decreased. During the rainy season, HP enhanced seedling growth, except the basal diameters and number of leaves in the seedlings‐2003 with HP‐24. During the dry season, the effects of HG and HP were similar for all the seedlings. In the following rainy season, the priming effect was lost while HG continued to promote seedling growth. The combined use of HP and HG and the shade projected by the patches resulted in a successful vegetation recovery strategy.  相似文献

The heavy metal-resistant bacteria from rhizospheric soils of wild Populus euphratica forest growing in arid and saline area of northwestern China were investigated by cultivation-dependent methods. After screening on medium sparked with zinc, copper, nickel and lead, 146 bacteria strains with different morphology were isolated and most of them were found to be resistant to at least two kinds of heavy metals. Significant increase in fresh weight and leaf surface area of Arabidopsis thaliana seedlings under metal stress were noticed after inoculated with strains especially those having multiple-resistance to heavy metals such as Phyllobacterium sp. strain C65. Investigation on relationship between auxin production and exogenous zinc concentration revealed that Phyllobacterium sp. strain C65 produced auxin, and production decreased as the concentration of zinc in medium increased. For wheat seedlings treated with zinc of 2 mM, zinc contents in roots of inoculated plants decreased by 27% (P < 0.05) compared to the uninoculated control. Meanwhile, zinc accumulation in the above-ground tissues increased by 22% (P < 0.05). The translocation of zinc from root to above-ground tissues induced by Phyllobacterium sp. strain C65 helped host plants extract zinc from contaminated environments more efficiently thus alleviated the growth inhibition caused by heavy metals.  相似文献

The correlation between auxin and RNA metabolism was investigated in lentil roots. IAA and NAA both cause a considerable rise in the RNA level of germinating lentil roots, though no effect of IAA was found on the DNA level. In untreated germinating roots various sections were isolated and a direct relation found between RNA and auxin content, and an indirect relation between RNA content and auxin oxidase activity. In excised roots, incubated for 24 hours, the loss of RNA is paralleled by a loss of endogenous auxin. Excised roots treated with 10?4M IAA or M 10?4 NAA loose little RNA. The findings suggest that in lentil roots the RNA levels may be controlled by auxin levels, which in turn may be controlled by the levels of auxin oxidase.  相似文献

Tissue cultures from both juvenile and adult stems of English ivy, Hedera helix L., were established in White's medium supplemented by coconut water and auxin (usually naphthalene acetic acid). With repeated transfers, cultures were habituated in less than a year to grow well without coconut water by using an auxin and kinetin. Cultures from juvenile seedlings were less demanding in requirements for growth. In all types of cultures occasionally small areas of rapidly growing cells were noticed. These when isolated gave rise to rapidly growing cultures with many cells of unusual appearance. Abnormally long cells and chain-type cells were abundant. When 0.1 mg/ liter of kinetin was added to the medium, these cells grew well without auxin and coconut water.  相似文献

Cucurbit seedlings potentially develop a peg on each side of the transition zone between the hypocotyl and root. Seedlings grown in a horizontal position suppress the development of the peg on the upper side of the transition zone in response to gravity. It is suggested that this suppression occurs due to a reduction in auxin levels to below the threshold value. We show in this study that the free indole-3-acetic acid (IAA) content is low, while IAA conjugates are significantly more abundant in the upper side of the transition zone of gravistimulated seedlings, compared to the lower side. A transient increase in mRNA of the auxin-inducible gene, CS-IAA1, was observed in the excised transition zone. The result suggests that the transition zone is a source of auxin. Cucumber seedlings treated with auxin-transport inhibitors exhibited agravitropic growth and developed a peg on each side of the transition zone. Auxin-transport inhibitors additionally caused an increase in CS-IAA1 mRNA accumulation at the transition zone, indicating a rise in intracellular auxin concentrations due to a block of auxin efflux. To study the involvement of the auxin transport system in peg formation, we isolated the cDNAs of a putative auxin influx carrier, CS-AUX1, and putative efflux carrier, CS-PIN1, from cucumber (Cucumis sativus L.) plants. Both genes (CS-AUX1 in particular) were auxin-inducible. Accumulation of CS-AUX1 and CS-PIN1 mRNAs was observed in vascular tissue, cortex and epidermis of the transition zone. A reduced level of CS-AUX1 mRNA was observed in the upper side of the gravistimulated transition zone, compared with the lower side. Summary Dark-grown, 3-day-old soybean seedlings were sprayed with 1 mM 2,4-dichlorophenoxyacetic acid 24 hours before harvest. Mitochondria from 2,4-D-treated lower hypocotyls were found to be larger and showed greater incorporation in vivo, of amino acids into protein and phosphate into phospholipids and RNA, than mitochondria from untreated tissue. Mitochondria isolated from 2,4-D-treated hypocotyls showed an enhanced energy-dependent incorporation of amino acids into protein, although the incorporation of nucleoside triphosphates into the RNA of isolated mitochondria was not affected. No effect of 2,4-D, applied in vitro, was noted, and no enhancement of mitochondrial respiratory efficiency followed auxin treatment. A method of isolating mitochondria with a very low level of bacterial contamination is reported.  相似文献

Summary Water extracts of foliage ofAster macrophyllus, Lonicera tatarica, Solanum dulcamara, Solidago gigantea, Prunus serotina, andRubus idaeus var.strigosus, collected from the ground vegetation of a red pine (Pinus resinosa Ait.) plantation, were tested for potential allelopathic effects on seed germination, radicle elongation, and growth of red pine seedlings. Seed germination was not significantly affected by extracts, but radicle elongation of newly germinated seeds was reduced by up to 48% compared to controls. All extracts variously inhibited height growth, formation of secondary needles, and dry weight increment of young red pine seedlings during a seven-week experiment. Total dry weight of seedlings treated with Lonicera extract was only 46% that of control plants. Phosphorus concentration in needles treated with Lonicera or Solidago extracts was significantly lower than that of controls. Growth inhibition was not mediated by pH effects or osmotic potentials of the extracts. The data suggest that allelopathic inhibition of red pine is possible, but additional field experiments are needed to evaluate the significance of allelopathy in red pine plantations.Research conducted by the College of Agricultural and Life Sciences. Water extracts of fresh leaves, buds, and leaf litter of Populus balsamifera (balsam poplar) were tested at different dilutions for allelopathic effects on nodulation, nitrogenase activity and growth of nodulated green alder (Alnus crispa) seedlings, and on growth of unnodulated green alder seedlings. All extracts inhibited height growth, root elongation and dry weight increment of nodulated and unnodulated green alder seedlings to some degree during a 2-month experiment. Foliar nitrogen content of both nodulated and unnodulated seedlings was significantly lower in extract treated plants than in controls. Growth inhibition was about 25% less in nodulated than in unnodulated seedlings. The number of nodules per plant in seedlings treated with any balsam poplar extract was only 51% that of control plants. Acetylene reduction by seedlings treated with bud and leaf litter extracts indicated a decrease of 62% compared to controls. Growth inhibition was not mediated by pH or osmotic effects of the extracts. The possible ecological significance is discussed.  相似文献

The effect of the defoliant thidiazuron (TDZ) on basipetal auxin transport in petiole segments isolated from cotton (Gossypium hirsutum L. cv LG102) seedlings was examined using the donor/receiver agar block technique. Treatment of intact seedlings with TDZ at concentrations of 1 micromolar or greater resulted in a dose-dependent inhibition of 14C-IAA transport in petiole segments isolated 1 or 2 days after treatment. Using 100 micromolar TDZ, the inhibition was detectable 19 hours after treatment and was complete by 27 hours. Both leaves and petiole segments exhibited a marked increase in ethylene production following treatment with TDZ at concentrations of 0.1 micromolar or greater. The involvement of ethylene in this TDZ response was evaluated by examining the effects of two inhibitors of ethylene action: silver thiosulfate, 2,5-norbornadiene. One day after treatment, both inhibitors effectively antagonized the TDZ-induced inhibition of auxin transport. Two days after TDZ treatment both inhibitors were ineffective. The decrease in IAA transport in TDZ treated tissues was associated with increased metabolism of IAA. The transport of 14C-2,4-dichlorophenoxyacetic acid was also inhibited by TDZ treatment. This inhibition was not accompanied by increased metabolism. Incorporation of TDZ into the receiver blocks had no effect on auxin transport. The ability of the phytotropin N-1-naphthylphthalamic acid to stimulate IAA uptake from a bathing medium was reduced in TDZ-treated tissues. This reduction is thought to reflect a decline in the auxin efflux system following TDZ treatment.  相似文献

The effects of the auxin-inhibiting substances raphanusanin ((3R*,6S*)-3-[methoxy (methylthio) methyl]-2-pyrrolidinethione, raphanusanin B)and benzoxazolinone (6-methoxy-2-bezoxazolinone, MBOA) on apical dominance of pea(Pisum sativum L. cv. Alaska) seedlings were studied.Application of raphanusanin B or MBOA to the apical bud, internode, or lateralbud of pea seedlings released apical dominance in either intact orindole-3-acetic acid (IAA )-treated, decapitated plants. These results suggestthat the auxin-inhibiting substances raphanusanin B and MBOA have activity inreleasing apical dominance. Conversely, the auxin transport inhibitors2,3,4-triiodobenzoic acid (TIBA) and 1-naphthylphthalamic acid (NPA) did notstimulate lateral bud growth when they were applied directly to the lateralbud,although application to the apical bud or internode released apical dominance.Therefore, the mode of action of raphanusanin B and MBOA in apical dominance isclearly different from that of auxin transport inhibitors. Raphanusanin B andMBOA may suppress the synthesis of growth-inhibiting factor(s) of the lateralbud induced by endogenous auxin transported from the apical bud or exogenouslyapplied auxin, and/or the action of the factor(s).  相似文献

Trichoderma species are commonly used as biological control agents against phytopathogenic fungi and some strains are able to produce metabolites that enhance plant growth. In the current study we evaluated the production of potential growth-promoting metabolites, rhizosphere competence and endophytism for 101 isolates of Trichoderma from Colombia, and assessed the relationship of these factors to the enhancement of early stages of growth on bean seedlings. Twenty percent of these Trichoderma strains were able to produce soluble forms of phosphate from phosphoric rock. Only 8% of the assessed strains showed consistent ability to produce siderophores to convert ferric iron to soluble forms by chelation. Sixty percent of isolates produced indole-3-acetic acid (IAA) or auxin analogues. The production of any of these metabolites was a characteristic of specific strains, as the ability to produce these metabolites varied greatly within species. Moreover, the production of these substances did not correlate with enhanced growth on bean seedlings, measured as the combined increase in length of roots and aerial parts in the V3 stage of growth. Seven Trichoderma isolates significantly improved the growth of bean seedlings. However, metabolite production varied widely in these seven strains, and some isolates did not produce any of the assessed growth-promoting metabolites. Results indicated that growth was enhanced in the presence of rhizosphere competent and endophytic strains of Trichoderma, and these characteristics were strain-specific and not characteristic for species.  相似文献

Avena seedlings were germinated and grown while continuously rotated on the horizontal axis of a clinostat. The coleoptiles of these gravity-compensated plants were phototropically more responsive than those of plants rotated on a vertical axis. When the plants were compensated after unilateral irradiation, phototropic curvature of the shoot progressed for the next 6 hours, with the rate of curving decreasing about 3 hours after irradiation. The decrease in rate was less in the plants gravity-compensated before irradiation than in those vertically rotated. In the period 70 to 76 hours after planting, the growth rate of the compensated coleoptiles was significantly less than that of the vertically rotated seedlings. The greater phototropic curvature, the decreased growth rate, and the slower rate of straightening of the curved, compensated shoot can be correlated with several consequences of compensation: an increase in sensitivity to auxin, a lowering of auxin content in the coleoptile tip, and possibly, from an interaction between compensation and phototropic stimulation, an enhanced difference in auxin transport between the illuminated and shaded halves of the unilaterally irradiated shoot.

The phototropic response of the vertically rotated seedling was significantly different from that of the vertical stationary, indicating the importance of vertically rotated controls in clinostat experiments.

Soybean seedlings treated with ethylene exhibited small increases in ribonucleic acid content in the elongating section of the hypocotyl. Chromatin isolated from the elongating section of ethylene-treated seedlings showed a 35 to 60% increase in the capacity for RNA synthesis. The ethylene-induced response was saturated at 1 microliter/liter of ethylene and was fully expressed after 3 hours. Auxin caused marked accumulation of RNA and DNA in the elongating and basal tissue of the hypocotyl. Chromatin isolated from these auxin-treated tissues showed an 8- to 10- fold increase in RNA synthetic capacity as measured in vitro. Ethylene added with auxin reduced the auxin enhancement of nucleic acid synthesis in the elongating and basal tissues. Both ethylene and auxin treatment of the seedlings inhibited nucleic acid accumulation and chromatin activity in the apical tissue. Ethylene did not appear to mediate the auxin effects on nucleic acid synthesis in soybean hypocotyl with the possible exception of inhibition in the apical tissue.  相似文献

Striga senegalensis seeds were cultured on Knop's and modified Murashige-Skoog's medium. An aqueous root exudate/extract of the host (Sorghum vulgare) was found essential for the germination of seeds in vitro. The seedlings grew better on a medium containing sucrose than on mineral salts alone. Only limited growth was achieved on Knop's medium. Better growth was supported by Murashige-Skoog's medium. By transfer to larger culture vessels seedlings were kept in the latter for several months. The seedlings developed chlorophyllous shoots in light and flowered after 4–4½ months. The successful culture of these seedlings in a simple medium of only mineral salts and sugar indicates that the parasitic seedling depends on the host for only water, mineral salts and sugar and not for more elaborate substances.  相似文献

Summary Mutant lines of Arabidopsis thaliana resistant to the artificial auxin 2,4-dichloro phenoxyacetic acid (2,4-D) were isolated by screening for growth of seedlings in the presence of toxic levels of 2,4-D. Genetic analysis of these resistant lines indicated that 2,4-D resistance is due to a recessive mutation at a locus we have designated Axr-1. Mutant seedlings were resistant to approximately 50-fold higher concentrations of 2,4-D than wild-type and were also resistant to 8-fold higher concentrations of indole-3-acetic acid (IAA) than wild-type. Labelling studies with (14C)2,4-D suggest that resistance was not due to changes in uptake or metabolism of 2,4-D. In addition to auxin resistance the mutants have a distinct morphological phenotype including alterations of the roots, leaves, and flowers. Genetic evidence indicates that both auxin resistance and the morphological changes are due to the same mutation. Because of the pleiotropic morphological effects of these mutations the Axr-1 gene may code for a function involved in auxin action in all tissues of the plant.  相似文献

It is widely reported that some humic substances behave as exogenous auxins influencing root growth by mechanisms that are not yet completely understood. This study explores the hypothesis that the humic acids’ effects on root development involve a nitric oxide signaling. Maize seedlings were treated with HA 20 mg C L−1, IAA 0.1 nM, and NO donors (SNP or GSNO), in combination with either the auxin-signaling inhibitor PCIB, the auxin efflux inhibitor TIBA, or the NO scavenger PTIO. H+-transport-competent plasma membrane vesicles were isolated from roots to investigate a possible link between NO-induced H+-pump and HA bioactivity. Plants treated with either HA or SNP stimulated similarly the lateral roots emergence even in the presence of the auxin inhibitors, whereas NO scavenger diminished this effect. These treatments induced H+-ATPase stimulation by threefold, which was abolished by PTIO and decreased by auxin inhibitors. HA-induced NO synthesis was also detected in the sites of lateral roots emergence. These data depict a new scenario where the root development stimulation and the H+-ATPase activation elicited by either HA or exogenous IAA depend essentially on mechanisms that use NO as a messenger induced site-specifically in the early stages of lateral root development.  相似文献

 Hypocotyls of the diageotropica (dgt) mutant of tomato (Lycopersicon esculentum Mill.) do not elongate in response to exogenous auxin, but can respond to gravity. This appears paradoxical in light of the Cholodny-Went hypothesis, which states that shoot gravicurvature results from asymmetric stimulation of elongation by auxin. While light-grown dgt seedlings can achieve correct gravitropic reorientation, the response is slow compared to wild-type seedlings. The sensitivity of dgt seedlings to inhibition of gravicurvature by immersion in auxin or auxin-transport inhibitors is similar to that of wild-type plants, indicating that both an auxin gradient and auxin transport are required for the gravitropic response and that auxin uptake, efflux, and at least one auxin receptor are functional in dgt. Furthermore, dgt gravicurvature is the result of asymmetrically increased elongation as would be expected for an auxin-mediated response. Our results suggest differences between elongation in response to exogenous auxin (absent in dgt) and elongation in response to gravistimulation (present but attenuated in dgt) and confirm the presence of two phases during the gravitropic response, both of which are dependent on functional auxin transport.
