Feeding 50% ethanolic extract of A. aspera to male rats resulted in reduced sperm counts, weight of epididymis, serum level of testosterone and testicular activity of 3beta-hydroxysteroid dehydrogenase, while motility of the sperm and activity of the HMG CoA reductase were not affected. Cholesterol level in the testis, incorporation of labelled acetate into cholesterol, 17-ketosteroids in urine and hepatic and fecal bile acids were increased. The results suggest that ethanolic extract of A. aspera caused reproductive toxicity in male rats and the action may be by suppressing the synthesis of androgen. 相似文献

To study the efficacy of ethanolic extract of B. monosperma bark in cafeteria and atherogenic diet fed rats and monosodium glutamate (MSG) obese rats, different doses (200, 400 and 800 mg/kg) of ethanolic extract of B. monosperma bark showed dose dependent decrease in body weight, daily food intake, glucose, lipids, internal organs' weight and fat pad weight in cafeteria and atherogenic diet fed rats and monosodium glutamate obese rats. A 9-month laboratory study was carried out to determine the impact of herbivory by a moth with shoot-boring larvae, Dichrorampha odorata Brown and Zachariades (Lepidoptera: Tortricidae) on growth and reproductive ability of its host plant, Chromolaena odorata (L.) King and Robinson (Asteraceae), a major invasive alien plant species in southern Africa. Newly hatched D. odorata larvae were inoculated onto 0 (control), 50 and 100% of the shoot tips of C. odorata in the laboratory. At all treatment levels, the basal stem diameter of C. odorata was not affected by D. odorata larval feeding. Larval feeding by D. odorata significantly reduced the height of the main shoot and flower production in C. odorata relative to the control treatment but promoted branching by increasing the number of shoot tips. However, the differences in plant height and number of flowers between the 50 and 100% inoculation levels were not statistically significant. Dichrorampha odorata is the first shoot-tip attacking agent that is being released as a biological control agent against C. odorata in South Africa. In general, the impacts of D. odorata on the weed were relatively small even though statistically significant. The findings of this study suggest that high levels of damage by the moth will modestly reduce the height, flower production, and the competitiveness of C. odorata, thereby contributing to the biological control of the weed in South Africa. 相似文献

This report examined the effects of larval tapeworm infection on the reproductive functions in both male and female host rats. Female rats were matched by age, then randomly assigned to control and treatment groups (infected with larval tapeworms). Estrous cycles were determined by vaginal smear with 95% of the control group exhibiting 4-day normal cyclicity and only 55% of the treated group exhibiting normal cycles. Female fertility was then evaluated for the normally cycling rats based on the percentage of successful matings on the evening of proestrus, number of implantation sites on Day 8 of pregnancy, and number of pups born at term. The normally cycling rats exhibited 96% successful mating, 12.95 +/- 1.80 implantation sites, and 11.20 +/- 1.80 pups born. Five months after larval tapeworm infection, the fertility parameters were decreased to 79%, 9.10 +/- 1.20, and 7.50 +/- 1.50, respectively. The control females were then used in a study of male fertility after larval tapeworm infection employing the same parameters used to test female fertility. At the onset of the study, control groups exhibited 95% successful mating, 12.50 +/- 1.50 implantation sites, and 11.60 +/- 1.60 pups born at full term. After the 5-month infection period, the parameters were substantially reduced to 29%, 6.20 +/- 0.80 implantation sites, and 5.10 +/- 0.80 pups, respectively. Average testosterone concentrations in serum and testis from control male rats were 8.80 +/- 0.95 ng/ml and 3.88 +/- 0.25 ng/mg protein, respectively. After the 5-month infection period, these levels were reduced to 2.47 +/- 0.31 ng/ml and 1.28 +/- 0.12 ng/mg protein, respectively.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

The role of natural antioxidants in preventing of age-relating diseases is evident. The vegetable industry generates a large amount of waste, which is a good source of antioxidants.The aim of the study was the investigation of the antioxidant effect of long-term consumption of ethanolic yellow onion husk extract in ageing laboratory rodents.Twenty male Wistar albino rats were divided randomly into two groups (n = 10): a control group and an experimental group that received ethanolic yellow onion husk extract (2 mL/rat diluted with distilled water; activity of 4.44 µmol-equiv. quercetin) for 188 days. Oxygen radical absorbance capacity and ferric reducing antioxidant power assays were used to determine the total antioxidant capacity of the extract, which amounted to 941.4 ± 32.7 µmol equiv. Trolox/g raw material and 167.4 ± 16.4 µmol-equiv. quercetin/g raw material, respectively. Oral intake of the onion husk extract affected the indicators of the antioxidant system of the liver and the brain but not of the blood and plasma, mainly due to elevations in the activity of catalase and superoxide dismutase in the liver by 44.4% and 79.1%, respectively, and in the brain by three-fold and 79.1%, respectively.The availability, cheapness and high antioxidant potential of onion waste qualifies it a good source of functional ingredients and bioactive substances applicable in the food and pharmaceutical industries. 相似文献

Hexavalent chromium is an environmental contaminant which may be associated with reproductive abnormalities in male rats. In the present study, we examined the effect of hexavalent chromium on male reproductive function of rats. Male Wistar rats received a daily intraperitoneal injection of potassium dichromate (1 or 2 mg/kg body weight) for fifteen consecutive days. A decrease in testis weight and an increase in seminal vesicles and prostate weights were demonstrated after chromium treatment. Moreover, a dose-dependent increase in blood and testis chromium levels as well as an increase in FSH and a decrease in LH and testosterone serum levels were detected in treated rats. Histological analysis revealed pronounced morphological alterations with enlarged intracellular spaces, tissue loosening and dramatic loss of gametes in the lumen of the seminiferous tubules of treated rats. In addition, a decreased sperm motility and number of epididymal spermatozoa together with an increased sperm abnormality rate was found in chromium-treated rats in comparison to controls. In rats receiving the higher chromium dose, histological images presented considerably increased areas filled with seminal vesicle and prostate secretions. The mucosal crypts of seminal vesicles and the typical invaginations of prostate were altered. The results suggest that subacute treatment of potassium dichromate promotes reproductive system toxicity and affects testicular function of adult male rats. 相似文献

An experiment was conducted to determine the effect of feeding tall fescue seed infected by an endophytic fungus Acremonium coenophialum on the reproductive performance of male rats. Thirty 70-day-old rats were randomly allocated to four treatments: (I) fed 50% rat chow and 50% healthy fescue seed ad libitum (control; N=9; (II) same as I but restricted to the daily feed intake of III (N=7; (III) 50% fungal-infected fescue seed and 50% chow (N=7); and (IV) a mixture of 50% laboratory chow, 25% healthy fescue seed and 25% fungal-infected fescue seed (N=7). The rats were fed these diets for 42 days. During this time, body weights were taken weekly and feed intake was taken daily. At the end of the experiment, the rats were sacrificed and the testes and epididymides were excised and measured. Sperm parameters were assessed (concentration, percentage of motility and progressive motility) at the site of the cauda epididymis; the testes were homogenized and assessed for daily sperm production potential (DSP). Concentrations of spermatozoa (x 10(6)) among the various treatments were 645.6, 486.5, 387.4 and 457.1; motility and progressive motility measurements ranged from 48 to 50% and 2.3 to 2.4 (0-4), respectively. DSP values (per gram) and testicular weight were reduced (P<0.05) in Diet III. The data suggests that 50% fungal-infected fescue seed in the diet of rats does influence the DSP, testicular parechyma and epididymal weight in the rat. 相似文献

The current research was focused on evaluation of the cytotoxic and suppressive action of ethanolic extract of Equisetum arvense (EA1) against human pancreatic carcinoma cell line ASPC-1 after treatment with 25 µg/mL, 50 µg/mL, 100 µg/mL and 200 µg/mL EA1, using MTT assay and Antioxidant activity. Detailed investigations led to reveal the ability of cell patronage through the dreadful upshot of free radicals. The current approach followed MTT assays to examine the long-lasting ability and growth of cells as EA1 restrained the cell viability and growth of ASPC-1. At the end, EA1 showed its potential cytotoxicity and reduced the cellular proliferation of ASPC-1 cells through a pattern, which appeared to be concentration dependent. Our results can form the basis to explore the molecular mechanisms underlying Ethanolic Extract of Equisetum arvense induced cell death in pancreatic cancer cell lines and may serve as an alternative anticancer agent for the treatment of pancreatic carcinoma (PC) with no or least side effects to the patient. 相似文献

Cananga odorata (Lam.) Hook.f. & Thomson forma genuina (Annonaceae) is a tropical tree, grown for the production of ylang-ylang essential oil, which is extracted from its fresh and mature flowers. Despite its economic and social importance, very little information is available on its variability and the possible factors causing it. Therefore, the relationship between the genetic structure, revealed by amplified fragment length polymorphism (AFLP), and the essential oil chemical composition, determined by GC/MS analysis, of ylang-ylang grown in semi-managed systems in three Indian Ocean islands (Grande Comore, Mayotte, and Madagascar) was investigated. Our results revealed a low genetic variation within plantations and contrasted situations between islands. Variations of the chemical composition could be observed within plantations and between islands. The genetic differentiation pattern did not match the observed pattern of chemical variability. Hence, the chemical variation could not be attributed to a genetic control. As Grande Comore, Madagascar, and Mayotte present different environmental and agronomic conditions, it can be concluded that the influence of these conditions on the ylang-ylang essential oil composition is consistent with the patterns observed. Finally, several strategies were proposed to valorize the chemical composition variations. 相似文献

The potential haematological effects associated with the administration of ethanolic leaf extract of Ageratum conyzoides was investigated in rats. 27 rats were randomly divided into four groups. The first group had 6 rats and served as control, the remaining 3 experimental groups and had 7 rats each. These later groups were gavaged with the extract of Ageratum conyzoides in concentrations of 200 mg/kg, 400 mg/kg and 500 mg/kg respectively for 30 days at a dose of 0.1 ml/body weight. The control group was gavaged with 0.9% sodium chloride at a dose of 0.1 m1/body weight as placebo. The extract at the doses administered was found to increase in a dose-related fashion PCV and Hb ([P < 0.01] for 200 mg/kg and [P < 0.001] for 400 mg/kg and 500 mg/kg), RBC ([P < 0.05] for 400 mg/Kg and 500 mg/kg) and marginal increases that were not significant for 200 mg/kg); MCH and MCV ([P < 0.05] and [P < 0.01] for 400mg/kg and 500mg/kg respectively) 200 mg/kg was not significant. MCHC recorded no significant change. WBC recorded marginal increases that were not significant, similarly, the differential white blood cell recorded marginal increases that were not significant, except lymphocytes that recorded significant increase in group 4 [P < 0.05]. Marginal Decreases in body weight were also observed, these decreases were however not significant. The result of this study thus indicate haematopoietic potentials of the extract and could possibly remedy anaemia. 相似文献

Methanolic extract of M. oleifera root was found to contain some alkaloids (total alkaloid 0.2%). Effects of multiple weekly (35, 46, 70 mg/kg) and daily therapeutic (3.5, 4.6, 7.0 mg/kg) i.p. doses of the crude extract (CE) on liver and kidney functions and hematological parameters in mice were studied. No alteration in hematological and biochemical parameters at low and moderate dose level of daily and low dose level of weekly treatment of the extract was observed. However, the extract at moderate dose level in weekly treatment changed serum aminotransferase and plasma cholesterol levels significantly. High dose in addition to the above parameters changed total bilirubin, non protein nitrogen, blood urea and plasma protein. High dose of daily treatment and moderate and high dose of weekly treatment of CE increased WBC count and decreased clotting time significantly. The results indicate that the weekly moderate and high dose (> 46 mg/kg body wt.) and daily/therapeutic high dose (7 mg/kg) of CE affects liver and kidney functions and hematological parameters whereas the weekly dose (3.5 mg/kg) and low and moderate daily/therapeutic dose (3.5 and 4.6 mg/kg) did not produce adverse effects on liver and kidney functions. 相似文献

This study examined the effects of rhizomes of Alpinia calcarata Roscoe (Zingiberaceae) on male sexual competence and fertility, using a hot water extract (HWE) and rats. Different doses of HWE (150, 250 and 500 mg/kg) were orally administrated to male rats and their sexual behaviour was monitored (for 15 min) 3 h later using receptive females. Fertility was determined in a separate group (with the highest dose) using a noncompetitive copulation test. In the sexual behaviour study, the HWE impaired the number of rats ejaculating and markedly prolonged the latency for ejaculation. Further, the number of rats mounting and intromitting, and the latencies for mounting and intromission were inhibited. Collectively, these observations indicate a strong aphrodisiac action. The other parameters remained unchanged indicating non-impairment in libido, sexual arousability, sexual vigour and sexual performance or penile erectile ability. However, a slight impairment was evident in sexual motivation (with the highest dose) in a partner preference test. In the fertility test, HWE induced profound oligozoospermia but fertility was uninhibited. The highest dose of HWE also elevated the serum testosterone level and the number of spontaneous penile erections rapidly and markedly. Further, the HWE was nontoxic. It is concluded that A. calcarata rhizomes possess a strong and safe oral aphrodisiac activity. 相似文献

The effects of neonatal exposure to different doses of diethylstilbestrol (DES) on the reproductive functions of male rats at adulthood were evaluated. Sprague-Dawley rats (5-8/group) received sc injections of 25 microl olive oil containing DES (Sigma Chemical Co., St. Louis, MO) at a dose of 10 microg, 1 microg, 100 ng, 10 ng, or 1 ng per rat on alternate days from Postnatal Days 2-12. Control animals received olive oil only. All animals were allowed to develop until 83-91 days of age; however, when they were 70 to 80 days old, four male rats each from the 10 microg, 1 microg, 100 ng, and control groups were cohabited with untreated 60- to 70-day-old females (1:1) for 12 days. At the end of cohabitation, both mated and unmated male rats were weighed, and blood and tissue samples were collected and processed. Results revealed that although sperm motility patterns and sperm morphology were adversely affected in the 10- microg group, other reproductive parameters, including 1). daily sperm production (DSP)/testis; 2). absolute and relative weights of the testis, epididymis, and seminal vesicle; and 3). sperm numbers in both regions of the epididymis declined significantly in a dose-dependent manner in the 10- and 1- microg groups. Conversely, in the <1- microg groups, none of these parameters (except DSP/testis and weight of the epididymis in the 100-ng group, and sperm numbers in the epididymis of the 100- and 10-ng groups) was different from controls. Generally, plasma testosterone levels decreased in the 10- and 1- microg groups, FSH level increased in the 10-microg group, and prolactin and LH levels were unaltered. In the fertility study, although each male in the 1-microg, 100-ng, and control groups produced a copulatory plug and impregnated a female, none could do so in the 10-microg group. The mean number of pups per litter was reduced to eight in the 1-microg group, in contrast to 15 each in the 100-ng and control groups. In conclusion, exposure of neonatal male rats to DES altered sperm motility patterns, sperm fertility (as evident from the reduced number of pups in the 1-microg group), and sexual behavior (as evident from the absence of copulatory plugs in the 10-microg group) and reduced weights of reproductive organs, DSP/testis, and sperm numbers in the epididymis. The effect of water extract of licorice (Glycyrrhiza uralensis), one of the most widely used medicinal plants in Oriental nations and in Europe, on male reproductive function was investigated in rats. Licorice extract was prepared as in Oriental clinics and orally administered at doses of 500, 1,000 or 2,000 mg/kg, the upper-limit dose (2,000 mg/kg) recommended in the Toxicity Test guideline of the Korea Food and Drug Administration, to 6-week-old male rats for 9 weeks. Licorice extract neither induced clinical signs, nor affected the daily feed consumption and body weight gain. There were no significant changes in testicular weights, gross and microscopic findings, and daily sperm production between vehicle- and licorice-treated animals, in spite of slight decreases in prostate weight and daily sperm production at the high dose (2,000 mg/kg). In addition, licorice did not affect the motility and morphology of sperm, although the serum testosterone level tended to decrease without significant difference, showing a 28.6% reduction in the high-dose (2,000 mg/kg) group. The results suggest that the no observed adverse-effect level of licorice extract is higher than 2,000 mg/kg, the upper-limit dose, and that long-term exposure to licorice might not cause profound adverse effects. 相似文献

Methanolic extract of Albizia lebbeck bark when administered orally at the dose level of 100 mg/rat/day to male rats of proven fertility for 60 days did not cause any significant loss in their body weights but the weights of reproductive organs, i.e. testis, epididymides, seminal vesicle and ventral prostate were decreased in a significant manner when compared to controls. Sperm motility as well as sperm density were reduced significantly which resulted in reduction of male fertility by 100%. Marked decline in the germ cell population was noticed. Population of preleptotene, pachytene, secondary spermatocytes and step-19 spermatid were declined by 60.86%, 65.81%, 71.56% and 66.55%, respectively. Cross-sectional surface area of sertoli cells as well as the cells counts were found to be depleted significantly. Leydig cells nuclear area and number of mature Leydig cells were decreased by 60.03% and 51.56%, respectively. Serum testosterone levels showed significant reduction after A. lebbeck extract feeding. Oral administration of the extract did not affect red blood cell (RBC) and white blood cell (WBC) count, haemoglobin, haematocrit and glucose in the blood and cholesterol, protein, triglyceride and phospholipid in the serum. In conclusion, A. lebbeck bark extract administration arrests spermatogenesis in male rats without noticeable side effects. 相似文献

Acute and sub-acute toxicity of ethanolic extract of Canthium mannii Hiern stem bark on Mus musculus
Acute and sub-acute toxicity of ethanolic extract (ETE) of C. mannii was assessed on white mice (Mus musculus). After 48 h of extract administration, no death was registered. It was deduced that the LD50 was indisputably higher than 16 g/kg body weight. The sub-acute toxicity test was based on the daily administration of three doses of ETE (300, 600 and 1200 mg/kg body weight) for four weeks; 1% DMSO served as negative control. As for the first experiment, no sign of toxicity was registered. Conversely, the sub acute doses stimulated and increased the weight-rate of mice after 7 days of treatment. Except for the spleen weight, the doses administrated did not modify the weight index. It was observed that, subacute doses induced and increased (a) the food (particularly) and water consumption according to time and (b) the number of red and white blood cells. It was thought that, ETE can stimulate the haematopoietic function. Finally, no time variation of the activity of alanine aminotransferase and aspartate aminotransferase enzyme was observed in the serum of euthanized mice. The results showed the innocuity of ETE of C. mannii and thus validated his utilization in cameroonian traditional pharmacopoea. 相似文献

The ethanolic extract of seeds of S. cumini increased body weight and decreased blood sugar level in alloxan diabetic albino rats. Level of significance for decrease in blood sugar after feeding alcoholic extract of S. cumini seeds in various doses was highly significant. The extract feeding showed definite improvement in the histopathology of islets. The most important finding is that the blood sugar level, which once dropped to normal levels after extract feeding was not elevated when extract feeding was discontinued for 15 days. 相似文献
