Culture of Steinernema sp. was maintained on Corcyra cephalonica larvae. Steinernema sp. (at 50, 500, 1000, 2500, 5000, 10,000 and 20,000 ij’s /500?g soil) was concomitantly inoculated with 500 J2 of Meloidogyne incognita/500?g soil to the eggplant seedlings in the pots filled with 4?kg sterilised soil. The simultaneous inoculation of M. incognita with either of the inoculum levels (1000, 2500, 5000 and 10,000 J3/500?g soil) of Steinernema sp. significantly reduced the damage caused by M. incognita in terms of plant growth parameters, viz. plant length, dry weight, number of flowers and weight of fruits. Moreover, the highest improvement in plant growth parameters, viz. plant length, dry weight, number of flowers and weight of fruits, was recorded in plants inoculated with 5000 J3 of Steinernema sp./500?g soil followed by 2500, 1000 and 10,000 J3/500?g soil. The highest reduction in the reproduction factor and number of galls/root system was recorded in the plants treated with 5000 J3 Steinernema sp./500?g soil followed by 2500, 1000 and 10,000 J3/500?g soil. Comparison of concomitant and sequential inoculations showed that the sequential inoculation (both prior and after) of Steinernema sp. at different inoculum levels (1000, 2500, 5000, 10,000 and 20,000 ij’s/500?g soil) was more effective in the management of root-knot nematode than the concomitant inoculation. Therefore, the application of Steinernema sp. might be useful for suppression of nematode pest on eggplant and may be used as an alternative for chemicals.  相似文献

在玻璃温室内遮荫条件下,研究种皮和播种深度(0、2、5、10和15 cm)对辽东栎种子萌发和幼苗早期生长的影响.结果表明: 种皮对辽东栎种子萌发具有明显的抑制作用,去除种皮后,种子萌发率显著升高,不同播种深度处理种子萌发率平均升高19.4%,且萌发指数和活力指数显著增大,萌发进程加快.去皮种子和非去皮种子的萌发率均在2 cm播种深度下最大,分别为78.9%和62.2%;萌发指数和活力指数均在2 cm播种深度下最大,而萌发速率系数在5 cm播种深度下最大.去皮处理幼苗的单株叶面积和干质量与非去皮处理相比均显著增大,但比根长显著减小.去皮和非去皮处理幼苗株高均在5 cm播种深度下最大,分别为13.8和14.2 cm;基径随播种深度增加逐渐增大,而主根长、侧根数和最大侧根长均减小;幼苗干质量受播种深度影响较小.  相似文献

Pathogenic effect of root-knot nematode Meloidogyne arenaria was studied on balsam (Impatiens balsamina) by inoculating the different inoculum levels of root-knot nematode. It was observed that the inoculum levels up to 2000 J2 of root-knot nematode did not show significant reduction in plant growth characters as compared to control. Although the significant reduction in plant growth characters was recorded at and above 3000 J2 of root-knot nematode, progressive increase in the host infestation as indicated by the number of galls as well as the population of root-knot nematode was recorded with an increase in the level of inoculum. However, the rate of nematode multiplication was reduced with the increase in the inoculum density of M. arenaria. It can be concluded from these results that the damaging threshold level of M. arenaria on balsam was found to be as 3000 J2/plant.  相似文献

A field experiment on winter wheat in autumn 1991 investigated the effect of the rhabditid nematode, Phasmarhabditis hermaphrodita, applied to soil at five dose rates (108 - 1010 infective larvae ha-1) immediately after seed sowing, on slug populations and damage to seeds and seedlings. The nematode was compared with methiocarb pellets broadcast at recommended field rate immediately after drilling and no molluscicide treatment. Slug damage to wheat seeds and seedlings was assessed 6 and 13 wk after drilling. Seedling survival increased and slug grazing damage to seedlings declined linearly with increasing log nematode dose. These two measures of slug damage were combined to give an index of undamaged plant equivalents, which also increased linearly with increasing log nematode dose. ANOVA showed that, after 6 wk, there were significantly more undamaged plant equivalents on plots treated with the two highest nematode doses (3 × 109 and 1 × 1010 ha-1) than on untreated plots, but the number of undamaged plant equivalents on methiocarb-treated plots was not significantly greater than that on untreated plots. Slug populations were assessed by refuge trapping and soil sampling. Deroceras reticulatum was the commonest of several species of slugs recorded. During the first 4 wk after sowing, significantly more slugs were found under refuge traps on plots treated with certain doses of P. hermaphrodita than under traps on untreated plots and more showed signs of nematode infection than expected from the prevalence of infection in slugs from soil samples, suggesting that the presence of P. hermaphrodita altered slug behaviour. Application of P. hermaphrodita had no significant impact on numbers or biomass of slugs in soil during a 27 wk period after treatment, except after 5 wk when slug numbers were inversely related to log nematode dose. However, by this time, numbers in soil samples from untreated plots had declined to levels similar to those in plots treated with the highest dose of nematodes. During the first 5 wk after treatment, c. 20% of slugs in soil samples from untreated plots showed symptoms of nematode infection. It is suggested that this represented the background level of infection in the experimental field rather than spread of infection from treated plots. The apparent lack of impact of P. hermaphrodita on slug numbers and biomass in soil suggests that its efficacy in protecting wheat from slug damage was through inhibition of feeding by infected slugs.  相似文献

Influence of different inoculum levels of 0, 10, 100, 1000 and 10,000 individuals of Hirschmanniella oryzae on nematode reproduction and plant growth of rice cv. Giza171 and biochemical changes of infected plants was studied under screen-house conditions. Rate of nematode build up (Pf/Pi) was negatively correlated with the progressive increase in nematode inoculum levels. The percentage reduction in growth parameters, rice grain yield and the amount of total and reducing sugars were markedly affected showing a negative correlation with the tested inocula. The conspicuous reductions of plant growth, yield and total and reducing sugar contents were obtained by using 1000 and 10,000 nematodes per pot. The inoculum level of 1000 nematodes per pot was identified as critical population at which control programme must be started.  相似文献

Penetration of second-stage juveniles (J2) of Meloidogyne incognita into tomato root explants and in vitro propagated peach plantlet roots were compared. Five inoculum levels were used: 25, 50, 75, 100, and 200 J2 for tomato; and 50, 100, 200, 500, and 1,000J2 for peach. The greatest root penetration into tomato was 30% at the 75 J2 level, but the maximum penetration into peach roots was only 8% at the 200 J2 level. The difference (P = 0.05) in penetration of M. incognita at all inoculum levels into these two hosts indicates that penetration versus inoculum density for in vitro studies need to be determined for different plant species.  相似文献

The root-knot nematode, Meloidogyne incognito (Kofoid et White) Chitwood is an important pathogen of vegetables. Five commercial cultivars of lettuce (Lactuca sativa L.) were evaluated under greenhouse conditions for resistance to Meloidogyne incognita, Benguet population. Plants were inoculated with 1000 eggs collected from 'Apollo' tomato (Lycopersicon esculentum) roots. The degree of galling and number of egg masses were assessed 4 and 8 weeks after inoculation. Host plant response was classified as immune, highly resistant, resistant, moderately resistant, intermediate, moderately susceptible, and highly susceptible based on the resistance index of Kouamè et at., 1998 [RI = (gall2 + egg2)]. Inoculation of 1000 eggs/plant significantly affected the growth and yield of the five lettuce cultivars 4 and 8 weeks after inoculation. A significant interaction was observed between treatment and cultivar during the two evaluation periods in terms of marketable and non-marketable yield, plant height, root weight, number of galls and number of egg masses. A reduction in growth and yield was observed in the cultivars Ballon, Lollo Rosa and Red Wave. Significant differences were noted in the number of galls and egg masses among the different cultivars tested. The highest average number of galls was obtained from the cultivars Red Wave, Ballon and Lollo Rosa. Cultivar Ballon had the highest average number of recovered nematode while Gilaben had the lowest with 15 and 4 per roots, respectively after 4 weeks inoculation. After 8 weeks, nematode was highest in cultivar Red Wave (615) and lowest in Great Lakes (70). Based on the host response, cultivars Great Lakes and Gilaben were rated highly resistant and resistant, respectively, while Red Wave, Ballon and Lollo Rosa were rated intermediate.  相似文献

Effects of the root-knot nematode (Meloidogyne incognita) on lentil (Lens culinaris) were studied under greenhouse conditions. The plants were inoculated with 250, 500, 1000, 2000 and 4000 J2 per plant. Plant growth, yield, nodulation, seed weight, chlorophyll, nitrogen, phosphorus and potassium, (NPK) contents, as compared to control, were found decreased in all the nematode infected plants. The extent of reduction increased with an increase in inoculum levels. The reductions were significant at 500 J2 and at higher inoculum levels, i.e. 1000, 2000 and 4000 J2 per pot over the control. An increase in inoculum level caused enhancement in galling, egg mass production and nematode population. At higher inoculum levels, the population of the nematode in the root as well as in the soil increased to a greater magnitude than at lower inoculum levels. On the contrary, reproduction factor (RF) and rate of population increase (RPI) decreased with increasing inoculum levels.  相似文献

An investigation was carried out to study the pathogenicity of root knot nematode Meloidogyne incognita on okra and its management through various organic amendments. The inoculum level of 1000 juveniles per plant showed significant reduction in various plant growth parameters, which reveals that M. incognita is a potential pathogen of okra. With the increase in inoculums level of M. incognita (J2), there was a progressive decrease in various plant growth parameters. The maximum reduction in plant growth parameters was observed at an inoculum level of 4000 juveniles per plant. The efficacy of five organic amendments viz. groundnut cake, castor cake, sunflower cake, linseed cake and sawdust was tested against root knot nematode M. incognita. Amending the soil with different oil cakes was found to be effective in reducing the nematode soil population, number of females, number of egg masses as well as root gall formation in okra. The highest increase in plant growth (13%) and maximum reduction in number of galls (54%), number of females (57%) and number of egg masses (55%) was recorded on application of groundnut cake.  相似文献

Postinfection development of Meloidogyne chitwoodi from second-stage juveniles (J2) to mature females and egg deposition on ''Nugaines'' winter wheat required 105, 51, 36, and 21 days at 10, 15, 20, and 25 C. At 25 C, the J2 induced cavities and hyperplasia in the cortex and apical meristem of root tips with hypertrophy of cortical and apical meristem cell nuclei, 2 and 5 days after inoculation. Giant cells induced by late J2 were observed in the stele 10 days after inoculation. Clusters of egg-laying females were common on wheat root galls 25 days after inoculation. Juveniles penetrated wheat roots at 4 C and above, but not at 2 C, when inoculum was obtained from cultures grown at 20 C, but no penetration occurred at 4 C when inoculum was stored for 12 hours at 4 C before inoculation. In northern Utah, J2 penetrated Nugaines wheat roots in the field in mid-May, about 5 months after seedling emergence. M. chitwoodi eggs were first observed on wheat roots in mid-July when plants were in blossom. Only 40% of overwintered M. chitwoodi eggs hatched at 25 C.  相似文献

Accumulation of the phytoalexin glyceollin I in roots of soybean (Glycine max [L.] Merr.) following inoculation with race 1 of Heterodera glycines Ichinohe, the soybean cyst nematode (SCN), was determined in a whole-root system by high performance liquid chromatography (HPLC) and in a cross-section system by a radioimmunoassay procedure. In the whole-root system, roots were harvested from controls and nematode-inoculated seedlings immediately after inoculation and at 2-day intervals for 8 days. The roots were extracted with ethanol, and the extracts were subjected to HPLC. Glyceollin I was not detected in roots of either resistant cultivar Centennial or susceptible cultivar Ransom immediately after inoculation with SCN but steadily accumulated in large quantity in roots of Centennial. Accumulation of glyceollin I in roots of Ransom following nematode inoculation was minimal. In the cross-section system, 3-day-old soybean seedlings were inoculated with juvenile nematodes, and root segments containing a single nematode were dissected from inoculated plants at 4-hour intervals under a dissecting microscope. The root segments were embedded in ice and cut into 16-micrometer sections with a cryostat microtome. The spatial and temporal distribution of glyceollin I was determined with a radioimmunoassay procedure specific for the phytoalexin. Glyceollin I was found to accumulate in tissues immediately adjacent to the head region of the nematode in Centennial but not in Ransom. Glyceollin I was detected 8 hours after nematode penetration, and the concentration increased steadily up to 0.3 micromole per milliliter in Centennial 24 hours after penetration.  相似文献

In this study, three concentrations (250, 500, and 1000 ppm) of abamectin 2% suspension concentration (SC) were used as cucumber seeds treatment. The seeds were treated with abamectin to reduce nematodes reproduction and their ability to penetrate the roots, then seed germination and plant growth were observed. All the concentrations didn’t negatively affect seeds germination wherever the germination percent reached 80% at the concentration (1000 ppm) after 20 days of sowing. The effect of abamectin on root-knot nematode was studied by recording numbers of nematodes in 100 g/soil, numbers of the galls, egg mass on the root, and the nematode reproduction factor. All concentrations significantly affected the nematode reproduction parameters compared to control. Abamectin at (500 ppm) was the most effective concentration on reducing nematodes parameters, i.e., 26.57, 38.83, 47.40 %, and 3.15 for the above-mentioned parameters, respectively at the end of experimint. No significant difference between 500 ppm and 1000 ppm. We recommended using the abamectin in (500 ppm) concentration as a seed application to control Meloidogyne incognita in cucumber plants under greenhouse conditions to reduce its environmental toxic effect.  相似文献

该文在玻璃温室内的遮阴环境下,采用盆播方法研究了辽东栎种子特征(大小和种皮)和播种深度(0、3、6、10 cm)对种子萌发和幼苗生长的影响。结果表明:(1)辽东栎大种子萌发率、萌发速率、萌发值和萌发指数在所有播种深度均高于小种子。其中非去皮种子萌发率、萌发速率和萌发指数在0 cm播种深度的不同大小种子间差异显著,去皮种子所有萌发参数在6和10 cm播种深度的不同大小种子间均差异显著;去皮可促进大种子萌发,但抑制小种子萌发;不同大小种子所有萌发参数均在0 cm播种深度最大,在10cm播种深度最小。(2)不论有无种皮,大种子萌发幼苗的叶片数、单株叶面积、总干物质质量和根冠比在所有播种深度均大于小种子萌发幼苗;去皮种子萌发幼苗的株高、基径、叶片数、单株叶面积和总干物质质量在所有播种深度均不同程度地小于非去皮种子萌发幼苗,但前者根冠比在0、3、6 cm播种深度大于后者;随着播种深度增大,幼苗株高、叶片数、单株叶面积、总干物质质量和根冠比等生长参数均呈减小趋势,但基径随播种深度增大而增大。  相似文献

以2年生杜鹃(Rhododendron simsii)苗为研究对象,采用J1(杜鹃花类菌根真菌混合接菌)、J2(杜鹃花类菌根真菌和马尾松外生菌混合接菌)、J3(不接菌,对照)等3种接菌组合与不同氮磷钾施肥组合研究菌根真菌与氮磷钾肥对杜鹃苗生长及养分吸收的影响。结果表明,杜鹃幼苗的生长指标和氮磷钾含量总体表现为接菌处理优于不接菌处理,J2处理下施用尿素1.1 g·株-1、钙镁磷1 g·株-1、氯化钾0.7 g·株-1对杜鹃苗生长及氮磷钾含量积累的促进作用最为明显。数据表明,J2处理促进N向叶积累,促进K向茎叶积累,促进P向根茎积累;J1处理促进N和P向根茎积累,促进K向茎叶积累。  相似文献

线虫与微生物的相互作用研究往往是在悉生培养体系 (gnotobiotic microcosm)中进行 ,为了研究在自然或开放土壤条件下土壤线虫与微生物的相互作用 ,作者在开放盆栽体系中接种土壤食细菌线虫 (原小杆线虫 ,Protorhabditis sp) ,研究在小麦不同生育期、在有和无根系作用下食细菌线虫对土壤微生物量和微生物群落结构的影响。结果表明 :接种线虫分别使 SMBC、SMBN、SMBP提高了 2 6 .4 %、32 .9%、2 1.8% ,这种促进作用除个别无根系土和非根际土处理外 ,均达到显著性差异。根际土中的 SMBC、SMBN、SMBP>非根际土 >无根系土。从方差解释比例 v来看 ,SMBN受线虫的影响最大 (v=2 4 % )、其次是 SMBC(v=16 % )、然后是 SMBP(v=12 % ) ,线虫对 SMBC的促进作用在根际土中最突出。接种线虫对土壤细菌、真菌和放线菌的数量有明显的影响。在苗期的无根系土和根际土中 ,接种线虫显著降低了细菌的数量、特别在根际土中尤为突出 ,但在其它处理中却增加了细菌的数量。接种线虫对真菌和放线菌数量的促进作用比对细菌更为明显 ,接种线虫后真菌和放线菌数量的总平均值分别比未接种提高了 4 8.5 %和 6 8.2 % ,而细菌数量的总平均值没有变化。细菌数量与微生物量 C相关散点图表明二者相关性在根际土、非根际土和无根系土中均未  相似文献

The strength and direction of plant response to inoculation with arbuscular mycorrhizal fungi (AM fungi) is dependent on both abiotic and biotic contexts, often generating patterns of AM fungal mediation of plant adaptation. However, knowledge of plant‐community level effects of these interactions in grassland restoration is limited. We conducted a field inoculation experiment by inoculating five plant species native to a drier prairie and five plant species native to a moister prairie with mycorrhizal fungal communities from each prairie type. Species were paired by genus or family to account for phylogenetic effects. The inoculated plants were transplanted to study plots seeded with a restoration seed mix. Plots were manipulated to create either moister or drier conditions similar to environments of the plant species and mycorrhizal communities. In both transplanted and seeded plant species, we found that only drier prairie‐range species benefited from moisture‐regime matched AM fungal inoculum. Other seeded prairie plant species demonstrated a negative response to inoculation, likely due to the earlier successional stage of these species. Additionally, nonseeded plants benefited from inoculation in different ways: native nonseeded plants had highest cover with drier prairie inoculum in drier conditions, while nonnative plants had highest cover with moister prairie‐origin inoculum. These results suggest that use of local AM fungi may be particularly important in restorations at drier sites, even at relatively small differences in moisture availability. Further, specific knowledge of relative responsiveness of seeded plant species and nonseeded plant species to AM fungal inoculation will be useful in planning restorations.  相似文献

Winter wheat was grown for six successive years (Expt 1) and for three successive years (Expt 2) in field experiments on different soil types. Artificial inoculum of the take-all fungus (Gaeumannomyces graminis var. tritici cultured on autoclaved oat grains) was incorporated in the soil of some of the plots just before, or at, sowing of the first winter wheat crop. Expt 1 tested the incorporation of similar amounts of inoculum (212 kg ha-1) at different depths. Expt 2 tested different amounts of inoculum at the same, shallow depth. Early sowing (September), late sowing (October) and spring inoculation were additional treatments, applied to the first crop only, in Expt 2. Seasonal factors apart, the disease outcome in the first year after inoculation depended on amounts and placement of applied inoculum, as well as date of sowing. Deeper inoculum resulted in less disease (Expt 1). Severe take-all was produced in Expt 2 by incorporating inoculum shallowly in sufficient quantities (400 kg ha-1 or more). Less inoculum (200 kg ha-1) generated less disease, especially in earlier-sown plots. Differences in disease amongst inoculum treatments were greatest in the first year and diminished subsequently, particularly where sowing had been early in the first year. In Expt 1, where first crops exposed to artificial inoculum developed moderate-to-severe disease, disease in subsequent second and/or third crops was less. In the fourth crop a second peak of disease occurred, coinciding with a first peak in sequences without added inoculum. Take-all decline (TAD) appeared to be expressed in all sequences thereafter. In Expt 2 in sequences without added inoculum, TAD occurred after a peak of disease in the second crop. Where 400 kg ha-1 or more of inoculum were added, disease was severe in the first year and decreased progressively in successive years. Disease was less patchy in plots that received artificial inoculum. However, it remains uncertain mat severe disease caused by artificial inoculation achieved an early onset of true TAD. The infectivity of the top 12 cm of soil in the first 3 yr of Expt 1, determined by bioassay, depended on the depth of added inoculum and amount of disease in subsequent crops. However, at the time of the naturally occurring peak of disease severity (in either inoculated or non-inoculated plots) it did not predict either disease or TAD. Differences and similarities amongst epidemics developing naturally and those developing from different amounts and placement of applied inoculum have been revealed. The epidemiological implications of adding inoculum and the potential value of artificially-created epidemics of take-all in field trials are discussed.  相似文献

Damaging threshold levels of root-knot nematode Meloidogyne incognita and root-rot fungus Fusarium solani on plant growth parameters, viz., plant length, fresh and dry weight of chilli were determined by conducting their pathogenicity trials in pot experiments. The results revealed a significant reduction in the plant growth parameters at and above the inoculum level of about 1000?J2 per plant of M. incognita and the highest reduction was recorded at 8000?J2 per plant. Significant reduction in plant growth parameters was recorded at 1.00?g mycelial mat of F. solani per plant, while the highest reduction was observed at 8.00?g mycelial mat per plant. Seven-month-old Scots pine seedlings were inoculated with water or culture filtrate (controls), with 10,000, or 20,000 (experiment 1), and with 2,500 (experiment 2) Bursaphelenchus xylophilus B.C. isolate nematodes and maintained under defined experimental conditions. Controls did not develop pine wilt disease over a 2-month period. In experiment 1, less than 50% of the inoculum was recovered from the nematode-inoculated seedlings in the first 48 hours, after which the nematode population of both treatments increased exponentially resulting in pine death and approximately equal populations at 216 hours after inoculation. In the second experiment, plant mortality, which was always preceded by 2-3 days of chlorosis and associated stem vascular necrosis, first occurred 14 days after inoculation. The nematode population increased until about day 40 after inoculation and declined thereafter. Nematodes extracted from the roots 2 weeks after inoculation accounted for ca.15% of the total number of nematodes per pine. The study indicates that the rate of nematode reproduction is a factor in pine wilt disease. However, the lack of a linear correlation between the number of nematodes and the timing of pine mortality suggests that the timing of pine death may also depend on the location of nematode damage to the host tissue.  相似文献

Although fertilisers enriched in rare earth elements (REE) are widely used in agricultural practice, little is known yet about behaviour of the REE in soil–plant system. Among REE, europium (Eu) may be of highest interest. Eu may serve as an indicator of biogeochemical processes due to a change of its valence state under different environmental conditions. Since chemical characteristics of Ca and REE are similar, Eu may compete with Ca for organic ligands. In the present work we studied the influence of Eu and Ca on the growth and mineral nutrition of wheat seedlings. An application of Eu favoured germination and root growth, whereas in combination with Ca it produced a more sustained leaf growth. Eu affected uptake and distribution of certain nutrients in different parts of a plant. The content of Eu in all parts of wheat seedlings at the end of germination significantly increased. Transfer of the seedlings germinated in the Eu-rich medium into soil resulted in the release of Eu from the plants. Nevertheless, 9 days after the sowing, Eu concentration in the seedlings was still much higher than in the control plants.  相似文献
