A simple procedure for spectrophotometric determination of amylase activity in coloured solutions is described. Starch cross-linked with a bifunctional reagent in the presence of black drawing ink is used as an insoluble chromolytic substrate. The absorbances of reaction mixture filtrates are read in the near-infrared region (at 800-900 nm) where the majority of coloured substances does not absorb; on the contrary, black drawing ink released from the substrate during the action of amylase absorbs strongly even at these wavelengths.  相似文献

CM-cellulose, cross-linked with epichlorhydrin in the presence of black drawing ink, is used as an insoluble chromolytic substrate. Summary A new insoluble chromogenic substrate for the determination of proteolytic activity was developed. This substrate was prepared by incorporating black drawing ink into casein and heating this complex at 200°C for 4 h. Summary A new insoluble chromolytic substrate for the spectrophotometric determination of alpha-amylase activity (starch cross-linked with 1,4-butanediol diglycidyl ether in the presence of black drawing ink; “black starch”) has been shown to work well also in the presence of dextran, Ficoll and polyethylene glycol; on the other hand these phase-forming polymers interfere with some commonly used alpha-amylase assays.  相似文献

A white heifer ("Snow") was born in 1991 from coloured registered Jersey parents. She produced six calves sired by coloured Jersey bulls: three white bull calves, two white heifer calves, and one coloured bull calf. One of the white bull calves was mated with 40 Hereford × Friesian yearling heifers (white face, predominantly black body with some white patches). The 38 resulting calves included 16 white and 22 coloured calves. Twelve of the 16 white calves were heifers and four were bulls. Red or black spotting was recorded on some white calves. The results are consistent with an autosomal dominant mutant causing the white phenotype. The mutation appears to have arisen spontaneously in Snow, then passing to her white progeny and white grand-progeny. The white individuals varied from entirely white in a few cases, to most having some residual small areas of red or black pigmentation in patterns not typical of other reported white spotting patterns of cattle.  相似文献

ABSTRACT. Glossina morsitans morsitans Westw. and G. pallidipes Aust. flying around and landing on coloured screens and traps were sampled using electrocuting nets. External colour affected both attractiveness and efficiency of traps, such that yellow and green traps were unattractive and inefficient; black and red, attractive and inefficient; white, moderately attractive, and very efficient; and blue traps, attractive and efficient. The order of attractiveness of coloured screens was similar to that for traps. Landing responses were generally strongest on black surfaces, and weakest on white, but the results for blue were variable. Carbon dioxide and acetone odours improved trap catches and also eliminated most catch differences between traps differently coloured on their outer surfaces. The relative performances of traps coloured differently on inside surfaces only were not affected significantly by these odours, and in all cases black or red target areas inside the trap were required for optimum trap performance. When acetone and 1-octen-3-ol odours were used, catches were improved but the relative performance of differently coloured traps and screens was not changed. There were no obvious species differences in colour responses although numbers of G. morsitans were too low for statistical comparisons.  相似文献

Poly(ethyleneimine) (PEI) microcapsules containing laccase from Trametes hirsuta (ThL) and Trametes versicolor (TvL) were printed onto paper substrate by three different methods: screen printing, rod coating, and flexo printing. Microcapsules were fabricated via interfacial polycondensation of PEI with the cross-linker sebacoyl chloride, incorporated into an ink, and printed or coated on the paper substrate. The same ink components were used for three printing methods, and it was found that laccase microcapsules were compatible with the ink. Enzymatic activity of microencapsulated TvL was maintained constant in polymer-based ink for at least eight weeks. Thick layers with high enzymatic activity were obtained when laccase-containing microcapsules were screen printed on paper substrate. Flexo printed bioactive paper showed very low activity, since by using this printing method the paper surface was not fully covered by enzyme microcapsules. Finally, screen printing provided a bioactive paper with high water-resistance and the highest enzyme lifetime.  相似文献

The melanocyte-stimulating hormone receptor (MC1-R) is a central regulator of mammalian coat colour, encoded by the extension locus. In cattle, the dominant extension allele E(D) is associated with the production of black pigment in coloured areas. Genotyping of the MC1-R gene in a bull with mosaic expression of red vs. black pigment verified the existence of the E(D) allele, in spite of the fact that the majority of the animal is red coloured. No further mutations were found within the E(D) variant of the MC1-R gene, which was inherited from a completely red mother (genotype E(D)/e).  相似文献

The heat shock response has been suggested as a potential biomarker in toxicology. A vast amount of stimuli have been shown to induce heat shock proteins and new techniques to measure the response are constantly being assessed. In this study we use a novel immunocytochemistry technique to measure heat shock protein 70 (hsp70) induction in L929 cells exposed to cadmium chloride. Hsp70 induction was quantifiably measured using a soluble coloured substrate and qualitatively measured using a coloured substrate that precipitated at the location of hsp70. Using the insoluble coloured substrate hsp70 was identified predominantly within the cytoplasm of control cells. At intermediate cadmium concentrations hsp70 was observed to translocate to the nucleus. At these intermediate concentrations a heterogeneous heat shock response was observed. At lethal concentrations a strong heat shock response was observed with a widespread cellular response. This study demonstrates the potential of this immunocytochemistry technique to measure toxicological effects in cells by identifying the response quantitatively and qualitatively.  相似文献

In view of alleged positive effects of coloured (red) hailnets on phytochrome, photosynthesis, yield and fruit quality, the objective of the present work was to investigate a range of red and green hailnets using apple as a model crop with cvs. ‘Pinova’ and ‘Fuji Kiku 8’. Light transmission of green or red hailnets peaked between 500 and 570 nm (green) or above 570 nm (red–orange) and was reduced by 12% (white) or 14% (red–white), 18% (red–black) and 23% (green–black) hailnets; there were no effects on phytochrome. Leaf chlorophyll concentration increased under coloured hailnets by up to 46% under the green–black hailnet, while air temperature was reduced by 0.2°C. Under sunny conditions, photosynthesis of ca. 18 μmol CO2 m−2 s−1 was not reduced under coloured hailnets, in contrast with a 21% reduction under cloudy conditions with a concomitant reduction in transpiration by 13%. Vegetative growth was affected in different ways: shaded trees showed smaller trunk diameter, but increased the number and length of their 1-year shoots under coloured hailnets, particularly with cv. ‘Fuji’ when grown under green–black hailnet. Hailnets reduced flower induction in June and return bloom in the next spring to the same extent as they reduced the light transmission. Overall, tree growth under coloured hailnets was genetically influenced, with cv. ‘Fuji’ being more prone and sensitive to adverse effects of coloured hailnets than cv. ‘Pinova’, but is also influenced by the environment. Gradients of the electrostatic potential and electron density of methyl fluoride are displayed as coloured regions on isopotential surfaces. Shading is used to achieve 3D perception and also to encode the direction of the gradient. Colouring the density by the magnitude of the gradient is demonstrated to provide an intrinsic mechanism for colouring the molecular density, so that the underlying atomic structure is vividly displayed in hues commonly used in molecular ball and stick models. The algorithm for contouring a 4D object and drawing coloured, shaded isopotential surfaces is described.  相似文献

This paper investigates how the pattern influences the discrimination of different locations of two or more areas of black, white or colour. The coloured patterns were made from two calibrated coloured papers that give contrast only to green receptors, or alternatively only to blue receptors. The patterns are fixed during training. It is found that the discrimination of translocation of two areas of colour involves green receptors and also blue receptors, and the resolution depends strongly on the pattern. Patterns that offer horizontal strips and up-down differences in locations are well resolved, even with no green contrast. Resolution of left-right reversal is greatly improved when the patterns promote fixation in the horizontal plane, as if green contrast is essential to stabilize the eye in yaw. The addition of radial bars with green contrast, a central black spot or a black surround, is particularly effective. The additions promote fixation, and would aid the detection of natural symmetrical objects. Accepted: 30 May 1999  相似文献

Individual white and black mollies Poecilia latipinna spent significantly more time near the larger group when given the choice between two shoals of similar colouration to themselves. When given the choice between a large and a small shoal of dissimilar colouration to themselves, black test fish spent significantly more time with the larger shoal while white test fish showed no preference for either group. Both white and black mollies chose the smaller of two shoals when given the choice between a large dissimilarly coloured shoal and a small similarly coloured shoal. The results indicate that mollies actively discriminate between shoals on the basis of both body colour and shoal size. However, body colour segregation appears to have a stronger influence on shoal choice.  相似文献

Summary A method of classification is presented, which divides histochemical visualization reactions into categories based on general reaction types. This scheme is dependent upon the reaction between two elements, the substrate and the probe. The substrate represents a tissue component(s) with one or more reactive groups that can combine directly with the probe. Alternatively, the substrate reactive groups are chemically modified (activation) with a suitable reagent before reaction with the probe. Probes are of three types: those that yield a coloured product, those that result in a colourless product, and those that produce a coloured product only after a further reaction.Methods used in carbohydrate histochemistry are divided into one, two and three probe reactions. Two probe reactions are further subdivided into sequences involving one or two coloured products (one and two dye sequences); three probe reactions into sequences involving one, two or three coloured products (one, two and three dye sequences). This classification permits the rationalization and organization of methods, and provides a framework for the examination of existing methods and the development of new ones.  相似文献

Fragmentarily preserved shells – mainly pro-ostraca, in several cases also phragmocones – occurring together with arm hooks and the ink sac of the Carnian (Late Triassic) coleoid cephalopod Phragmoteuthis bisinuata (Bronn) from Lunz (Austria) are examined with the scanning electron microscope and energy-dispersive spectrometer. The pro-ostracum bears black, shiny, pitch-like sheets. The black sheets, the ink sac content and the arm hooks have a granular ultrastructure of 0.1–1 μm grain size. The arm hooks and black sheets are micro-laminated; each lamina consists of fibres. The ink consists of an agglomerate of grains. On the ventral (internal) side of the pro-ostracum, the black sheets occasionally bear agglomerates of homogeneous, ink-like material along with heterogeneous structures. The pro-ostracum has crystal-shaped units with lamello-columnar ultrastructure of the inner layer and plate ultrastructure of the outer layer. The black sheets, the material on their inner surface, the ink and the arm hooks consist of carbon, occasionally with minor amounts of sulfur. The shell is of calcium carbonate.Based on their organic composition, position in the shell and lamello-fibrillar ultrastructure, the black sheets are considered to be remains of the mantle, sometimes with ink sac and soft body debris. The carbon composition and granular ultrastructure of arm hooks, ink, and soft tissue remains indicate that the non-mineralized structures are pseudomorphosed by carbon (carbonization), possibly due to C-accumulating bacteria.  相似文献

The success of printing technology in the electronics industry primarily depends on the availability of metal printing ink. Various types of commercially available metal ink are widely used in different industries such as the solar cell, radio frequency identification (RFID) and light emitting diode (LED) industries, with limited usage in semiconductor packaging. The use of printed ink in semiconductor IC packaging is limited by several factors such as poor electrical performance and mechanical strength. Poor adhesion of the printed metal track to the epoxy molding compound is another critical factor that has caused a decline in interest in the application of printing technology to the semiconductor industry. In this study, two different groups of adhesion promoters, based on metal and polymer groups, were used to promote adhesion between the printed ink and the epoxy molding substrate. The experimental data show that silver ink with a metal oxide adhesion promoter adheres better than silver ink with a polymer adhesion promoter. This result can be explained by the hydroxyl bonding between the metal oxide promoter and the silane grouping agent on the epoxy substrate, which contributes a greater adhesion strength compared to the polymer adhesion promoter. Hypotheses of the physical and chemical functions of both adhesion promoters are described in detail.  相似文献

Feathers confer protection against biophysical agents and determine flying ability. The geometry and arrangement of the barbs, together with the keratin and pigments deposited in the feathers, determine the mechanical stability of the vane, and its stiffness and resistance to abrasive agents. In colour‐polymorphic species, individuals display alternative colour morphs, which can be associated with different foraging strategies. Each morph may therefore require specific flying abilities, and their feathers may be exposed to different abrasive agents. Feathers of differently coloured individuals may thus have a specific structure, and colour pigments may help resist abrasive agents and improve stiffness. We examined these predictions in the barn owl (Tyto alba), a species for which the ventral body side varies from white to dark reddish pheomelanic, and in the number and size of black spots located at the tip of the feathers. White and reddish birds show different foraging strategies, and the size of black feather spots is associated with several phenotypic attributes. We found that birds displaying a darker reddish coloration on the ventral body side deposit more melanin pigments in their remiges, which also have fewer barbs. This suggests that wear resistance increases with darkness, whereas feathers of lighter coloured birds may bend less easily. Accordingly, individuals displaying a lighter reddish coloration on the ventral body side, and those displaying larger black spots, displayed more black transverse bars on their remiges: as larger‐spotted individuals are heavier and longer‐winged birds also have more transverse bars, these bars may reduce feather bending when flying.
