The aims of this study were to characterise the composition of five seaweed species (Ascophyllum nodosum, Fucus serratus, Fucus vesiculosus, Laminaria hyperborea and Sargassum muticum), their extracts and commercial formulations, using thermogravimetry (TGA), energy dispersive X-ray microanalysis (EDX), Fourier-transform infrared spectroscopy (FTIR) and pyrolysis gas chromatography/mass spectrometry (Py-GC/MS). Analyses of the samples by TGA and EDX provided information on the proportions of algal cell wall, inorganic fractions and minerals. The main carbohydrate constituents of the five species and extracts were identified by their pyrolysis products, e.g. 1-(2-furanyl) ethanone, 5-methyl-2-furcarboxaldehyde, 2-hydroxy-3-methyl-2-cyclopenten-1-one, diannhydromannitol, 1,6-anhydromannopyranose and 1,6-anhydromannofuranose, using Py-GC/MS. The differences in relative intensities of the infrared bands of the five species were enhanced, especially after acid extraction compared with alkaline or neutral treatments, resulting in improved understanding of the compositional changes. In addition four commercial formulations and two acidic extracts of A. nodosum were evaluated for composition using the techniques. The dry matter, pH, electrical conductivity, ash, carbon and nitrogen content of the six preparations showed significant differences in composition. Variations in fatty acid, alginic acid, mannitol, laminarin and fucoidan content of the six formulations were reported. The results have shown that TGA, EDX, Py-GC/MS and FTIR are complementary techniques for rapid evaluation of seaweed materials and products.  相似文献

Methods for estimating the absolute polyphenol content of the brown algae Ascophyllum nodosum (L.) Le Jol. and Fucus vesiculosus (L.) have been developed and tested. Polyphenols were extracted almost quantitatively from Ascophyllum nodosum using aqueous acetone, whereas this procedure was somewhat less efficient with Fucus vesiculosus. Colorimetric methods based on the Folin-Denis reagent, Brentamine Fast Red 2G Salt, and vanillin-H2SO4 were applied to acetone-free extracts for determination of polyphenol content relative to suitable reference compounds. Gravimetric methods based on hide powder and on haemoglobin were employed to derive ‘estimation factors’ (EFs) which allow calculation of the absolute polyphenol content from the relative polyphenol content. The values calculated for absolute polyphenol content are considered to be reasonably accurate, despite imprecisions in the methods and despite often large standard deviations, and re-emphasize the potential physiological and ecological significance of brown algal polyphenols. Although the precise EFs calculated here are not valid for other brown algae, the methods are considered to be generally applicable to other Phaeophyceae.  相似文献

The vertical zonation of the flat periwinkles Littorina obtusata (L.) and L. mariae Sacchi et Rastelli is different, but overlaps. At a sheltered location on the Isle of Cumbrae, Scotland, L. obtusata, although most abundant on Fucus vesiculosus L. and Ascophyllum nodosum (L.) Le Jol. on the midshore, ranged over the entire shore from the Pelvetia zone down to the upper limits of Laminaria. Littorina mariae exhibited a much more restricted distribution being associated predominantly with Fucus serratus L. on the lower shore. In laboratory trials with adult plants, flat periwinkles found fucoid algae both more attractive and more palatable than other seaweeds. Littorina obtusata also preferred the reproductive receptacles of fucoid algae to their vegetative tissue. Tiny germlings of a diversity of seaweeds were readily consumed by L. obtusata. In choice experiments germling Ulva lactuca L. was preferred to adult U. lactuca, whereas adult Fucus serratus was preferred to germling F. serratus. Contrasting foraging behaviours between different species of flat periwinkle are discussed in relation to differences in their radular dentition.  相似文献

The content of extractable polyphenols in the brown algae Ascophyllum nodosum (L.) Le Jol. and Fucus vesiculosus (L.) was measured at ≈28-day intervals for one year. Colorimetric methods based on the Folin-Denis, Brentamine, and vanillin-H2SO4 reactions were used to estimate relative contents of polyphenols, and these values were converted to absolute contents using the gravimetric method introduced earlier. Relatively little error was introduced by variations in the qualitative composition of the extracted polyphloroglucinols.There appeared to be a significant temporal correlation between polyphenol content and the reproductive state of the algae. The content of polyphenols in A. nodosum was at a minimum (≈9–10% of dry matter) during the period of maximum fruit body shedding (late May), and reached a maximum (≈12–14% of dry matter) during the ‘winter season’. In F. vesiculosus, the minimum (≈8–10% of dry matter) was one to two months later, just before the period of maximum fertility, and thereafter rose to a maximum (≈11–13% of dry matter) during the period of sterility. These results furthermore suggest that the bulk of the polyphenols are not readily accessible as reserve components, and indicate that modifications may be needed in the ‘chemical defense’ and ‘waste product’ hypotheses concerning the significance of brown algal polyphenols.  相似文献

Lipid metabolism and environmental effects on this process havebeen studied in the marine brown algae Fucus vesiculosus andAscophyllum nodosum. These algae showed very similar patternsof lipid metabolism during 24 h incubations. Labelling from[1-14C]acetate showed the major labelled lipids to be the ß-alanineether lipid and the neutral lipid fraction in both algae. Ofthe glycolipids, only sulphoquinovosyldiacylglycerol was welllabelled and the phosphoglycerides were all poorly labelled.The major labelled fatty acids were palmitate and oleate, againin both algae, although Fucus vesiculosus also showed significantlabelling of stearate and behenate. Although the amount of fattyacid labelling increased with time, the proportion of labelin palmitate and oleate remained approximately constant. Verylong chain fatty acids (arachidic, behenic) were increasinglylabelled with time. Lowered incubation temperatures decreased labelling of the saturatedfatty acids. Cu2+ increased the proportion of oleate labelledin both algae, and of linoleate in Fucus vesiculosus. This cationdecreased the percentage labelling of stearate and myristatein Ascophyllum nodosum. The fucose-containing, sulfated polysaccharides from Ascophyllum nodosum and Fucus vesiculosus were isolated by extraction with water adjusted to pH 2. Pure fractions were carefully separated by fractional precipitation with ethanol from aqueous solutions containing magnesium or calcium chloride. Progress in the fractionation efforts and purity of the fractions isolated were established by free-boundary and cellulose acetate clectrophoresis. Ascophyllan, two “complexes”, and a galactofucan were isolated from A. nodosum. An ascophyllan-like fraction, and a “complex” were isolated from F. vesiculosus. Mild, acid hydrolysis (0.02m hydrochloric acid, 1 h, 80°) converted each of the “complexes” into an electrophoretically faster-moving and a slower-moving component. The “complex” from F. vesiculosus comprised a greater proportion of the extract than did the two “complexes” from A. nodosum. In addition, the Fucus “complex” was richer in fucose*. Poor correlation exists between levels of total phenols and of ultraviolet-absorbing materials in extracts of the brown algae Ascophyllum nodosum (L.) Le Jol. and Fucus vesiculosus L. The uv extinction sp coefficient of a major exuded polyphenol fraction increases rapidly and markedly in sea-water solution. Consequently, uv spectrophotometry cannot provide a useful measure of either total polyphenols or total organic matter in extracts or exudates from these brown algae. Moreover, considerable errors may be introduced when uv spectrophotometry is used to estimate dissolved organic matter in sea water which receives significant exudation from brown macrophytes.  相似文献

The potential of algae to acclimate to environmental stress is commonly assessed using chlorophyll a fluorescence, with changes in parameters of photosynthesis versus irradiance (P/E) curves measured either as rapid light curves (RLC) or steady-state light curves (LC). Here, effects of emersion on primary photosynthesis of four brown macroalgae (Ascophyllum nodosum, Fucus serratus, Sargassum muticum, Laminaria digitata) were compared by applying both RLC and LC. When LC were used, photosynthetic performance was enhanced during emersion in A. nodosum and F. serratus as shown by increases in qP, rETRmax and E k. By contrast, emersion had no impact on photosynthetic parameters of S. muticum and L. digitata. Relative changes in the NPQ?CrETR relationship were reduced in A. nodosum, F. serratus and S. muticum, but remained unaffected in L. digitata. As none of the species developed their potential NPQmax, corresponding values could not be determined from RLC. Using RLC, observed photosynthetic performance of F. serratus and L. digitata was reduced upon emersion, whilst values for NPQmax were enhanced. Only results derived from LC provide evidence for a potential physiological adaptation of brown macroalgae to their natural habitat; it is recommended using the LC protocol to detect environmental impacts on photosynthesis.  相似文献

The growth rate of five species of intertidal Fucales (Pelvetia canaliculata (L.) Dec. et Thur., Fucus spiralis L., Fucus vesiculosus L., Fucus serratus L., Ascophyllum nodosum (L.) Le Jolis) was measured at temperatures from 2.5 to 35 °C. An increase in temperature immediately causes a high growth rate, and during the first hour it increases linearly with temperature; at 35 °C it is 20 times the control at 7 °C. This acceleration of growth is based mainly on stored photosynthate. After the first few hours the growth rate decreases rapidly, particularly at the highest temperatures. After 2–3 weeks a temperature optimum below 17.5 °C is indicated. High temperatures, 30–35 °C, were lethal to all species, with a survival time corresponding to their vertical zonation in the natural habitat.  相似文献

The nature and seasonal extent of microbial fouling on Ascophyllum nodosum (L.) LeJol., Fucus vesiculosus L., Polysiphonia lanosa (L.) Tandy and Chondrus crispus Stackh. were observed by scanning electron microscopy. Bacterial filaments and smaller rod and coccoid forms dominated the fouling communities on all species, with pennate diatoms constituting a minor component in contrast to results with plastic substrates on which pennate diatoms dominated and preceded bacterial colonization. The total percent microbial coverage on the surfaces of all four seaweed species was determined by monthly stereological analyses of representative composite micrographs. These showed a simultaneous decline between April and May which could represent the die-off of the cold water bacterial flora when water temperature increased past the threshold for obligate psychrophiles. Microbial colonization patterns were directly correlated (P = 0.005) with maximum coverage in April and November–December and reduced levels from May to October. Significant inverse (P < 0.041) correlations between total percent coverage and water temperature indicate distinct seasonal cycles, however, the patterns of dominance by filamentous bacteria and rod and coccoid forms were markedly different. Total coverage patterns of both rhodophytes showed no apparent seasonal cycle and were not related to water temperature. Rod and coccoid bacteria were apparently suppressed year round on P. lanosa relative to the other species. These interspecific differences in seasonal fouling patterns are discussed in light of possible modes of regulation, especially algal antibiosis.  相似文献

Biological interactions affecting densities of settling and newly-settled Semibalanus balanoides (L.) have been investigated by manipulative field experiments on the Isle of Man.The effects of sweeping by fucoid clumps of different species and Patella browsing have been compared on moderately-exposed shores. Patella allowed barnacle settlement by preventing growth of competing green algae, but reduced post-settlement densities. Small clumps of Fucus spiralis L., F. vesiculosus L, and F. serratus L. all reduced settlement considerably more so than limpets. F. serratus had the greatest sweeping effect.Interactions between macroalgae and Semibalanus balanoides have been investigated at all levels on sheltered shores and low down on more exposed shores. In the Fucus spiralis and F. vesiculosus zones, post-settlement numbers were higher than in adjacent areas where the canopy was removed. Barnacles did not settle readily in the Ascophyllum zone in either experimental or control areas. Settlement occurred in the upper part of the Fucus serratus zone in experimental areas where the canopy was removed but not in control areas. No settlement occurred in either treatment or control areas lower in the F. serratus zone. At all levels on the shore fucoid canopies seemed to reduce cyprid settlement, but the effect was greatest amongst F. serratus where there was total prevention. High on the shore the effect of enhanced post-settlement survival under the canopy outweighs reduction of cyprid settlement thus there are greater numbers in the controls. Competition with red algal turfs was shown to set the lower limit of the barnacle zone on a vertical pier face.  相似文献

Year-round studies of photosynthesis and respiration in the fucoid brown algae Ascophyllum nodosum and Fucus vesiculosus in the White Sea were performed. The annual specific production of the macrophytes was determined to be 1314 and 1642 cal/g of wet weight for A. nodosum and F. vesiculosus, respectively. The total annual production of fucoids (4.88 × 1011 kcal) comprised about 0.8% of the phytoplankton production in the White Sea.  相似文献

The increase in length of Pelvetia canaliculata (L.) Dec. et Thur., Fucus spiralis L., F. vesiculosus L., F. serratus L., and Ascophyllym nodosum (L.) Le Jolis was measured in various concentrations of lead, cadmium, and mercury during a period of 9–10 days. Concentration ranges of the three metals were 45 2600 μg 1, 1.5 1040 μg 1 and 0.9 1250 μg 1, respectively.Significant reductions of growth rate compared with the controls were observed at ? 810 μg 1 of lead, ? 450 μg/l of cadmium, and ? 10 μg/l of mercury, and regressions of growth reduction on log concentration were indicated.Growth was significantly enhanced in Pelvetia canaliculata and Ascophyllum nodosum when exposed to cadmium, and in this case there was a significant regression of growth iincrease on log concentration. The growth of Pelvetia canaliculata was also enhanced at all concentrations of lead.  相似文献

Ascophyllum nodosum L. dominates rocky intertidal shores throughout the temperate North Atlantic. Six microsatellite loci were developed for A. nodosum using enriched libraries. The number of alleles ranged from 9 to 24 and heterozygosities from 0.2213 to 0.7785. Ascophyllum is monotypic. There was no cross‐reactivity observed with Fucus serratus, F. vesiculosus or F. evanescens.  相似文献

The Fucaceae is a family of brown seaweeds that dominate and frequently co‐occur on North Atlantic rocky shores. We developed nine polymorphic microsatellite markers for the fucoid seaweeds Fucus vesiculosus, F. serratus and Ascophyllum nodosum using a combined, enriched library. Six of these loci were polymorphic in at least two species, showing from two to eight alleles with heterozygosities ranging from 0.41 to 0.85. Loci were also tested on F. spiralis, revealing five polymorphic microsatellite loci in this species.  相似文献

Colonization and growth of Ascophyllum nodosum (L.) Le Jol. were investigated in two Maine estuaries from 1972 to 1978. Following denudation of intertidal rock, substrata were initially colonized by Fucus vesiculosus L.; eventually, Ascophyllum supplanted Fucus, and became dominant in terms of percentage cover. Ascophyllum settled first and most densely in the low intertidal zone, but its fastest growth occurred in the mid-intertidal zone. Some, but not all, Ascophyllum germlings produced a vesicle within one year of colonization. The mean annual growth of A. nodosum was variable among sites, zones and years.  相似文献

The settlement patterns of spores of Polysiphonia lanosa (L.) Tandy on Ascophyllum nodosum (L.) Le Jolis and Fucus vesiculosus L. were studied using a flow tank. Settlement sites were defined as ‘sheltered’ or ‘exposed.’ Surface area calculations revealed non-random settlement on A. nodosum, with higher than expected spore frequencies on the thallus and lateral pits and lower than expected frequencies on the vesicles. Settlement of F. vesiculosus was random and significantly lower than on A. nodosum. On the shore, survival of sporelings from September (post-sporulation) to May (pre-sporulation) was highly non-random on both basiphytes. On A. nodosum, lateral pits ('sheltered') showed the highest survival frequency. Here the proportion of surviving sporelings increased over the study period, whereas the proportion on open thallus area ('exposed') decreased. On F. vesiculosus also preferential survival occurred on ‘sheltered’ sites such as vesicle/thallus interfaces and wounds. Between September and May, all P. lanosa sporelings were lost from ‘exposed’ areas (thallus surface and vesicles). Overall, frequencies of surviving sporelings were much greater on A. nodosum than on F. vesiculosus. These results are discussed with reference to basiphyte morphology, epiphyte removal mechanisms and the survival stratagy of P. lanosa.  相似文献

There is some controversy concerning the presence of steryl glycosides and acylated steryl glycosides in eucaryotic algae. These two classes of sterol compounds were investigated in species belonging to the three major groups of eucaryotic algae: green algae (Ulva gigantea, Cladophora rupestris), brown algae (Fucus vesiculosus, Ascophyllum nodosum), and red algae (Rhodymenia palmata, Porphyridium sp.). All these algae contain both steryl glycosides and acylated steryl glycosides. The sterol components of these compounds vary according to the alga but they are always the same as the free sterols of the alga in question. The most common sugar moiety is glucose. In the acylated steryl glycosides, the fatty acid is mainly palmitic acid. The percentage of these compounds (as a percentage of the total sterol content) is often low.  相似文献

Plant secondary metabolites provide resistance against herbivory if they are capable of decreasing herbivore performance or deterring feeding. Here we test the effects of phlorotannin-containing polar extracts from the brown alga Fucus vesiculosus on the assimilation efficiency and feeding preferences of the herbivorous isopod Idotea baltica. We test the effects separately for each sex of the isopod to test the hypothesis that the digestive efficiencies of the sexes may have diverged. Phlorotannin-containing polar extracts were incorporated into an agar-based artificial food in varying amounts. We also varied the quality of the food, using high-quality food containing animal proteins and low-quality food containing only algal material. The total assimilation efficiency as well as the assimilation efficiencies of carbon and nitrogen decreased with an increasing concentration of phlorotannin-containing extracts. The negative effects on carbon and nitrogen assimilation were higher when extracts were incorporated in the high-quality food. Females were consistently more efficient than males in all assimilation measures; there were no interactions between sex and food quality or phlorotannin concentration, suggesting that the digestive efficiency of the sexes had diverged but that the two sexes responded similarly to phlorotannin enriched artificial food. Although phlorotannin-containing extracts were harmful to performance, they did not deter feeding in preference tests. On the contrary: the isopods preferred the food containing more phlorotannins, suggesting that phlorotannins may act as cues in host recognition. Thus phlorotannins were inefficient deterrents, and I. baltica seems to be adapted to utilizing phlorotannin-containing foods despite their detrimental effects on assimilation. We suggest that F. vesiculosus provides a predation refuge and temporal constancy in availability for I. baltica, both of which select for the ability to utilize it as a food. Phlorotannin-containing polar extracts from F. vesiculosus are still harmful digestibility reducers, and may thereby prevent complete specialization and generate selection for utilization of a wider host range.  相似文献

Wrack (dead, washed-up seaweed and seagrass) buried in soft substrata may increase the organic content and alter the physical structure of sediments. These effects may influence the composition and structure of macrofaunal assemblages in the sediment. Such influences can be expected to vary according to the type and amount of wrack as well as the presence of invasive seaweeds in the wrack. In this study, we deliberately buried different amounts of the invasive species Sargassum muticum in isolation or mixed to the native species Ulva sp. and Fucus vesiculosus, in two intertidal sandflats to test some hypotheses about the response of macrofaunal assemblages. We tested whether (1) diversity of detritus (i.e. different mixtures), and (2) the amount of detritus of S. muticum influenced the composition and the relative abundance of macrofaunal assemblages. We also assessed whether the sediment organic carbon and the biomass of benthic microalgae varied depending on the diversity of detritus and the amount of detritus of S. muticum. Finally, we tested if these effects of wrack were consistent across sites. Results indicated that buried wrack affected the composition and structure of macrofaunal assemblages in short-term (i.e. 4 weeks), but there were no differences depending on detritus diversity or the amount of S. muticum. In addition, sediment organic matter and microalgal biomass were not affected by the addition of wrack. They instead varied greatly among small spatial scales (i.e. plots). Wrack composition or abundance of the invasive species S. muticum played thus a small role in shaping the structure of macrofaunal assemblages or the biomass of benthic microalgae in these intertidal sediments, probably because these sediments are frequently affected by various inputs of organic matter and benthic assemblages are already adapted to organically enriched sediments.  相似文献
