Measurements of photosynthesis, dark respiration, and leaf chlorophyll content were made in the laboratory on both shallow (1 to 5 m) and deep (25 to 33 m) leaves of Cymooceu nodosa (Ucria) Aschers, and Posidonia oceanica (L.) Delile in Malta in April and August. Light saturated photosynthetic rates in Cymodocea were similar in spring (18 μg C cm?2h?1) and summer (25μg Ccm?2h?1) if the 9 C increase in water temperature in summer is taken into account: however, photosynthetic rates in Posidonia were higher in spring than in summer, especially in shallow leaves which fixed ≈ 10 μg C cm?2h?1 in spring but less than half that in summer when rates of carbon accretion were close to compensation point. Levels of irradiance at which photosynthesis was light saturated ( were ≈ 3 mW cm?2 PAR for Cymodocea and 2 mW cm?2 PAR for Posidonia: underwater irradiance at the lower depth limit for these plants (≈33 m) was ≈3 mW cm?2 PAR. corresponding closely to the saturation irradiances. Compensation irradiance for both species was between 0.3 and 0.5 mW cm?2 PAR.Photosynthesis in both species had a temperature optimum at about 30 C (slightly higher in Cymodocea in summer). Dark respiration rates were generally similar in spring and summer, in the region of 3 μg C cm?2 h?1 in Cymodocea and 1.5 to 2 μg C cm?2 h?1 in Posidonia. Increase in dark respiration rates with increased temperature was considerably greater in spring than in summer in both species. Photosynthesis was directly proportional to chlorophyll content in Posidonia in the range encountered (up to 58 μg Chl cm?2) and the summer reduction in photosynthesis was closely correlated with reduction in chlorophyll content. It seems unlikely that environmental factors such as seasonal changes in light intensity, nutrient availability or water temperature were directly responsible for this loss of chlorophyll and it is suggested that this is a manifestation of general leaf senescence, probably induced by daylength changes but possibly enhanced by increased water temperature. Cymodocea showed a similar reduction in chlorophyll content in summer but this was not reflected in reduced photosynthesis. Thus, although Cymodocea may grow rapidly throughout the spring and summer with an overall productivity of 3.6 g C m?2 day?1 in shallow water, the luxuriant growths of Posidonia must develop in the first half of the year when a dense meadow may produce up to 2.1 g C m?2 day?1 in shallow water, declining to ?0.6 g C m?2 day?1 in summer.  相似文献

A novel environmentally friendly octahedrally coordinated 2D polymeric complexes bis(1,5-diaminotetrazole) -dichlorozinc(II) (Zn(DAT)2Cl2) was first designed based on the the crystal data of bis(1,5-diaminotetrazole)- dichlorocopper(II) (Cu(DAT)2Cl2). Density functional theory (DFT) was used to predict the optimized geometries at TPSSTPSS/6-311G(d, p) level. Densities and detonation properties were evaluated using the electron cloud enclosed volume and VLW equation of state (VLW EOS), respectively. Calculation results show that the density of Zn(DAT)2Cl2 (2.117 g?·?cm?1) is a bit more than that of Cu(DAT)2Cl2 (2.106 g?·?cm?1). The calculated high positive heat of formation (HOF) predicts that the stabilities of the title compounds decrease in the order Zn(DAT)2Cl2 > Cu(DAT)2Cl2, which agrees with the result of bond dissociation energies (BDE). Even though they have the same molecule structures, their first scission steps are different. Furthermore, the title two compounds show good detonation velocities and pressures compared with that of bis-(5-nitro-2H-tetrazolato-N 2) tetraamminecobalt(III) perchlorates (BNCP), and they are potential candidates for high-energy-density materials (HEDM).  相似文献

This study investigates aspects of the life history of the polychaete Thoracophelia furcifera on a sandy beach in southern Brazil. Two fixed transects perpendicular to the shoreline in the intertidal zone were sampled fortnightly from May 2008 to April 2009 at low tide. Five T. furcifera samples were collected along each transect and sediment temperature and the salinity of interstitial water were recorded. The material was washed over 0.5- and 0.088-mm sieves, and the width of setiger 8 of each specimen was measured. A total of 5,870 organisms were examined and the estimated parameters of the von Bertalanffy growth curve were L 3.60?mm (Wd8S), K 0.63?year?1, C 0.3 and WP 0.97 (Rn 0.132). Life span was 2.6?years, instantaneous mortality rate Z was 3.8?year?1 and the growth index φ′ 0.91. Mean density ranged from 644.44?±?191.77 to 2,783.33?±?453.64 ind m?2 and mean biomass ranged from 2.52?±?0.55 to 9.52?±?1.83?g?m?2. Recruitment occurred from April to July and ovigerous females were found from June to November. Annual secondary production was 6.582?g?m?2?year?1, mean biomass was 5.638?g?m?2 and turnover rate was 1.167. The high values for density, secondary production and biomass suggest that T. furcifera constitute an important food source. These features of T. furcifera’ life strategy demonstrate the significant role this species plays in ecosystem dynamics.  相似文献

Cutaneous evaporation is the main avenue by which cattle dissipate heat via the involvement of sweat glands and other skin components. The difference in skin morphology between B. indicus and B. taurus has been recognized, as well as differences in their ability to tolerate heat. The objective of this study was to compare skin morphology between B. indicus, B. taurus, and their crossbreds. Skin samples of Sahiwal (B. indicus) (n?=?10, reddish brown skin) and Holstein Friesian (HF) (B. taurus) (n?=?10, black and white skin) and crossbred of HF75% (n?=?10, black and white skin) and HF87.5 % (n?=?10, black and white skin) were biopsied for histological study, followed by measurement of skin components. The results indicated that breed significantly affected sweat gland morphology. The shape of the sweat gland, as indicated by the ratio of length/diameter, in Sahiwal was baggier in shape compared to HF (5.99 and 9.52) while values for crossbreds were intermediate (7.82, 8.45). The density and volume of sweat glands in Sahiwal (1,058 glands/cm2; 1.60 μ3?×?10?6) were higher than in HF (920 glands/cm2; 0.51 μ3x10?6) and crossbreds, both HF 75 % (709 glands/cm2; 0.68 μ3?×?10?6) and HF 87.5 % (691 glands/cm2; 0.61 μ3?×?10?6) respectively. However, capillary surface area was greater for HF (2.07 cm2) compared to Sahiwal (1.79 cm2); accordingly, the lower genetic fraction of HF in crossbred cattle showed less capillary surface area (1.83 and 1.9 cm2 for HF75% and HF87.5 %) (P?<?0.01). Nerve density was not significantly different between Sahiwal and HF but was higher in the crossbred (P?<?0.01) cattle. Moreover, the effect of skin color (black and white) was evaluated and it was found that there was an interaction (P?<?0.01) between breed and skin color on the skin components. This study reveals that there are differences in skin morphology among B. indicus, B. taurus and their crossbreds, with these differences being more or less related to the genetic fraction of HF. This may imply that capability for cutaneous evaporative heat loss and tolerance to heat in crossbred cattle could be related to skin morphology.  相似文献

Aims

X-ray Micro Computed Tomography (CT) enables interactions between roots and soil to be visualised without disturbance. This study examined responses of root growth in three Triticum aestivum L. (wheat) cultivars to different levels of soil compaction (1.1 and 1.5?g?cm?3).

Methods

Seedlings were scanned 2, 5 and 12?days after germination (DAG) and the images were analysed using novel root tracking software, RootViz3D?, to provide accurate visualisation of root architecture. RootViz3D? proved more successful in segmenting roots from the greyscale images than semi-automated segmentation, especially for finer roots, by combining measurements of pixel greyscale values with a probability approach to identify roots.

Results

Root density was greater in soil compacted at 1.5?g?cm?3 than at 1.1?g?cm?3 (P?=?0.04). This effect may have resulted from improved contact between roots and surrounding soil. Root diameter was greater in soil at a high bulk density (P?=?0.006) but overall root length was reduced (P?=?0.20). Soil porosity increased with time (P?<?0.001) in the uncompacted treatment.

Conclusions

RootViz3D? root tracking software in X-ray CT studies provided accurate, non-destructive and automated three dimensional quantification of root systems that has many applications for improving understanding on root-soil interactions.  相似文献

Antiviral effects of acyclic nucleoside phosphonates PMEA, (S)-HPMPC, PMEDAP, and ribavirin on double-stranded DNA Cauliflower mosaic virus (CaMV) were evaluated in Brassica pekinensis plants grown in vitro on liquid medium. A double-antibody sandwich ELISA was used for relative quantification of viral protein and PCR for detection of CaMV nucleic acid in plants. Ribavirin and PMEA had no significant antiviral effect. (S)-HPMPC at concentration 50?mg?l?1 and PMEDAP at concentrations 50 and 12.5?mg?l?1 significantly (P?<?0.05) reduced CaMV concentration in plants within 42?C63?days to levels detectable neither by ELISA nor by PCR. A phytotoxicity experiment resulted in progressive yellowing of leaves and dwarfing in plants cultured 42?days on media with concentrations 12.5, 25 and 50?mg?l?1 of (S)-HPMPC and PMEDAP. Laser-Raman spectra of L-α-dimyristoylphosphatidylcholine (DMPC) liposomes in the spectral range 1000–1200 cm?1 were obtained as a function of temperature from ?80 to +50°C. The triplet found in this spectral region was resolved into Lorentzian components by means of an iterative computer program. The peak intensities, band widths, and band areas of the resolved 1062 cm?1 and 1130 cm?1 bands, assigned to CC stretching vibrations of trans segments, were evaluated as a function of temperature. While the peak intensities of the bands decrease substantially with temperature, the band widths show a considerable increase. The change in band areas is therefore smaller than the change in peak heights. Experiments with all trans carboxylic acids showed that in these compounds the area of the Raman bands at 1062 cm?1 and 1130 cm?1 is proportional to the number of trans bonds. The variation with temperature of the number of trans and gauche bonds in the studied phospholipid is reflected by the change of the area of the 1130 cm?1 Raman band.  相似文献

A series of twenty-two BODIPY compounds were synthesized, containing various meso-phenyl and meso-thienyl groups, and their spectroscopic and structural properties were investigated using both experimental and computational methods. Further functionalization of the BODIPY framework via iodination at the 2,6-pyrrolic positions was explored in order to determine the effect of these heavy atoms on the photophysical and cytotoxicity of the meso-aryl-BODIPYs. BODIPYs bearing meso-thienyl substituents showed the largest red-shifted absorptions and emissions and reduced fluorescence quantum yields. The phototoxicity of the BODIPYs in human carcinoma HEp2 cells depends on both the presence of iodines and the nature of the meso-aryl groups. Six of the eleven 2,6-diiodo-BODIPYs investigated showed at least a sevenfold enhancement in phototoxicity (IC50 = 3.5–28 μM at 1.5 J/cm2) compared with the non-iodinated BODIPYs, while the others showed no cytotoxicity, while their singlet oxygen quantum yields ranged from 0.02 to 0.76. Among the series investigated, BODIPYs 2a and 4a bearing electron-donating meso-dimethoxyphenyl substituents showed the highest phototoxicity and dark/phototoxicity ratio, and are therefore the most promising for application in PDT.  相似文献

Phenolic acids are plant metabolites important in phytotherapy and also in cosmetology. In this study, proliferating shoot and callus cultures of Aronia melanocarpa were established and maintained on Linsmaier and Skoog (L-S) medium containing different levels of α-naphthaleneacetic acid (NAA) and 6-benzyladenine (BA), ranging from 0.1 to 3.0 mg l?1. Methanolic extracts from the biomass of these cultures and from the fruits of soil-grown plants were used to determine the amounts of free phenolic acids and cinnamic acid using the high-performance liquid chromatography (HPLC) method. Out of a total of twelve analyzed compounds, all of the extracts contained four of them: caffeic acid, p-hydroxybenzoic acid, syringic acid, and vanillic acid. Moreover, shoot extracts also contained salicylic acid (o-hydroxybenzoic acid), while callus extracts contained p-coumaric acid. On the other hand, fruit extracts also contained both salicylic acid and p-coumaric acid. The total amount of the analyzed compounds in extracts from both shoot and callus cultures depended on the L-S medium used, and varied between 103.05 and 150.95 mg 100 g?1 dry weight (DW), and between 50.23 and 81.56 mg 100 g?1 DW, respectively. Both types of culture contained higher levels of phenolic acids than the fruit extracts (32.43 mg 100 g?1 DW). In shoot cultures, p-hydroxybenzoic acid and salicylic acid were the predominant metabolites (reaching 55.14 and 78.25 mg 100 g?1 DW, respectively), while in callus cultures, p-hydroxybenzoic acid (25.60 mg 100 g?1 DW) and syringic acid (41.20 mg 100 g?1 DW) were the main compounds. In fruit extracts, salicylic acid (15.60 mg 100 g?1 DW) and p-hydroxybenzoic acid (5.29 mg 100 g?1 DW) were predominant.  相似文献

Impacts of long-term drying and associated vegetation change on anaerobic decomposition, methane production, and pore water composition in peat bogs are poorly documented. To identify some of these impacts, we analyzed peat humification, pore water solutes, in situ and in vitro respiration rates, and Gibbs free energies of methanogenesis in a bog near a drainage ditch established in 1923. We compared drained peat under open bog vegetation and forest with a bog reference site. Drainage and tree growth induced an enrichment in carboxylic, aromatic, and phenolic moieties in the peat. Short-term in vitro respiration rates significantly decreased with humification (R 2?>?0.6, p?<?0.01). Dissolved inorganic carbon (DIC) and CH4 concentrations also attained lower maxima in drained areas. However, near the water table in situ respiration intensified as indicated by steeper increases in DIC and CH4 concentrations than at the reference site, especially under forest. Maximum in situ CO2 production derived from inverse pore water modeling was 10.3?nmol?cm?3?d?1 (forest) and 6.3?nmol?cm-3?d-1 (bog) and was one to two orders of magnitude slower than in vitro anaerobic respiration. In the highly decomposed shallow peats under forest, methane production was suppressed and DOC concentration elevated. Raised H2 concentrations (up to 200?nmol?l?1) and in situ Gibbs free energies of down to ?60?kJ?mol?1?(CH4) suggested an inhibition of hydrogenotrophic methanogenesis by an unidentified factor at these sites. The study documents that several changes in biogeochemical process patterns do occur post-drainage, especially when tree growth is triggered. Most importantly, the establishment of forest on intensely humified peats can lower in situ methane production.  相似文献

It was demonstrated previously that polar and non-polar surface extracts of the brown alga Fucus vesiculosus collected during winter from the Kiel Bight (Germany) inhibited bacterial attachment at natural concentrations. The present study describes the bioassay-guided identification of the active metabolites from the polar fraction. Chromatographic separation on a size-exclusion liquid chromatography column and bioassays identified an active fraction that was further investigated using nuclear magnetic resonance spectroscopy and mass spectrometry. This fraction contained the metabolites dimethylsulphopropionate (DMSP), proline and alanine. DMSP and proline caused the anti-attachment activity. The metabolites were further quantified on the algal surface together with its associated boundary layer. DMSP and proline were detected in the range 0.12–1.08 ng cm?2 and 0.09–0.59 ng cm?2, respectively. These metabolites were tested in the concentration range from 0.1 to 1000 ng cm?2 against the attachment of five bacterial strains isolated from algae and sediment co-occurring with F. vesiculosus. The surface concentrations for 50% inhibition of attachment of these strains were always <0.38 ng cm?2 for DMSP and in four cases <0.1 ng cm?2 for proline, while one strain required 1.66 ng cm?2 of proline for 50% inhibition. Two further bacterial strains that had been directly isolated from F. vesiculosus were also tested, but proved to be the least sensitive. This study shows that DMSP and proline have an ecologically relevant role as surface inhibitors against bacterial attachment on F. vesiculosus.  相似文献

Ulva spp. are used in a wide range of commercial applications, including bioremediation, food, bioenergy, pharmaceuticals, and agriculture. The sulfated polysaccharide ulvan obtained from Ulva spp. is of interest for triggering plant defenses against disease. However, the cultivation of Ulva spp. is still in its infancy. This study verified the feasibility of cultivating Ulva lactuca and Ulva flexuosa at two sites on the tropical Brazilian coast. We investigated the following: (a) methods to induce sporulation, (b) comparison of seeding ropes inoculated in vitro versus seeding at sea over 40 days, (c) production and harvest cycles at 15 and 30 days, (d) growth productivity of U. flexuosa at sea and in outdoor tanks, and (e) comparison of ulvan yields from biomass cultivated in tanks and the sea. High nutrient treatment was the most efficient method to induce sporulation (7,540?±?3,133 spores m?1). Sea-based cultivation of U. flexuosa was only successful at one site. Seeding of ropes in vitro was more efficient than seeding at sea (0.31?±?0.20 g m?2 day?1), and 15-day harvest cycles were most efficient (20.1?±?1.8 % day?1; 0.46?±?0.11 g m?2 day?1). Despite differences in plant growth in tanks (27.9?±?4.4 % day?1) and at sea (20.1?±?1.8 % day?1), the dry biomass and ulvan yields (17.7?±?5.0 %) did not differ between these systems. Cultivation of U. flexuosa was feasible at sea using in vitro seeding with a production cycle of 15 days in Brazilian tropical waters and tanks with high irradiance and enriched seawater.  相似文献

In the Yucatan Peninsula coast, a large diversity of seaweed species are found, and recent studies have reported the presence of metabolites with pharmaceutical importance. In this study, a biological screening of brown seaweed extracts from Dictyota ciliolata, Padina sanctae-crucis, Sargassum fluitans, and Turbinaria tricostata was carried out. Their cytotoxicity and antiproliferative activities were evaluated by the sulforhodamine B assay on human embryonic kidney (HEK 293), human breast cancer (MCF-7), human prostate cancer (LNCaP), and human hepatic cancer (Hep-G2) cell lines. Seaweed extracts were also tested for their anti-trichomonal (Trichomonas vaginalis) and anti-giardicidal (Giardia lamblia) properties. Fucan fractions were extracted using successive maceration with ethanol/water and freeze-dried. Organics extracts were obtained from ethanol residue from liquid–liquid fractionation. A total of four ethanol extracts, four fucan-rich fractions, four ethanolic extracts, and 12 organic fractions were obtained. Only the ethanolic extracts from Turbinaria tricostata and D. ciliolata were active against LNCaP (CC50 of 24.4 and 29.3 μg mL?1, respectively). Interestingly, the activity found in the extracts from D. ciliolata and Turbinaria tricostata was maintained when both extracts were subjected to a liquid–liquid fractionation with hexane on the LNCaP cell line (CC50 of 24.4 and 25.2 μg mL?1, respectively). The antiproliferative assays showed that both dichloromethane and ethanolic fractions from P. sanctae-crucis were active against MCF-7, with IC50 of 26.1 and 29.8 μg mL?1, respectively. The objective of this study was to identify loci associated with tail biting or being a victim of tail biting in Norwegian crossbred pigs using a genome-wide association study with PLINK case?Ccontrol analysis. DNA was extracted from hair or blood samples collected from 98 trios of crossbred pigs located across Norway. Each trio came from the same pen and consisted of one pig observed to initiate tail biting, one pig which was the victim of tail biting and a control pig which was not involved in either behaviour. DNA was genotyped using the Illumina PorcineSNP60 BeadChip whole-genome single-nucleotide polymorphism (SNP) assay. After quality assurance filtering, 53,952 SNPs remained comprising 74 animals (37 pairs) for the tail biter versus control comparison and 53,419 SNPs remained comprising 80 animals (40 pairs) for the victim of tail biting versus control comparison. An association with being a tail biter was observed on Sus scrofa chromosome 16 (SSC16; p?=?1.6?×?10?5) and an unassigned chromosome (p?=?3.9?×?10?5). An association with being the victim of tail biting was observed on Sus scrofa chromosomes 1 (SSC1; p?=?4.7?×?10?5), 9 (SSC9; p?=?3.9?×?10?5), 18 (SSC18; p?=?7?×?10?5 for 9,602,511?bp, p?=?3.4?×?10?5 for 9,653,881?bp and p?=?5.3?×?10?5 for 29,577,783 bp) and an unassigned chromosome (p?=?6.1?×?10?5). An r 2?=?0.96 and a D???=?1 between the two SNPs at 9?Mb on SSC18 indicated extremely high linkage disequilibrium, suggesting that these two markers represent a single locus. These results provide evidence of a moderate genetic association between the propensity to participate in tail-biting behaviour and the likelihood of becoming a victim of this behaviour.  相似文献

Rubredoxins contain a mononuclear iron tetrahedrally coordinated by four cysteinyl sulfurs. We have studied the wild-type protein from Clostridium pasteurianum and two mutated forms, C9S and C42S, in the oxidized and reduced states, with Mössbauer, integer-spin EPR, and magnetic circular dichroism (MCD) spectroscopies. The Mössbauer spectra of the ferric C42S and C9S mutant forms yielded zero-field splittings, D=1.2?cm?1, that are about 40% smaller than the D-value of the wild-type protein. The 57Fe hyperfine coupling constants were found to be ca. 8% larger than those of the wild-type proteins. The present study also revealed that the ferric wild-type protein has δ=0.24±0.01?mm/s at 4.2?K rather than δ=0.32?mm/s as reported in the literature. The Mössbauer spectra of both dithionite-reduced mutant proteins revealed the presence of two ferrous forms, A and B. These forms have isomer shifts δ=0.79?mm/s at 4.2?K, consistent with tetrahedral Fe2+(Cys)3(O-R) coordination. The zero-field splittings of the two forms differ substantially; we found D=?7±1?cm?1, E/D=0.09 for form A and D=+6.2±1.3?cm?1, E/D=0.15 for form B. Form A exhibits a well-defined integer-spin EPR signal; from studies at X- and Q-band we obtained g z =2.08±0.01, which is the first measured g-value for any ferrous rubredoxin. It is known from X-ray crystallographic studies that ferric C42S rubredoxin is coordinated by a serine oxygen. We achieved 75% reduction of C42S rubredoxin by irradiating an oxidized sample at 77?K with synchrotron X-rays; the radiolytic reduction produced exclusively form A, suggesting that this form represents a serine-bound Fe2+ site. Studies in different buffers in the pH?6–9 range showed that the A:B ratios, but not the spectral parameters of A and B, are buffer dependent, but no systematic variation of the ratio of the two forms with pH was observed. The presence of glycerol (30–50% v/v) was found to favor the B form. Previous absorption and circular dichroism studies of reduced wild-type rubredoxin have suggested d-d bands at 7400, 6000, and 3700?cm?1. Our low-temperature MCD measurements place the two high-energy transitions at ca. 5900 and 6300?cm?1; a third d-d transition, if present, must occur with energy lower than 3300?cm?1. The mutant proteins have d-d transitions at slightly lower energy, namely 5730, 6100?cm?1 in form A and 5350, 6380?cm?1 in form B.  相似文献

The synthesis and characterisation of a series of dinuclear and polynuclear coordination compounds with 4-allyl-1,2,4-triazole are described. Dinuclear compounds were obtained for Mn(II) and Fe(II) with composition [M2(Altrz)5(NCS)4], and for Co(II) and Ni(II) with composition [M2(Altrz)4(H2O)(NCS)4](H2O)2. The crystal structure of [Co2(Altrz)4(H2O)(NCS)4](H2O)2 was solved at room temperature. It crystallizes in the monoclinic space group P21/n. The lattice constants are a = 18.033(3) Å, b = 13.611(2) Å, c = 15.619(3) Å, β = 92.04(2)° Z = 4. One cobalt ion has an octahedrally arranged donor set of ligands consisting of three vicinal nitrogens of 1,2-bridging triazoles (CoN = 2.14–2.15 Å), one terminal triazole nitrogen (CoN = 2.12 Å) and two N-bonded NCS anions (CON = 2.08 Å). The other Co(II) ion has the same geometry, but the terminal triazole ligand is replaced by H2O (CoO = 2.15 Å). The crystal structure is stabilised by hydrogen bonding through H2O molecules, S-atoms of the NCS anions and the lone-pair electron of the monodentate triazole. The magnetic exchange in the Mn, Co and Ni compounds is antiferromagnetic with J-values of ?0.4 cm?1, ?10.9 cm?1 and ?8.7 cm?1 respectively. The Co compound was interpreted in terms of an Ising model. For [Zn2(Altrz)5(NCS)2]∞[Zn(NCS)4], [Cu2(Altrz)3(NCS)4]∞ and [Cd2(Altrz)3(NCS)4]∞ chain structures are proposed. The somatic and gonad productions of the cirolanid isopod Excirolana armata were analyzed by taking monthly samples from December 2003 to November 2005 on Una beach, S?o Paulo state (24°S), southeastern Brazil. Sampling was performed along three fixed transects established from the base of the foredunes to the waterline. Weight-specific growth rate was used to estimate the E. armata somatic production for 2004 and 2005, separately. The gonad production was estimated based on the monthly reproductive potential (mean number of eggs/embryos per female × monthly abundance of ovigerous females with near-release broods) for 2004. The annual somatic production of E. armata population varied from 15.57 to 17.25?g AFDW m?1?year?1 and the somatic production/biomass ratio (P s/B) from 3.55 to 3.14?year?1 for 2004 and 2005, respectively. The P s/B ratios were higher for males (4.02 and 3.19?year?1 for 2004 and 2005) than for females (3.10?year?1 for both years). The annual gonad production (P g?=?1.07?g AFDW m?1?year?1) contributed about 15 and 6% to the total production (P s?+?P g) of females and the population, respectively. The proportion of gonad to somatic production of females (P g/P s) increased with individual size (ca 90% in the 7.5?mm size class), and the annual weight-specific gonad production (P g/B ratio) was estimated to 0.24?year?1. The high P s/B ratios estimated for E. armata derive from the fast growth of individuals and show the importance of this population to the energy flow on Una beach ecosystem. However, the low percentage of juveniles verified in this population and in other studies of populations of the genus Excirolana is discussed as an important source of underestimation of P s/B ratio.  相似文献

The synthesis of a number of leucyl derivatives of substituted anilides and their properties as substrates and inhibitors of Zn2+-Mg2+ leucine aminopeptidase (EC 3.4.11.1) at pH 8.5 and 30 °C are described. The compounds include leucyl-X where X is o-, m-, or p-aminobenzenesulfonic acid, o-, m-, or p-anisidine, and m- or p-aminobenzenesulfonyl fluoride. The latter two sulfonyl fluorides, designed to be active site-directed irreversible inhibitors, turned out to be good substrates for leucine aminopeptidase. The Km and V values of the above compounds as substrates for leucine aminopeptidase are reported. N-Leucyl-m-aminobenzenesulfonate exhibits desirable properties (solubility much greater than Km, Δ? at 295 nm of 2000 m?1 cm?1, and V of 300 μmol min?1 mg?1) as a substrate for a spectrophotometric assay of leucine aminopeptidase. With the exception of N-leucyl-p-aminobenzenesulfonate, all of the above compounds are inhibitors of the hydrolysis of leucyl-p-nitroanilide by leucine aminopeptidase with Ki values approximately their Km values when they are used as substrates. Despite wide variability in steric bulk, chemical composition, and electrical charge of the substituted anilides, the Km values of the above compounds vary over a narrow range (0.5 to 4.8 mm), which indicates that the leucyl moiety plays the predominant role in the determination of Km values. Although the Km values of m- substituents are similar to those of o- substituents, the V values for m-substituents are much greater than those for o- substituents, which suggests that o-substituents interfere with the catalytic process. N-Leucyl-p-aminobenzenesulfonate and N-alanyl-p-aminobenzenesulfonate as well as the nonsubstrate p-aminobenzenesulfonate stimulate rather than inhibit the proteolysis of leucyl-p-nitroanilide. The stimulation has no effect on V but lowers the Km for the hydrolysis of leucyl-p-nitroanilide, which is compatible with these compounds' serving as nonessential activators.  相似文献

Insertion of nickel ions into the empty catalytic site of horse liver alcohol dehydrogenase yields an active enzyme with 65% metal substitution and about 12% intrinsic activity. The electronic absorption spectrum is characterized by bands at 357 nm (2900 M?1 cm?1, 407 nm (3500 M?1 cm?1), 505 nm (300 M?1 cm?1), 570 nm (?130 M?1 cm?1), and 680 nm (?80 M?1 cm?1). The absorption and CD spectra are similar to those of nickel(II) azurin and nickel(II) aspartate transcarbamoylase and prove coordination of the nickel(II) ions to sulfur in a distorted tetrahedral coordination geometry. Changes of the spectra upon ligand binding at the metal or conformation changes of the protein induced by coenzyme, or both, indicate alterations of the metal geometry.The chromophoric substrate trans-4-(N, N-dimethylamino)-cinnamaldehyde forms a ternary complex with Ni(II) liver alcohol dehydrogenase and the coenzyme analogue 1,4,5,6-tetrahydronicotinamide-adenine-dinucleotide, stable between pH 6 and 10. The corresponding ternary complex with NADH is only stable at pH > 9.0. The spectral redshifts induced in the substrate are 11 nm larger than those found in the zinc enzyme. We suggest direct coordination of the substrate to the catalytic metal ion which acts as a Lewis acid in both substrate coordination and catalysis.  相似文献
