Transfection of Vibrio cholerae by bacteriophage phi 149 DNA   总被引:3,自引:0,他引:3  
DNA isolated from Choleraphage ø149 of Group IV was infectious when mixed with competent V.cholerae cells. The cells were competent during mid-log phase of growth. The infectivity of phage DNA was destroyed by deoxyribonuclease but not by ribonuclease or pronase. About 5 min is required for the establishment of the DNase resistant state. The dose response curve for transfection suggested that 2 to 3 molecules of DNA are required to produce one infections center. An infectivity of 5 × 104 infectious center per μg of DNA was obtained.  相似文献

Using Electron Paramagnetic Resonance Spectroscopy, Al3+ was shown to produce a dramatic decrease of membrane lipid fluidity on the microorganism Thermoplasmaacidophilum at a pH > 2. The ability of Al3+ to alter lipid fluidity was enhanced with increasing pH (from 3 to 5). At pH 4, 10?2 M Al3+ increased the lower lipid phase transition by 39°C, and a detectable change was observed with AlCl3 concentrations as low as 10?5 M. The ability of Al3+ to increase the lower lipid phase transition temperature of T.acidophilum is the largest of any cation/lipid interaction yet reported.  相似文献

The epididymis of adult rats metabolize 3H-testosterone by experiments in vivo. Thirty minutes after the injection of 100 μCi 3H-testosterone, some 10 per cent of the total radioactivity of the epididymis was found in the water-soluble fraction, whereas 90 per cent was found in the ether soluble fraction (free steroids). The free steroids were examined further and the following androgenic metabolites identified: testosterone (17β-hydroxy-4-androsten-3-one) 8, 9%, androstendipne (4-androstene-3, 17-dione, 2,7%,5α-A-dione (5α-androstane-3, 17-dione) 6,5%, DHT (17β-hydroxy-5α-androstan-3-one) 47, 2%, 3β-diol (5α-androstane-3β, 17β-diol) 4, 4%, 3α-diol (5α-androstane-3α,17β-diol) 20, 8% and androsterone (3α-hydroxy-5α-androstan-3-one) 3,4%. The relative amount of each metabolite is given in per cent of total radioactivity in the ether soluble fraction.  相似文献

Pretreatment of Chang liver cells with N-ethylmaleimide (0.5 or 1 mM) stimulated Na+-independent uptake of leucine at low concentrations (?1 mM). The stimulatory effect of N-ethylmaleimide on the uptake of leucine measured in Na+-replete medium was completely blocked by the addition of b-2-aminobicyclo[2,2,1]heptane-2-carboxylate (5 mM), which shows that the L system participates in the stimulation. The Na+-dependent uptake of glycine was depressed by N-ethylmaleimide pretreatment. The stimulation of the Na+-independent component of leucine uptake continued for at least 30 min after N-ethylmaleimide treatment, while the inhibition of glycine uptake was progressive with time and the Na+-dependent uptake of leucine became depressed later, after the treatment. It has been demonstrated that treatment of cells with N-ethylmaleimide is capable of increasing the Na+-independent influx of leucine and at the same time slightly decreasing the efflux of it. These results suggest that N-ethylmaleimide attacks the Na+-independent system of amino acid transport at the reactive SH groups(s) of relevant protein(s) in favor of specific activation of that system in this cell.  相似文献

A new enzyme, which catalyzes the transfer of a methyl group to tRNA to form 5-methylaminomethyl-2-thiouridylate, was isolated from E.coli by a procedure including affinity chromatography. The purified enzyme was nearly homogeneous upon disc electrophoresis. Using methyl-deficient tRNAGlu of E.coli as substrate, the 5-methylaminomethyl-2-thiouridylate residue synthesized was mostly found in the anticodon loop, showing a coincidence of the modification site invitro with that invivo.  相似文献

An exogenous supply of N6,O2′-dibutyryl cyclic adenosine 3′,5′-monophosphate (DBcAMP) in vivo produces regression of one type of Walker 256 mammary carcinoma cell population (DBcAMP-responsive); a second type of cell population continues to grow despite DBcAMP treatment (DBcAMP-unresponsive). A correlation was found between altered cAMP-binding of the tumor cytosol and DBcAMP-unresponsiveness. It was found that there was: a) a higher apparent dissociation constant (Kd) for cAMP-binding in unresponsive tumor cytosol in vitro, and b) unsaturability of cAMP-binding by unresponsive tumor cytosol in response to elevated cAMP levels in vivo. Cycloheximide abolished the saturation of cAMP binding in vivo as well as tumor regression produced by DBcAMP.  相似文献

The replication defective transducing phage λplac5O29P3 carries a portion of the E.colilac operon in the b2 region of the lambda phage. This lac operon segment contains the lac promoter, the lac operator, and the β-galactosidase z gene, but does not contain the lac repressor i gene. The z gene can be expressed from both the inserted lac promoter and the phage promoter. When E.coli strain 594 (z?, i+) or JC6256 (Δlac) is infected by λplac5O29P3 in the absence of additional cyclic AMP, β-galactosidase synthesis is shown to be expressed from the phage promoter. When 594 (λ+) or JC6256 (λ+) is infected by λplac5O29P3 in the presence of additional cyclic AMP and IPTG, β-galactosidase synthesis is shown to be expressed from the inserted lac promoter.The ability to separate the phage promoter from the inserted lac promoter for β-galactosidase expression will simplify the interpretation whenever λplac5 is used.  相似文献

The cellular distribution of 35S from 35S- thioacetamide was determined in rabbit liver subcellular fractions following its in vivo administration. Of the various fractions isolated, only the nucleolar fraction contained 35S counts that were insoluble in 10% trichloroacetic acid but soluble in trichloroacetic acid if the fraction was treated with trypsin but not RNase or DNase. These results demonstrate that a protein bound form of thioacetamide is present in the nucleolus following in vivo administration of this drug.  相似文献

The epididymis of adult rats metabolizes 3H-testosterone by experiments invitro. After incubation of slices from epididymal tissue for 2 hrs at 37°C, 8% of the total radioactivity was found in the water-soluble fraction, whereas 92% in the ether soluble fraction (free steroids). The free steroids were examined further and the following metabolites identified: testosterone (17β-hydroxy-4-androsten-3-one) 10,4%, androstendione (4-androstene-3,17-dione) 6,2%, 5α-A-dione (5α-androstane-3,17-dione) 7,3%, DHT (17β-hydroxy-5α-androstane-3-one) 39,3%, 3α-diol (5α-androstane-3α,17β-diol) 22,7%, 3β-diol (5α-androstane-3β,17β-diol) 4,6% and androsterone(3α-hydroxy-5α-androstan-17-one) 8,9%. The relative amount of each metabolite is given in per cent of the total radioactivity in the ether soluble fraction. When segments (caput, corpus, cauda) of epididymis were incubated in the same way, differences in steroid metabolism were demonstrated. Characteristic for caput epididymidis was high formation of DHT (58,4%) and 3α-diol (23,5%). Corpus epididymidis showed lower formation of DHT (50,6%) and 3α-diol (12,7%), but an approximately 3 times higher formation of 5α-A-dione (12,0%) than caput (3,4%) and cauda (3,5%). Cauda epididymis showed the lowest formation of DHT (38,3%), whereas 3α-diol (29,1%) and androsterone (11,4%) formation were relatively high. The ratio between 17β-hydroxy metabolites (DHT and androstanediols) and 17-keto metabolites were much higher in the caput (8,8) than in the corpus (3,2) and cauda (3,6), indicating a higher 5α-reductase activity in this segment.  相似文献

(22S)-[22-3H1]-Cholesterol was incubated with an adrenocortical preparation and the isolated (22R)-[22-3H1]-22-hydroxycholesterol had a small loss of radioactivity, proving that direct replacement of the hydrogen from the now hydroxylated position occurred.In addition [1-3H1]-4-methylpentanol was isolated, which also had incurred a relatively small loss of its specific activity, thereby excluding (20R)-3β,20-dihydroxycholest-5-en-22-one as an important metabolite in the degradation of cholesterol to pregnenolone by adrenal tissue.  相似文献

Cyclic 3′, 5′-adenosine monophosphate (cAMP) has been identified in the ciliated gill epithelium of the marine mussel Mytilusedulis. In concentrations which stimulate the rate of particle transport by frontal gill cilia, DA and 5HT stimulate levels of cAMP within the gill. The stimulation occurs in as early as 15 sec and is graded from 10?6M to 10?4M. DA plus 5HT is not additive at maximal effective concentrations of both amines. ACH does not mimic the DA or 5HT stimulation of cAMP. Theophylline alone has a weak effect on cAMP levels; however, the effect of theophylline is potentiated in the presence of DA or 5HT. Dibutyryl cAMP produces a gradual stimulation in the rate of particle transport. It is suggested that the dopaminergic and serotonergic excitatory control of particle transport by frontal gill cilia of Mytilusedulis is mediated through a cAMP second messenger system.  相似文献

Phenoxybenzamine was antinociceptive in the mouse tail electrical stimulation assay (ED50, 36.8 mg/kg) with a peak effect at 2 12 hours after subcutaneous injection. Naloxone antagonized this antinociception action of phenoxybenzamine in a dose-related manner. Dose-ratio analysis of naloxone's antagonism of phenoxybenzamine antinociception gave a pA2 value of 6.15, similar to that found for the benzomorphinan mixed agonist-antagonists. This is in agreement with the sodium response ratio found for phenoxybenzamine, 4.3, in in vitro assays of phenoxybenzamine inhibition of 3H-naloxone binding to mouse brain homogenate (5). These findings suggest that phenoxybenzamine binds to the opiate receptor both in vivo as well as in vitro in a manner similar to the mixed agonist-antagonists.  相似文献

Human red cells (RBC) respond to moderate Ca2+-loading with increased ATP consumption and stimulation of glycolytic flux. 1. Ca2+-induced metabolite transitions at different pH-values showed a clearcut crossover at the glyceraldehyde-3-phosphate dehydrogenase/3-phosphoglycerate kinase (GAPDHPGK)-steps. 2. The behavior of glycolytic metabolites in iodoacetate-treated, GAPDH-inhibited, and in phosphoenolpyruvate-loaded RBC ruled out activation of hexokinase, phosphofructokinase and pyruvate kinase. 3. Glycolytic stimulation is linked to Ca2+-extrusion rate and not to the loaded Ca2+. 4. Adenine nucleotides and inorganic phosphate could be ruled out as the connecting link between glycolytic activation and Ca2+-extrusion. 5. NADH oxidation was observed at all pH-values studied when the RBC were incubated either at low or high extracellular potassium. NADH is product-inhibitor of GAPDH. The concentration (34 μM) of thermodynamically free NADH calculated from the GAPDHPGK equilibrium reactants was in the inhibitory range: any decrease in NADH is therefore followed by activation of GAPDH. NADNADH ratio seems to be the connecting link between ATP consuming ion transport and ATP generation by glycolysis.  相似文献

A linear analogue of the luteinizing hormone-releasing hormone, longer than a decapeptide, is described for the first time, which is equivalent in potency to the best known inhibitors of ovulation, and which constitutes an important new lead to the design of inhibitors of even greater potency. At a dosage of 200 μg/rat, the undecapeptide [(1, D-Phe2, D-Trp3, D-Trp6]-LH-RH caused 100% inhibition of ovulation. The related analogues, [(1, D-Phe2, D-Trp3, D-Trp6]-LH-RH and [(Gly-Pro)1, D-Phe2, D-Trp3, D-Trp6]-LH-RH, were less active, invivo. All of these undecapeptides inhibited the action of 0.6 ng/ml of LH-RH by greater than 50% at the very low level of 10 ng/ml.  相似文献

Glutathione S-transferase activity was found in sperm of the rat and DBA2J and C57 BL6J mice. In rat sperm activities with benzo(a)pyrene 4,5-oxide, styrene 7,8-oxide, and 1-chloro-2,4-dinitrobenzene were 0.88, 1.07, and 26.1 nmoles/min/mg protein, respectively. Δ5-3-Ketosteroid isomerase activity of rat sperm was 4.9 nmoles/min/mg protein. These specific glutathione S-transferase and Δ5-3-ketosteroid isomerase activities in sperm represent 0.4–4.1% of rat liver cytosol values. Human semen also contained significant glutathione S-transferase activity. It is postulated that these enzymes could function in the metabolism and detoxification of certain electrophilic xenobiotics, if present in sperm.  相似文献

Cytochromes P-450 and b5 were observed in the microsomal fraction of interstitial tissue of rat testes. Microsomal cytochrome b5 was reduced by the NADH coupled with the activities of Δ5-3β-hydroxysteroid dehydrogenase with Δ54 isomerase through conversion of pregnenolone to progesterone. Activities of NADPH-supported 17α-hydroxylase and C-17-C-20 lyase which converted progesterone to androstenedione were stimulated by either the presence of NADH or the oxidative reaction by the dehydrogenase upon Δ5-3β-hydroxysteroids. Androstenedione production enhanced by the reaction of the dehydrogenase was decreased by addition of the antibody against NADH-cytochrome b5 reductase which was purified from rat hepatic microsomes, suggesting the active participation of cytochrome b5 in the androgen synthesis.  相似文献

A class of non-histone chromatin proteins that were bound tightly to DNA and could not be dissociated from the chromatin by high salt and urea was isolated from HeLa cell nuclei and separated from DNA by DNase digestion. These ‘tight’ proteins retained their ability to bind to single- and double-stranded DNA as assayed by nitrocellulose filter binding. Polyacrylamide gel electrophoresis showed that the most prominent proteins possessed molecular weights of about 60 000 D. In asynchronously growing HeLa cell cultures about 13 of the cell nuclei were immunoreactive to fluorescein-labeled antinucleoside antibodies. The immunoreactive cells were the fraction in S phase. Cycloheximide treatment of the cultures raised the fraction of immunoreactive nuclei to over sol23. Exposure of the fixed cycloheximide-treated cell to tight proteins prior to staining with the antibody reduced the fraction of immunoreactive cells to the normal S phase level. Immuno-reactivity induced by X-irradiation or by the intercalating mutagen hycanthone was also suppressed by tight proteins. Cycloheximide treatment preferentially reduced the cellular content of tight proteins, suggesting that these proteins undergo a metabolic turnover with a half-life of about 5 h.  相似文献

The in vitro and in vivo metabolism of 1,2- 3H-progesterone was studied in estrogen-stimulated and control vaginae of ovariectomized mice. Employing two-dimensional thin-layer chromatography, gas-liquid chromatography and metabolite “trapping” techniques, the major and minor pathways for progesterone metabolism were determined in vitro and shown to involve saturation of the Δ4-double bond to yield 5α-pregnane compounds and reduction of the C20 and C3 ketone groups to form 20α- and 3α- and 3β-hydroxy derivatives, respectively. The quantities of 20β-hydroxy metabolites and 5β-epimers that were detected were considered not to be significant. The major metabolites formed by untreated tissues following in vitro incubation in the presence of both high (10?6M) and low (10?8M) progesterone concentrations were 3α-hydroxy-5α-pregnan-20-one and 5α-pregnane-3,20-dione. Although these two derivatives were also found in sizable quantities in estrogen-treated tissues, a marked increase (5-fold) in the rate of C20 ketone reduction at high progesterone concentrations (10?6M) to yield 20α-hydroxy-4-pregnen-3-one was demonstrated. Following intravaginal administration of 3H-progesterone in vivo, only progesterone and 3α-hydroxy-5α-pregnan-20-one were retained in appreciable quantities through 2 hr, suggesting rapid loss of 20α-hydroxy-4-pregnen-3-one and the 5α-pregnanediols from this tissue under in vivo conditions.  相似文献

The regulation of the in vitro synthesis of the N-terminal portion of the β-galactosidase molecule (α-peptide) has been investigated using DNA fragments of the lactose operon as template. DNA fragments of about 789 base pairs were isolated after endonuclease (Hin II) digestion of either λplac5, λh80dlacps or λh80dlacUV5 phage DNA or DNA from the recombinant plasmid PMC3. The regulation of the expression of these fragments is similar to that observed for the synthesis of β-galactosidase using total phage or plasmid DNA as template, indicating that the regulatory regions on the fragments are intact and functional. Thus, the synthesis of the α-peptide required an inducer due to the presence of lac repressor in the E. coli S-30 extract used. In addition a dependency on adenosine 3′,5′-cyclic monophosphate (cAMP)1 for α-peptide synthesis was obtained with the fragments isolated from λplac5 and λh80dlacps DNAs, whereas little effect of cAMP was seen with the fragment isolated from λh80dlacUV5 phage DNA or PMC3 plasmid DNA containing a UV5 promotor region. However, a significant difference in the effect of guanosine-3′-diphosphate-5′-diphosphate (ppGpp) was observed. With the total phage DNA as template, ppGpp resulted in a 2–4 fold stimulation whereas with the fragment, or PMC3 plasmid DNA, directed synthesis of the α-peptide no significant stimulation by ppGpp was seen.  相似文献

After intravenous administration of 3H-naloxone to rats, particulate bound radioactivity accumulated in the brain is selectively associated with opiate receptor binding sites, providing a means of labeling the opiate receptor in vivo. The regional distribution of 3H-naloxone bound in vivo closely parallels regional differences in opiate receptor binding in vitro with highest levels in the corpus striatum, negligible receptor-associated binding in the cerebellum and intermediate levels in other regions. 3H-Naloxone binding in vivo is saturable with the same total number of binding sites determined in vivo as by in vitro procedures. Nalorphine is markedly more potent than morphine in inhibiting 3H-naloxone binding in vivo and non-opiates are ineffective. The half-life for dissociation of 3H-naloxone bound to particles in vivo is the same as its dissociation rate after binding occurs in vitro, and sodium stabilizes 3H-naloxone bound in vivo from initial rapid dissociation as predicted from the known properties of the opiate receptor in vitro.  相似文献
