A simple chromatographic method to separate the arsenic metabolites in plasma and urine following iv administration of 74As to dogs is described. Plasma and urine samples were chromatographed on a cation-exchange resin column. Inorganic, monomethylated and dimethylated arsenic compounds were eluted with 0.5 n HCl, H2O, and 20% (vv) NH4OH solution respectively. Experiments on two dogs slow ~90% of the 74As in urine present in the form of dimethylarsinic acid 3 days after administration. It is also the predominant species in plasma 2 hr after dosing.  相似文献

In perfused human umbilical arteries both angiotensin I and II induced vasoconstriction with a monophasic response. Angiotensin I and II induced vasoconstrictions at doses ≥10?8M and 10?9 M respectively. Captopril inhibited the angiotensin I response while the angiotensin II receptor blocker Sar1-Ala8 AII inhibited the effect of both angiotensins. PGI2 attenuated the angiotensin II response in a dose dependent pattern. PGE2 and PGF in concentrations below the critical levels for creating pressure responses per se, also attenuated the angiotensin II response. The cyclooxygenase inhibitor indomethacin potentiated the angiotensin II response indicating that endogenous production of prostanoids is of importance in the modulation of angiotensin effects.  相似文献

In this study we examined the conformation and side chain environments of angiotensins I, II, III, and [Sar1-Ile5-Ala8]angiotensin II using laser Raman spectroscopy. The positions of the amide I bands for all four peptides were found between 1664 and 1673 cm?1. D2O exchange studies confirmed the positions of the amide I and amide III bands. The positions of the amide I bands for all the angiotensins were found at approximately 1665 cm?1 and the amide III bands were all located between 1265 and 1278 cm?1. From the positions and intensities of the amide I and III bands we concluded that all peptides share the same overall conformation consisting of β-turn structure. Spectral analysis indicated that although the spectra for all the peptides were qualitatively identical there was evidence that the angiotensin conformations were more flexible in the aqueous phase than the solid phase. Examination of the 850830 cm?1 tyrosine doublet suggested that the tyrosine residue in the peptides is exposed to the solvent environment and becomes more exposed as the peptide length is decreased. Therefore, there are some localized conformational differences among the angiotensins. The conformational data yielded by this study leads us to conclude that the various biological properties ascribed to the angiotensins are not due to different conformations of the peptides. The biological differences could perhaps be attributed to localized interactions of the individual amino acid residues with themselves and with the hormone receptors.  相似文献

Two nitroxide radicals (TEMPO, I; OXAN, II) and a spin labeled penicillin (III) were reduced by Staphylococcus aureus. A short induction period preceded zero order reduction of these substrates leading to a Km of 8 × 10?4M, 6.67 × 10?5M and 5.7 × 10?4M and Vmax of 106, 26 and 11 μ mole/min mg bacteria for I, II and III, respectively.  相似文献

The present study describes an improved method for measuring angiotensin III in arterial blood. This was accomplished by SE-sephadex column to separate angiotensin II from angiotensin III prior to radioimmunoassay. The arterial concentration of angiotensin III measured before and after 24 to 48 hours sodium depletion by acute cannulation of parotid gland was 12.4 ± 1.7 fmol/ml (SEM, n=7) and 49.8 ± 10.3 fmol/ml (SEM, n=7) respectively. The arterial concentration of Val4-angiotensin III obtained from continuous infusion of Val4-angiotensin III at rates of 24 and 48 nmol/h in sodium deficient sheep were 245 ± 32.5 fmol/ml (n=6) and 330 ± 11.4 fmol/ ml (n=7) respectively. The clearance rate of exogenous Val4-angiotensin III in sodium deficient sheep after correction for endogenous level was calculated to be 140 ± 13.6 L/h (SEM, n=13). This was in the same order as Ile5-angiotensin II and Ile4-angiotensin III reported earlier in sodium replete sheep. Prolonged intravenous infusion of Val4-angiotensin III at a rate of 48 nmol/h in sodium- deficient sheep suppressed plasma renin concentration to the same extent as equimolar infusions of angiotensin II. This suggests that angiotensin III may inhibit renin secretion by a similar mechanism to angiotensin II.  相似文献

Angiotensin II receptors in bovine adrenal cortex were solubilized with 1% digitonin solution. Binding of 3H-angiotensin II to the solubilized receptors could be assayed by gel filtration on Sephadex G-50 column. Scatchard analysis indicated two classes of binding sites with Kd of 15 and 170 nM. Maximal number of binding sites were estimated at approximately 120 and 470 fmole/mg protein for the high and low affinity binding sites respectively. Pharmacologically active angiotensin II analogues including angiotensin II, Sar1-Ile8-angiotensin II, desAsp1-angiotensin II, desAsp1-Ile8-angiotensin II were all active in inhibiting the specific 3H-angiotensin II binding with relative affinities similar to those in membrane preparations. The inactive angiotensin II precursor, angiotensin I was much weaker in inhibiting the specific 3H-angiotensin II binding thus indicating the specificity of angiotensin II receptors in the solubilized state was maintained.  相似文献

We have used [125I] angiotensin II to investigate the presence of specific angiotensin II receptors in beef heart sarcolemmal membranes. The observed binding is saturable, reversible and specific. The apparent equilibrium dissociation constant is 2.23 ± 0.15 (x ± SEM) and the maximal number of binding sites per mg membrane protein is 32.8 ± 5.4 fmol (x ± SEM). The specific binding is 80–100% of the total [125I] angiotensin II bound and is directly proportional to membrane protein concentration over the range of 33–173 μg protein per ml. Angiotensin II and its antagonists competed for binding in a potency order of (agent, Ki): angiotensin II, 0.9nM > Sar1 Ala3, 7 nM > Sar1-Ile3, 51 nM > Sar1-Leu3, 427nM > angiotensin I, 1709 nM. The ability to characterize and quantify these receptors should now provide a method for investigating the mechanisms underlying the effects of angiotensin II on myocardial tissues.  相似文献

The effects of angiotensin II and angiotensin III on mean arterial pressure, serum aldosterone, and serum corticosterone were studied in normal and sodium depleted, conscious rats. In normal rats, angiotensin III was 76% (p > 0.10) as potent as angiotensin II on aldosterone release but only 31% (p < 0.001) as potent on blood pressure. Following sodium depletion, the pressor responses to both angiotensin II and III were reduced (p < 0.001) (65% and 86% respectively). In addition, the release of aldosterone by both peptides was potentiated by sodium depletion as indicated by an increase in the slope of the dose-response curves. However, in the sodium depleted rats, angiotensin III was only 20% (p < 0.001) as potent as angiotensin II in stimulating aldosterone release. Small changes in serum corticosterone were noted following infusions of both peptides, but unlike the case with aldosterone, sodium depletion did not alter the serum corticosterone responses to the peptides. These invivo experiments taken with invitro studies support the interpretation that angiotensin III could function to control aldosterone release in altered sodium states either as a circulating hormone if present in concentrations far in excess of those of angiotensin II or as a local hormone formed in the adrenal from angiotensin II.  相似文献

When des-(Asp1, Arg2, Val3)-angiotensin II was infused iv at rates of 308–5,550 pmol/kg·min for 10–120 min into 5 normal men and 2 patients with Bartter's syndrome, no significant change was observed in blood pressure (BP), plasma renin activity (PRA) or plasma aldosterone (PA), and the lowest dose did not inhibit a captopril-induced increase in PRA in the normal men, although des-(Asp1, Arg2)-angiotensin II was reported in the same 5 normal men to cause a decrease in PRA and an increase in PA in this dose range and a rise in BP at 2,220 and 5,550 pmol/kg·min. However, an iv infusion of the pentapeptide at 9,000 pmol/kg·min for 15 min significantly raised BP in the 5 normal men but not in patients with Bartter's syndrome. BP returned to the pretreatment level 60 min after the cessation of the infusion, although the duration of the pressor actions of angiotensin II, angiotensin III and des-(Asp1, Arg2)-angiotensin II were reported to be within 5 min in man. At the same dose level none of the 7 examined subjects showed any significant change in PRA or PA. Des-(Asp1, Arg2, Val3, Tyr4)-angiotensin II was infused iv at a rate of 41,480 pmol/kg·min into one of the normal men, but it caused no significant change in BP, PRA or PA. These results suggest that the pentapeptide and probably the tetrapeptide do not possess renin-suppressing and steroidogenic actions in man but the pentapeptide does elecit a minimal pressor action with a prolonged duration.  相似文献

Two additives, glycerol and dimethyl sulfoxide (Me2SO), were investigated for toxic and protective effects for the intraerythrocytic stages of Plasmodium chabaudi. After incubation for 15 min, at 0 °C in Me2SO and at 37 °C in glycerol, with various concentrations of these additives, half the blood from each treatment was cryopreserved in glass capillary tubes cooled at approximately 3600 °C min?1 by plunging into liquid nitrogen. Warming was rapid, approximately 12000 °C min?1, produced by agitation in a water bath at 40 °C for 1 min. The effect of dilution in phosphate-buffered saline (PBS) supplemented with various concentrations (5 to 25% vv) of glucose was also investigated in conjunction with the two cryoprotectants. Survival of both the frozen and the unfrozen control parasites was assayed by the mean time taken for the parasitemia in groups of five mice to reach a level of 2% following intraperitoneal injection of 106 parasitized erythrocytes into each mouse. Glycerol was toxic at concentrations above 10% vv and Me2SO above approximately 15%. The use of glucose in the recovery medium resulted in a substantial improvement in the survival of frozen and unfrozen parasites previously incubated in either cryoprotectant. The amount of glucose required varied with the concentration of additive used, and optimum survival of cryopreserved parasites was obtaind with 10% vv glycerol or 15% vv Me2SO and with 15% wv glucose in the diluent medium.  相似文献

Isolation of ACTH1-39,ACTH1-38 and CLIP from the calf anterior pituitary   总被引:2,自引:0,他引:2  
Calf anterior pituitaries were defatted and homogenized and peptides were adsorbed from the homogenate supernatant onto octadecylsilyl-silica. After elution, the resulting extract was subjected to gradient elution reversed-phase high pressure liquid chromatography (RP-HPLC) using aqueous acetonitrile containing 0.1% (vv) trifluoroacetic acid (TFA). Radioimmunoassay of column fractions for corticotropin (ACTH) revealed three major areas of immunoreactivity. Each was purified to homogeneity by gradient elution RP-HPLC employing aqueous acetonitrile containing either 0.13% heptafluorobutyric acid (vv) or 0.1% TFA (vv). Amino acid analysis and exopeptidase and trypsin digestions revealed the three forms of corticotropin to be ACTH1–38, corticotropin-like intermediary lobe peptide, (CLIP, ACTH18–39) and ACTH1–39. 3H-labeled ACTH1–39 did not give rise to either 3H-ACTH1–38 or 3H-CLIP during isolation.  相似文献

1H nuclear magnetic resonance spectra of 1 - (II) and 3-deazaadenosines (III) together with adenosine (I) in dimethylsulfoxide have been examined. Features of coupling constants indicate that the furanose rings of I, II, and III have similar conformational preferences and that conformations about the 4′-C–5′-C bond are preferentially gauche-gauche. Nuclear Overhauser effect and spin-lattice relaxation-time measurements demonstrate that II predominantly adopts the syn-conformation similar to that of I, whereas that of III has a greater anti (freely rotating) component. The results suggest that the syn-conformation in II as well as I is stabilized presumably through a hydrogen bond between the 3-N and 5′-hydroxyl group.  相似文献

A fluorescent analog of angiotensin II was synthesized by reacting fluorescein 5′-isothiocyanate with (Asp1, Ile5)-angiotensin II. Nα-(N-Fluoresceinthiocarbamoyl)-(Asp1, Ile5)-angiotensin II was purified by chromatography on DEAE-cellulose and Sephadex G-25. Analysis of the analog by thin-layer chromatography, thin-layer electrophoresis, and reversed-phase high-performance liquid chromatography indicated that the analog was free of angiotensin II and fluorescein 5′-isothiocyanate. N-Terminal sequence analysis demonstrated that fluorescein 5′-isothiocyanate reacted with the N-terminal aspartic acid residue of angiotensin II. Nα-(N-Fluoresceinthiocarbamoyl)-(Asp1, Ile5)-angiotensin II has an absorption maximum at 492 nm, and the value of the molar extinction coefficient, ?, is 7.7 × 104m?1 cm?1. The fluorescence emission maximum occurs at 520 nm. Infusion of the analog (0.69 μg/min/kg body wt) directly into the renal artery of an anesthetized rat reduced the blood flow by 12 to 27% within 2 min. Infusion of angiotensin II (0.48 μg/min/kg body wt) reduced renal arterial blood flow by 35 to 53% within 2 min. Saralasin, a partial agonist and antagonist of angiotensin II, inhibited the biologic effect of the fluorescent analog and angiotensin II by 75 and 70%, respectively. The purity, spectral properties, and in vivo biologic activity of Nα-(N-fluoresceinthiocarbamoyl)-(Asp1, Ile5)-angiotensin II indicate that this analog should facilitate characterization of angiotensin II receptors.  相似文献

The interactions of transcobalamin II (TC II), intrinsic factor (IF) and R-type binding protein of cobalamin (Cb1, vitamin B12) with the hydrophobic chromatography matrix Phenyl-Sepharose CL-4B were investigated. IF-Cb1 and R-Cb1 complexes were not adsorbed on Phenyl-Sepharose at room temperature or at 4°C with buffer containing 50 mM sodium phosphate, pH 7.4 containing 150 mM sodium chloride. The TC II-Cb1 complex adsorbed and could be eluted with buffer containing 50% vv glycerol. IF without Cb1 adsorbed and was eluted with 50% glycerol at room temperature and 4°C. At room temperature, R binder without Cb1 eluted with buffer, but later than the R-Cb1 complex. At 4°C, R binder completely adsorbed to the matrix. TC II-without Cb1 bound to the matrix at 4°C and room temperature and could not be eluted with glycerol. These results suggest that Cb1 binding proteins can be separated and identified based on their hydrophobic properties. In addition, upon binding Cb1, TC II, IF and R-type binders undergo a conformational change such that the protein-Cb1 complex shows reduced hydrophobicity.  相似文献

The effect of Mg2+ concentration and phosphorylation of light-harvesting chlorophyll ab-protein on various chlorophyll fluorescence induction parameters of isolated pea thylakoids has been studied. (1) Lowering the Mg2+ concentration from 3 to 0.4 mM decreases only the variable fluorescence (Fv) and the area above the induction curve while at the same time increasing the slow exponential component of the rise (βmax). (2) A further decrease in Mg2+ concentration from 0.4 to 0 mM decreases the initial (F0) fluorescence level such that the ratio FvFm increases slightly as does the area above the induction curve and βmax. (3) Thylakoid membranes, phosphorylated at 5 mM Mg2+, show an equal decrease in Fv and F0, no change in the area above the induction curve and an increase in βmax. At 2 mM Mg2+, however, phosphorylation induced a more extensive quenching of Fv so that the FvFm ratio was lowered and the area above the induction curve decreased while βmax increased. (4) When phosphorylated membranes were subsequently suspended in an Mg2+-free medium the effect on F0 due to phosphorylation was found to be additive to that due to the absence of Mg2+. The effect of membrane phosphorylation on fluorescence is discussed in relation to the control of excitation energy distribution and shows that different mechanisms operate depending on the background Mg2+ levels. At high Mg2+ the phosphorylation seems to affect the absorption cross-section of Photosystem II while at lower Mg2+ levels there is an additional effect of increased spillover from Photosystem II to I.  相似文献

There are five oxidation-reduction states of horseradish peroxidase which are interconvertible. These states are ferrous, ferric, Compound II (ferryl), Compound I (primary compound of peroxidase and H2O2), and Compound III (oxy-ferrous). The presence of heme-linked ionization groups was confirmed in the ferrous enzyme by spectrophotometric and pH stat titration experiments. The values of pK were 5.87 for isoenzyme A and 7.17 for isoenzymes (B + C). The proton was released when the ferrous enzyme was oxidized to the ferric enzyme while the uptake of the proton occurred when the ferrous enzyme reacted with oxygen to form Compound III. The results could be explained by assuming that the heme-linked ionization group is in the vicinity of the sixth ligand and forms a stable hydrogen bond with the ligand.The measurements of uptake and release of protons in various reactions also yielded the following stoichiometries: Ferric peroxidase + H2O2 → Compound I, Compound I + e? + H+ → Compound II, Compound II + e? + H+ → ferric peroxidase, Compound II + H2O2 → Compound III, Compound III + 3e? + 3H+ → ferric peroxidase.Based on the above stoichiometries and assuming the interaction between the sixth ligand and heme-linked ionization group of the protein, it was possible to picture simple models showing structural relations between five oxidation-reduction states of peroxidase. Tentative formulae are as follows: [Pr·Po·Fe-(II) $?PrH+·Po·Fe(II)] is for the ferrous enzyme, Pr·Po·Fe(III)OH2 for the ferric one, Pr·Po·Fe(IV)OH? for Compound II, Pr(OH?)·Po+·Fe(IV)OH? for Compound I, and PrH+·Po·Fe(III)O2? for Compound III, in which Pr stands for protein and Po for porphyrin. And by Fe(IV)OH?, for instance, is meant that OH? is coordinated at the sixth position of the heme iron and the formal oxidation state of the iron is four.  相似文献

Differences in a number of testicular traits were examined in 12 Brahman cross (F2 generation 12 and 34 BX) bulls fed either poor-quality native pasture (NP) hay or NP hay with a protected protein supplement. Supplementation for 60 days significantly (P < 0.05) increased roughage dry matter intake (7.7 v 5.6 kg/head/day), enabling maintenance of liveweight, whereas control animals lost 40 kg. There were significant (P < 0.05) decreases in scrotal circumference (1.5cm) and testicular consistency (0.8 score) in the control group, in which testes weights at slaughter were significantly (P < 0.05) less (373 g v 459 g), with corresponding lower epididymal weights (37.4 g v 43.5 g). Estimates of daily sperm production per gram (DSPG) were similar for both groups, and testis daily sperm production (DSP) was somewhat but not significantly (P>0.05) lower in the control group (4.3 × 109v 6.0 × 109) as a result of lower testis weights. Total epididymal sperm storage capacity was also lower in control bulls (17.2 × 109v 27.0 × 109), but only significantly (P < 0.05) in relation to cauda sperm reserves (8.5 × 109v 13.6 × 109). Luteinizing hormone (LH) and testosterone responses to gonadotrophin releasing hormone (GnRH) treatment were similar for both groups, although LH responses to GnRH were greater in 12 BX than in 34 BX bulls.  相似文献

The purpose of this study was to determine the effects of chronic administration of the thromboxane synthetase inhibitor, UK 38,485, on noradrenergic neurotransmission. Male Sprague Dawley rats (n=14) were treated once daily with either UK 38,485 (100 mg/kg; n=7) or the vehicle of UK 38,485 (olive oil; n=7) by gavage. The dose of UK 38,485 chosen was sufficient to inhibit ex vivo platelet TXB2 production by >90% for 24 hours. One week into the treatment animals were prepared for in situ perfusion of their mesenteric vascular beds. Vasoconstrictor responses to both exogenous norepinephrine and periarterial nerve stimulation were determined both before and during an infusion of angiotensin II (9ng/min) into the superior mesenteric artery. UK 38,485 significantly (P<0.02) attenuated the vascular response to periarterial nerve stimulation without altering the vascular response to either norepinephrine or angiotensin II. UK 38,485 did not influence the baseline perfusion pressure, the mean arterial blood pressure or the potentiation of neurotransmission by angiotensin II. These data indicate that in the in situ rat mesentery UK 38,485 attenuates the release of neurotransmitter from sympathetic nerve terminals.  相似文献

Superior antitumor activity of 1-β-D-arabinofuranosylcytosine (ara-C) conjugates of prednisolone and prednisone against L1210 leukemic mice, based on ara-C content, has encouraged us to synthesize 5′-(cortisone-21-phosphoryl)-1-β-D-arabinofuranosylcytosine (I) and 5′-(cortisone-21-phosphoryl)-1-β-d-arabinofuranosylcytosine (II) by condensation of N4,2′,3′-triacetyl-1-β-d-arabinofuranosylcytosine 5′-monophosphate with cortisol and cortisone in the presence of N,N′-dicyclohexylcarbodiimide at room temperature followed by removing the acetyl groups in 2 N methanolic ammonia in 20% yield. The conjugates I and II inhibited the invitro growth of L1210 by 50% (ED50) at 0.25 μM and 0.07 μM, respectively, while ara-C showed ED50 0.1 μM. However, the conjugates I and II exhibited 287% and 238% of TC at 50 mg/kg/day × 5 doses against L1210 leukemic mice, respectively, while ara-C at 25 mg and 50 mg/kg/day × 5 gave the respective 127% and 110% of TC.  相似文献
