Fourteen thermophilic and thermotolerant fungal strains isolated from composting soils produced plant cell wall-acting esterases in a medium containing corn cobs and oat spelt xylan. The concentrated and dialyzed protein extracts of these fungi were fractionated using isoelectric-focusing, gels sliced and eluted protein in each slice was assayed for esterase activity against p-nitrophenyl acetate. A total of 84 esterases detected on the basis of pI were found to show distinct preferential substrate specificities towards p-nitrophenyl acetate, p-nitrophenyl ferulate and p-nitrophenyl butyrate, and were putatively classified as acetyl esterases and esterases types I and II. None of the esterases were active against p-nitrophenyl myristate. In addition, these esterases were characterized as acid, neutral or alkaline active.  相似文献

A glucose-containing mineral medium supplemented with 0.01% yeast extract is described upon which all the species of thermophilic and thermotolerant fungi tested will grow. Thirteen of the 21 species do not require the yeast extract supplement for growth. Using this solid, supplemented mineral medium, the pH and temperature optima for growth of all strains were measured. No correlation was found between temperature optimum and pH optimum among members of the group tested.  相似文献

Efficient protoplast isolation from fungi using commercial enzymes   总被引:6,自引:0,他引:6  
Several commercial polysaccharases have been compared for their ability to liberate protoplasts from fungi. These enzymes were found to contain side activities capable of hydrolysing fungal cell walls. Protoplasts have been commonly isolated from fungi using enzyme systems prepared by workers in their own laboratories. However, these procedures are time consuming and considerable variation may be found between different batches of enzyme. The present study shows that high yields of protoplasts can be prepared from a variety of fungi using relatively cheap commercial enzymes. The yields obtained were normally as good as or better than those previously produced.  相似文献

The extent to which veterinary pathogenic and human opportunistic species of thermophilic and thermotolerant fungi disseminate in aerosols from heated effluents of nuclear production reactors of the Savannah River Plant (SRP), South Carolina, has been measured. Aerosol samples were taken at 140 sites, from directly over thermal effluents to more than 100 kilometers from the SRP boundary. Sampling methods included settle plates (134 sites, 1, 103 plates for a total of 2, 805 hrs.), liquid impingement (78 sites, 1 cubic meter each), filtration (79 sites, 1 cubic meter each), and a particle sizing cascade impactor (Andersen Sampler) (17 sites, 1 cubic meter each). Soils, foams, and microbial mats from thermal effluents (a total of 191 samples), and vegetation (560 samples) were sampled to study distribution of particular species. Sampling was done under a variety of conditions: hot weather and cold, wet and dry, day and night, windy and calm, reactor(s) operating and not, disturbed vegetation and undisturbed. At 102 of the aerosol sampling sites, sophisticated meterological analyses were used to allow sampling of air in the plume which originated from thermal effluents. Soil, foam, microbial mat, vegetation, and aerosol samples were quantitatively plated for detection of viable units; filters were halved and then both plated and observed microscopically.Significant dissemination of thermophilic and thermotolerant fungi from thermal effluents was not detected. Thermophilic and thermotolerant fungi were widely distributed in soil, air, and on vegetation. Dactylaria gallopava, the indicator species and dominant fungus in microbial mats lining SRP thermal effluents and the cause of epidemic fatal phaeohyphomycosis in flocks of turkeys and chickens in South Carolina, Georgia, and elsewhere, was isolated from air at a maximum of 50 meters from effluents (8 viable units per cubic meter of air in the plume of an effluent during a strong wind).By acceptance of this paper, the publisher and/or recipient acknowledges that the U.S. Government's right to retain a nonexclusive, royalty-free license in and to any copyright covering this paper, along with the right to reproduce and to authorize others to reproduce all or part of the copyrighted paper.  相似文献

Three thermophilic cellulolytic fungi, Chaetomium thermophile var. coprophile, Sporotrichum thermophile, and Thermoascus aurantiacus were studied to determine the conditions for a high rate of cellulose degradation. The range of temperature over which good growth occurred was determined first in a temperature gradient incubator; the optimum temperature was then established in shake flask cultures. T. aurantiacus had the highest optimum growth temperature range (46 to 51 C), whereas S. thermophile had the broadest range over which good growth occurred (36 to 43 C). Optimum temperatures for the three organisms, T. aurantiacus, S. Thermophile, and C. thermophile were 48, 40, and 40 C, respectively. It was found that the addition of an organic carbon and nitrogen source to a cellulose mineral solution medium markedly increased the rate of cellulose degradation. The surfactant, Tween 80, which has been reported to be of value in the production and recovery of the enzyme, cellulase, was shown to be detrimental to the degradation of cellulose in culture. In the medium used, S. thermophile gave the highest rate of substrate utilization; 56% of the cellulose was hydrolyzed in 72 h. The average degree of polymerization of cellulose decreased from 745 to 575.  相似文献

Sequences from 86 fungal genomes and from the two outgroup genomes Arabidopsis thaliana and Drosophila melanogaster were analyzed to construct a robust molecular phylogeny of thermophilic fungi, which are potentially rich sources of industrial enzymes. To provide experimental reference points, growth characteristics of 22 reported thermophilic or thermotolerant fungi, together with eight mesophilic species, were examined at four temperatures: 22 °C, 34 °C, 45 °C, and 55 °C. Based on the relative growth performances, species with a faster growth rate at 45 °C than at 34 °C were classified as thermophilic, and species with better or equally good growth at 34 °C compared to 45 °C as thermotolerant. We examined the phylogenetic relationships of a diverse range of fungi, including thermophilic and thermotolerant species, using concatenated amino acid sequences of marker genes mcm7, rpb1, and rpb2 obtained from genome sequencing projects. To further elucidate the phylogenetic relationships in the thermophile-rich orders Sordariales and Eurotiales, we used nucleotide sequences from the nuclear ribosomal small subunit (SSU), the 5.8S gene with internal transcribed spacers 1 and 2 (ITS 1 and 2), and the ribosomal large subunit (LSU) to include additional species for analysis. These phylogenetic analyses clarified the position of several thermophilic taxa. Thus, Myriococcum thermophilum and Scytalidium thermophilum fall into the Sordariales as members of the Chaetomiaceae, Thermomyces lanuginosus belongs to the Eurotiales, Malbranchea cinnamomea is a member of the Onygenales, and Calcarisporiella thermophila is assigned to the basal fungi close to the Mucorales. The mesophilic alkalophile Acremonium alcalophilum clusters with Verticillium albo-atrum and Verticillium dahliae, placing them in the recently established order Glomerellales. Taken together, these data indicate that the known thermophilic fungi are limited to the Sordariales, Eurotiales, and Onygenales in the Ascomycota and the Mucorales with possibly an additional order harbouring C. thermophila in the basal fungi. No supporting evidence was found for thermophilic species belonging to the Basidiomycota.  相似文献

The thermophilic fungi Thermomyces lanuginosus, Malbranchea cinnamomea, Myceliophthora fergusii and the thermotolerant Aspergillus terreus were cultivated on various carbon sources, and hemicellulolytic and cellulolytic enzyme profiles were evaluated. All fungi could grow on locust bean galactomannan (LBG), Solka floc, wheat bran and pectin, except T. lanuginosus, which failed to utilize LBG for growth. Different levels of cellulase and hemicellulase activities were produced by these fungal strains. Depending on the carbon source, variable ratios of thermostable hydrolytic enzymes were obtained, which may be useful in various applications. All strains were found to secrete xylanolytic and mannanolytic enzymes. Generally, LBG was the most efficient carbon source to induce mannanase activities, although T. lanuginosus was able to produce mannanase only on wheat bran as a carbon source. Xylanolytic activities were usually highest on wheat bran medium, but in contrast to other investigated fungi, xylanase production by M. fergusii was enhanced on pectin medium. Preliminary thermostability screening indicated that among the investigated species, thermotolerant glycosidases can be found. Some of the accessory activities, including the α-arabinosidase activity, were surprisingly high. The capability of the produced enzymes to improve the hydrolysis of lignocellulosic pretreated substrate was evaluated and revealed potential for these enzymes.  相似文献

Screening of 40 mesophilic and 13 thermophilic fungi indicated that enzyme activities capable of degrading oat spelt xylan extensively were produced by only a few of the mesophilic species investigated. The relatively low degree of hydrolysis effected by the enzymes from thermophilic organisms could be explained, in part, by their lack of -xylosidase. Several strains of Aspergillus awamori and Aspergillus phoenicis were notable in producing high xylanase and -xylosidase and low protease activities. Of the fungl tested, 13 produced activities capable of removing O-acetyl, arabinosyl, 4-O-methylglucuronyl, feruloyl and coumaroyl substituents from the backbone of xylan polysaccharides as well as endo-1,4--d-xylanase and -1,4-xylosidase. When the growth medium contained oat spelt xylan as carbon source, higher levels of xylanase, -xylosidase and acetyl xylan esterase were found than in cultures containing meadow fescue grass but the latter were richer in ferulic acid and coumaric acid esterases and 4-O-methylglucuronidase. No single organism or carbon source used was capabie of producing high levels of all the debranching enzymes as well as high levels of enzymes capable of cleaving the glycosidic linkages of the xylan backbone. The best ballnce of enzymes was obtained in cultures of A. awamori IMI 142717 and NRRL 2276 and A. phoenicis IMI 214827. To detect thermotolerant fungus strain for decolorization of alcohol distillery wastewater (WAD), 38 fungus strains were studied. Ability of ligninolytic enzyme production was examined at 35 and 43 degrees C on agar media containing 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) and MnCl2. At 43 degrees C, four of Pycnoporus coccineus strains showed their higher potential for WAD decolorization both on agar media and in liquid media. Immobilized mycelia on polyurethane foam removed total phenol about threefold higher than free mycelia did in shaking condition at 43 degrees C. Moreover, color removed by immobilized mycelia nearly 50% higher than free mycelia did.  相似文献

A thermotolerant species of Phormidium produced extracellular anti-microbial material during batch culture. Ten species of fungi were obtained from poultry droppings in Nigeria. Six of these are true thermophiles while the other four are thermotolerant. Aspergillus fumigatus Fresenius, Mucor pusillus Lindt and Thermoascus aurantiacus Stolk are known human pathogens. Except for M. pusillus, all the thermotolerant species had a higher occurrence at 45 degrees C while the thermophilic varieties were readily obtained at 50 degrees C.  相似文献

The phylogenetic relationship among Geosmithia argillacea, Talaromyces emersonii, Talaromyces byssochlamydoides and other members of the Trichocomaceae was studied using partial RPB2 (RNA polymerase II gene, encoding the second largest protein subunit), Tsr1 (putative ribosome biogenesis protein) and Cct8 (putative chaperonin complex component TCP-1) gene sequences. The results showed that these species form a distinct clade within the Trichocomaceae and Trichocoma paradoxa is phylogenetically most closely related. Based on phenotypic and physiological characters and molecular data, we propose Rasamsonia gen. nov. to accommodate these species. This new genus is distinct from other genera of the Trichocomaceae in being thermotolerant or thermophilic and having conidiophores with distinctly rough walled stipes, olive-brown conidia and ascomata, if present, with a scanty covering. Species within the genus Rasamsonia were distinguished using a combination of phenotypic characters, extrolite patterns, ITS and partial calmodulin and β-tubulin sequences. Rasamsonia brevistipitata sp. nov. is described and five new combinations are proposed.  相似文献
