目的调查Z:ZCLA长爪沙鼠原种群普通级长爪沙鼠的遗传多样性。方法用本实验室自行建立的长爪沙鼠19个生化基因位点的乙酸纤维素膜电泳技术并结合基本群体遗传学指标研究了普通级Z:ZCLA长爪沙鼠100个家系的遗传多态性。结果Z:ZCLA长爪沙鼠在Es-1、Car-2、Hbb、Gpi-1、Cs-1、Ce-2I、dh-l、Mod-1呈单态,在Es-2、Es-3、Es-4、Es-6、Es-8、Es-9、Es-10、pd-1、gm-1、Trf、Akp-1个位点上呈现多态性,等位基因从2~4个不等,平均等位基因数3.0,平均杂合度0.512,平均多态信息量0.455。结论提示目前本群遗传多样性水平处于中度多态。  相似文献

An outbred stock of Swiss:SE mice has been surveyed for genic heterogeneity at 12 loci, encoding biochemical polymorphisms in mice. Using horizontal starch-gel electrophoresis, 6 loci (Es-1, Es-2, Es-3, Es-5, Trf, Dip-1) revealed no genic heterogeneity within the total sample of 289 male mice examined. The other 6 loci (Mpi-1, Mup-1, Hbb, Gpi-1, Pgm-1, Ldr-1) each showed a 2-allelic variation within each of the 3 stock units (pavillions) involved. A pavillion effect on the observed genotype numbers was found for Pgm-1 and Gpi-1 (P less than 0.10). Inadequate genotyping may have occurred at Gpi-1 and Ldr-1, as suggested by a 'week effect' on the observed genotype numbers (P less than 0.05). For studying long-term genetic changes within outbred stocks, a routine monitoring procedure using biochemical polymorphisms is recommended.  相似文献

Cramer (1981) reported that Pg-1 (urinary pepsinogen) in the rat was loosely linked with albinism in the linkage group I. We performed a three point test on the loci of pg-1, c, and Hbb. We could reconfirm that pg-1 was autosomal trait with two co-dominant alleles of pg-1a and pg-1b. But in progeny of ((WF X IS) X WF) backcross, pg-1 was linked to neither c nor Hbb, while a close linkage between c and Hbb was detected. Also pg-1 was not linked to Mup-1 (LG II), a (LG IV), Es-3 (LG V), and h (LG VI). pg-1 will be one of the most valuable genetic markers of the rats, since pg-1 was highly polymorphic among inbred strains of rats, and not linked to LG I, II, IV, V, and VI.  相似文献

In the course of inspection of the biochemical marker genes in inbred strains of mice maintained in our laboratory, a female mouse of the NZB strain was found to be heterozygous for the Es-1 locus. Namely, it was Es-1a/Es-1b type. After this finding, many heterozygous mice were found among her sisters and the descendants. However, these heterozygotes (Es-1a/Es-1b) showed no heterozygosity for other 11 characters, i.e., the 6 biochemical markers (Hbb, Trf, Es-2, Id-1, Mod-1, Gpd-1) and the 5 coat colour markers (A, B, C, D, AND S) were idential as those previously described. It was, moreover, observed that they possessed the immunological characteristics typical of the NZB mice. Therefore, it could be concluded that the heterozygosity had been originated from a single mutation at the Es-1 locus, i.e., from Es-1a to Es-1b or vice versa. With regard to the alleles at the Es-1 locus, an investigation was carried out in two sub-strains of the NZB mice having different breeding history and the followings were clarified. One substrain imported from Karolinska Institute, Sweden, had been fixed with the Es-1a allele and the other imported from England was found to be Es-1b/Es-1b type. The NZB mice which displayed the heterozygosity had been derieved from the Karolinska sub-strain. Importance of biochemical marker genes for inspection of proper maintenance of inbred strains has been discussed.  相似文献

To make the genetic profile of the LIBP/1 inbred strain obtained from Kunming mice, the most widely used outbred stock in China, 26 loci were examined. The genotypes of four kinds of coat color genes were a/a, B/B, c/c and D/D. The results of testing 21 biochemical marker genes showed Akp-1b, Amy-1a, Car-2a, Ce-2a, Es-1b, Es-3a, Es-10a, Es-11a, Gpd-1a, Gpi-1a, Gus-1b, Hbbs, Idh-1a, Ldr-1a, Mod-1a, Mup-1b, Pep-3b, Pgm-a, Sep-1b, Tam-1c, and Trfb. A new esterase locus (Es-13) has been identified in Mus musculus. Strains AEJ/GnRk, LG/J, SJL/J, and SWR/J carry a recessive allele, Es-13 b, for a locus possibly involved in the posttranslational modification of a kidney esterase. All other strains observed carried the dominant Es-13 a allele. Es-13 was mapped on Chr 9 by recombinant inbred lines and by conventional backcrossing experiments. Backcross data produced the following gene order and map distances: Lap-1 (31.6±7.5 cM) Es-13 (2.6±2.6 cM) Mod-1.  相似文献

Linkage tests on the faded gene were carried out with some coat color and biochemical markers, It was shown that the faded locus was not closely linked to the following loci: Idh-1 (chromosome 1), a (2), Car 2 (3), Mup-1 (4), Pgm-1 (5), Hbb (7), Gpi-1 (7), Es-1 (8), Trf (9), Es-3 (11), s (14), Sod-1 (16) and Ce-2 (17). The complement protein C6 has been shown to be genetically polymorphic in the rat. Isoelectric focusing of plasma samples from 19 inbred strains demonstrated two electrophoretically distinguishable migration patterns, each consisting of three bands. Breeding studies with the use of the BN and DA strains showed that the C6 patterns were inherited in a manner consistent with the co-dominant autosomal expression of two alleles (C6 A and C6 B). The distribution of the C6 alleles in a backcross mating was compared with eight independently segregating marker genes: RT1.A, RT2, Gdc -1, Igk-1, Hbb, Svp-1, Fh-1, and Es-6. There was no detectable linkage between C6 and any of these eight loci.  相似文献

Immunofixation after isoelectric focusing revealed two forms of mouse C6, C6A and C6M, both of which consist of two major protein bands and one or more acidic minor bands. They were distinguishable by their different isoelectric point (pI) ranges: C6M has more acidic pI ranges (pH < 6.2) than C6A (pH < 6.3). C6A was found in common inbred mice of Mus musculus domesticus, while C6M was found in inbred and wild mice of M. m. molossinus (Japanese wild mice, an Asian subspecies). Breeding experiments showed that these two forms of C6 were controlled by a single codominant autosomal locus. We propose the designation C-6 for this locus with two alleles, C-6 a and C-6 m , which encode for C6A and C6M, respectively. Linkage analysis indicated that the locus is not closely linked to the following loci: Idh-1, agouti, Amy-1, brown, Gpd-1, Mup-1, Pgm-2, Pgm-1, albino, Hbb, Es-1, Mod-1, Sep-1, Es-3, Igh-1, beige, Es-10, Sod-1, and C-3.  相似文献

Electrophoretic analysis of 24 loci specifying a number of enzymatic and non-enzymatic proteins was carried out in populations of Primorye house mice. No variability in Ldh-B, Ldh of testis, fumarase, alpha-Gpd, Aat-2, Sod-2, Gp-1, GP-3 was found. Distribution of the same main variant in all samples is represented by Sdh, Mor-1, Mor-2, Ldh-A, GP-2, Est-D, Gpd-x, Pgl, Ldr and alb. The second variant of these loci was found in one or two animals. Two and more variants encountered with significant frequency were detected for Id-1, Hbb, Aat-1, Est-1, Est-2, Mod-2. Hybrid origin of the Primorye house mouse is assumed. Genogeography of Id-1, Hbb, Aat-1 and Est-1 points to involvement of the Mus-2C castaneus form in development of their gene pool, the effect of Mus-2A musculus being not excluded. Genogeography of Sod-1 confirms the involvement of both castaneus and musculus. Intensive mutual penetration of these forms (more than 300 km from north to south) indicates that they did not achieve the level characteristic of musculus and domesticus in the course of their differentiation.  相似文献

Samples of the Antarctic limpet, Nacella concinna (Strebel,1908), were collected from four sites in the South Orkney Islandsand from Stromness in South Georgia during 1988. At three SouthOrkney sites, both littoral and sub-littoral samples of limpetswere taken. Shell dimensions were measured and littoral andsub-littoral limpets were shown to have significantly differentshell shapes. Foot and digestive gland tissues were subjectedto electrophoresis and five polymorphic loci {Es-1, lcd-1, lcd-2,Gpi, Got-1 and Pgm-1) and two monomorphic loci (Es-2 and Got-2)were scored. Genetic identities between littoral and sub-littoralforms and also between sites were calculated. At the loci scored,the littoral and sub-littoral forms were virtually identicaland this suggests that the shell shape differences between theforms are the result of environmentally induced phenotypic plasticity.However, significant genetic differences and some morphologicaldifferences were apparent between the sample of limpets fromSouth Georgia and all other sites. Genetic identity values suggestedthat the South Georgia and South Orkney N. concinna are geographicallyseparated populations of a single species rather than distinctsub-species. Corn stocks showing virus-induced aberrant ratio (AR) at the "A" locus were found to have recessive alleles at the R and/or C loci. Since by the known pedigree these loci should be homozygous dominant, the results suggest an inactivation of maize genes by a mechanism as yet unknown. The presence of recessive alleles at these additional loci can explain the segregation ratios obtained in these particular stocks.  相似文献

A further polymorphic rat esterase with broad tissue expression and restricted substrate specificity is described and tentatively called Es-6. Inbred rat strains have either fixed allele Es-6F or fixed allele Es-6S. Es-6 is not linked to the established esterase cluster consisting of the eight esterase loci Es-1, Es-2, Es-3M, Es-4M, Es-4W, Es-5 (=Es-3W), Es-7, and Es-8 in LG V of the rat or to RT1, Gc, c, a, and h. We have examined a polymorphism for the soluble glutamate oxaloacetate (GOT-1) isozyme system which was found in the Asian mouse Mus castaneus. Variants of GOT-1 segregate as though they are controlled by codominant alleles for a single autosomal locus which we have designated Got-1. No close linkage of genes for soluble and mitochondrial forms of the enzyme, GOT-1 and GOT-2 respectively, was observed. Furthermore, no close linkage of Got-1 and the loci c, Gpi-1, Mod-2, Mod-1, Ld-1, Gpd-1, Pgm-1 or Gpo-1 was observed. Our results demonstrate the utility of sampling Mus from diverse populations to extend the repertoire of polymorphic loci and the genetic linkage map.  相似文献

Polyacrylamide gel electrophoresis was used to resolve allozymes in the cosmopolitan blood-feeding stable fly,Stomoxys calcitrans (L.). Nineteen of 38 loci were polymorphic (53%). Mean heterozygosities among all loci and among only polymorphic loci were 0.096 and 0.182, respectively. These gene diversity measures are about half those among other muscid Diptera. Variation in gene frequencies was examined in 10 natural stable fly populations from Iowa and Minnesota. Gene frequencies were homogeneous at five of eight loci among six populations in 1990 and eight of eight loci among four populations in 1992. Wright'sF statistics showed no significant departure from random mating among stable flies. It was concluded that gene flow compensates for any local differentiation in stable fly populations.  相似文献

Most conifer species occur in large continuous populations, but radiata pine, Pinus radiata, occurs only in five disjunctive natural populations in California and Mexico. The Mexican island populations were presumably colonized from the mainland millions of years ago. According to Axelrod (1981), the mainland populations are relicts of an earlier much wider distribution, reduced some 8,000 years ago, whereas according to Millar (1997, 2000), the patchy metapopulation-like structure is typical of the long-term population demography of the species. We used 19 highly polymorphic microsatellite loci to describe population structure and to search for signs of the dynamics of population demography over space and time. Frequencies of null alleles at microsatellite loci were estimated using an approach based on the probability of identity by descent. Microsatellite genetic diversities were high in all populations [expected heterozygosity (H(e)) = 0.68-0.77], but the island populations had significantly lower estimates. Variation between loci in genetic differentiation (F(ST)) was high, but no locus deviated statistically significantly from the rest at an experiment wide level of 0.05. Thus, all loci were included in subsequent analysis. The average differentiation was measured as F(ST) = 0.14 (SD 0.012), comparable with earlier allozyme results. The island populations were more diverged from the other populations and from an inferred common ancestral gene pool than the mainland ones. All populations showed a deficiency of expected heterozygosity given the number of alleles, the mainland populations more so than the island ones. The results thus do not support a recent important contraction in the mainland range of radiata pine.  相似文献

ABSTRACT. The taxonomic attribution of four Leishmania stocks isolated from humans in Ecuador has been explored by both multilocus enzyme electrophoresis and random amplified polymorphic DNA. For three loci, MLEE results showed patterns suggesting a heterozygous state for a diploid organism, while the corresponding homozygous states are characteristic of the Leishmania panamensis/guyanensis complex and Leishmania braziliensis . RAPD profiles exhibited for several primers a combination of the Leishmania panmensis/guyanensis complex and L. braziliensis characters. These data hence suggest that the four stocks are the results of hybridization between L. panamensis/guyanensis and L. braziliensis . MLEE data show that the results cannot be attributed to either mixture of stocks, or an F1 in the framework of a simple Mendelian inheritance.  相似文献

采用ISSR分子标记对分布于山东省长岛县南部的北长山、南长山、大黑山和小黑山4个岛屿上的藜天然种群共81个个体进行了遗传多样性及遗传结构的研究。13个引物共检测到157个可重复的位点。遗传多样性研究结果表明:种内的多态位点比率为66.24%,具有较高的遗传多样性(Shannon(I)指数在物种水平上为0.332 0);种群间有一定的遗传分化,根据Gst值,种群间的遗传多样性占总群体的9.27%。遗传距离分析表明,XHS种群和NCS种群的遗传一致度最高,与地理距离无相关性。  相似文献

The cytoplasmic malic enzyme (Mod-1) catalyzes the oxidative decarboxylation of malate: malate + NADP+----pyruvate + CO2 + NADPH + H+. Using a cDNA clone of Mod-1 as a probe, two new DNA markers not at the Mod-1 locus (restriction fragment length polymorphisms, RFLP) were detected by Southern blot analysis that showed extensive homology to Mod-1 sequences. Linkage of each restriction fragment length polymorphism to loci other than Mod-1 was assessed using the BXD (C57BL/6J x DBA/2J) recombinant inbred strains and confirmed by backcrosses. One polymorphic site, designated D9Rti1, was found to be closely linked to the phosphoglucomutase (Pgm-3) locus on Chromosome 9. The other hybridization site, designated D1Rti2, was closely linked to the isocitrate dehydrogenase (Idh-1) locus on Chromosome 1. The data presented imply that Mod-1 homologous sequences are tightly linked to three different metabolic enzymes.  相似文献
