The ultrastructure of follicle cells in the ovary at different developmental stages of Branchiostoma has been observed in detail with a transmission electron microscope. The results indicate that only one kind of follicle cell exists with structural features related to steroid hormone biosynthesis: (i) oval or round mitochondria with tubules; (ii) smooth surfaced endoplasmic reticulum; (iii) several large lipid droplets in the cytoplasm; (iv) a well de-veloped Golgi complex and tubular rough surfaced endoplasmic reticulum, as can be found in mammalian theca interna cells. In addition, as steroid hormone synthesizing cells, they obviously play an important role in the phagocytosis of relict gametes and cellular debris and may have a nutritive function for the oocytes. They can produce abundant secre-tory granules in stages III-IV ovaries. In mature ovaries they transform into flat epithelial cells with numerous micro-filaments which may play a role in ovulation.  相似文献

Summary The olfactory organ of Helisoma trivolvis is located on the surface of the body at the base of the cephalic tentacles. An evagination of skin, the olfactory plica, at the base of the tentacle extends over the olfactory organ dorsally. The epithelium of the olfactory organs contains unspecialized epithelial cells, ciliated epithelial cells, basal cells, mucous secretory cells, and sensory dendrites. The surface of the epithelium has a complex brush border of thick plasmatic processes, which branch to form several terminal microvillar twigs. Long slender cytoplasmic processes form a dense spongy layer among the plasmatic processes beneath the level of the terminal twigs. Bipolar primary sensory neurons clustered beneath the epithelium of the olfactory organ send dendrites through the epithelium to the free surface. Some sensory endings have a few short cilia, but most bear only microvilli. Cilia of sensory endings and epithelial cells extend beyond the brush border of the epithelium. Small axons arise from the perikarya of the sensory neurons and enter a branch of the olfactory nerve. HRP tracing indicates that the axons pass to the cerebral ganglion without interruption. Histochemical tests indicate that the sensory neurons are neither aminergic nor cholinergic.  相似文献

Summary The endoplasmic reticulum of mononuclear placental cells from 10 cows in different stages of pregnancy has been studied with the electron microscope. Basicaly the cryptal cells are provided with rough surfaced endoplasmic reticulum. In addition, whorls of rough or smooth cytomembranes encircle lipid droplets or plain cytoplasmic matrix. The trophoblastic cells also contain rough surfaced endoplasmic reticulum. Furthermore, skein-like conglomerations of smooth tubules are encountered in some cells. The cephalic sensory organ (CSO) in planktonic veliger larvae of Littorina littorea is situated dorsally between the velar lobes at the level of the shell aperture. It consists of ciliated primary sensory cells, adjacent accessory cells and supporting epithelial cells. Cell bodies of the ciliated cells originate in the cerebral commissure and their dendrites pass to the epidermis. The flask-shaped sensory cells are characterized by a deep invaginated lumen with modified cilia arising from the cell surface in the lumen. These cilia are presumed to be non-motile because they lack striated rootlets and show a modified microtubular pattern (6 + 2, 7 + 2 and 8 + 2). The adjacent accessory cells never possess an invaginated lumen; occasionally cilia and branched microvilli arise from the apical surface. These cells may be sensory, but there is no obvious direct connection with the nervous system. The supporting epithelial cells are part of the epidermis and flank the apical necks of the sensory and accessory cells. Morphological evidence suggests that the CSO may function in chemoreception related to substrate selection at settlement, feeding or other behaviour.  相似文献

Summary Olfactory receptor molecules are assumed to be integral membrane proteins which may be visualized on fracture faces of the membrane as intramembrane particles (IMPs). In the present study, the plasma membrane of the receptor dendrites and ciliated epithelial cells in the teleost fish Alburnus alburnus were studied by freeze-fracture electron microscopy. The IMP diameters on the membrane P-faces of both receptor dendrites and ciliated epithelial cells ranged from 5 nm to 11 nm. The average IMP densities on membrane fracture faces of the ciliated and microvillous sensory dendrites were 3130±780 for the cilia, 2070±550 for the microvilli, 2390±1190 on the knob regions and 3050±1130/m on the lateral dendrite membranes. The IMP densities on the P fracture faces of the cilia and knob regions were compared with the densities found on the lateral membranes of each individual dendrite. The ratios ranged from 0.5 to 0.96 in the case of the cilia/lateral membrane and from 0.5 to 0.90 in that of the knob/lateral membrane, indicating that, in contrast to the average densities, it is the lateral membrane which has the higher IMP densities and not the cilia. The great variations in the average IMP densities, as well as the considerable variety of the ratios, may be explained by the maturation and turnover of the olfactory sensory neurons.  相似文献

The fine structure of the epithelial cells of one component of the prostatic complex of the mouse—the ventral lobe—has been investigated by electron microscopy. This organ is composed of small tubules, lined by tall simple cuboidal epithelium, surrounded by smooth muscle and connective tissue. Electron micrographs of the epithelial cells of the ventral lobe show these to be limited by a cell membrane, which appears as a continuous dense line. The nucleus occupies the basal portion of the cell and the nuclear envelope consists of two membranes. The cytoplasmic matrix is of moderately low density. The endoplasmic reticulum consists of elongated, circular, and oval profiles representing the cavities of this system bounded by rough surfaced membranes. The Golgi apparatus appears localized in a region between the apical border and the nucleus, and is composed of the usual elements found in secretory cells (3, 9). At the base of the cells, a basement membrane is visible in close contact with the outer aspect of the cell membrane. A space of varying width, which seems to be occupied by connective tissue, separates the epithelial cells from the surrounding smooth muscle fibers and the blood vessels. Bodies with the appearance of portions of the cytoplasm, mitochondria, or profiles of the endoplasmic reticulum can be seen in the lumina of the acini and on the bases of these pictures and others of the apical region the mechanism of secretion by these cells is discussed. The fine structural organization of these cells is compared with that of another component of the mouse prostate—the coagulating gland.  相似文献

HeLa cells from conventional culture media have been studied in thin sections with the electron microscope; in many cases cells were examined in sets of sections cut in series. The fine structure of the cells is described including three unusual features not hitherto reported. It has been found that numerous cells contained rows of parallel smooth surfaced cisternae spaced about 150 mµ apart and communicating with rough surfaced elements of the endoplasmic reticulum. These cisternae resembled "annulate lamellae" but did not contain regular arrays of pores. In many cells an area of juxtanuclear cytoplasm was occupied by a membranous structure composed of closely applied pairs of narrow cisternae either arranged in concentric rings or else extending in several directions in a haphazard manner. Sparse particles were present on the outer membranes of each pair of cisternae. Communications between the double cisternae and other membrane-bounded structures were not observed. A small number of cells contained areas of cytoplasm devoid of organelles and filled with amorphous fuzzy material. The observations recorded are discussed.  相似文献

Opheliid nuchal organs are composed of ciliated cells, retractor muscles, and sensory cells. The perikarya of sensory cells are located in the posterior portion of the brain, and their distal processes extend along the body wall, as the nuchal nerve, and terminate just anterior to the ciliated region. The nuchal nerve of the juvenile is composed of 30–35 dendrites; the adult nuchal nerve has 35–40 dendrites. The ends of the sensory dendrites form sensory bulbs which are clustered around the olfactory chamber, and each bulb bears a modified cilium. Sensory cilia lose their axonemes and extend as microvillous-like structures into the olfactory chamber. Supportive cells delineate approximately the posterior and dorsal portions of the chamber with sensory bulbs forming the remaining ventral and anterior portions. On the lateral aspect of the chamber, cuticular matrix extends into it, and in this area supportive cells bear microvilli which extend into the matrix. The adult nuchal organ is larger than that of the juvenile, and the sensory portion of the olfactory chamber wall is expanded. Expansion of the sensory area is apparently the result of size increase in sensory bulbs and by intrusion of supportive cells between sensory bulbs.  相似文献

Most of the sensory cells found in the chemoreceptor of the ommatophore of Helix pomatia are typical bipolar cells. The chemoreceptor is deveded by a furrow into two parts; within the ventral subdivision the layer of sensory cell bodiesis thicker than in the dorsal part. According to the differentiations of the apical surface of the dendrites, it is possible to distinguish six different classes: a) dendrites with one cilium and 75 nm thick cytofila (sometimes dendrites of identical appearance posses more than one cilium); b)dendrites with several cilial and 150 nm thick cytofila; c) dendrites with several cilia, 50 nm thick cytofila, and long, striated rootlets; d) dendrites with several cilia bur without cytofila; e) dendrites with 130 nm thick cytofila but without cilia; and f) dendrites with 65 nm thick cytofila but without cilia; dendrites of this class are the only ones with a cytoplasm more electron dense than that of the surrounding supporting cells. All these dendrites are connected to the surrounding supporting cells by terminal bars, each consisting of zonula adhaerens, aonula intermedia and zonula septata. The perikarya of the sensory cells measure approximately 15 mum by 8 mum and enclose 10 mum by 6 mum large nuclei. Axons, originating from these perikarya, extend to the branches of the digital ganglion. In the distal part of this gangloin the axons come into synaptic contact with interneurons, but in our electron micrography it was not possible to coordinate processes and synapses with the corresponding neurons.  相似文献

Summary Electron-microscopic cytochemical studies on satellite cells of normal human skeletal muscle were carried out using the concanavalin Aperoxidase (Con A-HRP) coupling method. Con A-binding sites, which probably correspond to glycoproteins, were found to be associated with the cell surface, smooth surfaced vesicles, nuclear envelope and endoplasmic reticulum of the satellite cells and were also identified at the cell surface of the adjacent muscle fiber. The possible relationships of these observations to the functions of satellite cells are discussed.  相似文献

Portions of the livers of fingerling rainbow trout were studied by light and electron microscopy. The histology, cytology and ultrastructure of mesothelial cells, serosal fibroblasts, hepatocytes, sinusoidal endothelial cells, endothelial cells of central veins and blood cells were described. Mesothelial cells and fibroblasts constituted a very thin capsule. Hepatocytes contained extensive areas of rough surfaced endoplasmic reticulum, consisting mainly of parallel cisternae and pools of glycogen. One or two nuclei and numerous mitochondria occurred in the areas of endoplasmic reticulum, but never in the pools of glycogen. Hepatocyte surface possibilities included hepatocyte to hepatocyte, hepatocyte to bile canaliculus, hepatocyte to space of Disse and hepatocyte to serosa. The trout liver was compared compared to channel catfish liver and to rat liver. Functional implications of the structural features were discussed.  相似文献

Five malleoli extend ventrally from the proximal segments of each hind leg of mature and immature solpugids. Each malleolus contains the dendrites of 72,000 bipolar sensory neurons with somata in a ganglion in the leg just dorsal to the malleolus. The dendrites terminate in a sensory groove which extends along the ventral edge of the sense organ. The sensory groove has a slit-like opening to the outside, suggesting that these sense organs are chemoreceptors. The malleoli also contain long processes from microvillar cells with somata in the sensory ganglion. The microvillar cell processes extend into the malleolus to the ciliary ending of the dendrites where each process has an enlarged microvillar ending which surrounds the outer segments of 15 to 22 dendrites and releases a sense secretion.  相似文献

The olfactory epithelium contains three cell types: basal cells, supporting cells and sensory neurons. Electron microscopy as well as immunofluorescence microscopy with intermediate-filament antibodies were used to study the rat olfactory epithelium in order to obtain more information about these different cell types and to try to investigate their histogenetic origins. We found mitoses in the basal-cell layer, as well as multiple centrioles and tonofilaments in some basal cells. As revealed by electron microscopy, the supporting cells contained tonofilaments and reacted strongly with antibodies to keratin, in line with their known epithelial nature. When antibodies to other intermediate-filament types were used, i.e. glial fibrillary acidic protein, vimentin, desmin and neurofilaments, no reaction was seen in the cells of the olfactory epithelium, with the exception of occasional staining of a few axons in the subepithelial layer by neurofilament antibodies. In particular, the cell bodies, dendrites and most axons of the sensory neurons were negative for a variety of antibodies against neurofilaments. Olfactory sensory neurons therefore belong to the very few cells in adult animals which seem to lack intermediate filaments. We discuss whether this finding is related to the fact that these cells are also unique among neurons in that they are not permanent cells but constantly turn over.  相似文献

The localization of concanavalin A (Con A) binding sites in Purkinje cell somata and dendrites has been studied using a peroxidase labeling technique. In the somata, the nuclear, Golgi, and endoplasmic reticulum (ER) membranes are rich in Con A binding sites. The hypolemmal cisternae, which are continuous with the ER from the soma and throughout the dendritic tree of Purkinje cells, are also rich in Con A binding sites. Other cisternae seen in these dendrites do not bind detectable amounts of Con A. The results suggest that a cisternal system, rich in carbohydrate, may be continuous from the nuclear envelope to distal dendritic segments of Purkinje cells. Such a system could play a role in the movement of materials from Purkinje somata to dendrites.  相似文献

A group of patients, suffering from sequelae of acute radiation sickness (ARS), and liquidators was studied 5 years after exposure to a complex of factors resulting from the Chernobyl A.P.S. disaster. Studied were: the antibody titres to antigens of the cytoplasm of thymus epithelial reticulum cells and to Hassall's corpuscles the levels of serum immunoglobulins M, G, A; and the content of serum alpha 1-thymosin. Patients with ARS sequelae and liquidators showed a high level and incidence of autoantibodies to antigens of cytoplasm of thymus epithelial reticulum cells and to Hassall's corpuscles. There were no significant differences between the antibody levels in the blood of patients with ARS sequelae and liquidators. The antibodies were found to belong to IgM class; there was a correlation between the serum IgM titres and the rate of the indirect immunofluorescence reaction with autoantibodies to antigens of the cytoplasm of the thymus epithelial reticulum cells. To identify autoantibodies cryostat sections of human and mouse, (CBA x C57BL/6) F1, thymus as well as the epithelial and stromal cell culture of mouse thymus can equally be used.  相似文献

Summary Mucosal cells of the rat jejunum vary greatly in the relative abundance and kind of endoplasmic reticulum (ER). Many cells have few rough surfaced cisternal ER elements; these cells frequently are located in basal villous regions and generally are less differentiated than those near apices. Profiles of rough ER cisternae were often encountered in stacks of 3–4 parallel units in apical cells. A number of differentiating basal cells were found to contain ER-annulate lamellar associations arranged similarly to the ER stacks of apical cells. The annulate lamellae of this complex resemble those described for other rapidly differentiating or embryonic cells and may be derived from the nuclear envelope.
