Distribution of DOPA-oxidase isozymes in larval Culex pipiens pipiens, Culex pipiens quinquefasciatus and their hybrids was determined by agarose gel electrophoresis. Three zones of enzyme activity were detected in the subspecific and hybrid populations. Zone I was cathodic whereas Zones II and III were anodic. Electrophoretic variability was evident only in Zone II which was dimorphic for enzyme activity. Zones I and III were monomorphic. The subspecific and hybrid populations were distinguishable from one another only in Zone II. In all three populations, melanization was most intense in Zone II, less in Zone I, and least in Zone III.  相似文献

The purpose of this study was to examine the role of prostaglandins in the redistribution of renal cortical blood flow that occurs following reductions in renal perfusion pressure. The distribution of blood flow to the renal cortex was examined using radio-labeled microspheres (15 +/- 1 micron). It was found that in animals not treated with a prostaglandin synthesis inhibitor a decrease in renal perfusion pressure to the limit of renal blood flow autoregulation was associated with a decrease in fractional flow to the outer cortex (Zone I) and an increase in fractional flow to the inner cortex (Zones III and IV). A further decrease in renal perfusion pressure below the limit of autoregulation produced a further decrease in the fractional flow to Zone I and a further increase in fractional flow to Zones III and IV. In contrast, in animals treated with the prostaglandin synthesis inhibitor meclofenamate (5 mg/kg, i.v. bolus) a reduction in renal perfusion pressure to the limit of renal blood flow autoregulation produced no change in fractional blood flow to any of the 4 cortical zones. A further decrease in renal perfusion pressure, however, did produce a fall in fractional blood flow to Zone I and an increase in fractional flow to Zones III and IV. In conclusion, the results of this study indicate that within, but not below, the limit of renal blood flow autoregulation prostaglandin synthesis is an important factor in the regulation of renal cortical blood flow distribution.  相似文献

The distribution of Hyalomma species on domestic animals was studied in four zoogeoghraphical zones. Nine hundred and ninety-two Hyalomma ticks were collected from sheep, goats, cattle and camels. A total of seven tick species consisting of: Hyalomma anatolicum Koch, 1844 (28.93%), H. excavatum Koch, 1844 (12.5%), H. asiaticum Schulze & Schlottke, 1930 (13.5%), H. marginatum Koch, 1844 (27.01%), H. detritum Schulze, 1919 (9.67%), H. schulzei Olenev, 1931 (4.03%) and H. dromedary Koch, 1844 (4.33%) were recorded. The results indicated that H. anatolicum, H. asiaticum, H. marginatum and H. detritum were present in all zones whereas H. excavatum was absent in Zone I. The result also showed that H. dromedari was presented in Zones III and IV whereas H. schulzei was present in Zones II and III. During this study, the tick species collected are candidates for investigation as vectors of pathogenic organisms in Iran.  相似文献

The basalis of the primate endometrium: a bifunctional germinal compartment   总被引:7,自引:0,他引:7  
Radioautographic analysis of epithelial and stromal cell proliferation in the primate endometrial functionalis and basalis (rhesus monkey) has identified horizontal zonal patterns of mitotic activation and inhibition during natural menstrual cycles. At 1 h after a single i.v. injection of [3H]thymidine, mitotic activity in endometrial biopsies (hysterotomy) was determined on 9 days from the late proliferative to the late luteal phase (-2 days to + 14 days relative to the estrogen [E2]peak). Labeling indices (LIs) were determined within glandular segments of the 4 horizontal endometrial zones: Transient functionalis Zone I (luminal epithelium) and Zone II (uppermost gland); Germinal basalis: Zone III (middle gland) and Zone IV (basal gland). The size of the dividing epithelial populations (LI) differed zonally. During E2 dominance (-2 days to +3 days), the epithelial LIs of functionalis I (10 +/- 0.3%) and II (9.8 +/- 1.0%) were greater than those of basalis III (5.8 +/- 0.2%) and basalis IV (3.7 +/- 0.8%). During progesterone (P) dominance (+5 days to +14 days), epithelial mitosis was strongly inhibited in functionalis I (4.3 +/- 1.9%), functionalis II (0.8 +/- 0.2%), and basalis III (1.4 +/- 0.5%). Thus germinal basalis III was linked functionally with transient functionalis I and II by periovulatory uniformity in epithelial proliferation and postovulatory mitotic inhibition. A unique mitotic pattern set basalis IV apart from other zones by a steady rise in LI from 1% (-2 days) to 11% (+10 days). The LIs for stromal fibroblasts remained quite uniform in basalis IV but varied in other zones. Thus the postovulatory primate basalis was a distinct bipartite compartment in which the mitotic rate in basalis IV glandular epithelium increased steadily whereas that of basalis III was strongly inhibited. The remarkable enhancement of epithelial mitotic activity in basalis IV may reflect expansion of the stem-progenitor cell population for gestational growth or for post-menstrual regeneration.  相似文献

1. The nuclei of the cells of the whole rat brain have been fractionated in a B-XIV zonal rotor with a discontinuous gradient of sucrose. Five fractions were obtained. Zone (I) contained neuronal nuclei (70%) and astrocytic nuclei (23%). Zone (II) contained astrocytic nuclei (81%) and neuronal nuclei (15%). Zone (III) contained astrocytic nuclei (84%) and oligodendrocytic nuclei (15%). Zone (IV) contained oligodendrocytic nuclei (92%) and zone (V) contained only oligodendrocytic nuclei. 2. The content of DNA, RNA and protein per nucleus was determined for each zone. Although the amount of DNA per nucleus is constant (7pg) the RNA varies from 4.5 to 2.5pg/nucleus and the protein from 38 to 17.6pg/nucleus. The neuronal nuclei have the greatest amounts of protein. The oligodendrocytic nuclei have the least content of RNA and protein. 3. The effects of pH, ionic strength, and Mg(2+) and Mn(2+) concentration on the activity of the nuclear system for synthesis in vitro of RNA have been investigated for unfractionated nuclei. From these studies a standard set of conditions for the assay of nuclear RNA polymerase has been established. 4. The activity of the RNA polymerase in each of the zonal fractions has been determined in the presence and in the absence of alpha-amanitin. Zone (II) is the most active, followed by zone (I). The nuclei of zones (IV) and (V) have comparable activity, which is 40% of that of zone (II). 5. The extent of incorporation of each of the four labelled nucleoside triphosphates by the nuclei from each zone has been measured. These values have been used to calculate the base composition of the RNA synthesized in vitro in each class of nucleus. 6. The effect of changes in the condition of assay of RNA polymerase in the different classes of nuclei has been investigated. Significant differences in the response to concentrations of metal ions and ammonium sulphate have been observed. 7. Homopolymer formation in each zone of brain nuclei has been determined. The extent of formation of the four homopolymers roughly parallels the RNA polymerase activity.  相似文献

Intra-articularly administered sodium aurothiomalate (Myochrysine) produced aurosomes containing characteristic electron dense contents (indicating the presence of gold), in the chondrocytes of rabbit articular cartilage. At first the aurosomes were bounded by a membrane but later the electron dense contents were seen lying free in the cytoplasmic matrix. Such deposits were detectable up to 14 months after injection of Myochrysine but none were found at later time intervals (18 months and 2 years). There was a reduction in the population of superficial chondrocytes (Zone I) while those in deeper zones (Zones II and III) showed an increased content of intracytoplasmic filaments. It is thought that these are regressive or degenerative changes produced by gold.  相似文献

Limulus polyphemus hemocyanin is a 3.3 x 10(6)-Mr protein containing 48 subunits in an assemblage of eight hexamers. The molecule can be dissociated into monomers and dimers at pH 8.9 in the presence of 0.01 M EDTA. These subunits are heterogeneous and can be separated into five zones (I--V) by DEAE-Sephadex chromatography. Reassembly experiments were carried out with varied subunit mixtures, based on different combinations of the five chromatographic zones, in order to study the structural role of the diverse subunits in the eight-hexamer molecule. The reassembly products were analysed by electron microscopy and ultracentrifugation. No structural role for zone I could be found. Zone V and possibly zone II are needed to form structures larger than hexamers. Absence of zone III causes irregular aggregation of hexamers. Zone IV and perhaps zone II are needed to make eight-hexamer molecules from four-hexamer molecules. From these results we conclude that there is a high degree of subunit specificity in the inter-subunit contacts in the native Limulus hemocyanin molecule.  相似文献

将中国北方13个省按纬向划分为5个区域:32°~36°N(区域Ⅰ)、36°~40°N(区域Ⅱ)、40°~44°N(区域Ⅲ)、44°~48°N(区域Ⅳ)和48°~52°N(区域Ⅴ),然后利用Savitzky-Golay滤波算法平滑了1982~1999年NOAA/AVHRR NDVI和MSAVI时间序列影像,基于经验正交函数(EOF)分析提取了不同区域植被NDVI和MSAVI主分量,估测了1982~1999年中国北方不同纬度带的植被生长季开始、结束和长度,最后对1982~1999年不同区域的生长季参数进行了线性拟合,分析了不同区域的植被生长季变化趋势.研究表明,不同纬度带的植被生长季开始日期均表现出不同程度的提前趋势,区域Ⅳ的植被生长季开始提前趋势最大;生长季结束日期呈现推迟的趋势,区域Ⅱ的植被活动结束日期的推迟趋势最大,而区域Ⅲ最小.整个生长季长度呈延长趋势,延长日期在10 d以上.  相似文献

Abstract

The purpose of this study was to investigate the distribution of total and bioavailable metals (Cd, Cu, Cr, Fe, Mn, Pb and Zn) in sediments of the Abelardo L. Rodríguez (ALR) dam located in the eastern part of the city of Hermosillo, Sonora, Mexico. Seventy two sediment samples were collected in 2009 during four sampling campaigns in February (spring), May (summer), September (end of summer) and December (winter) in five different areas within the dam surface (Zones I, II, III, IV and V), including the Gate Station. Determination of heavy metals was carried out by flame atomic absorption spectrophotometry (FAAS). The results indicate high levels of total heavy metals in the following order: Fe>Mn>Zn>Pb>Cu>Cr>Cd. This is indicative of the impact of human activities located in areas surrounding the reservoir, specifically urban and industrial. The distribution and state of accumulation of trace metals in the sediment is largely dominated by the residual and Fe/Mn oxides geochemical phases. Fraction I (exchangeable) also presented high concentrations of metals (Cu, Fe, Mn and Zn). From enrichment factor analysis, the study area is prevalently enriched in Cd, Cu and Pb in Zones I, II, III, IV and Gate Station. This indicates that the sediments are impacted by anthropogenic activities such as downloads, domestic and industrial wastewater. Geoaccumulation index (Igeo) indicates that Zones I, II, III, IV and V (including Gate Station) do not show contamination by Cr, Fe, Mn and Zn. However, there is a moderate to heavy contamination by Cd, Cu and Pb (Igeo: 2–4) in all areas of study. The comparison between the results obtained with the sediment quality criteria (LEL and SEL), indicate that Zones I, II, III, IV, V and Gate Station, are contaminated with Cd, Cu and Pb, and severely contaminated by Fe.

The elevated levels of heavy metals detected in the sediment of the ALR Dam require special attention, since in the exchangeable fraction, the metals are specifically adsorbed on the sediment and can be released when the ionic composition of water changes. However, additional studies are required in this reservoir on the chemistry and toxicology of metals for a full assessment of potential risks posed to biota and man.  相似文献

Germ nuclei of Blepharisma japonicum begin meiosis within a few hours when cells of complementary mating types conjugate. We synchronized the onset of conjugation and treated cells in different stages of meiosis with 10 micrograms/ml cycloheximide which strongly inhibits protein synthesis in this ciliate. Cycloheximide arrested meiosis at six stages: I, between pairing of cells and swelling of germ nuclei; II, leptotene; III, zygotene; IV, pachytene; XI, interkinesis; XII, prometaphase II. Five of these arrests were reversible. Puromycin (250-500 micrograms/ml) also inhibited the progress of meiosis, though to lesser extents. We propose that the progression of meiosis of B. japonicum requires at least six proteins which are synthesized sequentially during meiosis.  相似文献

1. A comparison is made of gel electrophoretic patterns of the "whey" proteins of the milk of red (Macropus rufus) and eastern grey (Macropus giganteus) kangaroos at various stages of lactation. Qualitative and quantitative changes occur with time during the mature phase of lactation of both types. Their onset is related solely to the stage of lactation. "Whey" proteins are isolated and characterised and the nature of protein changes determined for the first time. 2. The anodic electrophoretic pattern is divided into 6 main zones (designated A F in order of decreasing mobility) and 2 cathodic zones (G and H) that are only detected in the milk of M. giganteus. 3. Zones A, B and C are milk specific. Zone B is present throughout lactation in both species and is an alpha-lactalbumin. Zones A and C are present only in late lactation, zone C, usually, but not always, appearing first. Zone A is an alpha-lactalbumin in M. giganteus, but is not an alpha-lactalbumin in M. rufus. Zone C appears to be the same protein in both species and is possibly a beta-lactoglobulin. 4. Zone D is kangaroo serum albumin and zone E is possibly a beta 2-microglobulin. Zone F contains three main iron (III) binding bands whose relative intensity varies with stage of lactation. Their intensity differs from the corresponding blood serum transferrin bands. 5. Zone H of Macropus giganteus is a lysozyme. 6. Lactose is present in the milk, but is not the principal sugar. 7. The significance of the results is discussed.  相似文献

Composite-tissue (e.g., hand allograft) allotransplantation is currently limited by the need for immunosuppression to prevent graft rejection. Inducing a state of tolerance in the recipient could potentially eliminate the need for immunosuppression but requires reprogramming of the immunological repertoire of the recipient. Skin is the most antigenic tissue in the body and is consistently refractory to tolerance induction regimens using bone marrow transplantation alone. It was hypothesized that tolerance to skin allografts could be induced in rats by injecting epidermal cells with bone marrow cells during the first 24 hours of life of the recipients. Brown Norway rats (RT1n) served as donors for the epidermal cells, bone marrow cells, and skin grafts. Epidermal cells were injected intraperitoneally and bone marrow cells were injected intravenously into Lewis (RT1l) newborn recipient rats. In control groups, recipients received saline solution with no cells (group I, n = 12), bone marrow cells only (group II, n = 15), or epidermal cells only (group III, n = 15). In the experimental group (group IV, n = 18), recipients received epidermal and bone marrow cells simultaneously. Skin grafts were transplanted from Brown Norway (RT1n) rats to the Lewis (RT1l) rats 8 weeks after cell injections. Skin grafts survived an average of 8.5 days in group I (10 grafts), 9.2 days in group II (12 grafts), and 12 days in group III (14 grafts). Grafts survived 15.5 days (8 to 26 days) in group IV (15 grafts). The difference was statistically significant (p < 0.05). Hair growth was observed in some accepted grafts in group IV but never in the control groups. This is the first report of prolonged survival of skin allografts in a rat model after epidermal and bone marrow cell injections. Survival prolongation was achieved across a major immunological barrier, without irradiation, myeloablation, or immunosuppression. It is concluded that the presentation of skin-specific antigens generated a temporary state of tolerance to the skin in the recipients that could have delayed the rejection of skin allografts.  相似文献

In Argentina, due to climatic conditions, Fusarium head blight (FHB) caused by Fusarium graminearum, affected the 1993/94 wheat crop. To evaluate the severity of this disease, samples of wheat where gathered from four zones of the wheat area. Sanitary conditions and mycotoxin contamination were determined. One zone (IIN) was intensely affected by FHB with 90% of samples in grade III (bad quality). No samples were grade I (good quality). The other zones were less affected falling into grade I or II (moderate quality). In all samples tested F. graminearum was the most prevalent species singly or in combination with others. Zone II N, with a DON mean level of I1.26 ppm, did not fulfil aceptability limits, whereas zones IIS, III and IV with overall means of 2.12, 1.57 and 1.0 ppm, respectively, did. Statistical analysis showed a close relation between percentage FHB and DON contamination (r:-0.71, p<0.01) in infected samples.  相似文献

Summary The techniques of light and electron microscopy have been employed in a study of the protective coverings of the egg of Drosophila melanogaster. Data obtained during this investigation suggest the involvement of the follicle cells, in the production of one of these coverings and justify its classification as a secondary coat. The secondary coat of D. melanogaster is highly organized and has been divided into three Zones (I, II, IIII). The follicle cells enveloping the oocyte exhibit two phases of secretory activity each involving hypertrophy of the Golgi complex and rough endoplasmic reticulum, and the production of protein and polysaccharide components. The first phase concerns the elaboration of the material which gives rise to the homogeneous lamina referred to as Zone I. The second results in the release of an electron dense component which becomes organized into two laminae separated by struts or pillars; this construction is referred to as Zone II. At the completion of this secretory phase, the follicle cells assume a squamous morphology and a third Zone, composed of a homogeneous substance, appears between the follicle cells and Zone II.This investigation was supported by grant GM-08776 to one of us (EA) from the National Institutes of Health, United States Public Health Service.  相似文献

The histology and ultrastructure of the salivary glands of male and female H.a. anatolicum ticks have been examined m unfed and feeding ticks with special emphasis on aspects related to the feeding process. The salivary glands of H.a. anatolicum consisted of three types of acinus (acinus I, II and III) in females and an additional type IV acinus in males. The type I acinus was agranular and showed slight morphologic changes during feeding. The presence of cells with ultrastructural features characteristic of epithelia involved in the secretion of hyperosmotic fluids supports the hypothesis that these acini secrete hygroscopic saliva during questing stages to absorb water from an unsaturated atmosphere. There were five granular cell types (a, b, c1c3) in type II acinus, three granular cell types (d, e, and f) in type III acinus, and one granular cell type (g) in type IV acinus. The cells a, d and e secreted most of their granules early in feeding and are considered to be cement precursors. The b and c cells appeared to synthesise and secrete their products throughout feeding and so are likely to secrete anticoagulants, enzymes and other pharmacologically active agents required during feeding. The interstitial cells, which were insignificant in acinar types II, III and IV of unfed ticks, became more distinct during feeding. The type III acinus in females showed remarkable cell transformations, during the course of feeding. The ablumenal interstitial cells of type III acinus, in females formed a basal labyrinth of extraordinary complexity by interdigitating with the basolateral membranes of transformed f cells to form a network of extracellular channels to excrete fluid during feeding. There was an enormous increase in the secretory granules of g cells as the feeding advanced. The secretory granules were released by a process of exocytosis, by direct fusion with the apical membranes and through channels connecting several granules.  相似文献

The uterine endometrium of menstruating primates (rhesus monkey, human) consists of a germinal basalis that regenerates a transient functionalis during each menstrual cycle. The endometrium is further subdivided into 4 zones that differ histologically and in epithelial mitotic rate along the longitudinal axes of the uterine glands and microvasculature (Bartelmez et al: Contrib. Embryol. Carnegie Inst., 34:99-146, 1951; Bartelmez: Am. J. Obstet. Gynecol., 74:931-955, 1957; Padykula et al.: Biol. Reprod., 32:1103-1118, 1118, 1984; Biol. Reprod., in press, 1988). The zones are defined as follows: functionalis I, luminal epithelium; functionalis II (upper straight gland segments); basalis III (middle gland segments), and basalis IV (bottoms of the glands). The surrounding stroma and microvasculature also differ zonally. Ultrastructural epithelial differences are evident among the 4 zones during 3 distinct functional states during natural menstrual cycles and after ovariectomy: 1) basal level after ovariectomy and 2) estrogen dominance and 3) progesterone dominance. Zonal structural differences persist at a minimal level of differentiation after ovariectomy and thus zonation is an inherent property. During estrogen dominance, distinctive ultrastructural differences are evident among the 4 zones, such as epithelial cell heterogeneity in functionalis I and homogeneity in functionalis II. Also a distinctive glandular cell type occurs in basalis III and IV that is recognized by a highly irregular cisternal rough endoplasmic reticulum that permeates the cytoplasm. During progesterone dominance, ultrastructural differences exist among the 4 zones except for similarity between the epithelial cells of functionalis II and basalis III. Postovulatory epithelial cells of functionalis I and II and basalis III become postmitotic via progesterone inhibition but intracellular differentiation continues progressively. Postovulatory epithelial mitotic activity in basalis IV escapes progesterone inhibition as the [3H]thymidine labeling index continues to increase from 1 to 12% during the menstrual cycle (Padykula et al.: Reprod., 30(Suppl.1):92 (Abstr. 123), 1984). This post-ovulatory proliferation coupled with progressive differentiation in basalis IV may represent a stem-progenitor set of cells for postmenstrual endometrial regeneration or alternatively for creation of the maternal placenta.  相似文献

Myocardial cells were isolated after treatment with collagenase (0.05%) and hyaluronidase (0.1%) by discontinuous-gradient centrifugation on 3% Ficoll. Nuclei derived from these myocardial cells were then fractionated on a discontinuous sucrose density gradient with the following steps: (I) 2.0M/2.3M, (II) 2.3M/2.4M, (III) 2.4M/2.5M, (IV) 2.5M/2.6M, and (V) 2.6M/2.85M. The myocardial nuclei were sedimented in the interfaces of gradient fractions (II) and (III). Nuclei from whole ventricles that had been treated with the enzymes before isolation sedimented into five major subsets of nuclei. These findings suggest that nuclei sedimented in the isopycnic gradient at fractions (II) and (III) are most probably derived from myocardial cells. However, this procedure is laborious and lengthy, and the recovery of myocardial-cell nuclei is low. An alternative method was developed to isolate an enriched fraction of myocardial-cell nuclei from whole ventricular tissue without exposing the tissues to enzyme digestion. These ventricular nuclei could be fractionated into five nuclear subsets by using the same discontinuous sucrose density gradient as that described above. The content of DNA, RNA and protein per nucleus for each band was determined. Although the DNA content per nucleus was constant (10pg), that of RNA varied from 1.5 to 4.5pg and that of protein from 16 to 24pg. Nuclei from each band were examined by light-microscopy: large nuclei occurred in the ligher regions whereas smaller nuclei were found in the denser regions of the gradient. From the size distribution pattern of myocardial-cell nuclei compared with that of total ventricular nuclei, it was found that nuclear subsets (II), (III), and (IV) were similar to myocardial nuclei. Electrophoretic analyses of the proteins solubilized in sodium dodecyl sulphate/phenol or Tris/EDTA/2-mercaptoethanol/phenol obtained from each nuclear subset indicate that these fractions are similar, with limited qualitative differences. These findings indicate that isolation of an enriched fraction of myocardial-cell nuclei could be achieved by discontinuous-sucrose-density-gradient centrifugation.  相似文献
