We examined the effects of brassinolide (BL) and/or an auxin (indole-3-acetic acid) on ethylene production and elongation in the primary roots of maize (Zea mays). When these two hormones were applied exogenously, both increased ethylene production. Before the tenth hour after treatment began, the influence of IAA was more evident than that of BL; the reverse was found beyond 10 h. When these hormones were treated simultaneously, the increase in level of ethylene was greater than the sum of effects by each hormone. Such a positive interaction was also recorded for changes in the activity of ACC synthase and the expression of its gene. For ACC oxidase, however, the two hormones had no apparent influence. When applied separately, neither affected root elongation nor proton extrusion. However, when given in combination, both phenomena occurred. Our results suggest that BL interacts with IAA to promote ethylene biosynthesis and elongation in roots. Therefore, it is possible that brassinolide acts by inducing auxin, which then stimulates both ethylene production (at the early stage) and root development.  相似文献

Hypocotyl explants of Beta vulgaris L. were grown on defined agar media with different combinations of IAA and kinetin at varying concentrations of nitrogen or sucrose. The cultures were kept in light (18 h a day) at 27°C for 5 weeks. Root initiation and callus growth were recorded and the callus tissue was analysed for N and K. Root formation was found to increase with increasing nitrogen concentration (from 5 mM to 23.3 mM) in the medium at 10.0 mg/1 of IAA, whereas no stimulation was found at 0.1 mg/1 of IAA. When raising the sucrose level from 20 g/1 to 100 mg/1 at 10.0 mg/1 of IAA and 1.0 mg/1 of kinetin, root initiation was also stimulated. At a lower kinetin and auxin level, however, no increase was recorded. Callus growth was affected by changes in the nitrogen or sucrose concentration of the culture media. The nitrogen content of the callus tissue increased with rising nitrogen concentration of the media. When raising the sucrose level instead of the nitrogen level, the nitrogen content of the tissue decreased.  相似文献

Flooding of soil, tilting of seedlings, application of ethrelto stems, and combinations of these treatments, variously alteredthe rate of growth and stem anatomy of 2-year-old Pinus densifloraseedlings. Either flooding or tilting increased stem diametergrowth and induced formation of abnormal xylem. Whereas floodingdecreased the rate of dry weight increment of roots and needlesand increased growth of bark tissues, tilting of stems did not.However, tilting decreased the rate of height growth, stimulatedtracheid production, and induced formation of well-developedcompression wood with rounded, thick-walled tracheids, witha high lignin content but without an S3 layer in the tracheidwall. Ethylene appeared to have an important regulatory rolein stimulating growth of bark tissues as shown by thicker barkin flooded seedlings or those treated with ethrel. Ethyleneappeared to have a less important role in regulating formationof compression wood. Flooding increased the ethylene contentsof stems and induced formation of rounded, thick-walled tracheids.However, these tracheids lacked such features of well-developedcompression wood tracheids as a thick S2 layer, high lignincontent, and absence of an S3 layer. Furthermore, applicationof ethrel to vertical stems greatly increased their ethylenecontents but did not induce formation of well-developed compressionwood. Furthermore, ethrel application blocked development ofcertain characteristics of compression wood when applied totilted seedlings. For example an S3 wall layer was absent intracheids of tilted seedlings but present in tracheids of tilted,ethrel-treated seedlings. Also lignification of tracheids wasincreased on the under side of tilted stems, but reduced intilted, ethrel-treated seedlings, further de-emphasizing a directrole of ethylene in the formation of compression wood. Ethreltreatment induced formation of longitudinal resin ducts in thexylem whereas flooding or tilting of stems did not. The relation hetween the effects of kinetin and gibberellin on retardation of leaf senescence was studied. Leaf discs were incubated for five days in several hormone concentrations, the chlorophyll was extracted and its amount estimated spectrophotometrically. Investigation of leaves from actively growing plants of Taraxacum megalorrhizon and Tropaeolum majus showed that an interaction existed between the effect of both hormones. Leaf age, light intensity and day length had a marked effect on the degree of response to the hormonal treatments, hut the change in response effected by these conditions remained similar for both hormones. Possible interpretations of the interaction observed between the effects of kinetin and gibberellin are discussed.  相似文献

Effects of uniconazole-P, a triazole-type growth retardant,on endogenous levels of IAA, cytokinins, ABA and ethylene inrice seedlings were investigated. Endogenous levels of IAA andABA were similar between control and uniconazole-P-treated riceshoots. Evolution of ethylene was promoted slightly, being 1.8times greater under 0.3 ppm uniconazole-P treatment than thatof control. The most obvious effect was the increase of trans-Zand trans-RZ in shoots. Shoots treated with uniconazole-P (10mg/m2 nursery box) contained 3.4 times and 3 times more trans-Zand trans-RZ than control, respectively. No significant differencesof cytokinin levels were recognized in roots except for cis-RZ.The increase of ethylene and active forms of cytokinins, andthe decrease of gibberellin in the shoots may be the basis forphysiological phenomena caused by uniconazole-P, namely thepromotion of flowering in woody plants and the enhancement offemaleness in cucumber. Cytokinins are a class of plant growth regulators that regulate several developmental processes in plants, and recently their role in counteracting the deleterious effects of abiotic stresses has been noted. The impacts of kinetin (10 µM, KN; an artificial cytokinin) on growth, photosystem II photochemistry, and nitrogen metabolism in tomato seedlings exposed to two levels (UV-B1, ambient+?1.2 kJ m?2 day?1, and UV-B2, ambient+?2.4 kJ m?2 day?1) of enhanced UV-B radiation were analyzed under open field condition. The growth, pigment contents, carbonic anhydrase activity, photosynthetic O2 yield, and values of chlorophyll a fluorescence parameters: F v/F 0, F v/F m or φP0, ψ 0, φE 0, and PIABS declined, whereas the values of energy flux parameters (ABS/RC, TR0/RC, ET0/RC, and DI0/RC) of PS II, efficiency of water splitting complex (F 0/F v), and respiratory rate of O2 uptake increased under UV-B stress. Likewise, UV-B exposure at both doses significantly inhibited the activity of enzymes involved in nitrogen metabolism: nitrate reductase, nitrite reductase, glutamine synthetase, and glutamate synthase. In contrast, an enhancing effect on glutamate dehydrogenase activity was observed under UV-B stress. Exogenous KN resulted in a significant attenuation in UV-B-induced negative effects on growth, pigments, photosynthesis, and nitrogen metabolism. The study concludes that exogenous KN improved the growth performance of tomato seedlings by attenuating the damaging effects of UV-B radiation on photochemistry of PS II and nitrogen metabolism, and the alleviating effect against the low dose (UV-B1) of UV-B was more pronounced.  相似文献

This study explores the unique growth-regulatory roles of two naturally occurring auxins, indole-3-acetic acid (IAA) and 4-chloroindole-3-acetic acid (4-Cl-IAA), and their interactions with gibberellin (GA) during early pea (Pisum sativum L.) fruit development. We have previously shown that 4-Cl-IAA can replace the seed requirement in pea pericarp growth (length and fresh weight), whereas IAA had no effect or was inhibitory. When applied simultaneously, gibberellin (GA3 or GA1) and 4-Cl-IAA had a synergistic effect on pericarp growth. In the present study, we found that simultaneous application of IAA and GA3 to deseeded pericarps inhibited GA3-stimulated growth. The inhibitory effect of IAA on GA-stimulated growth was mimicked by treatment with ethephon (ethylene releasing agent), and the inhibitory effects of IAA and ethylene on GA-mediated growth were reversed by silver thiosulfate (STS), an ethylene action inhibitor. Although pretreatment with STS could retard senescence of IAA-treated pericarps, STS pretreatment did not lead to IAA-induced pericarp growth. Although 4-Cl-IAA stimulated growth whereas IAA was ineffective, both auxins induced similar levels of ethylene evolution. However, only 4-Cl-IAA-stimulated growth was insensitive to the effects of ethylene. Gibberellin treatment did not influence the amount of ethylene released from pericarps in the presence or absence of either auxin. We propose a growth regulatory role for 4-Cl-IAA through induction of GA biosynthesis and inhibition of ethylene action. Additionally, ethylene (IAA-induced or IAA-independent) may inhibit GA responses under physiological conditions that limit fruit growth.  相似文献

Effects of Low Irradiance Stress on Gibberellin Levels in Pea Seedlings   总被引:9,自引:0,他引:9  
Using gas chromatography-selected ion monitoring with internalstandards we analyzed endogenous levels of GA1, GA8, GA19, GA20,GA29, GA44 and GA53 in shoots of pea cv. Alaska grown underdifferent levels of irradiance: high irradiance, 386±70µmolm-2s-1, control (100%); medium (50%); low (10%); darkness (0%).The average plant heights for medium and low irradiance anddark grown plants were 157%, 275%, and 460% of the control plants,respectively. Plants grown in medium and low irradiance developedthe same numbers of internodes as control plants but plantsin darkness developed fewer internodes and exhibited suppressedleaf expansion. The endogenous levels of GA1 GA8 and GA29 werehigher in medium and low irradiance grown plants than thoseof the high irradiance control. In particular, the GA20 levelof low irradiance plants was markedly higher (7.6-fold) thanthat of control plants. In dark-grown plants GA1, and GA8 levelsalso slightly increased but GA20 and GA29 levels decreased andthe levels of GA19, GA44 and GA53 did not change. Feeding ofGA1, and a GA biosynthesis inhibitor (uniconazole) to plantsgrown at reduced irradiance and in darkness suggests that theresponsiveness of plants to GA1, also increased at low irradianceand in darkness. Although elongation growth is reduced by ethylene, swelling responses do not occur. Ethylene reduces neither transport nor metabolism of applied IAA in either mesocotyl or coleoptile. We propose that maintenance of high auxin levels within these tissues sustains polar transport and contributes to the relative insensitivity of maize to applied ethylene.  相似文献

In de-rooted seedlings of Amaranthus caudatus L., betacyanin synthesis induced by white light or cytokinin was inhibited by abscisic acid (ABA) or a mixture of gibberellins A4 and A7 (GA4/7). The GA4/7 and ABA effects were additive. Thus ABA inhibited the cytokinin action but had no effect on the gibberellin response.  相似文献

The effect of coumarin, IAA, ethylene, kinetin and gibberellic acid on roots of maize and wheat was investigated. Sterile attached and detached roots and isolated elongation zones were used. In some experiments a semi-sterile procedure was followed. The effects of the different regulators separately or in various combinations together with coumarin were studied on the root growth with regard to division, elongation and swelling of the cells. The ethylene production in isolated elongation zones was measured after treatment with coumarin, IAA, PCIB, kinetin, colchicine and dinitrophenol. The results show the following: 1) Each substance produces a specific morphologic pattern. 2) Changes in polarity were demonstrated for auxin-induced swelling in cell divisions and cell expansion and for coumarin-induced swelling in cell divisions. Other cell expansion in swollen parts was due to cylindric cells increasing in width while retaining their cylindric form. 3) Coumarin-induced inhibition could not be counteracted by IAA, PCIB, carbon dioxide, kinetin, gibberellic acid or Cycocel. 4) The ethylene production in isolated elongation zones increases noticeably after kinetin treatment, less strongly after auxin treatment and least after coumarin treatment. The production of ethylene does not seem to be correlated with the morphogenetic effect of the different substances. 5) The isolated elongation zones reacted to a) IAA and kinetin with an increase in length in some cases and b) gibberellic acid with a reduction of their width. 6) The inhibitory effect of coumarin on the growth in length of the elongation zones was diminished by kinetin. The swelling produced by coumarin in these zones was reduced by gibberellic acid. The effects just mentioned of kinetin and gibberellic acid were considered as indirect ones. - From the present findings it was concluded that concomitant effects of auxin, ethylene, cytokinins and gibberellins are not obligatory for coumarin to exert its morphogenetic effects on root growth. In discussing the results some pitfalls in studies of growth reactions after application of hormones to roots containing meristem were emphasized.  相似文献

In hypocotyl segments of mung bean (Phaseolus mungo L.) seedlings, exogenously supplied indoleacetic acid was rapidly conjugated mainly into indoleacetylaspartic acid, which was inactive in inducing ethylene production. Kinetin is known to stimulate indoleacetic acid-induced ethylene production. The mechanism of kinetin action on indoleacetic acid-induced ethylene production by hypocotyl segments of mung bean seedlings was studied in relation to indoleacetic acid uptake and indoleacetic acid metabolism. Kinetin enhanced indoleacetic acid uptake during the initial 2-hour incubation and markedly suppressed the conversion of indoleacetic acid to indoleacetic acid conjugates throughout the whole 7-hour incubation. As a result, there was more free indoleacetic acid and less conjugated indoleacetic acid in the segments treated with kinetin than in those receiving no kinetin. A close relationship was demonstrated between the rate of ethylene production and the level of free indoleacetic acid, which was regulated by kinetin.  相似文献

Paclobutrazol [(2RS,3RS)-1-(4-chlorophenyl)methyl-4,4-dimethyl-2-(1h-1,2,4-trizol-1-yl)penten-3-ol] effectively decreased vegetative growth of rice (Oryza sativa L.) seedlings and increased the chlorophyll content. The number of veins in a leaf, the calculated number of stomata per leaf, and the length of guard cells were not altered by the paclobutrazol treatment, suggesting an effect on cell elongation. The allocation pattern of carbohydrates was changed by either gibberellin (GA) or paclobutrazol treatment. GA3 induced more shoot growth and less accumulation of starch than the control and paclobutrazol-treated seedlings. Photosynthetic ability was not affected by either paclobutrazol or GA3 treatment. Paclobutrazol-treated plants allocated a smaller amount of photosynthates for vegetative shoot growth and stored more as starch in the crowns than the control and GA3-treated plants. The same starch degrading activity in the crown tissue of paclobutrazol-treated seedlings as in control plants suggests that the accumulated starch is utilized in a normal activity for growth including leaf emergence, tiller formation, and root production, resulting in improved seedling quality. Factorial experiments using the three variables nitrate, ammonium, and kinetin at six different concentrations each (nitrate 4.64 to 215 mM; ammonium 2.15 to 100 mM; and kinetin 0.1 to 4.64 mg/l) were set up to measure the effects of each of these factors, and their interactions, on the fresh weight, protein, and enzyme activities of callus of Paul's Scarlet Rose. Optimum fresh weight values were obtained with nitrate at 46.4 mM. Ammonium inhibited growth at concentrations above 2.15 mM, and kinetin had no significant effect. Significant interaction between nitrate and ammonium effects on growth was found. Kinetin did not interact significantly with either nitrate or ammonium to influence the fresh weight. The specific activity of glutamate dehydrogenase (NAD) in the aminating reaction increased with increasing ammonium concentrations to 21.5; at higher concentrations the activity remained high. Glutamine synthetase specific activity was constant over a large range of nitrate and ammonium concentrations, increasing only when nitrate went from 46.4 to 100 mM. Glutamine synthetase was sensitive to the nitrate: ammonium interaction. Specific activity decreased at progressively higher ammonium levels when nitrate concentration increased. No glutamate synthase activity was detected at optimal nitrate concentrations.  相似文献
