Males of the cucujid grain beetles, Cryptolestes ferrugineus (Stephens), C. pusillus (Schönhen), C. turcicus (Grouvelle), Oryzaephilus mercator (Fauval) and O. surinamensis (L.), produce seven structurally related macrolides (“cucujolides”). Species-specific combinations of these macrocyclic lactones function as aggregation pheromones. To study the biosynthesis of these cucujolides, mixed sex cultures of the rusty grain beetle, C. ferrugineus, were fed oats impregnated with radiolabelled substrates. Culture volatiles were trapped on Porapak Q, eluted by back-flushing with solvent, concentrated and purified to constant specific activity by high performance liquid chromatography and preparative gas chromatography. The incorporation of radiolabelled acetate and mevalonate into cucujolide I [4(E),8(E)4,8-dimethyldecadien-10-olide] provided evidence that this pheromone is of terpenoid origin. The efficient incorporation of radiolabelled fatty acid substrates into cucujolides II 3(Z)dodecen-11-olide], III 5(Z)tetradecen-13-olide], IV [3(Z),6(Z)dodecadien-11-olide] and V [5(Z),8(Z)tetradecadien-13-olide] provided evidence that these cucujolides are of fatty acid origin. Radiolabelled palmitic acid was incorporated into cucujolides II, III, IV and V; radiolabelled oleic acid was incorporated into II and III; and radiolabelled linoleic acid was incorporated into IV. Radiolabelled acetate was incorporated into cucujolides II, III and V, indicating that C. ferrugineus can produce these cucujolides de novo.  相似文献

Sexual characters may reveal the quality of a potential mate, including the mate's level of infection with parasites. Females that prefer males with low levels of infection or no infection may benefit in several ways. Direct benefits may include avoidance of infection, acquistition of larger nuptial gifts or enhancement in fecundity due to differences in male fertility. Females may also benefit indirectly by producing offspring that are more resistant to infections. We measured female preference for odours produced by male grain beetles, Tenebrio molitor, that were either infected by a tapeworm, Hymenolepis diminuta, or uninfected. This parasite is not transmitted directly between conspecifics. Females were attracted to odours of all males, but they were less attracted to those from parasitized males. To the contrary, females were preferentially attracted to infected females. Males did not show any biased attraction to odours from infected and uninfected male beetles. Females that mated with highly infected males produced fewer offspring than females mated to uninfected males, indicating parasitic infection inflicts multiple costs to males. The number of potential annual generations of the rusty grain beetle,Cryptolestes ferrugineus, was simulated in wheat stored in granaries for all crop districts in the prairie provinces of Canada each year from 1952 to 1990 using a population dynamic model driven by ecological variables. Granary size was assumed to be 6 m in diameter. Historical data for temperatures at harvest and times when storage began were used in the simulation model. A second model, which predicted the rate of temperature change at the centre of a 6-m-diameter bulk of wheat, determined environmental parameters for the population dynamic model. (Grain moisture content was assumed constant at 14.5% wet mass basis.) The combined model shows that the initial storage temperature is the most important factor responsible for predicting the number of generations and levels of infestation ofC. ferrugineus. This finding was largely validated by historical grain storage and infestation data. For various years initial grain temperature ranged from 17.7 to 37.4 °C and harvest dates were between 1 August and 20 October. The number of generations annually in simulations based on field conditions ranged from 0.35 to 6.77 with a mean of 3.29. Three or more generations result in a severe infestation and every year at least three simulated generations were completed in some crop districts. In one year, at least three generations were completed in every crop district. Harvest temperature and date permit prediction of crop districts that will potentially have the largest populations of C.ferrugineus so that early monitoring of wheat for infestations can be targeted to areas most at risk. The biosynthesis of chrysomelidial and plagiodial was studied in the rove beetle subtribe Philonthina (Staphylinidae). Glandular homogenates were found to convert synthetic (2E,6E)-[trideuteromethyl-5,5-(2)H(5)]octa-2,6-diene-1,8-diol (10) into nor-chrysomelidial (14) and nor-plagiodial (13). The overall transformation requires; i) oxidation of the substrate at C(1) and C(8), ii) cyclization of the resulting dialdehyde to nor-plagiodial followed by iii) isomerization to give nor-chrysomelidial. The oxidase requires molecular oxygen as a cofactor and operates with removal of the pro-R hydrogen from C(1) and C(8) of synthetic (1R,8R,2E,6E)-[1,8-(2)H(2)]-2,6-dimethyl-octa-2,6-diene-1,8-diol (15), producing a dialdehyde along with H(2)O(2). Unlike enzymes from iridoid-producing leaf beetle larvae, the Philonthus enzyme is able to oxidize saturated substrates such as citronellol. Crude protein extracts prepared from Philonthus glands by ammonium sulfate precipitation, were found to produce hydrogen peroxide at a rate of 0.085+/-0.003 ng H(2)O(2) (ng protein)(-1) hr(-1) with nerol as an oxidase substrate. The cyclase operates with opposite stereochemistry to the enzyme(s) from Phaedon cochleariae and other herbivorous leaf beetles, specifically removing the C(5)-H(R) hydrogen atom from (4R,5S,2E,6E)-[4,5-(2)H(2)]-2-methyl-octa-2,6-diene-1,8-diol (17). These findings have enabled us to construct a detailed account of iridoid biosynthesis in rove beetles, which resembles the biosynthetic route in leaf beetle larvae, but exhibits distinct stereochemical differences.  相似文献

We report on juvenile hormone (JH) biosynthesis from long‐chain intermediates by specific reproductive tissues and the corpora allata (CA) prepared from adult longhorned beetles, Apriona germari. The testes, male accessory glands (MAGs), ovaries, and CA contained the long‐chain intermediates in the JH biosynthetic pathway, farnesoic acid (FA), methyl farnesoate (MF), and JH III. The testes and ovaries, but not CA, produced radioactive JH III after the addition of 3H‐methionine and, separately, unlabeled methionine, to the incubation medium. We inferred that endogenous FA is methylated to MF in the testes and ovaries. Addition of farnesol led to increased amounts of FA in the testes, MAGs, ovaries, and CA, indicating oxidation of farnesol to FA. Addition of FA to incubation medium yielded increased JH III, again indicating methylation of FA to MF in the testes, MAGs, ovaries, but not CA. Addition of MF to incubation medium also led to JH III, from which we inferred the epoxidation of MF to JH III. JH biosynthesis from farnesol in the testes, MAGs, and ovaries of A. germari proceeds via oxidation to FA, methylation to MF, and epoxidation to JH III. This is a well‐known pathway to JH III, described here for the first time in reproductive tissues of longhorned beetles. © 2010 Wiley Periodicals, Inc.  相似文献

Wheat stored in upright concrete bins at seven grain elevators in central Kansas was sampled intermittently for insects over a 2.5-yr period by collecting samples from the upper half of the grain mass, from the discharge spout at the base of the bins, and from residue remaining in empty bins before the 2000 wheat harvest. Samples were taken from the grain mass with a power vacuum sampler (PV) and from the discharge spouts (DS) by dropping grain onto the reclaim belt beneath the bins. The density and species distribution in the residue samples were compared with those found in the DS samples and samples from the grain mass (PV). Cryptolestes spp. dominated the insect populations in all types of samples, constituting >40% of all insects in the PV samples in three of five time periods and >60% of all insects in DS samples in four of the five time periods. Cryptolestes spp. was an early colonizer, being found in the grain mass shortly after new grain was added. Rhyzopertha dominica appeared to be slower to colonize grain and grain residue, but sometimes developed large populations (i.e., 2.4 +/- 0.7 adults/kg between July and December 2000). Sitophilus spp. weevils often were present in grain masses, were often abundant in grain in the discharge spouts (i.e., 11.1 +/- 2.9 adults/kg between July and December 2000), and were abundant in grain residue in empty bins in May/June 2000 (5.3 +/- 0.7 adults/kg). Differences in density and species distribution of insects in grain in the upper part of the grain mass and those in the discharge spouts indicated that the populations were not closely related. Grain in discharge spouts usually was densely infested, and parasitic wasps, natural enemies of several of the beetles, were found when the density of the pest insects was greater than approximately 10/kg. The population of natural enemies appeared to increase when the density of pest insects increased after a lag of about one month, and decreased when the population of pest insects decreased. Grain in discharge spouts appeared to provide an incubation chamber for pest insects, and removing this grain periodically should reduce the resident populations. Residue in empty bins often was densely infested compared with samples from the grain masses. Cleaning the empty bins before refilling with newly-harvested wheat resulted in a significantly-reduced density of pest insects in discharge spouts later, and the effect lasted at least 12 wk after filling.  相似文献

The movement and distribution of adult Cryptolestes ferrugineus (Stephens) (Coleoptera: Laemophloeidae) in grain provide important information for detection of insect pests and for simulations of their distribution in grain bins. Adult movement and distribution were determined in 100 by 100 by 1000-mm wheat (14.5 +/- 0.2% moisture content) columns at four insect densities, three temperature gradients, and dynamic (changing) temperature conditions. Insect density was a minor factor influencing insect movement and distribution in grain columns with temperature gradients. Dispersal resulted in a uniform distribution at a higher insect density (higher than two adults per kilogram of wheat), and aggregation occurred at a low insect density. Adults wandered in the first 6 h after introduction, and there were fewer adults wandering in the vertical direction than in the horizontal direction. Adults moved faster in the vertical direction than in the horizontal direction, and the maximum speed of the movement was 6 m/d in the horizontal direction, and >10.8 m/d in the vertical direction through wheat. Adults could detect temperature gradients in <1 h and preferred warmer temperatures when they had a choice. Insect distribution in horizontal wheat columns at any temperature gradient was unstable for 24 h. Twenty-four hours after introduction, adults gradually overcame their positive geotactic behavior if the upper temperature was more biologically suitable or was not <27.5 degrees C. Adults responded faster to higher temperature gradients than to lower temperature gradients. There was a similar pattern of adult distribution in 144 h.  相似文献

松墨天牛Monochamus alternatus Hope是传播松材线虫病的主要媒介,引诱剂是抑制松墨天牛种群数量的无公害调控有效方式之一。本研究通过在中国浙江富阳的林间试验测定了6种小蠹类引诱剂与1种天牛引诱剂不同组合联用对松墨天牛成虫以及其他蛀干害虫的诱捕效果。结果表明: 横坑切梢小蠹Tomicus minor引诱剂3-carene-10-ol、云杉八齿小蠹Ips typographus引诱剂2-methyl-3-buten-2-ol、欧洲榆小蠹Scolytus multistriatus引诱剂4-methyl-3-heptanol和纵坑切梢小蠹T. piniperda引诱剂verbenol分别与松墨天牛引诱剂联用后,对松墨天牛的引诱效果无显著性差异(P<0.05)。西部松大小蠹Dendroctonus brevicomis 引诱剂exo-brevicomin或黄杉大小蠹D. pseudotsugae引诱剂1-methylcyclohex-2-en-l-ol与松墨天牛引诱剂联用后对松墨天牛的诱捕效果有减弱的趋势。另外,与单独使用中国天牛引诱剂相比,小蠹类引诱剂与其联用后在诱捕其他蛀干害虫的种类和个体数量上均有所减少。  相似文献

Auxin is a crucial phytohormone, controlling multiple aspects of plant growth and responses to the changing environment. However, the role of local auxin biosynthesis in specific developmental programs remains unknown in crops. This study characterized the rice tillering and small grain 1 (tsg1) mutant, which has more tillers but a smaller panicle and grain size resulting from a reduction in endogenous auxin. TSG1 encodes a tryptophan aminotransferase that is allelic to the FISH BONE (FIB) gene. The tsg1 mutant showed hypersensitivity to indole‐3‐acetic acid and the competitive inhibitor of aminotransferase, L‐kynurenine. TSG1 knockout resulted in an increased tiller number but reduction in grain number and size, and decrease in height. Meanwhile, deletion of the TSG1 homologs OsTAR1, OsTARL1, and OsTARL2 caused no obvious changes, although the phenotype of the TSG1/OsTAR1 double mutant was intensified and infertile, suggesting gene redundancy in the rice tryptophan aminotransferase family. Interestingly, TSG1 and OsTAR1, but not OsTARL1 and OsTARL2, displayed marked aminotransferase activity. Meanwhile, subcellular localization was identified as the endoplasmic reticulum, while phylogenetic analysis revealed functional divergence of TSG1 and OsTAR1 from OsTARL1 and OsTARL2. These findings suggest that TSG1 dominates the tryptophan aminotransferase family, playing a prominent role in local auxin biosynthesis in rice.  相似文献

Simulation studies were conducted to determine temperature accumulations below defined thresholds and to show the impact of controlled aeration on populations of the rusty grain beetle, Cryptolestes ferrigineus (Stephens), a major secondary pest of stored wheat, Triticum aestivum (L.). Recorded data from weather stations in Texas, Oklahoma, Kansas, eastern New Mexico, and eastern Colorado (356 total) were used to determine hours of temperature accumulation below 23.9 degrees C in June and July, 15.6 degrees C in September and October, and 7.2 degrees C in December. At an airflow rate of 0.0013 m3/s/m3 (0.1 cubic ft3/min/bu), which requires 120 h of temperatures below the specified threshold to complete an aeration cycle, summer cooling at 23.9 degrees C in bulk-stored wheat could be completed throughout the hard red winter wheat zone except for extreme southern Texas. An early-autumn cooling cycle at 15.6 degrees C could not be completed throughout most of Texas and Oklahoma before the end of September. The late-autumn cooling cycle could be completed in all states except Texas by the end of November. Five geographic regions were delineated and the times required for completion of the summer, early-autumn, and late-autumn cooling cycles within each region were estimated. Population growth of the rusty grain beetle was modeled for San Antonio, TX; Abilene, TX; Tulsa, OK; Topeka KS; and Goodland, KS, by predicting the numbers of adults in the top, outer middle, outer periphery, and the center of the bin during a 1-yr storage season. Populations of C. ferrugineus in San Antonio and Austin were predicted to exceed the Federal Grain Inspection Service (FGIS) threshold of two beetles per kilogram of wheat in all four levels of the bin during late autumn, decline during the winter, and increase the following spring. In Midland, TX, and Oklahoma City, OK, populations were predicted to exceed the threshold only in the top and outer middle of the bin, whereas populations in the Kansas locations were not predicted to exceed the threshold at any time.  相似文献

Summary The microflora of sugary kefir grains was principally mesophilic and consisted chiefly of lactic acid bacteria [Lactobacillus casei, Lactobacillus hilgardii (=brevis),Leuconostoc mesenteroides ssp.dextranicum, Streptococcus lactis] and a small proportion of yeasts (Zygosaccharomyces florentinus, Torulospora pretoriensis, Kloeckera apiculata, Candida lambica andC. valida). Few coliforms and faecal streptococci were observed. Observation by scanning electron microscopy revealed that the filamentous yeasts adhered to the bacteria on the periphery of the grain.Lactobacillus hilgardii, the single microorganism isolated which was able to produce a gelling polysaccharide, was important in the biosynthesis of the grain. Pieces of gel produced by this strain, and transferred in a yeast extract-sucrose solution, grew and resembled the household kefir grains. This represents a new, cheap way of producing immobilized cells by self-embedding in a neutral polysaccharide.

Résumé La microflore des grains de kefir sucré est essentiellement mésophile et comprend principalement des bactéries lactiquesLactobacillus casei, Lactobacillus hilgardii (= brevis),Leuconostoc mesenteroides ssp.dextranicum, Streptococcus lactis ainsi qu'une petite proportion de levures (Zygosaccharomyces florentinus, Torulospora pretoriensis, Kloeckera apiculata, Candida lambica etC. valida). On a observé peu de coliformes et de streptocoques fécaux. L'observation au microscope électronique à balayage a révélé que les levures filamenteuses adhèrent aux bactéries sur la périphérie du grain.Lactobacillus hilgardii, le seul microorganisme isolé susceptible de produire un polysaccharide gélifiant, est important dans la biosynthèse du grain. Des morceaux de gel produit par cette souche et transférés dans une solution d'extrait de levure et de saccharose, croissent et ressemblent aux grains de kefir domestique. Ceci représente une manière nouvelle, peu coûteuse de produire des cellules immobilisées par auto-piégeage dans un polysaccharide neutre.

The parasitic wasp Cephalonomia tarsalis parasitizes larvae of the saw-toothed grain beetle Oryzaephilus surinamensis, which feed on wheat grains. In contrast to most other host–parasitoid systems studied so far, the grain beetles are highly mobile within their habitat, bulk of grains in grain stores. This should increase the wasps’ problem to locate the hosts. To study the host-finding strategy of C. tarsalis females, the reaction of wasps to different grain and host-derived odour sources was tested in a four-chamber-olfactometer. These experiments revealed that wasps were attracted by healthy grains and mechanically damaged grains. In direct comparison, healthy and mechanically damaged grains are equally attractive. Both potential sources of host-derived odours, host faeces and trail-traces of larvae on filter paper were attractive to the wasps. The response to trail-traces vanished 30 min after larvae had been removed from the filter paper. With respect to the specificity of the odours, it turned out that wasps were attracted to odours from the seed–host complexes from O. surinamensis and Oryzaephilus mercator and the non-host complex of larvae of the granary weevil Sitophilus granarius in wheat grains. Odours from the seed–host complex were preferred. From these results, we hypothesize that host habitat location in C. tarsalis is achieved by using grain-derived odours. Within the habitat, wasps search for kairomones from host faeces and host trails. Following these larval trails finally leads wasps to their hosts.  相似文献

In the naucorid bug, Alphelocheirus, the plastron hairs are twice as thick and nearly twice as dense (ca. 4 × 106/mm2) as they had been thought to be by previous workers. From experiments and calculations it seems clear that when the plastron is subjected to excess pressures it is wetted long before there is any question of the collapse of the hair pile itself. The plastron of Aphelocheirus is thus like the plastrons of other insects in that it is wetted long before the structures supporting the air film collapse.A plastron has been independently evolved in at least five subfamilies of the Naucoridae. A plastron is here recorded for the first time in bugs of the family Helotrephidae.It has been claimed that the plastron-bearing elmid beetles are unable to fly. Many, if not most, of these beetles fly after they emerge from their pupal cells. However, once they have begun to live under water they cease to be able to fly: the flight muscles degenerate, and this degeneration seems to be irreversible.The structure of the plastron scales of several kinds of weevils is described. The resistance of the plastron of the rice water weevil, Lissorhoptrus, to wetting at excess pressures is examined. An explanation is advanced to account for the fact that weevils and other plastron-bearing beetles that live in still waters can often swim whereas those, like elmids and dryopids, that live in running waters cannot swim.  相似文献
