1. Effect of pH on liver lactate dehydrogenase (LDH) and its isozymes was examined in the goldfish acclimated to different temperatures and some purification of the LDH was attempted. 2. The optimal pH and the Km value at 30 degrees C of the enzyme were independent of acclimation temperature. 3. the optimal pH of isozyme was more basic in the order of LDH-1, LDH-2, LDH-3, LDH-4 and LDH-5. Km values of isozymes at 30 degrees C were higher in the order of LDH-1, LDH-3 and LDH-5. 4. There was no change in the enzyme activity during thermal acclimation.  相似文献

1. LDH activity and isozyme pattern were examined in the liver and epididymal fat pad of animals in 12 different sublines of the Upjohn Chinese hamster colony, which was established to produce animals with spontaneous diabetes. 2. Considerable divergence was observed and the animals could be divided into 3 groups according to LDH-H activity. Each group was significantly different from the other in epididymal fat pad LDH-1, 2,3 and 5 and liver LDH-3, 4 and 5. 3. The variance in LDH isozyme pattern bore no relationship to the state of diabetes but appeared to arise from other genetic determinants. However, within a single subline, a significant correlation between blood sugar and epididymal fat pad LDH-5 was observed.  相似文献

Summary Lactate dehydrogenase (LDH) isozyme patterns were analysed by polyacrylamide (PAA) slab gel electrophoresis in extracts prepared from various rabbit skeletal muscles of defined fibre composition and by PAA microelectrophoresis of microdissected, histochemically typed single muscle fibres. The results obtained by electrophoresis of whole muscle extracts generally agreed with the data obtained from single fibre electrophoresis, i.e. the LDH isozyme pattern corresponded to that of the predominant fibre type. Type I Fibres from soleus and semitendinosus muscles were characterized by a unique pattern of all 5 LDH isozymes with a predominance of LDH-1, 2 and 3. The major fraction (80%) of the type II fibres from extensor digitorum longus and tibialis anterior muscles contained only LDH-5 (M4). About 20% of the type II fibres contained in addition to LDH-5 small amounts of LDH-4 and LDH-3. The fraction of fibres containing LDH-5, LDH-4, and LDH-3 was similar (ca. 20%) in the histochemically defined IIA and IIB subpopulations In view of the fact that the major fractions of rabbit IIB fibres display low and of IIA fibres high aerobic oxidative capacities (Reichmann and Pette 1982), these data indicate that the expression of the H-subunit of LDH is not correlated with the aerobic-oxidative capacity of the fibre. It also appears not to be correlated with the presence of different myosin isoforms in IIA and IIB fibres.  相似文献

Uterine secretions were collected from 20 mature cows during estrus (day 0), metestrus (day 5), diestrus (day 10) and proestrus (day-1). Lactate dehydrogenase (LDH) and LDH isoenzymes activity were evaluated. No significant cyclic variations of LDH activity was found in the uterine secretions while the mean of the enzyme activity was higher during the estrogenic period of the cycle. The relative activity of LDH-1, LDH-2 and LDH-3 isoenzymes were higher during proestrus and estrus whereas LDH-5 activity was more important during metestrus. The LDH-3 seems to have the higher relative activity in uterine secretions of the cow.  相似文献

1. Electrophoresis of extracts of turkey spermatozoa for lactate dehydrogenase activity revealed the usual five tissue LDHs (LDH-1 to LDH-5). 2. The presence of LDH-X (the spermatozoan-specific isozyme) was not obvious. 3. Only one band was present on electrophoresis of fowl spermatozoan extracts and it coincided with LDH-1 (heart type). 4. Kinetic investigations, the use of inhibitors and the heat-stability test confirmed that the fowl spermatozoan LDH was probably LDH-1 and not LDH-X.  相似文献

Extensive extraction studies with Triton X-100 revealed only LDH-1 (B4) but no trace of LDH-5 (A4) in one-cell and two-cell mouse and rat embryos. The LDH isozyme pattern of preimplantation mouse embryos changes from the maternally inherited B subunit isozyme (LDH-1) to a pattern dominated by LDH-5 when mouse blastocysts are cultured under conditions that prevent hatching but allow trophoblast giant cell transformation. During differentiation of mouse blastocysts in vitro, implantation is therefore not essential for the appearance of the A subunit form of LDH (LDH-5) coded for by the embryonic genome. A total of 65 cases of ovarian tumor admitted for surgical intervention were prospectively enrolled in this study, including 40 cases with benign ovarian tumors and 25 cases of ovarian cancer. The 25 malignancies were comprised of 22 cases of common epithelial origin and three cases with Krukenberg tumors. Serum total LDH level and its isozyme activity were measured preoperatively in each patient. The mean value (+/-SD of serum LDH in the malignant group was 876.3 (+/- 450.4) IU/1, which was significantly higher than 364.8 (+/- 87.9) IU/1 in the benign group. Twenty-two cases of ovarian malignancy (88%) and 6 cases of the benign group (15%) had elevated serum LDH levels above 450 IU/1. Analysis of the LDH isozyme pattern demonstrated that there was a significant shift to LDH-4 and LDH-5 fractions in ovarian malignancy when compared to the benign group (with the mean value of 9.5%, 14.6% vs. 6.5% and 4.6%, respectively). The elevated LDH level could yield 88% and 85% in sensitivity and specificity for detecting ovarian malignancy, while the abnormal isozyme pattern could yield 84% and 77.5%, respectively. From combined analysis of total LDH levels and isozyme patterns, the true positive detection rate and true negative detection rate could both reach 100%. In conclusion, the present findings imply that total serum LDH level and isozyme activity measurement may provide significant aids in evaluation and detection of human ovarian cancers among patients with ovarian tumors.  相似文献

1. The concentration of myoglobin (Mb) and the isozymic distribution and activity of lactate dehydrogenase (LDH) in heart and pectoralis muscle were investigated at three stages of maturation of the Pigeon Guillemot, Cepphus columba. 2. Mb is not detectable in chick pectoralis; it is present in fledgling pectoralis muscle and increases four-fold in adult pectoralis. Mb concentration in heart muscle is similar in chick and fledgling and doubles in the adult. 3. LDH activities in pectoralis muscle of fledgling and adult increase to about three times that of the chick. LDH activities in heart of chick, fledgling and adult are similar to one another. 4. All five isozymes of LDH are present in both heart and pectoralis muscle at all stages; the heart muscle shows predominantly LDH-1 isozyme, and the pectoralis, LDH-5. The relative amounts of the five isozymes in the heart extract were constant during maturation but pectoralis LDH isozymes changed during maturation towards a more even distribution of the five isozymes in the adult. 5. Changes in Mb and LDH in the Pigeon Guillemot correlate with the animal's maturation from a sedentary nest sitter to an active diver and flyer. The adult pectoralis muscle probably has both aerobic function for wing-propelled short dives and flying and anaerobic capacity for longer dives.  相似文献

Changes in the activity of serum lactate dehydrogenase (LDH) and the serum LDH isozyme pattern in healthy young men given a high carbohydrate diet (480-636 g/day, 80% of the total energy) for 21 days were examined. Serum total LDH activity showed no significant change in four healthy young volunteers who received high carbohydrate diet for 21 days. However, the percentage of LDH-4 increased significantly (P less than 0.01) from 8.5 +/- 2.4 to 10.9 +/- 1.9% after 21 days, as did also the percentage of LDH-5 (P less than 0.01) from 5.1 +/- 1.9 to 10.7 +/- 2.9%. The percentage of M-type LDH activity increased 30% during the experiment (P less than 0.05). It is concluded from the results that the high carbohydrate intake affects the percentage of LDH-4 and LDH-5, but not the total serum LDH activity.  相似文献

Lactic dehydrogenase (LDH) and alpha-hydroxybutyrate dehydrogenase (HBD) and LDH isozyme patterns were studied in serum from newborns and patients with ovarian cancer or myocardial infarction. LDH and HBD activities from newborns and patients with ovarian cancer or myocardial infarction were significantly increased, compared with those from patients with benign ovarian tumor. These increases were accompanied with a decrease of LDH-H and an increase of LDH-M in serum from newborns and patients with ovarian cancer, while an increase of LDH-H in serum from patients with myocardial infarction was dominant. However, the raised HBD activities in serum from patients with benign ovarian tumor did not affect the LDH isozyme patterns. From analysis of linear regression, a negative correlation between LDH-1 or -2 and HBD activity in serum from patients with ovarian cancer was observed while there was a positive correlation between LDH-4 and HBD activity. Similar patterns in serum from newborns were observed. On the other hand, a positive correlation between LDH-1 and HBD activity and a negative correlation between LDH-4 and HBD activity were found in serum from patients with myocardial infarction.  相似文献

Summary Adult human testicular tissue contains up to six previously undescribed lactate dehydrogenase (LDH) isoenzymes in addition to the five LDH isoenzymes normally found and the sixth found in spermatogenic cells and spermatozoa, LDH-X. Additional LDH isoenzymes were also found in spermatozoa but not in seminal fluid or in serum. After electrophoresis one additional LDH isoenzyme of testicular tissue was localized between LDH-1 and LDH-2, two between LDH-2 and LDH-3, two between LDH-3 and LDH-4, and two between LDH-4 and LDH-5. These localizations indicate that the additional LDH isoenzymes are tetramers combining the A and B subunits of the five normal LDH isoenzymes and the C subunit of LDH-X. The additional LDH isoenzymes may be important in the metabolism of spermatogenic germ cells and spermatozoa.  相似文献

To determine whether long-term reductions in neuromuscular activity result in alterations in metabolic capacity, the activities of oxidative, i.e., succinate dehydrogenase (SDH) and citrate synthase (CS), and glycolytic, i.e., alpha-glycerophosphate dehydrogenase (GPD), enzyme markers were quantified in rat soleus muscles 1, 3, and 6 mo after a complete spinal cord transection (ST). In addition, the proportional content of lactate dehydrogenase (LDH) isozymes was used as a marker for oxidative and glycolytic capacities. The myosin heavy chain (MHC) isoform content of a fiber served as a marker of phenotype. In general, MHC isoforms shifted from MHC1 toward MHC2, particularly MHC2x, after ST. Mean SDH and CS activities were higher in ST than control at all time points. The elevated SDH and CS activities were indicative of an enhanced oxidative capacity. GPD activities were higher in ST than control rats at all time points. The increase in activity of SDH was larger than GPD. Thus the GPD-to-SDH (glycolytic-to-oxidative) ratio was decreased after ST. Compared with controls, total LDH activity increased transiently, and the LDH isozyme profile shifted from LDH-1 toward LDH-5, indicative of an enhanced glycolytic capacity. Combined, these results indicate that 1) the metabolic capacities of soleus fibers were not compromised, but the interrelationships among oxidative and glycolytic capacity and MHC content were apparently dissociated after ST; 2) enhancements in oxidative and glycolytic enzyme activities are not mutually exclusive; and 3) chronic reductions in skeletal muscle activity do not necessarily result in a reduced oxidative capacity.  相似文献

利用SOD和LDH同工酶电泳分析,研究大豆黄酮对衰老小鼠的抗氧化作用。结果显示大豆黄酮没有改变SOD和LDH同工酶谱的特征,但对因衰老引起的小鼠脑组织LDH和SOD同工酶活性、各组分的相对活性和比活力的变化有不同程度的改善作用,即LDH同工酶中LDH-2、LDH-3的活性明显下降,LDH-1的活性下降最为明显,而LDH-4的活性有所下降,但不显著,LDH-5的活性几乎没有变化,SOD同工酶的SOD-1和SOD-2的活性有不同程度的升高。这表明大豆黄酮是通过抑制LDH同工酶H亚基的合成来降低LDH的活性,而对M亚基的合成没有影响,并且能够促进SOD同工酶SOD-1和SOD-2的合成,不影响其遗传稳定性。  相似文献

The lactate dehydrogenase isoenzyme pattern has been determined in different murine lymphocytic cell populations. In each cell population, the LDH activity was predominantly found in the LDH-4 and LDH-5 fractions. The percentage LDH-5 activity was significantly higher in B cells than in T cells. The same is true for lymphocytes from the spleen versus lymph node lymphocytes. The percentage LDH-5 activity is significantly higher in peripheral T lymphocytes than in thymocytes. Enrichment of the more mature thymocytes of the thymocyte cell pool by either cortisone treatment in vivo or gradient centrifugation on bovine serum albumin (BSA) results in a decrease of LDH-1 and LDH-2 fractions. In the cortisone-treated group, the shift in the LDH pattern is accompanied by a significant increase of LDH-5 and LDH-4 fractions, whereas in the BSA group only the LDH-4 fraction increases.  相似文献

The pattern of lactate dehydrogenase isozymes was investigated by means of electrophoresis in erythrocytes of CBA/Lac and DBA/2J mice homozygous for b and a alleles of the Ldr-1 locus. It is found that differences in the pattern of LDH isozymes, homozygous for the genes Ldr-1a and Ldr-1b, consist in increased activity of the isozyme LDH-4 in mice homozygous for the gene Ldr-1a (DBA/2J) within 12-14 days of postnatal development. Inhibition of the reaction between 125I-LDH-1 and the respective antibodies has demonstrated that increased LDH-4 activity during development is related to the higher content of B-subunits of LDH. It is suggested that the mechanism of the action of the gene Ldr-1 involves changes in the rate of the synthesis and degradation of B-subunit of LDH.  相似文献

Lactate dehydrogenase (LDH; E.C. 1.1.1.27) isozymes were compared in three inbred strains of mice, and two strains of wild mice, as well as the F1 hybrids and other genetic crosses involving two of the inbred strains. The strains examined were NZB/B1NJ, 129/J and C57BL/6J, Mus musculus molossinus and M. musculus castaneus. Genetic crosses were made between the xenotropic virus-producing NZB and the non-virus producing 129/J mice. Tissue specificity of LDH in these strains was studied using homogenates of kidney, liver, spleen and thymus. Polymorphism of the enzyme was studied by agarose gel electrophoresis. Enzyme polymorphism in the tissues of NZB and 129/J has not been previously reported. The liver and spleen tissues of 129/J showed the absence of LDH-1 and LDH-2 isozymes. Thymic homogenates of NZB showed a lack of expression of LDH-1, LDH-2 and LDH-3 isozymes. The F1, F2 and the backcross progeny from genetic crosses involving NZB, and 129/J mice showed an isozyme pattern more similar to the non-virus-producing 129/J strain than the virus-producing NZB. Evidence of genetic regulation at the LDH-B subunit appears to be the reason for the differential expression of the isozymes in NZB and 129/J strains. The other inbred strain of mice, C57BL/6J, also showed a greater similarity to the 129/J strain than NZB. The two strains of wild mice were similar in their expression of LDH-isozymes between each other and to the 129/J strain, with respect to the liver and spleen tissues.  相似文献

本文以聚丙烯酰胺凝胶电泳法分析华南兔(Lepus sinensis sinensis)11种组织乳酸脱氢酶同工酶的分布待征。分析结果:骨骼肌和肝组织5条同工酶带俱全;脑、肺、卵巢和盲肠等组织各含有4条谱带(LDH-1,-2,-3和-4);胃和肾组织含有3条谱带(LDH-1,-2,和-3);眼晶状体和睾丸组织也含有3条谱带,但前者是LDH-3,-4和-5,后者是LDH-1,-2和-5;谱带最少的是心肌组织,只有2条(LDH-1和-2)。此外,还对各组织中的亚基活性分布及电泳图谱特征进行了分析。  相似文献

Lactate dehydrogenase (LDH) isoenzyme profiles in human platelets and the sera of patients with type I and II diabetes mellitus and vascular complications, as well as normal subjects were measured utilizing a recently established, modified micromethod. LDH-3 was the predominating species in platelets (37.5 +/- 3.0%), with LDH-2, 1, 4 and 5 following in decreasing order of concentration. The LDH-3/LDH-4 ratio in platelets varied from 6.2 to 1.38. Type I and type II diabetic patients with vascular complications showed a significantly higher ratio for LDH-3/LDH-4 (3.99 +/- 1.20 for DM I, 2.16 +/- 0.25 for DM II patients) than the mean ratio for normal subjects (1.14 +/- 0.08). This platelet-specific LDH isoenzyme pattern may be the result of frequent in vivo platelet-vessel wall interactions in the diabetic patients whose platelets are known to be hyperaggregable in in vitro test systems. Since non-diabetic patients patients with vascular complications also displayed a similarly elevated LDH-3/LDH-4 ratio, a wider classification is preferable, although the measurement of the LDH isoenzyme pattern will be helpful in assessing diabetic vascular complications.  相似文献

Neoplastic thymocytes from rat thymic lymphoma-leukemias induced by the rat-adapted Gross leukemia virus (RAGV) were analyzed for a variety of differentiation markers. The neoplasms from individual rats all expressed the antigenic phenotype MP+, W3/13+, Thy-1+, RT-1+, RT-7+, W3/25-. However, approximately two-thirds of the neoplasms were positive for the OX 8 antigen, and one-third were negative. The OX 8- neoplasms only involved the thymus, whereas approximately 40% of the OX 8+ neoplasms involved the spleen as well as the thymus. Virtually all OX 8+ and OX 8- neoplastic cells contained terminal deoxynucleotidyl transferase (TdT), and both OX 8+ and OX 8- lymphomas expressed the lactate dehydrogenase (LDH)-5' isozyme and the primary, but not the secondary, ADA isozyme. This enzymatic phenotype is characteristic of thymocyte precursors, but not thymocytes. Our results therefore indicate that RAGV-induced lymphomas arise from transformed prethymic TdT+ cells which contain the LDH-5' and the primary ADA isozymes. These preleukemic cells presumably migrate to the thymus where they express the RT-7 pan-T-cell antigen and, in some instances, the OX 8 antigen during the development of overt leukemia. The OX 8+ neoplasms, being more differentiated than their OX 8- counterparts, then migrate to peripheral lymphoid tissues.  相似文献
