In this study, we aimed to produce a citrus fruit with no seed and with edible peel. Therefore, the seedless Yoshida navel oranges (Citrus sinensis ‘Yoshida navel’) and Jangsil kumquat (Fortunella japonica) with edible peel were used as the starting materials. Since two varieties of different genera and different flowering periods would be difficult to crossbreed, protoplast fusion was conducted using the polyethylene glycol method. Yoshida navel orange and Jangsil kumquat were used as an embryogenic callus line and a mesophyll line, respectively. The regenerated plants were analyzed by flow cytometry to identify tetraploids, which were further evaluated by polymerase chain reaction, using simple sequence repeat markers specific for the nuclear and cytoplasmic organellar DNAs of the two parents, to confirm allotetraploids and cybrids. Sixteen allotetraploids were finally produced and characterized phenotypically for leaf morphology and fruit quality. All allotetraploids contained mitochondria originating from Yoshida navel orange; 12 contained chloroplasts derived from navel orange whereas four contained chloroplasts of kumquat origin. We investigated both plant and fruit characteristics of allotetraploids derived from the protoplast fusion of navel orange and kumquat. The sizes of the allotetraploid leaves were intermediate between those of both parents. However, in contrast to the spindle-shaped leaves of both parents, those of the allotetraploids were obovate. Petiole wings were absent in the allotetraploids, a known kumquat trait. Similar to navel oranges, flowering time was mid-May, and pollen was sterile, whereas fruit size, external shape, soluble solids content, and acidity were all intermediate between the two parents. Peel thickness and the number of segments were similar to those of kumquat, and flesh weight was similar to that of navel orange. Collectively, these results indicated that the intergeneric allotetraploids derived from navel orange and kumquat inherited favorable traits of both parents and could be produced and selected despite a lack of extensive parental variation. This is one of the first reports showing fruiting results of protoplast-fused citrus plants. The study reported characteristics of the fruits from both the original plants as well as from the protoplast-fused plant. Detailed characteristics of the allotetraploid fruit produced by protoplast fusion would be very helpful for future polyploid studies.

Pollen grains of thirty one species and two varieties of the genus Caragana Fabr. were observed under LM and SEM.The results revealed that the pollen exine ornamentation of the species examined could be divided into two types: perforate and reticulate.The later type consists of two subtypes according to lumina size and muri width. In the first subtype the lumina is round with d iameter equal to muri width,and the exine is perforate in two polar areas,while in the second subtype lumina is irregular or polygonal,with diameter sm aller than muri width.The pollen volume was divided into four types.The genus Caragana is a natural group which was indicated by the general resemblance of pollen morphology. In this genus the pollen morphological classification was not in concordance with the arrangement of the sections and series except Ser.Pygmeae and some species. The pollen variation at infraspecific level was obvious,especially the species distributed in Qinghai Xizang Plateau, such as C.jubata, C.bicolor and C. erinacea . The evolutionary trend of pollen morphology is from perforate to reticulate, which is congruous with the evolution from the pinnate leaf group to the almate leaf group in the genus.  相似文献

Quantitative variation for leaf trichome number is observed within and among Gossypium species, varying from glabrous to densely pubescent phenotypes. Moreover, economically important cotton lint fibers are modified trichomes. Earlier studies have mapped quantitative trait loci (QTLs) affecting leaf pubescence in Gossypium using allotetraploids. In this study, we mapped genes responsible for leaf trichome density in a diploid A genome cross. We were able to map 3 QTLs affecting leaf pubescence based on trichome counts obtained from young leaves (YL) and mature leaves (ML). When the F(2) progeny were classified as pubescent versus glabrous, their ratio did not deviate significantly from a 3:1 model, suggesting that glabrousness is inherited in a simple Mendelian fashion. The glabrous mutation mapped to linkage group A3 at the position of major QTL YL1 and ML1 and appeared orthologous to the t1 locus of the allotetraploids. Interestingly, a fiber mutation, sma-4(ha), observed in the same F(2) population cosegregated with the glabrous marker, which indicates either close linkage or common genetic control of lint fiber and leaf trichomes. Studies of A genome diploids may help to clarify the genetic control of trichomes and fiber in both diploid and tetraploid cottons.  相似文献

在光学显微镜和扫锚电镜下对锦鸡儿属31种2变种植物花粉形态进行了观察。根据外壁表面纹饰,可分:类型I,表面具小穿孔;类型II, 具网状纹饰。 在类型II中据网眼大小和网脊宽度特征又分为两个亚类型。 根据花粉体积大小,可分4个类型。本属植物花粉形态较为一致,表明是一个自然类群。组、 系的花粉形态特征与一般形态变异不一定相对应,即形态上有明显区别的种类,在花粉形态上未必有很大区别,仅一些种类及矮锦鸡儿系 Ser. Pygmaeae的花粉形态相近。 本属花粉形态在种下等级的变异, 特别是青藏高原分布的种类的变异较为明显,例如 C.bicolor,C. jubata,C. erinacea 与各自的近缘种类。花粉形态为分类处理提供了参考。本属植物花粉形态是从表面具小穿孔向具网状纹饰演化,对应于羽状叶类群向掌状叶类群演化。  相似文献

本文采用聚丙烯酰胺凝胶垂直板电泳和等电聚焦技术,对棉属(Gossypium)A基因组2个二倍体种、D基因组10个二倍体野生种和四倍体2(AD)基因组的3个种进行过氧化物酶同工酶酶谱分析。种间酶谱关系符合形态学,细胞学和遗传学的研究结果,但G.gossypioides,G.thurberi和G.trilobum的酶谱与D基因组其他种有较大差异却与A基因组相似。由二倍体种酶液组成的体外人工混合体与自然四倍体的比较分析表明,四倍体棉种G.darwinii,G.barbadense和G.hirsutum是A基因组和D基因组的异质组合,G.raimondii而不是G.thurberi或G.trilobum为四倍体种祖先基因组的最可能的D亚基因组供体。对过氧化物酶同工酶分析为棉属种间亲缘关系和四倍体起源的研究提供生化遗传依据的可行性进行了阐述。  相似文献

Abstract In a preliminary trial 187 collections of muskmelon comprising dessert and non-dessert types and wild Cucumis species were screened against cucumber green mottle mosaic virus (CGMMV) under field conditions. Based on the reaction to CGMMV, 64 were studied further in detail. Thirteen collections were found to be resistant in two field screenings (summer and rainy seasons). Results on screening under artificial conditions against pure isolate of CGMMV indicated that Cucumis figarei , C. myriocarpus 1, C. myriocarpus 3, C. africanus 1, C. africanus 2, C. meeusii, C. ficifolius and C. zeyheri 2 were resistant to virus. Immune nature of resistance was later confirmed by back inoculation technique and, transmission electron microscopic study of C. figarei, C. ficifolius, C. meeusii, C. africanus 1 and C. zeyheri 2. Phoot ( C. melo var. momordica ) and Kachri (non-dessert types), FM 1 and FM 2 (Cornell breeding lines) and C,. myriocarpus 2 were also resistant to CGMMV with very mild symptoms.  相似文献

? There is limited evidence on how shifts in plant physiological performance influence vegetation variations in the paleorecord. ? To evaluate δ(13) C of pollen from C(3) plants as an indicator of community-level physiology, small quantities (10-30 grains) of untreated pollen and sporopollenin from herbarium specimens of Ambrosia (A.?tomentosa and A.?psilostachya) and Artemisia (A.?frigida, A.?ludoviciana and A.?dracunculus), genera abundant in grassland pollen profiles, were isolated by micromanipulation. Their δ(13) C values were measured using a spooling-wire microcombustion device interfaced with an isotope-ratio mass spectrometer. Leaf δ(13) C was also measured. Carbon isotope discrimination (Δ) for untreated pollen, sporopollenin and leaves was compared with historic records of seasonal precipitation amount, vapor pressure deficit and the Palmer Drought Severity Index (PDSI). ? Each species showed positive correlations between Δ of untreated pollen and sporopollenin. Sporopollenin Δ was most strongly correlated with PDSI. Correlations among leaf Δ and moisture indicators were stronger for Ambrosia than Artemisia. ? These results suggest that sporopollenin Δ indicates the level of moisture stress in C(3) plants. Therefore, δ(13) C analysis of pollen promises to help address important paleoecological questions, such as how community-level physiology contributes to shifts in vegetation composition.  相似文献

Oxidative stress induced by excessive accumulation of reactive oxidative species (ROS) during cryopreservation is thought to be one factor contributing to cryodamage of biological materials. To explore the role of oxidative stress in the cryopreservation of plant pollen, germination, ROS, and malondialdehyde (MDA) levels of pollen from 20 ornamental plant species were compared before and after cryopreservation. The results showed that the germinability of cryopreserved pollen from 13 out of the 20 species was not significantly different from that of fresh pollen (group 1), only one increased significantly, while the other six declined significantly (group 2). The MDA content in cryopreserved pollen from nine species in group 1 showed no significant difference from that of fresh pollen, while four species in group 2 rose significantly. This suggested that pollen viability and MDA levels were negatively correlated. ROS generation in cryopreserved pollen from nine species in group 1 was unchanged compared to fresh pollen, while five species in group 2 increased significantly. This suggested that pollen viability was negatively correlated with ROS generation. Additionally, both ROS and MDA levels in pollen from four species in group 2 increased significantly. In conclusion, pollen from most species possesses some cryostorage tolerance, but some species are severely damaged by cryostorage. Oxidative stress induced by the cryostorage in liquid nitrogen (LN) may be a key factor for the decreased viability in pollen following cryopreservation.  相似文献

细胞周期蛋白(cyclin)B是真核细胞周期运转中调控G2期至M期转化的关键因子.本实验根据斑马鱼细胞周期蛋白 B1基因的剪切方式,设计3对特异于金鱼和银鲫细胞周期蛋白B基因外显子区兼并引物,首次扩增出异源四倍体鲫鲤及其原始亲本红鲫和鲤鱼细胞周期蛋白B基因2条大小分别约为2.4 kb和2.1 kb的片段.测序及比对分析表明:这3种鱼的细胞周期蛋白B基因2个片段均包含8个外显子和7个内含子.内含子剪切位点符合GT/AG规则,推测细胞周期蛋白B基因2个片段可能是细胞周期蛋白B基因的2种存在形式.异源四倍体鲫鲤与其原始父母本细胞周期蛋白B基因片段序列的比较结果表明:无论是外显子区还是内含子区,异源四倍体鲫鲤与其原始亲本都具有较高的遗传相似性,在1 025 bp的外显子序列中相同的碱基位点数达963个,为异源四倍体鲫鲤来源于红鲫和鲤鱼提供了分子证据;同时,异源四倍体鲫鲤与其原始亲本差异碱基位点的存在又表明这一独特的多倍体物种与其原始亲本存在着进化上的变异.此外,还分别以细胞周期蛋白B基因外显子和内含子序列构建了包括异源四倍体鲫鲤及其原始父母本在内的系统进化树.结果初步表明:对于亲缘关系较近的物种,用外显子和内含子序列构建的系统进化树与传统的物种进化树一致;而对于亲缘关系较远的物种,用内含子序列构建的进化树与传统的物种进化顺序存在较大差异.  相似文献

* BACKGROUND AND AIMS: High-temperature environments with >30 degrees C during flowering reduce boll retention and yield in cotton. Therefore, identification of cotton cultivars with high-temperature tolerance would be beneficial in both current and future climates. * METHODS: Response to temperature (10-45 degrees C at 5 degrees C intervals) of pollen germination and pollen tube growth was quantified, and their relationship to cell membrane thermostability was studied in 12 cultivars. A principal component analysis was carried out to classify the genotypes for temperature tolerance. * KEY RESULTS: Pollen germination and pollen tube length of the cultivars ranged from 20 to 60 % and 411 to 903 microm, respectively. A modified bilinear model best described the response to temperature of pollen germination and pollen tube length. Cultivar variation existed for cardinal temperatures (T(min), T(opt) and T(max)) of pollen germination percentage and pollen tube growth. Mean cardinal temperatures calculated from the bilinear model for the 12 cultivars were 15.0, 31.8 and 43.3 degrees C for pollen germination and 11.9, 28.6 and 42.9 degrees C for pollen tube length. No significant correlations were found between pollen parameters and leaf membrane thermostability. Cultivars were classified into four groups based on principal component analysis. * CONCLUSIONS: Based on principal component analysis, it is concluded that higher pollen germination percentages and longer pollen tubes under optimum conditions and with optimum temperatures above 32 degrees C for pollen germination would indicate tolerance to high temperature.  相似文献

Summary Embryogenic pollen were selectively isolated from buds after cold treatment at 10 °C for 10 days; it was immaterial whether the buds were taken from short day and low temperature (SD and LT; 8 hours light, 18 °C) or long day and high temperature (LD and HT; 16 hours light, 24 °C) plants. However, in buds from SD and LT plants the differentiation of embryogenic pollen could be detected as early as 7 days after the cold treatment, and pollen from these plants formed embryos at higher frequency (up to 4% of cultured pollen) than those from LD and HT plants (up to 1% only).The embryogenic pollen, in isolated buds, differentiated by way of pollen dimorphism. During cold treatment a fraction of pollen remained small, retained clear cytoplasm and was capable of embryogenesis in comparison to gametophytic pollen which enlarged and acquired granular cytoplasm. In our experiments cold treatment was a key factor in the induction of pollen dimorphism. This aspect of cold treatment in pollen embryogenesis is reported for the first time and was possible on the basis of selection of embryogenic pollen by density gradient centrifugation. The ratio of embryogenic pollen was about one fifth of the total population.The nutritional requirements of isolated pollen for embryogenesis were rather simple. These pollen formed embryos which readily developed into plantlets on a mineral medium supplemented with sucrose provided the pH was 6.8.  相似文献

Polyploidization is an important mechanism for introducing diversity into a population and promoting evolutionary change. It is believed that most, if not all, angiosperms have undergone whole genome duplication events in their evolutionary history, which has led to changes in genome structure, gene regulation, and chromosome maintenance. Previous studies have shown that polyploidy can coincide with meiotic abnormalities and somatic cytogenetic mosaics in Arabidopsis allotetraploids, but it is unclear whether this phenomenon can contribute to novel diversity or act as a mechanism for speciation. In this study we tested the hypothesis that mosaic aneuploidy contributes to the formation of incipient diversity in neoallopolyploids. We generated a population of synthesized Arabidopsis allohexaploids and monitored karyotypic and phenotypic variation in this population over the first seven generations. We found evidence of sibling line-specific chromosome number variations and rapidly diverging phenotypes between lines, including flowering time, leaf shape, and pollen viability. Karyotypes varied between sibling lines and between cells within the same tissues. Cytotypic variation correlates with phenotypic novelty, and, unlike in allotetraploids, remains a major genomic destabilizing factor for at least the first seven generations. While it is still unclear whether new stable aneuploid lines will arise from these populations, our data are consistent with the notion that somatic aneuploidy, especially in higher level allopolyploids, can act as an evolutionary relevant mechanism to induce rapid variation not only during the initial allopolyploidization process but also for several subsequent generations. This process may lay the genetic foundation for multiple, rather than just a single, new species.  相似文献

Leaf cuticular membranes can be released cleanly and in one piece after a 10 min treatment of leaf in a 100 C acetolysis mixture: acetic anhydride, 9; and concentrated H2SO4,1 v/v. To examine the effects of maternal and paternal parentage and the size of the pollen load on seed size and weight and on progeny performance we conducted a controlled crossing experiment using a natural population of Campanula americana. We found that seed size was positively correlated with early seedling performance for all but one of traits we measured (days to emergence), but was not significantly correlated with any of the later vegetative measures or reproductive output. We detected significant effects due to the maternal parent for the vegetative traits days to emergence, days to first leaf, and final plant height, as well as total seed weight, and mean seed weight per fruit. Significant paternal effects were found for all of the seedling traits except number of leaves after vernalization. The progeny from fruits receiving high pollen loads significantly outperformed the progeny from fruits receiving low pollen loads for the traits days to first and second leaf, numbers of leaves after vernalization, and days to first flower. These results not only demonstrate the importance of parentage and seed weight on progeny performance, but also indicate that variations in the size of the pollen load may be important in seedling establishment in natural populations.  相似文献

Onion seedlings of two open-pollinated cultivars were grown either at a constant 10°C or outdoors in spring and were selected for high, medium or low relative leaf growth rate, RL. Flowering plants within each RL group were crossed using controlled pollination for the 10°C selections and as a fly-pollinated polycross for the outdoor selections. The pollen donors of the 10°C selections were also selfed. Despite the wide variation of RL in the parent populations, there was no indication of heritability of RL. Mean RL was significantly lower for the progeny from self-rather than cross-pollinations as was initial leaf area and rate of seedling emergence.  相似文献

Background and Aims

Flow cytometry has been used to measure nuclear DNA content in pollen, mostly to understand pollen development and detect unreduced gametes. Published data have not always met the high-quality standards required for some applications, in part due to difficulties inherent in the extraction of nuclei. Here we describe a simple and relatively novel method for extracting pollen nuclei, involving the bursting of pollen through a nylon mesh, compare it with other methods and demonstrate its broad applicability and utility.

Methods

The method was tested across 80 species, 64 genera and 33 families, and the data were evaluated using established criteria for estimating genome size and analysing cell cycle. Filter bursting was directly compared with chopping in five species, yields were compared with published values for sonicated samples, and the method was applied by comparing genome size estimates for leaf and pollen nuclei in six species.

Key Results

Data quality met generally applied standards for estimating genome size in 81 % of species and the higher best practice standards for cell cycle analysis in 51 %. In 41 % of species we met the most stringent criterion of screening 10 000 pollen grains per sample. In direct comparison with two chopping techniques, our method produced better quality histograms with consistently higher nuclei yields, and yields were higher than previously published results for sonication. In three binucleate and three trinucleate species we found that pollen-based genome size estimates differed from leaf tissue estimates by 1·5 % or less when 1C pollen nuclei were used, while estimates from 2C generative nuclei differed from leaf estimates by up to 2·5 %.

Conclusions

The high success rate, ease of use and wide applicability of the filter bursting method show that this method can facilitate the use of pollen for estimating genome size and dramatically improve unreduced pollen production estimation with flow cytometry.  相似文献
