Abstract— Glial cells were isolated from bovine white matter by differential centrifugation. The fatty aldehyde and fatty acid compositions of ethanolamine glycerophosphatides (EGP), serine glycerophosphatides (SGP) and choline glycerophosphatides (CGP) were determined by gas-liquid chromatography. The fatty acid compositions of the sphingo-lipids including sphingomyelin, cerebroside and cerebroside sulphate, and of minor lipid components including cholesterol esters and triglycerides, were also determined by gas-liquid chromatography. The relative proportions correlated closely with the results obtained by O'B rien and S ampson (1965 b ) for adult human brain. The fatty aldehyde compositions of the glycerophosphatides were more closely related to the corresponding fatty acid compositions of the plasma membrane than of the mitochondria. Long-chain fatty acids (19–26 carbon atoms) were detected in sphingomyelin, cerebroside and cerebroside sulphate; this indicates that chain-elongation beyond C18 occurs in the glial cells.  相似文献

The compositions of the major lipids and their constituent fatty acids and fatty aldehydes from cerebral gray matter, white matter, and myelin, spinal cord myelin, and sciatic nerve were determined in a 57 yr old woman who died of Refsum's disease. There were deficiencies of ethanolamine glycerophosphatides (EGP) in gray matter and frontal lobe myelin, and a lipid with the chromatographic properties of lyso-EGP accumulated in all tissues. The proportions of the remaining lipids were nearly normal in the central nervous system tissues. In the sciatic nerve the proportions of sphingolipids were small; this observation is consistent with the severe demyelination noted on pathologic examination. Cholesteryl esters were not detected in any tissue. Phytanate (3,7,11,15-tetramethylhexadecanoate) was present in the glycerophosphatides from each tissue. Higher proportions of phytanate were found in choline glycerophosphatides (CGP) than in EGP or in serine glycerophosphatides (SGP). Hydrolysis with phospholipase established that phytanate was confined to the 1-position of CGP. More phytanate was found in CGP from myelin than from gray or white matter. Fourfold higher proportions of phytanate were found in CGP from sciatic nerve than in CGP from the central nervous system: in sciatic nerve, 24% of the fatty acids of CGP consisted of phytanate. The proportions and compositions of sphingolipid hydroxy fatty acids and odd-numbered fatty acids were normal in each tissue. These findings argue against a defect in sphingolipid alpha-hydroxy acid metabolism in Refsum's disease. The results are consistent with the view that the accumulation of phytanate is responsible for the demyelination.  相似文献

Abstract— Myelin was isolated from bovine optic nerves by differential ultracentrifugation and its lipid composition was analysed. Optic nerve myelin contained 76·3 per cent lipid. The major lipids were cholesterol, ethanolamine glycerophosphatides (EGP) and cerebroside. Serine glycerophosphatides (SGP), sphingomyelin and cerebroside sulphate were present in smaller proportions. EGP and SGP contained 34·6 and 0·5 per cent aldehydes. The major fatty aldehydes were palmitaldehyde, stearaldehyde and octadecenaldehyde. The fatty acids of EGP, SGP and choline glycerophosphatides (CGP) were chiefly 16:0, 18:0 and 18:1, with small proportions of 20 and 22 carbon polyunsaturates. The sphingolipids contained predominantly saturated and monounsaturated fatty acids of chain lengths of 20–26 carbon atoms. Optic nerve myelin and white matter myelin resembled one another closely in overall lipid composition and in the fatty acid compositions of their constituent lipids. Optic nerve myelin and white matter myelin are chemically similar membranes, but both of these differ in their lipid composition from spinal root myelin.  相似文献

The ethanolamine glycerophosphatides (EGP) were isolated from rat mucosal lipids by column and thin-layer chromatography and were found to contain alkyl acyl EGP. After hydrolysis and acetylation, the resulting alkyl diacetyl glycerols were separated by gas-liquid chromatography and the paraffinic moieties were shown to consist of C18- and C20-saturated and C18-monounsaturated residues. The fatty acid compositions of the 1,2-diacyl glycerols and 1-alkyl-2 acyl glycerols obtained by treatment of the phosphatides with phospholipase C were determined. The 1-alkyl 2-acyl glycerols were rich in polyunsaturated acyl residues.  相似文献

The content and fatty acid composition of phospholipids and the in vivo labeling of lipids by [3H]glycerol and [3H]serine was studied in the retina and the optic tectum of young chickens. The tectum had a higher content of phospholipids and a significantly lower ratio of choline (CGP) to ethanolamine (EGP) glycerophospholipids than the retina. Lipids of the chicken optic system were characterized by a high proportion of polyenoic fatty acids of the n-6 series compared to other species. Intravitreally injected [3H]glycerol was incorporated into all glycerol-containing lipids of the retina, especially in CGP and EGP. Most of the label from [3H]serine was found in serine glycerophospholipids (SGP). The time-dependent distribution of both precursors among retinal lipids was consistent with de novo synthesis as well as metabolic interconversions of lipids. Thus, [3H] from serine also appeared in EGP and CGP, indicating the presence and activity of SGP decarboxylase and EGP-n-methyl transferase. Lipids labeled with both precursors in retina were subsequently found in the tectum, via axoplasmic transport. Even though different lipid classes were labelled by each precursor the proportion of lipids transported to the tectum was similar in both cases (about 1% of the label present in retina).  相似文献

Abstract— —Glial cells were isolated from bovine white matter by differential centrifugation with'Ficoll'and their lipid composition was analysed. The preparations contained 20.8 per cent lipid and 792 per cent protein. The major lipid components were cholesterol, ethanolamine glycerophosphatides (EGP), cerebroside and serine glycerophosphatides (SGP). Sphingomyelin, cerebroside sulphate and inositol glycerophosphatide were present in lower proportions. EGP contained the largest proportion of aldehydes (17 per cent) and SGP contained 12 per cent. Choline glycerophosphatides contained only a trace of aldehyde. No gangliosides were present in the filial cell preparations.  相似文献

To investigate the relative turnover of esterified polyunsaturated fatty acids in diacylglycerophospholipids and plasmalogens in isolated cardiac myocytes, we characterized the phospholipid composition and distribution of radiolabel in different phospholipid classes and in individual molecular species of diradyl choline (CGP) and ethanolamine (EGP) glycerophospholipids after incubation of isolated cardiac myocytes with [3H]arachidonate or [14C]linoleate. Plasmalogens in CGP (55%) and EGP (42%) quantitatively accounted for the total plasmalogen content (39%) of cardiac myocyte phospholipids. Plasmalogens comprised 86% and 51% of total arachidonylated CGP and EGP mass, respectively, and [3H]arachidonate was primarily incorporated into plasmalogens in both CGP (65%) and EGP (61%) classes. The specificity activity of [3H]arachidonylated diacyl-CGP was approximately 2- to 5-fold greater than that of [3H]arachidonylated choline plasmalogen, whereas comparable specific activities were found in the [3H]arachidonate-labeled ethanolamine plasmalogen and diacyl-EGP pools. Of the total linoleate-containing CGP and EGP mass, 54% and 57%, respectively, was esterified to plasmalogen molecular species. However, [14C]linoleate was almost exclusively incorporated into diacyl-CGP (96%) and diacyl-EGP (86%). The specific activities of [14C]linoleate-labeled diacyl-CGP and diacyl-EGP were 5- to 20-fold greater than that of the [14C]linoleate-labeled plasmalogen pools. The differential incorporation of polyunsaturated fatty acids in plasmalogens and diacylglycerophospholipids demonstrates that the metabolism of the sn-2 fatty acyl moiety in these phospholipid subclasses is differentially regulated, possibly fulfilling separate and distinct physiologic roles.  相似文献

The effects of dietary fish oil containing n - 3 polyunsaturated fatty acids on the fatty acid compositions of the alkylacyl and alkenylacyl species of choline glycerophospholipids (CGP) and ethanolamine glycerophospholipids (EGP) were studied in rat heart and compared with the corresponding diacylglycerophospholipids. After a 7 week feeding period, all phospholipid classes from the fish oil group exhibited much higher levels of the n - 3 polyunsaturated fatty acids including eicosapentaenoic acid (20:5(n - 30)), docosapentaenoic acid (22:5(n - 3)) and docosahexaenoic acid (22:6(n - 3)), as well as lower levels of the n - 6 series (18:2, 20:4, 22:4 and 22:5), relative to animals given sunflower seed oil-enriched in 18:2(n - 6). However, the docosahexaenoic acid rather than eicosapentaenoic acid provided a much greater contribution to the n - 3 accumulation (fish oil group) in the ether-containing CGP, as indicated by the 20:5(n - 3)/22:6(n - 3) molar ratios of 0.32, 0.26 and 0.56 in the alkylacyl, alkenylacyl and diacyl classes, respectively. In addition to accumulating very high levels of docosahexaenoic acid (e.g., 47.2 mol% of fatty acids in alkenylacylglycerophosphoethanolamine of fish oil group), both ether-linked classes of EGP exhibited significantly higher levels of docosapentaenoic acid than the diacylglycerophosphoethanolamine (GPE) and all classes of CGP. These findings may bear relevance to possible beneficial effects of dietary fish oil on pathophysiological states (including myocardial ischemia) in cardiac tissue and their mediation via platelet-activating factor, 1-alkyl-2-acetylglycerophosphocholine (PAF) and arachidonic acid (20:4(n - 6))-derived eicosanoids.  相似文献

Phospholipid fatty acid composition and phospholipid subclass distribution of control and arachidonate-depleted rat polymorphonuclear leukocytes (PMN) were compared. The 20:4-depleted PMN contained significantly higher amounts of 16:1, 18:1 and 20:3 (delta 5,8,11) and lower amounts of 18:2 and 20:4 than the phospholipids from control cells. Choline-containing glycerophospholipids (CGP) were the major phospholipids of both control and 20:4-depleted cells representing 34% and 37% of the total phospholipids, respectively. Significant amounts of ethanolamine-containing glycerophospholipids (EGP) (29% and 30%) and sphingolipids (20% and 18%) were also present in both cell types. Serine-containing glycerophospholipids (SGP) together with inositol-containing glycerophospholipids (IGP) constituted 16% and 13% of the phospholipids in control and 20:4-depleted cells, respectively. CGP from control cells had significantly higher amounts of 16:0 and 18:2 and lower amounts of 18:0 and 20:4 than EGP, whereas CGP from 20:4-depleted cells has higher amounts of 16:0 and 16:1 and lower amounts of 20:3 than EGP. Analysis of the subclass composition of CGP and EGP revealed that both control and 20:4-depleted cells contained significantly large amounts of alkylacyl-GPC and alkenylacyl-GPE. Small amounts of alkylacyl-GPE and alkenylacyl-GPC were also observed. The predominant fatty acyl residues found in the 1,2-diacyl-GPC, alkylacyl-GPC of control cells were 16:0, 18:0, 18:1, 18:2, and 20:4, while those of 20:4-depleted cells were 16:0, 16:1, 18:0, 18:1, and 20:3. More than 60% of CGP-bound 20:4 of control cells and about 70% of the CGP-bound 20:3 of 20:4-depleted cells were found in their alkylacyl species.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The effect of feeding rats for 30 days with diets containing high levels of linoleic acid (sunflower oil, SO) or -linolenic acid (perilla oil, PO) was studied in the liver, kidney and brain. The PO group showed a higher labeling of choline glycerophospholipids (CGP) in liver and kidney but no difference with the SO group in ethanolamine glycerophospholipids (EGP) labeling. The brain displayed the lowest incorporation of both precursors and no difference between the two diets. Analyses of brain CGP and EGP fatty acid compositions showed that in the PO group the ratio n-6/n-3 was lower than in the SO group, mainly as a consequence of lower levels of n-6 fatty acids. The mole % of docosahexaenoate (DHA) in these lipids was the same for both groups and only triacylglycerols (TAG) displayed a higher DHA. Therefore, at least in the brain, the magnitude of fatty acid changes observed in CGP and EGP for the PO group does not affect the uptake/incorporation of the precursors into phospholipids.  相似文献

Thin-layer chromatography showed that only lecithin was present. 2. The distributions of phosphorus and the fatty acids at the 3 and 2 positions were determined. 3. Cultures of Tetrahymena pyriformis W incorporate exogenous 3-[14C]-cilienic acid and gamma-[1(-14)C] linolenic acid, terminal products of unsaturated fatty acid synthesis, into glycerophosphatides without randomization of the radiolabel. There was no difference in the rate of loss of each of the two acids at 15 or 28.5 degrees C. Differential turnover of these fatty acids, therefore, does not appear to be the cause of the shift in fatty acid pattern observed with temperature reduction.  相似文献

Myelin was isolated from the brain of adult fruit bats (Rousettus aegyptiacus) in a discontinuous sucrose gradient. Cholesterol comprised 189.0 mol/100 mol lipid phosphorus, galactolipids 60.3 mol/100 mol phosphorus and plasmalogens 32.5 mol/100 mol phosphorus. Choline and ethanolamine glycerophosphatide were present in nearly equal amounts followed by serine glycerophosphatide, sphingomyelin and inositol glycerophosphatides.The fatty acid composition of sphingomyelin and non-hydroxy cerebroside was determined by gas-liquid chromatography. Fatty acids were mainly saturated or mono-unsaturated with a small percentage of polyunsaturated fatty acids present.The lipid composition and sphingolipid fatty acid distribution in bat myelin was fairly similar to that of other species.  相似文献

Fatty acid compositions of glycerophosphatides of developing chick embryonic brain and liver were compared. In brain, ethanolamine and serine glycerophosphatides contained 30-40% polyunsaturated fatty acids, lecithin almost none (except for arachidonic). In the liver, these acids were equally distributed in the phospholipid fractions. The principal polyunsaturated fatty acids of the ethanolamine and serine glycerophosphatides in brain, liver, and yolk were 22:6, 20:4, and 18:2, respectively. During embryonic development of brain from the 8th day of incubation to hatching, the fatty acid composition of individual glycerophosphatide fractions remained constant. Because of the relative increase of ethanolamine glycerophosphatides and decrease of lecithin, total glycerophosphatides showed a decrease in 16:0 and an increase in 18:0. Substantial amounts of palmitaldehyde and stearaldehyde were present on the 8th day of incubation in the brain ethanolamine glycerophosphatide fraction. During the 3rd week of incubation, the liver showed a two-fold increase in the relative amount of 18:2 in all glycerophosphatide fractions. A decrease of 16:0 in the lecithin fraction and consequently in total glycerophosphatides was also observed during this period. No significant changes in glycerophosphatide fatty acids were observed in the yolk throughout incubation.  相似文献

The formation of phospholipid hydroperoxides was monitored in human red blood cell (RBC) membranes that had been peroxidized with an azo initiator. Peroxidation of RBC membranes caused a profound decrease in the amount of polyunsaturated fatty acids and concomitantly hydroperoxides, as primary products of peroxidation, appeared in the phospholipids. Hydroperoxides were predominantly generated in choline glycerophospholipid (CGP), while the extent of formation of ethanolamine glycerophospholipid (EGP) hydroperoxides was low and their presence was transient. Hydroxy and hydroperoxy moieties in CGP were identified as 9-hydroxy and 13-hydroxy octadecanoic acid, derived from linoleic acid, by gas chromatography-mass spectrometric analysis. No consistent generation of hydroperoxide from arachidonic acid was evident in CGP. The CGP-hydroperoxide accounted for approximately 76% of linoleic acid consumed during peroxidation of RBC membranes. The prominent generation of phospholipid hydroperoxides was observed in the linoleic acid-rich membranes from rabbit RBC, indicating that the level of linoleic acid in phospholipids determins, in part, the extent of formation of phospholipid hydroperoxides. Aldehydic phospholipids, as secondary products of peroxidation, were detected in oxidized membranes. EGP was the most prominent aldehydic phospholipid, while negligible amounts of aldehydic CGP were formed. This study indicates that the process of oxidation of individual phospholipids clearly differs among phospholipids and depends on the structure of each.  相似文献

Carp (Cyprinus carpio L.) whole brain was used to investigate the thermal acclimation changes under normoxic conditions of three-subclasses (alkenylacyl-, alkylacyl- and diacyl-subclasses) of choline glycerophospholipids (CGP), ethanolamine glycerophospholipids (EGP) and inositol glycerophospholipids (IGP) as well as their acyl chain profiles and molecular species composition. The alkenylacyl subclass of CGP and IGP and the alkylacyl subclass of CGP and EGP varied significantly during summer (25°C) acclimation compared to winter (5°C). The levels of alkenylacyl and alkylacyl-CGP, alkylacyl-EGP and alkenylacyl-IGP were 17.3-, 3.7-, 3.5- and 1.3-fold higher in the summer, respectively, while the alkenylacyl EGP was moderately lower. The levels of diacyl subclasses from CGP and IGP were considerably lower in the summer to compensate for the higher proportion of alkenylacyl and alkylacyl subclasses. Significant changes of ether phospholipids and the reorganization of the molecular species composition of all lipid subclasses may be associated with the fine tuning of the physical properties of the cellular membranes in carp brain due to temperature acclimation. The overall acyl chain profile of the three subclasses of carp brain phospholipids showed differences in composition depending upon the subclass of the individual phospholipid. Generally, the polyunsaturated fatty acid (PUFA) chain composition increased relative to monounsaturated fatty acid (MUFA) and saturated fatty acids (SFA) during winter acclimation. Docosahexaenoic acid (DHA) was richer in the winter compared to summer. However, no DHA was found in ether-containing species of IGP from either winter or summer, except for 2% in alkylacyl-IGP during the summer. The above observations suggest that the content of ether phospholipids (alkenylacyl and alkylacyl) as well as the reorganization of the molecular species composition of all phospholipids may serve to maintain a functional fluid-crystalline state to preserve the signaling functions in carp brain.  相似文献

1. The total yield of fatty acids from the whole envelopes was markedly higher than that obtained from the ordinary cell walls. In both samples the major fatty acids were C(16) and C(18) acids. 2. The whole envelopes contained C(18) acids and long-chain (C(19)-C(26)) fatty acids, in a higher proportion than did the ordinary cell walls. Fifteen fatty acids with more than 18 carbon atoms were identified, among which 2-hydroxy-C(26:0) and C(26:0) acids predominated. 3. A complex sphingolipid containing inositol, phosphorus and mannose was isolated from the whole cell envelopes. The main fatty acids of this lipid were 2-hydroxy-C(26:0) and C(26:0) acids. It was concluded that this sphingolipid is present both in the ordinary cell wall and in the plasma membrane of baker's yeast. 4. The neutral lipids amounted to over 50% and the glycerophosphatides to about 30% of the total fatty acid content of the whole envelope. The major fatty acids in these lipids were C(16:1), C(18:1) and C(16:0) acids. The proportion of fatty acids with more than 18 carbon atoms was lowest in the neutral lipids, whereas the neutral glycolipids contained the highest percentage of these fatty acids. Acidic glycolipids amounted to 14% of the total fatty acid content of the whole envelope. The presence of a cerebroside sulphate in this lipid fraction was demonstrated, whereas the high content of 2-hydroxy-C(26:0) acid found is caused by the complex inositol- and mannose-containing sphingolipid.  相似文献

Pure preparations of phosphatidylcholine were isolated from spinach leaf chloroplasts, spinach leaf microsomes, and cauliflower inflorescence. The isolated phosphatidylcholine was treated with snake venom phospholipase A, and the fatty acid distribution and composition of the fatty acid methyl esters prepared from the lysophosphatidylcholine and the freed fatty acid were determined by gas-liquid chromatography. The results showed that saturated fatty acids were preferentially esterified at position 1 and unsaturated fatty acids at position 2. The phosphatidylcholine from cauliflower was also treated with phospholipase C. The resulting diglycerides were fractionated on AgNO(3)-impregnated thin-layer plates. The diglyceride fractions were transesterified and the fatty acid composition of each was determined by gas-liquid chromatography. The predominant species contained linolenic acid only (22% of the total), linolenic and oleic acids (19%), and linolenic and palmitic acids (37%). These molecular species could not be accounted for by random distribution of the fatty acids.  相似文献

Abstract— Quantitative analyses of white matter from four brains of patients with multiple sclerosis (MS) and four control brains were carried out for total and soluble proteins, individual lipid fractions, and their corresponding fatty acids. In three specimens from two of the MS brains there were reductions of cerebrosides and of the C20:1 acid in the ethanolamine glycerophosphatide (EGP) fraction and a slight increase of tetraenes and trienes, while all other components were present in concentrations similar to those in the controls. In three other samples from two of the MS brains, galactolipids were deficient to a greater extent than cholesterol, EGP or CGP (choline glycerophosphatide), while proteins were within the control range. In samples where thinning of myelin was observed in Luxol-blue stained sections, there were proportional decreases of all components. The percentage of C20:1 acid in the EGP fraction was reduced in two of three myelin preparations from corresponding samples of MS white matter, and that of C24:1 acid in the cerebroside fraction was reduced in all three MS myelin preparations when compared with the two controls. The data suggest that inadequacy of the fatty acid elongation process together with deficits of cerebrosides represent one of the early biochemical lesions in the white matter of the MS brain.  相似文献

The cellular fatty acid composition of 14 strains of Caulobacter species and types, two species of Prosthecomicrobium, and two species of Asticcacaulis was determined by gas-liquid chromatography. In most of these bacteria, the major fatty acids were octadecenoic acid (C18:1), hexadecenoic acid (C16:1) and hexadecanoic acid (C16:0). Some cyclopropane and branched chain fatty acids were detected in addition to the straight chained acids.
