The contribution of three exoglucanases from a commercial Trichoderma viride cellulase to transcellobiosylation, and the tolerance of these enzymes to acetonitrile co-solvent were studied. The enzymatic reactions were performed with p-nitrophenyl-β-d-cellobioside as the starting substrate. Among these enzymes, the least anionic exoglucanase (Exo I) showed the highest transcellobiosylation activity and acetonitrile tolerance. Exo I retained considerable activity even in 30% MeCN/water and produced p-nitrophenyl-β-d-cellotetraoside at about 1.5% conversion from the initial substrate in 30% MeCN/water. The residual activity of Exo I after incubation in MeCN/water mixture was almost identical to that of the crude cellulase and a considerable amount of the transcellobiosylation properties of the crude cellulase seemed to be attributable to this Exo I component.  相似文献

在摇瓶试验基础上,采用里氏木霉(Trichoderma reesei)HC-415菌株进行5L自控罐产纤维素酶深层发酵试验。在通气量为 0.2—0.6vvm、搅拌速度为 400r/min、发酵液pH控制在5.8—6.1的条件下,发酵液的羧甲基纤维素(CMC)酶酶活最高为325.0mg糖/ml,滤纸糖酶(FPA)酶活最高达17.9mg糖/ml。发酵周期为108h。所得冻干纤维素酶粉CMC酶活最高3111IU/g,FPA最高135IU/g ,对发酵液得率平均6.7g/L。酶活总收率CMC酶活平均78.2%,FPA酶活平均73.5%。  相似文献

绿色木霉(Trichodermaviride)在pH控制发酵条件下,采用流加葡萄糖发酵策略,可显著提高综合滤纸酶活力(FPA)和内切酶(endo—β—1,4-glucanase,EG)、外切酶exo—β-1,4-glucanase,CBH)、纤维二糖酶(cellobiase,CB)酶活。在5L发酵罐中采用pH控制和流加葡萄糖工艺,可提高CB酶含量,改变酶组分之间的比例,使得FPA、EG、CB和CBH酶活分别达到50.0U/mL,210.0U/mL,4.0U/mL和2.5U/mL,比摇瓶发酵分别提高了6.7.4.2、19、2.5倍。  相似文献

野生型康氏木霉854-B_2分别经物理化学诱变因子,TDP辐射器和空间微重力辐射等因素的多级处理,得到1株形态发生明显改变的变异株B—7,其固体培养物的纤维素酶各组分酶活力,如滤纸糖酶活力(FPA)为34u/g,羧甲基纤维素酶活力(CMC—ase)为29.0u/g,β-葡萄糖苷酶活力(β-Glase)为29.0u/g,与854-β2相比,分别提高5.9倍,7.6倍和4.2倍。其固体培养的最佳条件是:pH6.0,28~30℃,96小时,最适培养基成分为:青草粉:麸皮=7:3,1.5~2.0倍水和(NH4)2SO41.5~2.0%(均以固体料计)。  相似文献

采用固态发酵和连续监测正交实验结果的方法,研究了培养温度、初始pH值、液料比和接种量对一株木霉(Trichodermasp.)发酵过程中微晶纤维素酶活、CMC酶活和滤纸酶活的影响及影响程度。指出液料比在整个发酵过程中是对产酶影响最大的因素,温度在发酵初期影响较大,初始pH和接种量的影响均不显著。总体看来,培养温度、初始pH值、液料比和接种量分别为30℃、4、7和5%是比较合适的。  相似文献

采用3,5-二硝基水杨酸(DNS)为显色剂,滤纸和CMC为底物,测定纤维素酶的酶活.考察了在不同温度、pH值、反应时间、底物浓度等反应条件下,有机复合物H对纤维素酶滤纸酶活(FPA)和CMC酶活(CMCA)的影响.结果表明,有机复合物H对纤维素酶的FPA活性和CMCA活性均有一定的促进作用,且在不同条件下的促进效果有较大差异.  相似文献

Trichoderma reesei (QM 9414) produced cellulase in continuous culture, on media containing xylose (1%) supplemented with sorbose (0.3%) to induce cellulase production. Maximum cell mass of 4.54 kg/m(3) occurred at pH 4.0 and a dilution rate of 0.0391 h(-1) where residual substrate was 0.43 kg/m(3), but no cellulase was produced. Maximum cellulase production of 0.69 FPU occurred at pH 3.5 and a dilution rate of 0.0110 h(-1), where cell mass production was 2.56 kg/m(3) and residual substrate was 0.15 kg/m(3). Monod kinetic constants, corrected for endogenous metabolism, were 0.091 h(-1), 0.469 kg/m(3), 0.00923 h(-1), and 0.470 kg cells/kg xylose at pH 3.5, for the maximum specific growth rate, Michaelis-Menten coefficient, endogenous metabolism coefficient, and yield coefficient, respectively. Specific growth rate fitted a maturation time model, which predicted decreasing maturation time with increasing pH.  相似文献

Trichoderma viride (Pers.) pre-inoculated wheat seedlings infected with Fusarium oxysporum Schlecht. (co-stressed) did not show wilting symptoms compared to Fusarium infected seedlings. Antagonistic activity of T. viride could be demonstrated against Fusarium infection by dual culture experiment. After seven days post infection, morphological and physiological parameters such as, root and shoot length, fresh and dry weight, relative water content, total soluble protein, total chlorophyll and carotenoid contents were observed to be increased in co-stressed compared to Fusarium infected seedlings. Accumulation of hydrogen peroxide was enhanced in Fusarium infected tissues compared to co-stressed. Trichoderma mediated activation of antioxidant enzymes such as, catalase, guaiacol peroxidase, ascorbate peroxidase, superoxide dismutase in co-stressed seedlings indicated their involvement in enhanced resistance against Fusarium infection, which is suggestive of playing crucial role in mitigating cellular toxicity developed due to excess H2O2. Thus, Trichoderma pre-inoculation protected wheat against Fusarium infection by stabilising oxidative stress.  相似文献

Chemical modification of histidine residues in ricin E was studied with regard to saccharide binding. The analytical data indicate that 6 out of 7 histidine residues in ricin E are eventually modified with diethylpyrocarbonate (DEP) at pH 6.0 and 25°C in the absence of specific saccharides. Modification of histidine residues greatly decreased the cytoagglutinating activity of ricin E, and only 10% of the residual activity was found after modification of 6 histidine residues/mol. The data of affinity chromatography using lactamyl- and galactosamine-cellulofine columns suggest that modification of histidine residues does not have much effect on the binding ability at the low affinity saccharide-binding site of ricin E but abolishes the binding ability at the high affinity saccharide-binding site. In the presence of lactose, one histidine residue/mol was protected from the DEP modification with retention of a fairly high cytoagglutinating activity. Such a protective effect was also observed for specific saccharides such as galactose and A^-acetylgalactosamine, but not for glucose, a non-specific saccharide. On treatment with hydroxylamine, the modified ricin E restored 67 % of the cytoagglutinating activity. Based on these findings, it is suggested that in the high affinity saccharide- binding site of ricin E there exists one histidine residue responsible for saccharide binding.  相似文献

The solvent effect on the equilibrium position and the initial rate of esterification of 1-hexanol with acetic acid catalyzed by a lipase has been experimentally investigated. A variety of non-polar and polar solvents have been considered and the results obtained indicate that the solvent effect on the equilibrium conversion is very important compared to that for transesterification reactions. A theoretically sound methodology using the group-contribution UNIFAC model for the prediction of solvent effects on the equilibrium position of enzymatic reactions is presented and it is applied to the reaction of 1-hexanol with acetic acid as well as to a similar reaction from the literature. The results obtained are better than those from empirical methods proposed in the literature such as correlations with the octanol-water partition coefficient of the solvent, as well as the solubility of water in the solvent. Moreover, the proposed methodology can be used for the determination of the equilibrium constant of the reaction. For the prediction of the solvent effect on the initial rate of enzymatic reactions it is found that it is more accurately determined using the product of the activities of the reactants, which can be predicted by the UNIFAC model, than the octanol-water partition coefficient of the solvent or the solubility of water in the solvent.  相似文献

From in vitro and in vivo screening tests for antagonism by isolates of Trichoderma against postharvest pathogens of yams (Dioscorea spp.), an isolate of Trichoderma viride Pers. ex S.F. Gray was selected as the most promising candidate for the biocontrol of postharvest rot of yams. Inoculation of white yam (Dioscorea rotundata Poir.) with conidiaspores of T. viride and subsequent storage of the tubers under the ambient environment conditions of a traditional yam barn, resulted in a drastic reduction in the frequency of occurrence of the normal tuber surface mycoflora over a 4‐month (December‐April) storage period. Trichoderma viride on the other hand, maintained a high frequency of occurrence during the same period. Furthermore, whereas up to 52.0%) rot was found among groups of tubers that were artificially inoculated with the postharvest pathogens of yams, Aspergillus niger Van Tiegh., Botryodiplodia theobromae Pat. or Penicillium oxalicum Currie and Thom, and also the group that was not inoculated with any organism (control), among groups of tubers that were inoculated with T. viride the rot was either totally suppressed or only a low percentage of rot occurred. The significance of the findings is discussed in relation to yam storage especially by farmers with limited resources.  相似文献

Induction of synthesis of cellulolytic enzymes in Trichoderma reesei QM 9414 by cellobiono-1,5-lactone (CBL) has been investigated in a replacement system lacking additional carbon source. CBL induced cellulase secretion optimally at pH 5 and a concentration of 70 g/ml. Higher concentrations lead to lower induction. De novo induction of cellulases was proven by the inhibitory effect of cycloheximide addition. Induction by CBL was shown to act synergistically on induction by sophorose, as it decreased the concentration of sophorose required for maximal induction. Maximal endo--1,4-glucanase activities induced by either sophorose or CBL were comparable. The CBL-induced cellulase system contained all the major cellulolytic enzymes of T. reesei, i.e. cellobiohydrolase I and II, and endoglucanase I, as shown by SDS-PAGE, Western blotting and detection with specific mono- and polyclonal antibodies. No differences were seen in the types of individual enzymes formed upon induction by either sophorose or CBL. Trichoderma reesei is a well-known cellulase producer and widely applied in enzyme industry. To increase its ability to efficiently decompose cellulose, the beta-glucosidase activity of its enzyme cocktail needs to be enhanced. In this study, a beta-glucosidase I coding sequence from Penicillium decumbens was ligated with the cellobiohydrolase I (cbh1) promoter of T. reesei and introduced into the genome of T. reesei strain Rut-C30 by Agrobacterium-mediated transformation. In comparison to that from the parent strain, the beta-glucosidase activity of the enzyme complexes from two selected transformants increased 6- to 8-fold and their filter paper activity (FPAs) was enhanced by 30% on average. The transformant's saccharifying ability towards pretreated cornstalk was also significantly enhanced. To further confirm the effect of heterologous beta-glucosidase on the cellulase activity of T. reesei, the heterologously expressed pBGL1 was purified and added to the enzyme complex produced by T. reesei Rut-C30. Supplementation of the Rut-C30 enzyme complex with pBGL1 brought about 80% increase of glucose yield during the saccharification of pretreated cornstalk. Our results indicated that the heterologous expression of a beta-glucosidase gene in T. reesei might produce balanced cellulase preparation.  相似文献

The solvent effect on the equilibrium position of the transesterification reaction of hexanol with ethyl acetate catalyzed by a lipase has been investigated in a variety of non-polar and polar solvents - and binary mixtures. The results obtained indicate that the solvent effect on the equilibrium conversion is very small as compared to that for the direct esterification reactions.

Equilibrium conversions were then predicted using the equilibrium constant for the reaction obtained from Gibbs free energy of formation information for reactants and products in combination with the UNIFAC activity coefficient model. A solvent independent equilibrium conversion was obtained, which was in good agreement with the observed average value for all solvents. This indicates that UNIFAC provides satisfactory estimates of the activity coefficients but its group contribution structure does not allow the prediction of the small differences in conversion among the solvents examined.

Finally plots of these conversions versus the solvent octanol/water partition coefficient or the solubility of water in the solvent, that provide the correct trend in direct esterification reactions, did not achieve the same for transesterification.  相似文献
