The ability to concentrate CO2 around Rubisco allows C4 crops to suppress photorespiration. However, as phosphoenolpyruvate regeneration requires ATP, the energetic efficiency of the C4 pathway at low photosynthetic flux densities (PFD) becomes a balancing act between primary fixation and concentration of CO2 in mesophyll (M) cells, and CO2 reduction in bundle sheath (BS) cells. At low PFD, retro‐diffusion of CO2 from BS cells, relative to the rate of bicarbonate fixation in M cells (termed leakiness φ), is known to increase. This paper investigates whether this increase in ? could be explained by incomplete inhibition of photorespiration. The PFD response of φ was measured at various O2 partial pressures in young Zea mays plants grown at 250 (LL) and 750 µmol m?2 s?1 PFD (HL). φ increased at low PFD and was positively correlated with O2 partial pressure. Low PFD during growth caused BS conductance and interveinal distance to be lower in the LL plants, compared to the HL plants, which correlated with lower φ. Model analysis showed that incomplete inhibition of photorespiration, especially in the HL plants, and an increase in the relative contribution of mitochondrial respiration at low PFD could explain the observed increases in φ.  相似文献

The plant growth regulator activity of epoxiconazole, a new triazole fungicide, was investigated by time-course, dose-response and histology experiments with Galium aparine L. (cleavers). Seven days after treatment with 125g ai ha–1 epoxiconazole (field rate), plant height was reduced by 43%. After seventeen days, leaflet area was reduced by 27% but leaflet fresh weight was not significantly influenced. This was partly because leaflet thickness had increased by 20% following epoxiconazole application. Chlorophyll concentrations were also increased on a unit area basis. Examination of leaflet anatomy showed that epoxiconazole elongated palisade, spongy mesophyll and upper epidermal cells. For example, 125g ai ha–1 caused a 35% increase in the length of spongy mesophyll cells. Epoxiconazole also prevented cell separation as there were significantly more palisade and spongy mesophyll cells per unit area than in leaflets sprayed with water. Stem development was reduced and 125g ai ha–1 inhibited the elongation of pith cells in stem tissue by 53%. However, the simultaneous application of gibberellin A3 (GA3) with epoxiconazole resulted in stem elongation similar to that of control plants. Botryococcus braunii, B race is a unique green microalga that produces large amounts of liquid hydrocarbons known as botryococcenes that can be used as a fuel for internal combustion engines. The simplest botryococcene (C30) is metabolized by methylation to give intermediates of C31, C32, C33, and C34, with C34 being the predominant botryococcene in some strains. In the present work we have used Raman spectroscopy to characterize the structure of botryococcenes in an attempt to identify and localize botryococcenes within B. braunii cells. The spectral region from 1600–1700 cm−1 showed ν(C=C) stretching bands specific for botryococcenes. Distinct botryococcene Raman bands at 1640 and 1647 cm−1 were assigned to the stretching of the C=C bond in the botryococcene branch and the exomethylene C=C bonds produced by the methylations, respectively. A Raman band at 1670 cm−1 was assigned to the backbone C=C bond stretching. Density function theory calculations were used to determine the Raman spectra of all botryococcenes to compare computed theoretical values with those observed. The analysis showed that the ν(C=C) stretching bands at 1647 and 1670 cm−1 are actually composed of several closely spaced bands arising from the six individual C=C bonds in the molecule. We also used confocal Raman microspectroscopy to map the presence and location of methylated botryococcenes within a colony of B. braunii cells based on the methylation-specific 1647 cm−1 botryococcene Raman shift.  相似文献

Peperomia has species that may be C3, show Crassulacean acid metabolism (CAM), or CAM-cycling. Species that show CAM progress from C3 to CAM through CAM-cycling during leaf development. In CAM and CAM-cycling species, CAM metabolism is predominately in the upper multiple epidermis and lower spongy mesophyll, whereas C3 metabolism is localized mostly in the palisade mesophyll. Using specific protein and cDNA probes prepared from P-enolpyruvate carboxylase (PEPc) and ribulose-1,5-bisphosphate carboxylase (Rubisco), we have now studied the differential distribution of photosynthetic metabolism in Peperomia leaves using the technique of tissue printing. The tissue printing studies detected Rubisco protein in leaves of C3 P. orba, but not PEPc. Young C3 leaves of P. scandens and P. camptotricha showed Rubisco protein, but not PEPc; however, the mature leaves of these two species that have CAM showed PEPc protein and RNAs in both the multiple epidermis and spongy mesophyll. In contrast, Rubisco protein and RNAs were present throughout the leaf. The tissue printing data are consistent with our previously published data showing the differential distribution of photosynthetic metabolism in leaves of Peperomia. Although the tissue printing technique is qualitative, coupled with quantitative data it has proven useful for the study of function related to structure.  相似文献

Structural Adaptation of the Leaf Mesophyll to Shading   总被引:1,自引:0,他引:1  
Structural characteristics of the mesophyll were studied in five boreal grass species experiencing a wide range of light and water supply conditions. Quantitative indices of the palisade and spongy mesophyll tissues (cell and chloroplast sizes, the number of chloroplasts per cell, the total cell and chloroplast surface area per unit leaf surface area) were determined in leaves of each of the species. The cell surface area and the cell volume in spongy mesophyll were determined with a novel method based on stereological analysis of cell projections. An important role of spongy parenchyma in the photosynthetic apparatus was demonstrated. In leaves of the species studied, the spongy parenchyma constituted about 50% of the total volume and 40% of the total surface area of mesophyll cells. The proportion of the palisade to spongy mesophyll tissues varied with plant species and growth conditions. In a xerophyte Genista tinctoria, the total cell volume, cell abundance, and the total surface area of cells and chloroplasts were 30–40% larger in the palisade than in the spongy mesophyll. In contrast, in a shade-loving species Veronica chamaedris, the spongy mesophyll was 1.5–2 times more developed than the palisade mesophyll. In mesophyte species grown under high light conditions, the cell abundance and the total cell surface area were 10–20% greater in the palisade mesophyll than in the spongy parenchyma. In shaded habitats, these indices were similar in the palisade and spongy mesophyll or were 10–20% lower in the palisade mesophyll. In mesophytes, CO2 conductance of the spongy mesophyll accounted for about 50% of the total mesophyll conductance, as calculated from the structural characteristics, with the mesophyll CO2 conductance increasing with leaf shading.  相似文献

The zebra finch (Taeniopygiaguttata) has become an increasingly important model organism in many areas of research including toxicology1,2, behavior3, and memory and learning4,5,6. As the only songbird with a sequenced genome, the zebra finch has great potential for use in developmental studies; however, the early stages of zebra finch development have not been well studied. Lack of research in zebra finch development can be attributed to the difficulty of dissecting the small egg and embryo. The following dissection method minimizes embryonic tissue damage, which allows for investigation of morphology and gene expression at all stages of embryonic development. This permits both bright field and fluorescence quality imaging of embryos, use in molecular procedures such as in situ hybridization (ISH), cell proliferation assays, and RNA extraction for quantitative assays such as quantitative real-time PCR (qtRT-PCR). This technique allows investigators to study early stages of development that were previously difficult to access.  相似文献

Palisade parenchyma cells and spongy parenchyma cells were isolated separately from Vicia faba L. leaflets. Extracts of the cell isolates were assayed for several enzymes involved in CO2 fixation and photorespiration. When compared on a chlorophyll basis, the levels of enzyme activities either were equal in the different cell types or were greater in the spongy parenchyma; this difference is a reflection, perhaps, of the higher protein-chlorophyll ratio in the latter tissue. The distribution of radioactivity in the products of photosynthesis by each cell type was the same at various times after exposure to NaH14CO3, and the kinetics of 14C incorporation into these compounds was similar. However, a larger percentage of radioactivity was incorporated by the cell isolates into the 80% ethanol-insoluble fraction and correspondingly less into the neutral fraction as compared to whole leaf. It was concluded that photosynthetic CO2 fixation is similar in the different mesophyll tissues from which these cells were derived.  相似文献

Anatomy and some physiological characteristics of the leaves in Polygonum cuspidatum Sieb. et Zucc., a dioecious clonal herb, were compared between two populations, one from a lowland in Shizuoka City (10 m above sea level), and another from a highland on Mt. Fuji (2500 m above sea level). Leaf mass per area (LMA) of the highland plants was about twice that of the lowland plants. The greater leaf thickness, thicker mesophyll cell walls and higher mesophyll cell density in the highland leaves contributed to the larger LMA. Although mesophyll area exposed to intercellular airspaces was greater in the highland leaves than in the lowland leaves by 30%, the surface area of chloroplasts facing intercellular airspaces was similar between these leaves. CO2 transfer conductance inside the leaf (gi) of the highland leaves (0·75 μmol m?2 s?1 Pa?1) is the lowest recorded for herbaceous plants and was only 40% of that in the lowland leaves. On the other hand, the difference in stomatal conductance was small. δ13C values in the leaf dry matter were greater in the highland leaves by 4‰. These data and the estimation of CO2 partial pressures in the intercellular air spaces and in the chloroplast suggested that the greater dry matter δ13C in the highland leaves, indicative of lower long‐term ratio of the chloroplast stroma to the ambient CO2 partial pressures, would be mainly attributed to their lower gi.  相似文献

Several photosynthetic parameters were examined in the different tissue layers of leaves from the recently characterized crassulacean acid metabolism/C4 intermediate plant, Peperomia camptotricha (Nishio and Ting, 1987). Light appears to control the development of certain photosynthetic characteristics within the tissue layers, while factors other than light seem to dictate others. Analysis of the chlorophyll content (including P700) and chlorophyll-proteins indicated that more light harvesting chlorophylls were associated with reaction centers in the tissues that were shaded by overlying tissue. Electron transport activity and chlorophyll-protein analysis indicated that the ratio of photosystem I to photosystem II in the spongy mesophyll (the abaxial tissue type and hence most shaded) was elevated relative to the overlying chlorophyll-rich median palisade mesophyll. The elevated photosystem I relative to photosystem II in the spongy mesophyll of Peperomia camptotricha may be related to C4 metabolism and an increased requirement for ATP.  相似文献

Near-infrared (NIR) optical imaging is a noninvasive and nonionizing modality that is emerging as a diagnostic tool for breast cancer. The handheld optical devices developed to date using the NIR technology are predominantly developed for spectroscopic applications. A novel handheld probe-based optical imaging device has been recently developed toward area imaging and tomography applications. The three-dimensional (3D) tomographic imaging capabilities of the device have been demonstrated from previous fluorescence studies on tissue phantoms. In the current work, fluorescence imaging studies are performed on tissue phantoms, in vitro, and in vivo tissue models to demonstrate the fast two-dimensional (2D) surface imaging capabilities of this flexible handheld-based optical imaging device, toward clinical breast imaging studies. Preliminary experiments were performed using target(s) of varying volume (0.23 and 0.45 cm3) and depth (1–2 cm), using indocyanine green as the fluorescence contrast agent in liquid phantom, in vitro, and in vivo tissue models. The feasibility of fast 2D surface imaging (∼5 seconds) over large surface areas of 36 cm2 was demonstrated from various tissue models. The surface images could differentiate the target(s) from the background, allowing a rough estimate of the target''s location before extensive 3D tomographic analysis (future studies).  相似文献

A histochemical study using light microscopy has been made ofthe distribution of acid phosphatase (EC 3.1.3.2 [EC] ) activity intransverse sections of fully expanded leaves of Lycopersiconesculentum grown in phosphate-deficient or sufficient media.Leaf tissues were prepared by two methods and were embeddedin paraffin wax. The location of acid phosphatase activity inleaf sections was determined by trapping orthophosphate releasedfrom p-nitrophenyl phosphate with lead acetate and subsequentlyconverting the lead phosphate to optically dense lead sulphide.In leaf sections from control tissue lead sulphide depositswere larpely confined to the spongy mesophyll cells. Whereasthe staining of the palisade cells was limited and of a granularnature, the staining of the spongy mesophyll cells was heavierand coincident with the outline of the individual cells. Moreover,the minor veins were more heavily stained than the surroundingmesophyll cells. Sections of phosphorus-deficient tissues wereheavily stained in both the palisade and spongy mesophyll layersand heavy deposits of lead sulphide were present in the regionsof the minor veins. It is suggested that the enhanced acid phosphataseactivity of the mesophyll cells in fully expanded leaves couldbe involved in the remobilization of phosphate within phosphorus-deficientplants, or be part of a phosphate transporting system, concentratingthe intracellular phosphate from the limiting supply in thesolution bathing the mesophyll cells. A realistic numerical three‐dimensional (3D) model was constructed to study CO2 transport inside a birch leaf. The model included chloroplasts, palisade and spongy mesophyll cells, airspaces, stomatal opening and the leaf boundary layer. Diffusion equations for CO2 were solved for liquid(mesophyll) and gaseous(air) phases. Simulations were made in typical ambient field conditions varying stomatal opening, photosynthetic capacity and temperature. Doubled ambient CO2 concentration was also considered. Changes in variables caused non‐linear effects in the total flux, especially when compared with the results of double CO2 concentration. The reduction in stomatal opening size had a smaller effect on the total flux in doubled concentrations than ambient CO2. The reduced photosynthetic capacity had a similar effect on the flux in both cases. The palisade and spongy mesophyll cells had unequal roles mainly due to the light absorption profile. Results from the 3D simulation were also compared to the classical one‐dimensional resistance approach. Liquid and gas phase resistances were estimated and found strongly variable according to changes in temperature and degree of stomatal opening. For the birch leaves modelled, intercellular airspace resistance was small (2% of the total resistance in saturating irradiance conditions at 25 °C at stomatal opening diameter of 4 µm) whereas the liquid phase resistance was significant (23% for mesophyll and chloroplasts in the same ‘base case’). The absorption of CO2 into water at cell surfaces caused additional (strongly temperature dependent) resistance which accounted for 36% of the total resistance in the base case.  相似文献

Leaves of Peperomia camptotricha contain three distinct upper tissue layers and a one-cell thick lower epidermis. Light and dark CO2 fixation rates and the activity of ribulose bisphosphate carboxylase/oxygenase and several C4 enzymes were determined in the three distinct tissue layers. The majority of the C4 enzyme activity and dark CO2 fixation was associated with the spongy mesophyll, including the lower epidermis; and the least activity was found in the median palisade mesophyll. In contrast, the majority of the C3 activity, that is ribulose bisphosphate carboxylase/oxygenase and light CO2 fixation, was located in the palisade mesophyll. In addition, the diurnal flux in titratable acidity was greatest in the spongy mesophyll and lowest in the palisade mesophyll. The spatial separation of the C3 and C4 phases of carbon fixation in P. camptotricha suggests that this Crassulacean acid metabolism plant may have low photorespiratory rates when it exhibits daytime gas exchange (that is, when it is well watered). The results also indicate that this plant may be on an evolutionary path between a true Crassulacean acid metabolism plant and a true C4 plant.  相似文献

Leaflets of Vicia faba L. were pulse labeled with 14CO2 and the kinetics of 14C-sucrose redistribution among individual tissues was followed. Sucrose specific activity in the whole leaf peaked about 15 minutes after labeling and declined with a half-time of about 80 minutes. In one experiment, leaflet discs taken at various times during the 12CO2 chase were quick frozen, freeze-substituted, and embedded in plastic. The tissue was sectioned paradermally and sections of palisade parenchyma, of spongy parenchyma, and of spongy parenchyma that contained veins were collected. Water extracts from these sections were assayed for sucrose specific activity. Sucrose specific activity in the palisade parenchyma was higher than that of the spongy parenchyma and reached a maximum in both tissues 9 to 15 minutes after labeling. Sucrose specific activity initially declined rapidly in the palisade parenchyma followed by a period during which little or no loss occurred. Sucrose specific activity in sections containing veins peaked at 15 minutes with a maximum value substantially higher than either mesophyll tissue, indicating that recently synthesized sucrose was preferentially exported from the mesophyll. Decline of activity in these sections containing veins continued for the remainder of the experiment. Sucrose specific activity in lower epidermal peels peaked several minutes after that of the whole leaflet and remained lower. Sucrose specific activity in upper epidermal peels was variable (probably due to contamination), but the limited data suggest that the sucrose specific activity there reached somewhat higher values than those of the lower epidermis. The experiments indicate that each leaf tissue contains a kinetically identifiable sucrose pool (which we refer to as “histological compartmentation”), and that further compartmentation may occur at the intracellular level. When particles move through fluids, they produce far-field pressure differences and near-field fluid deformations. Here we evaluate if a copepod, relying on mechanoreceptive antennulary setal hairs, can detect pressure changes caused by a variety of signal sources. We first provide a correction of the copepod mechanoreception model of Legier-Visser et al. (1986), showing how an object above a minimum size should be detectable. The pressure change P created by an object of this minimum size was 385 dynes/cm2, based on biomechanical relationships for a rigid seta bending with respect to the exoskeletal body and using the neurophysiological detection threshold of a 10 nm bend of the sensory seta (Yen et al., 1992). The P for: a 3 m particle = 0.01 dynes/cm2, a 50 m particle = 0.16 dynes/cm2, an escaping nauplius = 78 dynes/cm2, a revolving prey = 10–5 dynes/cm2, a 1 mm copepod escaping at 1 m/s at a distance of 1 mm from the mechanoreceptive sensory hairs of its captor = 312 dynes/cm2. Only the copepod escaping at high-speed close to the captor would create a pressure difference that could elicit a response. At this point, we conclude that pressure differences are rarely of a magnitude that is perceptible and that additional information must be derived for a copepod to detect prey. Other signals include fluid deformations as well as other types of stimuli (odor, shadows). Like most organisms, a copepod will rely on all sensory modalities to find food, avoid predators, and track mates, assuring their survival in the aquatic environment. It also is possible that the biomechanical model is insufficient for estimating pressure differences causing the cuticular deformation or that further analysis is necessary to improve our certainty of the sensitivity of the copepod seta.  相似文献

An enzymatic digestion procedure is presented which allows for the isolation of large quantities of mesophyll cells and minor veins of Nicotiana tabacum L. cv. ‘Wisconsin 38’. Isolated mesophyll cells exhibited CO2 fixation rates in excess of 150 μmoles CO2 mg chl-1hr-1, while bundles consistently fixed CO2 at much lower rates, viz. 10–20 μmoles CO2 mg chl-1hr-1. Various physical and biochemical parameters of the isolates have been compared with intact leaf disks.  相似文献

Light gradients were measured in leaves that had different types of anatomical development of the mesophyll but similar pigment content. Leaves of the legume, Thermopsis montana, had columnar palisade and spongy mesophyll whereas leaves of the monocot, Smilacina stellata, had spongy mesophyll only. Light gradients were measured at 550 nm in both types of leaves when they were irradiated with collimated or diffuse light. When irradiated with collimated light, light gradients were steeper in leaves with spongy mesophyll in comparison to those that had palisade tissue. On the other hand, light gradients were similar between both leaf types when they were irradiated with diffuse light. Thus, columnar palisade cells facilitated the penetration of collimated light over diffuse light. These results suggest that palisade tissue may help distribute light more uniformly to chloroplasts within the leaf. Moreover, the functional significance of palisade tissue may be related to the amount of collimated light within the natural environment.  相似文献

The viscosity change of myosin A concentrated solution with or without other components was measured as the incubation time elapsed at 30°C.

The viscosity of myosin A solution increased, but that of F-actin solution did not. The shear stress at 0.04 sec?1 was not increased to 1.0 dyne/cm2 in the former, but in the latter was below 0.5 dyne/cm2.

The viscosity of myosin B solution increased slightly, but that of native tropomyosin-free myosin B solution decreased remarkably. In both the shear stress at 0.04 sec?1 was greater than or equal to 15 dynes/cm2.

The speed of the viscosity increase in the presence of 3 mm pyrophosphate and 3 mm MgCl2 was higher in concentrated solution of myosin B than in that of native tropomysin-free myosin B. The shear stress at 0.04 sec?1 after 6 hr at 30°C was 11.5 and 8.2 dynes/cm2, respectively.

Morphological, anatomical and physiological summer and winter leaf traits of Cistus incanus subsp. incanus, C. salvifolius and C. monspeliensis growing at the Botanical garden of Rome were analyzed. With regard to differences between summer and winter leaves of the considered species, leaf thickness (L) was 21% higher in summer than in winter leaves (mean of the considered species) and this increase was mostly the result of the increased palisade parenchyma thickness over the spongy parenchyma one (24 and 16% higher in summer than in winter leaves, respectively). Leaf mass area (LMA) and leaf tissue density (LTD) were 38% and 17% higher in summer than in winter leaves, respectively (mean of the considered species). The photosynthetic rate (PN), stomatal conductance (gs) and chlorophyll content (Chl) of summer leaves were 54%, 17% and 14% lower, respectively, than in winter leaves. C. monspeliensis summer leaves had the highest LMA, LTD, adaxial cuticle thickness (14.6 ± 1.8 mg cm−2, 1091 ± 94 mg cm−3, and 5.8 ± 1.7 μm, respectively) and the lowest mesophyll intercellular spaces (fias 38 ± 3%). Moreover, C. monspeliensis had the highest PN in summer (2.6 ± 0.1 μmol m−2 s−1) and C. incanus the highest PN and WUE (84% and 59% higher than the other species) in the favorable period, associated to a higher fias (42 ± 2%). C. salvifolius had the highest PN (54% higher than the other species) in winter. The plasticity index could allow a better interpretation of the habitat preference of the considered species. The physiological plasticity (PIp = 0.39, mean value of the considered species) was higher than the morphological (PIm = 0.22, mean value) and anatomical (PIa = 0.13, mean value) plasticity. Moreover, among the considered species, C. salvifolius and C. incanus are characterized by a larger PIa (0.14, mean value) which seems to be correlated with their wider ecological distribution and the more favorable conditions of the environments where they naturally occur. The highest PIm (0.29) of C. monspeliensis indicates that it can play a high adaptive role in highly stressed environments, like fire degraded Mediterranean areas in which it occurs.  相似文献
