A field strain of Spodoptera littoralis Biosduval was selected against Cry1C toxin derived from Bacillus thuringiensis entomocidus for 10 subsequent generations under laboratory conditions. Selection pressure resulted in a 29‐fold resistance ratio compared with the susceptible strain. Inheritance of Cry1C resistance was partially dominant and autosomal on the basis of bioassay response to Cry1C toxin in a reciprocal cross between male and/or female F1. Consistent with earlier findings, resistance was recessive at high concentrations of Cry1C toxin. However, the dominance of resistance increased as the concentration of Cry1C decreased. Analysis of survival and growth of progeny from a backcross (F1 × resistance strain) suggested that resistance was controlled by either a single or a few loci in cotton leafworm.  相似文献

Hosts are often target to multiple simultaneous infections by genetically diverse parasite strains. The interaction among these strains and the interaction of each strain with the host was shown to have profound effects on the evolution of parasite traits. Host factors like genetic architecture of resistance have so far been largely neglected. To see whether genetic architecture differs between different kinds of infections we used joint scaling analysis to compare the genetic components of resistance in the red flour beetle Tribolium castaneum exposed to single and multiple strains of the microsporidian Nosema whitei. Our results indicate that additive, dominance and epistatic components were more important in single infections whereas maternal components play a decisive role in multiple infections. In detail, parameter estimates of additive, dominance and epistatic components correlated positively between single and multiple infections, whereas maternal components correlated negatively. These findings may suggest that specificity of host–parasite interactions are mediated by genetic and especially epistatic components whereas maternal effects constitute a more general form of resistance.  相似文献

Two point mutations (F1845Y and V1848I) in the voltage-gated sodium channel gene of Plutella xylostella are involved in the target-site resistance to sodium channel blocker insecticides (SCBIs). The contribution of the individual mutations to the SCBI resistance and the associated inheritance modes is as yet unclear. Through 2 rounds of single-pair crossing and marker-assisted selection, 2 P. xylostella strains (1845Y and 1848I) bearing homozygous F1845Y or V1848I mutant alleles were successfully established from a field-collected population, and the contribution of each mutation to SCBI resistance, as well as associated inheritance patterns, was determined. When compared with the susceptible SZPS strain, each of the mutations individually conferred equally high-level resistance to indoxacarb (378 and 313 fold) and metaflumizone (734 and 674 fold), respectively. However, dominance levels of resistance to SCBIs were significantly different between the 2 resistant strains. Resistance of the 1845Y strain to indoxacarb and metaflumizone was inherited as an autosomal and incompletely dominant trait (D values ranged from 0.43 to 0.76). In contrast, that of the 1848I strain followed an autosomal but incompletely recessive to semidominant mode (D values: −0.24 to 0.09). Our findings enriched the current understanding of inheritance and mechanisms of SCBI resistance in P. xylostella, and will help develop resistance management programs for P. xylostella and other economic pests.  相似文献

A field population of Typhlodromus pyri (Acari: Phytoseiidae) tolerant to mancozeb was selected in the laboratory. After 10 mancozeb selections the LC50 value for mancozeb was 73 times higher in the selected-10 strain compared to the standard susceptible strain. A genetic analysis using reciprocal crosses and backcrosses of female F1 progeny found no maternal effect. Resistance in the selected-10 strain was codominant in expression, dominance value was about −0.1. Backcrosses between F1 females and the susceptible strain indicate that the resistance to mancozeb could be principally conferred by a predominant gene, but additional factors would also be involved.  相似文献

Summary Protein synthesis by ribosomes from several cryptopleurine-resistant yeast mutants is also resistant to emetine and tubulosine. These mutants can be classified into two different types: Class I mutants which display high levels of resistance to emetine and tubulosine and Class II mutants that are only weakly resistant to tubulosine and are slightly more sensitive to emetine than those of Class I. Apparently all mutants have similar levels of resistance to cryptopleurine. The distinct phenotypes of Class I and Class II strains are expressed through their 40S ribosomal subunit. Genetic analysis has shown that the mutations to cryptopleurine resistance are allelic and that in a particular case (strain CRY6) the pleiotropic phenotype is a result of the expression of the cryl locus. It is suggested that Class I and Class II mutants arise from two independent mutational events within the cryl allele. in heterozygous (+/cryl) diploids both the sensitive and the resistant genes are expressed as shown by studies of the action of cryptopleurine on polyphenylalanine-synthesizing system derived from each parental sensitive and resistant haploid strain and heterozygous diploid strains. The apparent dominance of sensitivity over resistance which may be observed in vivo in heterozygous (+/cryl) diploids has been explained in terms of the mode of action of the inhibitors.  相似文献

Reciprocal crossing of the Mt Alford (A) strain of the cattle tick B. microplus with a susceptible (S) strain and phenotype analysis of F1, testcross and F2 progeny showed that high chlorpyrifos resistance in strain A was due to two genes that were complementary and jointly exhibited incomplete dominance. Diazinon resistance in the Gracemere (G) strain appeared to be similarly inherited. The 'average' degree of dominance ('average dominance', Dav) of high chlorpyrifos resistance over susceptibility, exhibited by F1 hybrids from A X S reciprocal crossings, was +0.54 on a -1 to +1 scale and was not significantly different from the parametric value of +0.5 (semi-dominance). The corresponding Dav values revealed by G X S crossings were +0.42 for diazinon resistance (significantly less than +0.5) and -0.031 for chlorpyrifos resistance (not significantly different from zero and therefore exhibiting zero dominance/recessivity). Resistance factors for chlorpyrifos in strains A and G for homozygotes were 74 and 35, respectively, and for F1 hybrids were 25-29 and 5-7, respectively. The resistance factors for diazinon in strain G for homozygotes and F1 hybrids were 174 and 37-41, respectively.  相似文献

Cross‐resistance, resistance mechanisms, and mode of inheritance of spinosad resistance were studied in the western flower thrip, Frankliniella occidentalis (Pergande). Spinosad (naturalyte insecticide) showed low cross‐resistance to prothiophos (organophosphorus insecticide) and chlorphenapyr (respiratory inhibitor) showed some cross‐resistance to thiocyclam (nereistoxin). The synergists PBO (piperonyl butoxide), DEM (diethyl maleate), and DEF (s, s, s‐tributyl phosphorotrithioate) did not show any synergism on the toxicity of spinosad in the resistant strain (ICS), indicating that metabolic‐mediated detoxification was not responsible for the spinosad resistance, suggesting that spinosad may reduce sensitivity of the target site: the nicotinic acetylcholine receptor and GABA receptor. Following reciprocal crosses, dose‐response lines and dominance ratios indicated that spinosad resistance was incompletely dominant and there were no maternal effects. The results of backcross showed that spinosad resistance did not fit a single‐gene hypothesis, suggesting that resistance was influenced by several genes.  相似文献

Summary The location of the characters for rifampicin-resistance in E. coli linked to the arg E marker was confirmed by mating experiments with rifampicin-resistant Hfr C mutants and a rifampicin-sensitive acceptor strain. During these tests, temporary dominance of rifampicin-sensitivity was observed. On the basis of the results obtained, the time required for complete phenotypic expression of rifampicin-resistance was determined, and the ratios of rifampicin-sensitivity to rifampicin-resistance in the merozygote phase of the cell were studied. It was found that the episomal marker of rifampicin-sensitivity influences the level of resistance in merodiploid cells, but does not fully restore the original sensitivity of the wild type.  相似文献

The diamondback moth (DBM), Plutella xylostella (L.), is a major pest of cruciferous crops. Tebufenozide, a novel nonsteroidal ecdysone agonist, exhibits good efficacy and has played an increasingly important role in the control of Lepidopteran pests in China. For its resistance management, the genetic basis of tebufenozide resistance was studied using a laboratory selected resistant strain of DBM (resistant ratio, RR = 268). A series of crosses with laboratory susceptible and resistant strains revealed that tebufenozide resistance in this pest was partially biased toward female heredity, with a large difference in RR for F1 (RR = 29) and rF1 progeny (RR = 147). The dominance calculated for these 2 cross progeny was ?0.788 and 0.09, respectively. Further analysis showed that the susceptible male and female larvae were similar in their sensitivity to tebufenozide, but the resistant female larvae showed significantly higher resistance than the resistant male larvae. The heredity of tebufenozide resistance in DBM might be linked with the W sex chromosome, which suggested that DBM has the ability to develop high levels of resistance to tebufenozide. This is the first report of sex‐linked inheritance of tebufenozide resistance in P. xylostella (L.).  相似文献

We studied inheritance of resistance to dicrotophos in greenhouse whitefly,Trialeurodes vaporariorum Westwood (Homoptera, Aleyrodidae). Compared with females from a field-collected susceptible strains (S), females from a resistant strain (R) ofT. vaporariorum derived from heavily treated cotton fields had a 28-fold greater LC50 to dicrotophos in laboratory bioassays. Concentration-mortality lines obtained from female progeny of reciprocal F1 crosses (R XS and S XR) were similar, suggesting that inheritance of dicrotophos resistance was autosomal and not influenced by maternal effects. Responses of F1 female progeny were similar to those of the parental S strain, indicating that the resistance was partially recessive (degree of dominance, D, was −0.61). Mortality observed in female progeny obtained from a backcross (F1♀ XR) corresponded more closely to expectations derived from polygenic models than to expectations from a monogenic model. The estimated number of effective factors (sensu Lande, 1981) contributing equally to resistance was three.  相似文献

Gravid female phytoseiid mites, Amblyseius fallacis (Garman), were selected for resistance to permethrin using a table-top Petri dish (PD) technique. After 55 selections, the LC50 of the R population increased 964-fold to 12 241 p.p.m. (PD) and 3.6-fold to 167 p.p.m. by a slide-dip (SD) method. A genetic analysis was conducted to examine the mode of inheritance by a series of single-pair reciprocal interstrain crosses and backcrosses of female F1 progeny. Concentration-mortality regressions (PD) for parental populations showed a 69.4-fold difference between R and S strains. Regressions for F1 females resulting from both interstrain crosses were intermediate between parental strains and not significantly different, showing no obvious maternal effect. Resistance in the R strain was not completely dominant or recessive and the estimated dominance, D, for the F1 females from combined data of both reciprocal crosses was-0.18, for the female R x male S cross it was 0.24 and for the female S x male R cross it was-0.01. We concluded that more than one gene was responsible for the resistance in the selected R strain. No incompatibility was observed in any reciprocal interstrain crosses or backcrosses between the S strain and R strain.  相似文献

Laboratory selection increased resistance to the Bacillus thuringiensis toxin Cry1C in a strain of diamondback moth (Plutella xylostella). The selected strain was derived from a field population that had evolved high levels of resistance to Bacillus thuringiensis subsp. kurstaki and moderate resistance to Cry1C. Relative to the responses of a susceptible strain of diamondback moth, the resistance to Cry1C of the selected strain increased to 62-fold after six generations of selection. The realized heritability of resistance was 0.10. Analysis of F(inf1) hybrid progeny from reciprocal crosses between the selected strain and a susceptible strain showed that resistance to Cry1C was autosomally inherited. The dominance of resistance to Cry1C depended on the concentration; inheritance was increasingly dominant as the concentration decreased. Responses of progeny from single-pair families showed that resistance to Cry1C and resistance to Cry1Ab were inherited independently, which enhances opportunities for managing resistance. However, compared with projections based on previously reported recessive inheritance of resistance to Cry1A toxins, the potentially dominant inheritance of resistance to Cry1C observed here could accelerate evolution of resistance.  相似文献

就小菜蛾Plutella xylostella对阿维菌素的抗性遗传方式和抗性品系的相对适合度进行了研究。室内选育的阿维菌素抗性品系与同源的敏感品系杂交、F1代自交、F1代与亲本回交,结果表明:杂交后的显性度(D)分别为-0.64和-0.52,说明小菜蛾对阿维菌素的抗性是常染色体、不完全隐性遗传;χ2检验证实,可能是多基因控制的抗性遗传。杂交F1代乙酰胆碱酯酶(AChE)、羧酸酯酶(CarE)和谷胱甘肽转移酶(GST)活性比抗性亲本有所降低,F2代及回交后代三种酶的活性继续降低。种群适合度研究表明,抗性品系相对于敏感品系有0.84的适合度。  相似文献

Summary Studies on the genetics of leaf blight caused byAlternaria triticina using generation mean analysis revealed that additive components played a major role, but that dominance components also contributed significantly in controlling the variability for leaf blight resistance in wheat crosses. Furthermore, the additive x additive type of epistasis was predominant in the first three crosses, whereas in the fourth cross additive x dominance (j) and dominance x dominance (1) components of epistasis were most significant. Because of this it may be desirable to follow a simple recurrent selection scheme for higher tolerance, to isolate resistant plants from the segregating populations derived from crosses of parents of diverse origin following the pedigree method of breeding. CPAN-1887 was very tolerant to leaf blight in the present study and should be utilized in hybridization programs to develop leaf-blight-resistant varieties.  相似文献

Six generations, consisting of three resistant parents, three susceptible parents, their 15 possible F1 crosses, 15 F2's, 15 BC1's (F1 x resistant female parent) and 15 BC2's (F1 x susceptible male parent) were analysed following Hayman (Heredity 12: 371–390, 1958) to evaluate the nature and type of gene action governing resistance to H. turcicum. The results showed that all types of gene effects, viz., additive, dominance and epistasis (i.e., additive x additive, additive x dominance and dominance x dominance) were operating in one cross or the other in controlling resistance. However, it was additive gene action and dominance x dominance type of epistasis with duplicate nature that were important in controlling resistance in most crosses. Depending upon the final objectives, one of the breeding methods, viz., recurrent selection, heterosis breeding, back-cross method or full-sib selection (bi-parental mating) may be followed.  相似文献

A half diallel was made amongst five wheat (Triticum aestivum L.) genotypes of which one was susceptible, while the others had adult-plant resistance, to stripe rust (Puccinia striiformis West.). The five parent and ten F1 progeny were grown in the glasshouse and were inoculated with three rust pathotypes at the seedling stage. The latent period was measured on the first leaf. Two procedures were used to analyze the half diallel. Both methods showed that the average effects of alleles were of much greater importance than was dominance in conditioning resistance in response to two of the pathotypes, while for the third pathotype dominance was important. Resistance was conditioned by partial dominance for two pathotypes whereas for the third it was determined by full dominance. Broad-sense heritabilities range from 60–73% and the number of genes involved was different (from 1 to 4), depending on the pathotype.  相似文献

In order to investigate on inheritance and gene action for resistance to yellow rust, the resistant line C.B227 was crossed with the susceptible variety Avocet. Parents (P1 and P2) and the resulting F1, F2 and F3 generations were planted in a randomised complete block design with two replications in the field. The plants were inoculated with 70E0A+ pathotype of yellow rust in the research station of Gharakhil, Iran, and evaluated for resistance at adult plant stage. Disease severity and infection type of flag leaf were recorded for each single plant and final coefficient of infection was calculated. The results of weighted ANOVA indicated that the difference among the generations was significant (p?<?0.01) for the trait final infection type. Generation mean analysis showed that dominant effect was more important than additive one. The degree of dominance indicated the presence of complete dominance. Additive, dominance and epistasic additive?×?additive [i] effects were important in genetic control of resistance. The results of generation variance analysis were consistent with generation mean analysis.  相似文献

The community composition and ecophysiological features of microbial autotrophic biofilms were studied in Fuente Podrida, a cold sulfur spring located in East Spain. We demonstrated how different ecophysiological strategies, such as resistance and/or utilization of sulfide and oxygen, light adaptation, or resistance to high water flow, allow each of the microorganisms described to efficiently colonize several areas within the environmental gradient. In the zone of the spring constantly influenced by sulfide-rich waters, biofilms were formed by purple bacteria, cyanobacteria, and filamentous colorless sulfur bacteria. Purple bacteria showed higher photosynthetic efficiency per pigment unit than cyanobacteria, although they were dominant only in anoxic areas. Two filamentous cyanobacteria, strain UVFP1 and strain UVFP2, were also abundant in the sulfide-rich area. Whereas the cyanobacterial strain UVFP2 shows a strategy based on the resistance to sulfide of oxygenic photosynthesis, strain UVFP1, additionally, has the capacity for sulfide-driven anoxygenic photosynthesis. Molecular phylogenetic analyses cluster the benthic strain UVFP1 with genus Planktothrix, but with no particular species, whereas UVFP2 does not closely cluster with any known cyanobacterial species. The colorless sulfur bacterium Thiothrix sp. extended throughout the zone in which both sulfide and oxygen were present, exhibiting its capacity for chemolithoautotrophic dark carbon fixation. Downstream from the source, where springwater mixes with well-oxygenated stream water and sulfide disappears, autotrophic biofilms were dominated by diatoms showing higher photosynthetic rates than cyanobacteria and, by a lesser extent, by a sulfide-sensitive cyanobacterium (strain UVFP3) well adapted to low light availability, although in the areas of higher water velocity far from the river shore, the dominance shifted to crust-forming cyanobacteria. Both types of microorganisms were highly sensitive to sulfide impeding them from occupying sulfide-rich areas of the spring. Sulfide, oxygen, light availability, and water velocity appear as the main factors structuring the autotrophic community of Fuente Podrida spring. 以兰州吐鲁沟公园的二点叶螨Tetranychus urticae 为敏感品系,分别用氧乐果、甲氰菊酯、四螨嗪喷雾处理15次,其抗性水平分别为38.5、479.8和67.3倍。将抗性品系分别与敏感品系进行杂交和回交的结果表明:抗氧乐果品系的显性系数DRS(R♀×S)为0.4700、DSR(S♀×R)为0.4749;抗甲氰菊酯品系的DRS(R♀×S)为0.5155, DSR为0.5237;抗四螨嗪品系的DRS为0.3134, DSR为0.2466。表明二点叶螨对这3种药剂的抗性均是由单个不完全显性基因所控制。在连续10个月不接触药剂的情况下,3个抗性品系的抗药性都有下降,抗氧乐果品系的抗性下降最快,只有敏感品系的抗性倍数的3.6倍;抗甲氰菊酯种群的抗性下降较慢,为敏感品系的95.9倍。再经甲氰菊酯、氧乐果、四螨嗪分别连续15次喷雾处理后,3个抗性种群的抗性水平又再度回升,抗甲氰菊酯品系回升较快,抗性为敏感品系的523.5倍,抗四螨嗪品系次之,抗氧乐果的品系抗性恢复最慢。  相似文献

Four caffeine-resistant haploid isolates, two resistant to 50 mM caffeine and two resistant to 100 mM caffeine, were genetically analyzed. Complementation and tetrad analysis indicated that all four mutations are alleles of the same locus. All four isolates demonstrated incomplete dominance when hybridized to the wild-type strain and dominance of high to low resistance when hybridized to one another. Differences in caffeine resistance were found between wild-type grande cells and its petite derivative.  相似文献
