In an open-field experiment, 50-year-old trees of Scots pine (Pinus sylvestris L.) were fumigated with low concentrations of SO2 and NO2 (10–15 nl I?1) during the growing season in four consecutive years (1988 to 1991). Results from the autumn and early winter of 1991 and 1992 are presented. The maximum photochemical efficiency of photosystem II (PSII), as indicated by the ratio of variable to maximum fluorescence (Fv/FM) was assessed in current and one-year-old needles from the top and the bottom of the canopy. Furthermore, simultaneous measurements of photosynthetic O2 evolution and chlorophyll fluorescence were made in current-year needles at 20°C. In general, the Fv/FM ratio as well as the gross rate of O2 evolution in needles of fumigated trees was not significantly different from that in needles of control trees during the fumigation period. However, both current and one-year-old needles sampled in November and December 1991 from the top of the canopy of fumigated trees had significantly lower Fv/FM values than corresponding needles of control trees. Similar differences in Fv/FM correlated with the treatments were observed in needles from the bottom of the canopy, indicating that the depression of Fv/FM in needles of fumigated trees was not due to an increased susceptibility to photoinhibition. In 1992, when no fumigation occurred, differences in Fv/FM between the treatments were not significant during autumn and early winter. The gross rate of O2 evolution at high irradiances was significantly lower in current-year needles of fumigated trees sampled in November and December 1991 than in those of control trees. Furthermore, a nearly identical linear relationship between the quantum yield of PSII electron transport determined from chlorophyll fluorescence and the quantum yield of O2 evolution (gross rate of O2 evolution/PPFD) was found during autumn and early winter. This appeared to be largely a result of changes in the thermal energy dissipation within PSII. The observed differences in photosynthetic characteristics correlated with the different treatments after the fumigation period is suggested to be mainly caused by increased sensitivity of the needles of fumigated trees to low and subfreezing temperatures. However, current-year needles of fumigated trees tended to have a lower N content than those of control trees, which may partly explain the differences in gross photosynthesis between fumigated and control trees.  相似文献

Mature Pinus sylvestris trees were exposed to air-pollution strees in an open field fumigation experiment. The trees were exposed to low-levels of sulphur dioxide and nitrogen oxides throughout the whole growing season (June to October 1991). Three age-classes (current, one- and two-year-old) of needles were collected from exposed and control trees and the expression of chloroplastic and cytosolic CuZn-superoxide dismutase (SOD; EC 1.15.1.1) genes was studied. The levels of chloroplastic and cytosolic CuZn-SOD mRNA were significantly (up to 2.3-fold) higher in exposed trees for all age-classes of needles as compared to control trees. No significant differences were observed between different needle age-classes in CuZn-SOD mRNA induction. The chloroplastic and cytosolic CuZn-SOD mRNA were induced to the same extent, however, the level of chloroplastic CuZn-SOD mRNA was higher than the cytosolic one. Small differences were observed in CuZn-SOD mRNA levels for individual trees both within exposed and control areas. Protein activity measurements of the CuZn-SOD isozymes did not reveal any significant differences between control and exposed trees. The results are discussed in relation to protection against oxidant stress from air pollutants.  相似文献

The application of gibberellin A4/7 (GA4/7) to the stem of previous-year (1-year-old) terminal shoots of Scots pine (Pinus sylvestris) seedlings has been observed to stimulate cambial growth locally, as well as at a distance in the distal current-year terminal shoot, but the distribution and metabolic fate of the applied GA4/7, as well as the pathway of endogenous GA biosynthesis in this species, has not been investigated. As a first step, we analysed for endogenous GAs and monitored the transport and metabolism of labelled GAs 4, 9 and 20. Endogenous GAs from the elongating current-year terminal shoot of 2-year-old seedlings were purified by column chromatography and high-performance liquid chromatography and analysed by combined gas chromatography-mass spectrometry (GC-MS). GAs 1, 3, 4, 9, 12 and 20 were identified in the stem, and GAs 1, 3 and 4 in the needles, by full-scan mass spectrometry (GAs 1, 3, 4, 9 and 12) or selected-ion monitoring (GA20) and Kovats retention index. Tritiated and deuterated GA4, GA9 or GA20 were applied around the circumference at the midpoint of the previous-year terminal shoot, and metabolites were extracted from the elongating current-year terminal shoot, the application point, and the 1-year-old needles and the cambial region above and below the application point. After purification, detection by liquid scintillation spectrometry and analysis by GC-MS, it was evident that, for each applied GA, unmetabolised [2H2]GA and [3H]radioactivity were present in every seedling part analysed. Most of the radioactivity was retained at the application point when [3H]GA9 and [3H]GA20 were applied, whereas the largest percentage of radioactivity derived from [3H]GA4 was recovered in the current-year terminal shoot. It was also found that [2H2]GA9 was converted to [2H2]GA20 and to both [2H2]GA4 and [2H2]GA1, [2H2]GA4 was metabolised to [2H2]GA1, and [2H2]GA20 was converted to [2H2]GA29. The data indicate that for Pinus sylvestris shoots (1) GAs applied laterally to the outside of the vascular system of previous-year shoots not only are absorbed and translocated extensively throughout the previous-year and current-year shoots, but also are readily metabolised, (2) the GA metabolic pathways found are closely related to the endogenous GAs identified, and (3) GA9 metabolism follows two distinctly different routes: in one, GA9 is converted to GA1 through GA4, and in the other it is converted to GA20, which is then metabolised to GA29. The results suggest that the late 13-hydroxylation pathway is an important route for GA biosynthesis in shoots of Pinus sylvestris, and that the stimulation of cambial growth in Scots pine by exogenous GA4/7 may be due to its conversion to GA1, rather than to it being active per se.  相似文献

Foliar elements were analysed in Scots pine, Sitka spruce and Norway spruce over a 6 year period before and during continuous exposure to SO2 and O3 in an open-air fumigation experiment. Sulphur dioxide treatment elevated foliar sulphur concentration in all species, and there were increases in foliar nitrogen in the two spruce species but not in pine. The concentrations of cations were frequently increased by SO2 treatment, but there was no correlation between the sulphur concentration of needles and their total cation charge. SO2-related elevations of foliar magnesium were correlated with the concentration of this element in soil solution, but the mechanism by which other cations were enhanced remains unclear. The only consistent effects on nutrient ratios were for SO2 treatments to increase sulphur/cation ratios.  相似文献

The possibility to induce nitrate reductase (NR; EC 1.6.6.2) in needles of Scots pine ( Pinus sylvestris L.) seedlings was studied. The NR activity was measured by an in vivo assay. Although increased NR activities were found in the roots after application of NO3, no such increase could be detected in the needles. Detached seedlings placed in NO3 solution showed increasing NR activities with increasing NO3 concentrations. Exposure of seedlings to NOx (70–80 ppb NO2 and 8–12ppb NO) resulted in an increase of the NR activity from 10–20 nmol NO2 (g fresh weight)−1 h−1 to about 400 nmol NO2 (g fresh weight)−1 h−1. This level was reached after 2–4 days of exposure, thereafter the NR activity decreased to about 200 nmol NO2 (g fresh weight)−1 h−1. Analyses of free amino acids showed low concentrations of arginine and glutamine in NOx-fumigated seedlings compared to corresponding controls.  相似文献

The carbohydrate metabolism of the needles of Scots pine (Pinus sylvestris) and Norway spruce (Picea abies) has been examined in trees that were exposed to SO2, and O3, in an open-air fumigation experiment located in the Liphook forest in southern England. Two-year-old seedlings were planted in 1985 in seven experimental plots. Five plots received fumigation treatments of SO2, O3 or a combination of these gases to give a 2 × 3 factorial design with one additional ambient plot Fumigation with SO2, occurred from May 1987 to December 1990 and O3, fumigation occurred from March to December 1988, May to December 1989 and February to December 1990. Five samples of needles for investigation of carbohydrate metabolism were taken between February and July 1989. The concentrations of soluble carbohydrates (including sucrose and hexoses) were greatly reduced in the needles taken from Scots pine growing in the treated plots, and were also reduced, but to a lesser extent, in the needles taken from Norway spruce. Little variation in the concentration of starch in the needles of either species was detected. The activities of the two final enzymes of sucrose synthesis, sucrose phosphate synthase and sucrose 6-phos-phate phosphatase, were greatly reduced in the needles of Scots pine and were also reduced, but to a lesser extent, in the needles of Norway spruce in the fumigated plots. These reductions could be correlated with decreases in rates of photosynthetic CO2 assimilation determined by independent groups of researchers working on the Liphook site.  相似文献

Abstract. The kinetics of in vivo chlorophyll fluorescence of photosystem II (PS II) was measured at room temperature and 77 K during frost hardening of seedlings of Scots pine (Pinus sylvestris L.), and after exposure of frost-hardened shoots to sub-freezing temperatures. A more pronounced decrease in variable fluorescence yield for the upper exposed than for the lower shaded surface of the needles suggested that some photoinhibition occurred during prolonged frost hardening at 50 μmol photons m?2 s?1 and 4°C. Reversible inhibition of photosynthesis after exposure to sub-freezing temperatures was initially manifested as an increase of steady-state energy-dependent fluorescence quenching (qE) and a reduction in the rate of O2 evolution. Further inhibition after treatment at still lower temperatures caused a progressive decline of steady-state photochemical quenching (qQ) and the rate of O2 evolution, whereas qE remained high. This implies an inactivation of enzymes in the photosynthetic carbon reduction cycle decreasing the consumption of ATP and NADPH, which is likely to cause an increase of membrane energization and a reduction of the primary electron acceptor (QA) of PS II. Alternatively, the changes in qQ and qE might be attributed to an inhibition of photophosphorylation. Severe, irreversible damage to photosynthesis resulted in a suppression of qE and of variable fluorescence yield, probably because the photochemical efficiency of PS II was impaired. Changes in the fast fluorescence kinetics at room temperature after severe freezing damage were interpreted as an inhibition of the electron flow from QA to the plastoquinone pool. It is suggested that irreversible freezing injury to needles of frost-hardened P. sylvestris causes damage to the QB,-protein.  相似文献

Naturally regenerated Scots pines (Pinus sylvestris L.), aged 28–30 years old, were grown in open-top chambers and subjected in situ to three ozone (O3) regimes, two concentrations of CO2, and a combination of O3 and CO2 treatments From 15 April to 15 September for two growing seasons (1994 and 1995). The gas exchanges of current-year and 1-year-old shoots were measured, along with the nitrogen content of needles. In order to investigate the factors underlying modifications in photosynthesis, five parameters linked to photosynthetic performance and three to stomatal conductance were determined. Elevated O3 concentrations led to a significant decline in the CO2 compensation point (Г*), maximum RuP2-saturated rate of carboxylation (Vcmax), maximum rate of electron transport (Jmax), maximum stomatal conductance (gsmax), and sensitivity of stomatal conductance to changes in leaf-to-air vapour pressure difference (?gs/?Dv) in both shoot-age classes. However, the effect of elevated O3 concentrations on the respiration rate in light (Rd) was dependent on shoot age. Elevated CO2(700 μmol mol?1) significantly decreased Jmax and gsmax but increased Rd in 1-year-old shoots and the ?gs/?Dv in both shoot-age classes. The interactive effects of O3 and CO2 on some key parameters (e.g. Vcmax and Jmax) were significant. This may be closely related to regulation of the maximum stomatal conductance and stomatal sensitivity induced by elevated CO2. As a consequence, the injury induced by O3 was reduced through decreased ozone uptake in 1-year-old shoots, but not in the current-year shoots. Compared to ambient O3 concentration, reduced O3 concentrations (charcoal-filtered air) did not lead to significant changes in any of the measured parameters. Compared to the control treatment, calculations showed that elevated O3 concentrations decreased the apparent quantum yield by 15% and by 18%, and the maximum rate of photosynthesis by 21% and by 29% in the current-year and 1-year-old shoots, respectively. Changes in the nitrogen content of needles resulting from the various treatments were associated with modifications in photosynthetic components.  相似文献

Young Scots pine trees naturally established at a pine heath were exposed to two concentrations of CO2 (ambient and doubled ambient) and two O3 regimes (ambient and doubled ambient) and their combination in open-top field chambers during growing seasons 1994, 1995 and 1996 (late May to 15 September). Filtered ozone treatment and chamberless control trees were also included in the treatment comparisons. Root ingrowth cores were inserted to the undisturbed soil below the branch projection of each tree at the beginning of the fumigation period in 1994 and were harvested at the end of the fumigation periods in 1995 and 1996. Root biomasses were determined from different soil layers in the ingrowth cores, and the infection levels of different mycorrhizal types were calculated. Elevated O3 and CO2 did not have significant effects on the biomass production of Scots pine coarse (Ø > 2 mm) or fine roots (Ø < 2 mm) and roots of grasses and dwarf shrubs. Elevated O3 caused a transient stimulation, observable in 1995, in the proportion of tuber-like mycorrhizas, total mycorrhizas and total short roots but this stimulation disappeared during the last study year. Elevated CO2 did not enhance carbon allocation to root growth or mycorrhiza formation, although a diminishing trend in the mycorrhiza formation was observed. In the combination treatment increased CO2 inhibited the transient stimulating effect of ozone, and a significant increase of old mycorrhizas was observed. Our conclusion is that doubled CO2 is not able to increase carbon allocation to growth of fine roots or mycorrhizas in nutrient poor forest sites and realistically elevated ozone does not cause a measurable limitation to roots within a period of three exposure years.  相似文献

Abstract. Net fluxes of gaseous sulphur compounds, water vapour and carbon dioxide to current- and one-year-old shoots of Scots pine were simultaneously measured in the field. The shoots were fumigated in temperature-controlled assimilation chambers with sulphur dioxide at concentrations ranging from 40 to 250 μg m−3 (15–95 ppb). The hypothesis that stomatal opening regulates the uptake of sulphur dioxide was tested. The following conclusions were reached concerning the dry deposition of sulphur dioxide to a dry coniferous shoot.
There was a marked diurnal variation in the uptake rate of sulphur dioxide. The net deposition velocity of sulphur was about three times higher during the day than during the night during July-October. Stomatal opening was not the primary factor controlling the dry deposition rate of sulphur dioxide. The net dry deposition rate was significantly smaller than the calculated rate based on stomatal conductances for water vapour.
Part of this deviation could be explained by re-emission of reduced sulphur compound(s) from the needles. The re-emission of sulphur was a light-dependent process and might be of great significance for the sulphur balance calculations.
A variable deposition velocity should be used in atmospheric transport models to account for the diurnal and seasonal variation in the surface resistance of a dry canopy.  相似文献

  1. For successful colonization of host roots, ectomycorrhizal (EM) fungi must overcome host defense systems, and defensive phenotypes have previously been shown to affect the community composition of EM fungi associated with hosts. Secondary metabolites, such as terpenes, form a core part of these defense systems, but it is not yet understood whether variation in these constitutive defenses can result in variation in the colonization of hosts by specific fungal species.
  2. We planted seedlings from twelve maternal families of Scots pine (Pinus sylvestris) of known terpene genotype reciprocally in the field in each of six sites. After 3 months, we characterized the mycorrhizal fungal community of each seedling using a combination of morphological categorization and molecular barcoding, and assessed the terpene chemodiversity for a subset of the seedlings. We examined whether parental genotype or terpene chemodiversity affected the diversity or composition of a seedling''s mycorrhizal community.
  3. While we found that terpene chemodiversity was highly heritable, we found no evidence that parental defensive genotype or a seedling''s terpene chemodiversity affected associations with EM fungi. Instead, we found that the location of seedlings, both within and among sites, was the only determinant of the diversity and makeup of EM communities.
  4. These results show that while EM community composition varies within Scotland at both large and small scales, variation in constitutive defensive compounds does not determine the EM communities of closely cohabiting pine seedlings. Patchy distributions of EM fungi at small scales may render any genetic variation in associations with different species unrealizable in field conditions. The case for selection on traits mediating associations with specific fungal species may thus be overstated, at least in seedlings.

Hydraulic resistance to water flow was measured in branches and stems of Scots pine trees ranging from 7 to 59 years of age in Thetford (East Anglia, UK). On the basis of these measurements, tree above-ground conductance was calculated and related to the amount of leaf area sustained by each tree. Branches at the crown bottom had a lower proportion of sapwood area and a lower total hydraulic conductance than branches of the same diameter at the tree top. Within branches, most of the hydraulic resistance was located near the needles. Tree above-ground conductance was positively related to tree diameter and inversely related to tree height. Compared with young trees, mature trees had about 4 times less above-ground conductance per unit of leaf area. Apparently, the increase in pathway length associated with tree height growth could be only partially compensated for by the increase in conductive capacities resulting from diameter growth. We argue that this reduction may account for reported decreases of stomatal conductance with tree age. It is suggested that the increase in branchiness associated with tree maturation may represent a compensation mechanism to reduce the overall resistance to water flow in the crowns.  相似文献

Four-year-old seedlings of Scots pine ( Pinus sylvestris L.) were exposed to filtered air (FA), and to FA supplemented with NH3 (60 and 240 μg m−3) in controlled-environment chambers for 14 weeks. Exposure to the higher NH3 concentration resulted in an increased activity of glutamine synthetase (GS, EC 6.3.1.2), and an increase in the concentrations of soluble proteins, total nitrogen, free amino acids and leaf pigments in the needles. The GS activity (μmol g−1 fresh weight h−1) in the needle extract increased to levels 69% higher than in FA and the soluble protein concentration to levels 22% higher. Total nitrogen concentration in the needles was 42% higher than in FA, while the free amino acid concentration was 300% higher, which was caused by an increase in arginine, glutamate, aspartate and glutamine. Chlorophyll a , chlorophyll b and carotenoid concentrations were 29, 38 and 11% higher, respectively. Neither the glutamate dehydrogenase (GDH, EC 1.4.1.2) activity nor the concentrations of free NH4+ and glucose in the needles were affected by exposure to NH3. After NH3 fumigation at 240 μg m−3 the starch concentration decreased by 39% relative to the FA. The results indicate that the metabolism of Scots pine acclimates to concentrations of NH3 which are 3 to 10 times higher than the average concentration in areas with intensive stock farming. The possible mechanisms underlying acclimation to NH3 are discussed.  相似文献
