The hemocyanin from the spiny lobster Panulirus interruptus, a hexamer with a molecular weight of approximately 540,000, was crystallized in space group P21 with two molecules in the unit cell and cell dimensions a = 119.8 A?, b = 193.1 A?, c = 122.2 A? and β = 118.1 °. With screened precession photographs a three-dimensional set of reflections was collected up to 10 Å resolution. Both the conventional and the fast rotation function programs were applied and gave results that were in excellent agreement with each other. The hemocyanin hexamer has 32 point group symmetry. Its 3-fold molecular axis runs approximately parallel to the crystallographic 2-fold screw axis.X-ray diffraction data to 5 Å resolution were collected by the oscillation method. Rotation function studies with data between 7 and 5 Å resolution confirmed the 10 Å studies and, furthermore, showed that the rotation axes relating subunits within one hexameric molecule can be distinguished from the rotation axes relating subunits belonging to different hexamers in the unit cell. The local 3-fold axis in the hexamer makes an angle of about 6 ° with the crystallographic 2-fold screw axis.For a mercury and a platinum derivative three-dimensional data sets were collected to 5 Å by the oscillation method. The difference Patterson of the platinum derivative could be solved. The eventual number of heavy-atom sites was 36 for the platinum derivative and 70 for the mercury derivative. From the well-occupied sites the point-group symmetry of the molecule could be established accurately. In addition, the centre of the hexamer could be located within 0.2 Å.Protein phases were obtained from isomorphous as well as anomalous differences. A “best” electron density map calculated with these phases showed the shape of the hexameric molecule as well as the boundaries of the six subunits. Correlation coefficients between the densities of the subunits showed little variation, suggesting a random distribution of the different subunit types (Van Eerd & Folkerts, 1981) over the six positions in the hexamer.The subunits are positioned at the corner of an antiprism. When viewed along the 3-fold axis the hexamer is roughly hexagonal in shape, with a diameter of approximately 120 Å. Viewed along one of the 2-fold axes the molecule is of rectangular shape with dimensions 95 Å × 120 Å. The subunit can be described as an ellipsoid of irregular shape with axes of 80 Å, 55 Å and 48 Å. Each subunit makes extensive contacts with three other subunits in the hexamer and, possibly, a much weaker contact with a fourth subunit.  相似文献

Summary Previous electrophysiological studies have shown that the olfactory organ (antennule) of the spiny lobster, Panulirus argus, has external chemoreceptors, which are selectively stimulated by adenosine 5-monophosphate (AMP) when present in seawater. Subsequent biochemical investigations revealed that AMP can be rapidly dephosphorylated by 5-ectonucleotidase/phosphatase activity associated with the olfactory sensilla (aesthetascs). In this study the deposition of cerium phosphate was used to examine the ultrastructural distribution of 5-ectonucleotidase/phosphatase activity in aesthetascs. Utilizing AMP as substrate, we found dephosphorylating activity to be associated with the outer membranes of both dendrites and auxiliary cells. Moreover, this activity was specifically localized to a narrow band that approximately corresponds to the transitional zone where dendrites develop cilia and branch extensively to form the outer dendritic segments. A similar distribution of the cerium phosphate reaction product was found when -glycerol phosphate was substituted for AMP. The alkaline-phosphatase inhibitor, levamisole, had no apparent effect on the deposition of reaction product when either AMP or -glycerol phosphate was used as substrate. 1.  The effects of the odorant compounds adenosine-5-monophosphate (5AMP), ammonium, betaine, L-cysteine, L-glutamate, DL-succinate, and taurine and of mixtures of these comounds on binding of taurine and 5AMP to dendritic membrane from the olfactory organ of spiny lobsters (Panulirus argus) were quantified to evaluate the contribution of inhibition of odorant-receptor binding to the generation of physiological responses to mixtures.
2.  Taurine binding sites belong to two affinity classes, while 5AMP binding sites belong to a single affinity class. Binding of either taurine or 5AMP was partially inhibited in an apparently noncompetitive, concentration dependent fashion by most odorant compounds, with 25–40 % inhibition by 1 mM of odorant. Mixtures of two or more odorant compounds also inhibited binding of taurine or 5AMP to its sites. However, the inhibition by mixtures was often significantly less than expected from the inhibition produced by a mixture's components assuming either a noncompetitive or competitive mechanism.
3.  By including this binding inhibition between compounds into models for predicting physiological responses to mixtures from the responses to the components, the predictive power of the models is significantly improved. This result strongly suggests that binding inhibition can influence the physiological responsiveness of chemoreceptor cells to mixtures.

Many biological processes such as reproductive and migratory behaviours have been associated with moon cycles. In this study, the nocturnal light levels associated with lunar cycle (INT) were correlated with daily catch rate of lobster P. argus, during seven lunar months of 2002 fishing period, to determine a possible relationship between these variables. The lobster catches were obtained from three fishing companies that develop their activities in the Gulf of Batabanó: EPICOL that fishes in Coloma area; PESCAHABANA in Batabanó area and PESCAISLA in Isla area. Daily catch per boat (CDB) was used as a measurement of daily catch variations (catch rate). The correlation was analyzed showing it in chronological graphs based on average of CDB per lunar phases, comparing lobster catch rate per lunar phases -with the Kruskal-Wallis test-. Spearman rank correlation coefficient and cross correlation techniques were also applied. Similarities between lobster catch rate and the lunar cycle were not found. Spearman rank correlation coefficient was modularly smaller than 0.1 in all cases and demonstrated quantitatively that correlation between CDB and INT does not exist. Kruskal-Wallis test detected differences only in Batabanó area but not when making the analyses for the whole Gulf of Batabanó. Finally, the cross correlations do not detected significance in any zone, as well. It is concluded that, in opposition to what other authors have reported, the catch rates of P. argus and the lunar cycle did not show significant correlation in the Gulf of Batabanó. This trend was independent of the fishing art, which varied according to the time of the year that was analyzed.  相似文献

Nutritional condition can affect survival and growth rate of crustaceans, and this is mostly affected by habitat conditions. This study describes the space-temporary nutritional changes in this commercially important species. With this aim, the variations in the nutritional condition (K) of lobsters from four zones (1, 2, 4 and 5) in the Gulf of Batabanó, Cuba, were determined. For this, the weight/length ratio (K=Pt/Lt) was calculated using animals captured in 1981 and 2010. The nutritional condition between areas and sexes, and years and sexes, was contrasted by a bifactorial ANOVA, and the overall length and weight of lobsters were compared using a t-Test for independent samples and unifactorial ANOVA. It was found that the nutritional condition was significantly greater in males than in females. In addition, significant variations between zones were detected for both years. Nutritional condition of lobsters from Zone five was the highest for 1981, while it was Zone two for 2010. Lobsters nutritional state showed significant variations between years, being greater in 1981 (2.34 +/- 0.84g/mm) than in 2010 (1.96 +/- 0.49g/mm). The inter-zones variations as well as the inter-annual ones seem to be related to the reported variations of the bottom type and the vegetation cover. Seasonal variations in the abundance and distribution of benthic organisms, that constitute food for lobsters, could also be influencing. The differences between sexes, however, were assumed as a consequence of the methodology used and the sexual dimorphism of the species. Due to other K estimation methods, that do not include morphometric measurements, these differences were not detected. We suggested that the P. argus nutritional condition is a good estimator of the habitat condition. Besides, according to the applied K estimation methodology, it was found that different groups of lobsters that have resemblant nutritional condition, did not necessarily observe similarities in the overall mean length or weight, so they could exist under different habitat conditions.  相似文献

Okra’s (Abelmoschus esculentus (L.) Moench) commercial cultivation is threatened in the tropics due to high incidence of yellow vein mosaic virus (YVMV) disease. Okra geneticists across the world tried to understand the inheritance pattern of YVMV disease tolerance without much success. Therefore, the inheritance pattern of YVMV disease in okra was revisited by employing six generations (\(\hbox {P}_{1}\), \(\hbox {P}_{2}\), \(\hbox {F}_{1}\), \(\hbox {F}_{2}\), \(\hbox {BC}_{1}\) and \(\hbox {BC}_{2}\)) of four selected crosses (one tolerant \(\times \) tolerant, two tolerant \(\times \) susceptible and one susceptible \(\times \) susceptible) using two tolerant (BCO-1 and Lal Bhendi) and two susceptible (Japanese Jhar Bhendi and PAN 2127) genotypes. Qualitative genetic analysis was done on the basis of segregation pattern of tolerant and susceptible plants in \(\hbox {F}_{2}\) and backcross generations of all the four crosses. It revealed that a single dominant gene along with some minor factors governed the disease tolerant trait in both the tolerant parents used. However, it was observed that genes governing disease tolerance identified in both the tolerant variety used was different. It could be concluded that the gene governing YVMV disease tolerance in okra was genotype specific. Further, duplicate gene action as evident from an approximate ratio of 15 : 1 (tolerant : susceptible) in the \(\hbox {F}_{2}\) population in the cross of two tolerant varieties gave a scope of increasing the tolerance level of the hybrid plants when both the tolerant genes are brought together. However, generation mean analysis revealed involvement of both additive and nonadditive effects in the inheritance of disease tolerance. Thus, the present study confirms that a complicated genetic inheritance pattern is involved in the disease tolerance against YVMV trait. The major tolerance genes could be transferred to other okra varieties, but the tolerance breaking virus strains might not allow them to achieve tolerance in stable condition. Therefore, accumulation of additional genes may be needed for a sustainable tolerance phenotype in okra.  相似文献
